为分析福建黄兔、新西兰兔、日本大耳白兔群体内的遗传变异情况和群体间的遗传相关性,本试验选取3个品种兔各25只,分别耳静脉采血并采用试剂盒抽提全血基因组,运用15个微卫星标记,结合荧光PCR技术和毛细管电泳方法获得目的片段,通过计算等位基因数、有效等位基因数、杂合度、多态信息含量和遗传距离,分析3个品种兔的遗传多样性。结果显示,3个品种兔在15个座位共检测到110个等位基因,平均等位基因数为7.3个,其中福建黄兔、新西兰兔和日本大耳白兔分别检测到95、95和88个等位基因,且分别在11、12和11个微卫星座位表现为高度多态,表明3个品种兔在筛选的15个微卫星座位均呈现较丰富的遗传多态信息。Hardy-Weinberg遗传平衡检验结果表明,福建黄兔、新西兰兔和日本大耳白兔分别有8、8和6个座位处于Hardy-Weinberg不平衡状态。Nei氏遗传距离分析显示,福建黄兔与新西兰兔、福建黄兔与日本大耳白兔、新西兰兔与日本大耳白兔的Nei氏遗传距离分别为0.1761、0.2347和0.0432。遗传距离越小时,相似度越高,亲缘关系越近,因此,新西兰兔与日本大耳白兔间的亲缘关系最近,福建黄兔与日本大耳白兔间的亲缘关系最远。  相似文献

新疆塔里木马鹿遗传多样性的微卫星分析   总被引:3,自引:1,他引:3  
利用5个微卫星标记对新疆塔里木马鹿遗传多样性进行了检测。统计了塔里木马鹿3个群体的等位基因组成、平均有效等位基因数(E)和平均基因纯合率(Rh),利用等位基因频率计算出各群体的平均遗传杂合度(h)、多态信息含量(PIC)。结果表明5个微卫星位点在库尔勒、阿拉尔和沙雅县塔里木马鹿3个群体的平均多态信息含量分别为0.5884、0.5754、0.5344,除微卫星位点BM5004外均为高度多态,可作为有效的遗传标记用于塔里木马鹿遗传多样性分析;塔里木马鹿三个群体总平均PIC、h、Rh和E分别为:0.5661、0.5995、0.4474和2.7。分析认为塔里木马鹿遗传变异度较高,遗传多样性相对丰富,具有较大的遗传潜力。  相似文献

Genetic variation at 59 protein coding loci (16 polymorphic) and 25 microsatellite loci was analysed for 11 indigenous south-east Asian goat populations, and the Australian feral population, to determine the magnitude of genetic differentiation and the genetic relationships among the populations. Significant deviations from Hardy–Weinberg equilibrium were detected in one or more populations for eight of the nine protein loci with codominant alleles, and for microsatellites for all except the two Sri Lankan populations and for all but four loci. For both marker types, average inbreeding coefficients ( F IS) were exceptionally high. Heterogeneity of deviations from Hardy–Weinberg equilibrium for the microsatellites showed no differences for among loci within populations as compared with among populations within loci. For protein loci, however, the former was higher, indicating selection affecting allele frequencies at some loci. The variance among protein loci was significantly higher than among microsatellite loci, further indicating selection at some protein loci. There was significant differentiation among populations for both protein and microsatellite loci, most likely reflecting the geography of south-east Asia, and the presumed spread of goats throughout the region. Phylogenies derived from pair-wise genetic distance estimates show some similar clustering for the microsatellite and protein based trees, but bootstrap support was generally low for both. A phylogeny based on the combined set of 38 protein and microsatellite loci showed better consistency with geography and higher bootstrap values. The genetic distance phylogeny and the Weitzman diversity tree derived from microsatellite data showed some identical clusters, and both identified the Ujung Pandang and Australia populations as contributing most to overall genetic diversity.  相似文献

塔里木马鹿微卫星遗传多样性与产茸量的相关性研究   总被引:1,自引:0,他引:1  
利用7个微卫星遗传标记,采用PCR扩增,12%非变性聚丙烯酰胺凝胶电泳、Sanguinetti银染法显色,对新疆塔里木马鹿亚种遗传多样性进行检测,统计各群体的等位基因组成、平均有效等位基因数(E)和平均基因纯合率(Rh),根据等位基因频率计算出各群体的平均遗传杂合度(h)、多态信息含量(PIC)和群体间的遗传距离,利用分子进化遗传分析软件(MEGA),采用邻结法(NJ)重建系统发生树。根据等位基因频率,利用PHYLIP(3.6)分析软件,采用最大似然法(ML)构建系统发生树,应用自举检验(bootstrap test)估计系统树中节点的自引导值(bootstrap value),并进行了系统发生分析。同时对部分马鹿群体个体基因型与产茸量之间进行了相关分析。结果表明:7个微卫星位点在3个塔里木马鹿中的多态信息含量除BMS2508和Celjp0023没有多态性,以及BM5004为中度多态外,其他4个微卫星均为高度多态,可作为有效的遗传标记用于3个塔里木马鹿群体遗传多样性和系统发生关系的分析。所有马鹿群体的平均PIC为(0.5196)、h(0.5552)和E(2.45),其基因多态性和遗传多样性相对丰富。塔里木马鹿35团群体与沙雅群体的血缘关系及遗传距离近于阿拉尔群体,3个塔里木马鹿群体的系统发育关系基本符合其地理分布和育成史。微卫星位点BM4208的166bp/185bp基因型和微卫星BM888的208bp/208bp基因型可以对塔里木马鹿35团群体高产茸量进行分子遗传标记。  相似文献

A total of 10 fluorescent labeling microsatellite markers detected by capillary electrophoresis were selected to analyze the genetic diversity of tree shrew(T.belangeri chinensis).Microsatellite Toolkit and PopGene softwares were used to calculate the genetic correlation indexes to assess the genetic diversity of the population.64 alleles with 6.4 alleles per locus were identified among 10 microsatellite loci.The average effective number of alleles, observed heterozygosity, expected heterozygosity and polymorphic information content were 3.7892, 0.6156, 0.6963, and 0.6367, respectively.Seven of all loci were high polymorphic, and three were moderate polymorphic.Five loci extremely significantly deviated from Hardy-Weinberg equilibrium(P<0.01).These novel polymorphic microsatellite markers indicated high genetic diversity, which will be used to evaluate genetic diversity in tree shrew.  相似文献

选取10个微卫星位点,结合荧光标记(FAM) PCR扩增技术,运用毛细管电泳检测扩增产物,利用Microsatellite Toolkit、PopGene软件计算遗传相关指标,探讨广西树鼩群体的遗传多态性。结果发现,在10个微卫星基因座中,共检测到64个等位基因,平均等位基因数为6.4个,平均有效等位基因数为3.7892,平均观察杂合度和平均期望杂合度为0.6156和0.6963,平均多态信息含量为0.6367,其中7个是高度多态位点,3个属中度多态位点。此外,在10个微卫星基因座中,有5个位点偏离Hardy-Weinberg平衡(P<0.01)。结果表明广西树鼩遗传多态性较丰富,所选微卫星位点基本可用于评估广西树鼩群体遗传多样性。  相似文献

Polymorphisms for seven microsatellite loci in three red deer subspecies (9 populations) found in XinJiang were detected by polymerase chain reaction (PCR), 12% nondenaturation polyacrylamide gel electrophoresis and the Sanguinetti silver staining method. Numbers of alleles, average effective numbers of alleles (E) and the average rate of homozygosity, allelic frequencies of seven microsatellite loci, polymorphism information content (PIC), mean heterozygosity (H) and genetic distances among the populations were calculated for each population. Dendrograms were constructed based on genetic distances by the neighbor‐joining method (NJ), utilizing molecular evolutionary genetics analysis software PHYLIP (3.6). The phylogenetic tree was constructed based on allelic frequencies using maximum likelihood (ML); the bootstrap value was estimated by bootstrap test in the tree. Lastly, phylogenesis was analyzed. The results showed that four of the seven microsatellite loci were highly polymorphic, but BMS2508 and Celjp0023 showed no polymorphism and BM5004 was a neutral polymorphism. It is our conclusion that the four microsatellite loci are effective DNA markers for the analysis of genetic diversity and phylogenetic relationships among the three red deer subspecies. The mean PIC, H and E‐values across the microsatellite loci were 0.5393, 0.5736 and 2.64, which showed that these microsatellite loci are effective DNA markers for the genetic analysis of red deer. C.e. songaricus populations from Regiment 104, 151 and Hami are clustered together. C.e. yarkandensis populations from Regiment 35, Xaya and Alaer are clustered together. These two clusters also cluster together. Lastly, C.e. sibiricus populations from Burqin, Regiment 188 and the first two clusters were clustered together. The phylogenetic relationship among different red deer populations is consistent with the known origin, history of breeding and geographic distributions of populations.  相似文献

应用结构基因座和微卫星DNA两种遗传标记探讨长江三角洲白山羊群体遗传结构。结果表明,检测的9个结构基因座位上7个存在多态性,座位的基因平均杂合度、多态信息含量及有效等位基因数分别为0.1767、0.1457和1.2837;7个微卫星位点上共检测到110个等位基因,位点的基因平均杂合度、多态信息含量及有效等位基因数分别为0.8867、0.8774和11.2907。群体内的遗传变异程度相对较高,反映出丰富的遗传多样性,而且微卫星DNA标记揭示的遗传变异高于结构基因座。  相似文献

研究旨在分析金银滩藏羊的微卫星DNA多态性,以期了解该品种的遗传多样性,为该品种的保种选育和品质的进一步提高提供有益资料。利用8对微卫星引物,以金银滩藏羊为研究对象,通过计算基因频率、多态性信息含量、有效等位基因数和杂合度,评估其品种内的遗传变异。结果表明:在8个微卫星座位中,共检测到83个等位基因,每个座位平均10.38个等位基因;平均杂合度0.893;平均有效等位基因数9.06;平均多态性信息含量0.871。结果提示,金银滩藏羊群体存在丰富的遗传多样性,所选微卫星标记可用于绵羊遗传多样性评估。  相似文献

利用微卫星DNA多态性预测引进肉用绵羊品种杂种优势   总被引:4,自引:2,他引:2  
旨在利用微卫星DNA在不同绵羊群体中的多态性预测引进肉用绵羊品种与地方绵羊品种的杂种优势。利用23个微卫星基因座对4个国外引进肉羊品种及3个地方绵羊品种的群体多态信息含量、有效等位基因数、杂合度和遗传距离进行遗传检测,并测定引进肉羊与地方肉羊品种的杂交效果。结果,23个微卫星座位在7个绵羊群体中均呈现高度多态,共检测到348个等位基因,平均每个座位等位基因数为15个;多态信息含量在0.532 1~0.936 1,有效等位基因数在2.572 8~9.345 8,平均杂合度在0.549 2~0.936 0,品种平均杂合度在0.714 2~0.829 5,可以用于绵羊遗传多样性的评估。从不同品种看,无角陶赛特的遗传变异最大,而小尾寒羊的遗传变异相对较小;经遗传关系分析,在引进的4个肉羊品种中,与小尾寒羊、蒙古羊及滩羊杂交优势由大到小依次为白萨福克、波德代、无角陶赛特、特克赛尔,与实际杂种优势测定结果一致。利用微卫星DNA多态性预测引进肉用绵羊品种与小尾寒羊、蒙古羊及滩羊的杂种优势是可行的,这将对今后绵羊育种具有重要的应用价值和指导意义。  相似文献

利用30个微卫星DNA标记对3个江苏地方鸡品种进行遗传多样性分析,通过计算等位基因频率、多态信息含量、平均杂合度、有效等位基因数和遗传距离,评估各品种内遗传变异和各品种间遗传相关。结果表明:江苏的3个品种在30个微卫星上共发现196个等位基因,其中96个等位基因为三个品种共有,47个等位基因为各品种所特有;3个地方鸡品种的平均杂合度:狼山鸡为0.655,鹿苑鸡为0.681,溧阳鸡为0.687;狼山鸡与溧阳鸡的遗传距离较小,而两鸡与鹿苑鸡的遗传距离较远。  相似文献

利用选自家禽基因组的10个微卫星标记,对四川常羽乌骨鸡5个群体(四川山地乌骨鸡白羽系、黑羽系;黄忠山地乌骨鸡;黄忠山地乌骨鸡绿壳蛋系;草科乌骨鸡)的遗传多样性进行了检测,计算了各群体的群体杂合度、群体间遗传距离,并根据遗传距离进行了聚类分析。结果表明所选择的微卫星标记在各群体中表现出较高的多态性;四川5个乌骨鸡群体遗传多样性比较丰富,群体平均杂合度为0.5383到0.6659;各群体间的遗传距离有一定的差异;聚类分析将5个群体聚为三类。  相似文献

The native breeds, because of their natural selection against harsh environment and adaptation to regional conditions are important to resource-poor farmers and pastoralists. The molecular characterization of genetic variation is a fundamental step to manage and conserve indigenous breeds. The aim of this study was to determine the genetic variation of Makui sheep. Totally, 100 sheep (60 ewes, 10 ram and 30 lambs) were used in this study. Polymerase Chain Reaction (PCR) was performed using microsatellite primers BM1329, OarAE101, OarFCB11, OarFCB128, OarFCB129, OarFCB20, OarFCB304, OarFCB5, OarHH35 and OarHH55 in a standard 25 μl reaction. All microsatellite loci were amplified and produced minimum 2 and maximum 14 alleles ranging from 90 to 185 bp in size. The mean number of alleles for each locus was 6.8. Loci OarFCB128 and OarAE101 produced the highest (8.5288) and the lowest (1.0304) effective number of alleles, respectively. The mean expected heterozygosity for all loci was 0.6893 (range 0.0295–0.8837). The highest (2.12) and the lowest (0.07) Shanon Index was observed in OarFCB11 and OarAE101, respectively. Only two (OarFCB5 and OarAE101) out of ten loci were in Hardy-Weinberg equilibrium. The present study was able to demonstrate a reasonable genetic variation and polymorphism across all microsatellite loci studied in the Makui sheep.  相似文献

应用微卫星标记对江苏省无锡市祖代鸡场有限公司太湖鸡的保种群体进行了遗传变异的分析,计算了群体的杂和度(H)和多态信息含量(PIC)。研究结果表明:30个微卫星标记在太湖鸡保种群体的杂和度为0.504~0.858之间,平均杂和度为0.688,群体的遗传多样性丰富;其中24个微卫星标记在太湖鸡中多态信息含量为高度多态,可作为有效遗传标记用于太湖鸡保种群的遗传分析,而且各座位杂和度的高低与PIC值的大小体现了较高的一致性。本研究结果为太湖鸡开发利用研究提供了基础,也为太湖鸡的合理保护提供了科学根据。  相似文献

以4个中国地方鸡品种遗传多样性的微卫星分析数据为例,分析样本量和性比对微卫星分析中群体遗传多样性指标的影响。结果表明:当样本量超过20后,期望杂合度值趋于稳定,样本量与期望杂合度无显著相关,而与平均等位基因数呈正相关,在微卫星分析中性比对群体遗传多样性指标不表现出显著影响;微卫星位点多态性的高低直接影响到检测所需的样本量,在使用平均等位基因数分析群体遗传多样性时,应该充分考虑样本量对检测结果的影响;期望杂合度受样本量变动的影响较小,可作为度量群体遗传多样性的一个最适参数,而且样本量以20~25较为适宜。  相似文献

This study was the first to analyze the polymorphic characteristics of a wide range of genetic markers in three Lithuanian horse breeds. Genetic variation at seven red blood cell alloantigen (blood group) loci, 10 biochemical genetic loci and 16 microsatellites was examinated in 31 Zemaitukai, 30 large-type Zemaitukai and 24 Lithuanian Heavy Draught horse and compared to that of other domestic horse breeds. Genetic variability of the Zemaitukai breeds based upon blood groups and biochemical loci was higher than average for domestic horse breeds. For microsatellite loci, the variability of the large-type Zemaitukai and the Heavy Draught breeds also was high compared to the mean variation for domestic breeds; however, for the Zemaitukai breed, variation levels were very near the average for horses.  相似文献

选用10对微卫星引物分析了黄羽肉鸡A、B、C3个品系的遗传结构和差异,并对微卫星标记所计算的个体间遗传距离与C×B、C×A杂交后代的初生至8周龄体重的杂种优势和杂种优势率进行了相关分析。结果表明,10个微卫星标记中有9个表现出多态性,多态信息含量(PIC)和群体杂合度(Heterozigosity)分别为0·4400和0·6811,各品系、各微卫星基因座上都表现出一定差异。利用9个多态微卫星标记计算得出的亲本个体间的遗传距离与后代杂种优势率间多数没有显著的相关(P>0·05),仅C×B组合中亲本个体间平均遗传距离与第8周龄体重杂种优势和杂种优势率间相关显著(P<0·05)。  相似文献

研究对甘肃高山细毛羊优质毛品系的15个微卫星位点进行了遗传多样性检测.计算了各位点的等位基因频率(P)、杂合度(H)、多态信息含量(PIC)和有效等位基因数(Ne).结果表明:15个微卫星位点中有1个未检测到多态,其余14个均表现出高度多态性.多态性标记在该群体中的平均等位基因数为10个,平均多态信息含量PIC=0.83,平均杂舍度H=0.85,平均有效等位基因数Ne=7.1.说明甘肃高山细毛羊优质毛品系的遗传多样性丰富,遗传变异程度较高,基因一致度较差,变异性较大,具有较大的选择潜力.这14个位点可以作为有效的遗传标记,用于甘肃高山细毛羊优质毛品系遗传多样性分析及其与各生产性状的相关性研究.  相似文献

利用微卫星标记分析了乌骨大骨鸡的遗传多样性和遗传结构,筛选了鸡基因组7条染色体上的7个微卫星标记位点,随机选取24羽乌骨大骨鸡个体,进行多态性检测,共检测到23个等位基因,每个座位等位基因数目从2个到5个不等,平均等位基因数为3.3个。该群体平均多态信息含量和平均杂合度分别为0.660 3和0.717 6。结果表明乌骨大骨鸡属多态性较丰富的群体。  相似文献

为了解麦洼牦牛现有群体的遗传多样性及种群遗传分化特征,试验通过生物信息学方法从已公布的牦牛基因组中筛选到9 466个三碱基重复微卫星位点,以191个麦洼牦牛基因组DNA样品为模板对各个位点进行PCR扩增和聚丙烯酰胺凝胶电泳基因分型。经过哈迪-温伯格平衡检验,在15个三碱基微卫星位点中,有5个位点偏离哈迪-温伯格平衡(P<0.05)。15个微卫星位点均为高度多态性(PIC>0.5),群体平均观测杂合度(Ho)、平均期望杂合度(He)和平均多态信息含量(PIC)分别达到0.5041、0.8152和0.7831。进一步通过Structure 3.0软件分析发现,麦洼牦牛群体结构单一,无分化特征,表明麦洼牦牛品种虽然具有较高的遗传多态性但是无分化迹象。  相似文献
