This study was conducted to evaluate the dietary use of vegetable oil sources in grass carp (Ctenopharyngodon idella). Fish were fed diets having the same lipid level from fish oil (FO), palm oil (PO), rapeseed oil (RO), soybean oil (SO) or linseed oil (LO). The results showed that dietary vegetable oils significantly decreased the feed utilization and a significantly lower growth was observed in SO group, while palm oil showed no effect on the growth performance. Dietary vegetable oils suppressed intestinal digestion by inhibiting the activities of digestive enzymes. Vegetable oils significantly elevated the activities of lipase, hepatic lipase and total lipase in liver, and a phenomenon of intense lipid accumulation emerged in liver of PO and SO groups. Furthermore, linseed oil significantly decreased plasma antioxidant capacity, whereas no significant difference was found between RO and FO groups. Dietary vegetable oils caused a significantly lower EPA and DHA in muscle, and further influenced fillet quality through an increase in cohesiveness, gumminess, chewiness and/or springiness, together with a decrease in hardness. Overall, our results indicated that rapeseed oil may be a suitable alternative oil source for grass carp from a point of growth, feed conversion ratio, antioxidative capacity and hepatic histology.  相似文献

An 8‐week feeding trial was conducted to investigate the effect of dietary selenium (Se) on feed intake, weight gain and antioxidant activity in juvenile grass carp (11.2 ± 0.03 g). Six Se levels (0.13, 0.41, 0.56, 1.12, 2.18 and 4.31 mg/kg) of semi‐purified diets were assayed in triplicate. The maximum weight gain, specific growth rate and feed intake were obtained in fish fed with 1.12 mg Se/kg diet. Hepatic glutathione peroxidase activity was markedly increased when dietary Se ≤1.12 mg/kg diet and reached a plateau when dietary Se ≥1.12 mg/kg diet. Hepatic superoxide dismutase and serum catalase activities in juvenile grass carp fed with 0.56, 1.12 and 2.18 mg Se/kg diets were all significantly higher than those in the other groups. The malondialdehyde content in liver and serum was firstly decreased and then increased with increasing dietary Se content, and the lowest content was observed in fish fed with 1.12 mg Se/kg diet. With the increase in Se level, the activities of serum alanine aminotransferase and aspartate aminotransferase were reduced. In addition, serum alkaline phosphatase activity and albumin content were highest in fish fed with 1.12 mg Se/kg diet. This study indicated that both the Se deficiency and excess of Se caused negative effect on the oxidative stress in juvenile grass carp and suggested that the health‐giving concentration of dietary inorganic Se was 1.12 mg/kg diet. Moreover, based on the broken‐line regression analysis of weight gain, the optimal concentration of dietary inorganic Se was 0.83 mg/kg for juvenile grass carp.  相似文献

A feeding trial was conducted for 8 weeks to evaluate the effects of supplemental phytic acid (PA) on the apparent digestibility and utilization of dietary amino acids (AAs) and minerals in juvenile grass carp. Five experimental diets consisted of graded levels of PA (0.2, 4.7, 9.5, 19.1 and 38.3 g kg?1, named as P0, P5, P10, P20 and P40). Triplicate groups of fish (initial weight, 22.37 ± 0.16 g) were fed twice daily (08:00 and 16:00 h). The crude protein content in whole body significantly (< .05) decreased in fish fed with 19.1 and 38.3 g PA kg?1 diet. Supplemental PA (>4.7 g kg?1) significantly reduced the apparent digestibility coefficient (ADC) of AAs (Asp, Thr, Ser, Glu, Gly, Ala, Cys, Val, Met, Ile, Leu, Phe, Lys, Pro, His and Arg) and the ADC of minerals (P, Ca, Mg, Zn, Cu, Fe and Mn) in grass carp. The contents of minerals (P, Ca, Mg and Zn) in whole body and bone were also found to be significantly reduced in dietary PA > 4.7 g kg?1, while the bone ash, serum Alkp and Zn contents were found to be significantly decreased when the PA supplementation level was above 9.5 g kg?1, and the contents of serum Ca and Mg were found to be markedly altered in higher PA‐supplemented groups. The results indicated that supplemental PA decreased the apparent digestibility and utilization of AAs and minerals, and thus reduced the feed utilization of grass carp, suggesting that the level of total PA should be below 4.7 g kg?1 in grass carp diet.  相似文献

A 95‐day feeding trial was conducted to determine the effects of n‐3 highly unsaturated fatty acids (n‐3 HUFA) on the growth, antioxidation and lipid metabolism in grass carp (Ctenopharyngodon idellus). Two isonitrogenous and isolipidic diets were formulated with either lard oil (LO) or fish oil (FO) as the main lipid source. The results showed that the intraperitoneal fat (IPF) ratio was significantly lower (P < 0.05) in FO group. The concentration of n‐3 HUFA in muscle, hepatopancreas and IPF was significantly higher in FO group (P < 0.05). The serum low‐density lipoprotein (LDL) content was significantly lower (P < 0.05), and glucose (GLU) content was significantly higher (P < 0.05) in FO group. The serum total superoxide dismutase (T‐SOD) activity was significantly higher (P < 0.05) in FO group, consistent with the serum malondialdehyde (MDA) content. The gene expression of IPF fatty acid synthase (FAS), acetyl‐CoA carboxylase (ACC), sterol regulatory element‐binding protein (SREBP‐1) and peroxisome proliferator‐activated receptor γ (PPARγ) was significantly lower (P < 0.05) and that of peroxisome proliferator‐activated receptor α (PPARα) was significantly higher (P < 0.05) in FO group compared with LO group. Similar trends were found in the hepatopancreas, except for PPARγ. It is suggested that n‐3 HUFA could inhibit lipid accumulation in grass carp by affecting the expression of lipid‐metabolism‐related genes.  相似文献

实验选取平均体质量(31.86±1.47) g的草鱼, 随机分为2个实验组, 对照组(CG)和饥饿组(SG), 每组3个平行, 饥饿处理15、30、45和60 d, 测定饥饿对鱼体的生长、肌肉生化组成、血液生化指标以及蛋白质代谢的影响。结果表明: 饥饿组肥满度、脏体比、肝体比显著低于对照组。随着饥饿时间的延长, 草鱼肌肉中的水分含量逐渐增加, 脂肪和蛋白质含量呈现降低趋势, 脂肪含量在4个时期没有明显的差异(P>0.05), 水分和蛋白质含量分别呈现差异性增加和降低的趋势。血清总蛋白在饥饿15 d时与对照组没有明显的差异(P>0.05), 其后3个时期对照组显著高于饥饿组(P<0.05), 但饥饿组后3个时期趋于稳定。随着饥饿时间的延长, 白蛋白、总胆固醇和甘油三脂含量在4个时期与对照组都有明显变化(P<0.05), 饥饿组呈现先降低后升高再降低的波浪形变化。丙氨酸氨基转移酶活性在15、60 d时与对照组没有明显的变化(P>0.05), 在30 d时先降低, 45 d时显著升高(P<0.05)。随着饥饿时间的延长, 与对照组相比RNA︰DNA在肌肉中没有明显的差异(P>0.05), 在肝胰脏中与对照组呈现显著性差异, 随着时间的延长饥饿组呈现先降低后增高的趋势。谷氨酸脱氢酶活性在肌肉中于饥饿15 d时与对照组没有明显差异(P<0.05), 其后比对照组显著增加, 随着时间的延长饥饿组谷氨酸脱氢酶活性呈现上升的趋势; 在15、45 d时与对照组没有明显的变化(P>0.05), 在30、45 d时显著升高(P<0.05)。肝胰脏中谷氨酸脱氢酶活性在4个时期比对照组有显著的增加; 与肌肉相比, 肝胰脏和血清中谷氨酸脱氢酶活性是一种有效的反应蛋白质代谢的指标。草鱼对体内贮能物质(蛋白质和脂肪)的动用并不是简单地与饥饿时间呈线性相关, 而是出现周期性和阶段性起伏的变化。  相似文献

Growth performance, digestive and absorptive capacities and target of rapamycin (TOR), ribosomal protein S6 kinase 1 (S6K1) and eIF4E‐binding protein (4E‐BP) gene expression in the hepatopancreas and intestine of juvenile grass carp (Ctenopharyngodon idellus) fed graded ratios of dietary alpha‐linolenic acid/linoleic acid (ALA/LNA) (0.01, 0.34, 0.68, 1.03, 1.41, 1.76 and 2.15) for 60 days were investigated. The results showed that ALA/LNA ratio of 1.03 significantly improved (i) per cent weight gain (PWG) and feed efficiency, (ii) hepatopancreatic trypsin, chymotrypsin, lipase, amylase and intestinal creatine kinase (CK) activities, (iii) hepatopancreatic trypsinogen‐2 and chymotrypsinogen mRNA levels. Meanwhile, fish fed with ALA/LNA ratio of 0.68 significantly enhanced, (iv) Na+/K+‐ATPase and γ‐glutamyl transpeptidase activities in whole intestine, and alkaline phosphatase activities in the proximal intestine (PI) and distal intestine, (v) amylase, intestinal Na+/K+‐ATPase alpha‐subunit isoform 1, Na+/K+‐ATPase alpha‐subunit isoform 8 and CK mRNA abundances, (vi) TOR and S6K1 gene expression in the hepatopancreas and intestine of juvenile grass carp. Based on the quadratic regression analysis of PWG, cholecystokinin and leptin contents in the PI, optimal dietary ALA/LNA ratio of juvenile grass carp (8.78–72.00 g) was estimated to be 1.08, 1.19 and 1.05, respectively.  相似文献

Six isonitrogenous and isoenergetic diets were used to test the influence of lipid source on growth performance, antioxidant status and lipid metabolism of juvenile Russian sturgeon, Acipenser gueldenstaedtii. Each diet was supplemented with 90 g kg?1 of lipid from each of six sources including fish oil (FO), beef tallow (BT), sunflower oil (SO), linseed oil (LO) and equal combinations of FO + SO + BT (FSB) or LO + SO + BT (LSB). After 56 days, fish fed LSB demonstrated highest weight gain, specific growth rate and lowest hepatosomatic index among all groups. The n‐6 polyunsaturated fatty acids (PUFAs) in the whole fish were highest in the SO group, and n‐3 PUFAs were highest in fish fed LO. The fish fed FO contained highest n‐3 highly unsaturated fatty acids. Triglyceride in the serum of fish fed LSB was lowest, but was not significantly different from that in the SO group. Triglyceride in the serum of fish fed FO and BT was highest among all groups. Lipase, malate dehydrogenase and lipoprotein lipase activities were highest in fish fed LSB. Serum malondialdehyde in fish fed LSB was significantly lower than in fish fed FO or SO, but no significant differences were found among fish fed LSB, BT, LO or FSB. Fish fed LSB showed higher catalase activity and total antioxidant capacity than fish fed FO or FSB. This study indicates that linseed, sunflower oil and BT mixed oil are a suitable lipid source and can benefit growth performance and antioxidation in juvenile sturgeon.  相似文献

A 56‐day feeding trial was conducted to elucidate the effects and mechanism action of dietary α‐linolenic acid (ALA, 18:3n‐3) on lipid accumulation and fatty acid profile of muscle, hepatopancreas and intraperitoneal fat (IPF) in juvenile grass carp using three isonitrogenous and isoenergetic semi‐purified diets containing 0.0% (control group), 1.0% and 2.0% ALA, respectively. The lowest intraperitoneal fat (IPF) ratio was found in 2.0% group. In the muscle, hepatopancreas and IPF, docosahexaenoic acid (DHA, 22:6n‐3) and eicosapentaenoic acid (EPA, 20:5n‐3) contents increased with the increase in dietary ALA. In the IPF, caspase 3, caspase 8 and caspase 9 showed the highest activities in 2.0% group, while the value of Bcl‐2/Bax (B‐cell leukaemia 2/Bcl‐2‐associated X protein) reached the lowest. Meanwhile, swelling of the IPF mitochondria was observed in 2.0% group. The gene expressions of fatty acid desaturase (FAD) and fatty acid elongase (ELO) in the hepatopancreas and muscle showed significantly higher levels in the treatment groups, whereas an opposite trend was existed in the IPF. Fatty acid synthase (FAS), sterol regulatory element binding protein‐1c (SREBP‐1c) in the IPF and hepatopancreas reached the lowest in 2.0% group. Overall, dietary ALA could promote n‐3 highly unsaturated fatty acids (HUFAs) synthesis and suppress the accumulation of lipid by decreasing the expression of related genes and promoting the apoptosis in IPF.  相似文献

To study the effects of manganese on growth performance, digestive and absorptive abilities, as well as the antioxidative capacity in the hepatopancreas and intestine, young grass carp (Ctenopharyngodon idellus Val.) (264 ± 1 g) were fed diets containing graded levels of manganese at 3.65 (control), 8.62, 13.48, 18.24, 22.97 and 27.86 mg kg?1 diet for 8 weeks. Per cent weight gain (PWG) and feed intake were the poorest in fish fed the basal diet (< 0.05). The activities of trypsin, lipase and alkaline phosphatase in the intestine were significantly enhanced with dietary manganese level at 13.48 mg kg?1 diet (P < 0.05). Additionally, in the hepatopancreas and intestine, the protein carbonyl and malondialdehyde contents were the lowest in fish fed the diet with dietary manganese level at 13.48 mg kg?1 diet (< 0.05), while the anti‐hydroxyl radical capacities, manganese superoxide dismutase (MnSOD), glutathione peroxidase and glutathione‐S‐transferase activities were significantly enhanced with dietary manganese level at 13.48 mg kg?1 diet (< 0.05). Moreover, the catalase activity and glutathione content in the intestine were the highest in fish fed the diet with dietary manganese level at 18.24 mg kg?1 diet (< 0.05). These results indicated that optimum dietary manganese promoted growth, enhanced the digestive and absorptive abilities, and improved the antioxidative capacity in young grass carp. Based on the quadratic regression analysis for PWG and intestinal MnSOD activity, the manganese requirements for young grass carp with the initial body weight of 264 g were 16.91 and 18.21 mg kg?1 diet respectively.  相似文献

This study investigated the effects of phenylalanine on growth, digestive and absorptive ability and antioxidant status of young grass carp (Ctenopharyngodon idella). Young grass carp were fed diets containing 3.4 (basal diet), 6.1, 9.1, 11.5, 14.0 and 16.8 g phenylalanine kg?1 diet with a fixed of 10.7 g tyrosine kg?1 diet for 8 weeks. Percent weight gain (PWG), feed efficiency and feed intake of fish were the lowest in fish fed the basal diet (< 0.05). Trypsin, lipase and amylase activities in the hepatopancreas, and antioxidants including glutathione contents and glutathione reducase activities in the hepatopancreas and intestine were all the highest in fish fed 11.5 g phenylalanine kg?1 diet (< 0.05). Trypsin, chymotrypsin and amylase activities in whole intestine, and creatine kinase, Na+, K+‐ATPase and alkaline phosphatase activities in the proximal intestine, and superoxide dismutase activities in the hepatopancreas and intestine were all the highest when phenylalanine at level of 9.1 g kg?1 diet (< 0.05). In conclusion, phenylalanine improved growth, digestive and absorptive ability, and antioxidant capacity of young grass carp. The phenylalanine requirement of young grass carp (256–629 g) based on PWG was 10.4 g kg?1 diet or 3.44 g 100 g?1 protein.  相似文献

A 6‐week study was conducted to determine the effects of different lipid sources in pelleted diets on juvenile mud crab Scylla paramamosain. Five isonitrogenous and isolipidic diets containing 8% level of fish oil (FO), lard (LD), safflower oil (SO), perilla seed oil (PO) or mixture oil (MO; VFO:VSO:VPO = 1:1:1), and a live food of marine bivalve Potamocorbula rubromuscula as the control diet (CF), were fed to groups of 25 juvenile crabs (average initial weight 7.4 g, carapace width 3.5 cm) in triplicate. The results showed that crabs fed MO had the highest survival (< 0.05). The moisture content was significantly higher in crabs fed LD, SO and PO (< 0.05). Crabs fed SO exhibited the lowest crude protein and lipid (< 0.05). Ash contents were obviously lower in LD group (< 0.05). Highest total lipid in the hepatopancreas and muscle was in LD and FO group respectively. Glucose, total cholesterol and low‐density lipoprotein were higher while high‐density lipoprotein was lower (< 0.05) in LD group. Tissue fatty acid compositions were consistent with those in diets. FO and MO diets had the same depression effect like CF on fatty acid synthase activity and mRNA expression in the hepatopancreas. The results of this study indicated that FO and mixed oil are suitable for preparation of pelleted diets with better effects for juvenile S. paramamosain compared with live food, and the ratio of n‐6/n‐3 fatty acids in pelleted diets must be <1.  相似文献

Recent studies in terrestrial animals have shown that feeding the oxidized lipids led to a reduction in triacylglycerols (TAG) and total cholesterol (TC) in liver and plasma. However, limited information is available on the effect of oxidized lipids on lipid metabolism in fish. In this study, four diets containing 0 g kg?1 (control: fresh fish oil), 30 g kg?1 (low‐oxidized oil, LOO), 60 g kg?1 (medium‐oxidized oil, MOO) and 90 g kg?1 (high‐oxidized oil, HOO) graded oxidized oil levels with the same dietary lipid level were fed to channel catfish for 86 days. The tissue lipid metabolism and fatty acid composition of the fish were investigated after this period. The results showed that plasma and liver concentrations of TAG and TC decreased with increasing dietary oxidized oil level (< 0.05). Decreasing liver lipoprotein lipase and hepatic lipase activities were observed with increasing dietary oxidized fish oil inclusion (< 0.05). The liver C22:6n?3 concentrations significantly decreased with increasing dietary oxidized oil level (< 0.05), while muscle lipid had a high proportion of polyunsaturated fatty acids. It suggests that the adverse effects of dietary oxidized oil may be induced by inhibiting lipid metabolism enzymes and, consequently, inhibition of cholesterol homoeostasis and fatty acid synthesis.  相似文献

The study was conducted to investigate the effects of dietary tea polyphenols (TP) on growth performance, biochemical and antioxidants responses, fatty acid composition, and lipid metabolism‐related gene expressions of large yellow croaker (Larimichthys crocea). Four diets were formulated with different levels of TP (0.00%, 0.01%, 0.02% and 0.05%). Results showed that growth performance of L. crocea were not different among dietary treatments. Compared with the control group, fish in 0.02% TP group had lower body and hepatic lipid content and lower total cholesterol content. The minimum content of triglycerides and low‐density lipoprotein‐cholesterol were found in 0.05% TP group. Hepatic n‐6 PUFA and n‐3 PUFA were significantly higher in TP supplementation groups. Malondialdehyde content was lower in TP supplementation groups, and superoxide dismutase activity was higher in 0.01% TP group than the control group. The mRNA expressions of carnitine palmitoyltransferase1, acyl‐CoA oxidase and peroxisome proliferators‐activated receptor α were up‐regulated in 0.01% and 0.02% TP groups, while lipoprotein lipase expression was down‐regulated in TP supplementation groups than the control group. Results suggested that 0.01%–0.02% TP supplementation could reduce the deposition of liver lipid of L. crocea caused by high‐lipid diet, which might be due to the increase in lipid oxidation related gene expressions.  相似文献

In order to investigate the effects of lysine and dissolved oxygen on grass carp, the grass carp were fed with 13, 15 and 17 g kg −1 lysine diet at about 6 mg L−1 (high dissolved oxygen, HO group) or 3.5 mg L−1 (low dissolved oxygen, LO group) dissolved oxygen level, for 8 weeks. The fish were fed to apparent satiation by hand. The results showed that apparent digestibility of protein, energy and dry matter were decreased significantly when grass carp were fed at 3.5 mg L−1 dissolved oxygen, and feed intake (FI) was also inhibited by low dissolved oxygen (< 0.05). Weight gain, protein retention, protein efficiency, feed conversion ratio and amino acid retention of fish at 6 mg L−1 dissolved oxygen level were significantly improved at 3.5 mg L−1 dissolved oxygen level (< 0.05). Weight gain, protein and amino acid retention, and feed efficiency of grass carp at the two dissolved oxygen levels were significantly improved by lysine supplementation (< 0.05). The dietary lysine level and dissolved oxygen of water had an interaction effect on feed conversion ratios (< 0.05). Grass carp fed at low dissolved oxygen level showed lower liver protein and fat contents. Plasma aspartate aminotransferase (AST) activity of grass carp fed at 3.5 mg L−1 dissolved oxygen level was significantly increased compared to 6 mg L−1 dissolved oxygen level (< 0.05). Our results show that low dissolved oxygen level of water is harmful to the liver of grass carp.  相似文献

The fatty acid synthase (FAS) gene was cloned from liver of grass carp (Ctenopharyngodon idella) by degenerate oligonucleotide primed PCR. The obtained cDNA fragment was 683 bp, which encoded 227 amino acids. Then, grass carps with initial body weight of (134.89 ± 12.12) g were fed diet supplemented with 0, 20, 40 g kg?1 fat to investigate the impacts of dietary fat levels on growth, liver FAS enzyme activity and mRNA expression. After 8 weeks feeding, final body weights of the three groups were 344.11, 347.23 and 373.02 g. Compared with control group (0 g kg?1), growth rate of 40 g kg?1 fat group was increased by 14.03%, and feed conversion rate decreased by 11.32% (P < 0.05), liver FAS enzyme activity of 20, 40 g kg?1 fat groups were reduced by 33%, 64% (P < 0.05), and FAS mRNA expression level reduced by 18%, 74% (P < 0.05), respectively. Results above showed that 40 g kg?1 fat addition can significantly improve growth performance of grass carp. Liver FAS activity and mRNA expression tended to be inhibited by the increasing dietary fat level. Fat containing high levels of polyunsaturated fatty acids had strongly inhibitory effects on liver FAS activity and gene expression.  相似文献

A 70‐day experiment was conducted to examine the effects of different macroalgal meals and lipid sources on growth, body wall composition and fatty acid (FA) profile of sea cucumber Apostichopus japonicus. Two macroalgal meals including Sargassum muticum (SM) and Gracilaria lemaneiformis (GL) and two lipid sources including fish oil (FO) and vegetable oil (VO) were formulated into four diets, i.e., S. muticum and fish oil (SF), S. muticum and vegetable oil (SV), G. lemaneiformis and fish oil (GF) and G. lemaneiformis and vegetable oil (GV). The results showed that the specific growth rates (SGR) of A. japonicus fed diets containing SM were significantly higher than those fed diets containing GL. No significant differences in SGR between the FO‐based and VO‐based groups were observed. Similar results were observed in the body wall lipid content. Most body wall FAs changed to resemble the dietary FA proportions because of the dietary effect. Concentrations of 20:4n‐6 of the SF and GF groups were significantly lower than the SV and GV groups, while levels of 20:5n‐3 and 22:6n‐3 were significantly higher than the SV and GV groups. The n‐3/n‐6 polyunsaturated fatty acids (PUFA) ratios of the SF and GF groups were significantly higher than the SV and GV groups. Moreover, the SF group had significantly higher 20:5n‐3 and 22:6n‐3 contents and n‐3/n‐6 PUFAs ratio than the GF group. These findings reveal that the SF diet can show beneficial effects on both growth performance and body wall n‐3 PUFAs content of A. japonicus.  相似文献

A 56‐day feeding trial was conducted to evaluate the potential use of faba bean meal (FBM) as a partial replacement of soybean meal (SBM) in the commercial diets for juvenile grass carp (Ctenopharyngodon idella) with initial average weight of 3.39 ± 0.03 g. Five isonitrogenous and isoenergetic diets were formulated to replace SBM by FBM protein at 0, 150, 300, 450 and 600 g kg?1 (designated as FBM0, FBM15, FBM30, FBM45 and FBM60, respectively). The results indicated that up to 420.3 g kg?1 of FBM can be used for grass carp without negative effects on growth and feed utilization. The higher FBM level would result in growth retardation. The apparent digestibility and intestinal enzymes activities significantly decreased with increased dietary FBM, and the lowest value was observed in diet FBM60. No significant differences were detected in protein contents of whole body, muscle and liver tissues among all treatments. Intraperitoneal fat index and lipid contents of whole body and liver tissues significantly decreased with increased dietary FBM. Serum antioxidant enzymes were significantly affected by dietary FBM level, and the lowest enzymes activities were found in diets FBM45 and FBM60. Results of this study indicated that FBM can successfully replace SBM protein in the commercial diets for grass carp, and the optimum level of SBM replacement with FBM was 450 g kg?1 on the basis of the maximum growth gain.  相似文献
