Solanum okadae Hawkes et Hjert is a wild diploid potato species endemic to Argentina and Bolivia, of potential breeding value. However, no genetic studies have been carried out with this species and its crossability relations with the common tetraploid potato, S. tuberosum L. ssp. tuberosum, are unknown. Furthermore, accessions from both countries differed in their morphological phenotypes. To ascertain the feasibility of incorporating this wild germplasm into cultivated potato, 2n pollen screening was carried out in 10 accessions and families derived from crosses between accessions; also, pollen-pistil compatibility relations were studied in reciprocal interspecific crosses. Plants of four of the 10 accessions produced 2n pollen (0.1 to 5.0%) and 4n pollen (0.0 to 3.0%). Parallel and tripolar spindles at Anaphase II were the cytological mechanisms involved in 2n pollen formation; lack of chromosome migration in both meiotic divisions originated the 4n pollen. Both full compatibility and incompatibility at various sites along the pistil were observed in the S. tuberosum × S. okadae combinations; most reciprocal combinations were incompatible. Compatible genotypes produced 2n pollen. However, only a few seeds were obtained and chromosome counts could not be carried out in the hybrid progeny because seedlings died at an early stage. The pollen-pistil barriers are incomplete and can be circumvented by the appropiate choice of parents. The identification of the post-zygotic barriers will be the focus of further studies.  相似文献

Wild potato germplasm represents a unique, diverse and accessible resource for disease and pest resistance, along with useful agronomic traits that may be introgressed into the cultivated potato (Solanum tuberosum L.). Hybridization of diploid wild Solanum species with haploids (2×) of cultivated potato (4×) is generally an effective technique for introducing genetic diversity and desirable traits into potato. However, in this study, hybridization barriers were found in crosses between S. tuberosum haploids and the wild species S. raphanifolium. Male sterility, likely due to nuclear-cytoplasmic interactions, was observed in some haploid tbr?×?S. raphanifolium hybrids. In addition, pollen-pistil incompatibilities were observed in backcross, F2, and reciprocal cross hybridization attempts that failed to produce seed. More crosses were successful when F1 clones were crossed as females to wild clones than to cultivated clones. When crosses were made in the other direction, with F1 hybrids used as male parents, seeds were almost never produced.  相似文献

For breeding programs of the tetraploid potato (Solanum tuberosum), both wild and cultivated diploid relatives are valuable sources of genetic diversity. While both types of germplasm are used in breeding programs, there are several advantages to using wild relatives. Diploid relatives are typically crossed with haploids (2n = 2x = 24) from tetraploid S. tuberosum to improve daylength adaptation. Most haploids are male sterile, so they are typically used as female parents. Cultivated diploids, such as members of the Phureja Group, produce male sterile hybrids when crossed as females to haploids; wild relatives, such as S. tarijense, often produce male fertile hybrids. Tuber yield following crosses of haploids to cultivated or wild relatives is often high. However, cultivated relatives generally produce hybrids with a high set of small tubers; hybrids from wild relatives are variable, but many are similar to cultivars in tuber size and set. While tubers of hybrids from cultivated relatives are typically rough, with deep eyes and raised internodes, those from wild relatives are often smooth. Tuber dormancy in hybrids with cultivated relatives is generally short, while that in hybrids with wild species is longer, allowing for storage over winter. Finally, resistance to several major diseases and stresses has been found in wild species and their hybrids with S. tuberosum haploids. The desirable traits in hybrids are transmitted to tetraploids via unilateral sexual polyploidization (4x × 2x or 2x × 4x crosses in which the diploid parent produces 2n gametes). Wild Solanum species are recommended for use in potato breeding programs as sources of genetic diversity that can be adapted easily following hybridization with S. tuberosum haploids.  相似文献

Patterns of variation for the solanidine-based glycoalkaloids have been determined for tuber material of genebank accessions of landraces of the cultivated potato and three closely-related wild species. Total levels were low in the cultivated taxa investigated,S. phureja, S. stenotomum, and the tetraploidS. tuberosum in its two formsS. tuberosum ssp.tuberosum andS. tuberosum ssp.andigena. The only solanidine-based glycolkaloids found in the tubers investigated in this study were-solanine and-chaconine, with one additional related compound, dehydrocommersonine, found in tubers of an accession ofS. canasense. The ratio of chaconine : solanine differed markedly from a mean of 0.98 inS. phureja to 2.28 in ChileanS. tuberosum ssp.tuberosum. The chaconine : solanine ratio was relatively constant within these two taxa, reflecting their relatively narrow genetic bases, but was much more variable within the taxa regarded as ancestral,S. stenotomum andS. tuberosum ssp.andigena. The wild speciesS. sparsipilum has been implicated in the origin of the tetraploid potato,S. tuberosum, but there was no support for this from the patterns of variation in chaconine : solanine ratio. The cline of chaconine : solanine ratios in cultivated potatoes along the spine of S. America does not appear to relate to the influence of local wild species through introgression, but may instead reflect differing selective pressures acting in different parts of the native range of the cultivated potato.  相似文献

Summary An extensive crossing program including 10 wild taxa and 23 accessions representing 6 major cultivated forms and landraces of theBrassica oleracea group was carried out. Data for crossability, germination, viability, fertility inF 1 andF 2 were studied as well as the meiotic chromosomal pairing inF 1 hybrids in some combinations. The fertility of hybrids between all cultivated forms and wild populations ofB. oleracea was high and it was concluded that all, including the Chinese endemicalboglabra form, belong to the same biological species. Among the other species,B. macrocarpa, B. montana, andB. rupestris had the lowest andB. cretica the highest fertility values in crosses with cultivated forms. The crossing data do not give any further information as to the genetic differentiation ofB. oleracea during the course of domestication. Introgression between wild and cultivated forms has probably occurred frequently. All members of theB. oleracea cytodeme belong to the primary and secondary gene pools of the 18 chromosomic crops and are as such of great importance for breeding purposes.  相似文献

Abstract

An Agrobacterium tumefaciens -mediated transformation procedure was developed for the non tuber-bearing, diploid wild potato species Solanum brevidens, and the tetraploid S. tuberosum cv. Pito. Cointegrative transformation vectors pGV2260 and pGV3850, and binary vector pGUS-INT were employed in transformation of S. brevidens and Pito, respectively. Leaf and stem explants of S. brevidens and microtuber discs of Pito were precultured 24 h, and cocultivated 48 h on solidified callus induction medium with a 24-h liquid culture of A. tumefaciens C58C1 diluted 1:10 with liquid MS medium. Explants were rinsed with cefotaxime solution (500 mg/1) to remove Agrobacterium, grown without the selective agent kanamycin on solified callus induction media for 14 days, and then exposed to kanamycin selection for the first seven days on callus induction medium and subsequently on shoot regeneration medium. Shoots regenerated faster from stem explants than from leaf explants. Up to 49% and 57% of the transformed leaf explants of S. brevidens and microtuber discs of Pito, respectively, produced transformed shoots.  相似文献

Seed protein profiles by SDS-PAGE were studied in 72 accessions ofSolanum melongena L. and its related taxa in subgenusLeptostemonum (Dunal) Bitter. Comparisons based onJaccard's similarity and UPGMA clustering revealed interrelationships broadly inconformity with the conventional taxonomic treatments. Thus,S. viarum Dunal (sectionAcanthophora Dunal), S.torvum Swartz (section Torva Nees) andS. sisymbriifolium Lam. (sectionCryptocarpum Dunal) were located on separate branches ofthe dendrogram while all taxa of section Melongena Dunal,except S. macrocarpon Dunal, wereclustered closer together. S.melongena, incanum and insanum,all members of the eggplant complex, were revealed to bear very high similaritywith each other. Cultivated Solanum tuberosum L. subsp. andigena is well known as a rich source of valuable traits for potato breeding, especially for resistance to diseases and pests. The golden potato cyst nematode, Globodera rostochiensis Woll., is considered to be one of todays most serious hindrances to potato production in Europe and North America. Thus, the breeding of new cultivars that have resistance to PCN is of great importance. The USPG (USA) and VIR (Russian) potato genebanks, as well as others, maintain many samples of primitive cultivated and wild potato species originating from Latin America. Many of these samples are assumed to be genetically duplicated because the material in both genebanks came from the same original source. A joint investigation of new genotypes of subsp. andigena forms resistant to potato cyst nematode (PCN) was carried out on samples of subsp. andigena at VIR with reputed duplicate samples at USPG. After careful screening, 14 samples which possessed resistance to PCN were identified. A high level of this resistance was transmitted to sexual progeny at a high frequency for all of the selections. Eleven of the accessions found to be resistant have reputed duplicates in USPG that were not previously known to be resistant. Thus, this research not only broadens the choice of parents available for resistance breeding, but identifies model materials for future research to test the parity of PCN resistance among reputed duplicate samples in the two genebanks.  相似文献

Amplified Fragment Length Polymorphisms (AFLPs) were employed within the taxonomically difficult Solanum nigrum L. complex in order to characterize the genetic diversity present in a collection of the Gatersleben Genebank, to classify taxonomically unknown material, and to correlate the clustering of the examined accessions with their geographic origin. The results from AFLP analyses using two primer combinations on 44 entries from five species led to the detection of four major clusters, simultaneously uncovering significant differences in the levels of genetic diversity within or between species. S. americanumexhibited the highest infraspecific variation despite close geographic origins, simultaneously being placed in a clearly separated cluster in comparison to the other examined species of the complex. In addition, these other species showed even less interspecific variation than was found at the infraspecific level in S. americanum. In terms of taxonomy, the application of AFLPs helped in the classification of 13 black nightshade accessions formerly only listed as Solanum sp. This also was confirmed by morphological determination. Furthermore, one accession formerly classified as S. physalifolium var. nitidibaccatum i) clustered with AFLPs and ii) was identified morphologically as S. villosum. Contradictory classifications remain for two further entries from the same species, found within the S. nigrum cluster after AFLP analyses, while belonging to S. physalifolium var. nitidibaccatum according to herbarium specimen. Finally, as indicated by the information on provenance in geographically separated subclusters in S. americanum and partially in S. villosum, clues on the currently unknown origin of accessions from the genebank seem feasible by AFLP data.  相似文献

Solanum stenotomum Juz. et Buk. (2n = 2x = 24) is considered to be the most primitive diploid cultivated species from which all the other Andean cultivated potatoes were originated (Hawkes 1990). To disclose chloroplast DNA (ctDNA) variability and the maternal origin of S. stenotomum, 36 accessions of S. stenotomum and 86 accessions of putative wild ancestral species were determined for ctDNA types and analyzed by high-resolution markers (seven ctDNA microsatellites and an H3 marker). High-resolution markers discriminated 57 different ctDNAs (haplotypes), which were classified into the W-type ctDNA group and C-, S- and A-type ctDNA group, and within the latter group S- and A-type ctDNAs were distinct from each other among many different haplotypes mostly having C-type ctDNA. This ctDNA relationship supported our previous findings obtained for mostly Andean cultivated species (Sukhotu et al. 2004). Compared with other putative ancestral wild species, S. stenotomum showed somewhat limited ctDNA diversity, having two major haplotypes 1 and 2 also found in different wild species in different places. Therefore, the ctDNA in S. stenotomum was of at least dual origins either by successive domestication from different species or else by introgression after initial S. stenotomum arose.  相似文献

In vivo interspecific pollinations were performed and immature seed development investigated by histological methods in order to study crossability barrier(s) in Cicer L. species wide hybridization. Seven of the eight wild annual Cicer species, belonging to the secondary and tertiary gene pools, were used in reciprocal crosses with the cultivated chickpea. It was confirmed that the zygote was formed in all interspecific crosses. The embryos showed continued and retarded growth at different rate in various crosses, but eventually aborted at an early pro-embryo stage in all crosses except C. arietinum L. ×C. echinospermum Dav. Reciprocal cross differences were observed in early embryo growth rate and could have implications in obtaining hybrids. This study further emphasizes the necessity for developing appropriate and efficient in vitro procedures for rescuing immature globular hybrid pro-embryos, which will make the wild Cicer gene resources amenable to chickpea improvement.  相似文献

A quantitative trait locus analysis was carried out to unravel the genetic basis of dormancy in wild barley (Hordeum spontaneum) from Israel. Two accessions, Ashkelon and Mehola, from divergent environments were crossed to produce a mapping population. A linkage map was produced from the F2 population, and F4 seeds were used for germination experiments. Five quantitative trait loci (QTL) were detected for dormancy across the different germination experiments. These QTL were found on chromosomes 1, 2, 5, 6 and 7. The variation explained by each QTL varied between 8 and 25%. Ashkelon alleles increased the germination except for the QTL on chromosome 5. Three out of these five QTL co-locate with QTL found earlier in cultivated barley crosses, although this does not necessarily imply that they would be the same loci. The level of dormancy is much higher in wild barley than in cultivated barley and wild barley may have alleles that have not yet been utilised in breeding for optimally dormant barley.  相似文献

Brassica juncea (2n = 36, AABB) is an amphidiploid derived from its diploid progenitor species B. rapa (2n = 20, AA) and B. nigra (2n = 16, BB). Resynthesis of B. juncea by exploiting the wider gene pool of the present day diploids offers a powerful tool for obtaining novel genetic variation. Hybridization between the two species is difficult because of the presence of crossability barriers. Barriers to hybridization were identified using Aniline Blue Fluorescence (ABF) method. Crosses were classified as having pre- or post-fertilization barriers based on whether less than or more than one per cent ovules showed pollen tube entry. Of the 23 crosses studied, seven crosses all with B. rapa as the female parent showed pre-fertilization barriers. Only 0 to 0.3 per cent ovules were fertilized in these crosses. The remaining 16 crosses, four with B. rapa as the female parent and 12 with B. nigra as the female parent, showed post-fertilization barriers. In crosses with pre-fertilization barriers use of bud pollination and stump pollination ensured or increased fertilization. These two simple techniques were used in combination with ovary-ovule culture to ensure recovery of hybrids. In crosses with post-fertilization barriers, ovary-ovule culture alone helped to obtain hybrids. Hybrids were obtained at higher frequency with the use of direct ovule culture compared to ovary-ovule culture. The F1 plants obtained in vitro were multiplied on MS medium + 2 mg/l Kn + 0.2 mg/l IAA. These F1 plants were confirmed as true hybrids through morphology, leaf isozymes and cytology. In all 17 hybrids were obtained of which three were amphidiploids.  相似文献

A fertile amphidiploid × Brassicoraphanus (RRCC, 2n = 36) between Raphanus sativus cv. HQ-04 (2n = 18, RR) and Brassica alboglabra Bailey (2n = 18, CC) was synthesized and successive selections for seed fertility were made from F4 to F10. F10 plants exhibited good fertility with 14.9 seeds per siliqua and 32.3 g seeds per plant. Cytological observation revealed that frequent secondary pairing occurred among 3 chromosome pairs in pollen mother cells of plants (F4) with lower fertility, but not of plants with high fertility (F10). GISH analysis indicated that these F10 plants included the expected 18 chromosomes from R. sativus and B. alboglabra, respectively, but they lost approximately 27.6% R. sativus and 35.6% B. alboglabra AFLP (amplified fragment length polymorphism) bands. The crossability of the Raphanobrassica with R. sativus and 5 Brassica species (13 cultivars) were investigated. Seeds or F1 seedlings were easy to be produced from crosses × Brassicoraphanus × R. sativus, and B. napus, B. juncea and B. carinata × Brassicoraphanus. Fewer seeds or seedlings were obtained from crosses × Brassicoraphanus × B. napus, B. juncea and B. carinata. However, few seeds were harvested in the reciprocals of × Brassicoraphanus with B. rapa and B. oleracea. The possible cause of fertility improvements and the potential of the present × Brassicoraphanus for breeding were discussed.  相似文献

Erianthus rockii, a wild relative of sugarcane, is drought and cold tolerant, and both are potentially important agronomic traits to the sugarcane industry worldwide. As such it is of interest as a source of parental germplasm to sugarcane breeders and is currently being used in sugarcane introgression programs in both China and Australia. To date morphological characters have been used to verify the putative hybrids produced. Two crosses were generated between two different Saccharum species and two E. rockii accessions. Over 400 AFLP markers were used to identify the intergeneric hybrid progeny as well as determine hybrid diversity. Both crosses generated hybrids but efficiency levels were very different and are probably related to the different Saccharum parent used in each cross. Cross 1 was between a Saccharum officinarum and E. rockii and generated 100% hybrid progeny. Cross 2, however, was between a sugarcane hybrid (S. officinarum × Saccharum spontaneum) and E. rockii and only 10% of the progeny were intergeneric hybrids. Inheritance of markers in the progeny was analysed and for both crosses there were equal numbers of markers from both parents indicating n + n transmission of chromosomes. This is the first verification of E. rockii hybrids with molecular markers. It may now be possible to exploit genes of value from E. rockii in sugarcane breeding programs.  相似文献

Genetic characterization of germplasm is important for setting objective guidelines for conservation. One common problem found in genebanks is determining the value of populations with insufficient or unreliable data regarding their geographic origin. In this study, a genetic analysis based on RAPD markers was conducted to characterize a `mystery' population of Solanum sucrense, a polysomic tetraploid potato (2n=4x=48), for which adequate documentation was lacking. The comparative analysis of genetic similarities between this mystery population and each one of 30 other S. sucrense populations in the genebank revealed that all populations within this species, including the mystery population, are significantly different from being duplicates, and are therefore worthy of separate conservation. RAPD markers also distinguished the mystery population from closely related tetraploid species S. oplocense, S. gourlayi and S. tuberosum ssp. andigena, suggesting that it is also not a duplicate of a population of these species. If RAPDs can clearly differentiate populations within highly heterogeneous tetraploids like S. sucrense, they should be generally useful for determining germplasm organization within potato species.  相似文献

Summary A collection of 400 Ae. tauschii (syn. Ae. squarrosa) Coss. accessions were screened for powdery mildew resistance based on the response patterns of 13 wheat cultivars/lines possessing major resistance genes to nine differential mildew isolates. 106 accessions showed complete resistance to all isolates, and 174 accessions revealed isolate-specific resistance, among which were 40 accessions exhibiting an identical response pattern as wheat cultivar Ulka/*8Cc which is known to possess resistance gene Pm2. Expression of both complete and isolate-specific resistance from Ae. tauschii was observed in some synthetic hexaploid wheats derived from four mildew susceptible T. durum Desf. parents, each crossed with five to 38 resistant diploid Ae. tauschii accessions. Synthetic amphiploids involving different combinations of T. durum and Ae. tauschii generally showed a decrease in resistance compared with that expressed by the Ae. tauschii parental lines.  相似文献

In order to study relationships within and among species of the highly complex polyploid genus Saccharum, 30 clones from 5 species (S. barberi, S. officinarum, S. robustum, S. sinense, and S. spontaneum) and 6 cultivated clones were analyzed using 15 sugarcane simple sequence repeat (SSR) markers and two multivariate statistical methods. The origins of two species, S. barberi and S. sinense, are unclear, and it has been hypothesized that they arose from crosses of S. officinarum × S. spontaneum. S. robustum is suggested to be ancestral to S. officinarum, which, when crossed with S. spontaneum, principally comprise the genomes of commercial cultivars. Although our analyses supported S. robustum as being an ancestor of S. officinarum, and show the composition of commercial clones as expected, our results concerning the species S. barberi and S. sinense indicated them to be much less related to the main Saccharum germplasm pool than previous molecular marker investigations, suggesting that introgression from other genera may exist in their ancestry. The differing results of our analysis of S. barberi and S. sinense are likely attributable to more equal and larger numbers of clones studied in each species. Given the extremely high ploidy, the use of only 15 SSR markers enabled clear distinction of Saccharum species and clones, and also identified likely mislabeled accessions. In addition, the replication of DNA extraction, polymerase chain reactions, and fragment electrophoresis increased the robustness of our analysis.  相似文献

Genetic variability of wild populations, closely related to domesticated species, constitute important genetic resources for plant breeding programs. In this paper, we analysed the variation of eight wild populations of Solanum lycopersicum var. cerasiforme in a common garden experiment for levels of plant infestation by whitefly, leaf trichome density as a defensive character preventing infestation by whitefly, and the effect of whitefly incidence into vegetative and reproductive plant characters. Number of adults of whitefly was recorded in the eight wild populations of S. lycopersicum var. cerasiforme, one population of the wild species S. habrochaites (C-360), and one of a cultivated variety of S. lycopersicum (Rio Grande). There were significant differences among the wild populations of S. lycopersicum var. cerasiforme in the average level of whitefly incidence and trichome density. Cultivated tomatoes had the higher incidence of whiteflies ( = 7.50 ± 0.14) followed by plants of S. lycopersicum var. cerasiforme ( = 2.02 ± 0.92) and plants of S. habrochaites with the lowest incidence ( = 0.36 ± 0.35). Whitefly incidence was negatively correlated with trichome density (r = − 0.38, p < 0.0001), suggesting that trichomes deter or limit the establishment of whiteflies. Additionally, a significant negative effect of whitefly incidence along the growing season upon plant growth rate (number of branches and height) and fruit production was detected.  相似文献
