Virus-neutralizing factors in tumor extracts and sera, age of the tumor, and the infecting dose of virus are important factors contributing to the infective titer of tumor tissue. An assay is described for a nonzviral factor from the medium of Rous sarcoma cells which stimulates rapid and sustained cellular overgrawth in crowded chick embryo cultures. The factor is nondialyzable and thermnolabile and is released in large amounts several days after the first visible transformation of newly infected chick embryo cells.  相似文献

Mediation of wound-related Rous sarcoma virus tumorigenesis by TGF-beta   总被引:19,自引:0,他引:19  
In Rous sarcoma virus (RSV)-infected chickens, wounding leads to tumor formation with nearly 100% frequency in tissues that would otherwise remain tumor-free. Identifying molecular mediators of this phenomenon should yield important clues to the mechanisms involved in RSV tumorigenesis. Immunohistochemical staining showed that TGF-beta is present locally shortly after wounding, but not unwounded controls. In addition, subcutaneous administration of recombinant transforming growth factor-beta 1 (TGF-beta 1) could substitute completely for wounding in tumor induction. A treatment protocol of four doses of 800 nanograms of TGF-beta resulted in v-src-expressing tumors with 100% frequency; four doses of only 10 nanograms still led to tumor formation in 80% of the animals. This effect was specific, as other growth factors with suggested roles in wound healing did not elicit the same response. Epidermal growth factor (EGF) or TGF-alpha had no effect, and platelet-derived growth factor (PDGF) or insulin-like growth factor-1 (IGF-1) yielded only occasional tumors after longer latency. TGF-beta release during the wound-healing response may thus be a critical event that creates a conducive environment for RSV tumorigenesis and may act as a cofactor for transformation in this system.  相似文献

Neuroretinal cells from 7-day-old chick embryos are transformed and induced to proliferate after infection with Rous sarcoma virus in vitro. Kinetic analysis of the hybridization of 71S RNA from Prague strain of Rous sarcoma virus with an excess of DNA from virus induced sarcomas indicated the presence of the majority of the viral genome sequences in cellular DNA with a very low average frequency per cell. About one-third of the viral sequences were at least partially complementary to DNA sequences with a higher average frequency on the order of 50 to 100 per cell. Normal chick embryo DNA was distinctly different, but contained sequences at least partially homologous to some fraction of the viral RNA.  相似文献

Purified preparations of Rous sarcoma virus (an avian tumor virus with an RNA genome) contain small amounts of double-stranded DNA. This DNA cannot be hybridized to viral RNA, but will reanneal completely with the DNA of avian cells. Extensive substitution of bromodeoxyuridine for thymidine in "viral" DNA does not photosensitize the biological activity of the virus. These observations indicate that the DNA associated with Rous sarcoma virus is derived from the DNA of the avian host cell, and is probably devoid of any function in the life cycle of the virus.  相似文献

The localization of gammadelta T cells within epithelia suggests that these cells may contribute to the down-regulation of epithelial malignancies. We report that mice lacking gammadelta cells are highly susceptible to multiple regimens of cutaneous carcinogenesis. After exposure to carcinogens, skin cells expressed Rae-1 and H60, major histocompatibility complex-related molecules structurally resembling human MICA. Each of these is a ligand for NKG2d, a receptor expressed by cytolytic T cells and natural killer (NK) cells. In vitro, skin-associated NKG2d+ gammadelta cells killed skin carcinoma cells by a mechanism that was sensitive to blocking NKG2d engagement. Thus, local T cells may use evolutionarily conserved proteins to negatively regulate malignancy.  相似文献

Murine sarcoma virus transformed mouse 3T3 cells, which are negative for murine leukemia virus and which yield sarcoma virus after superinfection with murine leukenmia virus, spotaneously give rise to flat variants front which murine sarcoma virus can no longer be rescued. The revertants support leukemia viruis growth and show an enhanced sensitivity to murine sarcoma superinfection and, like normal cells, do not release RNA-dependent DNA polymerase activity. Because revertants could be obtained with high frequency from progeny of single transformed cells, each cell that containts the sarconma virus genome seems to have the capacity to suppress or eliminate an RNA tumor virus native to its species of origin.  相似文献

Examination of multiphase melt inclusions in 91 sections of 26 lunar rocks revealed abundant evidence of late-stage immiscibility in all crystalline rock sections and in soil fragments and most breccias. The two individual immiscible silicate melts (now glasses) vary in composition, but are essentially potassic granite and pyroxenite. This immiscibility may be important in the formation of the lunar highlands and tektites. Other inclusions yield the following temperatures at which the several minerals first appear on cooling the original magma: ilmenite (?) liquidus, 1210 degrees C; pyroxene, 1140 degrees C; plagioclase, 1105 degrees C; solidus, 1075 degrees C. The glasses also place some limitations on maximum and minimum cooling rates.  相似文献

A suspension of a variant of Rous sarcoma was injected into four adult and eight newborn rhesus monkeys. Seven newborns developed tumors, three of which were diagnosed as fibrosarcomas, in 2 to 6 weeks; none of the adults have tumors after 11 weeks. Virus was demonstrated in two of the tumors by injecting a tumor-suspension preparation into the chick wing-web where tumors subsequently appeared. To the best of our knowledge this is the first time that sarcomas have developed in primates after a virus has been injected.  相似文献

 【目的】探讨pH对猪肉肌原纤维蛋白热诱导凝胶保水性及水的移动性影响。【方法】从猪肉中提取肌原纤维蛋白,用低场NMR(nuclear magnetic resonance)研究pH对肌原纤维蛋白热诱导凝胶中水的T2弛豫性质的影响。同时用离心法测量pH对肌原纤维蛋白凝胶保水性(water holding capacity,WHC)影响。【结果】NMR结果拟合后得到水有4个组分,合并为对应水的3种状态即不可移动水、可移动水和自由水。随着pH升高,pH偏离肌原纤维蛋白的等电点(pI),代表可移动水的T2弛豫时间显著增加,其所占峰的面积和凝胶的WHC也随之增加。主成分分析结果发现,处于等电点附近的样品在样品评分图上与其它pH样品显著不同。【结论】凝胶保水性的增加主要是可移动水的增加,凝胶WHC增加的原因可能是肌原纤维凝胶后孔径增加,从而可以容纳更多的水。  相似文献

The near identity of the partial amino acid sequence of human platelet-derived growth factor (PDGF) and that predicted for p28sis, the putative transforming protein of the simian sarcoma virus (SSV), suggests expression of a growth factor activity may be central for transformation by SSV. It is now reported that SSV-transformed cells but not control cells contain a growth factor activity that is identical to PDGF in immunoassay, in mitogenic dose response, and in specific mitogenic activity. The protein immunoprecipitated by antiserum to human PDGF has an apparent molecular weight of 20,000, identical to that of p20sis, the putative intracellular degradation product of p28sis. The results support the concept that expression of a PDGF-like molecule, which appears to be the product of the viral-sis gene, is responsible for the abnormal regulation of growth is SSV-transformed cells.  相似文献
