The objective of the present work was to develop species-specific microsatellite markers for P. scalare and to analyze the diversity and genetic structure of a wild population, from the Amazon River, and three commercial stocks (common, marble, and clown morphological variants), from farmers in Vieras-Minas Gerais. Through microsatellite-enriched genetic libraries, 11 microsatellite markers with adequate amplification patterns were characterized. Population genetic analysis identified eight polymorphic loci that generated 66 alleles ranging from two alleles (PSCA1B3) to nine (PSCA2H1). The polymorphic information content ranged from 0.031 to 0.827. High genetic differentiation was observed between the wild population and the stocks, and moderate differentiation between the three stocks. Deviation in the Hardy-Weinberg equilibrium was observed in one locus in the wild population, in five loci in the common morphological variant, in two in the marble, and in two in the clown morphological variant. Bayesian analysis of genetic structure revealed the existence of two clusters, one represented by the natural population and the other by the stocks. The developed microsatellite markers serve as a tool for the analysis of diversity and genetic structure and conservation studies of P. scalare. 相似文献

Three different live diets, Eucyclops serrulatus, Ceriodaphnia quadrangula and equal combination of E. serrulatus copepodid and C. quadrangula, were offered to angelfish ( Pterophyllum scalare) larvae viz 1‐week, 2‐week and 3‐week old at prey densities of 2, 5 and 10 individuals mL ?1. Results showed that 1‐week‐old P. scalare larvae consumed E. serrulatus copepodid at a rate of 31.3–56.7 ind. h ?1, C. quadrangula at 8.0–12.0 ind. h ?1, and mixture of E. serrulatus and C. quadrangula at 20.7–40.7 ind. h ?1. For 2‐week‐ and 3‐week‐old larvae, consumption rate increased accordingly. The electivity indices (E) of P. scalare (1‐week‐old larvae) for E. serrulatus copepodid were +0.18, +0.23 and +0.22 at prey densities of 2, 5 and 10 ind. mL ?1 respectively. Tendency towards E. serrulatus copepodid consumption reduced by aging P. scalare as indicated by the E values for 2‐ and 3‐week‐old larvae. However, growth and survival of P. scalare larvae was greatest when fed on combination of copepod E. serrulatus and C. quadrangula. 相似文献

Current ornamental fish packaging systems are characterized by very high fish loading densities and high metabolic wastes in the transport water after shipment. They focus mainly on management of the quality of transport water. Recent studies using the guppy as a model fish showed that post‐shipment mortality could be reduced through enhancement of the stress resistance of the fish, and hence emphases should also be placed on the preparation of the fish for transport and recovery of the fish after shipment. Farmers can contribute significantly by applying nutritional prophylaxis before harvesting. Exporters may use the salinity stress test to identify fish lots of good quality for transport, apply health prophylaxis to eradicate parasites and optimize other techniques such as starvation of the fish or addition of salt to the transport water to enhance the stress resistance of the fish. Importers may adopt proper acclimation procedure and allow fish to recover in low salinity water to reduce post‐shipment mortality. As the main bulk of post‐shipment mortality is stress‐mediated and occurs during the 1‐week recovery period, the industry should consider revising the basis of the current warranty system for their customers, from death on arrival to cumulative mortality at 7 days post shipment (or death after 7 days, DA7), in order to cut down fish losses after shipment. 相似文献

In this study, we evaluated the production potential of the polyculture of Freshwater Angelfish ( Pterophyllum scalare, Cichlidae) and Pacific white shrimp ( Litopenaeus vannamei). Two experimental trials were set up. The first trial entailed the use of a randomized design to investigate three treatments options: angelfish monoculture, shrimp monoculture, and fish and shrimp polyculture in 12 experimental 15 m 2 ponds. In the second trial, we investigated two treatments (polyculture of caged fish and uncaged fish) in 10 experimental units. In trial 1, polyculture negatively affected fish growth and survival. However, fish did not affect shrimp growth and the greatest profit was achieved in polyculture. In trial 2, caging fish improve the growth of both fish and shrimp. An economic analysis showed the greatest profit and benefit cost ratio for caged fish compared with uncaged fish. We conclude that growing P. scalare and L. vannamei together is a better strategy than shrimp monoculture in low‐salinity water. The use of caged fish in such a polyculture operation would enhance productivity and profitability. 相似文献

This study investigated the efficacy of Lippia grata essential oil (EO) against monogeneans of Colossoma macropomum and effects on the haematology after immersion baths. In the in vitro assays, the efficacy of 100, 250, 350 and 700 mg/L of L. grata EO and two controls were tested, of which one control was with the cultivation tank water and the other was with the tank water and 70% ethyl alcohol. Composition of majority bioactive compounds in the EO was carvacrol (48.12%), p‐cymene (24.39%) and γ‐terpinene (2.49%). Anacanthorus spathulatus, Notozothecium janauachensis, Mymarothecium boegeri and Linguadactyloides brinkmanni obtained of fish gills showed 100% immobilization in the in vitro assays when exposed to 700 and 350 mg/L after 30 min and 2 hr respectively. Fish showed tolerance to 700 mg/L of the EO, which was used in therapeutic in three consecutive baths, which caused lamellar hyperplasia in C. macropomum gills. The 700 mg/L of the EO in the therapeutic baths showed an efficacy of 95.1% against the monogeneans and caused decrease in haemoglobin, whereas control fish showed a reduction in mean corpuscular haemoglobin concentration. Results indicated that 700 mg/L of L. grata EO is a safe and effective concentration to treat C. macropomum infected with monogeneans. 相似文献

This study evaluated the effect of short-term baths with the essential oils (EOs) of Lippia sidoides, Ocimum gratissimum and Zingiber officinale on the survival, haematological, biochemical and histopathological parameters of native fish tambaqui ( Colossoma macropomum), challenged with Aeromonas hydrophila. After challenge, the fish infected with A. hydrophila received therapeutic baths of 60 min, for five consecutive days. Treatments were as follows: challenged and not treated; challenged and treated with 10 mg L ?1 of gentamicin; challenged and treated with EO of L. sidoides (2.5 and 5.0 mg L ?1); challenged and treated with EO of O. gratissimum (5.0 and 10.0 mg L ?1); challenged and treated with Z. officinale EO (5.0 and 10.0 mg L ?1). The highest survival rates were 89.5% (fish treated with 5.0 mg L ?1 of O. gratissimum EO) and 75.0% (fish treated with 5.0 mg L ?1 of L. sidoides EO). Reductions in the haematocrit percentage, haemoglobin and erythrocyte number were observed in fish treated with L. sidoides and Z. officinale EOs compared with control fish. Mild to moderate damage to the liver tissue of fish after challenge and EO treatments were observed. It is suggested that baths containing O. gratissimum and L. sidoides EOs consist in a viable treatment to increase survival rates of C. macropomum infected with A. hydrophila. 相似文献

Failure to inflate the swim bladder is regarded a major obstacle in the rearing of many fish species. We present a study of swim bladder non-inflation (SBN) in angelfish, Pterophyllum scalare. A normal developing primordial swim bladder was first discernable at the end of the first day post-hatch (p.h.) as a cluster of epithelial cells with a central lumen, surrounded by presumably mesenchymal cells. Initial inflation occurred on the fourth day p.h. Prior to inflation the swim bladder epithelium consisted of an outer squamous and inner columnar layer. Cells of the inner layer were filled at their basal region with an amorphous material, which disappeared upon inflation. A pneumatic duct was absent, and larvae presented no need to reach the water surface for inflation, suggesting that angelfish are pure physoclists. A model for the role of the amorphous material in normal initial inflation is proposed. Abnormal swim bladders were apparent from the fourth day p.h., and methylene blue (MB) at a concentration of 5 ppm significantly increased the prevalence of SBN. Histologically, abnormal swim bladders in larvae hatched in 5 ppm MB could not be distinguished from those in fish raised under routine conditions (0.5 ppm MB). Ornamental fish farming is globally a key sector of aquaculture. The purpose of this study was to report on the parasitic fauna of ornamental fish from seven farms in the East Azerbaijan province in northwestern Iran. Between November 2020 and February 2021, a total of 600 freshwater ornamental fish of six species, including goldfish (Carassius auratus), angel (Pterophyllum scalare), guppy (Poecilia reticulata), dwarf gourami (Colisa lalia), swordtail (Xiphophorus helleri), and cichlid (Cichlasoma sp.) were collected for parasitological analysis. Six ciliate protozoan species (Trichodina mutabilis, Trichodina nobilis, Trichodina spp., Heteropolaria lwoffi, Ichthyophthrius multifiliis, and Chilodonella spp.), three species of monogenean trematodes (Dactylogyrus dulkeiti, Dactylogyrus spp. and Gyrodactylus spp.), two species of crustacean arthropods (Lernaea cyprinacea and Argulus japonicus), and a nematode (Capillaria spp.) were identified among the examined fish. Parasites were detected in the fish from 5 of 7 farms, with an overall parasitic prevalence in 26.33% (158/600) of the fish. Fish infection rates with protozoans, monogenean trematodes, crustacean arthropods, and nematodes were 16.83, 13.17, 4, and 0.33% respectively. The highest prevalence was found in X. helleri infected with Gyrodactylus spp. and P. scalare infested with Trichodina spp. Moreover, 9/100 and 10/100 of goldfish were infected with A. japonicus and L. cyprinacea, compared to 2/500 and 3/500, respectively, for all other species in combination, which is a statistically significant result (p?<?0.01). Some mycobacterial species (particularly Mycobacterium marinum) found in aquarium environments may cause chronic diseases in fish and cutaneous infections in humans, the so-called 'fish tank granuloma'. The presence and distribution of mycobacterial species in clinically healthy aquarium fish and their environment has not been adequately explored. The present study analysed the occurrence of mycobacteria in a decorative aquarium (Brno, South Moravia) and in five aquaria of a professional fish breeder (Bohumin, North Moravia). After Ziehl-Neelsen staining, acid-fast rods (AFR) were observed in six (14.3%) and mycobacteria were detected by culture in 18 (42.9%) of 42 tissue samples from 19 fish. Sixty-five samples of the aqueous environment from all six aquaria were examined; AFR were found in 16 (24.6%) and mycobacteria were detected by culture in 49 (75.4%) samples. Forty-one (70.7%) of 58 selected mycobacterial isolates were identified biochemically as follows: M. fortuitum, M. flavescens, M. chelonae, M. gordonae, M. terrae, M. triviale, M. diernhoferi, M. celatum, M. kansasii and M. intracellulare. The clinically important species for humans and fish, M. marinum, was not detected. Mycobacterium kansasii was isolated from one sample of the aquarium environment from North Moravia, which is a region of the Czech Republic with endemic incidence of M. kansasii in water. The incidence of other conditionally pathogenic mycobacterial species in healthy fish and in all investigated constituents of the aquarium environment including snails and crustaceans used for fish feeding, was quite high. Accordingly, mycobacterial species from aquarium environments may serve as a possible source of infection for both aquarium fish and immunodeficient fish handlers. 相似文献

The juveniles of Macrobrachium rosenbergii (0.060 ± 0.004 g and 2.018 ± 0.071 cm) were fed during 60 days with diets containing different doses of Lippia alba essential oil (EOLA) (0.0—control, 1.0, or 2.0 ml EOLA/kg diet; in triplicate, with 20 prawns/replicate). After the experimental period, were verified the survival, growth parameters and the antioxidant enzymes, lipid peroxidation and Na/K‐ATPase activities in hepatopancreas and gills, respectively, of the animals. There were no significant differences on survival, growth parameters and gill Na/K‐ATPase activity. However, glutathione peroxidase and glutathione‐ S‐transferase activities were lower in hepatopancreas of prawns fed with 1.0 ml EOLA/kg diet compared to the control. Moreover, the activity of the enzyme superoxide dismutase was higher in hepatopancreas of prawns that received 2.0 ml EOLA/kg diet compared to those fed with 1.0 ml EOLA/kg diet. Although the addition of both EOLA doses has not improved the survival and growth parameters of M. rosenbergii, these doses contributed to decrease lipid peroxidation. Additionally, the dose of 2.0 ml EOLA/kg diet contributed for increasing the antioxidant enzymes activities in the hepatopancreas, improving antioxidant status, and therefore, it can be recommended as diet supplementation for M. rosenbergii. 相似文献

Mycobacteriosis in fish is a chronic progressive ubiquitous disease caused by Mycobacterium marinum, M. gordonae and M. fortuitum in most cases. The aim of this study was to describe the morphology and distribution of lesions in 322 freshwater ornamental fish across 36 species. Granulomatous inflammation was diagnosed by gross examination and histopathology testing in 188 fish (58.4%); acid‐fast rods (AFR) were determined in only 96 (51.1%) fish from 19 species after Ziehl–Neelsen staining. The most often affected organs with AFR were the kidney (81.2%), digestive tract (54.1%), liver (48.2%), spleen (45.9%) and skin (21.2%); sporadically, AFR were found in the branchiae (9.4%) and gonads (4.7%). In 14 randomly selected fish originating from four different fish tanks, the distribution of mycobacterial infection was studied by culture examination of the skin, gills, muscle tissue, digestive tract, liver, spleen and kidney. In 12 fish, the species M. marinum, M. gordonae, M. fortuitum, M. triviale, and M. avium subsp. hominissuis (serotypes 6 and 8 and genotype IS 901? and IS 1245+) were detected; mixed infection caused by different mycobacterial species was documented in five of them. 相似文献

通过检索BOLD SYSTEMS数据库和Gen Bank中锦鲤、金鱼、慈鲷、神仙鱼、七彩神仙鱼、血鹦鹉、花罗汉、龙鱼、孔雀鱼、鼠鱼、魟鱼等主要观赏鱼的DNA条形码和基因序列,应用MEGA软件计算种间遗传距离,在BOLD SYSTEMS数据库中检索相似序列,以分析DNA条形码在主要观赏鱼物种鉴定中的应用效果。结果表明该技术可以有效鉴别大多数种以上阶元的观赏鱼类,但对亲缘关系较近、种类繁多的多种慈鲷、孔雀鱼等观赏鱼物种难以获得准确的鉴定结果,对金龙、红龙等亚洲龙鱼的亚种以及人工繁育的不同品系的锦鲤、金鱼、神仙鱼、七彩神仙鱼、血鹦鹉、花罗汉等鱼类,无法通过COI基因条形码进行鉴别,需要研究其他分子生物学方法。 相似文献

There is a growing commercial interest in the fish, Puntius filamentosus , in the ornamental fish trade in India and elsewhere. The trade is, however, hampered by severe mortalities during transport of the fish owing to insufficient data available on the use of anaesthetics. To resolve this problem, we evaluated the efficacy of two anaesthetics, MS-222 and benzocaine, in sedating P. filamentosus in simulated transportation experiments and used stress response parameters such as cortisol and blood glucose levels to perform assessments. We observed that MS-222 at 40 mg L −1 and benzocaine at 20 mg L −1 were sufficient to induce sedation for 48 h. Above these concentrations, both the anaesthetics adversely affected the fish and resulted in mortalities. Both anaesthetics significantly lowered the blood cortisol and glucose levels compared with the unsedated controls. Importantly, the anaesthetics treatment significantly lowered the post-transport mortality in the fish. The results of the study show that MS-222 and benzocaine could be used as sedatives to alleviate transport-related stress in P. filamentosus to improve their post-transport survival and hence reduce economic loss. 相似文献

‘Gold standard’ OIE reference PCR assay was utilized to detect the presence of infectious spleen and kidney necrosis virus (ISKNV) in freshwater ornamental fish from Malaysia. From total of 210 ornamental fish samples representing 14 species, ISKNV was detected in 36 samples representing 5 fish species. All positive cases did not show any clinical signs of ISKNV. Three restriction enzymes analyses showed that the fish were infected by identical strains of the same virus species within Megalocytivirus genus. Major capsid protein (MCP) genes of 10 ISKNV strains were sequenced and compared with 9 other reference nucleotide sequences acquired from GenBank. Sequence analysis of MCP gene showed that all strains detected in this study were closely related to the reference ISKNV with nucleotide sequence identity that was ranging from 99.8% to 100%. In addition, phylogenetic analysis of MCP gene revealed that viruses from genus Megalocytivirus can be divided into three genotypes: genotype 1 include reference ISKNV and all other strains that were detected in this study, genotype 2 include viruses closely related to red sea bream iridovirus (RSIV), and genotype 3 include viruses closely related turbot reddish body iridovirus (TRBIV). 相似文献

A jet fish pump with a throat of ø60 mm was designed to study its performance in the transport of different fish species and the physiological changes in fish thereafter. Experiments were conducted to investigate the fish transport rate and energy required to transport each ton of fish when transporting Carassius auratus, commonly known as the Chinese goldfish, Megalobrama amblycephala, or Wuchang bream, and Ctenopharyngodon idella, the grass carp. Fish were examined for external injuries as well as for several important enzymes and hormones which are indicators of tissue injury and stress. The results showed that the transport rate for all three species of fish rose dramatically with an increase in the primary stream rate. In this experiment, the transport rates of C. auratus, M. amblycephala and C. idella reached 2357 ± 37.2 kg ∙ h −1, 2888 ± 41.6 kg ∙ h −1, and 2060 ± 40.2 kg ∙ h −1, respectively. However, both injury rate and energy required to transport each ton of fish increased no matter whether the primary stream rate was too low or too high. Considering both transport rate and injury rate, the mean primary stream rate of 80 m 3 ∙ h −1 was determined to be the optimal operating condition in this experiment. Fish were stressed and most likely some of their organs were damaged. However, most physiological indexes almost fully recovered after several hours. 相似文献

目前我国水产业以养殖为主,捕捞为辅。在2022年的水产品产量中,淡水养殖占47.91%,海水养殖占33.15%,捕捞占18.94%。在水产养殖中,鱼类养殖产量占52.16%,呈现出举足轻重的地位。“良种良养良销”是水产业中三个重要的组成部分,“良种”是水产业的龙头,“良养”是水产业的保障,加工、销售等“良销”环节是拉动水产业发展的重要动力。20世纪50年代以来,我国水产业取得了长足发展,为广大人民提供了丰富的水产品,解决了“吃鱼难”的问题。然而,目前我国水产业中有种无业、有业无种等现象还比较严重,没有从根本上解决“吃好鱼”及“吃放心鱼”的问题,为了加快我国水产业的发展,作者认为很有必要建立和实施“良种良养良销”体系,以推动我国水产业高质量发展。 相似文献

This study aimed to investigate the effects of dietary supplementation of lemongrass ( Cymbopogon citratus) (LEO) and geranium ( Pelargonium graveolens) (GEO) essential oils on growth, immunity, antioxidant responses and disease resistance of Oreochromis niloticus. Fish (3.04 ± 0.003 g) were fed the basal diet (G 1), or the basal diet supplemented with LEO at 200 and 400 mg kg ?1 of the diet (G 2 and G 3), or GEO at 200 and 400 mg kg ?1 of the diet (G 4 and G 5), respectively for 12 weeks. Growth indices and feed utilization were significantly improved with G 2, G 3 and G 5 groups. Also, a significant increase in protein content of the whole body was recorded in all groups. G 2 and G 5 showed a significant rise of both catalase activity and reduced glutathione content with a significant incline in the MDA level. Lysozyme activity and total IgM levels were significantly increased in G 2 and G 5 groups. G 3 and G 5 groups had a significantly lower intestinal total bacteria, Coliforms, Escherichia coli and Aeromonas spp counts. Following challenge with A. hydrophila, all groups had a high survival rate. In conclusion, LEO and GEO could be a candidate dietary supplementation in fish improving growth performance, feed utilization, oxidative status, immune responses and disease resistance. 相似文献
