Crowded uterine conditions were induced by unilateral hysterectomy-ovariectomy (UHO) in 42 gilts to determine the effect of recombinant porcine somatotropin on fetal and placental growth. Gilts were randomly assigned across three replicates to one of three treatments: Control (C; n = 14), daily injections of 1 mL saline from d 0 to 64 of gestation, Early (E; n = 12), 5 mg of rpST/d from d 0 to 30, followed by 1 mL saline from d 31 to 64, and Late (L; n = 16), 1 mL saline/d from d 0 to 29, followed by 5 mg of rpST/d from d 30 to 64 of gestation. Blood was collected from each gilt via jugular venipuncture at d 0 and every 15 d thereafter. Gilts were hysterectomized on d 65 of gestation. Length of placental attachment and fetal crown-rump length were measured. Placentas and fetuses were weighed. Placental length, wet weight, and dry weight were recorded. Treatment with rpST (either E or L) increased (P < 0.0001) maternal plasma IGF-I concentrations relative to controls. Treatment with rpST did not affect placental wet weight or placental DNA content. However, E and L treatments increased the percentage of placental protein (P = 0.01) and placental dry matter (P = 0.10) and increased contact area of uterine-placental interface (P = 0.01). Despite changes in placental composition and morphology, weights of fetuses collected from L-treated gilts did not differ from controls, whereas weights of fetuses collected from E-treated gilts tended to be less than controls (P < 0.06). Administration of rpST increased maternal IGF-I concentrations and placental surface area but failed to increase fetal growth in the UHO model. Therefore, mechanisms that are independent of maternal IGF-I or placental contact area may control early fetal growth under crowded uterine conditions.  相似文献

Two experiments were conducted to examine responses of gilts to treatment with and withdrawal of exogenous porcine somatotropin (PST). In Exp. 1, 36 prepubertal gilts (79.7 +/- .9 kg; 159.1 +/- .7 d) were allotted randomly to receive daily either 0 micrograms PST (C) or 70 micrograms PST/kg initial BW for either 21 (PST-3) or 42 d (PST-6). Gilts were examined for estrus daily by a mature boar starting on d 22 and continuing for up to 50 d. Gilts that expressed estrus were mated and removed from treatment. PST-treated gilts had higher ADG (P less than .01) and lower feed/gain (P less than .02) than C gilts. Following initiation of boar exposure, C gilts (mean interval to estrus = 2.0 d) exhibited estrus earlier than PST-3 (24.8 d) and PST-6 (24.0 d) gilts (P less than .07); however, only two C gilts were observed in estrus compared with six PST-3 and six PST-6 gilts. In Exp. 2, 40 prepubertal gilts (72.6 +/- 1.0 kg; 141.1 +/- .7 d) were allotted randomly to receive daily either 0 mg PST (C) or 5 mg PST for 30 d. On d 31, half the gilts were comingled with unfamiliar penmates and examined for estrus daily by a mature boar for up to 45 d. Estrual gilts were removed from treatment.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Seventy-two finishing pigs (initial weight = 57.6 kg) were utilized to determine the effects of porcine somatotropin (pST) and dietary lysine level on growth performance and carcass characteristics. Pigs were injected daily with 4 mg pST in the extensor muscle of the neck and fed either a pelleted corn-sesame meal diet (.6% lysine, 17.8% CP) or diets containing .8, 1.0, 1.2 or 1.4% lysine provided by additions of L-lysine.HCl. All diets were formulated to contain at least twice the required amounts of other amino acids. Control pigs received a placebo injection and the .6%-lysine diet. Increasing levels of dietary lysine resulted in increased ADG and improved feed conversion (quadratic, P less than .01) for pST-treated pigs. The calculated daily lysine intake was 16.6, 13.6, 19.6, 25.1, 29.6 and 33.6 g for the control and pST-treated pigs fed .6, .8, 1.0, 1.2 and 1.4% lysine, respectively, over the entire experiment. Breakpoint analysis indicated that cumulative ADG and feed conversion were optimized at 1.19 and 1.22% lysine, respectively. Longissimus muscle area and trimmed ham and loin weights increased as dietary lysine was increased among pST-treated pigs (quadratic, P less than .01). Breakpoint analysis indicated that 1.11% lysine maximized longissimus muscle area, whereas trimmed ham and loin weights were maximized at .91 and .98% lysine, respectively. Adjusted backfat thickness was not affected by dietary lysine, but pST-treated pigs had less backfat (P less than .05) than control pigs did. Percentage moisture of the longissimus muscle increased (linear, P less than .05), as did percentage CP (quadratic, P less than .05), whereas fat content decreased (linear, P less than .05) as lysine level increased. Similar trends in composition were observed for muscles of the ham (semimembranosus, semitendinosus, and biceps femoris). Shear-force values from the longissimus and semimembranosus were lowest for control pigs, but they increased as dietary lysine level increased among pST-treated pigs. Sensory panel evaluations indicated that juiciness and tenderness decreased (linear, P less than .05) as dietary lysine level increased. Plasma urea concentrations decreased linearly (P less than .01) on d 28 as lysine level increased, whereas plasma lysine and insulin were increased (quadratic, P less than .01). Plasma glucose and free fatty acid concentrations on d 28 tended to increase (quadratic, P less than .10) with increasing dietary lysine level.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

Effects of estradiol benzoate (EB) and zearalenone (Z) on luteal maintenance and plasma hormone concentrations were studied in 45 gilts. Gilts were allocated to receive either 20 mg Z, 2 mg EB or no treatment (C) on d 1 to 5 (T1), 6 to 10 (T2) or 11 to 15 (T3) of an estrous cycle (five per treatment). Onset of estrus was designated as d 0 of the estrous cycle. Zearalenone was added to the daily ration and EB was administered via an intramuscular injection. Blood samples were collected every 10 min over a 4-h period on the first 2 d prior to onset of treatment; the first, third and fifth days of treatment; and the first two and the fifth day after the end of the treatment periods. Gilts receiving EB and Z during T2 and T3 had longer (P less than .05) inter-estrous intervals than C gilts. The range in inter-estrous intervals for Z and EB treatments was 28 to 74 and 27 to 63 d, respectively. Mean plasma progesterone concentrations were elevated (P less than .05) during T2 and T3 in EB and Z-treated gilts when compared with C females. Estradiol benzoate treatment during T2 and T3 reduced (P less than .05) mean plasma luteinizing hormone (LH) concentrations more than C or Z treatments. Mean plasma concentrations of 13, 14-dihydro-, 15-keto-prostaglandin F2 alpha (PGFM) during T3 were higher (P less than .05) in C and Z gilts on d 13 and 15 post-estrus when compared with EB gilts.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Effects of recombinant porcine somatotropin (rpST) on metabolic rate were studied in two trials with 24 crossbred barrows (Yorkshire x Landrace) in each. The barrows weighed about 80 kg (SE within trials 2.2 kg) at the start of the measurements and in each trial 12 pigs received 4 mg of rpST and 12 received a placebo. The diet contained 2.57 Mcal NE/kg and 20% CP (about 1% lysine). Animals were fed approximately 2.8 times maintenance (280 kcal ME.kg-.75.d-1). Heat production (gaseous exchange of CO2 and O2) and activity were measured continuously. Heat production associated with activity was calculated from the regression of heat production on activity. Animals treated with rpST exceeded controls in rate of gain by 252 g/d (P less than .001) and in metabolic rate by 14.5 kcal.kg-.75.d-1 (P less than .01). The rpST treatment increased rectal (+ .2 degrees C) and surface (+ .8 degrees C) temperatures. Activity-related heat production in treated pigs was increased, but this was only partly related to the increase in metabolic rate with rpST. The daily patterns of total and activity-related heat production were similar between pigs in both experimental treatments.  相似文献

Recombinant bovine somatotropin (bST) was administered at 0, 10.3, or 20.6 mg per cow per day to 32 Holsteins for 38 wk. Fifteen currently treated cows had been treated in the previous lactation. Eighteen hematological variables and blood concentration of insulin-like growth factor I (IGF-I) were measured at five sample periods. The objectives of the study were to test the effects of bST treatments on hematological profiles and to relate blood IGF-I concentration to these variables. Results indicated little influence (P greater than .10) of previous bST treatments on hematological profiles measured in the current lactation. Current bST treatments, however, altered hematocrits (P = .10), mean corpuscular volume (P = .03), mean corpuscular hemoglobin (P = .009), and fraction of blood lymphocytes (P = .09). A week x bST treatment interaction also contributed to variation in hematocrit (P = .09), mean corpuscular hemoglobin (P = .05), fraction of neutrophils (P = .02), and fraction of lymphocytes (P = .04). Blood IGF-I concentration influenced fractions and counts of neutrophils (P = .06, .09), lymphocytes (P = .04, .02), and monocytes (P = .08, .05). Further in vivo and in vitro studies are warranted because this preliminary evidence suggests that bST and(or) IGF-I contribute to regulation of hematopoiesis in mature dairy cows.  相似文献

This study was conducted to determine the effect of daily recombinant porcine somatotropin (rpST) injection (0 or 120 micrograms/kg BW) and dietary CP level of the feed (14 or 26% CP) on pancreatic characteristics of growing pigs. Daily injection of rpST did not affect pancreatic weight (P = .885) but did decrease pancreatic amylase content (P = .005). The ratios of amylase:protein and amylase:trypsin were also lowered by daily rpST injection (P = .002 and P = .0002, respectively). There were protein x rpST and protein x rpST x sex interactions for the ratio of amylase:chymotrypsin. The CP content of the diet had a greater effect than the injection of rpST on pancreatic characteristics. Pigs consuming the 26% CP diet had significantly higher pancreatic weight (P = .003) and greater total pancreatic chymotrypsin (P = .006) than pigs consuming the 14% CP diet. The ratios of trypsin and chymotrypsin to DNA were also higher in pigs fed the 26% CP diet (P = .007 and P = .005, respectively). These responses were not influenced by sex. Recombinant porcine somatotropin seemed to have a slight effect on porcine pancreatic characteristics; however, dietary protein had a greater effect on pancreatic characteristics in market-weight hogs.  相似文献

To determine whether recombinant porcine somatotropin (rpST) alters reproduction, 40 crossbred gilts weighing 59.1 +/- .5 kg at 125 +/- 1 d of age were assigned randomly to an experiment arranged as a 2 x 2 factorial. Eight gilts were given daily injections of diluent until they reached 104 kg BW (DW), and eight received diluent injections until puberty (DP). Twelve gilts were given rpST (4 mg/d) until 104 kg BW (PW) and 12 were given rpST injections until puberty (PP). All gilts were individually fed on an ad libitum basis an 18% CP corn-soybean meal diet (1.2% lysine and 3.1 Mcal/kg of ME). Beginning at 5 mo of age, gilts were exposed 20 min daily to mature boars. Serum concentrations of progesterone were measured weekly from 5 to 8 mo of age to verify age of puberty. Gilts observed in pubertal estrus were mated to two different boars 10 h apart. At 47 +/- 1 d of gestation, gilts were slaughtered to assess fetal development. After 60 d of treatment, serum LH and FSH profiles were determined in blood samples drawn at 20-min intervals for 4 h from eight diluent- and eight rpST-treated gilts fitted with indwelling jugular catheters. By 28 d, feed intake, feed/gain, and blood urea nitrogen were decreased (P less than .005) by rpST. Treatments did not affect (P greater than .05) the proportion of gilts attaining first ovulation (DW = 6/6; DP = 10/10; PW = 7/9; PP = 14/14) or conception rate (DW = 5/6; DP = 7/10; PW = 4/6; PP = 11/12).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

An experiment was conducted to determine the effects of equine somatotropin on the reproductive axis of the stallion during the nonbreeding season. Adult stallions were treated with equine somatotropin (20 μg/kg body weight [BW]; n = 5) or saline (n = 4) daily for 21 days starting in January. During the last week of treatment, stallions were subjected to low- and high-dose injections of luteinizing hormone (LH), as well as low- and high-dose injections of gonadotropin-releasing hormone (GnRH) and thyrotropin-releasing hormone (TRH). Two months after the onset of somatotropin treatment, semen was collected from all stallions every other day for 14 days. Treatment with equine somatotropin increased (P < .001) daily IGF-1 concentrations but had no effect (P > .1) on concentrations of LH, follicle-stimulating hormone (FSH), or testosterone. The testosterone responses to injections of LH were similar (P > .1) between treatments. Likewise, the LH, FSH, prolactin, and testosterone responses to the injections of GnRH/TRH were similar (P > .1) between groups. At seminal collections, stallions treated with somatotropin exhibited greater volumes of gel-free semen (P < .01) and gel (P < .05) and had decreased time until ejaculation (P < .05). In conclusion, somatotropin treatment for 21 days may alter the long-term accessory gland contribution to seminal volume but does not appear to alter pituitary gonadotrope function or testicular testosterone secretion.  相似文献

The dose-dependent effects of porcine somatotropin (pST) on growth performance and composition of carcass gain were investigated in 150 growing pigs. The experiment involved two genotypes (barrows from the Pig Improvement Company [PIC] and a University of Nebraska [NEB] gene pool line) and two sexes (PIC barrows and boars). At 30 kg, pigs were randomly assigned within each genotype and sex subclass to receive daily i.m. injections of 50, 100, 150, or 200 micrograms of pST/kg BW or an equivalent volume of an excipient. A diet (3.5 Mcal of DE/kg) supplemented with crystalline amino acids and containing 22.5% CP was available on an ad libitum basis until pigs were slaughtered at approximately 90 kg live weight. Excipient-treated PIC barrows exhibited faster and more efficient growth (P less than .001) and a higher capacity for carcass protein accretion (P less than .001) but similar rates of lipid deposition compared to excipient-treated NEB barrows. Within the PIC genotype, control boars grew at a rate similar to that of barrows, but they were more efficient (P less than .05) and deposited more carcass protein (P less than .05) and less lipid (P less than .001). Carcass protein accretion rate increased (P less than .001) up to approximately 150 micrograms of pST.kg BW-1.d-1, whereas lipid deposition decreased (P less than .001) with each incremental dose of pST. Although differences between PIC boars and barrows for all criteria were negated with increasing pST dose, they were maintained between the two genotypes. Polynomial regressions suggested that a slightly higher pST dose was required to optimize the feed:gain ratio compared with rate of gain and that the dose (micrograms per kilogram BW per day) was a function of the genotype and sex (feed:gain: 185, 170, and 155; rate of gain: 155, 155, and 125 for NEB barrows, PIC barrows, and PIC boars, respectively).  相似文献

Treatment of pigs with recombinant porcine somatotropin (rpST) causes a marked increase in feed utilization with increased weight-gain over untreated controls. Physiological parameters such as creatinine clearance were increased by rpST treatment. Clearance of drugs eliminated by hepatic extraction, like indocyanine green (ICG), were also increased by rpST treatment. However, clearance of intravenous (i.v.)-dosed propranolol (PPL) was not affected by rpST treatment and data from oral (p.o.) - dosing was inconclusive because of the low bioavailability, probably because of a high first-pass effect. The very low oral bioavailability indicates that intestinal metabolism of PPL is probably quite high. Analysis of urinary metabolites indicated production of the two phenolic isomers, but there was no metabolite corresponding to N-dealkylase activity; although the latter metabolite could have been eliminated in the bile with subsequent fecal elimination. PPL was an excellent in vitro substrate for measuring hepatic DME activity in vitro ; two phenolic and one N-dealkylated metabolite were formed. The overall conclusions regarding this study must be that the effects of rpST on drug bioavailability and elimination were equivocal. As ICG and creatinine clearances were both increased significantly, one cannot rule out the probability that rpST would increase drug elimination in pigs as a result of increased hepatic uptake and/or renal clearance. One can only speculate that clearance of concurrently administered drugs would be increased. This would reduce residue levels, but it might also reduce efficacy.  相似文献

Effects of recombinant porcine somatotropin (rpST) on growth, feed intake, feed conversion, back-fat thickness and lean percentage were examined in growing Meishan pigs. The experiment comprised 42 barrows of which 20 were administered 14 mg rpST twice a week i.m., starting at 40 kg, the others received a placebo. Pigs were fed ad libitum a diet containing 9.2 MJ net energy and 156 g crude protein kg−1 and were slaughtered at 90 kg liveweight. From 40 to 90 kg liveweight, rpST effects were: daily gain +17.9%; feed intake −5.1%; feed conversion −17.5%; backfat thickness −29.8%; lean percentage +16.0%. The effects of rpST in Meishan are much larger than in a similar experiment with leaner western pigs. Development of synthetic breeds with Meishan in combination with the use of rpST in cross-bred fattening pigs may be a way to economically exploit the high fertility of Meishan.  相似文献

One hundred twenty crossbred beef steers averaging 377 kg were used in a 2 x 4 factorial experiment to determine the dose-response effects of recombinant bovine somatotropin (rbST) implants on growth performance and carcass characteristics. The implant dosages were 0 (sham), 40, 80, or 160 mg/wk administered s.c. in the tailhead region on a weekly basis throughout the experiment. The steers were fed a high-concentrate diet, which contained either a degradable (DP; soybean meal) or an escape (EP; corn gluten and blood meal) protein source. No dietary protein effect or dietary protein x rbST level interactions were detected. Recombinant bST decreased both DMI (P less than .10) and feed/gain (P less than .05) in a linear dose-dependent manner. Dosage of rbST did not significantly affect (P greater than .10) ADG or final weight of the steers. Recombinant bST decreased backfat depth (P less than .10), marbling score (P less than .05), and quality grade (P less than .10) and increased yield grade (P less than .10) in a linear dose-dependent manner. Soft tissue composition of the 9-10-11th rib section was altered (P less than .01) by rbST administration in a linear dose-dependent manner. The percentage of protein in the rib section was increased by 9.4% and fat was decreased by 11.8% at the 160 mg/wk rbST level compared with the sham-implanted steers. Recombinant bST did not affect (P greater than .10) dressing percentage, hot carcass weight, kidney, pelvic, and heart fat, or longissimus muscle area.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The soybean phytoestrogen genistein has a range of estrogenic actions demonstrated in various species; however, only limited research has been done to investigate its effects in swine. The objective of this study was to characterize the effects of a graded dose of genistein on estrogen-sensitive uterine and cervical tissues in ovariectomized gilts. Thirty-four postpubertal gilts were ovariectomized and assigned randomly to 1 of 6 treatment groups 15 d postovariectomy. Treatment groups received vehicle, estradiol benzoate (2 mg/d), or genistein (50, 100, 200, or 400 mg/d) via intramuscular injection at 12-h intervals for 10 d. Following the treatment period, gilts were euthanized, and uterine and cervical tissues were collected and processed for chemical or histological analysis. Uterine and cervical tissue mass, as indicated by wet, dry, and protein weights and total DNA content (expressed per 100 kg of BW), increased as the dosage of genistein increased (P < 0.001 for each regression). Uterine and cervical wet weights were increased by a dosage of 200 mg of genistein/d (P < 0.001 and P < 0.01, respectively) but not by 100 mg of genistein/d (P = 0.38 and P = 0.14, respectively) compared with those of control gilts. Height of epithelial cells lining the uterine glands and the lumen of uterus and cervix increased when gilts were treated with estradiol benzoate or 400 mg of genistein/d (P < 0.01). When the gilts were treated with estradiol benzoate or 400 mg of genistein/d, immunohistochemical staining demonstrated an increase in the percentage of cells that stained positive for progesterone receptor in the uterine glands and in the cells lining the vaginal cervix (P < 0.05). In gilts treated with 400 mg of genistein/d, the percentage of cells stained positive for proliferating cell nuclear antigen increased in the epithelium of the uterine glands, uterine lumen, and vaginal cervix (P < 0.05). Tissue growth was stimulated by genistein in a dosage-dependent manner, although no dosage of genistein induced a response as great as that of estradiol benzoate. Estrogen-sensitive tissues of the ovariectomized gilt, such as the cervix and uterus, are affected by injection of large dosages of the phytoestrogen genistein. The sensitivity of the uterus of the gilt to estrogenic substances makes it a potential model to examine the impact of environmental endocrine modulators on reproductive tissues.  相似文献

Two hundred and twenty-four pigs (112 boars, 112 gilts) housed in pens of seven pigs per pen were used in a 2 x 2 x 2 factorial design, with the factors of vaccination with a gonadotropin-releasing factor (GnRF) vaccine (Improvac; 0 or 2 mL at 13 and 17 wk of age), porcine somatotropin (pST; 0 or 5 mg/d from 17 wk of age), and gender. Pigs were weighed and feed intake was measured from 17 wk of age until slaughter at 21 wk of age. Body composition was estimated by dual-energy X-ray absorptiometry in two focus pigs per pen at 17 and 21 wk of age. Testes and ovary weights at slaughter were decreased by Improvac treatment (P < 0.001), but were not altered by pST treatment (P > 0.44). Daily gain was lower for gilts than boars (1,128 vs. 1,299 g/d, P < 0.001) and was increased by pST (1,172 vs. 1,255 g/d, P = 0.003) and Improvac (1,150 vs. 1,276 g/d, P < 0.001) treatments. Feed intake (as-fed basis) was lower in gilts than in boars (2,774 vs. 3,033 g/d, P = 0.002), was decreased by pST (3,037 vs. 2,770 g/ d, P = 0.002), and was increased by Improvac treatment (2,702 vs. 3,105 g/d, P < 0.001). As a result of the differences in feed intake and daily gain, feed conversion efficiency (gain:feed) was lower for gilts than for boars (0.403 vs. 0.427 P = 0.025), was improved by pST (0.385 vs. 0.452, P < 0.001), but was unchanged by Improvac treatment (0.423 vs. 0.410, P = 0.22). Carcass weight was lower in gilts than in boars (75.3 vs. 77.0 kg, P = 0.012), was unchanged by pST treatment (75.9 vs. 76.4 kg, P = 0.40), and was increased by Improvac treatment (75.1 vs. 77.2 kg, P = 0.003). Lean tissue deposition rate was lower in gilts than in boars (579 vs. 725 g/d, P < 0.001), was increased by pST (609 vs. 696 g/d, P < 0.001) and by Improvac treatment (623 vs. 682 g/d, P = 0.014). Fat deposition rate tended to be lower in gilts than in boars (214 vs. 247 g/d, P = 0.063), decreased by pST treatment (263 vs. 198 g/d, P < 0.001), and increased by Improvac treatment (197 vs. 264 g/d, P < 0.001). For pigs treated with both pST and Improvac, daily gain and lean tissue deposition rate was greater than for pigs that received either treatment alone, whereas fat deposition rate and feed intake did not differ from untreated control pigs. In conclusion, Improvac increased growth rate through increased lean and fat deposition, but concomitant use of Improvac and pST increased lean gain above either alone, while negating the increase in fat deposition in pigs treated with Improvac.  相似文献

One hundred sixty pigs were used in a 2 x 2 factorial design to compare the performance response to daily injection of porcine somatotropin (PST); (0 or 2 mg/d) in animals fed a 14% CP corn-soy diet (control, C) to those fed a diet with 10% added fat (F) and calorie:protein and lysine:protein ratios similar to that of the C diet. Treatments, assigned randomly to 20 pens (n = 5/treatment) of eight pigs each, were initiated at 90 kg body weight and continued for 28 d. Responses to PST and dietary fat were typical. These include improved gain and feed efficiency and decreased feed intake. The effects of dietary fat on intake and efficiency were accounted for largely by the difference in energy density of these diets. Across diets, PST treatment resulted in a 13% improvement in ADG (P less than .001), a 13% decrease in feed intake (P less than .0001) and a 22% improvement in efficiency (P less than .0001). Of particular interest were the additive (PST x diet interaction, P less than .2) effects of PST and dietary fat on gain in these animals. Pigs treated with PST that were fed the F diet had greater rates of gain than did PST-treated pigs fed the C diet (P less than .05). Treatment with PST increased ADG by 9% in pigs consuming the C diet vs 16% in pigs fed the F diet. Similarly, dietary fat resulted in 4 and 11% increases in ADG in pigs treated with 0 or 2 mg of PST/d.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The objective of this study was to examine the effects of somatotropin (ST) on porcine reproductive and metabolic statuses during early pregnancy. Four pregnant crossbred gilts received 6 mg of recombinant porcine somatotropin (rpST) daily from days 10 to 27 after artificial insemination while six pregnant gilts served as controls. Blood samples were taken on days 8, 10, 12, 14, 18, 22, and 27 prior to rpST injections (8:00 h) and subsequently at 9:00, 10:00, 12:00, 14:00, 16:00, 18:00, and 20:00 h. On all remaining days of treatment, samples were taken once daily before injections (8:00 h). The samples were assayed for the metabolic hormones: ST, insulin-like growth factor I (IGF-I), insulin, thyroxine (T4), triiodothyronine (T3), and cortisol; for metabolites: free fatty acids (FFA) and glucose; and for the reproductive hormones: luteinizing hormone (LH), progesterone, estradiol-17β, estrone sulfate, and prostaglandin F2. Delivery of rpST daily induced a 20- to 40-fold increase in plasma ST concentrations. Moreover, repeated administration of rpST resulted in a continuous increase in plasma IGF-I concentration (P<0.001), from 191.0±22.3–340.0±15.3 ng/mL 24 h after initial injection to 591.3±46.8 ng/mL after final injections. Mean serum insulin tended to be greater in rpST-treated gilts. Blood concentrations of T4 were reduced (P<0.05) from day 14 of gestation in treated gilts while T3 concentrations remained unchanged. Concentrations of both glucose and FFA were greater (P<0.01) and cortisol concentrations were unchanged in treated gilts. Changes in reproductive steroid hormones were minimally affected. Circulating progesterone (P=0.078), and estradiol-17β (P=0.087) concentrations tended to be lower in treated animals. These data show that treatment of pregnant gilts with rpST during early gestation mainly impacts metabolic rather than reproductive status.  相似文献

First-litter commercial cross-bred gilts were treated with levamisole (1.5, 2.5, or 3.5 mg/kg of body weight) weekly during the last 4 weeks of gestation, because similar treatment of dairy heifers had improved postpartum maternal health and neonatal survival. In the gilts, differences in reproductive performance were not found on the basis of pig survival at birth, pig survival at weaning, birth weight, or weaning weight. Also, differences between treated and control gilts were not found in response of circulating lymphocytes to mitogen stimulation (phytohemagglutinin A, concanavalin A, and pokeweed mitogen). In all gilts, the lymphocyte response to mitogen stimulation was decreased during the first week after farrowing.  相似文献
