BACKGROUND

RNA interference (RNAi) technology is an environmentally friendly strategy for controlling insect pests. Lipopolysaccharide-binding protein (LBP) recognizes lipopolysaccharides, which are a major outer membrane constituent of Gram-negative bacteria. We propose that the LBP gene is a potential target for termite management; however, to date, no studies have examined this gene in termites.

RESULTS

In this study, we cloned the LBP gene of Coptotermes formosanus (Cf) and found that the mortality rate of termite workers significantly increased, and the repellence of these workers to Gram-negative bacteria was suppressed after knockdown of CfLBP using double-stranded RNA (dsRNA) injection and feeding. Moreover, the mortality rate of termite workers fed with CfLBP dsRNA and three Gram-negative bacteria (provided separately) was over 50%, which was much higher than that of termites treated with either CfLBP dsRNA or Gram-negative bacteria. Finally, we found that CfLBP impacts the IMD pathway to regulate the immune response of C. formosanus to Gram-negative bacteria.

CONCLUSION

CfLBP plays a important role in the immune defense of termites against Gram-negative bacteria. It can be used as an immunosuppressant for RNAi-based termite management and is an ideal target for termite control based on the combined use of RNAi and pathogenic bacteria. © 2023 The Authors. BACKGROUND: Among the termite infestations in the United States, the Formosan subterranean termite, Coptotermes formosanus Shiraki (Isoptera: Rhinotermitidae), is considered to be the most devastating termite pest. This pest most likely invaded North America as a result of the disembarkation of wooden military cargo at the port of New Orleans that arrived from Asia during and after World War II. It has now spread over other states, including Texas, Florida, South Carolina and California. Devastation caused by C. formosanus in North America has been estimated to cost $ US 1 billion a year. Over the past decades, organochlorines and organophosphates, the two prominent classes of termite control agents, have been banned owing to environmental and human health concerns. At the present time, phenylpyrazoles, pyrethroids, chloronicotinyls and pyrroles are being used as termite control agents. Mammalian toxicity and seeping of these compounds into groundwater are some of the drawbacks associated with these treatments. The instruction for the application of these termiticides indicate ground water advisory. Hence, with the increasing spread of termite infestation there is an increased need to discover effective, environmentally friendly and safe termite control agents with minimal mammalian toxicity. RESULTS: Chromene analogs derived from a natural‐product‐based chromene amide isolated from Amyris texana were tested in a collaborative discovery program for effective, environmentally friendly termite control agents. Several chromene derivatives were synthesized and characterized as a novel class of potential termiticides, followed by bioassays. These compounds exhibited significantly higher mortalities compared with untreated controls in laboratory bioassays. BACKGROUND: The effectiveness of chlorantraniliprole and other insecticides (bifenthrin, fipronil, indoxacarb, imidacloprid and chlorfenapyr) were tested against Coptotermes gestroi (Wasmann). Four experiments were conducted: a topical bioassay, a horizontal transfer study, an insecticide bioavailability test and a feeding bioassay. RESULTS: The topical bioassay showed that chlorantraniliprole was significantly less active to C. gestroi at 24 h post‐treatment compared with the other insecticides tested. Nevertheless, it is likely that a lesser amount of chlorantraniliprole was required to cause 50% mortality of C. gestroi at 7 and 14 days post‐treatment. The exposure duration and donor:recipient ratio affect the mortality of recipient termites. Mortality after exposure to chlorantraniliprole in sandy clay was significantly lower than in sand; however, by 14 days, > 90% of donor and recipient termites died in both substrates, irrespective of concentration. Fipronil and imidacloprid showed faster action, and high to moderate toxicity to C. gestroi. Termite workers also ceased to feed after exposure for 1 h to 50 mg kg?1 chlorantraniliprole‐treated sandy clay. CONCLUSION: Chlorantraniliprole demonstrated delayed toxicity at the lowest label rate (50 mg kg?1) in sandy clay. Its slow action will enable greater transfer of toxicant between nestmates, while feeding cessation will promote greater social interaction between healthy and exposed termites. Warehouse moth (Ephestia elutella) larvae in diapause were exposed at 15°C to methyl bromide at 8 mg litre?1 for 14.5 h and then immediately exposed at a lower concentration. The exposure at 8 mg litre?1 killed 44–69% of the larvae treated. Subsequent concentrations down to 1.1 mg litre?1 obeyed Haber's rule (concentration × time= k, a constant for mortality), but a higher concentration-time product (ct) was required for over 90% kill at 0.8 mg litre?1. Only concentrations down to 1.9 mg litre?1 obey Haber's rule if there is no prior exposure at a higher concentration. Although minimum effective concentrations are lower at 15°C than at 25°C, exposure at a higher concentration depresses the subsequent level to a similar extent at each temperature. The contribution to the efficacy of a treatment, of low concentrations persisting at the end of fumigation, is thus likely to be even greater at moderate to low temperatures than at 25°C. The implications for the development of resistance to methyl bromide are discussed.  相似文献

The effects of hexaflumuron consumption on the mortality of workers of the Western subterranean termite, Reticulitermes hesperus Banks, were observed following various exposures to a substrate treated with 14C-labeled hexaflumuron at 5 g kg(-1). Uptake of hexaflumuron by workers was rapid, peaking at approximately 280 ng hexaflumuron per termite 12 days after exposure. The onset of mortality began at day 8, with cumulative mortality reaching approximately 96% by day 45. Excretion of hexaflumuron from worker termites was rapid following various exposure periods, half-lives ranging from 2.1 to 4.7 days. Trophallaxis among worker termites was efficient, hexaflumuron levels in recipient termites approaching two-thirds of levels present in donor termites feeding continuously on hexaflumuron-treated filter paper. The effect of donor-to-recipient group ratios was negligible on the amounts of hexaflumuron transferred from donors to recipients. In laboratory tests, hexaflumuron diffused from a feeding source within 7 days. The presence of additional food sources appeared to inhibit movement of hexaflumuron. Movement of hexaflumuron by cannibalism and coprophagy occurred, but was significant only when termites were starved. Hexaflumuron also appeared to adversely affect egg development by preventing hatch. Hexaflumuron was readily transferred through termite groups, effectively suppressing laboratory populations of R hesperus. Variable efficacy in field situations employing baiting with hexaflumuron in southern California may be a consequence of sporadic feeding at bait stations, the rapid clearance of hexaflumuron from individual termites, and the difficulties in bait presentation (low foraging activity, poor bait station foraging fidelity) inherent in the foraging characteristics of the Western subterranean termite.  相似文献

The toxicity of phosphine, methyl bromide, 1,1,1-trichloroethane (methyl chloroform) and carbon dioxide and mixtures of phosphine + methyl bromide, methyl bromide + methyl chloroform, phosphine + carbon dioxide, and methyl bromide + carbon dioxide to one- to two-day-old pupae of Tribolium castaneum Herbst was studied. Joint action ratios estimated at LD50 and LD90 for a 24-h exposure indicated no enhancement of toxicity for mixtures of phosphine and methyl bromide, or methyl chloroform and methyl bromide on the pupae. Carbon dioxide up to 40% in air enhanced the toxic action of phosphine as well as of methyl bromide. Higher levels of carbon dioxide, however, failed to improve the toxicity of phosphine or methyl bromide proportionately. Carbon dioxide used alone produced a maximum of 11% mortality of the pupae exposed to 10–70% levels for 24 h. The order of toxicity of the fumigants both at LD50 and LD90 was phosphine > methyl bromide > methyl chloroform.  相似文献

BACKGROUND: Vapormate? is a cylinderised non‐flammable mixture of ethyl formate (16.7% by weight) and carbon dioxide (CO2) that has been developed as a rapid fumigant of stored grain. Four field trials were undertaken on wheat in 50 t farm silos to demonstrate the feasibility of dynamic application. To assess treatment efficacy, each trial tested mixed stages of Rhyzopertha dominica F. (>11 000), Tribolium castaneum (Herbst.) (>1500) and Sitophilus oryzae (L.) (>13 000) in mesh cages positioned through the centre of the grain bulk and on the grain surface. Ethyl formate and CO2 concentrations were measured in the silo during fumigation and in ambient air outside the 6 m fumigation zone. Application rates of 420, 660 and 940 g m?3 of ethyl formate/CO2 formulation and exposure times of 24, 3 and 72 h, respectively, were examined using wheat of 10.4–11.7% moisture content and grain temperatures between 2 and 32 °C. RESULTS: All life stages of R. dominica and T. castaneum were fully controlled under all conditions tested, and mortality of all life stages of S. oryzae was greater than 98%. CONCLUSION: Dynamic application of vaporised ethyl formate and CO2 to 50 t silos proved safe to operators and rapidly effective against stored‐grain insects on cold to warm grain. BACKGROUND: Non-repellent insecticides, including fipronil and indoxacarb, are becoming increasingly important for soil treatments to manage the eastern subterranean termite, Reticulitermes flavipes (Kollar). The effects of these insecticides on termite walking and tunneling behavior could significantly reduce their efficacy against colonies. RESULTS: Groups of R. flavipes were exposed to several concentrations of commercial formulations of fipronil and indoxacarb, and the ability of treated termites to tunnel in soil and walk was assessed. Increasing insecticide concentration resulted in a reduction in the ability of R. flavipes to walk, tunnel and form tunnel branches; the importance of these effects on the use of non-repellent insecticides is discussed. CONCLUSION: Exposure of R. flavipes to 1, 10 or 50 mg L−1 of fipronil or 50, 100 or 200 mg L−1 of indoxacarb significantly reduced termite walking and tunneling and the number of tunnel branches. Distance walked (ca 73 mm) by untreated control termites did not change over time for at least 16 h after treatment; control termites formed ca 150 cm of tunnels with ca 40 branches. BACKGROUND: Research has shown that the family of grapefruit flavors called nootkatones have significant repellant and toxic effects to Formosan subterranean termites (Coptotermes formosanus Shiraki). RESULTS: Nineteen synthetic nootkatone derivatives, along with three commercially available nootkatone derivatives, were tested for repellent activity against C. formosanus by a choice assay in a petri dish with a two‐step triage procedure. Based on the repellency threshold value, the relationships between structure and activity are discussed. CONCLUSION: Four derivatives of nootkatone have very high repellency and toxicity to C. formosanus, 9 times the potency of the primary compound nootkatone. Four other compounds have between 2 and 3 times the repellency of nootkatones, and three compounds are equal in their repellency to nootkatone. Copyright © 2010 Society of Chemical Industry  相似文献

Ephestia elutella larvae in diapause were exposed at 25°C to methyl bromide at 12 mg litre?1 for 3.5 or 7.5 h and then immediately exposed to a lower concentration. The minimum effective concentration (that at which Haber's rule, concentration × time = k, a constant for mortality, still applied) was about 3 mg litre?1 in tests with no previous exposure toa high concentration, but it was about 2.5 mg litre?1 for individuals surviving a 3.5 h exposure to 12 mg litre?1, and was about 1.6 mg litre?1 for those surviving a 7.5 h exposure to 12 mg litre?1. These exposures to 12 mg litre?1, respectively, killed 2–20% and 50–75% of larvae exposed, and hence the smaller the proportion of survivors of exposure to a high concentration, the lower the minimum effective concentration needed against them. Thus the low concentration persisting at the end of a practical fumigation should contribute significantly to the success of the treatment and be much more effective than any similar low concentration present soon after the introduction of gas.  相似文献

Destruction of yam (Dioscorea spp.) tubers by termites is a major constraint to yam production in Central Benin. To obtain the basic information for the development of appropriate strategies of termites management in yam, a structured questionnaire was used to interview 142 farmers about their knowledge of termite species of yam, susceptibility of yam landraces, and management practices in 14 villages through the study area. A total of 45 vernaculars names of termites were recorded corresponding to 10 species. All the names given to termite species had a signification, mainly related to morphological aspects, type of damage, and termite caste. Eleven features were used by farmers to identify yam termites. The infestation of yam fields mostly occurred from October to April with Amitermes evuncifer and Trinevitermes oeconomus being the most damaging termite species. Overall, eight factors were identified by farmers as favouring the proliferation of termites in yam fields with drought being the most frequently reported factor. Farmers identified 11 yam landraces that were highly susceptible to termites attack, while 12 yam landraces were listed as resistant. Application of chemicals was the most commonly reported control method, followed by destruction of termite nest, and application of botanical extracts. According to farmers, the high multiplication rate of termites is the most important constraint in managing the issue of termites in yam.  相似文献

Abstract

Seed‐bed types, fertilizers, cassava stem portions, planting time and yam cultivars were studied for the incidence of termites in cassava and yam fields. Planting cassava on ridges reduced the incidence of termites and significantly (P<0.05) increased tuber yield. The bottom stem portion of cassava stick was the least attacked by termites. Planting cuttings when rains became steady reduced termite incidence. NPKMg (12:12:17:2) suppressed termites in yam and improved yield significantly. Delay in harvesting yam tubers increased termite infestation. Yield loss was highest for Dioscorea rotundata cv. Nwopoko, with losses of more than 5 metric tonnes/ha when harvest was done after December in termite infested soil. Chinese yam showed the greatest resistance to termites.  相似文献

BACKGROUND

The Asian needle ant, Brachyponera chinensis, is an invasive ant currently spreading in urban and natural habitats throughout the eastern United States. Recent studies have documented the negative impact of B. chinensis on native ecosystems and human health, yet effective control strategies are lacking. Control difficulties are, in part, due to the unique biology of B. chinensis, which is a predatory ant and a termite specialist. Given that subterranean termites are an important nutritional resource for B. chinensis, the current study evaluated the potential of termite cuticular extract to improve the target-specificity and efficacy of commercial bait used for B. chinensis control.

RESULTS

The efficacy of bait augmented with termite cuticular extracts was evaluated in laboratory and field trials. In laboratory assays, B. chinensis colonies were offered granular bait treated with termite cuticular extract. Results demonstrated that the acceptance of commercial bait is significantly increased by the addition of termite cuticular extract or synthetic (Z)-9-pentacosene, a major component of termite cuticular extract. Foraging activity of Asian needle ants was significantly greater on baits augmented with termite cuticular extract or (Z)-9-pentacosene relative to standard bait. Furthermore, bait augmented with termite cuticular extract worked substantially faster relative to standard bait. To evaluate population effects, field studies were conducted in forested areas invaded by B. chinensis. Bait treated with termite cuticular extract scattered on the forest floor provided rapid control of B. chinensis and ant densities throughout the treated plots declined by 98% within 14 days.

CONCLUSION

The incorporation of termite cuticular extracts and individual cuticular hydrocarbons such as (Z)-9-pentacosene into traditional baits used for B. chinensis control may offer a novel tool to manage this increasingly problematic invasive ant. © 2023 The Author. Coptotermes gestroi is a subterranean termite from Asia, introduced accidentally to Brazil, where it is responsible for substantial damage in urban areas. The foraging range of this species frequently overlap with that of Heterotermes tenuis, a native termite also considered to be a severe pest. The greatest challenge in controlling these termites is to develop baits suitable for both species. A critical issue is to select compounds that act as common phagostimulants. We examined whether the feeding preferences of C. gestroi for some nutrients were similar to those already reported for H. tenuis. A double choice design was used. Consumption was evaluated by change in the dry mass of paper discs after termite feeding. C. gestroi fed preferentially on discs treated with a solution of sucrose + yeast. However, insects did not choose urea at 0.015 g/ml, one of the preferred solutions for H. tenuis. These results indicate that the two termites have different feeding preferences, and we suggest future use of sucrose + yeast in baits for management of C. gestroi.  相似文献

BACKGROUND: The ethyl formate/carbon dioxide (CO2) formulation Vapormate? is a rapid‐acting fumigant being developed for the control of stored‐grain insects. The effects have been investigated of concentration, exposure times of 1, 3, 24 and 72 h and two grain temperatures, 15 and 25 °C, on its efficacy against mixed‐stage cultures of Sitophilus oryzae (L.) Tribolium castaneum (Herbst) and strongly phosphine‐resistant Rhyzopertha dominica (F.) strain QRD569. RESULTS: High mortalities (≥92%) of mixed‐stage cultures of all three species were obtained when grain was fumigated with the formulation (193 g m?3 ethyl formate) for 1 h. Complete control of R. dominica QRD569 and T. castaneum was achieved with 63 and 76 g m?3 ethyl formate respectively, with exposure for 24 h, whereas mean mortality of S. oryzae was 86% under the same conditions. Mortalities of S. oryzae juvenile stages were significantly lower than adults under the conditions tested, which was due to pronounced tolerance of mid‐stage pupae to the fumigant. Reducing grain temperature from 25 to 15 °C had no effect on insect mortality. CONCLUSION: Ethyl formate/CO2 formulation is highly effective against stored‐grain insects over a range of concentrations and exposure times. Efficacious fumigations were conducted in as little as 1 h, and a strongly phosphine‐resistant R. dominica strain was readily controlled with the fumigant. Copyright © 2009 CSIRO, Australia. 嗜卷书虱Liposcelis bostrychophila Badonnel是一种发生在储粮环境中的重要害虫,其抗性问题十分突出。在实验室条件下,采用高CO2(35%CO2、21%O2、44%N2)、低O2+高CO2(35%C02、1%O2、64%N2)、DDVP和PH3等4种不同的处理,对嗜卷书虱连续处理20代,每代处理2次,每次处理保持对成虫死亡率35%的选择压力,得到了4个不同的抗性品系HCO2-R、HCLO-R、DDVP-R和PH3-R,抗性指数分别为3.3、5.2、10.2和4.5。经过对抗性发展趋势的分析,嗜卷书虱对这几种环境胁迫都有一定的抗性潜力。对抗性品系的现实遗传力(h^2)进行分析,其h^2分别为0.388、0.155、0.341和0.594。抗性风险评估结果表明,分别采用高CO2低O2+高CO2、DDVP和PH,处理,在50%的选择压力下,抗性增加10倍所需的代数分别为62.30、44.17、26.46和38.48代;而在90%的选择压力下,抗性增加lO倍所需的代数分别为28.1l、20.08、11.98和17.39代。因此对于采用气调处理,嗜卷书虱对低O2+高CO2处理的抗性风险比用高CO2处理大,而采用药剂处理,嗜卷书虱对DDVP的抗性风险要比PH3处理大。  相似文献

BACKGROUND: Isothiocyanates (ITCs) extracted from Armoracia rusticana Gaertn., May & Scherb. have been shown previously to have insecticidal activity. Allyl isothiocyanate (AITC), a major component of ITCs with high volatility, was therefore extracted using different methods and tested as a fumigant against four major pest species of stored products, maize weevil Sitophilus zeamais (Motsch.), lesser grain borer Rhizopertha dominica (F.), Tribolium ferrugineum (F.) and book louse Liposcelis entomophila (Enderlein). RESULTS: Whereas there was no significant difference between hydrodistillation and supercritical carbon dioxide fluid extraction in extraction rate for AITC from A. rusticana, both methods resulted in higher extraction efficiency than water extraction. AITC fumigation showed strong toxicity to the four species of stored‐product pests. Adult mortality of 100% of all four pest species, recorded after 72 h exposure to AITC fumes at an atmospheric concentration of 3 µg mL?1, showed no significant difference from that of insects exposed to phosphine at 5 µg mL?1, the recommended dose for phosphine. CONCLUSIONS: The results suggest good insecticidal efficacy of AITC against the four stored‐product pests, with non‐gaseous residuals on stored products. 广州市古树名木受白蚁为害率为16%,其中以家白蚁为害最大,占总为害率的95.6%,受害树种以樟树为最严重。本文作者使用凯雷牌白蚁粉,彩蚁路施药法,分群孔施药法和钻巢+诱杀法,于4-7月份对受害树木进行防治试验,结果证明分群孔施药法和钻巢+诱杀法处理效果比较理想。作者还对广州市古树名木的保持提出了建议。  相似文献

Heat treatment of dates at the time of harvest, was examined as an alternative to fumigation with methyl bromide, which has been phased out in Israel under the Montreal Protocol. In laboratory studies, the influence of 40°, 45°, 50° and 55°C on the levels of disinfestation and mortality ofCarpophilus hemipterus larvae was examined over a 2-h exposure period. The ratio of the number of insects found outside the feeding sites to the total number of insects, was greatest at 50°C (92.3%), significantly greater than at 40° and 55°C. At 50° and 55°C, 100% mortality was obtained. Since drying temperatures for most date varieties is between 45° and 50°C, and because percent disinfestation and control was most effective at 50°C, these findings were examined under field conditions. Field trials were carried out at a commercial drying station where the crated dates are placed on the ground inside a plastic-clad hothouse. The stacks were covered with plastic liners to form ‘drying ducts’ through which heated air was sucked using fans positioned at the opposite end of the ducts. It was shown that from 1 to 2 h were required for the dates to reach the set temperature of 50°C. During the following 2-h aeration, the dates were exposed to 50°C heated air, after which an examination of infested dates inserted into the drying ducts, and natural infestations showed that successful control and emigration were obtained. 采用水蒸气蒸馏法提取黄金香柳枝叶中的挥发油,得率为0.972%(以鲜重为基础)。通过GC-MS分析,从黄金香柳枝叶中共鉴定出18种成分,占总相对含量的99.056%,其中主要成分是甲基丁香酚(92.185%)和肉桂酸甲酯(3.795%)。黄金香柳枝叶挥发油对6种供试细菌表现出一定的抑菌作用,其中对黄瓜角斑病菌和根癌农杆菌的抑菌活性最好,抑菌圈直径分别为(9.0±0.0)mm和(8.5±0.1)mm,但仍弱于阳性对照硫酸链霉素对黄瓜角斑病菌(18.0±1.0)mm和根癌农杆菌(21.7±1.0)mm的活性。  相似文献
