利用RAPD技术对20个姜(Zingiber officinale)品种进行了遗传多样性的研究。从200多个引物中筛选出25个有效引物,共扩增出171条DNA带,其大小分布在100~3 500 bp之间,其中111条为多态性,约占总数的64.91%,平均每个引物的扩增带为6.8条。利用25个有效引物扩增的DNA带数计算出品种间的J系数,并进行UPGMA聚类分析,在此基础上,建立了中国姜品种亲缘关系系统树。在相似性系数0.67处,20个品种分为Ⅰ、Ⅱ、Ⅲ和Ⅳ类。Ⅰ类可分为A、B和C组;Ⅳ类可分为D和E组。其中,A组可再分为A1和A2亚组;B组可再分B1和B2亚组。各亚组内相似性系数较大,表明它们之间的亲缘关系较近,但也存在着一定的差异。  相似文献

目标起始密码子多态性(start codon targeted polymorphism,SCo T)是一种新型的遗传分子标记技术。本研究采用单因素水平设计方法,优化并建立梨的适宜SCo T分子标记体系,通过SCo T标记技术,分析了安徽省砀山县的43份梨(Pyrus spp.)的遗传多样性和亲缘关系。结果表明,优化后梨SCo T-PCR反应体系:10×Easy Taq Buffer(含2 mmol/L Mg2+)2.5μL,模板DNA终浓度30 mg/L,上下游引物终浓度1.0μmol/L,d NTPs终浓度0.2 mmol/L,Taq DNA聚合酶1.0 U,总体积为25μL。随机选取2个DNA模板进行引物筛选,最终从67条引物中筛选出16条扩增条带清晰且有差异性的引物,共扩增出145条带,其中多态性条带有126条,多态性比率为86.1%,每条引物平均扩增出9.1条带。SCo T标记的43个梨材料遗传相似性系数为0.517~0.931,平均值为0.685。聚类分析表明,在遗传相似系数为0.610的水平上,43份梨材料分为A和B两组;在遗传相似系数为0.746的水平上,A组又分为6个亚组(Ⅰ~Ⅵ)。所研究的43份梨材料具有丰富的遗传多样性,且能够被SCo T分子标记有效地检验,为进一步研究利用安徽省砀山县梨种质资源提供了依据。  相似文献

基于ISSR指纹的甘薯食用品种的遗传多样性分析   总被引:2,自引:0,他引:2  
运用ISSR分子指纹技术检测甘薯基因组DNA的多态性,是鉴定甘薯遗传多样性和亲缘关系有效的方法之一。本研究选用6个ISSR标记对17份甘薯品种进行了DNA指纹扩增,共扩增出55条谱带,其中多态性谱带50条,多态性水平为90.91%;UBC825引物扩增出带型最多,有16种;根据Nei’s遗传距离计算获得的聚类树状图表明,在GS为0.625时,参试的17份甘薯品种分为4个组群;遗传相似性分析表明,顶芽菜用品种间遗传相似系数最高0.877,叶柄食用品种和烤薯型品种之内的遗传相似系数分别为0.778和0.740。结果表明甘薯主要食用间具有较好的遗传多样性,但同类型品种间却又有较高的遗传相似性,这类品种需要引进或创制新资源。  相似文献

利用ISSR标记技术分析了28份香蕉种质的遗传多态性。从100个ISSR引物中筛选出8个多态性引物,共扩增出55条DNA带,其中46条为多态性带,占83.6%,平均每个引物扩增的DNA带数为6.88条。依相似系数0.73的水平,将香蕉28个品种划分为6大类。其中云南BB(BB)和东莞高把大蕉(ABB)在相似系数为0.94时,二者的亲缘关系较近。Pisang Ceylan(AAB)和FHIA-18(AAAB)相似系数水平接近为1,表明二者亲缘关系最近。本研究为香蕉遗传关系的建立及品种鉴定提供理论基础。  相似文献

用随机引物对河南省角菊头蝠(Rhinolophus cornutus)、不同产地的中菊头蝠(Rhinolophus affinis)和马铁菊头蝠(Rhinolophus ferrumequinum)进行DNA多态性研究,以探讨它们之间的亲缘关系。从20个随机引物中优化出15个引物对基因组DNA进行扩增,10个标本共扩增出184条DNA谱带,平均每个引物扩增出12条谱带,其中多态性谱带173条,多态率为94%。聚类结果表明:对同种蝙蝠,同一地理区域的个体之间分化较小,不同地理区域的个体之间分化较大;中菊头蝠与角菊头蝠之间的亲缘关系较近,而与马铁菊头蝠之间的亲缘关系较远。  相似文献

利用SSR和RAPD对来自福建、黑龙江、河北和内蒙古4个地理群体的80个马铃薯晚疫病菌(phytophthora infestans)株进行了遗传多样性分析。13对SSR引物共扩增出76条谱带,多态性条带比率78.9%,相似系数变化范围0.00~0.42之间;筛选出的14条RAPD引物共扩增出189条谱带,多态性条带比率95.2%,相似系数变化范围0.04~0.66之间。遗传多样性分析表明,在4个群体中,福建群体的多样性更为丰富。遗传相似性分析显示,黑龙江和内蒙古两个群体间的遗传相似性最高,而福建和河北两个群体间的遗传相似性最低。聚类分析显示,来自南方福建的菌株与来自北方黑龙江、河北和内蒙古的菌株亲缘关系较远,且福建群体分布于更多的聚类组,显示出更高的遗传变异度。  相似文献

本实验对影响SRAP-PCR体系中的Mg2+、dNTPs和引物浓度等因子进行了体系优化,建立了一套适合白簕SRAP检测的25μL反应体系:1.5mmol/LMg2+,1.5mmol/LdNTPs,1μmol/L引物,10ng模板DNA,1.5UTaqDNA聚合酶;进一步以白梗簕菜为模板,利用优化的体系进行多态性标记引物组合分析,共筛选出17对引物;利用这些引物对7个白簕品种进行SRAP遗传多样性分析,共扩增出461条谱带。其中多态性谱带155条,多态性谱带比率为33.6%,白簕品种间的相似系数为0.7077~0.9474。经UPGMA聚类分析结果显示,所检测的7个白簕品种分为两大类,其中亲缘关系较近的青梗簕菜和细叶密刺簕菜聚成了一组;而其余的4个种,包括白梗簕菜、细叶小刺簕菜、紫柄簕菜、大叶大刺簕菜和红梗簕菜,聚成另一组。  相似文献

为了解广西地方葛根种质资源之间的亲缘关系,本研究以SCoT分子标记对44份广西葛根种质资源的基因组DNA进行PCR扩增,并进行遗传多样性分析。结果表明,25条SCoT引物共扩增出223个条带,其中具有多态性的条带有194条,平均每条引物获得7.76个多态性条带,多态性比例为86.99%。聚类分析表明,44份葛根种质资源的遗传相似系数在0.587~0.982之间。在系数0.65处,44份葛根分为两大类,37号材料单独成为一类;在系数为0.74处,可聚为六类。综上所述,SCoT分子标记适用于葛根种质资源遗传多样性分析,本研究结果为广西葛根种质资源鉴定评价和新品种选育奠定了一定的理论基础。  相似文献

离子束介导的水稻早熟变异株系AFLP分析   总被引:1,自引:0,他引:1  
用AFLP分子标记技术对N+离子束介导玉米总DNA获得的2个稳定遗传至第6代的水稻早熟变异株系C(08-5-121-6-1)和D(08-5-121-6-2)进行分析。首先从64对引物中筛选出10对多态性较好的引物,然后用这10对引物对早熟变异株系C和D分别扩增。结果显示,变异株系C、D和阴性对照水稻B(豫粳6号)与阳性对照玉米A(郑单14)之间的AFLP扩增图谱相似率分别为15.7%、16.2%和11.6%,说明变异株系与对照豫粳6号的AFLP扩增图谱存在显著差异。变异株系C和D与阴性对照豫粳6号相比,分别扩增出50条和58条差异带,其中新带分别为25条和35条,缺失带分别为19条和15条,变异株系C和D分别扩增到与阳性对照玉米相一致的目的带6条和8条,说明变异株系C和D基因组DNA与玉米基因组DNA相似性高于对照水稻。  相似文献

利用SSR标记分析藜麦品种的遗传多样性   总被引:1,自引:0,他引:1  
为了解藜麦种质资源的多样性,本研究利用SSR引物对所搜集的41个藜麦种质的多态性及其亲缘关系进行了分析。结果表明,从54对SSR引物中筛选出了16对能明显扩增出稳定的多态性条带的引物,共检测出139个等位基因条带,每一对引物的等位基因个数为3~13,平均为8.7;16对引物的多态信息含量(PIC)变幅为0.208~0.432,平均为0.366。UPGMA聚类分析显示,41份材料的遗传相似系数(GS)在0.374~0.906之间,平均相似系数为0.626。在阀值(GS)约为0.665时,41份材料可分为4大类。其中614929与B.B.Quinoa浙Ⅰ间的遗传相似系数最小,为0.374,表明来源于不同地区的遗传距离较远,遗传基础较广泛。藜麦品种资源间的亲缘关系的揭示为藜麦资源保存和新品种选育提供了理论依据。  相似文献

【目的】以富士(Fuji)、 秦冠(Qinguan)嫁接在平邑甜茶(Malus hupehensis Rehd.)上的当年生盆栽苗为试验材料,采用砂培方法,研究了缺氮胁迫和干旱对富士和秦冠生长情况、 光合参数、 植株各部位氮磷钾含量及氮素利用效率的影响,分析比较了低氮干旱条件下富士和秦冠生长及氮素利用的差异,以期为果树生产高效肥水利用提供理论指导。【方法】试验共设四个处理: 正常氮正常水(ZZ)、 低氮正常水(DZ)、 正常氮干旱(ZG)、 低氮干旱(DG)。氮素和水分均设置两个水平,分别为正常氮(6 mmol/L NO-3-N)、 低氮(0.3 mmol/LNO-3-N)、 正常供水(保持盆中砂子相对含水量为饱和含水量的80%~85%)、 干旱处理(保持盆中砂子相对含水量为饱和含水量的60%~65%)。【结果】富士和秦冠的生物量(茎和叶)、 株高茎粗等生长指标以及光合速率、 气孔导度、 蒸腾速率均为正常氮正常水(ZZ)>低氮正常水(DZ)>正常氮干旱(ZG)>低氮干旱(DG),并且相对应处理下秦冠的以上指标均高于富士;正常供水下,缺氮处理使富士、 秦冠的根冠比比正常氮处理均有所增加,富士提高了2.05%,秦冠提高了22.40%。富士和秦冠的氮、 磷、 钾含量均表现出正常氮正常水(ZZ)>低氮正常水(DZ)>正常氮干旱(ZG)>低氮干旱(DG); 氮、 钾元素含量在植株各部位的分布顺序依次是叶>根>茎,磷元素则是根>叶>茎;光合氮素利用效率(PNUE)和氮素利用效率表现为秦冠处理之间差异极显著,富士处理之间差异不显著;秦冠的PNUE和NUE明显高于富士,在低氮正常水(DZ)处理下,秦冠氮肥利用率比富士高42.07%,在低氮干旱(DG)处理下高64.14%;低氮胁迫下富士和秦冠的NUE显著提高,并且秦冠提高的幅度高于富士。【结论】施用氮肥能够显著提高富士与秦冠的干物质量,同等水肥条件下,秦冠生长优于富士;水分亏缺会减少叶片对氮的吸收,干旱条件下适度增施氮肥,可提高果树的抗旱能力;低氮干旱胁迫下秦冠的生长指标、 光合指标及氮素利用效率指标均优于富士,表现出较强的抗低氮干旱胁迫的能力。  相似文献

Laser-induced breakdown spectroscopy (LIBS) is a new technique for the analysis of plant material. This study investigates the application of LIBS to pasture-based plant samples. The LIBS measurements were obtained from pelletized pasture samples (100 samples) that had been also analyzed by inductively coupled plasma–optical emission spectroscopy (ICP-OES) following microwave digestion for calibration and comparison purposes. Comparisons for elements sodium (Na), potassium (K), magnesium (Mg), calcium (Ca), manganese (Mn), iron (Fe), copper (Cu), zinc (Zn), boron (B), phosphorus (P), and sulfur (S) showed that LIBS could be used for almost all the standard profile total elements with concentrations down to low mg/kg levels (observed error of Na: 0.024 percent, K: 0.18 percent, Mg: 0.016 percent, Ca: 0.073 percent, P: 0.017 percent, Mn: 31 mg/kg, Fe: 150 mg/kg, Zn: 6.6 mg/kg, and B: 1.1 mg/kg). Elemental analysis at less than mg/kg levels was not possible using LIBS. The elements S and Cu were particularly difficult to analyze with reliability using LIBS at the concentration levels found in the plant samples. Replacing microwave digestion and subsequent ICP analysis with a direct analysis of dried plant samples using LIBS has the potential to improve the productivity and reduce the cost of testing.  相似文献

Staff members of the Department of Botany of Palacký University in Olomouc and Gene Bank Department – Workplace Olomouc, Research Institute of Crop Production in Prague, Czech Republic, conducted an expedition in seven European countries (Austria, Czech Republic, France, Germany, Italy, the Netherlands, Switzerland) in August/September 1999 to collect wild Lactuca spp. germplasm and study its geographic distribution, ecology and biodiversity. During the mission, more than 600 locations were visited resulting in the collection of 602 seed samples (accessions) of wild Lactuca species and 13 seed samples of related genera (Chondrilla and Mycelis). Lactuca serriola f. serriola, L. serriola f. integrifolia, L. saligna and L. viminea subsp. chondrilliflora were prevalent in southern Europe (Italy, France), however, only L. serriola was common in central and western Europe (Austria, Czech Republic, Germany, Netherlands, Switzerland). The greatest diversity of Lactuca species was found in France, where also the most seed samples (165) were collected. The most characteristic habitats with a high density of Lactuca spp. populations were observed along roads and highways, grassy ditches, ruderal communities, and dust-heaps. Natural infections by powdery mildew (Erysiphe cichoracearum) and downy mildew (Bremia lactucae) on some wild Lactuca spp. were observed. Recent observations concerning the geographic distribution, population structure, habitats, and natural occurrence of diseases of Lactuca spp. are discussed. This assemblage of genetic resources of Lactuca spp. can serve as the basis of future studies of species diversification, spatial population structure, plant microevolution, domestication processes, and genetic variability of host-parasite interactions.  相似文献

The responses of tallgrass prairie plant communities and ecosystem processes to fire and grazing are well characterized. However, responses of invertebrate consumer groups, and particularly soil-dwelling organisms, to these disturbances are not well known. At Konza Prairie Biological Station, we sampled soil macroinvertebrates in 1994 and 1999 as part of a long-term experiment designed to examine the effects and interactions of annual fire, mowing, and fertilization (N and P) on prairie soil communities and processes. For nearly all taxa, in both years, responses were characterized by significant treatment interactions, but some general patterns were evident. Introduced European earthworms (Aporrectodea spp. and Octolasion spp.) were most abundant in plots where fire was excluded, and the proportion of the total earthworm community consisting of introduced earthworms was greater in unburned, unmowed, and fertilized plots. Nymphs of two Cicada genera were collected (Cicadetta spp. and Tibicen spp.). Cicadetta nymphs were more abundant in burned plots, but mowing reduced their abundance. Tibicen nymphs were collected almost exclusively from unburned plots. Treatment effects on herbivorous beetle larvae (Scarabaeidae, Elateridae, and Curculionidae) were variable, but nutrient additions (N or P) usually resulted in greater densities, whereas mowing usually resulted in lower densities. Our results suggest that departures from historical disturbance regimes (i.e. frequent fire and grazing) may render soils more susceptible to increased numbers of European earthworms, and that interactions between fire, aboveground biomass removal, and vegetation responses affect the structure and composition of invertebrate communities in tallgrass prairie soils.  相似文献

We have studied the effect of anaerobically digested (ADMSW) and composted municipal solid waste (CMSW) on mineralization and foodweb dynamics to verify the hypothesis that ADMSW would immobilize N right after addition to soil in contrast to addition with CMSW. Another hypothesis was that the mesofauna (enchytraeids and microarthropods) would stimulate N release from the decomposer community. We measured excretion of -N and urea-N from the mesofauna and hypothesized that enchytraeids would release urea. ADMSW and CMSW were amended to pots with sandy loam and barley. The pots were divided into treatments with or without mesofauna. Mesofauna, plant N and biomass, soil N and ergosterol (fungal biomass) were measured over a 113-day period of four equidistant samplings. Soil respiration, N mineralization and N release by the mesofauna were modelled from concurrent studies. ADMSW- and CMSW-treated soils initially (<20 days) immobilized N. The amendments did not increase plant growth substantially, and this was probably due to N-limitation in the early stages of plant growth. Enchytraeid abundance was about three times higher in ADMSW- than CMSW-treated soils, indicating that ADMSW contained more labile compounds, bacteria, and microfauna. The mesofauna did not affect N-content, but the cumulated -N excreted by the mesofauna was estimated to be substantial and about one-fifth of total plant N in ADMSW. An explanation for this discrepancy might be that in the absence of the mesofauna, other members of the detrivore and microbivore community performed the mesofauna’s function. Neither enchytraeids nor microarthropods excreted urea.  相似文献

Abstract

A 3-year study was carried out to investigate quality parameters in 14 tree fruit and berry species grown in southern Norway. The species were blueberry, apple, aronia, sour cherry, sweet cherry, red raspberry, strawberry, blackcurrant, gooseberry, red currant and elderberry, harvested along with wild bilberry, cloudberry and lingonberry. Significant differences between species were identified for all quality parameters. The coefficient of variation between species was lowest for pH (12.5%), dry matter (18.9%) and soluble solids (25.3%), followed by titratable acids (59.3%), total phenolics (83.8%), antioxidant capacity FRAP (85.7%) and antiradical power by the DPPH-assay (97.8%), total monomeric anthocyanins (132%) and ascorbic acid (137%). Average coefficient of variation within species were lower and ranged from 4 (pH) to 62% (ascorbic acid). Only the FRAP values were significantly affected by harvesting year with lower levels in 2004 than in 2005 and 2006. There were significant interactions between species and harvesting year for dry matter, soluble solids, pH, ascorbic acid and FRAP. The results indicate generic ranges in composition within species independent upon growing location and climate, and the composition of the tree fruits and berries is not likely to deviate from these ranges. It is concluded that desirable composition of tree fruits and berries and their products should primarily be achieved by selection among species rather than searching fors broadened variation within individual species.  相似文献

Potassium (K) fixation and release in soil are important factors in the long-term sustainability of a cropping system. Changes in K concentration and characteristics of K fixation and release in rhizosphere and nonrhizosphere soils in the rapeseed (Brassica napus L.)–rice (Oryza sativa L.) rotation were investigated using a rhizobox system. The concentrations of different forms of K in both rhizosphere and nonrhizosphere soils decreased with plants compared to without plants, regardless of K fertilizer application. Potassium uptake by crops mainly came from the rhizosphere soil. In the treatment without K fertilizer (–K), the main form of K supplied by the soil to the crops was 1.0 mol L?1 nitric acid (HNO3) nonextractable K, followed by nonexchangeable K, and then exchangeable K. In the treatment with K fertilizer (+K), the main K forms supplied by the soil to the crops were exchangeable K and nonexchangeable K. The amount and rate of K fixation after one cycle of the rapeseed–rice rotation was greater in rhizosphere soil than in nonrhizosphere soil. The amount and rate of K fixation of soil in the +K treatment were significantly less than in the –K treatment. The cumulative amounts of K released with 1.0 mol L?1 ammonium acetate (NH4OAc) and 1.0 mol L?1 HNO3 extraction increased with the increasing numbers of extractions, but the K-releasing power of soil by successive extraction decreased gradually and finally became almost constant. The release of K was less in rhizosphere soil than in nonrhizosphere soil. The release of K in the +K treatment was similar to that in the –K treatment in rhizosphere soil, but the K release in nonrhizosphere soil was greater with the +K than the –K treatment. Overall, the information obtained in this study will be helpful in formulating more precise K fertilizer recommendations for certain soils.  相似文献
