The role of the gut in acquired resistance to Fasciola hepatica in the rat was assessed by comparison of the number of flukes recovered 4 and 10 weeks after oral or intraperitoneal challenge infection of male outbred Wistar rats previously infected by the oral or intraperitoneal routes.Previous infection given by either route generated signigicant protection against both oral and intraperitoneal challenge. The rats were resistant to Fasciola challenge in the presence of the primary infection or after its removal by anthelmintic treatment.It was concluded that passage of juvenile flukes through the gut was not essential for either the acquisition or the expression of acquired resistance to F. hepatica in the rat.  相似文献

The objective of the present study was to develop and evaluate a local vaccine (strain TPV-1) against Fowl pox (FP) in chickens. Two separate groups of chickens were vaccinated with FP vaccine through oral (coated on oiled rice) and wing web stab routes, respectively. The results showed that the haemagglutination-inhibition (HI) antibody titres in both vaccinated groups were comparable and significantly higher (P < 0.05) than the control chickens. It was further revealed that 14 days after vaccination HI GMT of ≥2 log2 was recorded in chickens vaccinated by oral and wing web stab routes whereas 35 days after vaccination the HI antibody titres reached 5.6 log2 and 6.3 log2, respectively. Moreover, in both groups the birds showed 100% protection against challenge virus at 35 days after vaccination. The findings from the present study have shown that oral route is equally effective as wing web stab route for vaccination of chickens against FP. However, the oral route can be used in mass vaccination of birds thus avoid catching individual birds for vaccination. It was noteworthy that strain TPV-1 virus could be propagated by a simple allantoic cavity inoculation and harvesting of allantoic fluid where it survived exposure at 57°C for 2 hours. If the oral vaccination technique is optimized it may be used in controlling FP in scavenging and feral chickens. In conclusion, the present study has shown that FP vaccine (strain TPV-1) was safe, thermostable, immunogenic and efficacious in vaccinated chickens.  相似文献

Immunity to colonization of the respiratory tract by Mycoplasma bovis (formerly Mycoplasma agalactiae subsp. bovis was induced in calves by inoculation of formalin inactivated organisms. Animals inoculated intramuscularly and then intratracheally with inactivated mycoplasmas had significantly fewer M. bovis in their lungs, compared with non-vaccinated animals, 3 weeks after intratracheal challenge with viable organisms. In contrast, there was no significant difference in the numbers of M. bovis isolated from the lungs of control animals and of calves given two intramuscular inoculations of inactivated organisms. These results indicate that stimulation of the local immune system is important in the development of resistance to M. bovis respiratory infection following vaccination with inactivated organisms.  相似文献

All mammals are colonised by Escherichia coli generally at birth and these organisms become part of their intestinal flora for the rest of their lives. New types are acquired generally by an oral route. Some E coli are pathogenic and some may have a far more enhanced ability to colonise the human intestine than most others. Recently enterohaemorrhagic E coli have emerged. They can cause a number of intestinal illnesses in humans including bloody diarrhoea and haemolytic uraemic syndrome. These organisms produce a number of virulence factors particularly the Shiga-like toxins (verotoxins). The intestines of animals may be the reservoir of these organisms for human infection, and cattle particularly have been shown to harbour them. Food, especially undercooked meat products, have been associated with a number of outbreaks throughout the world. While a certain serotype O157.H7 has been associated with many outbreaks throughout the world, other serotypes, particularly 0111.H-, have also been reported. This latter serotype appears to be more common in Australia.  相似文献

The development of oral and intraperitoneal infections with Fasciola hepatica in young and old rats showed that the gut was involved in the expression of age resistance. The role of the gut was assessed by comparison of the number of flukes recovered 4 and 10 weeks after oral or intraperitoneal infection of 2-, 6-, 15-, and 35-week-old male Wistar rats and 6- and 15-week-old male DA rats with encysted metacarcariae of F. hepatica.Rats of both strains behaved similarly in their response to F. hepatica infection. For both routes of infection the number of flukes recovered decreased as host age increased. In 22- and 6-week-old rats equal numbers of flukes were recovered at 4 and 10 weeks after oral or intraperitoneal infection. In 15-week-old rats, fluke burdens 4 weeks after infection were significantly greater following intraperitoneal infection than oral infection. A significant loss of flukes from the intraperitoneal infection of 15-week-old Wistar rats occured between 4 and 10 weeks after infection. In 35-week-old Wistar rats there was no significant effect for route or age of infection.As intraperitoneal infection (to by-pass rejection at the gut level) only partially eliminates the age response, additional age related mechanism(s), able to reject flukes at some time after they have entered the peritoneal cavity, must be operative in the peritoneal cavity and/or the liver.  相似文献

1. By comparison with birds examined 1 d after slaughter, those held uneviscerated at 4 °C for a further 7 d showed a statistically‐significant difference in flavour (P < 0·01) when examined by a trained taste panel and the difference increased progressively up to 24 d when the experiment ended.

2. Changes in the composition of the microflora in the small intestine and caecum were investigated because of previous evidence that meat flavour development may be influenced by the metabolic activities of intestinal organisms. The only marked effect of storage was a decline in the numbers of lactose‐positive coliform bacteria in the small intestine; there was no clear evidence of any microbial growth in the gut.

3. Although birds examined at day 8 by a sensory panel or by means of a tenderometer were slightly more tender than those tested at day 1, the differences were not statistically significant; nor were there any significant changes in texture during the remainder of the storage period. Possible reasons for the lack of any marked tenderisation are discussed.

A study was undertaken on weaned 4–5 month old farmed red deer to test the efficacy of moxidectin and abamectin anthelmintics, given by three different routes of administration, compared with an untreated control. Faecal samples were collected on days 0, 7 and 14 for a faecal egg count reduction test (FECRT), blood samples were collected on days 0, 0.5, 1, 2, 3, 5, 7, 10 and 14 for pharmacokinetics, and the deer were killed on days 14 or 15 for total nematode count.The control group averaged 1264 adult Ostertagia-type nematode parasite species and treatment efficacy was 77.4% for moxidectin injection, 26% for oral moxidectin and 27.6% for pour-on moxidectin, while the treatment efficacy was 72.4% for abamectin injection, 70.1% for oral abamectin (Hi-Mineral) and 34.1% for pour-on abamectin. Both moxidectin and abamectin injections were significantly more efficacious than their equivalent pour-ons. There was a significant difference in efficacy between oral abamectin (Hi-Mineral) and oral moxidectin (P < 0.01).The control group averaged 2956 adult lungworm (Dictyocaulus eckerti) and 50 Oesophagostomum venulosum in the large intestine and treatment efficacy against these nematodes was 100% for all treatments. There were negligible numbers of other gastro-intestinal nematodes.At slaughter, there was a significant correlation (P = 0.02) between FEC and Ostertagia-type nematodes in the untreated controls. Relatively few eggs were found in faeces from treated animals at 7 and 14 days post-treatment despite significant worm burdens in all six treatment groups, suggesting egg-laying suppression in resistant nematodes, and all three different FECRT calculations tended to overestimate the efficacy of the treatments compared with actual nematode counts.Peak plasma concentrations (Cmax) for both actives were measured 12 h after treatment for injection and oral and at 5 days for pour-on. Cmax (ng/ml) for moxidectin injection, oral and pour-on were 71.8, 8.3 and 0.4, respectively, and for abamectin injection, oral and pour-on were 62.1, 30.3 and 10.0, respectively. Area under the curve (AUC) estimates for moxidectin injection, oral and pour-on were 106.6, 12.9 and 6.1, respectively, and for abamectin injection, oral and pour-on were 162.7, 57.5 and 74.3, respectively.The results demonstrate that significant anthelmintic resistance to moxidectin and abamectin is present on this deer farm. However, the injection was the most effective route of administration in young deer for both anthelmintics, although <80% efficacious. We conclude that the FECRT is unreliable in deer when anthelmintic resistance is present.  相似文献

Summary

Following EDS'76 virus (BC14 virus) infection of breeder chickens by the conjunctival route, vertical transmission occurred in the first week after infection. In the progeny which had been infected with EDS'76 virus by the vertical route, increasing haemagglutination inhibiting (HI) litres to BC14 virus and increasing numbers of birds with HI litres were observed from 3 weeks to 15 weeks of age. Sixty‐one per cent of the hens and 77 per cent of the cocks had 2log HI BC14 virus litres exceeding 4 at an age of 15 weeks.

Some birds which had been serologically negative throughout the rearing period, seroconverted between 25 and 28 weeks of age. This phenomenon occurred in hens as well as in cocks. Simulation of stress twice during the laying period by injection of corticosteroid hormone did not increase the number of birds serologically positive to EDS'76 virus.

EDS'76 was observed in the group of hens that was vertically infected, since egg production was significantly depressed between 28 and 34 weeks of age. Probably this was mainly the result of a production drop in the hens showing seroconversion at 27 or 28 weeks of age.

In the group of fowl vertically infected with EDS'76 virus, serologically positive birds appeared to be protected for the greater part to BC14 virus challenge at 50 weeks of age, while negative birds seemed to be fully susceptible. Chicks hatched from eggs collected in the third and fourth week after infection of the dams had maternal antibodies. Fertility and hatchability of apparently normally shelled eggs seemed not to be affected after BC14 virus infection of the dams. Intensive contact with contaminated faeces is probably an indispensable condition for lateral transmission of the virus.  相似文献

Experimental infection with Salmonella stanley was produced by oral, intravenous and intramuscular routes in day-old chicks. The earliest evidence of the presence of the organisms was in duodenal mucosa six hours after oral infection. Following oral infection the organisms were detected in the duodenum from six hours to five days, in the caecum from 12 hours to nine days, liver, spleen and blood from 24 hours to seven days. The resistance to infection was found to be significant after 10 days old, but not up to six days old. The work confirmed that the survival time of birds given S stanley by the intravenous or intramuscular routes was inversely proportional to the dose up to a maximum beyond which the survival time was not further decreased by dose increase. The presence of S stanley in tissues and blood was detected by isolation and by the fluorescent antibody technique.  相似文献

Necrotic enteritis is a complex disease condition of broiler chickens, commercial layer pullets, and turkeys and requires the presence of a toxigenic strain of Clostridium perfringens, alteration of bird diets, and damage to the intestinal epithelium. All of these alone, but especially in combination, result in significant alterations of the intestinal microflora. The intestinal microflora is part of a complex ecosystem that is involved in augmenting intestinal development, immune surveillance, and competitive exclusion against pathogenic organisms. However, when the microflora fails to protect the mucosa from pathogens, antimicrobials are used to reduce the numbers of pathogenic organisms or to prevent their colonization of the intestine. We were interested in studying the effects of monensin on the bacterial community within the ileum of chickens because ionophore antimicrobials are commonly used to prevent Eimeria infections in broilers. We used two 16S ribosomal DNA community analysis protocols, terminal restriction fragment length polymorphism analysis combined with 16S rDNA clone libraries. These methods showed that monensin caused significant alterations in the microbial community structure of the ileum. Whereas the ileal bacterial community of control birds primarily consisted of lactobacilli, monensin-treated birds had communities rich in clostridia. None of the birds exhibited signs of intestinal disease or mucosal lesions, which suggested that the clostridia were avirulent. Although the therapeutic benefits of monensin may largely result from its anticoccidial effects, its ability to foster a competitively exclusive bacterial community may contribute to the intestinal health of birds, thus preventing the development of necrotic enteritis.  相似文献

The immunogenicity of a sonicated extract (SE) and of outer membrane proteins (OMP) of Salmonella enteritidis was tested in birds of about 8 weeks of age. The dose, route of vaccination and the adjuvant used varied in different groups of birds. Two vaccine doses with or without adjuvant were given parenterally or orally 3 weeks apart. OMP vaccines gave significantly higher antibody titres than SE vaccines, as indicated by ELISA. The vaccines adjuvanted with oil produced higher antibody titres than those without any adjuvant. A dose of 1 mg of vaccine produced higher antibody titres than 0.5 mg of vaccine. Adjuvanted vaccine given subcutaneously elicited higher antibody responses than oral vaccines given without adjuvant. The birds were challenged with virulent S. enteritidis organisms at the end of the second week after a booster dose. None of the birds given 1 mg of OMP vaccine subcutaneously shed the organisms when tested by culturing cloacal swabs, although a few birds vaccinated with 0.5 mg of OMP vaccine did so. In general, adjuvanted OMP vaccines gave better protection than SE vaccines.  相似文献

Twenty-five day-old White Leghorn chickens were each infected orally with 500 (Group I), 1000 (Group II) and 2000 (Group III) infective eggs of Ascaridia galli and were killed 30 days after the infection. A high percentage of the infecting dose (6.5%) established as adult worms in the intestine of chickens receiving the lowest level of primary infection, but as the amount of primary infection given to birds increased, there was a significant fall in the percentage establishment of adult worms in the intestine. A similar pattern of worm establishment was shown by chickens of the same age and receiving similar levels of primary infections, but which were treated with betamethasone at a dose of 2 mg/kg body weight commencing 5 days before and continuing up to 15 days after the infection. Betamethasone-treated birds, however, showed more establishment of worms in the intestine, but lower weight gains in comparison to the birds which were not treated. Different levels of primary infections given, with or without treatment with betamethasone, had no effect on the sex ratio of the resultant male/female worm populations, which became established in almost equal numbers in the intestine of chickens.  相似文献

There was no difference in the establishment or rate of development of Oesophagostomum columbianum in young lambs after infective larvae were administered either orally or by injection directly into the rumen. However, the linear distributions in the intestine of encysted third-stage larvae differed according to the route of infection. The distributions of fourth-stage larvae, after migration to the large intestine, did not differ. It is suggested that the distribution of parasitic third-stage O. columbianum is host-dominated, depending on the site of exsheathment and the rate of passage of ingesta, but that of fourth-stage and adult worms involves active site selection by the parasite.  相似文献

The effects of simulatenous infection of pigs with Oesophagostomum spp. and Trichinella spiralis on the interpretation of the Enzyme-Linked Immunosorbent Assay (ELISA) for trichinosis were examined. Extinction values were observed from four groups of pigs. The first group acted as uninfected controls, the second was infected only with Oesophagostomum spp., the third with T. spiralis alone and the fourth by both nematodes.It was found that the pigs infected with T. spiralis could be differentiated from the others, but that those infected with both species had lower extinction values than the group with T. spiralis alone. The differences may be related to the numbers of T. spiralis larvae able to establish and develop into adults in the small intestine of the host. Those infected with Oesophagostomum spp. alone showed no rise in extinction values, and it was concluded that there was no cross-reaction in the ELISA between thos pecies and T. spiralis antigen.  相似文献

Day-of-hatch broiler chicks housed in isolation units were each given, by oral gavage, 0.1 ml of Mucosal Starter Culture (MSC) or saline control. Each of the treated and control chicks was subsequently given a composite culture of three strains of bacitracin-resistant Clostridium perfringens (Cp) previously isolated from chickens with symptoms of necrotic enteritis. Some chicks were maintained on a corn-based diet provided ad libitum. Others were given the feed supplemented with 50% rye (a predisposing factor for necrotic enteritis). At 7, 14, and 21 days after receiving Cp, chicks were euthanatized, and cecal contents were diluted and plated on selective agar containing bacitracin. For chicks on corn feed, Cp numbers were similar in control birds and birds given MSC in three of four trials. In two of the trials that demonstrated no effect of MSC on Cp numbers, enterotoxin presence was determined. The number of birds with detectable Cp enterotoxin in their small intestine and the mean toxin levels were lower in the MSC-treated birds. In a fourth trial with birds on corn-based feed, mean Cp numbers and the number of Cp-positive birds were lower in the MSC-treated birds. For the two trials involving chickens on rye-supplemented feed, Cp numbers and the percentage of Cp-positive birds were significantly reduced in MSC-treated birds compared with control birds. Enterotoxin in birds receiving the 50% rye diet was at low levels or not detected in control and MSC-treated birds. Results suggest that MSC may reduce intestinal proliferation of Cp, a causative agent of necrotic enteritis in poultry and of foodborne disease in humans.  相似文献

Experimental infections of Salmonella typhimurium, S. agona, S. enteritidis or S. infantis were studied in chicks infected with Eimeria tenella. In all experiments, 4-d old birds were given 5 daily oral doses of approximately 10(4) salmonella organisms per bird. One day before this inoculation one group of birds received a single dose, in the range 1 X 10(4) to 4 X 10(4) sporulated oocysts of E. tenella per os whilst the other group was not inoculated and served as the control. Chicks were examined post morten 7, 10 or 14 d after receiving coccidia. With S. typhimurium infection, the number of salmonellas in the caeca of the E. tenella-infected birds was greater than in those infected only with salmonella. With S. agona and S. enteritidis, the counts of salmonella in the caeca and the numbers of birds positive for salmonella in the caeca and liver were greater in the E. tenella-infected birds. The rate of infection of S. infantis was not increased by caecal coccidiosis.  相似文献

A feeding trial was conducted to evaluate the effects of mannanoligosaccharide (MOS) on the growth performance, nutrient digestibility and gut development of broilers given a corn or a wheat-based diet over a 21-day experimental period. Dietary MOS improved the growth performance of birds given the wheat-based diet compared to that of birds given the corn-based diet during 7–21 days of age. In line with this, the ileal digestibility of starch was increased by MOS at 21 days of age. The addition of MOS modulated the development of gut microflora. From day 7 to day 21, the numbers of mucosa-associated coliforms along the small intestine were decreased; whereas the numbers of mucosa-associated lactobacilli were increased by MOS, regardless of the cereal type in the diets. Dietary MOS also reduced the counts of coliforms and Clostridium perfringens in the caeca of birds by 21 days of age. Villus height at the jejunum was not affected by MOS but the crypt depth and the muscularis thickness were reduced. The specific activities of maltase and alkaline phosphatase were increased in birds given the MOS-supplemented diet; whereas the development of leucine aminopeptidase was delayed by MOS. All these changes in the mucosal morphology and function were dependent on the type of cereal and/or the age of the birds.  相似文献
