Eighteen steers were used to evaluate the effect of supplemental corn oil level to steers grazing endophyte-free tall fescue on fatty acid composition of LM, stearoyl CoA desaturase (SCD) activity and expression as well as cellularity in s.c. adipose. Corn oil was supplemented (g/kg of BW) at 0 (none), 0.75 (medium), and 1.5 (high). Cottonseed hulls were used as a carrier for the corn oil and were supplemented according to pasture availability (0.7 to 1% of BW). Steers were finished on a rotationally grazed, tall fescue pasture for 116 d. Fatty acid composition of LM, s.c. adipose, and diet was determined by GLC. Total linoleic acid intake increased linearly (P < 0.01) with corn oil supplementation (90.7, 265.1, and 406.7 g in none, medium, and high, respectively). Oil supplementation linearly reduced (P < 0.05) myristic, palmitic, and linolenic acid percentage in LM and s.c. adipose. Vaccenic acid (C18:1 t11; VA) percentage was 46 and 32% greater (linear, P = 0.02; quadratic, P = 0.01) for medium and high, respectively, than none, regardless of tissue. Effect of oil supplementation on CLA cis-9, trans-11 was affected by type of adipose tissue (P < 0.01). In the LM, CLA cis-9, trans-11 isomer was 25% greater for medium than for none and intermediate for high, whereas CLA cis-9, trans-11 CLA isomer was 48 and 33% greater in s.c. adipose tissue for medium and high than for none, respectively. Corn oil linearly increased (P 0.05) the percentage of total SFA, MUFA, or PUFA but linearly increased (P = 0.03) n-6:n-3 ratio from 2.4 to 2.9 in none and high, respectively. Among tissues, total SFA and MUFA were greater in s.c. adipose than LM, whereas total PUFA, n-6, and n-3 fatty acids and the n-6:n-3 ratio were lower. Trans-10 octadecenoic acid, VA, and CLA trans-10, cis-12 were greater (P < 0.01) in s.c. adipose than in LM. Oil supplementation did not alter (P > 0.05) stearoyl CoA desaturase activity or mRNA expression. Corn oil supplementation to grazing steers reduced the percentages of highly atherogenic fatty acids (myristic and palmitic acids) and increased the percentages of antiatherogenic and anticarcinogenic fatty acids (VA and cis-9, trans-11 CLA).  相似文献

Feeding sheep concentrate-based diets increases the oleic acid content of their tissues, whereas the cis-9, trans-11 conjugated linoleic acid (CLA) content is increased by feeding forage diets. Both these metabolic transformations could be attributable to increased activity of stearoyl-CoA desaturase (SCD). Therefore, the effect of forage or concentrate feeding regimens on the fatty acid composition of sheep tissues were investigated to determine whether any changes are related to an alteration of SCD mRNA levels. Twenty-four ewe lambs were randomly allotted to one of three dietary treatment groups: 1) dehydrated grass pellets, 2) concentrate diet fed to achieve a growth rate similar to that of the dehydrated grass pellets, and 3) the same concentrate diet approaching ad libitum intake. As expected, animals fed ad libitum concentrates grew at a greater (P = 0.001) rate (280 g/d) than those fed either of the other two diets (180 g/d), which were similar. In samples of liver and the three adipose tissue depots studied, the concentration of oleic acid from sheep fed either level of the concentrate diet was greater (P < 0.001) than from animals fed forage. This was associated with an increase (P < 0.05) in the ratio of SCD to acetyl-CoA carboxylase mRNA in adipose tissue and liver. Compared with concentrate-fed, the forage-fed lambs had increased (P < 0.05) levels of the cis-9, trans-11 isomer of CLA and C18:1, trans-11 in all their tissues, although the levels of SCD mRNA were lower. It therefore seems that the increased oleic acid content of sheep tissues in response to concentrate-rich diets is associated with an increase in SCD gene expression. By contrast, the increased concentration of CLA in animals fed forage-based diets is associated with an increase in substrate (C18:1 trans-11) availability.  相似文献

Thirty-six Angus x Hereford heifers (365 +/- 60 kg) were used to determine the effects of supplemental dietary lipid sources on fatty acid composition of i.m., perianal (p.a.), and s.c. lipid depots. Lipid was supplied to diets as either corn oil or a rumen-protected conjugated linoleic acid (CLA) salt for two specific treatment periods of either the final 32 or 60 d on feed. Following an initial 56-d feeding period, heifers were fed one of three dietary treatments (DM basis): 1) basal diet containing 88% concentrate and 12% grass hay (CON), 2) basal diet plus 4% corn oil (OIL), or 3) basal diet plus 2% rumen-protected CLA salt (RPCLA) containing 31% CLA. The trans-10, cis-12 CLA concentration was greatest (P < 0.05) for heifers fed RPCLA and OIL diets and least (P < 0.05) for CON, regardless of time on dietary treatment. Heifers fed supplemental RPCLA had greater (P < 0.05) total CLA content than either CON- or OIL-fed heifers. Adipose tissue concentration of trans-11 vaccenic acid (TVA) was less (P < 0.05) for CON than OIL or RPCLA, which did not differ (P > 0.05). Percentages of C18:1 trans-10 were least (P < 0.05) in i.m. lipid compared with p.a. and s.c., which did not differ (P > 0.05). Following 60 d of lipid supplementation, heifers fed OIL and RPCLA had lower (P < 0.05) concentrations of oleic acid and total monounsaturated fatty acids (MUFA) compared with CON. The ratio of cis-9, trans-11 CLA:TVA was higher (P < 0.05) for heifers fed 60 vs. 32 d, but did not differ (P > 0.05) between adipose depots. Feeding OIL increased (P < 0.05) adipose concentration of C18:2 fatty acid, whereas feeding RPCLA increased (P < 0.05) total CLA isomers by 22%. Intramuscular lipid contained the lowest (P < 0.05) percentage of cis-9, trans-11 CLA, total CLA, C18:1 cis-9, C18:1 trans-10, and TVA. Total CLA and cis-9, trans-11 CLA isomers were increased (P < 0.05) in p.a. and s.c. adipose depots, whereas i.m. adipose tissue contained increased (P < 0.05) amounts of total PUFA. Results from this study indicate that short-term lipid supplementation to feedlot cattle can increase adipose tissue CLA concentrations, but only marginally (8.3 to 17.5%). Moreover, observed decreases in oleic acid and total MUFA concentrations of adipose tissues from heifers fed rumen-protected CLA or corn oil suggest that lipid supplementation may decrease delta9 desaturase activity in adipose tissues, which in turn would lower the conversion of TVA to cis-9, trans-11 CLA isomer.  相似文献

Our objective was to determine effects of dietary high-oleate (Oleate; 76% 18:1) or high-linoleate (Linoleate; 78% 18:2) safflower seeds on fatty acids in muscle and adipose tissue of feedlot lambs. White-faced ewe lambs (n = 36) were fed a beet pulp, oat hay, and soybean meal basal diet (Control), blocked by BW, and allotted randomly to dietary treatments. Cracked safflower seeds were used in isocaloric and isonitrogenous replacement of beet pulp, oat hay, and soybean meal so that Oleate and Linoleate diets contained 5.0% additional fat. Fatty acids were determined in semitendinosus, longissimus dorsi (longissimus), and adipose tissue from the tail head (tailhead adipose tissue), adjacent to the 12th rib (s.c. adipose tissue), and kidney and pelvic fat (KPH adipose tissue) depots. Fatty acid data were analyzed within muscle and adipose tissue as a split-block design. Single degree of freedom orthogonal contrasts were used to compare treatment effects. Average daily gain, feed efficiency, and carcass characteristics did not differ (P = 0.15 to 0.96) across dietary treatments. Adipose tissue saturated fatty acids were greater (P = 0.04) for Controls but were not different (P = 0.36) in muscle. Trans-vaccenic acid (18:1(trans-11)) increased (P < 0.0001) with safflower supplementation and was greater (P < 0.0001) in Linoleate than in Oleate for both tissue types. Linoleate lamb had greater (P < 0.0001) PUFA than Oleate lamb in muscle and adipose tissue. Conjugated linoleic acids (CLA; cis-9, trans-11 and trans-10, cis-12) were greater (P < 0.0001) in muscle and adipose tissue of lambs fed safflower seeds. Lambs fed Linoleate had greater (P < 0.0001) CLA in adipose tissue and muscle than lambs fed Oleate. Saturated fatty acids were greater (P < 0.0001) in s.c. adipose tissue than in tailhead adipose tissue. Mono- and polyunsaturated fatty acids were greater (P < 0.0001) in tailhead adipose tissue than in s.c. adipose tissue. Weight percentages of 18:1(trans-11) ranked tailhead adipose tissue = KPH adipose tissue > s.c. adipose tissue and semitendinosus > longissimus, whereas CLA ranked tailhead adipose tissue > s.c. adipose tissue > KPH adipose tissue and semitendinosus > longissimus. Feeding mono- and polyunsaturated fatty acids increased tissue 18:1(trans-11) and CLA, which is a favorable change in regard to current human dietary guidelines.  相似文献

Five beef cattle management regimens were evaluated for their effect on meat quality, fatty acid composition, and overall palatability of the longis-simus dorsi (LD) muscle in Angus cross steers. A 98-d growing phase was conducted using grass silage with or without supplementation of growth promotants (Revalor G and Rumensin) or soybean meal. Dietary treatments in the finishing phase were developed with or without supplementation of growth promotants based on exclusive feeding of forages with no grain supplementation, or the feeding of grain:forage (70:30) diets. Growth promotants increased (P < 0.01) shear force and tended (P = 0.06) to increase toughness of the LD muscle due to limited postmortem proteolytic activity (lower myofibrillar fragmentation index value; P = 0.02). Grain feeding increased DM and intramuscular fat content (P = 0.03 and P = 0.05, respectively) in the LD but decreased the sensory panel tenderness score (P = 0.01). 为探究饲养方式对滩羊羔羊肌肉和脂肪组织中脂肪酸组成的影响,试验选择24只健康且体重相近(14.16 kg±0.58 kg)的2月龄滩羊羔羊随机分成舍饲组和放牧组,每组12只,于4月龄屠宰取其背最长肌、股二头肌、膈肌、皮下脂肪、肾周脂肪、尾部脂肪,用气相色谱仪测定脂肪酸含量。结果表明:①在肌肉组织饱和脂肪酸中,与放牧组相比,舍饲组滩羊羔羊股二头肌丁酸、棕榈酸含量显著提高(P<0.05),膈肌辛酸、月桂酸、肉豆蔻酸含量显著降低(P<0.05)。在肌肉组织不饱和脂肪酸中,与放牧组比较,舍饲组滩羊羔羊背最长肌棕榈油酸、花生四烯酸含量显著降低(P<0.05),γ-亚麻酸、α-亚麻酸含量显著提高(P<0.05),二十碳五烯酸(EPA)、n-3多不饱和脂肪酸(n-3 PUFA)极显著提高(P<0.01);股二头肌花生四烯酸含量显著降低(P<0.05),十七碳一烯酸、α-亚麻酸、EPA、n-3 PUFA含量极显著提高(P<0.01);膈肌棕榈油酸含量显著降低(P<0.05)。②在脂肪组织饱和脂肪酸中,与放牧组比较,舍饲滩羊羔羊皮下脂肪十一碳酸、十五碳酸含量显著降低(P<0.05),十三碳酸含量极显著降低(P<0.01);肾周脂肪葵酸、肉豆蔻酸含量显著降低(P<0.05),丁酸、硬脂酸含量极显著提高(P<0.01);尾脂十一碳酸含量显著降低(P<0.05)。在脂肪组织不饱和脂肪酸中,与放牧组比较,舍饲滩羊羔羊皮下脂肪十八碳二烯酸含量极显著提高(P<0.01),肉豆蔻油酸、棕榈油酸、γ-亚麻酸含量极显著降低(P<0.01);肾周脂肪十八碳一烯酸、α-亚麻酸、n-3PUFA含量显著提高(P<0.05),肉豆蔻油酸含量显著降低(P<0.05),十七碳一烯酸、二十碳二烯酸、十八碳二烯酸含量极显著提高(P<0.01);尾脂肉豆蔻油酸、棕榈油酸含量显著降低(P<0.05),十八碳二烯酸含量极显著提高(P<0.01)。综上,不同饲养方式对滩羊羔羊肌肉和脂肪组织脂肪酸含量存在一定影响,舍饲滩羊羔羊股二头肌中饱和脂肪酸含量及肾周脂肪中不饱和脂肪酸含量显著高于放牧滩羊羔羊。  相似文献

There is no information about the effect of different pasture types on tissue fatty acid profiles of a native rustic lamb breed of the Chiloe Archipelago, the Chilota. Eight Chilota lambs were grazed on a ‘Calafatal’ pasture (CP), a typical secondary succession of Chiloé Archipelago (Chile) and eight Chilota lambs were located to graze on naturalized pasture (NP) of Chiloé. Botanical, chemical and lipid composition of the two types of pastures and of different lamb tissues (muscle, subcutaneous – and tail adipose tissues) and plasma were performed. Both pasture types induced high n‐3 polyunsaturated fatty acids (PUFA) and CLAcis‐9,trans‐11 proportions in Chilota meat. Thus, in muscle, Chilota lambs grazing CP showed higher sum PUFA, sum n‐6 PUFA proportion and n‐6/n‐3 PUFA ratio compared with Chilota lambs grazing NP. In tail fats of Chilota lambs grazing CP significantly higher proportions of 18:3n‐3, sum saturated fatty acids, sum PUFA, n‐3 and n‐6 PUFA were detected compared with Chilota lambs grazing NP. Feeding of different pasture types (CP vs. NP) caused significant differences in fatty acid composition of muscle and the two fat depots in Chilota lambs, but also point to tissue‐specific responses of de novo synthesized fatty acid deposition in the tissues.  相似文献

We hypothesized that stearoyl-CoA desaturase (SCD) enzyme activity would not correlate with fatty acid indices of SCD activity in steers fed different grains. Forty-five Angus steers (358 +/- 26 kg BW) were individually fed for 107 d diets differing in whole cottonseed (WCS) supplementation (0, 5, or 15% of DM) and grain source (rolled corn, flaxseed plus rolled corn, or ground sorghum grain) in a 3 x 3 factorial arrangement. Flaxseed- and corn-fed steers had greater (P < 0.01) G:F (0.119 and 0.108, respectively) than sorghum-fed steers (0.093). Marbling score was decreased by WCS (P = 0.04), and LM area was decreased (P < 0.01) by sorghum. Plasma 14:0, 16:0, 16:1n-7, and 18:2n-6 were greatest in corn-fed steers, whereas plasma 18:3n-3 and 20:5n-3 were greatest in the flax-seed-fed steers (P < 0.01). Plasma 18:1trans-11 was least in sorghum-fed steers, and plasma cis-9,trans-11 CLA was barely detectable, in spite of high intestinal mucosal SCD enzyme activity (118 to 141 nmol*g tissue(-1).7 min(-1)). Interfascicular (i.f.) and s.c. cis-9,trans-11 CLA remained unchanged (P > or = 0.25) by treatment, although 18:1trans-11 was increased (P < or = 0.02) in steers fed corn or flaxseed. Steers fed flaxseed also had greater (P < 0.01) i.f. and s.c. concentrations of 18:3n-3 than steers fed the other grain sources. Oleic acid (18:1n-9) was least and total SFA were greatest (P < 0.01) in i.f. adipose tissue of steers fed 15% WCS. Lipogenesis from acetate in s.c. adipose tissue was greater (P < 0.01) in flaxseed-fed steers than in the corn- or sorghum-fed steers. Steers fed flaxseed or corn had larger i.f. mean adipocyte volumes (P < 0.01) than those fed sorghum and tended (P = 0.07) to have larger s.c. adipocyte volumes. Several fatty acid indices of SCD enzyme activity were decreased (P < or = 0.03) by WCS in i.f. adipose tissue, including the 18:2cis-9,trans-11/ 18:1trans-11 ratio. The 18:2cis-9,trans-11/18:1trans-11 ratio also tended to be decreased (P = 0.09) in s.c. adipose tissue by flaxseed; however, SCD enzyme activities in i.f. and s.c. adipose tissue were not affected by dietary WCS (P > or = 0.47) or grain source (P > or = 0.37). Differences in SFA seemed to be independent of SCD enzyme activity in both adipose tissues, suggesting that duodenal concentrations of fatty acids were more important in determining tissue fatty acid concentrations than endogenous desaturation by SCD.  相似文献

Abstract

With the objective of studying the effects of production systems on meat quality, 75 Norwegian White Sheep lambs were subjected to one of the following treatments: continuous grazing on a semi-natural lowland pasture until slaughtering (Control); continuous grazing followed by either stall-feeding on concentrate and grass silage or grazing ryegrass pasture for 44 or 24 days before slaughtering (Conc44, Conc24, Rye44, Rye24). Loin samples of M. longissimus dorsi including the subcutaneous fat were analysed for sensory attributes and fatty acid composition. Compared with the control group, a lower intensity of acid taste (P<0.05) and a lower content of C18:3n-6 fatty acids (P<0.001) were observed in the Conc44 group. The n-6/n-3 ratio was higher (P<0.001) in meat tested from the concentrate treatments compared to the ryegrass treatments. These findings indicate that the fattening of lambs on improved pastures or a concentrate-based diet prior to slaughter may alter meat characteristics.  相似文献

Carotenoid pigments are good biomarkers of grass feeding in sheep. However, grazing lambs are often stall-finished because of grass shortage. We investigated the nature of the carotenoids present in sheep blood and their persistence in this tissue. Four treatments were compared: 1) feeding a concentrate-based diet (n = 10 lambs), 2) grazing followed by a long stall-finishing period (n = 10), 3) grazing followed by a short stall-finishing period (n = 10), and 4) grazing to slaughter weight (n = 10). The concentrate supply was regulated to have similar average daily gain for all treatments. The 40 lambs were allocated to either the grazing or the stall treatments on the basis of their birth date, birth weight, and body weight. The 30 grazing lambs were further allocated to long-stall, short-stall, or grass treatment on the basis of their body weight and plasma carotenoid content. Plasma content of total carotenoids was measured by spectrophotometry during the grazing and the stall periods for all lambs and at slaughter weight for the eight heaviest lambs of each treatment. Analysis of the nature and the concentration of individual carotenoids was performed by HPLC on pasture and stall diets and on blood of grazing lambs. The carotenoid content of the stall diet was 2 to 3% that of the pasture diet. Lutein, zeaxanthin, and beta-carotene accounted for 43 to 58%, 3 to 17%, and 0 to 7% of total plasma carotenoids in grazing lambs, respectively. Two unknown polar carotenoids, expressed in lutein equivalent, accounted for 10 to 22% and 0 to 9% of total carotenoids. Plasma carotenoid content during the grazing and the finishing periods varied among animals (P < 0.001). At slaughter weight, plasma carotenoid content was higher for grass-fed than for stall-fed, long-stall finished, or short-stall finished lambs (P < 0.001), and reliably distinguished grass-fed lambs from all the others. Plasma carotenoid content decreased exponentially with the interval from starting on the stall diet (P < 0.005). The deceleration parameter of the model increased linearly with lamb average daily gain during the stall-finishing period, suggesting that the turnover of carotenoids in the blood may depend on the level of intake of the stall-finishing diet. After 4 to 13 d on the stall diet, depending on the initial plasma carotenoid concentration, plasma carotenoid concentration of previously grazed, stall-finished lambs fell to the values of lambs fed a concentrate diet without grazing. Such a low persistence is of interest for discriminating grazing lambs from stall-finished grazing lambs.  相似文献

The aim of this study was to investigate the effects of castration on carcass characteristics and fatty acid profile of visceral fat and meat from lambs. Eighteen six-month-old Santa Inês male lambs (18.9?±?2.4 kg of body weight (BW)) were used. Animals were assigned to two treatments according to their sexual condition: uncastrated (U) or castrated (C). During a 98-day experimental period, animals were kept on an Andropogon gayanus grass pasture area of 1 ha and supplemented with 200 g/animal/day of concentrate mixture. Water and mineral salt were available ad libitum. The lambs were weighed fortnightly, and at the last day of the trial, they were slaughtered for evaluation of carcass characteristics and fatty acids profile of perirenal fat and longissimus lumborum muscle samples. Castrated lambs showed higher BW than U during most part of the experiment (p?<?0.05). Fat deposition was higher in C lambs as evidenced by their increased carcass fat cover. Meat from U lambs showed lower content of C16:0 and higher polyunsaturated fatty acids (PUFA) (p?<?0.05) than that from C (U, 14.3 and C, 10.5%). Conjugated linoleic acid (CLA) content was not affected by castration (p?>?0.05) (U, 0.74 and C, 0.76%). The cis-9, cis-12 C18:2n-6 (U, 10.6 and C, 6.86%) fatty acid and the PUFA:SFA (saturated fatty acids) ratio (U, 0.36 and C, 0.25) were higher in the muscle of U lambs (p?<?0.05), indicating that the meat from U animals may provide more benefits to human health than that of C.

Six Hereford steers (295 kg) cannulated in the proximal duodenum were used to evaluate the effects of forage and sunflower oil level on ruminal biohydrogenation (BH) and conjugated linoleic acid (CLA) outflow. Steers were fed one of six treatment diets in a 3 x 2 factorial arrangement of treatments (grass hay level: 12, 24, or 36% of DM; and sunflower oil level: 2 or 4% of DM) in a 6 x 6 Latin square design. The remainder of the diet was made up of steam rolled corn and protein/mineral supplement. Duodenal samples were collected for 4 d following 10-d diet adaptation periods. Data were analyzed with animal, period, forage level, sunflower oil level, and two-way interaction between forage and sunflower oil level in the model. Dry matter intake showed a quadratic response (P < 0.04), with an increase in DMI as forage level increased from 12 to 24% followed by a decrease in DMI when 36% forage was fed. Flow of fatty acids at the duodenum was higher (P < 0.03) for 4 vs. 2% sunflower oil diets, and similar among forage levels. Apparent ruminal digestibility of NDF increased in a linear manner (P < 0.04) as dietary forage level increased. Ruminal BH of dietary unsaturated 18-C fatty acids, oleic acid, and linoleic acid increased linearly (P < 0.05) as dietary forage level increased. Linoleic acid BH tended (P < 0.07) to be greater for 4 than 2% sunflower oil level. Duodenal flow of pentadecyclic, stearic, linolenic, and arachidic acids increased linearly (P < 0.05) as dietary forage level increased from 12 to 36%. Duodenal flow of linoleic acid decreased in a linear manner (P < 0.03) with increasing dietary forage level. Flow of trans-10 octadecenoate decreased linearly (P < 0.03) as dietary forage level increased, whereas trans-11 vaccenic acid flow to the duodenum increased (P < 0.01) linearly with increased dietary forage. Dietary forage or sunflower oil levels did not alter the outflow of cis-9, trans-11 CLA. Flows of cis-11, trans-13, and cis-9, cis-11 CLA increased linearly (P < 0.05) with increased dietary forage. Flows of cis-11, cis-13, and trans-11, trans-13 CLA decreased linearly (P < 0.05) with increased dietary forage. Increasing dietary forage levels from 12 to 36% in beef cattle finishing diets increased BH of unsaturated 18-C fatty acid and outflow of trans-11 vaccenic acid to duodenum without altering cis-9, trans-11 CLA outflow.  相似文献

Two experiments were conducted to determine the effectiveness of a rumen-protected CLA (pCLA) supplement and the impact of feeding this pCLA on carcass characteristics and tissue fatty acid composition of lambs. In Exp. 1, a CLA-80 preparation (80% pure CLA; contained similar proportions of cis-9, trans-11, and trans-10, cis-12 CLA), protected against rumen degradation, was fed to sheep, and the proportion of CLA reaching the duodenum was determined. A 3 x 3 Latin square design was used with 3 diets (1.4 kg of concentrate-based control diet, the same control diet plus 22 g of CLA-80, or the same control diet plus 110 g of pCLA/d), 3 feeding periods, and 3 rumen and duodenally cannulated sheep (Mule x Charolais males, 10 mo of age, BW 55.3 +/- 1.8 kg). After 7 d of feeding, sheep were ruminally infused with chromium EDTA and Yb acetate for 7 d, after which samples of duodenal digesta were collected every 6 h for 48 h to determine the quantity of CLA reaching the small intestine each day. The amounts of CLA cis-9, trans-11 and trans-10, cis-12, and combined isomers, flowing through the duodenum each day were greater (P = 0.01) in sheep fed pCLA. Approximately 65% of the pCLA avoided rumen biohydrogenation, with the ratio of the 2 main isomers remaining similar. In Exp. 2, 36 Mule x Charolais ewe lambs (approximately 13-wk old, average initial BW 29.3 kg) were fed 3 levels of the pCLA or Megalac, which were fed to provide an equivalent energy content at each pCLA level. Lambs were randomly assigned to 1 of 7 treatment groups, which were fed for 10 wk to achieve a growth rate of 180 g/d. Treatments included the basal diet and the basal diet plus 25, 50, or 100 g of pCLA/kg of diet or the equivalent amount of Megalac. In liver (P < 0.001) and all adipose tissue depots studied, the proportions of both CLA isomers increased (P = 0.02) with the amount of pCLA fed but were not altered with increasing of Megalac. Although there was no effect of treatment on cis-9, trans-11 CLA content, accumulation (P < 0.001) in the LM with increasing of pCLA supplementation was observed for the trans-10, cis-12 isomer. Although tissues had been enriched with CLA, there was no evidence of a reduction in adipose tissue or an increase in muscle mass in these sheep. However, an effect of pCLA on tissue fatty acid composition was consistent with an inhibition of stearoyl-CoA desaturase.  相似文献

Two lactating dairy cows fitted with a rumen cannula received successively diets containing 0%, 20%, 34% and again 0% of wheat on a dry matter basis. After 5, 10 and 11 days, ruminal pH was measured between 8:00 and 16:00 hours, and milk was analysed for fat content and fatty acid profile. Diets with 20% and 34% wheat induced a marginal and a severe subacute ruminal acidosis respectively. After 11 days, diets with wheat strongly reduced the milk yield and milk fat content, increased the proportions of C8:0 to C13:0 even- or odd-chain fatty acids, C18:2 n-6 and C18:3 n-3 fatty acids but decreased the proportions of C18:0 and cis-9 C18:1 fatty acids. Wheat also increased the proportions of trans-5 to trans-10 C18:1, the latter exhibiting a 10-fold increase with 34% of wheat compared with value during the initial 0% wheat period. There was also an increase of trans-10, cis-12 C18:2 fatty acid and a decrease of trans-11 to trans-16 C18:1 fatty acids. The evolution during adaptation or after return to a 0% wheat diet was rapid for pH but much slower for the fatty acid profile. The mean ruminal pH was closely related to milk fat content, the proportion of odd-chain fatty acids (linear relationship) and the ratio of trans-10 C18:1/trans-11 C18:1 (nonlinear relationship). Such changes in fatty acid profile suggested a possible use for non-invasive diagnosis of subacute ruminal acidosis.  相似文献

Our objective was to determine the effect of the duration of grazing before slaughter on the fatty acid composition of muscle fat and s.c. adipose tissue (SAT) of beef heifers. Sixty crossbred Charolais heifers (n = 15 per treatment) were assigned randomly to one of four dietary treatments: 45 animals (Treatments 1, 2, and 3, respectively) were housed at the beginning of the experiment, and 15 (Treatment 4) were fed at pasture. Two groups of 15 heifers were moved to pasture 40 d (Treatment 2) and 99 d (Treatment 3) before slaughter, respectively, resulting in preslaughter grazing periods of 0, 40, 99, or 158 d for Treatments 1, 2, 3, and 4, respectively. Before grazing the predominantly perennial ryegrass pasture, animals were housed and offered grass silage ad libitum and 3 kg of concentrate diet (650 g of grass silage/kg of total DMI). After slaughter, the fatty acid profile of the neutral (NL) and polar lipid (PL) fractions of muscle fat from the LM and the total lipids from SAT were analyzed by gas chromatography. Duration of grazing showed a quadratic tendency on mean carcass weight (P = 0.08), but did not affect growth (P = 0.27) or the lipid content (P = 0.13) of the LM. Increasing the duration of grazing led to a linear increase (P < 0.001) in the concentration (on fresh-tissue basis) of CLA in muscle fat (from 11.80 to 17.75 mg/100 g of muscle in NL, and from 0.52 to 0.82 mg/100 of g muscle in PL) and in SAT (from 3.98 to 10.23 mg/g of SAT; P < 0.001), and increased the concentration of C18:1trans-11 in both muscle fat fractions (P < 0.001) and in SAT (P < 0.001). In the total muscle lipids, the polyunsaturated to saturated fatty acid ratio (P:S) increased from 0.12 to 0.15 with increased duration of grazing following a linear (P < 0.05) and cubic pattern (P < 0.05). Increasing the duration of grazing led to a linear decrease in the n-6:n-3 ratio of muscle fat from 2.00 to 1.32 (P < 0.001), and from 2.64 to 1.65 in the SAT lipids (P < 0.001), mainly as a consequence of the increased concentration of C18:3n-3. It is concluded that muscle fat and SAT fatty acid profile was improved from a human health perspective by pasture feeding, and that this improvement depended on the duration of grazing.  相似文献

Our objective was to determine the effect of oil supplementation of pasture fed, beef cattle on the fatty acids, particularly CLA and PUFA, of muscle and s.c. adipose tissue. Forty-five Charolais crossbred heifers were blocked on BW and randomly assigned to 1 of 3 dietary regimens in a randomized complete block design (n = 15). The 3 treatments were: unsupplemented grazing (GO), restricted grazing plus a sunflower oil-enriched ration (SO), or restricted grazing plus a linseed oil-enriched ration (LO). Heifers were fed the experimental diets for approximately 158 d. Samples of LM muscle and s.c. adipose tissue were taken postmortem, the muscle fat was separated into neutral lipid and polar lipid (no separation was performed on the s.c. adipose tissue), and the fatty acid profile was determined by GLC. No effect of dietary treatment on carcass weight or total fatty acid concentration (mean 2,571 mg/100 g of muscle) in muscle fat was detected. Heifers offered SO had a greater (P < 0.001) proportion of CLA and C18:1trans-11 (1.90 and 9.35 vs. 1.35 and 6.89 g/100 g of fatty acids, respectively) in neutral lipid of muscle fat compared with those offered LO, which had a greater proportion of CLA and C18:1trans-11 than heifers offered GO (0.78 and 3.37 g/100 g of fatty acids, respectively). Similar effects were observed in the polar lipid and s.c. lipid. The PUFA:SFA ratio was greater in muscle fat and s.c. adipose tissue from supplemented heifers than in those offered GO (P < 0.001). Compared with LO, the PUFA:SFA ratio was greater (P < 0.05) in muscle fat of heifers offered SO, but there was no difference between SO and LO for this ratio in s.c. adipose tissue. The n-6:n-3 PUFA ratio was similar in muscle and s.c. adipose tissue for GO and LO, but it was greater (P < 0.05) for SO. It is concluded that supplementation of pasture-fed cattle with plant oil-enriched concentrates resulted in an increase in beef fat of some fatty acids considered to be of benefit to human health. Concentrates enriched with sunflower oil were more effective in increasing the CLA concentration, whereas linseed oil-enriched concentrates resulted in a more favorable n-6:n-3 PUFA ratio. The relevance to human health of the associated increase in C18:1trans-11 merits investigation.  相似文献

Because of a high prevalence of anthelmintic resistance and consumer demand for chemical free meat products, management tools to minimize the need for deworming are needed. The objective was to examine the effectiveness of grazing sericea lespedeza (SL) in a mixed grass or a pure forage system for control of gastrointestinal nematodes (GIN); in other words pasture systems included grass, grass plus SL, or SL alone (Experiments 2 and 3). Selective use of copper oxide wire particles (COWP) based on the FAMACHA(?) system was used to aid in GIN control. In Experiment 1, lambs co-grazed bermudagrass (BG; n=21) or SL in a mixed grass pasture (SLM; n=22) with dams for 14 days. In Experiment 2, lambs grazed BG (n=14), SLM (n=13), or pure SL (SLP; n=13) pastures for 56 days. In Experiment 3, doe kids grazed BG (n=12), SLM (n=13), or SLP (n=13) for 84 days. Animals were fed a 16% crude protein supplement based on NRC requirements and estimated forage quality of pastures, so that 454, 389, and 200 g/lamb (Experiment 2), or 454, 300, and 150 g of supplement/goat (Experiment 3) was fed to BG, SLM, and SLP, respectively. Animals were dewormed with COWP if FAMACHA(?) was >3. Coprocultures were conducted to identify GIN genus. In Experiment 1, FEC were reduced in lambs grazing SLM compared with BG pastures. In Experiment 2, FEC were reduced in SLP compared with BG lambs on all days, and reduced in SLM compared with BG lambs on day 56. Initially, Haemonchus contortus was the predominant nematode, but the population shifted to other species in the SL groups by the end of the study. The mean number of dewormings/lamb was 0.71, 0.20, and 0.21±0.13 for BG, SLM, and SLP groups, respectively. In goats in Experiment 3, Trichostrongylus spp. was the predominant nematode in May and June and H. contortus in July. There was little meaningful effect of forage treatments on GIN infection in kids. Because H. contortus was not the predominant nematode in kids, the integrated approaches used may not have been effective in controlling GIN. Grazing SL was effective for GIN control in lambs in early summer, but the effect was delayed in lambs grazing a mixed SL/grass pasture unless lambs initially grazed with dams. An integrated approach used that included SL grazing and COWP for deworming was effective in lambs.  相似文献

Despite the widespread adoption of clean grazing systems in lowland sheep flocks, detailed parasitological investigations had not previously been carried out on such flocks. A trial was therefore conducted on two commercial flocks: a traditional permanent pasture flock (A) and one operating a system of clean grazing (B), and on an East of Scotland College flock (C) which had operated a clean grazing system for eight years. Ewe and lamb worm egg output, pasture larval levels and lamb liveweight gains were monitored and tracer lambs were grazed during July and August on each farm. Under clean grazing conditions on farm C all parasitological parameters were lower than on both commercial farms. However, in the commercial flocks comparable contamination was evident from midsummer onwards and tracer lambs grazed during August on farm B had significantly greater worm burdens than on the other two farms. The differences observed between the flocks were thought to be due to greater residual contamination by overwintered larvae in both commercial flocks while the higher worm burdens in August on farm B probably resulted partly from incomplete control of the periparturient rise in ewe faecal egg output and partly to autoinfection of the lamb crop. It was concluded that farm C grazing was the cleanest. Considerable contamination was present on farm A while farm B occupied an intermediate position which resulted in considerable worm burdens in lambs grazing during the latter part of the season.  相似文献

The signature of carotenoid pigments stored in the caudal fat has been used to discriminate carcasses of lambs raised on pasture from those fed concentrates. However, the literature reports higher concentration of carotenoids in perirenal than in caudal fat, and it is likely that carotenoid concentration in fat increases with shrinkage time. Our objectives were to investigate the effects of measurement site and shrinkage time after slaughter on the reliability of the discrimination between grass- and concentrate-fed lambs. Thirty-two lambs were fed concentrate-based diets (stall, n = 16) or grazed a natural pasture in France (grass, n = 16). The concentrate supply was regulated to have similar ADG between the two treatments. Periodically lambs weighing about 35 kg were slaughtered. Reflectance spectrum and lightness (L*), redness (a*), and yellowness (b*) were measured at slaughter and after 24 h of shrinkage in the subcutaneous caudal and in the perirenal fat. The spectra were translated to have a value of reflectance at 510 nm = 0 and the integral of the translated spectrum was calculated between 450 and 510 nm (i.e., the zone of light absorption by carotenoids). The integral value of subcutaneous caudal fat at slaughter differed between stall- and grass-fed lambs (P < 0.05) but was not reliable to discriminate between feeding treatments. After 24-h shrinkage, the discrimination was better but was uncertain for 50% of the lambs. The integral value of perirenal fat at slaughter allowed us to discriminate all the lambs fed concentrates (16) from all those fed pasture (16). After 24-h shrinkage, the discrimination based on perirenal fat was partially compromised; 21.9% of the animals were not correctly discriminated. The integral value of the translated spectrum was highly (P < 0.001) correlated with the fat yellowness (b*), but the b* value was not reliable to discriminate between feeding treatments. However, plotting yellowness (b*) against redness (a*) values allowed us to discriminate 31 out of 32 lambs. Measuring reflectance spectrum of perirenal fat at slaughter allowed for complete discrimination of grass-fed from stall-fed lambs. However, because the integral values were affected by shrinkage time, measurements should be rigorously made immediately after slaughter.  相似文献

Three-year-old Angus x Gelbvieh beef cows, which were nutritionally managed to achieve a BCS of 4 +/- 0.07 (479 +/- 36 kg of BW) or 6 +/- 0.07 (580 +/- 53 kg of BW) at parturition, were used in a 2-yr experiment (n = 36/yr) to determine the effects of maternal BCS at parturition and postpartum lipid supplementation on fatty acid profile of suckling calf plasma and adipose tissue. Beginning 3 d postpartum, cows within each BCS were assigned randomly to 1 of 3 treatments in which cows were all fed hay and either a low-fat (control) supplement or supplements with either high-linoleate cracked safflower seeds (linoleate) or high-oleate cracked safflower seeds (oleate) until d 61 of lactation. Diets were formulated to be isonitrogenous and isocaloric, and safflower seed supplements were provided to achieve 5% of DMI as fat. Total concentration of fatty acids in plasma did not differ (P = 0.48) due to maternal BCS at parturition. Percentage of 20:5n-3 in plasma tended (P = 0.06) to be greater for calves suckling cows with a BCS of 6 at parturition. No other differences (P = 0.12 to 0.99) were noted in calf plasma fatty acid profile due to maternal BCS at parturition. Likewise, no differences were detected for total fatty acid concentration (P = 0.88) in calf adipose tissue due to maternal BCS at parturition. Weight percentage of 14:1 (P = 0.001) was greatest in adipose tissue of calves suckling cows fed control and oleate; however, the percentages of 14:0, 15:0, 16:0, 16:1, 17:0, and 18:3n-3 were greater (P < 0.001) in adipose tissue from calves suckling cows fed control compared with calves suckling cows fed linoleate or oleate. Percentages of 18:0, 18:1trans-11, 18:2n-6, and cis-9, trans-11 CLA were greater (P < 0.001) in adipose tissue from calves suckling cows fed linoleate compared with calves suckling cows fed control and oleate. Calves suckling cows fed oleate had greater (P < 0.001) percentages of 18:1trans-9, 18:1trans-10, and 18:1cis-9 in adipose tissue than calves suckling cows fed control or linoleate. Calf plasma and adipose tissue fatty acid profiles were reflective of milk fatty acids. Because fatty acids play an important role in metabolic regulatory functions, changes in milk fatty acid profile should be considered when beef cows are fed lipid supplements.  相似文献
