Internal parasitism is a pervasive constant that reduces returns in beef cattle production. Parasitism may influence production sufficiently so that data derived investigating performance response will be erroneous. The interaction of internal parasites of cattle with other facets of their lives makes it imperative that more parasite research be done on cattle to consider their impact. Results of trials designed to determine forage production at various stocking densities may not reflect the nutritive value of the forage, but instead the severity of parasite exposure. Relative resistance or susceptibility to certain parasites vary with the breed of livestock and the species of parasite. Extensive work to evaluate the ability of various breeds and sires to influence parasitic numbers has been conducted in sheep but not in cattle. Some parasite trials have ignored the effects of nutrition on the establishment and retention of parasites. This becomes especially important where multiple parasite exposure may occur. Susceptible cattle, even on an optimal diet, will become parasitized, but if reexposed to the same species of parasite, cattle on an optimal diet may be able to resist reinfection, whereas those on deficient diets will not. 相似文献

To control parasitic disease in cattle, one first has to determine what parasites he or she is trying to control--nuisance and biting flies in a milking parlor or ticks on range cattle? The determination may be based on geographic location, class of cattle, and their management. After determining what is there, then if the numbers are sufficient, justify treatment. Each livestock producer has a different array of parasites with which to deal, and in some circumstances, parasitic disease is below the economic threshold. Others may have occasional clinical disease associated with parasites, but most have situations in which the potential of economic loss is high enough to justify control. 相似文献

The complex relationship between livestock and their parasites is now much better understood. The pattern of infection and reinfection is a dynamic, but predictable, phenomenon that varies with geographic region, production system and grazing system used. Preventive programs that use anthelmintics offer the most efficient approach for grazing animals and confined species. Moxidectin (MXD) is a milbemycin endectocide compound active at extremely low dosages against a wide variety of nematode and arthropod parasites. Different pharmacological approaches are currently being tested to delay the bile-faecal elimination and to obtain increased systemic availability for endectocide molecules in ruminants. Loperamide (LPM) is an opioid derivative, whose main pharmacological action is to abolish intestinal propulsive peristaltic waves. The influence of LPM on the pattern of faecal excretion of MXD and on its plasma disposition following intravenous (i.v.) and subcutaneous (s.c.) administrations to cattle was evaluated in the current work. Parasite-free calves were treated with MXD given either alone at 200 microg/kg by i.v. (Experiment 1) and s.c. (Experiment 2) administrations or coadministered with LPM subcutaneously injected at 0.4 mg/kg. Blood and faecal samples were collected over a period of 20 (Experiment 1) and 40 (Experiment 2) days post-treatment. The recovered plasma and faecal samples were extracted and analysed by high-performance liquid chromatography (HPLC) using fluorescence detection. Significantly higher MXD plasma concentrations were obtained after the coadministration of MXD + LPM compared with treatments with MXD alone by both routes. The higher MXD plasma concentration profiles measured after the coadministration with LPM accounted for the significantly higher AUC values obtained following the i.v. (> 46%) and s.c. (> 38%) treatments. A reduced MXD body clearance was observed in the presence of LPM. The appearance of MXD in faeces was significantly delayed after the i.v. and s.c. coadministrations of MXD with LPM (T(1/2app)=5.87 and 10.6 h, respectively) than that observed after the treatment with MXD alone (T(1/2app)=3.48 and 5.12 h). A delayed MXD peak concentration in faeces collected from MXD + LPM-treated animals compared with those receiving MXD alone, was observed. The delayed intestinal transit time caused by LPM and a potential competition between MXD and LPM for the P-glycoprotein-mediated bile/intestinal secretion processes, may account for the enhanced MXD systemic availability measured in cattle in the current work. 相似文献

The plasma kinetics disposition of moxidectin following a subcutaneous administration with a long-acting formulation (Cydectin) 10%, Fort Dodge Animal Health, France) at the recommended dose of 1 mg kg(-1) body weight was evaluated in Charolais cattle breed (five females weighing 425-450 kg) for 120 days. Furthermore, its concentration was measured in hair for the same period. After plasma extraction and derivatization, samples were analysed by HPLC with fluorescence detection. Moxidectin was first detected at 1 h after treatment for plasma (2.00+/-1.52 ng ml(-1)) and at 2 days for hair (446.44+/-193.26 ng g(-1)). The peak plasma concentration (C(max)) was 55.71+/-15.59 ng ml(-1) and 444.44+/-190.45 ng g(-1) for plasma and hair, respectively. The mean calculated time of peak occurrence (T(max)) was 3.40+/-3.36 and 2 days for plasma and hair, respectively. The mean residence time (MRT) was 28.93+/-2.87 and 13.32+/-2.48 days for plasma and hair cattle. The area under concentration-time curve (AUC) was 1278.95+/-228.92 ng day ml(-1) and 2663.82+/-1096.62 ng day g(-1) for plasma and hair, respectively. At the last sampling time (120 days), the concentration was 1.91+/-0.26 ng ml(-1) and 0.69+/-0.52 ng g(-1) for plasma and hair, respectively. The bioavailability of this long-acting formulation of moxidectin is similar to that registered after subcutaneous administration of moxidectin in cattle at 0.2 mg kg(-1) body weight. For the first time the moxidectin pharmacokinetics parameters in hair after a subcutaneous administration was described. The moxidectin profile concentrations in hair reflected that registered in plasma. The previous studies of efficacy have to be correlated to the extended period of absorption and distribution by the LA formulation due to the fivefold higher dose rate in comparison with the 1% injectable formulation (0.2 mg kg(-1) body weight). 相似文献

Three groups of 11 naturally infected crossbred beef calves were injected subcutaneously with moxidectin 1 per cent injectable at 0.2 or 0.3 mg moxidectin/kg bodyweight or with the unmedicated vehicle. Nematode infections had been acquired during grazing from December to April. Based on the faecal egg counts and total worm counts of the control calves at necropsy (11 to 13 days after treatment) most of the calves had heavy parasitic burdens. Ostertagia ostertagi was predominant and the mean numbers of adults, developing fourth stage larvae (L4) and inhibited early L4 were 45,906, 10,061 and 68,918, respectively. Haemonchus placei and Trichostrongylus axei were also present in the abomasa. Three species of Cooperia, Oesophagostomum radiatum L4 and T colubriformis adults were found in the intestinal tract. Both dosages of moxidectin were equally effective (P < 0.05) against all the abomasal nematodes (99.9 to 100 per cent) and the intestinal tract nematodes (99.4 to 100 per cent). No adverse reactions to the moxidectin treatment were observed. The activity of moxidectin was evaluated in 1988 and 1989 against natural infections of internal parasites in 20 critical tests (n = 20 equids) and three controlled tests (n = 20 equids). Two formulations, injectable administered intramuscularly (i.m.) or intraorally (i.o.) and gel i.o., were given at dose rates of 0.2, 0.3 or 0.4 mg kg-1 body weight. For the critical tests (all three dose rates evaluated), removals of second instar Gasterophilus intestinalis were 93-100%, except (89%) for the injectable formulation (i.m.) at 0.2 mg kg-1. Removals of third instar G. intestinalis were 88-100% for the injectable formulation given i.m. or i.o. and 93-100% for the gel formulation, except (53%) for one batch (0.4 mg kg-1). Activity was 100% for third instar Gasterophilus nasalis, Parascaris equorum, Strongylus vulgaris and Strongylus edentatus. For Oxyuris equi, removals were 91-100%, except (27%) for one batch of the injectable formulation given i.o. at 0.3 mg kg-1. There was apparent activity against migrating S. vulgaris and S. edentatus at various dose rates and routes of administration for both formulations. At necropsy, there were local reactions observed at the injection site of three equids. In the controlled tests, dose rates were 0.2 or 0.4 mg kg-1. Removal of third instar G. intestinalis was highest for the injectable formulation given i.m. All formulations and dose rates were highly effective against S. vulgaris and S. edentatus, but variable and incomplete against O. equi. Removal was excellent on Habronema muscae and on migrating S. vulgaris and S. edentatus, although incomplete on S. vulgaris. Gasterophilus nasalis third instars and P. equorum were present in low numbers in some non-treated equids, but none were recovered from treated equids. Toxicosis was not evident. 相似文献

The occurrence of internal parasites in Washinton cattle was estimated. During the survey period in 1972 and 1973, more than 3,700 fecal samples from 29 herds and 4 feedlots, and viscera from 55 cattle were examined. The percentage of fecal samples with parasite eggs or oocysts from herds was as follows: oocysts, 77%; "strongyline" eggs, 44%; Nematodirus eggs, 3%; Moniezia eggs, 3%; Fasciola eggs, 2%; Trichuris eggs, 2%; and Dictyocaulus larvae, 1%. The fecal samples from feedlots contained principally strongyline eggs which varied markedly in numbers among groups of cattle in the lots. The predominant parasite in viscera was Ostertagia spp, which was recovered from all of the cattle examined. Other parasites identified in the survey were found less frequently in these examinations. Cattle in dairy and beef operations utilizing irrigated pastures were particularly prone to Fasciola infections, and more strongyline eggs were observed there than in fecal samples from other management systems. A grazing experiment with young cattle was conducted over two consecutive (1997, 1998) grazing seasons on semi-natural pasturelands in central-eastern Sweden. Comparisons were made between groups of animals that were either untreated and set-stocked, ivermectin bolus treated and set-stocked or untreated but moved in mid-summer (mid-July) to ungrazed pasture. The whole experimental area had remained virtually free of cattle during the previous two seasons and the cattle had been raised indoors since birth. To introduce low-levels of parasite infection into the experimental system, each animal received a ‘priming dose’ of approximately 10,000 infective trichostrongylid larvae at the time of turnout for both years. Results of the first year study showed that the level of parasitism was so low that it failed to induce any productivity losses in both groups of untreated cattle, which grew as well as those given boluses at turnout. In contrast, in 1998 both groups of untreated cattle suffered varying degrees of sub-clinical and clinical parasitism to result in an average of 30 kg liveweight depression, compared with the bolus treated cattle, at the end of the season. The only major departure between the two years was that in the latter, the cattle in the untreated groups were exposed to infective larval pickup, which had overwintered on pasture. Cattle in the move treatment grazed in the same sequence on pastures used by similar classes of animals during the previous year. That is, their pastures at turnout had not been grazed since mid-summer of the previous year. Clearly this early season (1997) grazing by young cattle resulted in sufficient overwintered larvae at the start of the following year (1998) to cause productivity losses of the same magnitude as those recorded for young cattle grazing on pastures contaminated for the entire grazing season of the previous year. This was confirmed by tracer tests that were carried out on all treatments, at the time of turnout and the mid-summer move in 1999. These results have major significance to organic cattle producers in Sweden who have a much higher tendency to practice a variety of grazing management techniques aimed at controlling nematode parasite infections in young cattle, than their conventional farming colleagues. It has been identified that one of these strategies is to simply use summer/autumn saved pastures for young stock at turnout, which if grazed by young stock prior to this, could prove to be counter-productive. 相似文献

The efficacy of moxidectin, a new endectocide against natural nematode infections, was evaluated. Twenty-five calves were divided into two groups of eight calves each and a third group of nine calves. Moxidectin was administered s.c. to two groups (I, 0.2 mg kg-1 body weight (BW); II, 0.3 mg kg-1 BW) and the third group (III) received the vehicle (placebo) s.c. Two pre-treatment and one post-treatment faecal nematode egg count determinations were made from all calves, and they were necropsied 2 weeks after treatment for the identification and enumeration of nematode parasites. Group III calves, which received the drug-free vehicle, harboured eight species of nematodes (Ostertagia ostertagi, Trichostrongylus axei, Cooperia oncophora, Cooperia punctata, Nematodirus helvetianus, Trichuris discolor, Oesophagostomum radiatum and Dictyocaulus viviparus). The mean total worm burden for this group was 8935. There was a significant reduction in the numbers of many species of nematodes (Ostertagia, Trichostrongylus, Cooperia and Nematodirus) in both treated groups. Cooperia oncophora was reduced by 94% in Group I and by 96% in Group II, while all other nematode species were reduced by 99%. Immature stages of Ostertagia and Nematodirus were significantly reduced in the two treated groups. Two weeks after treatment, the mean faecal egg counts of both treated groups were reduced by more than 98%. Extract The important external parasites of cattle in New Zealand include the cattle tick, Haemaphysalis longicornis, which occurs mainly in northern districts, the biting louse, Damalinia bovis, and the long-nosed sucking louse, Linognathus vituli. There are two other lice, Haematopinus eurysternus, and Solenopotes capillatus but they are much less common. The mange mite, Chorioptes bovis, occurs mainly in the winter months and causes lesions on the legs, tail and escutcheon, and in bulls it may cause severe scrotal mange similar to that seen in rams. Demodex folliculorum causes lesions deep in the skin and these constitute a serious problem in the leather industry. Stable flies, Stomoxys calcitrans, are blood suckers and their attacks worry cattle on pasture and in the milking sheds. At milking time they may inhibit milk let-down and the teat cups may be kicked off by restless cows. Warble flies, Hypoderma spp., are seen only in imported cattle and have never become established in New Zealand. 相似文献

The reindeer husbandry range of Scandinavia overlaps with sheep, goat, and cattle pastures. The aim of this study was to determine whether reindeer are suitable hosts for ovine or bovine nematode parasites, and thus may spread these parasites into the reindeer husbandry regions. To render worm-free, twelve 4-month-old male reindeer calves, six lambs, and six bovine calves were given ivermectin at 200 microg/kg body weight. Five weeks post-treatment, six reindeer calves were each artificially dosed with 10,000 third-stage larvae (L3) of gastrointestinal nematodes derived from sheep, and an additional six reindeer with L3 derived from cattle. Lambs and bovine calves received the same dose of ovine and bovine larvae as reindeer, from the same larval source, respectively. Faecal samples collected on five occasions after the larval dosing revealed that by the fourth week, all reindeer calves, lambs, and bovine calves were infected. Animals were slaughtered on days 40 (reindeer) or 47 (lambs and bovine calves) after the larval dosing. Reindeer calves were most susceptible to L3 derived from sheep. The overall mean intensity of Haemochus contortus, Trichostrongylus axei, and Teladorsagia circumcincta, did not differ between reindeer and sheep; however, early fourth-stage larvae of H. contortus were more abundant in reindeer (p = 0.002). The establishment of bovine-derived Ostertagia ostertagi was similar in reindeer (62%) and bovine calves (57%), but larval inhibition was much higher in reindeer (91%, p < 0.001) than in cattle (31%). Very poor establishment of bovine derived Cooperia oncophora was recorded in reindeer calves (2%) compared with bovine calves (59%). These results show that young reindeer are susceptible hosts to the important gastrointestinal parasites of sheep (T. circumcincta, H. contortus) and cattle (O. ostertagi), as well as being a suitable host for T. axei. 相似文献

Abomasa, blood samples and faecal samples for examination of nematode infections were collected from 125 dairy cows during the period November 1997-October 1998. Of these, 12 had no grazing history and were, therefore, excluded from this study. From the remaining 113, 88.5% had nematode eggs in the faeces. Larval identification of the positive cultures showed that Ostertagia spp. larvae were most frequent (97%), followed by Trichostrongylus spp. (29%), Oesophagostomum spp. (23%), Cooperia punctata (20%), Cooperia oncophora (4%), Haemonchus contortus (2%) and Bunostomum phlebotomum (1%). The geometric mean EPG was 2.4. Two cows excreted larvae of Dictyocaulus viviparus (0.1 and 0.6 LPG resp.). Worms were found in the abomasa of 108 cows (96%). In all these abomasa Ostertagia spp. was present (100%). Trichostrongylus axei was found in 47 abomasa (43.5%) and two cows (2%) were infected with Capillaria bovis. The geometric mean of the total abomasal worm counts was 1743 and of Ostertagia spp. alone 1615. Almost all male worms were Ostertagia ostertagi, only occasionally Skrjabinagia lyrata10,000) total worm burden.Ostertagia specific antibodies were highest in late summer and autumn and lowest in spring and early summer. The same pattern, although not so pronounced, was observed for the serum pepsinogen values. No clear seasonal pattern was found for the Cooperia specific antibodies. A study of cattle diseases was carried out in Vanuatu from 1971 to 1981. Tuberculosis was discovered in 4 herds and eradication was completed by 1981. The number of farms with brucellosis reactors increased from 2 in 1976 to 7 in 1978 despite eradication measures. Antibodies to serovars Leptospira icterohaemorrhagiae, australis, sejroe and canicola were demonstrated by the microscopic agglutination test. Antibodies were demonstrated against infectious bovine rhinotracheitis/infectious pustular vulvovaginitis and bovine virus diarrhoea/mucosal disease complexes. Of the 18 parasites identified, Haemaphysalis longicornis, Haemonchus, sp, Oesophagostomum phlebotomum and Neoascaris vitulorum were the most prevalent. As brucelossis is the only serious disease present, Vanuatu is in a favourable situation with regard to cattle diseases. 相似文献

Duplicate studies in France and Northern Ireland were carried out to determine the persistent efficacy of topical moxidectin and doramectin against natural infections of Cooperia oncophora. In each study, groups of 15 nematode-na?ve calves were treated either with topical doramectin or moxidectin, and put out to graze on pasture contaminated with C. oncophora infective larvae. The persistent efficacy for preventing establishment of infection was assessed by the time to faecal egg excretion of C. oncophora eggs. It was found that the moxidectin treatment prevented infection for less than 10 days and the effect of doramectin lasted for 24 days. 相似文献

The persistence of the broad-spectrum antiparasitic activity of endectocide compounds relies on their disposition kinetics and pattern of plasma/tissues exchange in the host. This study evaluates the comparative plasma disposition kinetics of ivermectin (IVM), moxidectin (MXD) and doramectin (DRM) in cattle treated with commercially available injectable formulations. Twelve (12) parasite-free male Hereford calves (180–210 kg) grazing on pasture were allocated into three groups of four animals each. Animals in each group received either IVM (Ivomec 1%, MSD AGVET, Rahway, NJ, USA), MXD (Cydectin 1%, American Cyanamid, Wayne, NJ, USA) or DRM (Dectomax 1%, Pfizer Inc., New York, NY, USA) by subcutaneous injection at a dose of 200 μg/kg. Jugular blood samples were collected from 1 h up to 80 days post-treatment, and plasma extracted, derivatized and analysed by high performance liquid chromatography (HPLC) using fluorescence detection. The parent molecules were detected in plasma between 1 h and either 70 (DRM) or 80 (IVM and MXD) days post-treatment. The absorption of MXD from the site of injection was significantly faster (absorption half-life (t½ab) = 1.32 h) than those of IVM (t½ab= 39.2 h) and DRM (t½ab= 56.4 h). MXD peak plasma concentration (Cmax) was reached significantly earlier (8.00 h) compared to those of IVM and DRM (4–6 days post-treatment). There were no differences on Cmax values; the area under the concentration–time curve (AUC) was higher for IVM (459 ng.d/mL) and DRM (627 ng.d/mL) compared to that of MXD (217 ng.d/mL). The mean plasma residence time was longer for MXD (14.6 d) compared to IVM (7.35 d) and DRM (9.09 d). Unidentified metabolites were detected in plasma; they accounted for 5.75% (DRM), 8.50% (IVM) and 13.8% (MXD) of the total amount of their respective parent drugs recovered in plasma. The comparative plasma disposition kinetics of IVM, MXD and DRM in cattle, characterized over 80 days post-treatment under standardized experimental conditions, is reported for the first time. 相似文献
