This study evaluated P. edulis f. edulis intraspecific variation at DNA level using AFLPs and SSRs. About 60 specimens were collected from Colombian commercial plantations in the departments of Antioquia, Boyacá, Cundinamarca, Huila, Quindío and Tolima. Ten specimens were also included from Corpoica’s La Selva research center Passiflora germplasm bank. A non-commercial purple passion fruit plant and three species from the Passiflora genus (Passiflora maliformis, Passiflora edulis f. flavicarpa and Passiflora ligularis) were included as controls. The six most informative ones were selected from the initial screening of 49 different AFLP primer combinations; 52–92 DNA fragments for each combination in each genotype (total 419 fragments) were obtained by using these six combinations. 17 SSR primers reported in P. edulis f. flavicarpa and Passiflora alata were tested on purple passion fruit. Eight of them were successfully amplified but none showed polymorphism. Relationships among individuals were assessed on AFLP data using cluster analysis by Dice coefficient, UPGMA algorithm and the multiple correspondence analysis ordination method based on Greenacre measurement. The similarity coefficients ranged from 0.75 to 1.00 giving an average of 0.96 within the different purple passion fruit materials, showing low genetic variability for the purple passion fruit material used in this study.  相似文献

The flavonoids present in passion fruit (Passiflora edulis) leaves were identified by a high-performance liquid chromatography-diode array detection-tandem mass spectrometry (HPLC-DAD-MS/MS) method. Sixteen apigenin or luteolin derivatives were characterized, which included four mono-C-glycosyl, eight O-glycosyl- C-glycosyl, and four O-glycosyl derivatives. With the exceptions of C-hexosyl luteolin and C-hexosyl apigenin, all the compounds exhibited a deoxyhexose moiety. Moreover, the uncommon C-deoxyhexosyl derivatives of luteolin and apigenin have been identified for first time in P. edulis by HPLC-DAD-MS/MS. The antioxidative capacity of passion fruit leaves was checked against DPPH radical and several reactive oxygen species (superoxide radical, hydroxyl radical, and hypochlorous acid), revealing it to be concentration-dependent, although a pro-oxidant effect was noticed for hydroxyl radical.  相似文献

Stir bar sorptive extraction (SBSE) in combination with GC-ECD/FPD analysis is here applied to the determination of the residues of 11 pesticides (hexachlorobenzene, lindane, chlorothalonil, parathion methyl, parathion ethyl, fenitrothion, malathion, dieldrin, alpha- and beta-endosulfan, and tetradifon) in herbal teas prepared with Passiflora alata Dryander spiked leaves. The method was optimized using spiked herbal teas in a range from 0.05 to 1 pg/microL for organochlorine pesticides and from 0.15 to 3 pg/microL for organophosphorus pesticides. The method is reproducible and repeatable with recoveries calculated from herbal teas prepared with spiked plant material versus spiked herbal teas, varying from about 30% for tetradifon to about 90% for parathion methyl and malathion. The limits of quantitation (LOQs) ranged from 0.017 pg/microL for lindane to 0.117 pg/microL for malathion.  相似文献

A new polysaccharide with a high molecular weight (greater than 1 x 106 Da) was extracted and characterized from the peels of Passiflora liguralis (granadilla) fruits. Chemical composition of the biopolymer, performed by using a high pressure anion exchange-pulsed amperometric detector (HPAE-PAD), showed the presence of six different sugar residues: xylose, glucose, galactose, galactosamine, an unknown component, and fucose in the relative ratio of 1:0.5:0.2:0.06:0.05:trace. The optical rotation of this xyloglucan was [alpha](D)(25) degrees C = -186.42 (concentration of 1.4 mg/mL of H(2)O), and the viscosity was dependent on the concentration and pH, showing a maximum value of 1.4 eta at a concentration of 3% in distilled water and a maximum value of 7.0 eta in citrate buffer solution. Thermogravimetric analysis indicated that this biopolymer was very stable at high temperatures, showing a degradation temperature at 280 degrees C. The characterization of the polysaccharide was also investigated by spectroscopic methods (1H NMR and IR) pointing out the complexity of this biopolymer and the presence of sugar residues in alpha-manno, alpha-gluco-galacto, and beta-gluco-galacto configurations. The formation of a biodegradable film using this novel xyloglucan was reported, and the anticytotoxic activity of the polysaccharide was studied in a brine shrimp bioassay. Considerable antioxidant activity (Trolox equivalent antioxidant capacity (TEAC) value of 0.32 microM/mg fresh product) was noted in the lipophilic extracts of Passiflora liguralis fruits, indicating, in this fruit, an alternative source of bioactive compounds.  相似文献

The Caatinga is a unique biome that encompasses nearly 11% of Brazil's territory and contains diverse vegetation composed of trees and bushes that have been scarcely studied from the economic perspective. In this context, Passiflora setacea may constitute a strategic alternative for agriculture because it produces fruit that has commercial potential and is tolerant to some diseases. Some species of Passiflora show more robust growth when associated with arbuscular mycorrhizal fungi (AMF) and are dependent on mycorrhization. In this study, the response of P. setacea to phosphate (P) fertilization and mycorrhization with Claroideoglomus etunicatum was evaluated. The experimental design included eight replicates of four randomized treatments: non-inoculated plants (NI), plants fertilized with phosphorus (P), mycorrhized plants (AMF), and plants that were both fertilized and mycorrhized (P+AMF). The plant height, leaf number, leaf area, fresh biomass (FB), and dry biomass (DB) of the shoots and roots, nutrient content, mycorrhizal colonization (MC), and the number of glomerospores (NG) in the rhizosphere were evaluated. The MC and the NG were reduced by phosphate fertilization. The development of the mycorrhized plants was significantly better than the NI and P treated plants for all of the variables tested. Inoculation with C. etunicatum promoted the growth of P. setacea seedlings even in the absence of phosphate fertilization, reducing production costs and strengthening the potential of P. setacea as a viable agricultural alternative for the semiarid region.  相似文献

Volatile organosulfur compounds (VOSCs) are high impact aroma chemicals characteristic of tropical fruits which are active as both free thiols and the respective thioesters. Using a simple and sensitive colorimetric enzyme assay, a thioesterase activity toward VOSCs has been identified in ripening purple passion fruit ( Passiflora edulis Sims). The assay was based on determining the release of free thiols from 2-methyl-3-furanthiol acetate using Ellman's reagent. The major thioesterase in the fruit was found to be a wall-bound protein in the mesocarp. The extracted enzyme activity was purified 150-fold and shown to be associated with a 43 kDa monomeric serine hydrolase which was selectively labeled with a fluorophosphonate suicide probe. MS-MS sequencing identified the thioesterase as a class 13 glycoside hydrolase, most similar to pectin acetylesterase, an enzyme involved in cell wall modifications in the peel of a number of fruit. Our results suggest that cell wall hydrolases in tropical fruit may have additional useful roles in biotransforming VOSCs.  相似文献

Pot and field experiments were conducted to (1) evaluate bioavailability of copper (Cu), lead (Pb), and zinc (Zn) in contaminated soil and phytoremediation potential by in situ plants, B. pilosa var. radiate and Passiflora foetida var. hispida, as inoculated with arbuscular mycorrhizal (AM) fungi, and (2) compare the results of pot and field experiments. The B. pilosa var. radiate plant inoculated with AM fungi had significantly greater Cu concentrations in the shoots and roots than noninoculated plants. Passiflora foetida var. hispida plant inoculated with AM fungi also had significantly greater Cu and Pb concentrations in the roots than noninoculated plants. As the root dry weight of Passiflora foetida var. hispida inoculated with AM fungi dramatically increased, the root Cu, Pb, and Zn content of Passiflora foetida var. hispida inoculated with AM fungi increased by 9–14 times, as compared with the noninoculated plants. The AM fungi have potential to either promote plant growth or increase heavy‐metal accumulation. The values of element translocation proportion from root to shoot were Zn > Cu > Pb for both plant species in pot and field experiments. For both plant species, the results of the pot and field experiments were significantly different. The concentration values of the pot experiment were greater in comparison to the field experiment, and some values were significantly greater than those in the field experiment.  相似文献

Piceatannol is present in passion fruit (Passiflora edulis) seeds in high amounts. In this study, we isolated the second major polyphenolic compound of passion fruit seeds and identified it as scirpusin B, which is a dimer of piceatannol. We investigated the antioxidant activities and vasorelaxing effects of these polyphenols. Their antioxidant effects were measured using an in vitro 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay, and their vasorelaxant effects were determined ex vivo in rat thoracic aorta. Both polyphenolic compounds exhibited potent antioxidant activities and significant vasorelaxant effects in endothelium-intact aortas. More specifically, scirpusin B exerted a greater antioxidant activity and vasorelaxant effect compared with that of piceatannol. Additionally, the vasorelaxation effects of the compounds were induced via the NO derived from the endothelium. This study provides the possibility that polyphenols in passion fruit seeds are effective against cardiovascular diseases (CVDs).  相似文献

Yellow passion fruit juice (PFJ, Passiflora edulis f. flavicarpa) is an important component of many tropical fruit beverages, but limited data exist on its antioxidant chemical composition and stability during processing and storage. PFJ fortified with ascorbic acid (450 mg/L) and sucrose (10%) was compared to a nonfortified control, and each was evaluated with and without vacuum deaeration to remove dissolved oxygen. Following pasteurization, juices were stored for 28 days at 37 degrees C to accentuate physicochemical changes. Pasteurization (85 degrees C for 30 min) resulted in minor changes to physicochemical attributes, but appreciable changes occurred during storage that resulted in termination of the study after 28 days. Oxygen control strategies proved to be ineffective for quality retention and indicated oxygen-independent reactions affecting juice color, phytochemical content, and antioxidant activity. Ascorbic acid and sucrose fortification had an overall preservation effect on total carotenoids, the former resulting in hyperchromic shifts in absorbance, indicating their chemoprotection. Pasteurization resulted in a 25% loss in l-ascorbic acid, which was completely destroyed after 14 days of storage; losses coincided with increased juice browning and formation of 5-hydroxymethylfurfural. Numerous polyphenolics were present in PFJ, and 16 of them were tentatively characterized on the basis of spectral similarities to known standards. Individually, polyphenolics increased during pasteurization, only to decline during storage at elevated temperatures. Antioxidant activity was measured in PFJ and in two subfractions (hydrophilic and lipophilic) after processing and storage, but antioxidant values were nonadditive. A significant chemical interaction affecting antioxidant capacity was found for hydrophilic juice components, but none was observed in the presence of lipophilic phytochemicals. Physicochemical attributes and overall quality of PFJ were retained following pasteurization but were significantly impacted by degradative reactions during accelerated storage.  相似文献

ABSTRACT

Salinity is one of the most important abiotic stresses limiting agricultural production worldwide. Here, we investigated the biometric, physiological and anatomical changes of Passiflora edulis plants propagated from seeds, cuttings and grafts and exposed to salt stress (0.3 – control and 6.3 dS m?1). Saline stress significantly reduced plant height, the number of leaves, total chlorophyll, stomatal conductance, shoot dry weight, total dry weight, total water consumption and root volume. Among the propagation methods evaluated, plants propagated from seeds were more sensitive to salinity, while plants propagated from cuttings showed smaller alterations when subjected to salinity. Similar behavior was observed for anatomical characteristics, since plants propagated from seeds presented more pronounced anatomical alterations, such as greater thicknesses of the leaf mesophyll, upper epidermis, lower epidermis, palisade parenchyma and spongy parenchyma and also was observed presence of calcium oxalate crystals in the form of druses in greater quantity in the root cortex, in comparison to plants propagated from cuttings. The approach used in this study allowed identifying the type of yellow passion fruit propagation (cuttings) that was best for physiological and anatomical adaptation to salt stress and will be potentially useful in programs for improvement of the crop.  相似文献

Isozyme variation was studied in 87 plants from 32 cultivated and wild accessions of banana passion fruit (P. tripartita var. mollissima, P. tripartita var. tripartita, P. tarminiana, and P. mixta), rosy passion fruit (P. cumbalensis), tin-tin (P. pinnatistipula), gulián (P. ampullacea), P. antioquensis, P. bracteosa, and P. manicata, from the Andes of Venezuela, Colombia, and Ecuador. Six polymorphic enzyme systems (IDH, PGDH, PGM, DIA, PRX, and ACP) revealed 31 zymotypes characterized by the presence or absence of 31 electromorphs. Cluster analysis separated clearly the accessions of P. tarminiana, P. tripartita, P. mixta, and P. cumbalensis from the less typical species of subgenus Tacsonia, which is consistent with morphological evidence. P. mixta showed the highest intraspecific variation and the closest affinity with P. tripartita. The accessions of these two species formed two clusters, one dominated by Colombian genotypes and the other dominated by Ecuadorian genotypes. One of the P. tripartita var. mollissima accessions clustered close to P. tarminiana accessions. The affinity between these three species is particularly interesting for conservation and use of banana passion fruit genetic resources. All the other species formed monospecific clusters.  相似文献

In Mexico and all over the world, the number of studies on the species of the Jatropha genus has increased because of the use of its seed oil to produce biodiesel. However, the knowledge of the taxonomy, distribution, and ethnobotany of these and related species is incomplete. This article presents the distribution of the genus in Mexico according to its internationally accepted taxonomic identity. The distribution is linked to environmental variables such as elevation, climate type, soil, and soil moisture patterns. In addition to showing the current taxonomical knowledge in Mexico, this article discusses its biogeography, its traditional uses and the research lines to follow in further study of the genus. The distribution results show that some species are broadly adapted; as a consequence, they are present in many different environments. Species such as J. dioica, J. curcas, J. cordata, J. cinerea and J. gaumeri are distributed in areas with well defined environmental conditions. However, the distribution of species such as J. riojae has not yet been ascertained. Three areas with high species richness were identified, and they are very important for the study and conservation of the genus. Mexico is a global center of diversity for this genus. Food and medicinal uses have been reported for some species. This article concludes with recommendations for further study to improve the knowledge of this genus in Mexico.  相似文献

Traffic of plants and animals of wild origin is a major threat to biological diversity creating a need for legal protection of traded species. Epiphytic vascular plants are a diverse guild which provides the horticultural market with many species. Many epiphytes are under protection intended to stop illegal trade at international and local levels. To measure the richness and volume of the epiphytes that are traded illegally on a local level, we monitored an illegal sale point in Xalapa, Veracruz, Mexico for 85 weeks. We recorded 27 traders, 207 species and 7598 plants. Nineteen species were known for Mexico but not native to Veracruz; two others had been collected in Veracruz but had never been reported before for Mexico. About 25% of epiphyte richness in Veracruz and 47% of the State’s epiphytic orchids are traded illegally. Most of the species traded could have been harvested in lower montane cloud forest (105 species), however all vegetation types of central Veracruz are potential epiphyte sources. Twenty-seven species protected by Mexican law were traded, along with 41 species endemic to Mexico and six endemic to Veracruz. We concluded that on a local level, the richness and volume of epiphytes illegally traded are high; in fact, the volume of orchids traded equals the annual average volume of legal Mexican orchid exports. Illegal epiphyte trade occurs worldwide, making necessary crucial increases of protective measures and the development of sustainable ways of harvesting epiphytes.  相似文献

Geographic location data for 383 accessions of five Passiflora species from five Andean countries (P. cumbalensis, P. mixta, P. tripartita var. mollissima, P. natistipula, and P. manicata) were linked to interpolated continental surfaces of monthly mean rainfall, monthly mean temperature, and monthly mean diurnal temperature range. This permitted us to identify the potential distribution of each species and document its climatic adaptation. Maps are presented showing regions where the climate is similar to that in areas where each species has been found, but from which no collections have yet been made, thus representing possible geographical gaps in collections. Some species showed evidence of intra-specific variation in their climatic adaptation. The study of Passiflora diversity in Andean countries continues and these maps based on passport data are expected to be useful tools for the planning of both ex situ and in situ conservation activities.  相似文献

A considerable number of bird species depend on tree cavities availability for nesting in temperate and tropical forests in Mexico. Tree cavity availability is reduced in heavily managed forests, making cavity nesting species particularly vulnerable to the high rates of forest loss and degradation that occur every day in the remaining wilderness areas of the country. We analyzed information about nesting behavior, distribution, and conservation status of resident landbird avifauna from 35 important and protected bird areas of Mexico. The main intention was to determine the proportion of resident bird species that nest in tree cavities and are more sensitive to intensive forest management practices. Our results revealed that 17% (112 species) of the resident landbird avifauna need tree cavities for nesting. Cavity nesters represented a higher proportion of endangered and threatened species than non-cavity nesters. The families Strigidae and Psittacidae represented the highest number of tree cavity nesting species in status. In the 35 avifaunas examined, the mean percentage of cavity nesting species ranged from 17% to 21%. The cloud forest reserve of “El Triunfo” with 43 species, is the area with the highest concentration of cavity nesting species in Mexico.  相似文献

DNA variation was studied at the intra- andinterspecific levels within the genus Passiflora, with particular emphasis on the subgenusTacsonia and its relationship with the subgenusPassiflora. AFLP fingerprints were generated for35 accessions from 21 species, 14 of which belong to subgenusTacsonia, and four to subgenusPassiflora. The morphologically intermediateP. manicata (subgenusManicata) was also included, as wereP. trinervia (subgenusPsylanthus) and P.alnifolia (subgenus Decaloba). The two latter were used as outgroup species. Twoprimer combinations generated 34–80 scorable fragments pergenotype for a total of 260 fragments. Polymorphism analyses spannedthe levels from individual genotype to section. SubgenusTacsonia presented wide genetic variability atboth intra- and interspecific levels, particularly within wildspecies. The classification based on these markers generally agreeswell with current taxonomic definitions of common species, but lesswith definitions of sections. A geographical component was evident inthe intraspecific variation of P.mixta, P.tripartita var. mollissimaand P. tarminiana, withwider variation in Ecuador and Peru than in Colombia and Venezuela.These same three species, plus P.cumbalensis, formed adjacent clusters, showingcloser affinity with each other than with the other species ofsubgenus Tacsonia. As expected, P. manicata clustered at anintermediate position between subgenera Tacsoniaand Passiflora.  相似文献

We analysed population trends of 24 wildfowl species in Mexico. Wildfowl numbers peaked during the early 1980s; lowest counts were recorded in 1997. Total wildfowl numbers (all species combined) and duck numbers (duck species combined) showed significant short-term (1981-2000) declines, while geese (goose species combined) showed a significant long-term (1961-2000) increase. Six wildfowl species suffered significant long-term declines, while four showed increases. During 1981-2000, 11 species declined, but none had significant increases. Redhead (Aythya americana), Mexican duck (Anas diazi), northern pintail (A. acuta) and black brant (Branta bernicla nigricans) should be given high conservation priority because of the high proportions of their North American populations in Mexico. Declining numbers of the later two species should trigger further investigation into the possibility of assigning them legal protection status. Other species with apparent declines in numbers should also be more closely monitored. For setting hunting limits in the country, the population status of each species should be accounted for, as well as the condition of breeding populations the previous spring. Other species with poor data or combined counts should be targeted for basic population studies. We suggest that the mid-winter counts be expanded to cover non-surveyed areas and conducted every year to more precisely detect wildfowl population change. Integrated to a site-selection analysis, the information presented here can provide the basis for a wildfowl conservation strategy in Mexico.  相似文献

Jacaratia mexicana A. DC. (Caricaceae) is a tropical tree distributed throughout Mexico and Mesoamerica. Some evidence in Mexico indicates the presence of an incipient domestication process in this species. Phylogeographical analyses can potentially determine contemporary patterns of gene flow, isolation between population lineages, as well as historical processes such as population bottlenecks or expansions on their geographical areas. In this study we reconstruct the phylogeographical patterns in populations of J. mexicana A. DC., in order to find differences between genetic variation among wild and cultivated populations utilizing chloroplast DNA and nuclear DNA sequences. We generate a Bayesian phylogenetic tree, to estimate the divergence time between clades using calibrated mutation rates. We also infer the demographic history of these populations using neutrality tests among wild and cultivated accessions. We identified higher levels of haplotype and nucleotide diversity for the cpDNA and ITS types in wild populations than in domesticated populations. These results indicate a reduction of genetic diversity derived from human selection on domestication traits. Neutrality test suggests population expansion detected by the significant negative values of Fu’s Fs in the cultivated populations of this specie. These process results in an excess of rare polymorphism with the fixation of certain advantageous mutation throughout time, this implication are in accordance with the role of the strong selection in the fruit traits of J. mexicana. The dated phylogeny constructed with BEAST program indicated a dispersion pattern for the J. mexicana ancestors across the South Pacific and South Eastern populations during the late Pliocene. Posterior dispersion and divergence in the clades from Central Mexico and North Pacific are in agreement with the episodes of mountain-building in different regions of Mexico.  相似文献

Though Neotropical countries are the most species rich in the world, their biodiversity is threatened by the loss of native vegetation. Land conversion in Mexico during the last 30 years has been extensive and is representative of that of other developing countries. However, the effects of land use change on the required size and configuration of an adequate biological conservation area network are largely unknown. It is shown here that endemic mammals in Mexico could have been protected considerably more economically if a conservation plan had been implemented in 1970 than is possible today due to extensive conversion of primary habitats. Analysis of the distributions of 86 endemic mammal species in 1970, 1976, 1993, and 2000 indicates that the distributions of 90% of the species shrank during this 30-year period. At each time step, optimal conservation area networks were selected to represent all species. 90% more land must be protected after 2000 to protect adequate mammal habitat than would have been required in 1970. In addition, under a realistic conservation budget, 79% fewer species can be represented adequately in a conservation area network after 2000 compared to 1970. This provides an incentive for rapid conservation action in Mexico and other biodiversity hotspots with comparable deforestation rates, including Burma, Ecuador, Indonesia, the Philippines, and Sri Lanka. Due to ongoing habitat degradation, the efficiency of a conservation plan decreases with delays in its implementation.  相似文献
