The effects of propiconazole, prothioconazole, tebuconazole, metconazole, and prothioconazole+tebuconazole (as a tank mix or a formulated premix) on the control of Fusarium head blight index (IND; field or plot-level disease severity) and deoxynivalenol (DON) in wheat were determined. A multivariate random-effects meta-analytical model was fitted to the log-transformed treatment means from over 100 uniform fungicide studies across 11 years and 14 states, and the mean log ratio (relative to the untreated check or tebuconazole mean) was determined as the overall effect size for quantifying fungicide efficacy. Mean log ratios were then transformed to estimate mean percent reduction in IND and DON relative to the untreated check (percent control: C(IND) and C(DON)) and relative to tebuconazole. All fungicides led to a significant reduction in IND and DON (P < 0.001), although there was substantial between-study variability. Prothioconazole+tebuconazole was the most effective fungicide for IND, with a C(IND) of 52%, followed by metconazole (50%), prothioconazole (48%), tebuconazole (40%), and propiconazole (32%). For DON, metconazole was the most effective treatment, with a [Formula: see text](DON) of 45%; prothioconazole+tebuconazole and prothioconazole showed similar efficacy, with C(DON) values of 42 and 43%, respectively; tebuconazole and propiconazole were the least effective, with C(DON) values of 23 and 12%, respectively. All fungicides, with the exception of propiconazole, were significantly more effective than tebuconazole for control of both IND and DON (P < 0.001). Relative to tebuconazole, prothioconazole, metconazole, and tebuconzole+prothioconzole reduced disease index a further 14 to 20% and DON a further 25 to 29%. In general, fungicide efficacy was significantly higher for spring wheat than for soft winter wheat studies; depending on the fungicide, the difference in percent control between spring and soft winter wheat was 5 to 20% for C(IND) and 7 to 16% for C(DON). Based on the mean log ratios and between-study variances, the probability that IND or DON in a treated plot from a randomly selected study was lower than that in the check by a fixed margin was determined, which confirmed the superior efficacy of prothioconazole, metconazole, and tebuconzole+prothioconzole for Fusarium head blight disease and toxin control.  相似文献

ABSTRACT A meta-analysis of the effect of tebuconazole (e.g., Folicur 3.6F) on Fusarium head blight and deoxynivalenol (DON) content of wheat grain was performed using data collected from uniform fungicide trials (UFTs) conducted at multiple locations across U.S. wheat-growing regions. Response ratios (mean disease and DON levels from tebuconazole-treated plots, divided by mean disease and DON levels from untreated check plots) were calculated for each of 139 studies for tebuconazole effect on Fusarium head blight index (IND; field or plot-level disease severity, i.e., mean proportion of diseased spikelets per spike) and 101 studies for tebuconazole effect on DON contamination of harvested grain. A random-effects meta-analysis was performed on the log-transformed ratios, and the estimated mean log ratios were transformed to estimate the mean (expected) percent control for IND ( C(IND) ) and DON ( C(DON)). A mixed effects meta-analysis was then done to determine the effects of wheat type (spring versus winter wheat) and disease and DON levels in the controls on the log ratios. Tebuconazole was more effective at limiting IND than DON, with C(IND) and C(DON) values of 40.3 and 21.6%, respectively. The efficacy of tebuconazole as determined by the impact on both IND and DON was greater in spring wheat than in winter wheat (P < 0.01), with a 13.2% higher C(IND) and a 12.4% higher C(DON) in spring wheat than in winter wheat. ABSTRACT A total of 126 field studies reporting deoxynivalenol (DON; ppm) content of harvested wheat grain and Fusarium head blight index (IND; field or plot-level disease severity) were analyzed to determine the overall mean regression slope and intercept for the relationship between DON and IND, and the influence of study-specific variables on the slope and intercept. A separate linear regression analysis was performed to determine the slope and intercept for each study followed by a meta-analysis of the regression coefficients from all studies. Between-study variances were significantly (P < 0.05) greater than 0, indicating substantial variation in the relationship between the variables. Regression slopes and intercepts were between -0.27 and 1.48 ppm per unit IND and -10.55 to 32.75 ppm, respectively. The overall mean regression slope and intercept, 0.22 ppm per unit IND and 2.94 ppm, respectively, were significantly different from zero (P < 0.001), and the width of the 95% confidence interval was 0.07 ppm per unit IND for slope and 1.44 ppm for intercept. Both slope and intercept were significantly affected by wheat type (P < 0.05); the overall mean intercept was significantly higher in studies conducted using winter wheat cultivars than in studies conducted using spring wheat cultivars, whereas the overall mean slope was significantly higher in studies conducted using spring wheat cultivars than in winter wheat cultivars. Study location had a significant effect on the intercept (P < 0.05), with studies from U.S. winter wheat-growing region having the highest overall mean intercept followed by studies from Canadian wheat-growing regions and U.S. spring wheat-growing regions. The study-wide magnitude of DON and IND had significant effects on one or both of the regression coefficients, resulting in considerable reduction in between-study variances. Seven spring wheat, 13 barley, 14 oats and 20 winter wheat genotypes were inoculated at flowering in 1993 and 1994 with mixed conidial suspensions of 8 isolates of Fusarium culmorum. Four weeks after inoculation, head blight was recorded in the field. After harvest, seed infection was assessed by a Freezing Blotter Test in the laboratory. Seed samples were also analyzed for deoxynivalenol (DON) content. Differences were found in head blight rating, the levels of infected seeds and the DON content between wheat, barley, and oats and between cultivars. Highly significant correlations were found between the percentage of heavily infected seed and the DON content. The weighted mean value of infected seeds and DON content were also significantly correlated. No significant correlation was found between head blight rating in the field and DON content. The level of infected seeds, as determined by the Freezing Blotter Test, was a better indication of the DON content in the seeds than the head blight rating. This visual assessment of levels of infected seeds gives a reliable estimate of resistance to Fusarium.  相似文献

小麦赤霉病是小麦穗期的主要病害之一。化学防控一直是小麦主产区防控赤霉病的主要措施。为明确几种新型杀菌剂对小麦赤霉病的防效和对小麦籽粒DON毒素含量的影响,于2018年进行了氰基丙烯酸酯类和三唑类杀菌剂单剂及其复配剂对赤霉病的防效试验。结果表明:30%戊唑·多菌灵悬浮剂(SC)1500 mL/hm^2处理对赤霉病病穗防效达92.40%,病指防效达93.20%,小麦籽粒DON毒素检出量较不用药对照降低80.38%;25%氰烯菌酯SC 2000 mL/hm^2处理对赤霉病的病穗防效达86.80%,病指防效达88.78%,小麦籽粒DON毒素检出量较不用药对照降低88.19%;48%氰烯·戊唑醇SC 900 mL/hm^2和40%丙硫·戊唑醇SC 600 mL/hm^2对小麦赤霉病的病穗防效分别为77.20%、78.00%,病指防效分别为80.27%和79.59%,对籽粒DON毒素检出量较不用药对照分别降低73.87%和81.42%。在小麦赤霉病较重发生的情况下,上述4种杀菌剂单剂或复配剂1次用药既能高效控制病情,又能有效控制小麦籽粒DON毒素不超标。本试验研究进一步阐明,氰烯菌酯、戊唑醇、丙硫菌唑等杀菌剂及其复配剂均能有效控制小麦赤霉病的危害,并能有效降低小麦籽粒DON毒素含量;吡唑醚菌酯单剂及其复配剂虽然对小麦赤霉病的病穗和病指防效也较高,但控制小麦籽粒DON毒素含量效果相对较差。研究结果为小麦穗期赤霉病化学防控提供了科学参考。  相似文献

A three-year field experiment with two wheat cultivars evaluated the effect of soil-applied silicon (Si), with and without fungicide spraying, on Fusarium head blight (FHB) control. Silicon treatment alone reduced FHB severity and the percentage of damaged wheat kernels, regardless of the cultivar. The best disease control was obtained for the cultivar with moderate disease resistance (MR), supplied with silicon and treated with fungicide during flowering. Silicon treatment alone promoted an increase in deoxynivalenol (DON) concentration in the disease-susceptible cultivar; however, in the MR cultivar, silicon amendment associated with fungicide treatment led to a reduction in DON concentration. Greenhouse experiments evaluated the effect of silicon combined with different timings of fungicide application on wheat defences against Fusarium graminearum. Plants supplied with silicon had a longer pathogen incubation period, lower FHB severity and lower DON concentration when compared to plants without silicon. In addition, silicon-supplied plants had higher soluble phenolic content and altered antioxidant enzyme activities (SOD, CAT, POX and PPO) that favoured early accumulation of hydrogen peroxide when compared to plants without silicon. Greater control of FHB and lower DON concentration in plants treated with silicon and fungicide before inoculation and up to 1 day after inoculation was associated with increased levels of defence-associated metabolites. Silicon contributed to the reduction of FHB and DON concentration in wheat, especially for the MR cultivar and, when combined with fungicide spraying, both MR and disease-susceptible cultivars had enhanced performances upon silicon amendment.  相似文献

Gibberella zeae (anamorph Fusarium graminearum ) causes head blight of cereals and contaminates grains with mycotoxins such as deoxynivalenol (DON). To determine the correlations among aggressiveness traits, fungal colonization and DON production, 50 progeny from a segregating population of G. zeae were inoculated onto a susceptible winter wheat cultivar in three field environments (year–location combinations). Aggressiveness traits were measured as head-blight rating and plot yield relative to noninoculated plots. Fungal colonization, measured as Fusarium exoantigen (ExAg) content, and DON production were analysed with two ELISA formats. Disease severity was moderate to high based on head-blight rating and relative plot yield. Fusarium ExAg content and DON production ranged from 0·26–1·41 units and from 4·18–43·70 mg kg−1, respectively. Significant ( P  = 0·01) genotypic variation was found for all traits. Heritability for Fusarium ExAg content was rather low because of high progeny–environment interaction and error. DON/ Fusarium ExAg ratio did not vary significantly ( P  > 0·1) among progeny. Correlation between DON production and Fusarium ExAg content across environments was high ( r  = 0·8, P  = 0·01), but no covariation existed between aggressiveness traits and DON/ Fusarium ExAg content ratio.  相似文献

In Europe and North America, deoxynivalenol (DON) is the most prevalent mycotoxin associated with wheat head blight caused by Fusarium graminearum and Fusarium culmorum. Because DON is toxic to plants and enhances the ability of the pathogen to spread within a spike, wheat lines with resistance to DON should be more resistant to head blight. Resistance to DON has been associated with resistance gene Fhb1 that confers resistance to spread within a spike. The objectives of this study were to determine if wheat lines resistant to head blight were also resistant to DON, if genes other than Fhb1 confer resistance to DON, and to identify lines able to fill grain in the presence of DON. Susceptible controls and diverse North American and European winter wheat lines with resistance to head blight were screened for molecular markers linked to known head blight resistance genes, and evaluated in a greenhouse for resistance to DON and relative yield after application of DON to spikes at flowering. Fhb1 appeared to have the unique ability to confer resistance to DON, as measured by the number of DON‐bleached primary florets. However, this resistance did not protect plants from the phytotoxic effects of DON on kernel formation as measured by the relative yield of treated spikes. Furthermore, measuring the relative yield loss following DON application may be useful for identifying lines with tolerance to head blight.  相似文献

Fusarium head blight of cereals has, in recent years, become one of the most important pre-harvest diseases worldwide. This paper examines the in vitro efficacy of fungicides to control Fusarium species in cereals and the efficacy in the field on both Fusarium infection of ripening ears as well as their impact on mycotoxin production. Field studies suggest that fungicides such as tebuconazole and metconazole give good control of both Fusarium infection of ears and control of deoxynivalenol (DON) production. However, azoxystrobin and related fungicides are less effective, and grain from treated crops has sometimes been found to have increased concentrations of DON and nivalenol. Studies of isolates of Fusarium culmorum from different parts of Europe showed that complex interactions occur between environmental factors, fungicide type and isolate in relation to growth inhibition and DON production. These studies confirmed the ineffectiveness of azoxystrobin and suggest that environmental stress factors, particularly water availability and temperature, and low fungicide doses may stimulate mycotoxin production by Fusaria in vitro and in wheat grain.  相似文献

Glasshouse studies were undertaken to determine if fungicides used for the control of Fusarium head blight (FHB) result in elevated concentrations of the trichothecene mycotoxin, deoxynivalenol (DON) in harvested wheat grain. Metconazole and azoxystrobin, at double, full, half or quarter the manufacturer's recommended dose rate, were applied to ears of wheat (cv. Cadenza), artificially inoculated with conidia of either Fusarium culmorum or F. graminearum. Metconazole demonstrated high activity against both pathogens, reducing significantly the severity of FHB and the DON concentrations at each of the four dose rates tested when compared to untreated controls. Applications of azoxystrobin significantly reduced FHB and DON compared to unsprayed controls. However, their effectiveness was significantly less than that of metconazole and no dose rate response was observed. Quantification of the amount of trichothecene-producing Fusarium present in harvested grain was determined using a competitive PCR assay based on primers derived from the trichodiene synthase gene (Tri5). Simple linear regression analyses revealed strong relationships between the amount of trichothecene-producing Fusarium present in grain and the DON concentrations (r 2=0.72–0.97). It is concluded that fungicides, applied for the control of FHB, affect DON concentrations indirectly by influencing the amount of trichothecene-producing Fusarium species present in wheat grain. There was no evidence that fungicide applications directly increase the concentration of DON in grain.  相似文献

Summary An infection of bread wheat by fusarium head blight contaminates the crop with mycotoxins, particularly deoxynivalenol (DON) and nivalenol (NIV). The toxicity and natural occurrence of these mycotoxins in wheat are reviewed. Based on 8 years data of fusarium head blight epidemics of wheat in the Netherlands, DON contamination of the grain was estimated. Fusarium head blight ratings averaged an infection of 1.7% of all spikelets; estimates for DON contamination averaged 0.9 mg kg–1. Taking a guideline level for DON in uncleaned bread wheat of 2 mg kg–1, in 1979 and 1982 a wheat crop was produced with estimated DON concentrations above the limit of tolerance. Human and animal exposure to mycotoxins in the Netherlands appears to be small but chronic. The information presented in this paper illustrates the need for an annual evaluation of the crop for fusarium head blight incidence and mycotoxin content, and the necessity of fusarium head blight resistant wheat cultivars.Samenvatting Aaraantasting van tarwe doorFusarium culmorum enFusarium graminearum leidt tot vorming van mycotoxinen in het graan, waarvan deoxynivalenol (DON) en nivalenol (NIV) de belangrijkste toxinen zijn. In dit artikel wordt een overzicht gegeven van de toxicologische aspecten, en het voorkomen van deze toxinen in tarwe. Informatie over DON en NIV in tarwe in West-Europa is schaars. Gebaseerd op gegevens vanFusarium epidemieën in de jaren 1979–1986 wordt een schatting gegeven van de concentratie DON in Nederlandse tarwe. Rekening houdend met de herkomst en verwerking van tarwe, blijken zowel in dierlijk als menselijk voedsel lage concentraties DON chronisch voor te komen. Op basis van een maximaal toelaatbare dagelijkse dosis DON van 3 g kg–1 lichaamsgewicht is de schatting van de dagelijkse opname van DON in het jaar volgend op de oogst van 1982 net op de grens. Fusarium culmorum causes head blight, produces toxins and reduces yield and quality of cereals. To prevent damage caused by fusarium head blight (FHB), azole fungicides are mainly applied. The occurrence of insensitivity to azoles is a major problem in agriculture. The present study shows that a tebuconazole insensitive strain of F. culmorum can be readily produced in the laboratory, but that the resulting strain of the fungus is of lower fitness in vitro. Insensitivity was confirmed microscopically and by cell viability and metabolic activity. The tebuconazole insensitive strain shows cross insensitivity to nine important azoles. In addition, plants inoculated with the insensitive F. culmorum strain showed no reduction of FHB symptoms and deoxynivalenol (DON) content after tebuconazole treatment, compared to an inoculation with the sensitive strain. Use of wheat cultivars carrying a high resistance level (i.e. cv. Toras) was the most effective method for reducing symptoms and decreasing DON content, independent from the level of fungicide insensitivity of the F. culmorum strain. In conclusion, resistant cultivars and a fungicide mixture which combines different mechanisms of action in fungal metabolism should be applied to avoid fungicide insensitivity of Fusarium spp. in future.  相似文献

Measurements of local environmental conditions, intensity of Fusarium head blight (FHB) in wheat spikes, biomass of Fusarium graminearum, F. culmorum, and F. poae (pathogens causing FHB) and concentration of the mycotoxins deoxynivalenol (DON) and nivalenol (NIV) in harvested wheat grain were obtained in a total of 150 location-years, originating in three European countries (Hungary, Ireland, United Kingdom) from 2001 to 2004. Through window-pane methodology, the length and starting time of temporal windows where the environmental variables were significantly associated with the biological variables were identified. Window lengths of 5 to 30?days were evaluated, with starting times from 18?days before anthesis to harvest. Associations were quantified with nonparametric Spearman correlation coefficients. All biological variables were significantly associated with at least one evaluated environmental variable (P?≤?0.05). Moisture-related variables (e.g., average relative humidity, hours of relative humidity above 80%) had the highest positive correlations with the biological variables, but there also was a significant negative correlation between average temperature and several biological variables. When significant correlations were found, they were generally for all window lengths, but for a limited number of window start times (generally before anthesis for disease index and after anthesis for the toxins and late-season fungal biomasses). Semi-partial Spearman correlation coefficients were used to evaluate the relationship between the environmental variables and the concentration of DON and NIV after the effects of FHB intensity and fungal biomass on the mycotoxins were removed. Significant semi-partial correlations were found between relative humidity variables and DON, and between temperature and relative humidity variables and NIV for time windows that started after anthesis (and not for any earlier time windows). Results confirm that the environment influences disease, fungal biomass, and mycotoxin production, and help refine the time windows where the association is greatest. However, variability in the relationships was high, indicating that no single environmental variable is sufficient for prediction of disease or mycotoxin contamination.  相似文献

以多菌灵为主的苯并咪唑类杀菌剂一直是小麦抽穗扬花期防控赤霉病的主要手段之一。本研究对2018年我国主要麦区采集的1 464株赤霉病菌菌株进行多菌灵抗性分子检测。共检测出多菌灵抗性菌株97株,抗性频率为6.63%,同时发现抗性菌株以F167Y突变频率最高,其次为E198Q和F200Y。通过比较不同省份间多菌灵抗性发生频率发现,长江中下游麦区赤霉病菌群体抗性频率明显高于黄淮麦区群体。本研究相比之前研究中的抗性频率大幅度上升,表明在多菌灵的选择压力下,多菌灵抗性种群发展迅速。为防止抗性群体的进一步发展,致使多菌灵防治赤霉病失效,应采用混配、复配药剂、不同作用机理的杀菌剂交替轮换使用来防治小麦赤霉病。  相似文献

Different sets of wheat genotypes were tested under field conditions by spraying inocula of isolates of seven Fusarium spp. and Microdochium nivale (formerly F. nivale) in the period 1998–2002. The severity of Fusarium head blight (FHB), Fusarium-damaged kernels (FDK), the yield reduction and the deoxynivalenol (DON) contamination were also measured to describe the nature of the resistance. The degrees of FHB severity of genotypes to F. graminearum, F. culmorum, F. avenaceum, F. sporotrichioides, F. poae, F.␣verticillioides, F. sambucinum and M. nivale were very similar, indicating that the resistance to F.␣graminearum was similar to that for other Fusarium spp. listed. This is an important message to breeders as the resistance relates not only to any particular isolate of F. graminearum, but similarly to isolates of other Fusarium spp. This holds true for all the parameters measured. The DON contamination refers only to DON-producers F. graminearum and F. culmorum. Highly significant correlations were found between FHB, FDK, yield loss and DON contamination. Resistance components such as resistance to kernel infection, resistance to DON and tolerance were identified in the more susceptible genotypes. As compared with western European genotypes which produced up to 700 mg kg−1 DON, the Hungarian genotypes produced only 100 mg kg−1 at a similar FDK level. This research demonstrates the importance of measuring both FDK and DON in the breeding and selection of resistant germplasm and cultivars.  相似文献

Determination of the Fusarium protein equivalent (FPE) levels in kernels for better characterisation of genotypes showing Fusarium head blight (FHB) resistance, and better detection of susceptibility to kernel infection among genotypes with slight symptom expression was carried out. Twelve wheat cultivars and eight hexaploid winter wheat lines derived from a cross of Triticum aestivum with related species T. macha, T. polonicum, and T. dicoccoides were evaluated for levels of spike and kernel infection, the content of the mycotoxin deoxynivalenol (DON) and FPE in kernels after artificial inoculation with the fungus Fusarium culmorum in the field in 2006–2007. The ELISA immunochemical method was employed for the quantitative analyses of DON and FPE. Three wheat lines had a significantly low infection of spikes and kernels compared to cvs Sumai 3 and Nobeoka Bozu, indicating the presence of specific resistance mechanisms to FHB. The significantly low AUDPC (area under the disease progress curve) and the high level of FPE and DON content in kernels indicated a lack of resistance in one wheat line (crossed with T. polonicum). The results showed highly significant correlations (P < 0.01) between FPE and DON content and between FPE and AUDPC. In addition, correlations between FPE and reductions in yield components were also highly significant. Quantification of Fusarium spp. in wheat kernels can be helpful for evaluating wheat genotypes for their levels of resistance to FHB.  相似文献

由禾谷镰孢菌引起的赤霉病是小麦上的重要病害,可严重影响小麦的产量并降低小麦的品质。苯醚甲环唑属于三唑类杀菌剂,是甾醇脱甲基化抑制剂,具有较高的抑菌活性。采用菌丝生长速率法测定了于2016—2017采集自河南省的107株禾谷镰孢菌对苯醚甲环唑的敏感性。结果表明:苯醚甲环唑对禾谷镰孢菌菌丝的生长具有较强的抑制效果,其有效抑制中浓度(EC50)值范围为0.0128~0.6079 mg/L,符合正态分布,平均EC50值为(0.2239 ± 0.1192) mg/L。因此,这些敏感性数据可以作为河南省禾谷镰孢菌对苯醚甲环唑的敏感性基线。通过对苯醚甲环唑与其他7种杀菌剂氟环唑、多菌灵、氰烯菌酯、氟唑菌酰羟胺、戊唑醇、丙硫菌唑及叶菌唑对20株禾谷镰孢菌的log10 EC50值之间的Spearman’s rho (ρ) 相关性分析发现:苯醚甲环唑与叶菌唑之间具有较低水平的相关性,与其他供试杀菌剂之间无相关性。本研究可为监测河南省禾谷镰孢菌对苯醚甲环唑的抗药性发展和防控小麦赤霉病合理用药提供依据。  相似文献

赤霉病是我国小麦上的重要病害,品种抗病性利用是控制病害发生的重要措施,明确小麦抗赤霉病资源的抗性类型,有利于小麦抗赤霉病育种。2003年和2004年对9个常用抗源在穗期进行单花滴注和喷雾接种,研究其抗侵染和抗扩展性,并对病穗中的脱氧雪腐镰刀菌烯醇(DON)的含量进行分析。结果表明,望水白和苏麦3号具较好的抗侵染和抗扩展能力,其中望水白的抗扩展性最好;感染赤霉病后,DON在5个抗源穗组织中的含量差异显著,DON在望水白和繁60096穗组织中积累量明显比在苏麦3号、延岗坊主和翻山小麦低。通过对望水白/安农8455遗传群体两年的病小穗率和病穗中DON毒素含量的比较,发现二者具有一定的相关性,且受环境影响比较大。  相似文献

Fusarium head blight of wheat is caused by a disease complex comprised of toxigenic pathogens, predominantly Fusarium spp., and a non-toxigenic pathogen Microdochium nivale, which causes symptoms visually indistinguishable from Fusarium and is often included as a causal agent of Fusarium head blight. Four field trials are reported here, including both naturally and artificially inoculated trials in which the effect of fungicide treatments were noted on colonisation by Fusarium and Microdochium, and on the production of deoxynivalenol (DON) mycotoxin. The pathogen populations were analysed with quantitative PCR and samples were tested for the presence of the mycotoxin DON. Application of fungicides to reduce Fusarium head blight gave a differential control of these fungi. Tebuconazole selectively controlled F. culmorum and F. avenaceum and reduced levels of DON, but showed little control of M. nivale. Application of azoxystrobin, however, selectively controlled M. nivale and allowed greater colonisation by toxigenic Fusarium species. This treatment also lead to increased levels of DON detected. nobreak Azoxystrobin application two days post-inoculation increased the production of DON mycotoxin per unit of pathogen in an artificially inoculated field trial. This result indicates the potential risk of increased DON contamination of grain following treatment with azoxystrobin to control head blight in susceptible wheat cultivars. This is the first study to show differential fungicidal control of mixed natural pathogen populations and artificial inoculations in field trials.  相似文献

A total of 82 fungal isolates was obtained from wheat kernel samples affected by fusarium head blight collected from 20 locations in southern Brazil. Polymerase chain reaction (PCR) assays were used to characterize trichothecene mycotoxin genotypes [deoxynivalenol (DON), nivalenol (NIV) and two acetylated derivatives of DON]. To identify isolates that producing DON and NIV, portions of the Tri13 gene were amplified. To identify 3-acetyl-deoxynivalenol (3-ADON) and 15-acetyl-deoxynivalenol (15-ADON) genotypes, portions of Tri3 and Tri12 were amplified. Nearly all of the isolates studied (76/82) were of the DON/15-ADON genotype. Six of the isolates were of the NIV genotype. The DON/3-ADON genotype was not observed. Portions of three genes were sequenced from representative isolates of the NIV and DON/15-ADON genotypes and compared with sequences from curated reference isolates of Fusarium in GenBank. blast queries for individual gene sequences and pairwise comparisons of percentage identity and percentage divergence based on 1676 bp of concatenated DNA sequence suggested that the isolates representing the DON/15-ADON genotype were Fusarium graminearum sensu stricto and the isolates representing the NIV genotype were Fusarium meridionale . This is the first detailed report of trichothecene mycotoxin genotypes of F. graminearum and F. meridionale in Brazil.  相似文献
