Diabetes mellitus is a common endocrinopathy in companion animals, characterised by hyperglycaemia, glycosuria and weight loss, resulting from an absolute or relative deficiency in the pancreatic hormone insulin. There are breed differences in susceptibility to diabetes mellitus in dogs, with the Samoyed breed being overrepresented, while Boxers are relatively absent in the UK population of diabetic dogs, suggesting that genetic factors play an important role in determining susceptibility to the disease. A number of genes, linked with susceptibility to diabetes mellitus in humans, are associated with an increased risk of diabetes mellitus in dogs, some of which appear to be relatively breed-specific. Diabetes mellitus in dogs has been associated with major histocompatibility complex (MHC) class II genes (dog leucocyte antigen; DLA), with similar haplotypes and genotypes being identified in the most susceptible breeds. A region containing a variable number of tandem repeats (VNTR) and several polymorphisms have been identified in the canine insulin gene, with some alleles associated with susceptibility or resistance to diabetes mellitus in a breed-specific manner. Polymorphisms in the canine CTLA4 promoter and in other immune response genes are associated with susceptibility to diabetes mellitus in a number of pedigree breeds. Genome wide association studies are currently underway that should shed further light on the genetic factors responsible for the breed profile seen in the diabetic dog population. 相似文献

Genetic testing offers precise and accurate data to horse owners concerning their horse’s health and offers potential economic value increases of foal crops via genome-assisted selection. Despite availability of at least 96 commercially available markers, fewer than 20 are commonly used. This project seeks to identify the barriers to the incorporation of genomic tools in diverse equine operations. Based on studies examining public opinion of human medical testing, we expected that lack of genetic education and few industry requirements for testing may contribute to neutral or negative opinions regarding genomic tools for horses. We developed a 20 total question survey to capture demographic information such as facility size, type, and primary breed, as well as horse owner familiarity, use, and outlook regarding equine genetic testing. Question types included multiple choice, Likert-type scale, and open-ended response. Recruitment through county extension agents and breed organizations supplied a broad sampling of the Florida horse industry. Responses were collected from February through May 2019. Data showed that 25% of horse owners had genetically tested their horses (n = 60). Of these consumers, 80% rated the quality of their experience as “excellent” or “good” (n = 15). There was a significant positive association between genetic education and likelihood to genetically test. Most respondents (65%) were interested in receiving information regarding genetic testing. Study participants preferred to find materials on genetic testing through Web-based articles and veterinarians. Results of this study will provide researchers with critical knowledge of their audience’s needs and a roadmap to tailor future educational resources. 相似文献

Pathogenesis of osteochondrosis dissecans: How does this translate to management of the clinical case? 下载免费PDF全文
Osteochondrosis (OC) is the most prevalent developmental orthopaedic disease in the horse. It is a complex disorder because of the interplay of factors that influence the formation of lesions and the ensuing natural healing process, the end result of which determines clinical outcome. The early pathogenetic mechanism of OC has long remained elusive, but recent research has provided compelling evidence for vascular disturbances leading to areas of ischaemic necrosis in the subarticular growth cartilage as the primary event. The aetiological factors that influence the process of the development of lesions are many and comprise both genetic and environmental factors. The genetic background of OC (dissecans) is highly complex with quantitative trait loci associated with some form of OC found on not fewer than 22 of the 33 chromosomes of the horse, and different manifestations and locations of OC having probably different genetic backgrounds. The environmental factors can be divided into nutritional factors and factors influencing the biomechanical loading of joints. Feeding large quantities of easily digestible carbohydrates is a clear risk factor and also the availability of sufficient copper for mares in the last part of pregnancy is important. Factors influencing the biomechanical loading of joints include the exercise regimen of foals, paddock size and roughness of terrain, and conformation. Equine OC is rarely a life‐threatening disease and the prognosis of (surgical) treatment is good to very good. However, the impact on economics of the equine industry and on horse welfare is large. This impact can only be lessened by the judicious management of both horse breeding and horse husbandry. Some of the measures that would alleviate the burden of OC conflict with (commercial) pressures from the industry and a balance between human‐imposed breeding and performance goals and the interest of the animal itself should be sought. 相似文献

In the last decades, several endangered breeds of livestock species have been re‐established effectively. However, the successful revival of the Dutch and Danish Landrace goats involved crossing with exotic breeds and the ancestry of the current populations is therefore not clear. We have generated genotypes for 27 FAO‐recommended microsatellites of these landraces and three phenotypically similar Nordic‐type landraces and compared these breeds with central European, Mediterranean and south‐west Asian goats. We found decreasing levels of genetic diversity with increasing distance from the south‐west Asian domestication site with a south‐east‐to‐north‐west cline that is clearly steeper than the Mediterranean east‐to‐west cline. In terms of genetic diversity, the Dutch Landrace comes next to the isolated Icelandic breed, which has an extremely low diversity. The Norwegian coastal goat and the Finnish and Icelandic landraces are clearly related. It appears that by a combination of mixed origin and a population bottleneck, the Dutch and Danish Land‐races are separated from the other breeds. However, the current Dutch and Danish populations with the multicoloured and long‐horned appearance effectively substitute for the original breed, illustrating that for conservation of cultural heritage, the phenotype of a breed is more relevant than pure ancestry and the genetic diversity of the original breed. More in general, we propose that for conservation, the retention of genetic diversity of an original breed and of the visual phenotype by which the breed is recognized and defined needs to be considered separately. 相似文献

Cytosine arabinoside (ara‐C) is a component of many protocols for the treatment of acute leukaemia and non‐Hodgkin lymphomas in humans. The aim of the study was to prospectively evaluate the efficacy of ara‐C in a myeloablative regimen in a cohort of canine lymphomas with bone marrow involvement. Seventeen dogs were enrolled. Eight were treated with a VCAA‐based protocol (Group 1) and nine with the same regimen added with ara‐C (Group 2). Ara‐C was administered on a 5‐day schedule as an i.v. continuous infusion at the dose of 150 mg m?2 per day for five consecutive days. During treatment complete remission (CR) was achieved in two dogs in Group 1 and in eight dogs in Group 2. CR rate was significantly higher in Group 2 (P < 0.01). Median survival was 72.5 days (range 6–174) in Group 1 and 243 days (range 73–635) in Group 2. Survival was significantly longer in Group 2 (P < 0.001). Both protocols were well tolerated, with a low incidence of adverse events. Ara‐C added to a VCAA‐based protocol appears to be safe and beneficial in dogs with stage V lymphoma. Incorporation of the nucleoside analogue might be crucial for the development of future therapeutic strategies in dogs. 相似文献

Finite element modelling of the canine and feline outer ear canal: benefits for local drug delivery?

Current therapeutic regimes of outer ear infections in dogs and cats aim at the application of efficient local therapeutics after cleaning of the acoustic meatus. One so far insufficiently answered question is if the local application of these substances results in an individually suitable drug concentration in the external ear canal. Thus, the purpose of the present study was to develop a finite element model to calculate the values of the different areas of the external acoustic meatus in dogs and cats in order to provide a tool for the benefit of an appropriate local drug dosage determination. A 3D finite element model (FEM), based on computer tomographic (CT) data sets of four dogs and two cats, was generated to determine areas and volumes of the outer ear canal. Furthermore, various ear therapeutics and cleansers were tested concerning their optimal distribution on 5 cm2 dog and cat skin. The data shows major variations of the area values of the external auditory canal in case of the different dogs but not in the examined cats. These results suggest that manufacturer's recommendations of the pharmaceuticals might be insufficient in terms of achieving an optimal drug concentration in the outer ear canal especially in larger dogs. In conclusion, the developed finite element model has shown to be suitable to calculate areas of the outer ear canal in cats and dogs and could be of help in context with the definition of optimal drug concentrations for a local drug delivery. 相似文献

Intestinal infections with Toxocara cati and Toxocara canis in their definitive host (felids and canids, respectively) are diagnosed by egg identification in faeces using coproscopical techniques. The Toxocara species is assumed to comply with the species from which the examined faeces were obtained, i.e. T. cati in cats and T. canis in dogs. We isolated and measured Toxocara eggs from faecal samples of 36 cats and 35 dogs from Switzerland and identified the Toxocara species by PCR. Amongst the isolates originating from dogs, 24 (68.5%) were determined as T. canis and 11 (31.5%) as T. cati. In all samples originating from cats, only T. cati was identified. Based on PCR identification, eggs of T. canis (n=241) and T. cati (n=442) were measured, revealing statistically significant different (p<0.001) mean sizes of 62.3 by 72.7 μm for T. cati and 74.8 by 86.0 μm for T. canis eggs. Considering that coprophagy is not unusual for dogs, a considerable percentage of Toxocara infections coproscopically diagnosed in dogs, as well as assumptions on anthelminthic resistance in regularly treated dogs, might in fact relate to intestinal passages of eggs following the uptake of other animals' faeces. 相似文献

Mast cell counts, epidermal thickness and hair follicle density were quantified in toluidine blue stained sections of normal skin from 20 different body regions in 10 dogs and compared to the predilection sites of canine atopic dermatitis. Mast cell distribution varied significantly from site to site (P < 0.0001) and counts in the superficial dermis were significantly higher than the deeper dermis (P < 0.05). Mast cell counts were highest in the medial and lateral pinna (mean 10.4–11.3 per high power field, HPF) and in the ventral interdigital skin of the hind and fore feet (mean 9.2–9.5 per HPF). Counts in these regions were at least 150% higher than all the other sites (means ranging between 2.9 and 6.0 per HPF). Variations in mast cell counts, epidermal thickness or hair follicle density did not adequately explain the predilection sites of canine atopic dermatitis. However, the results provide some evidence that cutaneous mast cell distribution may be involved in the frequent occurrence of ear and foot pruritus in this disease. 相似文献

In injection site sarcoma (ISS) in cats lateral as well as deep margins should be correctly planned for a successful surgical outcome. The discrepancy between clinical and computed tomography (CT) measurements of dimension in resectable tumour has led to possible bias that affects the subsequent surgical dose. The aim of this study was to prospectively investigate the agreement between clinical and CT measurements of dimension in newly diagnosed ISS in cats. Fifty‐three client‐owned cats that underwent both clinical and CT measurements of the length and width of ISS were included. CT measurements showed a tendency towards being larger than clinical dimensions, and this difference increased with increasing tumour size. Based on our results, in further studies focusing on ISS in cats, the kind of assessment used to define tumour dimensions (CT versus clinic) should be declared and specified to properly consider surgical results and prognostic impact of this variable. 相似文献
