Saskatchewan, in the summer of 1962, was the scene of an extensive outbreak of western equine encephalomyelitis (WEE) in horses. The results of mosquito survey work showed Culiseta inornata and Culex tarsalis respectively to be the two most abundant mosquito species during midsummer. These species are those reported to be most commonly associated with outbreaks of WEE. Five hundred and sixty-four pools of mosquitoes were examined for the presence of WEE virus. Six pools, three of C. tarsalis and one each of C. inornata, Aedes flavescens and Aedes dorsalis, yielded WEE virus. Positive mosquitoes were from St. Walburg (C. inornata), Saskatoon (C. tarsalis — two, A. dorsalis — one), Outlook (C. tarsalis) and Kisbey (A. flavescens).  相似文献

Serum protein electrophoresis (SPE) is a technique that could be considered one of the most useful diagnostic aids available to the clinician. The effect of storage time and temperature on the total proteins and electrophoretic fractions (albumin, α1-, α2-, β1-, β2-, and γ-globulins) was assessed in 24 healthy horses. All samples, collected by jugular vein puncture, were centrifuged and divided into 4 aliquots. The 1st aliquot was analyzed within 3 h from collection (time 0), the 2nd was refrigerated at +4°C for 24 h, the 3rd was refrigerated at +4°C for 48 h, and the last was frozen at −20°C for 48 h. One-way repeated-measures analysis of variance (ANOVA) showed a significant effect (P < 0.05) of the different storage conditions on the concentrations of all the parameters studied and significant variations in the percentages of albumin, α1-globulins, α2-globulins, and γ-globulins. Compared with time 0 the total protein concentration increased significantly after 48 h at −20°C, the albumin percentage decreased after 48 h at −20°C, the α1-globulin percentage increased after 24 h at +4°C, the α2-globulin percentage increased after 48 h at +4°C and at −20°C, and the γ-globulin percentage increased after 48 h at −20°C. The results should help veterinary practitioners handle and store equine serum samples appropriately. Further investigations at different storage times and temperatures could be useful.  相似文献

This study evaluated the CorTemp® ingestible telemetric core body temperature sensor in dogs, to establish the relationship between rectal temperature and telemetrically measured core body temperature at rest and during exercise, and to examine the effect of sensor location in the gastrointestinal (GI) tract on measured core temperature. CorTemp® sensors were administered orally to fasted Labrador retriever dogs and radiographs were taken to document sensor location. Core and rectal temperatures were monitored throughout the day in 6 resting dogs and during a 10-minute strenuous retrieving exercise in 6 dogs. Time required for the sensor to leave the stomach (120 to 610 min) was variable. Measured core temperature was consistently higher than rectal temperature across all GI locations but temperature differences based on GI location were not significant (P = 0.5218). Resting dogs had a core temperature that was on average 0.4°C above their rectal temperature with 95% limits of agreement (LoA) between 1.2°C and −0.5°C. Core temperature in exercising dogs was on average 0.3°C higher than their concurrent rectal temperature, with LoA of +1.6°C and −1.1°C.  相似文献

Whether proper heat shock preconditioning can reduce liver injury and accelerate liver repair after acute liver injury is worth study. So mice received heat shock preconditioning at 40°C for 10 minutes (min), 20 min or 30 min and recovered at room temperature for 8 hours (h) under normal feeding conditions. Then acute liver injury was induced in the heat shock-pretreated mice and unheated control mice by intraperitoneal (i.p.) injection of carbon tetrachloride (CCl4). Hematoxylin and eosin (H&E) staining, serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels and the expression levels of heat shock protein 70 (HSP70), cytochrome P450 1A2 (CYP1A2) and proliferating cell nuclear antigen (PCNA) were detected in the unheated control mice and heat shock-pretreated mice after CCl4 administration. Our results showed that heat shock preconditioning at 40°C for 20 min remarkably improved the mice’s survival rate (P<0.05), lowered the levels of serum AST and ALT (P<0.05), induced HSP70 (P<0.01), CYP1A2 (P<0.01) and PCNA (P<0.05) expression, effectively reduced liver injury (P<0.05) and accelerated the liver repair (P<0.05) compared with heat shock preconditioning at 40°C for 10 min or 30 min in the mice after acute liver injury induced by CCl4 when compared with the control mice. Our results may be helpful in further investigation of heat shock pretreatment as a potential clinical approach to target liver injury  相似文献

The newborn of some smaller animals rely upon heat produced by nonshivering thermogenesis in the brown fat to prevent a fall in body temperature after birth. Because of their pharmacological properties, some drugs may affect nonshivering thermogenesis. Therefore, in this study, the ability of newborn rabbits delivered under Innovar-Vet, ketamine hydrochloride, methoxyflurane and epidural anesthesia to maintain the rectal, subcutaneous interscapular and lumbar temperature was investigated at an ambient temperature of 35°C or 22°C and the results compared with control newborns delivered without anesthesia. When the newborns were exposed to 35°C, the anesthetics studied had no effect on the ability of the newborn to maintain the rectal temperature and the subcutaneous temperature over the interscapular fat pad was similar to the lumbar subcutaneous temperature thereby indicating that nonshivering thermogenesis was not activated. However, at an ambient temperature of 22°C Innovar-Vet or methoxyflurane reduced the temperature difference between interscapular and lumbar temperatures to 1.6°C compared to 2.5°C in controls and the difference between core temperature and ambient temperature to 3.5°C greater compared to 7.5°C in controls. Ketamine hydrochloride or lidocaine hydrochloride plus meperidine has less effect because these compounds lack adrenergic blocking properties. These data suggest that newborns delivered under anesthetics or tranquillizers that have adrenergic blocking properties require a warm (35°C) environment to prevent a fall in core temperature.  相似文献

An observational study was conducted from July to October 2004 to determine the association between in-transit losses of swine and internal trailer temperature after controlling for loading density, trip distance, herd size, and random trip effect. A convenience sample of 3 trucking companies was used to collect temperature, relative humidity, and global positioning data for 104 trips that delivered 21 834 pigs from 371 producers to Ontario abattoirs. The association between in-transit loss and trailer temperature was determined using the 90th percentiles of internal temperature for each trip. Average loading density was 0.36 m2/100 kg pig (range 0.28 to 0.50 m2/100 kg pig). Average in-transit loss was 0.12%; however, 94% of producers experienced no losses. As the 90th percentile of internal trailer temperature increased from a range of 8.6°C to 23.3°C to a range of 23.4°C to 26.1°C, average in-transit loss ratio increased approximately 3-fold, with an additional 2-fold increase as the range increased from 26.2°C to 28.9°C to 29.0°C to 30.5°C. As the 90th percentile of temperature increased by 1°C over the full range of temperatures in this study, in-transit loss was expected to increase 1.26 times. The in-transit loss was expected to decrease 0.81 times for each 50-km increase in distance traveled between the farm and the abattoir.  相似文献

The main purpose of the present study was to see if a kinetic model and a computer program for characterizing the rate of uptake and clearance of chemicals in aquatic organisms (Biofac, OECD’s Chemicals Testing Programme) was applicable to pharmacokinetic studies in fish. The program seems suitable for such studies.The effect of temperature upon rate of absorption of sulphadimidine from medicated water and elimination of the same drug was investigated in rainbow trout. A rise in the temperature from 7° C to 14° C more than doubled the calculated rate constants of absorption and elimination. The biological half life (t½ β) and the time to reach 90 % of steady state (t90% ss) at 14° C were approximately half the values at 7° C.  相似文献

Thermal stress can result in productivity losses, morbidity, and mortality if proper management practices are not employed. A basic understanding of the relationship between animals and the thermal environment is crucial to assess the environment’s impact on livestock performance. Therefore, the study objective was to evaluate whether different early life thermal stressors (ELTS) altered the temperature preference of pigs later in life. Twelve sows and their litters were randomly exposed to 1 of 3 ELTS treatments from 7 to 9 d of age: early life heat stress (ELHS; cycling 32 to 38 °C; n = 4), early life cold stress (ELCS; 25.4±1.1 °C without heating lamp; n = 4), or early life thermoneutral (ELTN; 25.4±1.1 °C with a heating lamp; n = 4) conditions. From 10 to 20 d, (weaning) all piglets were exposed to ELTN conditions. At weaning, pigs were randomly assigned to groups of 4 of the same sex and ELTS treatment. Temperature preference, where pigs freely choose a temperature, was assessed in 21 groups (n = 7 groups per ELTS treatment) using 1 of 3 thermal gradient apparatuses (22 to 40 °C). Testing began at 26 ± 1.3 d of age to give pigs time to acclimate to solid food after weaning and 1 group per ELTS treatment were tested simultaneously in each apparatus. Pigs were given 24 h to acclimate followed by a 24-h testing period. Behavior (active and inactive), posture (upright, sternal, and lateral lying), and location were documented every 20 min using instantaneous scan samples. Preferred feeding temperature was determined by the latency to empty a feeder in each location. Data were analyzed using PROC MIXED in SAS 9.4. A cubic regression model was used to calculate the peak temperature preference of pigs based on the temperature pigs spent most of their time. The preference range was calculated using peak temperature preference ±SE for each ELTS treatment group. Early life thermal stress altered where pigs spent most of their time within the thermal gradient (P = 0.03) with ELTN pigs preferring cooler temperatures (peak preference of 23.8 °C) compared with their ELCS exposed counterparts (peak preference of 26.0 °C; P < 0.01). However, ELHS exposed pigs (peak preference of 25.6 °C) did not differ in their temperature preference compared with ELTN or ELCS exposed counterparts (P > 0.05). In summary, ELCS exposure altered pig temperature preference later in life indicating that ELTS can alter temperature preference in pigs.  相似文献

Hot-iron branding uses thermal injury to permanently identify cattle causing painful tissue damage. The primary objective was to examine the physiological and behavioral effects of oral meloxicam (MEL), compared to a control, administered at the time of hot-iron branding in Angus and Hereford steers and heifers. The secondary objectives were to investigate breed and sex effects on pain biomarkers. A total of 70 yearlings, consisting of 35 heifers and 35 steers (Angus, Hereford, or Angus × Hereford), were enrolled in the study. Animals were blocked by sex, randomized across weight, and assigned to receive MEL (1 mg/kg) or a placebo (CON). Biomarkers were assessed for 48 h after branding and included infrared thermography (IRT), mechanical nociceptive threshold (MNT), accelerometry and a visual analog scale (VAS), and serum cortisol and prostaglandin E2 metabolites (PGEM). Wound healing was assessed for 12 wk. Hair samples to quantify cortisol levels were taken prior to and 30 d post-branding. Responses were analyzed using repeated measures with calf nested in treatment as a random effect, and treatment, time, treatment by time interaction, breed, and sex as fixed effects. There was a treatment by time interaction for PGEM (P < 0.01) with MEL having lower values than CON at 6, 24, and 48 h (MEL: 18.34 ± 3.52, 19.61 ± 3.48, and 22.24 ± 3.48 pg/mL, respectively; CON: 32.57 ± 3.58, 37.00 ± 3.52, and 33.07 ± 3.48 pg/mL; P < 0.01). MEL showed less of a difference in maximum IRT values between the branded (2.27 ± 0.29 °C) and control site (3.15 ± 0.29 °C; P < 0.01). MEL took fewer lying bouts at 0–12 h (4.91 bouts ± 0.56) compared with CON (6.87 bouts ± 0.55; P < 0.01). Compared with Hereford calves, Angus calves exhibited greater serum but lower hair cortisol, greater PGEM, more lying bouts, and less healed wound scores at 3, 4, and 5 wk. Compared with heifers, steers exhibited lower PGEM, lower branding site and ocular IRT, higher MNT, and lower plasma meloxicam levels. Steers spent more time lying, took more lying bouts and had greater VAS pain, and more healed wound scores at 5 wk than heifers. Meloxicam administration at branding reduced branding and control site temperature differences and reduced lying bouts for the first 12 h. Breed and sex effects were observed across many biomarkers. Changes from baseline values for IRT, MNT, lying time, step count, VAS pain, and wound scoring all support that branding cattle is painful.  相似文献

Feed efficiency (FE) is the amount of body weight gain for a given feed intake. Improving FE through selective breeding is key for sustainable finfish aquaculture but its evaluation at individual level is technically challenging. We therefore investigated whether individual routine metabolic rate (RMR) was a predictor of individual FE in the European sea bass Dicentrarchus labrax, a major species in European mariculture. The European sea bass has three genetically distinct populations across its geographical range, namely Atlantic (AT), West Mediterranean (WM), and East Mediterranean (EM). We compared FE and RMR of fish from these three populations at 18 or 24 °C. We held 200 fish (62 AT, 66 WM, and 72 EM) in individual aquaria and fed them from ad libitum down to fasting. FI was assessed for an ad libitum feeding rate and for a fixed restricted ration (1% of metabolic body weight·day−1, with metabolic body weight = body weight0.8). After being refed 12 wk in a common tank, individual RMR was measured over 36 h by intermittent flow respirometry. There was a significant effect of temperature whereby fish at 18 °C had greater mean FE (P < 0.05) and lower RMR (P < 0.001). There was also a significant effect of population, where AT fish had lower FE (P < 0.05) and greater RMR (P < 0.001) than WM and EM, at both temperatures. Despite these differences in temperature and population means, individual FE and RMR were not significantly correlated (P > 0.05). Therefore, although the results provide evidence of an association between metabolic rate and FE, RMR was not a predictor of individual FE, for reasons that require further investigation.  相似文献

Salmonella typhi murium, NVH 550 added in a concentration of 5 × 105 bacteria per ml could be detected in liquid manure and semi-solid manure, after 20 weeks and 12 weeks, respectively, at 10°C. In urine and solid manure, also held at 10°C, the test bacteria persisted for about 5 weeks. Salmonella typhi murium could not be isolated from solid manure held at 45°C for 14 days.Addition of silage effluent to final concentrations of 20, 40 and 60% reduced the survival times in mixtures with liquid manure, while the opposite effect was observed in the mixtures of silage effluent and semi-solid manure or urine.Inhibitory substances against Salmonella typhi murium or Sarcina lutea NVH 546 could not be demonstrated in any of the mixtures by the agar diffusion test.The effects of environmental factors on the survival time of Salmonella typhi murium in different types of manure are discussed.  相似文献

We investigated the influences of various reaction conditions on equine herpesvirus type 1 (EHV-1) disinfection by 5 commercial disinfectants (3 quaternary ammonium compounds [QACs] and 2 chlorine-based disinfectants) and 1 anionic surfactant. QACs at their highest recommended concentrations had no virucidal effect on EHV-1 with a 10-min reaction time at 0°C or a 1-min reaction time at room temperature. Chlorine-based disinfectants achieved EHV-1 disinfection with a 10-min reaction time at −10°C or a 30-sec reaction time at room temperature. In the presence of 5% fetal bovine serum, QACs (except for benzalkonium chloride) showed more stable virucidal effects than did chlorine-based disinfectants. The virucidal effect of the anionic surfactant was almost equivalent to that of the QACs.  相似文献

Paraoxonase-1 (PON-1) activity is a new inflammatory and oxidative marker. Technical effects and biological factors could affect the accuracy of PON-1 activity measurement. We investigated the effects of storage at different temperatures, repeated freeze–thaw cycles, interferences from hemolytic, lipemic, and icteric samples, and seasonal effects on PON-1 activity in horses. We evaluated 2 substrates with an automated spectrophotometer. Ten equine serum samples were stored under different conditions. Although storage at room (21°C) or refrigeration (4°C) temperature induced a statistically significant decrease (p < 0.05) in PON-1 activity, this is not diagnostically relevant. PON-1 activity in frozen samples (−20°C) was stable for short-term storage; diagnostically significant (p < 0.01) fluctuations were observed after 1 mo. Four repeated freeze–thaw cycles were assessed, and all cycles affected PON-1 activity (p < 0.01); however, this was diagnostically significant only after the 4th cycle. Hemolysis induced an overestimation of PON-1 activity; lipemia and hyperbilirubinemia did not change PON-1 activity. Thirty-four horses were sampled monthly for 1 y, and PON-1 activity was higher in autumn (p < 0.05) and winter (p < 0.05) than in spring and summer.  相似文献

A Saskatchewan strain of the mosquito Culex tarsalis, transmitted a local strain of western equine encephalitis virus from chick to chick, between four and 44 days after an infective blood meal. At incubation temperatures of 69 and 75°F, 120 transmissions occurred out of a possible 141, and all but seven of these were by single infected mosquitoes. At 75°F virus titers in individual mosquitoes were more uniform and transmission was more efficient, than at 69°F, although infection rates were similar at both temperatures. The minimum concentration of virus required to infect 50% of C.tarsalis was 102.5 intracerebral three-week old mouse LD50 per 0.03 ml of donor blood. These findings provide direct evidence that C. tarsalis of Saskatchewan is a highly efficient vector of western equine encephalitis virus.  相似文献

In part A of the investigation the heat resistance of isolated Diphyllobothrium plerocercoids was determined. Treatments for 5 min/at 56°C, 10 min. at 50°C, 20 min. at 48°C, and 30 min. at 47°C were found to be lethal as indicated by cessation of spontaneous movements. The number of plerocercoids in the tests was 640.In part B the consumption of pike, perch and burbot, which are known to be most important carriers of D. latum plerocercoids, and the customary methods of preparation of these fish species in Finland are first reviewed. Then the results of 85 frying and 7 boiling experiments are described, in which the temperature change at the sites of the least temperature rise was followed. During frying in the pan the inner temperature of the fish rose to the critical 56° C limit during 10 to 20 min., depending on the size of the fish. Normal boiling and frying in an oven were found to be rather safe in this respect.In all tests fish heated to the critical temperature of the plerocercoids were organoleptically found to be ready for the table.  相似文献

The complement-fixation test used at Onderstepoort was compared with the method used at A.D.R.I. on infected calf and sheep sera. In the first method, the tests are incubated at 37°C for 90 minutes and the test sera are inactivated at 53°C; whereas in the A.D.R.I. method, the test sera are inactivated at 60°C for 30 minutes, incubation is at 9°C for 18 hours, and guinea-pig complement is supplemented with 5 per cent fresh, non-inactivated, normal calf serum. Serial serum samples from one of six experimentally infected calves were negative in the Onderstepoort test, three calves gave only trace reactions and two showed maximum titres of 1:10 whereas all six had maximum serum titres of 1:10 to 1:80 in the A.D.R.I. test. A good correlation was obtained, however, between the results of the two methods with the sera of experimentally inoculated sheep although titres 3 to 8 times higher were obtained with the A.D.R.I.'s test. Post inoculation bleedings from each sheep reacted in both tests.  相似文献

Background

The duration of antacid‐induced hypergastrinemia after cessation of administration of omeprazole and famotidine apparently has not been determined in dogs.

Hypothesis

That serum gastrin will return to basal concentrations by 7 days after cessation of famotidine or omeprazole administration.

Animals

Nine healthy, adult, male, research colony dogs.

Methods

Randomized, cross‐over design. Serum gastrin was determined daily for 7 days to establish baseline concentrations. Famotidine (1.0 mg/kg q24h) or omeprazole (1.0 mg/kg q24h) was administered PO for 7 days followed by a 14‐day washout. Serum concentrations of gastrin were determined daily during 7 days of administration and daily for 7 days after cessation of administration. Each drug was evaluated in 8 of the 9 dogs.

Results

Omeprazole caused a significant increase in serum gastrin concentration (37.2 ± 7.3 to 71.3 ± 19.0 ng/L; P = .006). Famotidine induced a transient increase in serum gastrin (37.2 ± 7.3 to 65.5 ± 38.5 ng/L; P = .02) that peaked at administration day 3 and declined thereafter. By day 7 after cessation of both drugs, there was no difference in serum gastrin concentrations compared to those before administration (famotidine P = .99; omeprazole P = .99). During or after administration, gastrin concentrations above 3 times the upper reference range were rare (12 of 224 samples).

Conclusions and Clinical Importance

A 7‐day withdrawal from short‐term administration of famotidine or omeprazole is sufficient for serum gastrin to return to baseline concentrations. Withholding famotidine or omeprazole for longer before investigating pathologic causes of hypergastrinemia is unnecessary.  相似文献

An experiment was designed to determine the best practical method of thawing which could be applied to all semen processed by Canadian A.I. centers. Semen in Tris, Triladyl, whole milk, or Fresh Plus Extender packaged in 1 mL ampules or 0.5 mL French straws was used. Semen was thawed in water at 35°C, 20°C, or 5°C, or in a shirt pocket for 5, 5, 10, or 10 minutes, respectively. Post-thaw viability of sperm cells was assessed by determining the percentage of progressively motile cells, rate of progression, and percentage of intact acrosomes, at 0 hours and after 2 hours of incubation at 37°C. Post-thaw viability was significantly higher (P < 0.05) for the 35°C thaw than for any of the other thaw methods regardless of extender or packaging. Our data indicate that when AI center recommendations for thawing semen are unknown, semen in ampules and straws should be thawed in a 35°C water bath and maintained at that temperature until it can be inseminated.  相似文献

Background

Clipping the winter coat in horses is done to improve heat dissipation during exercise and make grooming easier. It is often combined with blanketing to keep the horse warm. The aims of the present study were to investigate how clipping and the use of blankets affect thermoregulation during exercise and recovery in horses.

Methods

One Gotland pony, one New Forest pony, and one warm-blooded horse exercised one after the other on a 6450 m long track. The horses walked, trotted and cantered according to a predetermined scheme, which took about 50 minutes including three stops. The scheme was repeated on five consecutive days when horses were: 1) unclipped 2) unclipped + blanket during recovery, 3) left or right side clipped, 4) clipped, and 5) clipped + riding blanket + blanket during recovery. Heart rate (HR) was measured with telemetry, respiratory rate (RR) by counting flank contractions, skin temperatures by thermistor probes, and rectal temperature with a digital thermometer. Skin wetness (SW) was estimated by ocular inspection (dripping = 5, dry = 0).

Results

Mean outdoor temperature varied from -1.1 to - 8.7°C. HR increased progressively during exercise with no difference between treatments. Maximum RR was 77 ± 30 breaths/min (unclipped) and 49 ± 27 breaths/min (clipped). The lowest skin temperature was 17.5 ± 2.7°C in a hind leg during exercise, which increased to 34.5 ± 0.1°C during recovery. Rectal temperature was elevated during recovery in unclipped, but not in clipped horses and skin temperature at base of tail was elevated during recovery except in unclipped horses without blanket. Moisture after exercise scored 3.2 ± 0.8 in unclipped and zero in clipped horses.

Discussion and conclusion

Leg skin temperature initially dropped at onset of exercise in clipped horses, and then increased after about 30 minutes due to internal heat from the working muscles. These changes were not significant when clipped horses had riding blankets, whereas unclipped horses became overheated as judged from respiratory rate and elevated rectal temperature. Providing clipped horses with blankets dampened the changes in leg skin temperature during exercise.  相似文献

In humans and other mammals, general anesthesia impairs thermoregulation, leading to warm core blood redistributing to the periphery. This redistribution is an important contributor to hypothermia that can be reduced with pre-warming before anesthesia. Additionally, sedation following premedication has been associated with hypothermia in dogs. In a prospective, randomized, cross-over study, 8 adult male and female rats (weighing 388 to 755 g) were sedated with intramuscular ketamine-midazolam-hydromorphone, then placed in an unwarmed cage or warmed box for 14 minutes, followed by 30 minutes of isoflurane anesthesia with active warming. Core body temperature was monitored throughout. After sedation, warmed rats gained 0.28°C ± 0.13°C and unwarmed rats lost 0.19°C ± 0.43°C, a significant difference between groups (P = 0.004). After anesthesia, warmed rats maintained higher core temperatures (P < 0.0001) with 2/8 and 6/8 of warmed and unwarmed rats becoming hypothermic, respectively. Pre-warming during sedation and active warming during general anesthesia is effective in minimizing hypothermia.  相似文献
