We determined the thyroxine (T4) concentration in 63 heifers on 0th (day of insemination), 6th and 21st day after first and in repeat-breeder cows after second and third insemination in relation with the breed, age and weight of animals. The examination was carried out with a feed ration balanced according to the Czechoslovak State Standard CSN 46 7070, with all-the-year-round housing, with keeping the uniform time of insemination (8.00--9.00 a. m.) as well as the time of blood sampling (10.00--12.00 a. m.). The animals were inseminated from August to November. The average T4 concentrations, with a successful insemination, fluctuated on 0th day after first, second and third insemination at levels of 56.15 +/- 13.6; 84.6 +/- 10.66 and 89.75 +/- 13.62 nmol X l-1 of serum. As the results show, the lowest T4 concentrations (P less than 0.001; P less than 0.001) on 0th day were recorded in animals becoming pregnant in first insemination. The comparison with non-pregnant animals did not show any statistically significant difference in T4 concentrations. On 0th day of the second insemination, the T4 concentrations in pregnant animals were higher (P less than 0.01) as compared with non-pregnant. On 6th and 21st day after successful first insemination the T4 content increased significantly (P less than 0.001). After second and third successful insemination we recorded in T4 concentrations a moderate decrease on 6th day (P less than 0.01 for third insemination), with insignificant increase on 21st day.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Thyroxine (T4) and cholesterol concentrations and their dynamic changes and differences were studied in 20 pregnant heifers 10 days before calving until 56 days after calving. Ten control heifers were fed the feed ration for pregnant heifers according to Czechoslovak Standard CSN 46 7070. The feed ration given to another ten (experimental) heifers was fortified with 300 mg of synthetic beta-carotene per head/day within the period of five months before insemination, during pregnancy and till the 56th day after parturition. As suggested by the regression function of straight line, no obvious differences were recorded in the dynamic changes of T4 before parturition and during the first days after parturition. T4 concentrations had a generally descending tendency in both groups, the lowest values being recorded in the 48th hour in the experimental group and the 24th hour in the control. A marked increase of T4 concentrations with the values significantly higher in the control group than in the experimental group was observed between the 21st and 35th day after calving. A medium increase of cholesterol concentrations was found in the heifers given rations fortified with beta-carotene. No significant differences in cholesterol concentrations were observed between the two groups, except in the 72nd hour and the seventh day. Significant T4: cholesterol correlations were recorded in the experimental animals in the first and second hours, on the 42nd day (P less than 0.05), and in the 72nd hour (P less than 0.001); in the control group these observations were made the seventh day before calving and in the sixth hour and on the 14th, 21st and 56th days (P less than 0.05) after calving. It is inferred from the results that beta-carotene interferes with the activity of the thyroid gland and the production of its hormones, and that the increases or decreases of the activity of this gland, caused by beta-carotene, influence the metabolism of cholesterol in the body.  相似文献

PGF2α either as a subcutaneous injection (8 or 10 mg) or intrauterine infusion (dose 5 mg) successfully synchronised oestrus in beef cattle AI programs. The beef cattle programs were managed by 1 of the following 2 systems.
  • 1 Two 10-day insemination periods, 1 for the first and the second for the repeat inseminations. Oestrus was detected and oestrus cows inseminated for the first 4 days of the first period. On the fifth day all cows not inseminated were treated with PGF2α. The treated cattle came into oestrus and were inseminated over the following 2 to 5 days.
  • 2 Two PGF2α treatments 10 days apart followed by two 4-day insemination periods. During the first 4-day period, cows exhibiting synchronised oestrus were inseminated. Cows returning to oestrus were inseminated during the second 4-day period, about 18 days after the first period.
Synchronisation of oestrus was not sharp, oestrus being distributed over 7 days, with the majority of cows being in oestrus on the third (42.3%) and fourth (22.4%) days. The route of administration of PGF2α (injection or infusion) did not effect oestrus synchronisation. PGF2α by either route of administration did not effect the fertility of cows inseminated at either the synchronised or subsequent oestrus. Pregnancy rates at the synchronised oestrus were higher when cows were inseminated after detection of oestrus than when they were inseminated without reference to oestrus at a fixed time 3 days after PGF2α treatment (mass injection). Oestrus synchronisation greatly reduced the input of time, labour and feed required during the total program. However, oestrus synchronisation did not alter the overall efficiency of artificial breeding programs measured in terms of the proportion of cattle entering a program that were inseminated and became pregnant. The main problem encountered was drug wastage through unknowingly treating cows and heifers that were not cycling.  相似文献

A system for the artificial insemination fo beef cattle was described. The system involved preparation of the cattle and facilities, visual oestrous detection twice a day, drafting each morning and insemination of oestrous cows twice a day. The AI program was evaluated during the course of the program and at pregnancy test 35 to 90 days after the end of the insemination period. The insemination period was from 25 to 60 days depending on efficiency, with programs from 25 to 35 days being favoured. Results for 12218 first inseminations (65.9%) and 2222 second inseminations (57.8%) in 56 AI programs compared favourably with results in dairy AI. Pregnancy rates in lactating cows (70.5%) were significantly higher than in nonlactating cows and heifers (66.1%). Overall efficiency of well-managed AI programs were represented by 63% of cows put into a program becoming pregnant in a 29-day insemination period. Seventy-two per cent of the cows put into the program were inseminated yielding an 88% pregnancy rate. The efficiency of visual oestrous detection was 95% but this estimate did not include false positives. Efficiency of AI programs were effected by management, nutrition, infection, inseminators, facilities and the origin and type of cattle put into a program. Environmental factors such as handling, stress and thunderstorms did not effect efficiency except via a nutritional effect.  相似文献

Conception of dairy cows was investigated in relation to changes in thyroxine (T4) and triiodothyronine (T3) concentrations in March (n = 15), June (n = 10) November (n = 7) after oestrus synchronization by cloprostenol (Oestrophan Spofa) at a dose of 0.05 mg per head. The cows were inseminated from 8.00 to 9.00 o'clock a.m. Blood was taken from 9.00 to 10.00 a.m. from v. jugularis on the day of Oestrophan treatment (-3rd day), on the day of insemination (day 0), and on the 6th and 21st day after insemination. The lowest percentage of pregnant cows (26.67%) was recorded after the March insemination, the highest (50.0%) after the June insemination. 42.86% of cows became pregnant in November. Concentrations T4 in pregnant animals on the day 0 of March insemination were 67.55 +/- 16.95 nmol.1-alpha of serum. Nonsignificant decrease to value 65.60 +/- 10.06 and 49.33 +/- 17.47 nmol.l-1 of serum were observed on the day of June and November insemination. In T3 concentrations an average decrease from the values of 2.53 +/- 0.67 nmol.l-alpha on day 0 of the March insemination to 1.48 +/- 0.67 nmol.l-alpha on day 0 of the June insemination was observed, as well as a significant decrease to 0.80 +/- 0.45 nmol.l-1 of serum (P less than 0.05) on the day of the November insemination. Considering the results we suppose that the conception of dairy cows has an indirect relationship to thyroid hormones.  相似文献

A pilot experiment with heifer heat synchronization with chlorsuperlutin-soked intravaginal tampons was conducted with 254 heifers in six herds in the North Moravian region. The rubber-foam tampons, cylindrical in shape (diameter 70 mm, height 70-90 mm), were introduced into the cranial part of the vagina, using an applicator and a vaginal speculum. Sixteen days after introduction, the tampons were removed by pulling the silon thread with which the tampons were cross-stitched. The heifers were inseminated on the second to fifth day after the removal of the tampons. The average performance in the application of the tampons was 38.5 tampons per hour, and in their removal 20.7 tampons per hour. The retention rate of the tampons was 97.6%; out of this, 79.8% of the tampons were pulled out by the silon thread. The signs of heat were good, 87.9% of the heifers could be inseminated intrauterinally. All the heifers with retained tampons were inseminated. The conception rate after the first insemination was 32.7%, after the second insemination 56.4%, and after two inseminations 68.1%. Out of the synchronized heifers, 8.1% were culled for slaughter. The drawbacks of this treatment include the low conception rate of the synchronized heifers, a high requirement for repeated insemination in the synchronized oestrus, and, consequently, a high sperm consumption.  相似文献

To examine the efficiency of retrograde sperm transport following intraperitoneal insemination, live and dead spermatozoa were used at different concentrations, and sperm recovery from cervical mucus (0.5 ml) 2, 6, 12 and 24 h following insemination was evaluated. Forty lactating Friesian cows, in their second to fourth lactation period, were used in this experiment. Thirty-six cows received intraperitoneally either live or dead spermatozoa. Each group of six cows received one of three total sperm numbers of 30, 45 and 90 million. Four cows were inseminated with 90 million spermatozoa into the uterus and served as a control group. All cows were inseminated towards the end of oestrus. After intrauterine insemination sperm recovery declined, but motile and/or immotile spermatozoa were recovered from all cows at any time. In cows inseminated intraperitoneally, sperm was recovered from the cervix at 6-24 h when 90 million were inseminated. A greater number of spermatozoa was recovered after dead rather than after live sperm inseminations. Only immotile, intact or broken spermatozoa and tail-less heads were recovered after intraperitioneal insemination using either live or dead spermatozoa. No sperm was recovered for 30 and 45 million inseminations. Our results show that, following intraperitoneal insemination, there is passive sperm transport from the peritoneal cavity to the genital tract close to the time of ovulation, and suggest a higher sperm retention in the genital tract when live as opposed to dead spermatozoa are used.  相似文献

Forty-seven heifers with the diagnosis anoestrus were treated with 15 mg to 60 mg megestrolacetat (Niagestin "NOVO") per os for 10 days. Thirty heifers showed oestrus within 10 days after treatment and 56% of these conceived after first insemination. There was no significant difference in fertility after different doses of megestrolacetat. Ten cycling heifers were synchronized with 50 mg megestrolacetat per os for 14 days. On the first day of treatment 5 mg oestradiolvalerat was given intramuscularily. All heifers came into heat and were inseminated 2--4 days after treatment. Fifty per cent were pregnant 55 days after insemination. Dosage of megestrolacetat for cows and heifers are discussed.  相似文献

The objective of this research was to determine the contribution of cow factors to the probability of successful insemination accounting for the serial number of inseminations in analysis. The investigation was performed with 101,297 insemination records in 51,525 lactations of different cows from 1368 herds obtained from the Dutch milk production recording database. Cows that had a first insemination (AI) between 40 and 150 days post-partum with one or more inseminations (≤6 inseminations) were selected. An insemination was defined successful when not followed by another insemination and when the cow calved between 267 and 295 days after insemination, or when the cow was culled between 135 and 295 days after the last insemination. Breed, parity, days in milk, lactation curve characteristics, milk production traits, moment of AI related to peak milk yield time (before or after peak milk yield), the last calf (female, male, twin or stillbirth) and season of insemination were selected as independent parameters for a model with successful rate of insemination as dependent parameter. A multivariable logistic regression model was used within cow and farm as a random effect. The probability of successful insemination was the highest in the first insemination and decreased in the following inseminations. However, the success rate of all inseminations increased in a later stage of lactation. The improvement in the successful inseminations in a later stage of lactation was better in multiparous cows than in first parity cows. Insemination in summer and before peak milk yield time reduced the success of insemination. The success rate was the lowest in 100% Holstein Friesian cows compared with other breeds and was the highest when the last calf was a female calf compared to a male calf, twin or stillbirth. In conclusion, the success of first and following inseminations depended on parity, breed, season of insemination, last calf status, daily milk yield at insemination date, serial insemination number and days in milk at insemination date.  相似文献

The effects of different treatments for oestrus synchronisation on the incidence of oestrus and fertility levels in dairy cows were studied in 2 experiments. In Experiment 1, 200 lactating cows were allotted to 5 groups and the treatments imposed were either; 1: Untreated controls, 2: An injection of 0.5 mg of cloprostenol followed 13 days later by a progesterone releasing intravaginal device (PRID) inserted for 12 days, 3: A PRID, with a capsule containing 10 mg of oestradiol benzoate (ODB) attached, inserted for 12 days, 4: A PRID inserted for 12 days with 0.5 mg of cloprostenol administered 24 h before PRID removal or, 5: As for 4 but 14 days after fixed-time insemination a second PRID was inserted for 12 days. Treated cows were inseminated 56 h after PRID removal and at an observed oestrus during the subsequent 30 days. The control group was inseminated at an observed oestrus during this 30-day period. For treatments 2, 3, 4 and 5, respectively, the percentage of cows showing oestrus by 60 h after PRID removal was 70, 40, 67 and 43 and conception rates to the fixed time insemination were 34, 33, 49 and 29%. Calving rates of cows inseminated at an observed oestrus during a 30-day period were 70, 75, 70, 83 and 82% for treatments 1, 2, 3, 4 and 5, respectively. In Experiment 2, 60 lactating cows were divided into 2 groups and the treatments imposed were either 1: An injection of 0.5 mg of cloprostenol followed 13 days later by a PRID inserted for 12 days or 2: As for 1 but 14 days after fixed-time insemination a second PRID was inserted for 12 days. Treated cows were inseminated 56 h after PRID removal and at an observed oestrus over a period from the first insemination to 6 days after removal of the second PRID. For treatments 1 and 2, respectively, 73 and 71% of cows showed oestrus by 60 h after removal of the first PRID and 40% and 46% conceived to the fixed time insemination. The conception rates to inseminations over the treatment period were 73 and 70% for treatments 1 and 2, respectively. None of the treatments resulted in conception rates which were lower than those of control cows provided that treated cows were reinseminated at observed oestrus. Treatment 4 provided the most practicable technique for oestrus synchronisation.  相似文献

OBJECTIVE: To compare the use of gonadotrophin releasing hormone (GnRH) and oestradiol benzoate (ODB) administered following a synchronised pro-oestrus on reproductive performance of lactating dairy cows and the submission rates of non-pregnant cows following resynchronisation. DESIGN: Cohort study. PROCEDURE: Lactating Holstein cows enrolled in a controlled breeding program were first treated with an intravaginal progesterone releasing insert (IVP4) for 8 days, 2.0 mg of ODB intramuscular (i.m.) at device insertion (Day 0), an analogue of PGF2alpha at device removal and either 1.0 mg of ODB i.m., 24 h after device removal (ODB group, n = 242), or 0.25 mg of a GnRH agonist (GnRH group, n = 152) injected i.m. approximately 34 h after device removal. Every cow was artificially inseminated between 49 and 56 h after removal of its insert (Day 10). Cows detected in oestrus 1 day after artificial insemination (AI) that were not detected in oestrus on the previous day were re-inseminated on that day. All cows treated on Day 0 were resynchronised for reinsemination by insertion of a used IVP4 device on Day 23. Oestradiol benzoate at a dose of 1.0 mg was administered i.m. at the time of device insertion. Inserts were removed 8 days later (Day 31) and 1.0 mg of ODB was injected i.m. 24 h later. Those cows detected in oestrus between Days 31 and 35 were artificially inseminated. On Day 46 these cows were resynchronised for a third round of AI by insertion of an IVP4 device, used previously to synchronise cows for the first and second rounds of AI, and administration of 1.0 mg of ODB i.m.. Eight days later inserts were removed. Cows detected in oestrus between Days 54 and 58 were artificially inseminated. Bulls were run with the herd between rounds of AI and removed after 21 weeks of mating. RESULTS: Treatment with ODB or GnRH at the first synchronised pro-oestrus did not significantly alter the reproductive performance over three rounds of AI or over a 21-week breeding period. Treatment also did not alter submission rates at the second round of AI or the proportion of non-pregnant and non-return cows ('phantom' cows) detected and did not result in significant differences in concentrations of progesterone in plasma 10 and 18 days after removal of inserts at the first round of AI. Treatment with GnRH reduced the proportion of cows detected in oestrus at the first round of AI (36.2 vs 97.5%; P < 0.001). CONCLUSION: Administration of GnRH compared to ODB at a synchronised pro-oestrus results in similar reproductive performance. Treatment with GnRH reduced the proportion of cows detected in oestrus following treatment. This may offer advantages to the way AI is managed by enabling insemination at a fixed-time and removing the need for the detection of oestrus.  相似文献

Maiden heifers and lactating cows of known ovarian status and of several breeds were treated with a synthetic prostaglandin, cloprostenol, or a synthetic progestagen, norgestomet, at the start of an artificial insemination (AI) program. Animals in the cloprostenol treatment received 2 injections 10 days apart. Over the next 26 days those animals that showed oestrous behaviour were inseminated. Synchronisation rates and calving rates to insemination over the first 7 days were calculated. Those in the norgestomet treatment received an implant of norgestomet plus an injection of norgestomet and oestradiol valerate. The implant was removed 10 days later and the animals were given an injection of pregnant mare serum gonadotrophin (PMSG). They were inseminated at 48 h (maiden heifers) or 56 h (lactating cows) after implant removal. Calving rates to fixed-time insemination were recorded. After completion of the AI program the animals in both treatments were joined with bulls. Overall calving rates (AI plus bulls) were calculated. By day 7 of the program, 82% of the maiden heifers and 76% of the lactating cows in the cloprostenol treatment had been detected in oestrus. By day 21 the respective figures were 99% and 81% Norgestomet treatment had an immediate and a prolonged effect on ovarian activity in those females classified as having inactive ovaries at the start of the AI program. Calving rates of those females to fixed-time AI and overall were similar to those of the females with active ovaries in both treatments. Their calving rates to fixed-time insemination, and overall calving rates for the lactating females, were significantly higher than the corresponding values of their contemporaries treated with cloprostenol and inseminated on observed oestrus over 7 days. For those females classified as having active ovaries at the start of the AI program, calving rates to first insemination and overall were similar for both treatments. Overall calving rates of lactating cows of each breed were, with one exception, higher in the norgestomet treatment than in the cloprostenol treatment. Although norgestomet treatment was more expensive than cloprostenol treatment, the advantage in calf crop resulted in an overall monetary advantage to the norgestomet treatment.  相似文献

To determine the effects of insemination season (IS; dry: May through October and rainy: November through April), lactation number (LN; heifers, primiparous or multiparous) and their interaction on non-return rate (NRR), conception rate (CR) to first-service and pregnancy loss (PL) of Jersey cattle in Turkey, over 1468 lactation records from 510 Jersey dairy cows were used. There was an interaction between LN × IS on NRR, CR to first-service (p < 0.01) and PL (p < 0.05). The NRR and CR to first-service of heifers inseminated during dry season and of primiparous inseminated during rainy season were higher (p < 0.01) than that of multiparous inseminated during both season. The NRR and CR of heifers and primiparous were higher (p < 0.05) than that of multiparous cows. The insemination of heifers during rainy season and of primiparous and multiparous during dry season increased (p < 0.01) the PL compared to the inseminations of heifers during dry season and of primiparous during rainy season. The PL was lower (p < 0.05) in primiparous than multiparous cows. The results demonstrate that the heifers have high fertility than the lactating, especially multiparous and that IS appeared to have a measurable impact on traits relating to conception of Jersey cattle in the each LN.  相似文献

An experiment was conducted to determine if concentrations of luteinising hormone or progesterone were different in pregnant or non-pregnant heifers for seven days before and 20 days after a successful or non-successful insemination. Heifers with an oestrous cycle length of 18 to 24 days only were used and they were bled at 08.00, 16.00 and 24.00 each day for seven days before and for 20 days after insemination with thawed semen (treatment 1) or semen diluent (treatment 2). Animals allocated to treatment 3 had the embryo nonsurgically flushed from the uterus at days 10 to 12 while animals allocated to treatment 4 were inseminated with semen diluent and then had a viable embryo transferred to the uterus between days 10 and 12. All animals were slaughtered between 19 and 21 days after insemination and pregnancy rate determined. There were no differences in basal luteinising hormone levels between treatments. Blood concentrations of progesterone were not different before insemination and for 16 days after insemination for pregnant (11 out of 15) and non-pregnant heifers (14) allocated to treatments 1 and 2. Between days 17 and 20, progesterone concentrations declined in non-pregnant heifers. Transfer of an embryo to non-pregnant heifers on day 10 to 12, did not affect progesterone concentrations, but non-surgical flushing of the embryo caused a decline in blood concentrations of progesterone. It was concluded that basal blood concentrations of luteinising hormone and progesterone, in samples taken three times daily were not different in pregnant or non-pregnant heifers before and for 16 days after insemination.  相似文献

The effect of two insemination policies after synchronization of oestrus on reproductive performance in two groups of cows was investigated. Oestrus was synchronized by two treatments with prostaglandin F2 alpha (PGF2 alpha) at a 14-day interval. Cows in Group 1 (n = 234) were inseminated twice on appointment 66 and 90 h after the second treatment. Cows in Group 2 (n = 222) were intensively watched for signs of oestrus after the synchronization protocol and inseminated on observed oestrus. Cows with abnormal discharge after synchronization were excluded from breeding and inseminated later during the study period. Service rate within 1 week after synchronization was higher in Group 1 than in Group 2 (89.3 versus 67.1%). Conception rates on first service did not differ between groups (33.2 versus 30.0%). Days to first service and days open were shorter in Group 1 (P < 0.05). The number of cows culled for infertility did not differ between the groups. Endometritis 14-20 days post-partum decreased the percentage of cows pregnant at the end of the study period in both groups but did not have a significant effect on conception rates and days open. It is concluded that additional inseminations required in the timed artificial insemination protocol were economically justified by the reduction in days open in comparison with insemination on observed oestrus after synchronization.  相似文献

Serum concentrations of thyroxine (T4), beta-carotene (beta K), vitamin A and conception after the first insemination on the zero, sixth and 21st day were studied in 63 heifers fed synthetic beta K supplement in groups I to IV at stanchion housing and low-carotene diet. The following amount of beta K was supplemented per head/day: I--0 mg (control, n = 17), II--100 mg (n = 16), III--200 mg (n = 14) and IV--400 mg (n = 16). The parameters were evaluated separately according to groups and conception. By comparing the T4 concentration between experimental groups and control, separately in non-gravid and gravid animals, a significant increase (P less than 0.05) was determined only on the zero day in the fourth group. At the same time, in the fourth group, in comparison with the others, insignificantly lower T4 values were recorded in non-gravid animals. beta K values on the zero day in gravid animals increased in a direct proportion to the higher supplement (P less than 0.05; P less than 0.01; P less than 0.001); in non-gravid animals the increase was slower (P less than 0.05; P less than 0.05; P less than 0.01), but differences in relation to gravidity were not significant. An increase of beta K had no significant effect on serum concentrations of vitamin A. Differences between groups as far as conception was concerned were insignificant. It follows from the results that increasing rates of beta K, with the exception of serum concentrations, had no significant effect on other parameters. Higher concentrations of T4 and a slower increase of beta K in non-gravid animals, however, point at mutual co-operation relation and its positive effect on conception in animals.  相似文献

The objective of this work was to evaluate the effect of two synchronization methods with prostaglandins F2α (PGF2α) on heifers and multiparous cows. Fourty-three Bos indicus cows (white and Red Fulani) were divided into four groups in a two-by-two factorial structure, parity x method of synchronization. The synchronization methods consisted of a two-dose regime which involved injection of animals on day 0 with PGF2α (Lutalyse) at 5 ml per cow intramuscularly. On day 11, the injection was repeated at the same dosage. On day 14 (72 h after the second injection), a fixed-time artificial insemination (AI) was done. On day 15 (96 h after the second injection), a second insemination was done. The one-and-a-half-dose regime consisted of an injection similar to the first treatment mentioned above on day 0. Thereafter, cows were observed for heat, and anyone showing heat was inseminated. A second dose was given on day 11 to all animals not having shown any heat. A fixed-time AI was done on days 14 and 15. Blood samples were collected on the day 0 of insemination for each cow while day 11 and day 21 after insemination. Progesterone was analysed by means of standard ELISA progesterone kits to determine its profiles after insemination. Results show no evidence of the effect of treatments on conception rates (P?>?0.05). Similarly, heifers and multiparous cows had similar conception rates (P?>?0.05). Between 3 weeks and 3 months of pregnancy, there was a loss of embryos of 28 % in heifers and 20 % in multiparous cows, but the difference between the two groups was not significant (P?>?0.05). It recommended that farmers do not synchronize animals with poor body condition score (BCS). They should also monitor weight gains of heifers, remove them from the herd when they have been mixed with young growing bulls and put them in a breeding herd. The two-dose regime is better to be used in areas where the inseminator cannot easily be available.

Beef cattle were treated to synchronize estrus using one of three procedures, and effects on subsequent endocrine responses and fertility were studied. Procedures were 1) feeding .5 mg.head-1.d-1 of melengestrol acetate (MGA) for 21 d (M), 2) feeding .5 mg.head-1.d-1 of melengestrol acetate for 21 d followed 14 d later by a single injection of prostaglandin F2 alpha (M + P) and 3) two injections of prostaglandin (PGF) 14 d apart (P). In Exp. 1, 94 beef cows were assigned to be artificially inseminated 12 h after detection of estrus. Procedures for synchronizing estrus did not affect the proportion of cows observed in estrus within 7 d (mean = 70.2%). However, conception rate of cows treated with MGA alone was lower (P less than .01) than that of cows treated with PGF alone (31.8 vs 78.3%). The conception rate of cows in the M + P group was intermediate (57.1%) but greater than that of cows treated with MGA alone (P less than .10). In Exp. 2, 18 heifers were observed for estrus four times daily and bled daily from 1 wk before predicted estrus until second estrus or 35 d post-treatment. Heifers treated with MGA alone maintained lower concentrations of progesterone and higher concentrations of estradiol-17 beta before first estrus than heifers treated with MGA and PGF or PGF alone (P less than .01). Conception rate following insemination was lower after long-term feeding of MGA than after two injections of PGF. Thirty nonlactating Holstein cows were superovulated to determine the effect of artificial insemination time on fertilization status and embryo quality. During the luteal phase of the estrous cycle, cows were administered 38 mg FSH-P in a 4-d descending dose regimen. Luteolysis was induced with two injections of prostaglandin on the last day of FSH-P treatment. All cows were continuously monitored for behavioral estrus using the HeatWatch estrus detection system. All cows were inseminated once with one .5-mL straw (50 x 10(6) sperm) at either 0 (n = 10), 12 (n = 10), or 24 h (n = 10) after the first standing event. The elapsed time (mean +/- SD) from the first prostaglandin dose to the first standing event was 39.4 h +/- 7.7 h. The (mean +/- SD) duration of behavioral estrus was 13.2 h +/- 4.1 h. The (mean +/- SD) number of standing events was 27 +/- 17. Five hundred twenty-nine embryos and ova were recovered nonsurgically 6 d after insemination. Fertilization rates were 29 (0 h), 60 (12 h), and 81% (24 h) (P < .01). Percentages of excellent and good, fair and poor, and degenerate embryos were not different (P > .05). Percentages of embryos with accessory sperm were 5 (0 h), 8 (12 h), and 41 (24 h) and differed between the 0 and 24 h and the 12 and 24 h inseminations (P < .01). Artificial insemination of superovulated, nonlactating Holstein cattle 24 h after onset of estrus increased fertilization rate and percentage of embryos with accessory sperm compared with insemination at 0 or 12 h after onset of estrus.
