ABSTRACT Berries of Vitis vinifera are reported to be susceptible to infection by Uncinula necator until soluble solids levels (brix) reach 8%, and established colonies are reported to sporulate until brix reach 15%. However, our analysis of disease progress on fruit of selected V. vinifera cultivars indicated that severity became asymptotic several weeks earlier in fruit development. When mildew-free fruit clusters of V. vinifera 'Chardonnay', 'Riesling', 'Gewürztraminer', and 'Pinot Noir' were inoculated at stages ranging from prebloom to 6 weeks postbloom, only fruit inoculated within 2 weeks of bloom developed severe powdery mildew. Substantial ontogenic resistance to infection was expressed in fruit nearly 6 weeks before fruit brix reached 8% and over 2 months before they reached 15%. Rachises of 'Chardonnay' and 'Riesling' fruit clusters developed severe powdery mildew when inoculated at bloom, and disease increased steadily over the next 60 days. The rachis of fruit clusters inoculated 31 days after bloom developed only trace levels of powdery mildew. Berry weight of all four cultivars at harvest was reduced when fruit clusters were inoculated at bloom or 16 days postbloom, primarily by splitting, rotting, and dehydration of mildewed berries, but the weight of later-inoculated berries was not reduced. Inoculation of berries just as ontogenic resistance increased markedly, approximately 3 to 4 weeks postbloom, resulted in the development of inconspicuous, diffuse, non-sporulating mildew colonies on berries, sometimes associated with a network of necrotic epidermal cells. Rather than a protracted and relatively static period of berry susceptibility lasting 3 months, fruit of V. vinifera appear to acquire ontogenic resistance rapidly after fruit set. A refocusing of disease management on this critical period of high fruit susceptibility should greatly improve the efficacy of fungicides directed against powdery mildew.  相似文献

ABSTRACT Vitis labruscana 'Concord' is a grape cultivar widely grown in the United States for processing into juice and other grape products. Concord grapes are sporadically but sometimes severely damaged by the grape powdery mildew pathogen, Uncinula necator. Although the foliage is often reported to be moderately resistant to powdery mildew, severe fruit infection occurs in some years. We observed the seasonal development of powdery mildew on leaves, rachises, and berries of unsprayed Concord grapevines. Inoculations of flower and fruit clusters revealed a brief period of berry susceptibility and a protracted period of rachis susceptibility. The rachis remained highly susceptible to infection, and the severity of rachis infection increased throughout the growing season until the rachis formed a periderm shortly before harvest. In contrast, berries were nearly immune to infection within 2 weeks after fruit set. Rachis and berry infections were detected before the disease was observed on foliage, and the incidence of rachis and berry infection often exceeded disease incidence observed on foliage until after fruit acquired substantial ontogenic resistance. Excellent control of fruit infection, and adequate control of leaf infection, was achieved by two fungicide applications targeted at the peak period of fruit susceptibility. Although Concord is thought to be moderately resistant to powdery mildew, the rachis is highly susceptible, and may be the avenue by which prebloom infections make their way onto the developing fruit. Late-season infection of the rachis neither spread to the fruit, nor did it cause fruit to drop prematurely, and may be of little economic consequence on fruit destined for processing. Although fruit of V. vinifera cultivars have been reported to remain susceptible to infection until berry sugar levels reach 8 to 15%, Concord fruit become nearly immune to infection nearly 6 weeks before this stage of development. Because powdery mildew does not become conspicuous on foliage until late summer, it is generally regarded as a late-season problem on Concord grapes, and previous management programs have reflected this belief. However, the greatest contribution to control of fruit infection is due to fungicides applied during the peak period of fruit susceptibility, from bloom until shortly after fruit set, long before the disease is observed on foliage.  相似文献

ABSTRACT The period of fruit susceptibility to Guignardia bidwellii (anamorph Phyllosticta ampelicida), the causal agent of grape black rot, was determined in the field. Intact fruit were inoculated weekly from bloom until 8 weeks later with a suspension containing 2 x 10(5) conidia per ml. Disease progress was monitored approximately every 2 days until 3 to 5 weeks after inoculation, depending on the year and variety. Fruit of Vitis x labruscana 'Concord' exhibited a period of maximum susceptibility from midbloom until 2 to 4 weeks later, although some berries became symptomatic when inoculated 4 to 5 weeks postbloom. Fruit of V. vinifera 'Chardonnay' and 'Riesling' exhibited a period of maximum susceptibility from midbloom until 3 to 5 weeks later, although some berries retained their susceptibility until 6 to 7 weeks postbloom. These susceptible periods were approximately 2 to 4 weeks shorter than previously assumed. Fruit age at the time of inoculation affected the length of the incubation period (time from inoculation until symptom appearance). When the incubation period was defined in terms of degree hours (base = 0 degrees C) accumulated after inoculation, DH(50) values (the number of degree hours required to reach 50% of final disease severity) increased by at least 50% as berries neared the end of their susceptible period. Newly symptomatic berries continued to appear for over 1 month after inoculation of older fruit. Thus, age-related or ontogenic, host resistance was manifested as both a decline in susceptibility and a significant increase in incubation period length. The control of black rot is likely to be improved by tailoring the intensity of fungicidal protection to the abbreviated period of fruit susceptibility defined in this study. Furthermore, the efficacy of management programs and the results of epidemiological studies are likely to be misinterpreted unless the variable effect of fruit age on incubation period length is recognized.  相似文献

ABSTRACT Grape berries become resistant to powdery mildew early in development and are nearly immune to infection within 4 weeks after bloom. In this study, ontogenic resistance did not reduce attachment, germination, or appressorium formation of Uncinula necator on 3- to 4-week-old berries of Vitis vinifera 'Chardonnay' or 3-week-old berries of V. labruscana 'Concord'. Pathogen ingress halted at the cuticle before formation of a penetration pore. As berries aged, hyphal elongation and colony growth slowed until finally no secondary hyphae formed on fully resistant berries. More appressoria formed per unit of hyphal length as berries aged, indicating that failure to penetrate older berries led to increased attempts to penetrate resistant fruit. Additionally, hyphae within the colonies began to die as berries aged. Finally, the number of degree-hours between germination and sporulation of the colony (latent period) increased and sporophore density decreased with berry age at time of inoculation. Thus, ontogenic resistance both slows, and eventually halts disease development on grape berries, and limits the likelihood of spread by reducing absolute supply of conidia and delaying their formation. It furthermore has a consistent, stable, and predictable impact on grape powdery mildew and operates in a similar fashion and to a similar degree in both V. labruscana and V. vinifera, although at a slightly earlier phenological stage in V. labruscana.  相似文献

ABSTRACT Resistance of mature berries of grapevine cultivars and selections to postharvest infection by Botrytis cinerea was assessed. Little or no resistance existed in most popular table grape Vitis vinifera cultivars, except in moderately resistant 'Emperor' and 'Autumn Black'. Highly resistant grapes were V. rotundifolia, V. labrusca, or other complex hybrids. Morphological, anatomical, and chemical characteristics of 42 genetically diverse cultivars and selections with various levels of resistance to B. cinerea were examined to determine which features were associated with resistance. We quantified the (i) density of berries within a cluster; (ii) number of pores and lenticels on the berry surface; (iii) thickness and number of cell layers in the epidermis and external hypodermis; (iv) amount of cuticle and wax; (v) berry skin protein content; (vi) total phenolic content of the skin before and after B. cinerea inoculation; and (vii) catechin and trans- and cis-resveratrol contents of the skin before and after inoculation. The number of pores was negatively correlated with resistance. Highly resistant cultivars had few or no pores in the berry surface. The number and thickness of epidermal and hypodermal cell layers and cuticle and wax contents were positively correlated with resistance. Other characteristics evaluated were not associated with resistance. trans-Resveratrol and cis-resveratrol were induced by B. cinerea inoculation only in sensitive and moderately resistant cultivars and selections.  相似文献

Bitter rot, caused by the fungus Greeneria uvicola, is one of the most important fruit rot diseases that threaten the burgeoning winegrape (Vitis vinifera) industry in the southeastern United States. Epidemiological studies were conducted to examine the period of fruit susceptibility of V. vinifera to G. uvicola, influence of temperature and duration of wetness on infection, and relative susceptibility of cultivars to bitter rot. In field studies, susceptibility of Merlot, Chardonnay, and Cabernet Franc fruit increased from bloom until véraison in 2003 and from bloom until 2 weeks before véraison in 2004. When detached V. vinifera fruit were inoculated and incubated at 14, 22, 26, and 30 degrees C for 6, 12, 18 or 24 h of wetness, 22.4 to 24.6 degrees C and 6 or 12 h of wetness were the optimal conditions for infection of fruit by G. uvicola. The relative susceptibility of 38 cultivars and selections, including 23 V. vinifera cultivars and five French-American hybrids, was determined in a detached fruit inoculation assay. A wide range in susceptibility was observed among the cultivars and selections. Fruit of cultivars of V. vinifera were significantly more susceptible than French-American hybrids. Isolates of G. uvicola differed in aggressiveness when tested on cv. Chardonnay.  相似文献

This work investigated the structural and biochemical changes during grape berry development which account potentially for the onset and increase in susceptibility to Botrytis cinerea. Using the cv. Sauvignon blanc, we quantified at seven developmental growth stages from herbaceous to over-mature berries: (1) fruit ontogenic resistance using three strains (II-transposa), (2) the morphological and maturity fruit characteristics and (3) preformed biochemical compounds located in the berry skin. From the mid-colour change stage onwards, susceptibility of unwounded fruit increased sigmoidally in both rot and sporulation severities at the berry surface. A principal component analysis identified a very close connection between fruit susceptibility and the level of fruit maturity. Berry susceptibility was significantly and positively correlated with the phenolic compounds in the skin cell walls and negatively correlated with the total tannin content in the skin and with water activity (Aw) at the fruit surface. On the berry, Aw decreased from 0.94 at bunch closure to 0.89 at berry maturity, with a relatively low value (0.90) at the stage of mid-colour change. Using artificial media, different Aw levels led to significant differences in mycelial growth (Aw ≤0.95 resulted in the lowest growth rate ≤0.34 mm day−1). Thus, besides the level of fruit maturity, both water activity on the fruit and the total tannin content in the skin may affect fungal growth and berry colonisation. The potential of these variables for use as indicators of grape berry susceptibility as well as associated mechanisms for the development of disease are discussed.  相似文献

ABSTRACT Little information is available concerning the expression of pathogenesis-related (PR) proteins in grapevine (Vitis vinifera) and their effect properties on the major fungal pathogens of grape. A systematic study was performed on the effect of total or individual grape proteins on mycelial growth, spore germination, and germ tube growth of Uncinula necator, Phomopsis viticola, and Botrytis cinerea. Two proteins, identified as PR proteins by immunological methods and by N-terminal sequencing as osmotin and thaumatin-like protein, exhibited strong antifungal activities in vitro, blocking the growth of Phomopsis viticola and Botrytis cinerea mycelia. In addition, they inhibited spore germination and germ tube growth of U. necator, Phomopsis viticola, and Botrytis cinerea. The presence of both proteins displayed a synergistic effect. The expression of osmotin and thaumatin-like protein was induced in grapevine leaves and berries infected with U. necator and Phomopsis viticola. Thaumatin previously was thought to occur exclusively in berries. Immunoblot analyses revealed the accumulation of the two PR proteins in infected leaves and berries, supporting a role in vivo in increasing the resistance of grapevine to fungal attack.  相似文献

ABSTRACT A fundamental principle of integrated pest management is that actions taken to manage disease should be commensurate with the risk of infection and loss. One of the less-studied factors that determines this risk is ontogenic, or age-related resistance of the host. Ontogenic resistance may operate at the whole plant level or in specific organs or tissues. Until recently, grape berries were thought to remain susceptible to powdery mildew (Uncinula necator) until late in their development. However, the development of ontogenic resistance is actually quite rapid in berries, and fruit become nearly immune to infection within 4 weeks after fruit set. Our objective was to determine how and at what stage the pathogen was halted in the infection process on ontogenically resistant berries. Adhesion of conidia, germination, and appressorium formation were not impeded on older berries. However, once berries were approximately 3 weeks old and older, few germlings were able to form secondary hyphae. Ontogenically resistant berries responded rapidly to infection by synthesis of a germin-like protein that had been previously shown to play a role in host defense against barley powdery mildew. On susceptible berries, cell discoloration around penetration sites indicated the oxidation of phenolic compounds; a process that was followed by localized cell death. However, the pathogen was still able to infect such cells prior to their death, continue secondary growth, and thereby colonize young berries. Formation of papillae was not involved in the differential resistance mechanism of older berries. In susceptible berries, papillae formed frequently at infection sites but did not always contain the pathogen, whereas in resistant berries, the pathogen was always halted prior to the formation of papillae. The host defense, which conditions ontogenic resistance, operates in the earliest stages of the infection process, in the absence of gross anatomical barriers, prior to the formation of a functional haustorium and prior to the development of a conspicuous penetration pore. We also found that diffuse powdery mildew colonies that were not visible in the field predisposed berries to bunch rot by Botrytis cinerea, increased the levels of infestation by spoilage microorganisms, and substantially degraded wine quality. Our improved understanding of the nature, causes, and stability of ontogenic resistance in the grapevine/ powdery mildew system has supported substantial changes in how fungicides are used to control the disease. Present applications are more focused on the period of maximum fruit susceptibility instead of following a calendar-based schedule. This has improved control, reduced losses, and in many cases reduced the number of fungicide applications required to suppress the disease. Particularly where fungicides are deployed in a programmatic fashion and ontogenic resistance is dynamic, there may be equivalent improvements to be made in other hostpathogen systems through studies of how host susceptibility changes through time.  相似文献

采用田间自然鉴定、田间接种鉴定及室内离体叶圆片接种鉴定三种方法,研究了10个欧亚种葡萄品种(系)、一个欧山杂种及其493株自交后代幼苗对葡萄霜霉病及白粉病的抗性及遗传关系,同时对其自交后代抗白粉病和霜霉病的遗传趋势进行了分析.结果表明,供试品种(系)及其自交后代对霜霉病与白粉病的抗性存在极显著相关,自交后代对两病表现伴随遗传现象,初步推断这可能是由葡萄抗病基因的多效性引起.在欧亚种葡萄品种自交后代群体中,虽然90%以上都是感病性中等或高的类型,但仍能够得到一定比例(10%以下)抗病性强的类型,这为利用欧亚种葡萄品种进行自交或品种间杂交选育优质抗病新品种提供了依据.  相似文献

The effects of different levels of inoculum of Aspergillus carbonarius and time of inoculation on berry infection and the development of aspergillus bunch rot on grapevines (cv. Sultana) were studied under field conditions. Inflorescences at full bloom were inoculated with aqueous spore suspensions of A. carbonarius containing 0 or 1 × 106 spores mL−1 in 2004/05 and 0, 1 × 102 or 1 × 105 spores mL−1 in 2005/06. In both years, the incidence of infection in inoculated berries was significantly higher than in uninoculated berries. Incidence of infection in berries from veraison until harvest was higher than at earlier stages of bunch development (berry set to berries that were still hard and green). Inoculation of bunches at veraison did not significantly increase A. carbonarius infection prior to harvest, at harvest, 6 days after harvest or when berries were over-ripe. Bunches inoculated at harvest did not significantly increase infection 6 days after harvest or when berries were over-ripe. Aspergillus carbonarius was isolated more frequently from the pedicel end (53·1%) than from the middle section (37·5%) and distal end (35·0%) of berries that were inoculated with 105 spores mL−1.  相似文献

Sphaeropsis rot, caused by Sphaeropsis pyriputrescens, is an important postharvest disease of apple in the United States. The objectives of this study were to determine the timing of apple fruit infection in the orchard in relation to development of Sphaeropsis rot in storage and to identify infection courts and mode of penetration by S. pyriputrescens on apple fruit. Fruit of apple cvs Red Delicious, Golden Delicious, and Fuji were inoculated in the orchard from 3 weeks after petal fall to 2 weeks before harvest at 5 to 6-week intervals in three consecutive seasons. All fruit were harvested and stored at 0?ºC to monitor decay development. Light and scanning electron microscopy were used to examine the infection courts and mode of penetration of the fungus on/in the host tissues. At harvest, the fungus was re-isolated from the stem (pedicel), sepal, anther, or filament of the inoculated fruit, but decay did not develop on fruit. Sphaeropsis rot developed on inoculated fruit during cold storage beginning 1–3 months after harvest. Stem-end rot was prevalent on cv. Golden Delicious, whereas calyx-end rot was prevalent on cv. Fuji. Both stem- and calyx-end rots were common on cv. Red Delicious. Infection also occurred at lenticels on fruit skin, particularly on cv. Golden Delicious, but at low incidence. Relationships between the incidence of Sphaeropsis rot in stored apple fruit and the timing of inoculation in the orchard varied with cultivar and year. On cv. Red Delicious apples, the incidence of Sphaeropsis rot generally increased as the timing of infection approached harvest. Histological studies indicated that infection took place through natural openings of plant organs such as stomata on stems and sepals and lenticels on fruit skin. Fungal penetration also was observed at micro-cracks on the stem and sepal and at trichome sockets where mechanical damage occurred in sepals. Direct penetration was observed on the stem and sepal of fruit, but most invasions were restricted between the cuticle and the epidermis. Our results indicate that wounding is not required for infection of apple fruit by S. pyriputrescens, though it may facilitate infections.  相似文献

 以霜霉病抗性葡萄品种左优红和感病品种霞多丽为材料,研究硫化氢(H2S)在葡萄应答霜霉病过程中的作用及其与过氧化氢(H2O2)的相互关系。结果表明,接种霜霉病菌后,抗性品种左优红和感病品种霞多丽叶片均出现H2S含量的猝发、H2S合成酶基因表达量增加、抗病相关蛋白多酚氧化酶(PPO)和β-1,3葡聚糖酶(Glu)活性显著升高现象;且H2S清除剂次牛磺酸(HT)可以显著抑制霜霉病菌诱导的PPO和Glu活性升高,并导致葡萄的感病率和病情指数显著升高;同时外源H2S可降低两个葡萄品种的感病率和病情指数。说明,H2S是参与葡萄抗霜霉病过程的新的信号物质。受霜霉病菌侵染后,葡萄叶片H2O2含量猝发早于H2S,并且H2O2清除剂抗坏血酸(AsA)可显著抑制霜霉病菌所诱导的H2S含量的上升,而H2S清除剂HT对霜霉病菌诱导的H2O2含量变化影响不显著,推测,H2S作用于H2O2的下游参与葡萄抵御霜霉病过程。  相似文献

ABSTRACT The course of colonization of leaf mesophyll by the causal agent of grapevine downy mildew, Plasmopara viticola, in a susceptible and a resistant grapevine genotype was examined in order to characterize the development of the pathogen in compatible and incompatible host-pathogen interactions. Within a few hours after inoculation, the pathogen was established in the susceptible Vitis vinifera cv. Müller-Thurgau and formed primary hyphae with a first haustorium. No further development occurred in the following 10 to 18 h. The next step, in which the hyphae grew and branched to colonize the intercellular space of the host tissue, was observed 1.5 days after inoculation. After 3 days, the intercostal fields were entirely filled with mycelium and sporulation was abundant under favorable environmental conditions. The first infection steps were essentially the same in the resistant V. rupestris. However, the invasive growth of P. viticola was delayed, and further development ceased before the intercostal fields were filled with mycelium.  相似文献

Summary. Solanum ptycanthum (Dun.) vegetative and reproductive growth was evaluated under shade and in the field with soybean interference. In full sunlight without soybean interference, a S. ptycanthum plant produced 243 g shoots and 5957 berries in 20 weeks, but only 38 g and 576 berries in 11 weeks. Plants grown in 94% shade produced 3 g of shoots and 23 berries in 20 weeks, and 1 g and 1 berry in 11 weeks. S. ptycanthum plants that emerged with the soybeans and were positioned between 75-cm rows produced 43 g shoots and 264 berries, whereas plants positioned in 75-cm rows and emerging 6 weeks after the soybeans produced 1 g and 16 berries. When grown between 37·5-cm rows for the same period of time this weed produced 12 g and 2 berries plant−1; plants positioned in 37·5-cm rows produced shoots less than 1 g in weight and one berry plant−1. Shoot growth and berry production of S. ptycanthum increased from 80 to 200% in a 2-week period between the initiation of soybean leaf abscission and maturity. In a season about 50 000 seeds weed−1 were produced in full sunlight; 20 000 or less were produced under soybean interference. When grown in irradiance levels from full sunlight to 94% shade, viable seeds were first detected 10 days after anthesis and essentially all seeds in the berry were viable 24 days after anthesis  相似文献

ABSTRACT Naturally established lowbush blueberry clones in four fields were evaluated for the incidence of leaf and flower blight, proportion of mummy berries, and yield reductions caused by Monilinia vacciniicorymbosi. The relationship between the phenology of flower and leaf bud development and susceptibility also was examined. Three fields were examined over one crop year and one field was studied in two subsequent crop years. The incidence of stems with blight was correlated to incidence of leaf blight in all fields and to incidence of flower blight in one field. Incidence of leaf and flower blight and the proportion of mummy berries produced were not correlated. Lowbush blueberry clones with higher incidence levels of leaf blight had reduced fruit set and lower berry weights. For healthy stems, leaf-to-fruit ratios had no effect on berry weight in most fields. In contrast, blighted stems with higher leaf-to-fruit ratios had higher berry weights in three fields. Stems with slowerdeveloping leaf and flower buds had less leaf and flower blight, respectively, than stems with faster bud development. Some blueberry clones may avoid infection by delaying production of susceptible tissue until after ascospore release by M. vaccinii-corymbosi.  相似文献

ABSTRACT Inflorescences of field-grown grapevines (Vitis vinifera L. cv. Gamay) were inoculated with a Botrytis cinerea conidia suspension or dried conidia at different stages during bloom in moist weather. Approximately 10% of the conidia germinated within 72 h, resulting in two to three times more latent infections than uninoculated controls in pea-size (7 mm in diameter) berries. In surface-sterilized pea-size berries, latent B. cinerea was present predominantly in the receptacle area. After veraison, latent B. cinerea also was found in the style and, in mature berries, latent colonies were distributed throughout the pulp. Inoculation at full bloom led to the highest disease severity (66%) at harvest, compared with 38% in controls. Stilbene stress metabolites in the flowers were measured by high-performance liquid chromatography. Resveratrol accumulated mainly after pre-bloom and full-bloom inoculation, but did not prevent infection. Piceid levels did not change following inoculation, while epsilon-viniferin was found in necrotic tissues only, and pterostilbene and alphaviniferin were not detected at all. B. cinerea conidia suspensions also were applied to various locations on flowers of pot-grown cvs. Pinot Noir and Chardonnay. Inoculation of the receptacle area, but not that of the stigma and ovary, resulted in latent infections. Stilbene synthesis was similar to the field results, with resveratrol accumulating mainly in the calyptra and receptacle area. Constitutive soluble phenolic compounds (mainly derivatives of quercetin and hydroxy-cinnamic acid) were present at high concentrations in the calyptra but at low levels in the receptacle area. These experiments confirmed bloom as a critical time for B. cinerea infection in grapes and suggest that the most likely site of infection is the receptacle area or cap scar exposed at anthesis. Stilbenes may have a limited role in inhibition of flower infection and latency in susceptible grape cultivars, and epsilon-viniferin may be a by-product rather than a deterrent of infection.  相似文献

ABSTRACT Production of grape (principally cultivars of Vitis vinifera) for high-quality wines requires a high level of suppression of powdery mildew (Uncinula necator syn. Erysiphe necator). Severe infection of either fruit or foliage has well-documented and deleterious effects upon crop and wine quality. We found that berries nearly immune to infection by U. necator due to the development of ontogenic resistance may still support diffuse and inconspicuous mildew colonies when inoculated approximately 3 weeks post-bloom. Fruit with diffuse mildew colonies appear to be healthy and free of powdery mildew in late-season vineyard assessments with the naked eye. Nonetheless, presence of these colonies on berries was associated with (i) elevated populations of spoilage microorganisms; (ii) increased evolution of volatile ethyl acetate, acetic acid, and ethanol; (iii) increased infestation by insects known to be attracted to the aforementioned volatiles; (iv) increased rotting by Botrytis cinerea; and (v) increased frequency of perceived defects in wines prepared from fruit supporting diffuse powdery mildew colonies. Prevention of diffuse infection requires extending fungicidal protection until fruit are fully resistant to infection. Despite a perceived lack of improvement in disease control due to the insidious nature of diffuse powdery mildew, potential deleterious effects upon crop and wine quality thereby would be avoided.  相似文献

Following inoculation of host and nonhost plants with Plasmopara viticola, the grapevine downy mildew, a histological survey was undertaken to identify the stage where its development is contained in nonhosts and in resistant host plants. Three herbaceous nonhost species, Beta vulgaris, Lactuca sativa, and Capsicum annuum, and three grapevine species displaying different level of resistance (Vitis vinifera [susceptible], Vitis riparia [partially resistant] and Muscadinia rotundifolia [totally resistant]) where inoculated by P. viticola using a controlled leaf disk inoculation bioassay. During the early steps of infection, defined as encystment of zoospores on stomata, penetration of the germ tube, and production of the vesicle with the primary hypha, there was no evidence of a clear-cut preference to grapevine tissues that could attest to host specificity. The main difference between host grapevine species and nonhosts was observed during the haustorium formation stage. In nonhost tissues, the infection was stopped by cell wall-associated defense responses before any mature haustorium could appear. Defense responses in resistant grapevines were triggered when haustoria were fully visible and corresponded to hypersensitive responses. These observations illustrate that, for P. viticola, haustorium formation is not only a key stage for the establishment of biotrophy but also for the host specificity and the recognition by grapevine resistance factors.  相似文献

ABSTRACT Grape berries are highly susceptible to powdery mildew 1 week after bloom but acquire ontogenic resistance 2 to 3 weeks later. We recently demonstrated that germinating conidia of the grape powdery mildew pathogen (Uncinula necator) cease development before penetration of the cuticle on older resistant berries. The mechanism that halts U. necator at that particular stage was not known. Several previous studies investigated potential host barriers or cell responses to powdery mildew in berries and leaves, but none included observation of the direct effect of these factors on pathogen development. We found that cuticle thickness increased with berry age, but that ingress by the pathogen halted before formation of a visible penetration pore. Cell wall thickness remained unchanged over the first 4 weeks after bloom, the time during which berries progressed from highly susceptible to nearly immune. Autofluorescent polyphenolic compounds accumulated at a higher frequency beneath appressoria on highly susceptible berries than on highly resistant berries; and oxidation of the above phenolics, indicated by cell discoloration, developed at a significantly higher frequency on susceptible berries. Beneath the first-formed appressoria of all germinated conidia, papillae occurred at a significantly higher frequency on 2- to 5-day-old berries than on 30- to 31-day-old fruit. The relatively few papillae observed on older berries were, in most cases (82.8 to 97.3%), found beneath appressoria of conidia that had failed to produce secondary hyphae. This contrasted with the more abundantly produced papillae on younger berries, where only 35.4 to 41.0% were located beneath appressoria of conidia that had failed to produce secondary hyphae. A pathogenesis-related gene (VvPR-1) was much more highly induced in susceptible berries than in resistant berries after inoculation with U. necator. In contrast, a germin-like protein (VvGLP3) was expressed within 16 h of inoculation in resistant, but not in susceptible berries. Our results suggest that several putative barriers to infection, i.e., cuticle and cell wall thickness, antimicrobial phenolics, and two previously described pathogenesis-related proteins, are not principal causes in halting pathogen ingress on ontogenically resistant berries, but rather that infection is halted by one or more of the following: (i) a preformed physical or biochemical barrier near the cuticle surface, or (ii) the rapid synthesis of an antifungal compound in older berries during the first few hours of the infection process.  相似文献
