This study was conducted to determine the optimal dietary biotin requirement of juvenile Megalobrama amblycephala. Quadruple groups of fish (initial average weight 2.01 ± 0.01 g) were fed thrice daily with six isonitrogenous and isoenergetic purified diets containing 0 (basal diet), 0.015, 0.049, 0.158, 0.624 and 2.49 mg kg?1 biotin, respectively, for 8 weeks. Results showed that survival rate, final weight, feed intake, specific growth rate, protein efficiency ratio and nitrogen retention efficiency all increased significantly (< 0.01) as dietary biotin levels increased from 0 to 0.049 mg kg?1, whereas the opposite was true for feed conversion ratio. Dressout percentage, condition factor, hepatosomatic index, viscera/body ratio all showed no significant difference (> 0.05) within the biotin range tested. Contrary to moisture content, whole‐body protein and lipid contents showed a positive correlation with dietary biotin levels. In addition, liver biotin content increased significantly (< 0.05) with increasing dietary biotin levels up to 0.624 mg kg?1. Hepatic pyruvate carboxylase (PC) and acetyl‐CoA carboxylase (ACC) activities both showed an increasing trend as dietary biotin levels increased. Based on the regression analysis of weight gain, hepatic PC and ACC activities, the optimal dietary biotin requirement of juvenile Megalobrama amblycephala is estimated to be 0.063, 0.071 and 0.075 mg kg?1, respectively.  相似文献

An eight‐week feeding trial was conducted to determine the dietary histidine requirement of juvenile blunt snout bream. The results showed that final body weight, weight gain rate and specific growth rate significantly increased with increasing dietary histidine levels up to 9.9 g/kg (p < .05) and decreased gradually thereafter, while feed conversion ratio showed a converse trend. The survival rate, condition factor, viscerosomatic and hepatosomatic index were not significantly affected by the graded dietary histidine levels (p > .05). About 9.9 g/kg dietary histidine level significantly improved whole‐body protein and lowered moisture content. The contents of plasma total protein, cholesterol, urea and triglyceride levels were not significantly affected by dietary histidine levels. About 9.9 g/kg dietary histidine level significantly upregulated target of rapamycin and insulin‐like growth factor mRNA levels (p < .05), and 12.1 g/kg and 14.2 g/kg dietary histidine levels significantly upregulated eukaryotic translation initiation factor 4E‐binding protein 2 and ribosomal protein S6 kinase‐polypeptide 1 mRNA levels (p < .05). Based on second‐degree polynomial regression analysis of weight gain rate, and specific growth rate against dietary histidine levels, the dietary histidine requirement for juvenile blunt snout bream was estimated to be 11.2 g/kg of diet, corresponding to 36.1 g/kg of dietary protein.  相似文献

A 16‐week feeding trial was conducted to assess the effects of replacing fish meal (FM) with cottonseed meal (CM) on the growth performance, feed utilization, plasma biochemical composition and target of rapamycin (TOR) pathway gene expression of juvenile blunt snout bream (Megalobrama amblycephala). Five isonitrogenous (36% crude protein) and isoenergetic (16 MJ kg?1) diets with graded replacing levels of CM (replacing 0, 25, 50, 75 and 100% FM with CM) and similar lysine and methionine concentrations were fed to triplicate groups of fish. Results revealed that specific growth rate (SGR), feed intake and protein efficiency ratio (PER) of group fed with diets replacing FM with CM up to 50% were significantly higher than others (< 0.05). However, final body weight and feed conversion ratio (FCR) remained similar up to 25% and thereafter significantly decreased and increased, respectively, as the replacement level increased (< 0.05). Replacement levels significantly decreased hepatosomatic index and viscerosomatic index (< 0.05), but did not significantly affect condition factor and whole body compositions. Free gossypol mainly accumulated in liver and significantly increased in diets CM75 and CM100 than CM0 (< 0.05). Replacement significantly influenced plasma urea contents (< 0.05). Meanwhile, increasing replacement of FM with CM in diets increased insulin‐like growth factor I (IGF‐1) gene expression in liver of blunt snout bream. Target of rapamycin (TOR) gene expression in diet CM100 was significantly lower than that in diets CM0 and CM75, while the eukaryotic translation initiation factor 4E‐binding protein 2 (4E‐BP2) gene expression was not affected by the replacement level of CM in diets. Therefore, based on final body weight and FCR, it suggested that up to 25% of FM (150 g kg?1 in basal diet) could be replaced by CM in diets, which was 112.5 g kg?1 FM and 192.9 g kg?1 CM, for juvenile blunt snout bream.  相似文献

Six extruded diets were formulated with a graded level of fish meal (FM); diet 1 and diet 2 were formulated with 80 g/kg FM; diet 3 and diet 4 were formulated with 40 g/kg FM; and diet 5 and diet 6 were devoid of FM. Hence, diet 2, diet 4 and diet 6 were supplemented with essential amino acids (EAAs). The diets were fed throughout an eight‐week feeding trial. The results revealed that specific growth rate (SGR), feed conversion ratio (FCR), body weight gain (BWG) and protein efficiency ratio (PER) were influenced by FM reduction and improved by the addition of EAA (p < 0.05). A survival rate (SR), whole body content of protein, lipid, moisture and ash, and plasma total protein (TP), albumin (ALB), triglyceride (TG) and urea (UN) did not influence by dietary treatments (p > 0.05). Additionally, target of rapamycin (TOR) pathway did not influence by dietary treatments (p > 0.05). Expression levels of hepatic peptide transporter 1 (Pept1) and peptide transporter 2 (Pept2) decreased against FM reduction and improved significantly in the groups fed diet 4 and diet 6. In summary, the findings revealed that diet containing plant proteins mixture supplemented with EAA could totally replace FM in the practical diet of blunt snout bream.  相似文献

A 9‐week feeding trial was conducted to investigate the dietary methionine requirement of juvenile Megalobrama amblycephala at a constant dietary cystine level. Six semipurified diets were formulated to contain graded dietary methionine levels from 3.9 to 15.4 g kg?1 in about 2.5 g kg?1 increments in the presence of 2.2 g kg?1 cystine. Results showed that specific growth rate (SGR) and protein efficiency ratio (PER) significantly increased with increasing dietary methionine levels from 3.9 to 12.4 g kg?1 and thereafter kept stable. Maximum protein productive value (PPV), nitrogen retention efficiency (NRE) and liver weight were observed in 8.5 g methionine kg?1 diet. Protein contents in whole fish body were positively correlated with dietary methionine level, while lipid contents were negatively correlated with it. Morphological index and hepatic glutamate‐pyruvate transaminase (GPT) activities were independent of dietary methionine levels. However, dietary methionine supplementation significantly improved haematological parameters, plasma methionine and total essential amino acid contents and hepatic glutamate‐oxaloacetate transaminase (GOT) activities. Analysis of dose response using broken‐line regression on the basis of SGR and PPV versus dietary methionine level estimated the optimum dietary methionine requirements of juvenile M. amblycephala to be between 8.5 and 8.4 g kg?1 of diet (25.0 and 24.7 g kg?1 of protein) in the presence of 2.2 g kg?1 cystine, respectively. Hence, the corresponding total sulphur amino acids requirements of this species were calculated to be 10.7 and 10.6 g kg?1 of diet (31.5 and 31.2 g kg?1 of dietary protein).  相似文献

A feeding trial was conducted to investigate the effects of dietary fructooligosaccharide (FOS) on growth performance, body composition, intestinal enzymes activities and histology of fingerling Megalobrama amblycephala. A total of 1200 fish (1.42 ± 0.01 g) were fed diets containing graded levels of FOS (0, 0.5, 1, 2, 4 and 8 g kg−1 diet) for 8 weeks in a recirculating system indoor. The weight gain, specific growth rate (SGR) and survival rate were all improved in dietary supplementation of FOS fed fish. Increasing FOS levels resulted in both higher whole‐body lipid and lower moisture contents, whereas ash and protein contents showed no significant differences among all the treatments. Intestinal amylase, protease, Na+, K+‐ATPase, alkaline phosphatase, γ‐glutamyl transpeptidase and creatine kinase activities all increased with dietary FOS levels up to 4 g kg−1 (< 0.05). Transmission electron microscopy analysis indicated that microvilli length in the mid‐intestine was significantly increased with increased dietary FOS levels (< 0.05). In conclusion, dietary supplementation of FOS could confer benefits on growth performance, intestinal digestive and absorptive ability, histology of fingerling Megalobrama amblycephala.  相似文献

A 9‐week feeding trial was conducted to estimate the dietary isoleucine requirement of juvenile blunt snout bream. Six isonitrogenous and isoenergetic experimental diets were formulated to contain graded isoleucine levels ranging from 5.3 to 20.1 g kg?1 dry diet. At the end of the experiment, weight gain (WG), specific growth rate (SGR), feed efficiency ratio (FER) and protein efficiency ratio (PER) increased with increasing dietary isoleucine level up to 11.1 g kg?1 dry diet, and dietary isoleucine level above 14.2 g kg?1 dry diet declined these performances. Dietary isoleucine levels (14.2 and 17.3 g kg?1 dry diet) significantly improved whole‐body protein content, but decreased whole‐body lipid, plasma triglyceride and cholesterol contents. Significantly lower visceral fat index (VFI) in fish fed with 14.2 g kg?1 dietary isoleucine was observed compared to those fed with deficient or excessive isoleucine. Dietary isoleucine supplementation significantly increased plasma isoleucine concentration, while plasma valine and leucine concentrations showed a reversed trend. Dietary isoleucine levels regulated the target of rapamycin (TOR) gene expression and improved plasma superoxide dismutase (SOD) activity in juvenile blunt snout bream. Based on second‐order polynomial regression model analysis of SGR and FER, the optimum dietary isoleucine requirement was estimated to be 13.8 g kg?1 dry diet (40.6 g kg?1 dietary protein) and 14.0 g kg?1 dry diet (41.2 g kg?1 dietary protein), respectively.  相似文献

This study aimed to investigate the optimum dietary carbohydrate/lipid (CHO/L) ratio for fingerling blunt snout bream Megalobrama amblycephala (average initial weight: 6.61 ± 0.03 g). Six isonitrogenous and isoenergetic diets were formulated to contain various CHO/L ratios ranging from 1.62 to 24.20. Each diet was tested in four replicates for 10 weeks. Specific growth rate (SGR), feed conversion ratio and nitrogen and energy retention all improved significantly (< 0.05) as dietary CHO/L ratios decreased from 24.20 to 5.64 but showed little difference (> 0.05) as dietary CHO/L ratios ranged from 2.45 to 5.64. Hepatosomatic index increased significantly (< 0.05) as dietary CHO/L ratios increased, whereas intraperitoneal fat ratio showed an opposite trend (< 0.05). Opposite to moisture content, lipid content of whole body and carcass all increased significantly (< 0.05) as dietary CHO/L ratios decreased. Liver lipid content showed little difference (> 0.05) among all the treatments, while liver glycogen content increased significantly (< 0.05) with increasing CHO/L ratios. High dietary carbohydrate enhanced the activities of liver hexokinase, pyruvate kinase and glucose‐6‐phosphate dehydrogenase but did not induce hyperglycaemia. Based on the second‐order polynomial regression analysis of SGR, the optimal dietary carbohydrate and lipid contents for fingerling blunt snout bream were 291.7 and 81.4 g kg−1, respectively, with a corresponding dietary CHO/L ratio of 3.58.  相似文献

To investigate the effects of dietary tryptophan on growth and glycometabolism in juvenile blunt snout bream, 450 fish (initial weight 23.33 ± 0.03 g) were fed six practical diets with graded levels of tryptophan (from 0.79 g/kg to 5.96 g/kg dry matter) for 8 weeks. Results showed that final weight, per cent weight gain (PWG), protein efficiency rate, feed intake and feed conversion ratio (FCR) were significantly improved by 2.80 g/kg diet. The maximum values of protein and ash were observed in 2.80 g/kg diet, while moisture was minimum. Lipid content of fish fed 3.95 g/kg diet was significantly higher than other diets. The highest plasma insulin‐like growth factor‐1 (IGF‐1) content was observed in 0.79 g/kg diet. In the liver, IGF‐1 mRNA levels were significantly downregulated by 2.80 g/kg dietary tryptophan, while glucokinase levels were by 3.95 g/kg, while glucose‐6‐phosphatase and phosphoenolpyruvate carboxykinase mRNA levels showed a converse trend compared with IGF‐1. Based on PWG and FCR, the optimal dietary tryptophan level was determined to be 1.99 g/kg (6.20 g/kg of dietary protein) and 1.96 g/kg (6.11 g/kg of dietary protein), respectively, using broken‐line regression analysis.  相似文献

A feeding trial was conducted to assess the effects of replacing fishmeal (FM) with canola meal (CM) on the growth performance, feed utilization and related gene expression of juvenile blunt snout bream (Megalobrama amblycephala). Fishes were fed with five isonitrogenous (330 g/kg crude protein) and isoenergetic (15 MJ/kg) diets for 16 weeks, which was graded replacing levels of fishmeal (replacing 0, 37.5, 75, 112.5 and 150 g/kg FM with CM). The results revealed that specific growth rate, feed efficiency ratio and protein efficiency ratio of the groups with replacement level up to half were significantly higher than the others, while feed intake was inverse. Peptide transporter 1 (PepT1) mRNA level in the gut was significantly up‐regulated in groups CM25 and CM50 compared to other groups. Relative gene expression of IGF‐1 among groups was extremely significant based on the nonparametric test. Replacement of FM with CM caused a reduction in the hepatic expression of target of rapamycin (TOR), while significant increase trends were observed in hepatic genes of protein kinase B (AKT) and eIF4‐binding protein (4E‐BP2). However, TOR gene expression in the gut was significantly up‐regulated first and then down‐regulated, while mRNA levels of 4E‐BP2 and S6K1 in the gut were first significantly decreased and then increased in CM75, finally significantly decreased to the lowest. The overall results indicated that up to 75 g/kg FM could be replaced by CM (350.5 g/kg CM) in the diets for juvenile blunt snout bream. Besides, these signalling molecules could explain further the mechanisms that juvenile blunt snout can only maintain optimal growth performance up to 75 g/kg FM replacement with CM.  相似文献

ABSTRACT

Fish meal is the main protein source in fish diet, but due to its unsustainability, it has to be replaced with more sustainable protein sources, such as soybean meal (SBM), in fish feeds. As SBM often produces negative effects on fish growth and health, we conducted an eight-week trial of complete fish meal replacement with SBM in the diet of juvenile (48.2 ± 0.55 g) blunt snout bream (Megalobrama amblycephala), a herbivorous cyprinid fish native to the Yangtze basin (China). SBM-fed fish exhibited: a worsened growth performance; improved survival rate (> 0.05); elevated serum triglycerides and total cholesterol but reduced low-density lipoprotein in plasma (< 0.05); mild lipid droplet accumulation and swelling of the liver; elevated goblet cell counts and mild signs of enteritis in the intestines. With negligible effects on growth and mild effects on health, M. amblycephala appears to be comparatively tolerant of SBM, but total fish meal replacement is not recommended. Future studies should aim to identify the SBM tolerance threshold in long-term growth trials.

This study aimed to investigate the effects of dietary choline supplementation on growth, lipid deposition and intestinal enzyme activities of Megalobrama amblycephala. Fish were fed four diets with two lipid levels (50 and 150 g kg?1) and two choline supplementations (600 and 1600 mg kg?1) for 8 weeks. Feed conversion ratio (FCR), viscerosomatic index (VSI), hepatosomatic index (HSI), intraperitoneal fat (IPF) ratio, whole‐body and muscle lipid contents, intestinal lipase activities and lipoprotein lipase (LPL) activities all increased significantly (< 0.05) as lipid levels increased, whereas the opposite was true for whole‐body and muscle moisture contents and intestinal amylase activities. VSI, IPF ratio and whole‐body lipid contents all decreased significantly (< 0.05) with increasing dietary choline supplementations. Weight gain, muscle moisture content all increased significantly (< 0.05) with increasing dietary choline supplementations when dietary lipid levels reached 150 g kg?1, whereas the opposite was true for FCR, IPF ratio, IPF and liver LPL activities. In addition, abnormal hepatocytes were found in the liver of fish fed 150 g kg?1 lipid with 600 mg kg?1 choline supplementation. The result of this study indicated that extra choline supplementation can improve growth performance, intestinal enzymes activities and reduce excessive lipid deposition of M. amblycephala fed high lipid.  相似文献

Six semipurified diets of graded histidine levels (from 4.1 to 14.2 g/kg) were fed to fish for 8 weeks. The results showed that the intestinal content of malondialdehyde (MDA), activities of total superoxide dismutase (T‐SOD) and glutathione peroxidase (GPx) were noticeably lower in 8.0 g/kg diet compared to control group. While the lowest activities of copper–zinc superoxide dismutase (Cu/Zn‐SOD) and manganese superoxide dismutase (Mn‐SOD) were observed in 9.9 g/kg diet, the intestinal activities of total antioxidant capacity (T‐AOC) and catalase (CAT) in 14.2 g/kg diet were noticeably higher than those in 8.0 and 9.9 g/kg diets. Plasma biochemical indexes were not significantly affected by dietary histidine levels. In the intestine, the Kelch‐like ECH‐associated protein 1 (Keap1) mRNA levels were increased in 8.0 g/kg diet, which suppressed the nuclear translocation of nuclear factor E2‐related factor 2 (Nrf2), and subsequently decreased CAT, GPx1, Cu/Zn‐SOD and Mn‐SOD expression levels. The lowest mRNA levels of interleukin 8 (IL‐8) and tumour necrosis factor‐α (TNF‐α) were observed in 8.0 g/kg diet, whereas the highest mRNA levels of transforming growth factor‐β (TGF‐β) and interleukin 10 (IL‐10) were observed in 8.0 g/kg diet. These results indicated that dietary histidine plays a major role in maintaining intestinal health in juvenile blunt snout bream.  相似文献

Six diets were formulated to investigate the success of fish meal (FM) replacement by plant proteins; diet 1 reflected a commercial feed (8% FM), diet 3 contained 4% FM, and diet 5 was devoid of FM. Whereas, diet 2, diet 4, and diet 6 reflected diet 1, diet 3, diet 5, respectively, and supplemented with essential amino acid (EAA). At the end of 8‐week trial, there was no significant difference in survival rate. Significantly higher final weight, weight gain rate, and specific growth rate were recorded in the group fed diet 2 compared with the other treatments (except diet 4) (p < 0.05). Feed conversion ratio of fish fed diet 2 was significantly lower than those fed diets 1, 3, and 5 (p < 0.05). The lowest feed intake and highest protein efficiency rate were found in fish fed diet 2 (p < 0.05). There was no significant difference in whole body compositions between treatments. Plasma aspartate transaminases, alanine aminotransferase, and glucose were significantly affected by dietary treatments (p < 0.05), while plasma protein and albumin contents were not influenced by the treatments. The relative expression of target of rapamycin (TOR) and phosphatidylinositol 3‐kinase, regulatory subunit 1 (alpha) (PIK3R1) in fish fed diet 3 (4% FM) were significantly down‐regulated compared with those fed diet 6 for TOR and diets 4 and 6 for PIK3R1 (p < 0.05). Insulin receptor substrate 1 (IRS‐1) and janus kinase 3 (JAK3) expressions were fluctuated, with the higher levels in fish fed diets 4 and 6. In conclusion, the findings of this study indicate that plant protein mixture supplemented with EAA could be used to substitute FM in practical diet for Megalobrama amblycephala.  相似文献

Six diets were designed to investigate the effects of dietary docosahexaenoic acid (22:6n‐3; DHA) levels (0.5, 1.3, 2.3, 4.2, 8.1 and 15.9 g/kg diets) on growth performance, fatty acid profile and expression of some lipogenesis‐related genes of blunt snout bream (Megalobrama amblycephala). Fish (average weight: 26.40 ± 0.11 g) were randomly fed one of six diets for 8 weeks. Results indicated that the final body weight (FBW) and specific growth rate (SGR) of fish fed 1.3 g/kg DHA were significantly higher than other groups except for the 2.3 g/kg DHA (p < .05). Compared with other groups, the number of lipid droplet clusters of the liver stained with oil red O in the 2.3 g/kg DHA group was the highest, which was consistent with the lipid contents of whole body and liver. The DHA proportion in liver and muscle significantly increased with the increasing dietary DHA levels (p < .05), which reflected fatty acid profiles of diets. The highest mRNA expressions of acetyl‐CoA carboxylase α (ACCα), fatty acid synthase (FAS) and sterol regulatory element‐binding protein‐1 (SREBP‐1) occurred in the 1.3 g/kg DHA group, followed by 2.3 g/kg DHA. In summary, the supplementation of 1.3–2.3 g/kg DHA could improve growth performance and lipogenesis, and the dietary DHA could improve DHA and PUFA proportion in liver and muscle.  相似文献

The 8‐week experiment was conducted to evaluate the effects of partial replacement of fish meal (FM) with soybean protein concentrate (SPC) on juvenile black sea bream, Acanthopagrus schlegelii (10.70 ± 0.04 g). Diets were formulated to replace FM protein by SPC at 0, 8, 16, 24, 32 or 40% (designated as T1, T2, T3, T4, T5 and T6, respectively). Diets except T1 were supplemented with phytase at 2000 phytase activity U kg?1. The results showed that survival rate, growth performance and feed utilization were not significantly affected by increasing dietary SPC. Fish fed diet T3 had higher feed intake compared to those fed T1, T2 and T5 diets. Whole body compositions of black sea bream were significantly influenced by SPC replacing FM except for protein, ash and phosphorus content. Condition factor of fish was significantly lower in T2 than that of fish in T3 group. Apparent digestibility coefficients (ADCs) of dry matter was higher in fish fed T6 diet than those of fish fed T1 and T2 diets, ADCs of phosphorus increased with dietary SPC level up to T3 and then decreased. The results obtained in this study indicate that FM protein could be effectively replaced by SPC protein with phytase in diet of black sea bream.  相似文献

This study evaluated effects of fishmeal replacement by cottonseed meal protein hydrolysate (CPH) on growth, antioxidant and immunity of Megalobrama amblycephala and its resistance to Aeromonas hydrophila. Fish (average weight: 38.66 ± 0.08 g) were divided into five groups and fed with five isonitrogenous (320 g/kg crude protein), isolipidic (70 g/kg crude fat) and isocaloric (17.8 MJ/kg gross energy) diets replacing fishmeal with 0%, 1%, 3%, 5% and 7% CPH, respectively. The control diet contained 60 g/kg fishmeal. Final weight and weight gain of fish fed 5% and 7% CPH were significantly lower than that of fish fed control diet (p < 0.05). 3% CPH significantly increased total protein, globulin, acid phosphatase, C3 and C4 contents of plasma and total‐superoxide dismutase and catalase activities of liver (p < 0.05), as well as upregulated the antimicrobial peptides 1 and 2 (Leap‐1 and Leap‐2) expression levels in liver and spleen (p < 0.05). After challenge, the lowest mortality was observed in fish fed 3% CPH, and it was significantly lower than that in fish fed the 7% CPH (p < 0.05). In conclusion, dietary fishmeal replacement by 3% CPH could increase antioxidative capacity, as well as enhance immunity of fish.  相似文献

An 8‐week feeding trial was performed to investigate effects of dietary selenium (sodium selenite, Na2SeO3; selenium nanoparticle, Nano‐Se; selenium yeast, Se‐yeast) on the growth, selenium status, antioxidant activities, muscle composition and meat quality of blunt snout bream. Na2SeO3 and Nano‐Se were supplemented at 0.2 mg Se kg?1, and Se‐yeast was supplemented at 0.1, 0.2, 0.4 and 0.8 mg Se kg?1 in basal diet; no Se was added as a control. The results indicated that groups of 0.2 and 0.4 mg kg?1 Se‐yeast had significantly higher weight gain and Nano‐Se, 0.2, 0.4 mg kg?1 Se‐yeast had significantly lower feed conversation ratio compared with the control group. The Se concentrations of whole body, muscle and liver linearly increased with increasing dietary Se‐yeast levels. Group of 0.4 mg kg?1 Se‐yeast significantly increased activities of catalase and glutathione peroxidase. Muscle colour of Nano‐Se, 0.2, 0.4 mg kg?1 Se‐yeast groups, the water‐holding capacity of 0.4, 0.8 mg kg?1 Se‐yeast groups showed significant differences compared with the control group. The results suggest that Se‐yeast and Nano‐Se had a better growth performance than Na2SeO3 at 0.2 mg Se kg?1, and supplementing appropriate Se‐yeast in diet can increase antioxidant activities and enhance meat quality of blunt snout bream.  相似文献
