Together with plants, soil microbial communities play an essential role in the development of stable ecosystems on degraded lands, such as postmining spoil heaps. Our study addressed concurrent development of the vegetation and soil fungal and bacterial communities in the course of primary succession in a brown coal mine spoil deposit area in the Czech Republic across a chronosequence spanning 54 years. During succession, the plant communities changed from sparse plants over grassland and shrubland into a forest, becoming substantially more diverse with time. Microbial biomass increased until the 21st year of ecosystem development and later decreased. Although there was a close association between fungi and vegetation, with fungi mirroring the differences in plant community assemblages, the development of the bacterial community was different. The early succession community in the barren nonvegetated soil largely differed from that in the older sites, especially in its high abundance of autotrophic and free‐living N2‐fixing bacteria. Later in succession, bacterial community changes were minor and reflected the chemical parameters of the soil, including pH, which also showed a minor change with time. Our results show that complex forest ecosystems developed over 54 years on the originally barren soil of the temperate zone and indicate an important role of bacteria in the initial stage of soil development. Although the arrival of vegetation affects substantially fungal as well as bacterial communities, it is mainly fungi that respond to the ongoing development of vegetation.  相似文献

In Eastern Spain, almond trees have been cultivated in terraced orchards for centuries, forming an integral part of the Mediterranean forest scene. In the last decades, orchards have been abandoned due to changes in society. This study investigates effects of changes in land use from forest to agricultural land and the posterior land abandonment on soil microbial community, and the influence of soil physico-chemical properties on the microbial community composition (assessed as abundances of phospholipids fatty acids, PLFA). For this purpose, three land uses (forest, agricultural and abandoned agricultural) at four locations in SE Spain were selected. Multivariate analysis showed a substantial level of differentiation in microbial community structure according to land use. The microbial communities of forest soils were highly associated with soil organic matter content. However, we have not found any physical or chemical soil property capable of explaining the differences between agricultural and abandoned agricultural soils. Thus, it was suggested that the cessation of the perturbation caused by agriculture and shifts in vegetation may have led to changes in the microbial community structure. PLFAs indicative of fungi and ratio of fungal to bacterial PLFAs were higher in abandoned agricultural soils, whereas the relative abundance of bacteria was higher in agricultural soils. Actinomycetes were generally lower in abandoned agricultural soils, while the proportions of vesicular–arbuscular mycorrhyzal fungi were, as a general trend, higher in agricultural and abandoned agricultural soils than in forests. Soils support highly diverse communities, but due to the great spatial complexity at very fine scales and small size of soil organisms, our knowledge of what structures belowground communities remains limited. We used a spatially nested sampling regime (distances between samples of 0.06–122 m) to develop an understanding of the relationship between soil mite and microbial community composition and properties of vegetation and underlying soil within two habitats: birch woodland and heather moorland. We found that variation in community composition of all four biotic groups investigated (Oribatida, Mesostigmata, fungi and bacteria – thus representing multiple trophic levels) was related to variation in soil properties and vegetation, but also space. These three factors (i.e. vegetation, soil properties and space) accounted for 17–36% of the variation in community composition of the biotic groups. The remaining, rather large, proportion of unexplained variation in community composition (64–87%) is likely to represent either random variation or variation related to unmeasured variables that are not spatially auto-correlated. Soil properties generally explained more variance in belowground communities than could vegetation composition. This should not be interpreted as evidence that soil properties exert a greater influence on belowground communities than vegetation composition per se, as the vegetation composition within a habitat is likely to vary at a larger spatial scale than the soil properties we measured. Vegetation composition had stronger explanatory power in the birch woodland and was more strongly related to fungal community composition than that of any other biotic group, particularly in heather moorland. This relationship may reflect the strong association between plants and their fungal symbionts and pathogens. We conclude that both vegetation composition and variation in soil properties influence belowground communities. However, the relative importance of these two factors depends on both habitat and the type of organism being studied.  相似文献

Soil micro-organisms play a vital role in grassland ecosystem functioning but little is known about the effects of grassland management on spatial patterns of soil microbial communities. We compared plant species composition with terminal restriction fragment length polymorphism (T-RFLP) fingerprints of soil bacterial and fungal communities in unimproved, restored and improved wet grasslands. We assessed community composition of soil micro-organisms at distances ranging from 0.01 m to 100 m and determined taxa–area relationships from field- to landscape level. We show that land management type influenced bacterial but not fungal community composition. However, extensive grassland management to restore aboveground diversity affected spatial patterns of soil fungi. We found distinct distance–decay and small-scale aggregation of fungal populations in extensively managed grasslands restored from former arable use. There were no clear spatial patterns in bacterial communities at the field-scale. However, at the landscape level there was a moderate increase in bacterial taxa and a strong increase in fungal taxa with the number of sites sampled. Our results suggest that grassland management affects soil microbial communities at multiple scales; the observed small-scale variation may facilitate plant species coexistence and should be taken into account in field studies of soil microbial communities.  相似文献

选取土壤侵蚀较严重的黑龙江省宾县宾州河流域为研究区,通过采集流域上、中和下游6个典型坡面的坡上、坡中和坡下部土壤样品,分析了坡耕地土壤微生物群落数量在流域和坡面尺度上的分布规律,并比较了土壤微生物群落数量空间分布与土壤侵蚀空间分布的关系。结果表明,在流域尺度上,土壤微生物总数量和细菌数量均表现为:下游>中游>上游土壤真菌数量表现为:上游>下游>中游土壤放线菌数量表现为中游最大,而上游和下游数量相当。在坡面尺度上,土壤微生物总数量和细菌数量均呈现:坡中部<坡上部<坡下部,土壤真菌数量呈现:坡下部<坡中部<坡上部,土壤放线菌数量呈现:坡中部<坡下部<坡上部。研究表明,流域土壤微生物总数量和土壤细菌数量的空间分布皆与侵蚀-沉积速率的空间分布相对应,反映出侵蚀-沉积速率是影响该流域土壤微生物群落数量的主要因素。  相似文献

土壤微生物是反映土壤环境质量的重要指标,为明确赤水河流域典型植被类型土壤微生物群落特征及优势菌属,为生态系统的恢复与管理提供理论依据,采用高通量测序技术研究了赤水河流域的5种典型植被类型(灌丛、针阔混交林、常绿阔叶林、杉木林和竹林)土壤的微生物群落结构及多样性,并探讨了其主要影响因子。结果表明:(1)不同植被类型的细菌和真菌丰富度Chao1指数差异均不显著,说明二者所观测到的物种总数没有差异。Shannon指数显示各植被类型微生物多样性存在一定差异,细菌多样性以竹林最低,显著性低于灌丛和针阔混交林(p<0.05); 真菌多样性以灌丛和杉木林显著性高于其他3种植被类型(p<0.05)。(2)5种植被类型土壤细菌优势门(相对丰度>10%)主要有变形菌门(Proteobacteria)、放线菌门(Actinobacteriota)和酸杆菌门(Acidobacteriota)。其中变形菌门(Proteobacteria)在灌丛、针阔混交林和常绿阔叶林中占绝对优势(相对丰度为29.70%~33.62%),而放线菌门(Actinobacteriota)则在杉木林和竹林中最为丰富,相对丰度占比分别为32.88%,29.88%。各植被类型土壤真菌门以子囊菌门(Ascomycota)为绝对优势菌群(相对丰度>49%)。(3)5种植被类型土壤细菌和真菌中优势菌属差异较大。在细菌属水平上,针阔混交林和竹林分别以未定名的Vicinamibacterales和芽孢杆菌属(Bacillus)为优势,而其他3种植被类型则以节杆菌属(Arthrobacter)最丰富。在真菌属水平上,灌丛和常绿阔叶林中优势关键属为子囊菌门(Ascomycota)未分类真菌属(unclassified_p_Ascomycota),杉木林以被孢霉属(Mortierella)最丰富,而针阔混交林和竹林中均以沙蜥属(Saitozyma)相对丰度最高。(4)NMDS分析表明,土壤细菌与真菌群落空间分布差异显著。(5)冗余分析表明,土壤含水量、pH值和TN(总氮)对土壤细菌群落结构具有显著性的影响,而pH值、容重、TOC(总有机碳)、TN和TP(总磷)对真菌群落结构影响显著。综合分析可知,针阔混交林土壤细菌群落较丰富,杉木林土壤真菌生长较旺盛,而灌丛土壤细菌和真菌多样性均较高,应采取有效措施提高主要林分土壤养分,从而激发微生物的生长,改善土壤环境。  相似文献

The aim of this study was to evaluate the long-term influence of contrasting rural land use types on the level, plot-scale variation and horizontal spatial structure of decomposition activities and the bacterial community in soil. Experimental data were collected in the southern boreal zone from topsoil layers of adjacent spruce forest, unmanaged meadow (former field) and organically cultivated field that all shared the same soil origin. The forest soil was sampled separately for the organic and mineral layers. A geostatistical design comprising 50 sampling points per plot area of 10 × 10 m2 was used. The measured microbiological characteristics included eight different hydrolytic soil enzyme activities involved in C, P and S cycles, bacterial 16S rDNA length heterogeneity profiles (LH-PCR) and total DNA yield as a relative estimate of microbial biomass.Effects of land use were pronounced on both the bacterial community structure and soil enzyme activities. Soil organic matter (SOM) content predicted well the major differences in soil enzyme activities and microbial biomass. Highest enzyme activities were generally found in the forest organic soil whereas the underlying mineral soil showed significantly lower activities with a pattern similar to those of the other mineral soils, especially the cultivated field. Bacterial LH-PCR fingerprints were distinct but at the same time remarkably similar between field and meadow soils whereas the forest organic layer differed clearly from the mineral soils. Within-plot variation of soil microbiological characteristics was best explained by the variation of SOM. Relative standard deviations of soil microbiological characteristics typically decreased in the order: forest organic layer ≈ forest mineral layer > meadow > field. However, bacterial fingerprints showed lowest variation within the meadow. Most of the microbiological variables studied showed no or only weak spatial structure at the scale sampled.  相似文献

Microbial communities vary across the landscape in forest soils, but prediction of their biomass and composition is a difficult challenge due to the large numbers of variables that influence their community structures. Here we examine the use of artificial neural network (ANN) models for extraction of patterns among soil chemical variables and microbial community structures in forest soils from three regions of the Atlantic Forest of Brazil. At each location, variations in soil chemical properties and FAME profiles of microbial community structures were mapped at 20 × 20 m intervals within 10 ha parcels. Geostatistical analyses showed that spatial variability in soil physical and chemical variables could be mapped at scale distances of 20 m, but that FAME profiles representing the microbial communities were highly variable and had no spatial dependence at the same scale in most cases. RDA analysis showed that FAME signatures representing different microbial groups were positively associated with soil pH, OM, P and base cations concentrations, whereas microbial biomass was negatively associated with the same environmental factors. In contrast, ANN models revealed clear relationships between microbial community structures at each parcel location, and generated verifiable predictions of variations in FAME profiles in relation to soil pH, texture, and the relative abundances of base cations. The results suggest that ANN modeling provides a useful approach for describing the relationships between microbial community structures and soil properties in tropical forest soils that were not able to be captured using geostatistical and RDA analyses.  相似文献

Soil fungi are highly diverse and act as the primary agents of nutrient cycling in forests. These fungal communities are often dominated by mycorrhizal fungi that form mutually beneficial relationships with plant roots and some mycorrhizal fungi produce extracellular and cell-bound enzymes that catalyze the hydrolysis of nitrogen (N)- and phosphorus (P)- containing compounds in soil organic matter. Here we investigated whether the community structure of different types of mycorrhizal fungi (arbuscular and ectomycorrhizal fungi) is correlated with soil chemistry and enzyme activity in a northern hardwood forest and whether these correlations change over the growing season. We quantified these relationships in an experimental paired plot study where white-tailed deer (access or excluded 4.5 yrs) treatment was crossed with garlic mustard (presence or removal 1 yr). We collected soil samples early and late in the growing season and analyzed them for soil chemistry, extracellular enzyme activity and molecular analysis of both arbuscular mycorrhizal (AM) and ectomycorrhizal/saprotrophic fungal communities using terminal restriction fragment length polymorphism (TRFLP). AM fungal communities did not change seasonally but were positively correlated with the activities of urease and leucine aminopeptidase (LAP), enzymes involved in N cycling. The density of garlic mustard was correlated with the presence of specific AM fungal species, while deer exclusion or access had no effect on either fungal community after 4.5 yrs. Ectomycorrhizal/saprotrophic fungal communities changed seasonally and were positively correlated with most soil enzymes, including enzymes involved in carbon (C), N and P cycling, but only during late summer sampling. Our results suggest that fine scale temporal and spatial changes in soil fungal communities may affect soil nutrient and carbon cycling. Although AM fungi are not generally considered capable of producing extracellular enzymes, the correlation between some AM taxa and the activity of N acquisition enzymes suggests that these fungi may play a role in forest understory N cycling.  相似文献

This study identified microbial functional groups like total culturable bacteria, potential N2-fixing free living bacteria, N2-fixing hydrocarbonoclastic bacteria, N-assimilating hydrocarbonoclastic bacteria, total fungi, actinobacteria, P-solubilizers, lipolytic microorganisms, and starch, cellulose, pectin and protein degrading microorganisms, isolated from the rhizosphere of four species of mangroves (Red, Black, White, and Button) from the natural protected area at the Terminos Lagoon, Campeche, México. Overall, microbial populations showed significant differences (P < 0.05) among the four mangrove species. The rhizosphere of White mangrove showed better chemical and textural soil properties, and harbored the highest microbial populations when compared to the remaining mangrove species. The principal component analysis indicated that two components accounted the 85.3% of the total variation. The most significant textural and chemical soil properties were the major components, CP1 (organic matter and total organic carbon) and CP2 (sand and clay). Microbial populations correlated (P < 0.05, Pearson coefficient) with sand and clay particles, and with some soil chemical properties such as organic matter. The total nitrogen and organic carbon significantly correlated with cellulose degraders, while phosphorus with N2-fixing bacteria, total fungi, and with pectin and starch degraders.  相似文献

Phospholipid fatty acid (PLFA) patterns were used to describe the composition of the soil microbial communities under 12 natural forest stands including oak and beech, spruce-fir-beech, floodplain and pine forests. In addition to the quantification of total PLFAs, soil microbial biomass was measured by substrate-induced respiration and chloroform fumigation-extraction. The forest stands possess natural vegetation, representing an expression of the natural site factors, and we hypothesised that each forest type would support a specific soil microbial community. Principal component analysis (PCA) of PLFA patterns revealed that the microbial communities were compositionally distinct in the floodplain and pine forests, comprising azonal forest types, and were more similar in the oak, beech and spruce-fir-beech forests, which represent the zonal vegetation types of the region. In the nutrient-rich floodplain forests, the fatty acids 16:1ω5, 17:0cy, a15:0 and a17:0 were the most prevalent and soil pH seemed to be responsible for the discrimination of the soil microbial communities against those of the zonal forest types. The pine forest soils were set apart from the other forest soils by a higher abundance of PLFA 18:2ω6,9, which is typical of fungi and may also indicate ectomycorrhizal fungi associated with pine trees, and high amounts of PLFA 10Me18:0, which is common in actinomycetes. These findings suggest that the occurrence of azonal forest types at sites with specific soil conditions is accompanied by the development of specific soil microbial communities. The study provides information on the microbial communities in undisturbed forest soils which may facilitate interpretation of data derived from managed or even damaged or degraded forests.  相似文献

Purpose

There have been a number of studies on the succession of vegetation; however, the succession of soil microbes and the collaborative relationships between microbes and vegetation during land restoration remain poorly understood. The objectives of this study were to characterize soil microbial succession and to explore the collaborative mechanisms between microbes and vegetation during the restoration of abandoned land through quantitative ecology methods.

Materials and methods

The present research was carried out in the succession of a 5-year abandoned land and its conversion to Hippophae rhamnoides shrubs, Larix principis-rupprechtii plantation, and a naturally regenerated forest (mixed forest). Soil bacterial, archaeal and fungal characteristics were tested by real-time quantitative PCR assays and terminal restriction fragment length polymorphism. The richness, diversity, and evenness indices were employed to analyze plant and microbial communities’ structure. The stability of plant and microbial communities was tested using Spearman’s rank correlation. The relationships between the regeneration scenarios and environmental factors were determined through canonical correspondence analysis.

Results and discussion

The aboveground biomass was significantly different among the sites. Soil bacterial, archaeal, and fungal rRNA gene abundances did not increase significantly with increasing soil organic carbon content. There were higher correlation coefficients between plant and total microbial communities on the richness, diversity, and evenness indices and ratios of positive to negative association compared to ones between plant and individual bacteria, archaea, and fungi. Soil bulk density, clay, pH, and litter were the primary significant environmental factors affecting the structure of plant and microbial communities. The positive relationships between plant and soil bacteria, fungi, and total microbe communities, as well as the negative relationships between plant and archaea, were demonstrated.

Conclusions

The results suggested that plants promote the growth of soil bacteria and fungi during the process of community succession on a small scale; however, plants inhibit the growth of soil archaea.  相似文献

为了探讨植被群落对喀斯特土壤养分和生物化学性质的影响,对贵州茂兰国家级自然保护区不同植被群落下土壤养分、基础呼吸、微生物量碳及酶活性进行比较研究。结果表明:不同植被群落之间土壤养分含量差异显著,土壤总有机碳、全氮、全磷、碱解氮和有效磷表现为裸露地<草丛<灌丛≤乔林;土壤基础呼吸表现为裸露地<灌丛<乔林<草丛,诱导微生物量碳为裸露地<灌丛<草丛=乔林,而熏蒸微生物量碳为裸露地<草丛<灌丛<乔林;不同植被群落下土壤蔗糖酶活性高于裸露地,但不同植被群落间差异不显著,土壤脲酶活性有着裸露地<草丛=灌丛<乔林的趋势,而土壤碱性磷酸酶活性表现为裸露地<灌丛<乔林<草丛;土壤呼吸商和诱导微生物量与熏蒸微生物量比值均表现为乔林=灌丛<草丛=裸露地。结果分析表明,不同植被群落下喀斯特土壤养分水平及其凋落物差异影响着土壤微生物群落结构及活性,而土壤微生物执行着土壤营养元素生物化学过程,其群落结构和活性又影响着土壤养分水平及养分有效性。  相似文献

Changes in the biomass and structure of soil microbial communities have the potential to impact ecosystems via interactions with plants and weathering minerals. Previous studies of forested long-term (1000s - 100,000s of years) chronosequences suggest that surface microbial communities change with soil age. However, significant gaps remain in our understanding of long-term soil microbial community dynamics, especially for non-forested ecosystems and in subsurface soil horizons. We investigated soil chemistry, aboveground plant productivity, and soil microbial communities across a grassland chronosequence (65,000-226,000 yrs old) located near Santa Cruz, CA. Aboveground net primary productivity (ANPP) initially increased to a maximum and then decreased for the older soils. We used polar lipid fatty acids (PLFA) to investigate microbial communities including both surface (<0.1 m) and subsurface (≥0.2 m) soil horizons. PLFAs characteristic of Gram-positive bacteria and actinobacteria increased as a fraction of the microbial community with depth while the fungal fraction decreased relative to the surface. Differences among microbial communities from each chronosequence soil were found primarily in the subsurface where older subsurface soils had smaller microbial community biomass, a higher proportion of fungi, and a different community structure than the younger subsurface soil. Subsurface microbial community shifts in biomass and community structure correlated with, and were likely driven by, decreasing soil P availability and Ca concentrations, respectively. Trends in soil chemistry as a function of soil age led to the separation of the biological (≤1 m depth) and geochemical (>1 m) cycles in the old, slowly eroding landscape we investigated, indicating that this separation, commonly observed in tropical and subtropical ecosystems, can also occur in temperate climates. This study is the first to investigate subsurface microbial communities in a long-term chronosequence. Our results highlight connections between soil chemistry and both the aboveground and belowground parts of an ecosystem.  相似文献

Silver nanoparticles hold great promise as effective anti-microbial compounds in a myriad of applications but may also pose a threat to non-target bacteria and fungi in the environment. Because microorganisms are involved in extensive interactions with many other organisms, these partner species are also prone to indirect negative effects from silver nanoparticles.Here, we focus on the effects of nanosilver exposure in the rhizosphere. Specifically, we evaluate the effect of 100 mg kg−1 silver nanoparticles on maize plants, as well as on the bacteria and fungi in the plant's rhizosphere and the surrounding bulk soil. Maize biomass measurements, microbial community fingerprints, an indicator of microbial enzymatic activity, and carbon use diversity profiles are used. Hereby, it is shown that 100 mg kg−1 silver nanoparticles in soil increases maize biomass, and that this effect coincides with significant alterations of the bacterial communities in the rhizosphere. The bacterial community in nanosilver exposed rhizosphere shows less enzymatic activity and significantly altered carbon use and community composition profiles. Fungal communities are less affected by silver nanoparticles, as their composition is only slightly modified by nanosilver exposure. In addition, the microbial changes noted in the rhizosphere were significantly different from those noted in the bulk soil, indicated by different nanosilver-induced alterations of carbon use and community composition profiles in bulk and rhizosphere soil.Overall, microorganisms in the rhizosphere seem to play an important role when evaluating the fate and effects of silver nanoparticle exposure in soil, and not only is the nanosilver response different for bacteria and fungi, but also for bulk and rhizosphere soil. Consequently, assessment of microbial populations should be considered an essential parameter when investigating the impacts of nanoparticle exposure.  相似文献

为了揭示京郊荒滩地植被恢复初期土壤理化性质与微生物群落结构的变化特征,以补播苜蓿(Medicago sativa L.)、补播无芒雀麦(Bromus inermis Leyss)为研究对象,自然撂荒地为对照,采用高通量测序方法研究不同植被恢复模式土壤细菌与真菌的群落结构。结果表明:(1)2种植被恢复方式下土壤全氮含量显著高于对照,补播无芒雀麦土壤全磷含量最高;(2)2种植被恢复方式短期内显著改善了土壤物理性质,土壤容重显著低于对照,补播无芒雀麦土壤田间持水量、毛管持水量与最大持水量最高,补播苜蓿土壤饱和导水率最高;(3)2种植被恢复方式短期内对土壤微生物群落结构产生影响,真菌中的子囊菌门、细菌中的蓝细菌门相对丰度增加,真菌与细菌的生物多样性降低;(4)相关性分析表明,土壤有机质含量与真菌中的子囊菌门、细菌中的变形菌门、蓝细菌门数量存在显著正相关关系。2种植被恢复方式短期内均不同程度的改善了土壤理化性质,增加了固氮及降解有机质的微生物的数量及活性。  相似文献

Although soil microorganisms play a central role in the soil processes that determine nutrient availability and productivity of forest ecosystems, we are only beginning to understand how microbial communities are shaped by environmental factors and how the structure and function of soil microbial communities in turn influence rates of key soil processes. Here we compare the structure and function of soil microbial communities in seven mature, undisturbed forest types across a range of regional climates in British Columbia and Alberta, and examine the variation in community composition within forest types. We collected the forest floor fermentation (F) and humus (H) layers and upper 10 cm of mineral soil at 3 sites in each of seven forest types (corresponding to seven Biogeoclimatic zones) in both spring and summer. Phospholipid fatty acid analysis was used to investigate the structure of soil microbial communities and total soil microbial biomass; potential activities of extra-cellular enzymes indicated the functional potential of the soil microbial community in each layer at each site.Multivariate analysis indicated that both structure and enzyme activities of soil microbial communities differed among the forest types, and significantly separated along the regional climate gradient, despite high local variation. Soil moisture and organic matter contents were most closely related to microbial community characteristics. Forests in the Ponderosa Pine and Mountain Hemlock zones were distinct from other forests and from each other when comparing potential enzyme activities and had the most extreme moisture and temperature values. Forest floors from the hot and dry Ponderosa Pine forests were associated with enzymes characteristic of water-stress and high concentrations of phenols and other recalcitrant compounds. The wet and cold Mountain Hemlock forests were associated with low enzyme activity.An influence of tree species was apparent at the three sites within the Coastal Western Hemlock zone; high bacterial:fungal biomass ratios were found under western redcedar (Thuja plicata) which also had high pH and base-cation levels, and under Douglas-fir (Pseudotsuga menziesii), which had high N availability. The response of soil microbial communities following changes in land-use is governed by multiple factors. The objectives of this study were to investigate (i) whether soil microbial communities track the changes in aboveground vegetation during succession; and (ii) whether microbial communities return to their native state over time. Two successional gradients with different vegetation were studied at the W. K. Kellogg Biological Station, Michigan. The first gradient comprised a conventionally tilled cropland (CT), mid-succession forest (SF) abandoned from cultivation prior to 1951, and native deciduous forest (DF). The second gradient comprised the CT cropland, early-succession grassland (ES) restored in 1989, and long-term mowed grassland (MG). With succession, the total microbial PLFAs and soil microbial biomass C consistently increased in both gradients. While bacterial rRNA gene diversity remained unchanged, the abundance and composition of many bacterial phyla changed significantly. Moreover, microbial communities in the relatively pristine DF and MG soils were very similar despite major differences in soil properties and vegetation. After >50 years of succession, and despite different vegetation, microbial communities in SF were more similar to those in mature DF than in CT. In contrast, even after 17 years of succession, microbial communities in ES were more similar to CT than endpoint MG despite very different vegetation between CT and ES. This result suggested a lasting impact of cultivation history on the soil microbial community. With conversion of deciduous to conifer forest (CF), there was a significant change in multiple soil properties that correlated with changes in microbial biomass, rRNA gene diversity and community composition. In conclusion, history of land-use was a stronger determinant of the composition of microbial communities than vegetation and soil properties. Further, microbial communities in disturbed soils apparently return to their native state with time.  相似文献

Soil microbial communities mediate the decomposition of soil organic matter (SOM). The amount of carbon (C) that is respired leaves the soil as CO2 (soil respiration) and causes one of the greatest fluxes in the global carbon cycle. How soil microbial communities will respond to global warming, however, is not well understood. To elucidate the effect of warming on the microbial community we analyzed soil from the soil warming experiment Achenkirch, Austria. Soil of a mature spruce forest was warmed by 4 °C during snow-free seasons since 2004. Repeated soil sampling from control and warmed plots took place from 2008 until 2010. We monitored microbial biomass C and nitrogen (N). Microbial community composition was assessed by phospholipid fatty acid analysis (PLFA) and by quantitative real time polymerase chain reaction (qPCR) of ribosomal RNA genes. Microbial metabolic activity was estimated by soil respiration to biomass ratios and RNA to DNA ratios. Soil warming did not affect microbial biomass, nor did warming affect the abundances of most microbial groups. Warming significantly enhanced microbial metabolic activity in terms of soil respiration per amount of microbial biomass C. Microbial stress biomarkers were elevated in warmed plots. In summary, the 4 °C increase in soil temperature during the snow-free season had no influence on microbial community composition and biomass but strongly increased microbial metabolic activity and hence reduced carbon use efficiency.  相似文献

Soil tillage practices affect the soil microbial community in various ways, with possible consequences for nitrogen (N) losses, plant growth and soil organic carbon (C) sequestration. As microbes affect soil organic matter (SOM) dynamics largely through their activity, their impact may not be deduced from biomass measurements alone. Moreover, residual microbial tissue is thought to facilitate SOM stabilization, and to provide a long term integrated measure of effects on the microorganisms. In this study, we therefore compared the effect of reduced (RT) and conventional tillage (CT) on the biomass, growth rate and residues of the major microbial decomposer groups fungi and bacteria. Soil samples were collected at two depths (0-5 cm and 5-20 cm) from plots in an Irish winter wheat field that were exposed to either conventional or shallow non-inversion tillage for 7 growing seasons. Total soil fungal and bacterial biomasses were estimated using epifluorescence microscopy. To separate between biomass of saprophytic fungi and arbuscular mycorrhizae, samples were analyzed for ergosterol and phospholipid fatty acid (PLFA) biomarkers. Growth rates of saprophytic fungi were determined by [14C]acetate-in-ergosterol incorporation, whereas bacterial growth rates were determined by the incorporation of 3H-leucine in bacterial proteins. Finally, soil contents of fungal and bacterial residues were estimated by quantifying microbial derived amino sugars. Reduced tillage increased the total biomass of both bacteria and fungi in the 0-5 cm soil layer to a similar extent. Both ergosterol and PLFA analyses indicated that RT increased biomass of saprophytic fungi in the 0-5 cm soil layer. In contrast, RT increased the biomass of arbuscular mycorrhizae as well as its contribution to the total fungal biomass across the whole plough layer. Growth rates of both saprotrophic fungi and bacteria on the other hand were not affected by soil tillage, possibly indicating a decreased turnover rate of soil microbial biomass under RT. Moreover, RT did not affect the proportion of microbial residues that were derived from fungi. In summary, our results suggest that RT can promote soil C storage without increasing the role of saprophytic fungi in SOM dynamics relative to that of bacteria.  相似文献
