沙漠化对科尔沁沙质草地土壤呼吸速率及碳平衡的影响   总被引:2,自引:0,他引:2  
在科尔沁沙地测定了不同类型沙漠化草地土壤呼吸速率的日进程和季节变化,以及植物碳储量和土壤有机碳储量,分析了沙漠化对沙质草地土壤呼吸速率及草地碳平衡的影响。结果表明:(1)沙漠化可导致草地土壤呼吸速率日变化幅度变小,日均土壤呼吸速率明显降低;与非沙漠化草地相比,在生长季轻度、中度、重度和严重沙漠化草地日均土壤呼吸速率依次下降6.4%、12.8%、33.0%和39.4%;(2)受沙漠化的影响草地碳储量明显下降,与非沙漠化草地相比,4种类型沙漠化草地碳储量依次下降50.3%、74.0%、86.4%和90.2%;其中,凋落物碳储量的下降幅度最大,土壤有机碳储量下降幅度次之,植物碳储量下降幅度最小;(3)沙漠化可使草地碳平衡受到严重破坏,草地一旦沙漠化,土壤呼吸消耗碳量将超过植物固定碳量,这时土壤有机碳库将作为后备碳源来弥补土壤呼吸过程中植物固碳量的不足。为了促进沙漠化草地碳库的恢复,应加强沙漠化草地的治理,促进草地植被的恢复重建。  相似文献

The determination of the phospholipid fatty acid (PLFA) pattern of soil organisms has become one of the most commonly used methods to study microbial community structure. Here we recapitulate the background of our work applying the PLFA method to soil in the early 1990s. We also stress that although the PLFA method was, and still is, a rapid and sensitive method to detect changes in the microbial community in soil, as with all popular methods it can be misused. We discuss problems in PLFA interpretation, the extent of turn-over of PLFAs in soil, and the flawed use of diversity indices to evaluate PLFA patterns.  相似文献

Heavy metal contamination in an area immediately surrounding a zinc smelter has resulted in destruction of over 485 hectares of forest. The elevated levels of heavy metals in these soils have had significant impacts on the population size and overall activity of the soil microbial communities. Remediation of these soils has resulted in increases in indicators of biological activity and viable population size, which suggest recovery of the microbial populations. Questions remain as to how the metal contamination and subsequent remediation at this site have impacted the population structure of the soil microbial communities. In the current study, microbial communities from this site were analyzed by the phospholipid fatty acid (PLFA) procedure. Principal component analysis of the PLFA profiles indicated that there were differences in the profiles for soils with different levels of metal contamination, and that soils with higher levels of metal contamination showed decreases in indicator PLFAs for mycorrhizal fungi, Gram-positive bacteria, fungi, and actinomycetes. PLFA profiles for remediated sites indicated that remediated soils showed increases in indicator PLFAs for fungi, actinomycetes, and Gram-positive bacteria, compared to unremediated metal contaminated soils. These data suggest a change in the population structure of the soil microbial communities resulting from metal contamination and a recovery of several microbial populations resulting from remediation.  相似文献

Analysis of phospholipid fatty acids (PLFAs) was performed to investigate effects of 2,4,6-trinitrotoluene (TNT) contamination and soil remediation on microbial biomass and community structure. A TNT-contaminated and an uncontaminated soil from a former ammunition plant were analysed before and after a humification/remediation process. TNT contamination reduced microbial biomass but indicated only minor differences in PLFA composition between the contaminated and uncontaminated soils. The humification process increased microbial biomass and altered soil PLFA patterns to a larger degree than did TNT contamination.  相似文献

Changes in the biomass and structure of soil microbial communities have the potential to impact ecosystems via interactions with plants and weathering minerals. Previous studies of forested long-term (1000s - 100,000s of years) chronosequences suggest that surface microbial communities change with soil age. However, significant gaps remain in our understanding of long-term soil microbial community dynamics, especially for non-forested ecosystems and in subsurface soil horizons. We investigated soil chemistry, aboveground plant productivity, and soil microbial communities across a grassland chronosequence (65,000-226,000 yrs old) located near Santa Cruz, CA. Aboveground net primary productivity (ANPP) initially increased to a maximum and then decreased for the older soils. We used polar lipid fatty acids (PLFA) to investigate microbial communities including both surface (<0.1 m) and subsurface (≥0.2 m) soil horizons. PLFAs characteristic of Gram-positive bacteria and actinobacteria increased as a fraction of the microbial community with depth while the fungal fraction decreased relative to the surface. Differences among microbial communities from each chronosequence soil were found primarily in the subsurface where older subsurface soils had smaller microbial community biomass, a higher proportion of fungi, and a different community structure than the younger subsurface soil. Subsurface microbial community shifts in biomass and community structure correlated with, and were likely driven by, decreasing soil P availability and Ca concentrations, respectively. Trends in soil chemistry as a function of soil age led to the separation of the biological (≤1 m depth) and geochemical (>1 m) cycles in the old, slowly eroding landscape we investigated, indicating that this separation, commonly observed in tropical and subtropical ecosystems, can also occur in temperate climates. This study is the first to investigate subsurface microbial communities in a long-term chronosequence. Our results highlight connections between soil chemistry and both the aboveground and belowground parts of an ecosystem.  相似文献

Overgrazing is one of the main causes of desertification in the semiarid Horqin sandy grassland of northern China. Excluding grazing livestock is considered as an alternative to restore vegetation in degraded sandy grassland in this region. However, few data are available concerning the impacts of continuous grazing and livestock exclusion on soil properties. In this paper, characteristics of vegetation and soil properties under continuous grazing and exclusion of livestock for 5 and 10 years were examined in representative degraded sandy grassland. Continuous grazing resulted in a considerable decrease in ground cover, which accelerates soil erosion by wind, leading to a further coarseness in surface soil, loss of soil organic C and N, and a decrease in soil biological properties. The grassland under continuous grazing is in the stage of very strong degradation. Excluding livestock grazing enhances vegetation recovery, litter accumulation, and development of annual and perennial grasses. Soil organic C and total N concentrations, soil biological properties including some enzyme activities and basal soil respiration improved following 10-year exclusion of livestock, suggesting that degradation of the grassland is being reversed. The results suggest that excluding grazing livestock on the desertified sandy grassland in the erosion-prone Horqin region has a great potential to restore soil fertility, sequester soil organic carbon and improve biological activity. Soil restoration is a slow process although the vegetation can recover rapidly after removal of livestock. A viable option for sandy grassland management should be to adopt proper exclosure in a rotation grazing system in the initial stage of grassland degradation.  相似文献

Phospholipid fatty acid (PLFA) and total soil fatty acid methyl esters (TSFAME), both lipid-based approaches used to characterize microbial communities, were compared with respect to their reliable detection limits, extraction precision, and ability to differentiate agricultural soils. Two sets of soil samples, representing seven crop types from California's Central Valley, were extracted using PLFA and TSFAME procedures. PLFA analysis required 10 times more soil than TSFAME analysis to obtain a reliable microbial community fingerprint and total fatty acid content measurement. Although less soil initially was extracted with TSFAME, total fatty acid (FA) content g−1 soil (DW) was more than 7-fold higher in TSFAME- versus PLFA-extracted samples. Sample extraction precision was much lower with TSFAME analysis than PLFA analysis, with the coefficient of variation between replicates being as much as 4-fold higher with TSFAME extraction. There were significant differences between PLFA- and TSFAME-extracted samples when biomarker pool sizes (mol% values) for bacteria, actinomycetes, and fungi were compared. Correspondence analysis (CA) of PLFA and TSFAME samples indicated that extraction method had the greatest influence on sample FA composition. Soil type also influenced FA composition, with samples grouping by soil type with both extraction methods. However, separate CAs of PLFA- and TSFAME extracted samples depicted strong differences in underlying sample groupings. Recommendations for the selection of extraction method are presented and discussed.  相似文献

The influence of soil pH on the phospholipid fatty acid (PLFA) composition of the microbial community was investigated along the Hoosfield acid strip, Rothamsted Research, UK - a uniform pH gradient between pH 8.3 and 4.5. The influence of soil pH on the total concentration of PLFAs was not significant, while biomass estimated using substrate induced respiration decreased by about 25%. However, the PLFA composition clearly changed along the soil pH gradient. About 40% of the variation in PLFA composition along the gradient was explained by a first principal component, and the sample scores were highly correlated to pH (R2 = 0.97). Many PLFAs responded to pH similarly in the Hoosfield arable soil compared with previous assessments in forest soils, including, e.g. monounsaturated PLFAs 16:1ω5, 16:1ω7c and 18:1ω7, which increased in relative concentrations with pH, and i16:0 and cy19:0, both of which decreased with pH. Some PLFAs responded differently to pH between the soil types, e.g. br18:0. We conclude that soil pH has a profound influence on the microbial PLFA composition, which must be considered in all applications of this method to detect changes in the microbial community.  相似文献

Soil incubations are often used to investigate soil organic matter (SOM) decomposition and its response to increased temperature, but changes in the activity and community composition of the decomposers have rarely been included. As part of an integrated investigation into the responses of SOM components in laboratory incubations at elevated temperatures, fungal and bacterial phospholipid fatty acids (PLFAs) were measured in two grassland soils contrasting in SOM quality (i.e. SOM composition), and changes in the microbial biomass and community composition were monitored. Whilst easily-degradable SOM and necromass released from soil preparation may have fuelled microbial activity at the start of the incubation, the overall activity and biomass of soil microorganisms were relatively constant during the subsequent one-year soil incubation, as indicated by the abundance of soil PLFAs, microbial respiration rate (r), and metabolic quotient (qCO2). PLFAs relating to fungi and Gram-negative bacteria declined relative to Gram-positive bacteria in soils incubated at higher temperatures, presumably due to their vulnerability to disturbance and substrate constraints induced by faster exhaustion of available nutrient sources at higher temperatures. A linear correlation was found between incubation temperatures and the microbial stress ratios of cyclopropane PLFA-to-monoenoic precursor (cy17:0/16:1ω7c and cy19:0/18:1ω7c) and monoenoic-to-saturated PLFAs (mono/sat), as a combined effect of temperature and temperature-induced substrate constraints. The microbial PLFA decay patterns and ratios suggest that SOM quality intimately controls microbial responses to global warming.  相似文献

Soil microbial communities mediate the decomposition of soil organic matter (SOM). The amount of carbon (C) that is respired leaves the soil as CO2 (soil respiration) and causes one of the greatest fluxes in the global carbon cycle. How soil microbial communities will respond to global warming, however, is not well understood. To elucidate the effect of warming on the microbial community we analyzed soil from the soil warming experiment Achenkirch, Austria. Soil of a mature spruce forest was warmed by 4 °C during snow-free seasons since 2004. Repeated soil sampling from control and warmed plots took place from 2008 until 2010. We monitored microbial biomass C and nitrogen (N). Microbial community composition was assessed by phospholipid fatty acid analysis (PLFA) and by quantitative real time polymerase chain reaction (qPCR) of ribosomal RNA genes. Microbial metabolic activity was estimated by soil respiration to biomass ratios and RNA to DNA ratios. Soil warming did not affect microbial biomass, nor did warming affect the abundances of most microbial groups. Warming significantly enhanced microbial metabolic activity in terms of soil respiration per amount of microbial biomass C. Microbial stress biomarkers were elevated in warmed plots. In summary, the 4 °C increase in soil temperature during the snow-free season had no influence on microbial community composition and biomass but strongly increased microbial metabolic activity and hence reduced carbon use efficiency.  相似文献

Liming is necessary for good nutrient availability and crop growth. Lime use in Ireland is now the lowest in half a century. A recent study shows that grassland mineral soils in Ireland has a mean pH of 5.4 and mean lime requirement (LR) of 9.3 t/ha ground limestone. There have been a number of studies in the USA to re-evaluate LR, but little activity in the European Union (EU) in recent years. The primary aim of our research was to compare five methods for estimating LR, which included the Shoemaker–McLean–Pratt (SMP) buffer method currently used in Ireland (IRL), the Sikora buffer method used at the University of Kentucky (UKY), Ca(OH)2 titration used at University of Georgia (UGA), the modified Mehlich buffer method used at Penn State University (PSU) and the UK RothLime model, using 57 representative grassland mineral soils from Ireland with a pH range from 4.8 to 6.6. The secondary aim was to explore an alternative to the SMP buffer that does not involve the use of toxic chemicals. The results show good agreement between the pH measured by the Irish and three US laboratories and reasonably good agreement in LR estimated by five methods. The main conclusions are: (1) a significant proportion of grassland on mineral soils in Ireland would benefit from liming to increase soil pH, (2) on average, LRs as recommended in Ireland are higher than those advised elsewhere , ( 3) the target pH in Ireland is high compared with that in other countries and should be reduced from pH 6.5 to 6.2, (4) the SMP buffer method should be replaced by a suitable alternative and, in principle, any of the four methods studied would be suitable, (5) to find the most suitable alternative for accurate LR advice it would be necessary to compare the different methods to the actual LR from incubation of representative soils with calcium hydroxide.  相似文献

The presence of plants induces strong accelerations in soil organic matter (SOM) mineralization by stimulating soil microbial activity – a phenomenon known as the rhizosphere priming effect (RPE). The RPE could be induced by several mechanisms including root exudates, arbuscular mycorrhizal fungi (AMF) and root litter. However the contribution of each of these to rhizosphere priming is unknown due to the complexity involved in studying rhizospheric processes. In order to determine the role of each of these mechanisms, we incubated soils enclosed in nylon meshes that were permeable to exudates, or exudates & AMF or exudates, AMF and roots under three grassland plant species grown on sand. Plants were continuously labeled with 13C depleted CO2 that allowed distinguishing plant-derived CO2 from soil-derived CO2. We show that root exudation was the main way by which plants induced RPE (58–96% of total RPE) followed by root litter. AMF did not contribute to rhizosphere priming under the two species that were significantly colonized by them i.e. Poa trivialis and Trifolium repens. Root exudates and root litter differed with respect to their mechanism of inducing RPE. Exudates induced RPE without increasing microbial biomass whereas root litter increased microbial biomass and raised the RPE mediating saprophytic fungi. The RPE efficiency (RPE/unit plant-C assimilated into microbes) was 3–7 times higher for exudates than for root litter. This efficiency of exudates is explained by a microbial allocation of fresh carbon to mineralization activity rather than to growth. These results suggest that root exudation is the main way by which plants stimulated mineralization of soil organic matter. Moreover, the plants through their exudates not only provide energy to soil microorganisms but also seem to control the way the energy is used in order to maximize soil organic matter mineralization and drive their own nutrient supply.  相似文献

The dynamics of fungal and bacterial residues to a one-season tillage event in combination with manure application in a grassland soil are unknown. The objectives of this study were (1) to assess the effects of one-season tillage event in two field trials on the stocks of microbial biomass, fungal biomass, microbial residues, soil organic C (SOC) and total N in comparison with permanent grassland; (2) to determine the effects of repeated manure application to restore negative tillage effects on soil microbial biomass and residues. One trial was started 2 years before sampling and the other 5 years before sampling. Mouldboard ploughing decreased the stocks of SOC, total N, microbial biomass C, and microbial residues (muramic acid and glucosamine), but increased those of the fungal biomarker ergosterol in both trials. Slurry application increased stocks of SOC and total N only in the short-term, whereas the stocks of microbial biomass C, ergosterol and microbial residues were generally increased in both trials, especially in combination with tillage. The ergosterol to microbial biomass C ratio was increased by tillage, and decreased by slurry application in both trials. The fungal C to bacterial C ratio was generally decreased by these two treatments. The metabolic quotient qCO2 showed a significant negative linear relationship with the microbial biomass C to SOC ratio and a significant positive relationship with the soil C/N ratio. The ergosterol to microbial biomass C ratio revealed a significant positive linear relationship with the fungal C to bacterial C ratio, but a negative one with the SOC content. Our results suggest that slurry application in grassland soil may promote SOC storage without increasing the role of saprotrophic fungi in soil organic matter dynamics relative to that of bacteria.  相似文献

Soils from a long term experiment, established in 1972, incorporating replicated treatments of burning every 2 and 4 years with control plots were sampled in 2005 to determine the changes in microbial community structure, measured using phospholipid fatty acids (PLFAs) and functional diversity measured using multiple substrate induced respiration (SIR) tests (MicroResp™). Microbial biomass (total PLFA) in the 2 year burn treatments was 50% less than both the control and 4-year burn treatments. There was also concomitantly less respiratory activity which mirrored the known changes in soil C and substrate quality. Contrary to other studies soil bacterial PLFAs were reduced as much as fungal PLFAs in the 2-year burn and the short term (6 h) SIR of arginine, lysine, galactose and trehalose were significantly inhibited in the 2-year burn soils. The data suggest that a 4-year burn is a more sustainable practice for maintaining the original structure and function of the forest belowground ecosystem.  相似文献

Saline soils around Lake Urmia in northern Iran constitute a stressed environment for plants and microbial communities, including arbuscular mycorrhizal (AM) fungi. Soil and root samples were collected from fields cultivated with the glycophytes Allium cepa L. and Medicago sativa L., and sites dominated by the halophyte Salicornia europaea L. Soil and root samples were analyzed for the AM fungal signature neutral lipid fatty acid (NLFA) 16:1ω5. The roots were also examined microscopically for mycorrhizal colonization. Each plant species was sampled across a salt gradient. Microscopic examination showed no AM fungal structures in the roots of S. europaea. The highest root colonization was recorded for M. sativa. The highest NLFA 16:1ω5 values were found in soil around M. sativa roots and the lowest in soil around S. europaea roots. We found evidence for stimulation of vesicle formation at moderate salinity levels in M. sativa, which is an indication of increased carbon allocation to mycorrhiza. On the other hand, we found a negative correlation between salinity and arbuscule formation in A. cepa, which may indicate a less functional symbiosis in saline soils.  相似文献

Microbial communities are important components of terrestrial ecosystems. The importance of their diversity and functions for natural systems is well recognized. However, a better understanding of successional changes of microbial communities over long time scales is still required. In this work, the size and composition of microbial communities in soils of a deglaciation chronosequence at the Damma glacier forefield were studied by fatty acid profiling. Soil fatty acid concentrations clearly increased with soil age. The abundances of arbuscular mycorrhizal fungi (AMF), bacteria and other soil fungi, however, were more affected by abiotic soil parameters like carbon content and pH than by soil age. Analysis of ratios of the different microbial groups (AMF, fungi, bacteria) along the soil chronosequence indicated that: i) the ratios of AMF to bacteria and AMF to fungi decreased with soil age; and ii) the ratio of fungi to bacteria remained unchanged along the soil chronosequence. These two pieces of evidence suggest that the evolution of this ecosystem proceeds at an uneven pace over time and that the role of AMF is less important in older, more organic and acidified soils than in mineral soils. In contrast to other studies, no successional replacement of bacteria with fungi in more acidified and organic soil was observed.  相似文献

Overwintering cattle outdoors causes soil surface disturbance, substantial increases of soil Ntot, Corg, and P and a shift in pH to alkaline levels. Since fungi predominate in unfertilized soils with acidic pH and have filamentous hyphae, we hypothesized that changes caused by overwintering cattle outdoors (trampling, excreta returns, and changes in soil chemistry) will lead to suppressed species richness, lower biomass, and alter the structure of fungal communities. The research was conducted on an upland pasture used more than 10 years for cattle overwintering. Both culture-dependent and -independent methods were used for the determination of either fungal species composition (cultivation; DGGE) or biomass (numbers of CFU; concentration of fungal PLFA marker 18:2ω6,9). Soils under three different levels of cattle disturbance (S - severe, M - moderate, C - no disturbance/control) were investigated during three subsequent years. In addition, the DGGE analysis of soils was completed by comparison with analysis of fresh cattle excrements (Ex). The composition of fungal communities showed significantly higher richness and a substantial shift in species composition in cattle-disturbed soils (S, M) in comparison to the non-disturbed soil (C). The number of separated DGGE bands was significantly higher in S (30.67 ± 1.63; mean ± SD) and M (25.50 ± 1.64) soils than in the C soil (19.33 ± 1.75). Sequencing of typical bands revealed common fungal genera - Alternaria, Penicillium, Fusarium, Rhizopus, Isaria, and Metarhizium. Profiles of the S soil were enriched by bands of rumen-born anaerobic fungi (Neocallimastix, Cyllamyces) occurring mainly in profiles of excrements, where relatively low band richness (14.33 ± 1.15) was observed. The increasing level of cattle disturbance induced an increase in the biomass of complex fungal community over the three-year experimental period from 3.39 ± 2.11 (mean ± SD) nmol of fungal PLFA per gram of the C soil to 5.87 ± 3.16 in the M soil and 9.21 ± 4.69 in the S soil. Concentrations of soil Ntot and Corg were evaluated as the parameters significantly correlating with biomass as well as composition of the fungal community.  相似文献

Plants often impact the rate of native soil organic matter turnover through root interactions with soil organisms; however the role of root-microbial interactions in mediation of the “priming effect” is not well understood. We examined the effects of living plant roots and N fertilization on belowground C dynamics in a California annual grassland soil (Haploxeralf) during a two-year greenhouse study. The fate of 13C-labeled belowground C (roots and organic matter) was followed under planted (Avena barbata) and unplanted conditions, and with and without supplemental N (20 kg N ha−1 season−1) over two periods of plant growth, each followed by a dry, fallow period of 120 d. Turnover of belowground 13C SOM was followed using 13C-phospholipid fatty acid (PLFA) biomarkers. Living roots increased the turnover and loss of belowground 13C compared with unplanted soils. Planted soils had 20% less belowground 13C present than in unplanted soils after 2 cycles of planting and fallow. After 2 treatment cycles, unlabeled soil C was 4.8% higher in planted soils than unplanted. The addition of N to soils decreased the turnover of enriched belowground 13C during the first treatment season in both planted and unplanted soils, however no effect of N was observed thereafter. Our findings suggest that A. barbata may increase soil C levels over time because root and exudate C inputs are significant, but that increase will be moderated by an overall faster C mineralization rate of belowground C. N addition may slow soil C losses; however, the effect was minor and transient in this system. The labeled root-derived 13C was initially recovered in gram negative (highest enrichment), gram positive, and fungal biomarkers. With successive growing seasons, the labeled C in the gram negative and fungal markers declined, while gram positive markers continued to accumulate labeled belowground C. The rhizosphere of A. barbata shifted the microbial community composition, resulting in greater abundances of gram negative markers and lower abundances of gram positive, actinobacteria and cyclopropyl PLFA markers compared to unplanted soil. However, the longer-term utilization of labeled belowground C by gram positive bacteria was enhanced in the rhizosphere microbial community compared with unplanted soils. We suggest that the activities of gram positive bacteria may be major controllers of multi-year rhizosphere-related priming of SOM decomposition.  相似文献

In grasslands, saprotrophic fungi, including basidiomycetes, are major decomposers of dead organic matter, although spatial distributions of their mycelial assemblages are little described. The aim of this study was to characterise the scale and distribution of saprotrophic fungal communities in a coastal grassland soil using terminal restriction fragment length polymorphism (T-RFLP).Soil fungi were sampled at Point Reyes, California, USA, by taking forty-five 26 mm diam. cores in a spatially defined manner. Within each sampled core, complete core sections at 1-2 cm and 14-15 cm depths were removed and sub-sampled for DNA extraction and amplification using the primer pairs ITS1F-FAM/ITS4 (general fungi) or ITS1F-FAM/ITS4B (basidiomycete-specific).Nonmetric Multidimensional Scaling showed that general fungal communities could be clearly separated by depth, although basidiomycete communities could not. There were no strong patterns of community similarity or dissimilarity for general or basidiomycete fungal communities at horizontal geographical distances from 25 cm to 96 m in the upper horizon. These results show considerable vertical, but little horizontal, variability in fungal community structure in a semi-natural grassland at the spatial scales measured here.  相似文献
