Barley spot blotch (SB), caused by Cochliobolus sativus, is an important barley disease which causes extensive grain yield losses. These losses may not always correlate directly with the amount of diseased leaf area. Two barley cultivars, Quebracho (susceptible to SB) and Carumbé (with intermediate susceptibility to SB), were compared in field experiments in 2003, 2004 and 2006. Plots of each cultivar were either inoculated with C. sativus or protected with fungicide under field conditions to generate contrasting treatments: i) diseased, and ii) free of disease, respectively. SB severity over the growing season, photosynthetic rate on leaves with no visible symptoms and grain yield were assessed for each treatment and year. There was no treatment effect on cv. Carumbé, while cv. Quebracho showed a significant yield reduction, even though SB severity during the grain filling period was <10 %. This yield reduction was associated with a reduced photosynthetic rate at the beginning of the grain filling period in cv. Quebracho. A similar experiment was conducted under greenhouse conditions, adding a treatment without inoculum or fungicide. There were no differences in photosynthetic rate or grain yield per plant among treatments. These results suggest a distinct physiological response to SB infection among cultivars affecting leaf photosynthetic rate, and SB severity may not be the best estimator of yield losses caused by SB.  相似文献

Initial experiment on the reactions of five Japanese cultivars of cucumber toColletotrichum orbiculare infection in the greenhouse revealed that cv Suyo and Gibai were susceptible and moderately susceptible, respectively, while cv Shogoin fushinari and Sagami hanjiro were resistant to infection byC. orbiculare; cv Ochiai fushinari was moderately resistant. The ability of 16 plant growth promoting fungi (some isolates belonged to species ofPhoma and some non-sporulating isolates) isolated from zoysiagrass rhizospheres to induce systemic resistance in the above five cucumber cultivars was tested by growing plants in potting medium infested with barley grain inocula of PGPF in the greenhouse. The second true leaves of 21-day-old plants were challenge inoculated withC. orbiculare and disease assessed. Nine, out of 16 isolates, caused significant reduction of disease caused byC. orbiculare in at least two cultivars.Phoma isolates (GS8-1 and GS8-2) and non-sporulating isolates (GU21-2, GU23-3, and GU24-3) significantly reduced the disease in all the five cultivars. The disease suppression in cucumber was due to the induction of systemic resistance, since the inducer(s) and the pathogen were separated spatially and that the inducer did not colonize aerial portions. The resistance induced by certain isolates in a susceptible cultivar was less than that in a resistant cultivar. Disease suppression caused by isolate GU21-2 was similar to theC. orbiculare induced control in certain cultivars. The average rate of expansion of lesion diameter on leaves due toC. orbiculare was slower due to induction with the selected plant growth promoting fungi compared to the uninduced control plants. ABSTRACT A leaf spot disease with unknown etiology has become more pronounced in spring and winter barley in Germany in recent years. The symptoms are similar to net blotch and Ramularia leaf spots, but the causal agents of these diseases are not identified. The symptom expression varied much on cultivars. Cultivars most affected by the disease of both spring and winter barley showed a significantly higher level of superoxide (O(2) ) production and lipid peroxidation (malondialdehyde), but a lower level of antioxidant potential expressed as superoxide dismutase (SOD) activity, catalase activity, and integral water-soluble antioxidant capacity (ACW) than insensitive cultivars. A high positive correlation between O(2) production and leaf spot development between ear emergence and milk ripeness was established in the most sensitive winter barley cv. Anoa (r(2) = 0.9622) and spring barley cv. Barke (r(2) = 0.9434). Leaf H(2)O(2) levels increased with the severity of leaf spots. The histochemical localization of O(2) and H(2)O(2) in the tissues adjacent to leaf spots indicated that these two active oxygen species (AOS) are involved in the formation of leaf spots. Reduction of symptom severity by applying strobilurin and azole fungicides was always associated with elevated SOD activity and ACW content and suppressed O(2) production. However, peroxidase activities were significantly higher in sensitive cultivars and in more severely affected tissues and decreased by applying fungicides. Thus, it is assumed that a possible genetic mechanism based on the imbalanced AOS metabolism contributes to formation of physiological leaf spots.  相似文献

ABSTRACT Pyrenophora teres, the causal agent of net blotch of barley (Hordeum vulgare L.), induces a combination of necrosis and extensive chlorosis in susceptible barley cultivars. Cell-free filtrates from both net and spot forms of P. teres; P. teres f. sp. teres, and P. teres f. sp. maculata were found to contain phytotoxic low molecular weight compounds (LMWCs) and proteinaceous metabolites which appear to be responsible for different components of the symptoms induced by the two forms of the pathogen in a susceptible cultivar of barley (cv. Sloop). Proteins induced only brown necrotic spots or lesions similar to those induced by the pathogens 72 h after inoculation. In contrast, LMWCs induced general chlorosis seen 240 h after inoculation but not the localized necrosis. Neither hydrolyzed or heat- or protease-treated proteinaceous metabolites induced the symptoms. This is the first report of the involvement of proteins produced by P. teres in symptom development during net blotch disease of barley.  相似文献

ABSTRACT Leaf blotch, caused by Rhynchosporium secalis, was studied in a range of winter barley cultivars using a combination of traditional plant pathological techniques and newly developed multiplex and real-time polymerase chain reaction (PCR) assays. Using PCR, symptomless leaf blotch colonization was shown to occur throughout the growing season in the resistant winter barley cv. Leonie. The dynamics of colonization throughout the growing season were similar in both Leonie and Vertige, a susceptible cultivar. However, pathogen DNA levels were approximately 10-fold higher in the susceptible cultivar, which expressed symptoms throughout the growing season. Visual assessments and PCR also were used to determine levels of R. secalis colonization and infection in samples from a field experiment used to test a range of winter barley cultivars with different levels of leaf blotch resistance. The correlation between the PCR and visual assessment data was better at higher infection levels (R(2) = 0.81 for leaf samples with >0.3% disease). Although resistance ratings did not correlate well with levels of disease for all cultivars tested, low levels of infection were observed in the cultivar with the highest resistance rating and high levels of infection in the cultivar with the lowest resistance rating.  相似文献

This paper evaluates, in the greenhouse and under natural conditions, barley yield losses due to defoliation treatments of the upper three leaves either healthy or infected at the boot stage by Pyrenophora teres f. teres. Defoliation was assumed as a loss of a similar leaf area caused by net blotch disease severity of 100%. Contribution to grain yield was defined herein as a difference between defoliation treatments and a treatment where plants lost all their upper three leaves. In contrast, yield losses referred to differences in yield between defoliations and the control. In the greenhouse, removal of the antepenultimate leaf did not affect any yield component. For main stems, defoliating upper three leaves reduced grain yield by 30% and this was mainly due to flag leaf removal. These losses were similar to those induced by net blotch disease under natural conditions, but were of 42% for all tillers. Grain yield losses due to disease severity were not equivalent to the defoliation effect of a similar healthy leaf area. On the other hand and for a significant contribution to grain yield, flag leaf was dependent on the presence of the other two leaves. Inoculation and defoliation of 21 cultivars induced similar grain yield losses of 32%. However, biotic stress reduced by 40% the contribution of their upper three leaves. Under field conditions, yield losses were not significant until barley plants lost more than one upper leaf and flag leaf contribution was equivalent to that of the remaining leaves. Characteristic roots, defined as leaf coefficients for plant performance, were 0.13, 0.06 and 0.01 for the flag, penultimate and antepenultimate leaves, respectively. Because antepenultimate leaves become trivial at the boot stage, we propose that coefficients of the remaining leaves should be used when modelling yield losses due to barley foliar diseases.  相似文献

为了明确鲁豫皖交界地区小麦禾谷孢囊线虫的致病类型,利用23个国际鉴别寄主品种和1个当地小麦品种温麦19对采自山东菏泽、安徽颍上、河南商丘和淮阳的4个小麦禾谷孢囊线虫群体致病型进行了鉴定。4个群体的致病型不同于国际上已命名的16个小麦禾谷孢囊线虫致病型。13个A组鉴别寄主对山东菏泽与安徽颍上群体具有相同的抗性反应,这2个线虫群体属于同一致病型;河南商丘群体与上述2个群体的毒性相似,但大麦寄主KVL191对山东菏泽群体和安徽颍上群体表现为抗病,而对河南商丘群体表现感病;河南淮阳群体的致病谱较宽,明显不同于其它3个群体。对照小麦品种温麦19对4个小麦禾谷孢囊线虫群体均表现感病。  相似文献

ABSTRACT Most barley cultivars are resistant to stripe rust of wheat that is caused by Puccinia striiformis f. sp. tritici. The barley cv. Steptoe is susceptible to all identified races of P. striiformis f. sp. hordei (PSH), the barley stripe rust pathogen, but is resistant to most P. striiformis f. sp. tritici races. To determine inheritance of the Steptoe resistance to P. striiformis f. sp. tritici, a cross was made between Steptoe and Russell, a barley cultivar susceptible to some P. striiformis f. sp. tritici races and all tested P. striiformis f. sp. hordei races. Seedlings of parents and F(1), BC(1), F(2), and F(3) progeny from the barley cross were tested with P. striiformis f. sp. tritici races PST-41 and PST-45 under controlled greenhouse conditions. Genetic analyses of infection type data showed that Steptoe had one dominant gene and one recessive gene (provisionally designated as RpstS1 and rpstS2, respectively) for resistance to races PST-41 and PST-45. Genomic DNA was extracted from the parents and 150 F(2) plants that were tested for rust reaction and grown for seed of F(3) lines. The infection type data and polymorphic markers identified using the resistance gene analog polymorphism (RGAP) technique were analyzed with the Mapmaker computer program to map the resistance genes. The dominant resistance gene in Steptoe for resistance to P. striiformis f. sp. tritici races was mapped on barley chromosome 4H using a linked microsatellite marker, HVM68. A linkage group for the dominant gene was constructed with 12 RGAP markers and the microsatellite marker. The results show that resistance in barley to the wheat stripe rust pathogen is qualitatively inherited. These genes might provide useful resistance against wheat stripe rust when introgressed into wheat from barley.  相似文献

Leaves of powdery mildew-susceptible barley (Hordeum vulgare cv. Ingrid) and related near-isogenic lines bearing various resistance genes (Mla12, Mlg or mlo5) were inoculated with Blumeria graminis f. sp. hordei race A6. Fungal attack induced several-fold increases in ethylene emission and electrolyte leakage in leaves of susceptible Ingrid beginning 3 days after inoculation. Activities of peroxidase, superoxide dismutase, glutathione S-transferase, ascorbate peroxidase and glutathione reductase enzymes were induced markedly in susceptible leaves 5–7 days after inoculation. Similar, but less pronounced pathogen-induced changes were detected in inoculated leaves of Mla-type resistant plants that show hypersensitive cell death upon inoculation, and, to an even lesser extent, in the Mlg and mlo lines, where no visible symptoms accompanied the incompatible interaction. Glutathione content increased only in susceptible barley 7 days after inoculation. Catalase activity, total ascorbate content and redox state were not influenced by inoculation in any of the genotypes. The activity of dehydroascorbate reductase was significantly reduced 3–5 days after inoculation in the susceptible parental plants and after 5 days in Mla and Mlg lines, while it was stable in the mlo barley. Slightly elevated levels of H2O2 were observed in the inoculated resistant plants. In contrast, H2O2 content decreased in the susceptible line 7 days after pathogen attack. These data indicate that high levels of antioxidants are involved in the compatible interaction of susceptible barley and powdery mildew by protecting the pathogen from oxidative damage.  相似文献

The effect of chocolate spot disease (incited byBotrytis fabae) on the host plant quality of two cultivars of faba beans (Vicia faba) for the black bean aphid (Aphis fabae), was investigated under greenhouse conditions. Faba bean cultivar Bolero is resistant to the aphid, whereas cv. Diana is susceptible. On healthy leaves of cv. Bolero, aphids completed their development one day later than the aphids reared on infected leaves of this cultivar and those which matured on healthy or infected leaves of cv. Diana. Mean weight of apterous virginoparae, mean relative growth rate, and intrinsic rate of natural increase were enhanced if the aphids were reared on infected leaves of either cultivar. Mean total progeny per female increased only on infected leaves of cv. Bolero, but not on cv. Diana.  相似文献

Two spring barley cultivars Bajo Aragon-1-1 and Martin 403-2 from the cyst nematode test assortment were each crossed with a susceptible, a pathotype-11 -resistant, and a pathotype-11 and 12-resistant cultivar, as well as with each other. A spring wheat cultivar, AUS 10894, from the cyst nematode test assortment was crossed with a susceptible and a resistant cultivar. From these crosses F! and F2 single plants were tested against pathotype 12 and/or pathotype 11 of Heterodera avenae in Denmark. Some plants of the spring barley cultivar Bajo Aragon-1-1 have two resistance genes, which probably are both dominant, but it is not out of the question that one might be recessive. Each gene is inherited independently of the other or with low linkage frequency. Neither gene is the same as or allelic to the resistance gene in the cultivar Ortolan, but one gene is allelic to or the same as the 191 gene in the cultivar Siri. The spring barley cv. Martin 403-2 has one dominant resistance gene which is not allelic to or the same as the resistance gene in Ortolan but is the same as or allelic to the 191 gene in the cultivar Siri. It is possible that there is one more gene, which might be recessive against pathotype 11. The spring wheat cv. AUS 10894 has one dominant resistance gene which might be the same as or allelic to the Loros gene.  相似文献

To assess the potential contribution of growing highly competitive crops to control Cyperus esculentus in the Netherlands, the influence of silage maize, hemp, winter barley and silage winter rye on emergence, growth and reproduction of this weed was studied in two field experiments. Growth and reproduction of C. esculentus (determined both as tuber production per plant and density of the weed in the subseuent crop maize) was reduced by each crop, compared to plots in which no crop was grown. In hemp, hardly any tuber production was observed. By growing a green manure crop after harvest of the winter barley the tuber production of the weed was reduced to about 40% of that in winter barley followed by fallow. Decreasing the light regime in a greenhouse experiment in the same order as was observed in the crops winter barley, maize and hemp, caused a similar reduction of tuber production to that found in the field. Therefore, it is suggested that competition for light is the main factor explaining the observed crop effects on the population dynamics of the weed. It is concluded that growing competitive crops as hemp in rotation may effectively complement chemical control of C. esculentus.  相似文献

Nitric oxide (NO) has been shown to be an essential regulatory molecule in plant response to pathogen infection in synergy with reactive oxygen species (ROS). At the present, nothing is known about the role of NO in disease resistance to nematode infection. We used a resistant tomato cultivar with different sensitivity to avirulent and virulent populations of the root-knot nematode Meloidogyne incognita to investigate the key components involved in oxidative and nitrosative metabolism. We analyzed the superoxide radical production, hydrogen peroxide content, and nitric oxide synthase (NOS)-like and nitrate reductase activities, as potential sources of NO. A rapid NO accumulation and ROS production were found at 12 h after infection in compatible and incompatible tomato-nematode interactions, whereas the amount of NO and ROS gave different results 24 and 48 h after infection amongst compatible and incompatible interactions. NOS-like arginine-dependent enzyme rather than nitrate reductase was the main source of NO production, and NOS-like activity increased substantially in the incompatible interaction. We can envisage a functional overlap of both NO and ROS in tomato defence response to nematode invasion, NO and H2O2 cooperating in triggering hypersensitive cell death. Therefore, NO and ROS are key molecules which may help to orchestrate events following nematode challenge, and which may influence the host cellular metabolism.  相似文献

Wild oat (Avena fatua L.) plants sprayed at the 2-or 3-leaf stages of growth with diclotop-methyl developed chlorosis over the entire leaf blade of all leaves. The leaves became necfrotic 7days after spraying Shool growth was inhibited. In wheat (Triticum aesicum L cv.Waldron) discrete chlorotic areas developed only where the herbicide convicted the 2nd or 3rd leaf with no visible injury so new growth uf'ter treutment. Growth inhibition of susceptible oat (Avena sativa L. cv. Garry) was sensitive to placement of diclutop-methyl near the upica and meristematic sites of the plant. Chlorosis and necrosis were independent of herbicide placement. Selective herbicide placement induced chlorosis only or both chlorosis and growth inhibition Root growth in wild oat and barley (Hordeum rulgare L. cv. Dickson) was strongly inhibited by 1–0 μM diclofop-methyl. Wild oat shoots were killed when seedlings were root-treated with 10 μM diclofop-melhyl. The 100 μM rool treatment killed barley shoots but only stunted the growth of wheat shoots by approximately 50%. In root-ireated wheat plants the shoots were turgid and developed a light purple colour, whereas in foliar-treated plants the shoots developed discrete chlorotic areas.  相似文献

For three seasons, six field trials were conducted in winter wheat ( Triticum aestivum ) at different sites in North Rhine-Westphalia, Germany. The aim was to evaluate the performance of three varieties, which differ in their shading behaviour, in suppressing weeds under organic farming (Ökologischer Landbau) conditions and grown at three row spacings. The experimental factors were cultivar, row width and row orientation. Key parameters of crop and weed development, including ground cover and biomass, were assessed at different growth stages and analysed with anova . Growth of the site-specific weed vegetation was significantly affected in five of six trials. Weed growth was reduced by cvs Astron and Pegassos compared with cv. Greif, and was lower at 12 cm compared with 24 cm row spacing. No effect of row orientation (East-West, North-South) on weed growth was observed. The more competitive cultivars Astron and Pegassos were taller than the less competitive cv. Greif and had higher ground cover and light interception, presumably induced by planophile leaf inclination and partly, in the case of cv. Astron, because of higher leaf area index (LAI). Narrow row spacing (12 cm), resulting in a more even spatial plant distribution, increased crop ground cover, LAI, dry matter and light interception. In two trials, the variety with planophile leaf inclination performed better at wider row spacing, suggesting that planophile wheat cultivars may be advantageous in wider row stands.  相似文献

A Japanese field isolate (Race I) of Erysiphe graminis f.sp, hordei was tested on 17 barley lines carrying the mlo powdery mildew resistance gene. Race I produced many successful infections with infection type larger than or equal to 2 on six lines (M66, MC20, SRI, SR7, A tem and Totem), On the remaining 11 lines it reacted with infection type 0. Colony numbers on the Mlo-lines were between 26% and 12 9% of the numbers on a susceptible cultivar Manchuria, These numbers were larger than, or similar to those produced by isolate HL3/5, which has the highest recorded infection efficiency on Mlo-resistant cultivars. The interaction between isolates and lines was highly significant. The isolate GE3, from which HL3/5 was derived by selection, gave rise to occasional colonies corresponding to less than 03° o of the number produced on cv. Manchuria.  相似文献

ABSTRACT Competition among eight Rhynchosporium secalis isolates was assessed during parasitic and saprophytic phases of the disease cycle in field experiments conducted at two locations and over two growing seasons. The eight isolates were inoculated onto six barley populations exhibiting varying degrees of resistance. Microsatellite analysis of 2,866 isolates recovered from the field experiments showed significant, and sometimes opposite, changes in the frequencies of R. secalis genotypes during the growing season (parasitic phase) and between growing seasons (saprophytic phase). Isolates that showed the most complex virulence in greenhouse seedling assays had the lowest fitness in the field experiment. Significant differences in isolate fitness were found on different host populations and in different environments. Selection coefficients were large, indicating that evolution can occur rapidly in field populations. Although inoculated isolates had the lowest overall fitness on the moderately resistant landrace cv. Arabi Aswad, some isolates were more virulent and consistently increased in frequency on this landrace, suggesting a risk of directional selection and possible erosion of the resistance following its widespread deployment in monoculture. These results provide the first direct evidence that R. secalis pathogen genotypes differ in their saprophytic ability and parasitic fitness under field conditions.  相似文献

Rice yellow mottle virus (RYMV) is a recent and major threat to rice production in Madagascar. A large scale screening of resistance to RYMV in rice germplasm in Madagascar was conducted by visual symptom scoring and virus-assessment through ELISA tests. The response to virus inoculation of 503 local or introduced rice accessions was assessed. Most of them were susceptible to RYMV. A few cultivars expressed partial resistance at a level similar to the partially resistant Oryza sativa japonica cv. Azucena. Only one O. sativa cultivar expressed high resistance characterised by a lack of symptoms and an undetectable level of virus. It was a Malagasy traditional indica cultivar, named Bekarosaka, which originated from the northwest of the country. It was selected by farmers for its field resistance to RYMV. The response of cv. Bekarosaka to four pathotypes of RYMV was similar to that of cv. Gigante, the only other highly resistant indica cultivar. The sequence of the middle domain of the eIF(iso)4G, the genetic determinant of Rymv1 resistance on chromosome 4, of cv. Bekarosaka was similar to that of cv. Gigante. Subsequently, cvs Bekarosaka and Gigante probably carried the same resistance allele Rymv1-2. Rymv1-2 resistance was efficient against isolates of the major strains of RYMV, but was readily overcome by a pathotype from the northwest of Madagascar.  相似文献
