Effects of administering killed Salmonella enterica serovar enteritidis (SE) vaccines to laying hens prior to induced molting on egg production and on shedding of SE were investigated. Forty hens were vaccinated with one of two SE vaccines available commercially in the United States and Japan. Twenty-five days after vaccination, feed was withdrawn for 2 wk from 20 vaccinated plus 10 unvaccinated hens to induce molt. Four days after molt induction, all hens were challenged with a dose of 2.4 X 10(9) of SE. For the 25 days following administration of the SE bacterins, egg production in vaccinated hens showed approximately a 15% decrease. After molt induction, egg production in molted hens ceased and then returned to normal levels 8 or 9 wk postvaccination. Through the 3-mo experimental period, the decreases in numbers of eggs laid in the unvaccinated/molted group and two vaccinated/molted groups were 225 (26.2%), 245 (28.4%), and 274 (31.9%), respectively, compared with 860 in the unvaccinated/unmolted group. There was no significant difference in egg lay at the P < 0.05 level among the former three groups. Hens in the vaccinated/molted groups shed about two logs less SE than hens in the unvaccinated/molted group 3 14 days postchallenge (P < 0.05 or 0.01). These results indicate that vaccination prior to induced molting might be effective in preventing the exacerbation of SE problems within flocks in which the potential for SE contamination may exist.  相似文献

Molting is used to increase productivity in the second laying cycle of commercial laying flocks. We report here the applicability of whole grain of bitter vetch as an alternative method for molt induction in laying hens. A total of 120 laying hens were molted by feed withdrawal (FW), a high-Zn diet (HZn), a moderate-Zn and Ca-P-deficient diet (MZn-CaP), or whole grain of bitter vetch (BV). Egg production ceased first in FW-treated hens and last in MZn-CaP-treated hens (P < 0.05). The rest period was longer (P < 0.05) in hens exposed to the BV diet, which proved to be comparable with the FW treatment for postmolt egg production. Egg mass for 4 to 16 wk after the molting was greater (P < 0.05) for hens molted by BV than hens molted by the MZn-CaP diet. Egg weight and yolk color were not (P > 0.05) significantly different among various molting induction methods. Albumen height and Haugh unit of eggs from hens molted by BV, for the most part of the experimental period, were significantly (P < 0.05) greater than for birds on the MZn-CaP diet and unmolted hens. In conclusion, use of BV grain as a single dietary ingredient proved to be effective in molt induction and improving postmolt egg production and internal egg quality when compared with unmolted hens.  相似文献

A study was undertaken to determine if a 2-week feed-removal protocol, as is used by industry to induce a molt in aging hens, would affect the course of a Salmonella enteritidis infection. White leghorn hens aged 69-84 weeks were deprived of feed to induce a molt, and on day 4 of the fast, the birds were orally infected with 5 x 10(6) S. enteritidis. S. enteritidis organisms were enumerated in the spleen on day 6 and from the alimentary tract on days 7, 14, 21, 28, and 35. Little difference was detected in numbers of S. enteritidis from spleens of molted and unmolted hens. Significantly more molted hens shed detectable intestinal S. enteritidis than unmolted hens on day 14 (one of two trials) and day 21 (one of two trials). Intestinal levels of S. enteritidis were increased 100- to 1000-fold in the molted birds on day 7 (one of two trials) and day 14 (two of two trials), and many of the hens exhibited bloody alimentary secretions. Histological examination of the intestinal tract of S. enteritidis-infected molted hens showed increased inflammation in the epithelium and lamina propria of colons and ceca, compared with unmolted infected hens.  相似文献

Increased resistance to Salmonella enteritidis (SE) organ infectivity in chickens can be conferred by the prophylactic administration of SE-immune lymphokines (ILK). Resistance is associated with an enhanced heterophilic accumulation within 4 h of ILK injection. In these studies, the role of IL-8 in ILK-mediated heterophil recruitment during SE infections in young chickens was investigated. Heterophil accumulation was enhanced 2-4 h after the i.p. injection of both ILK and SE (ILK/SE) when compared to the control chicks. An i.p. injection of a rabbit polyclonal anti-human IL-8 antibody significantly (P < 0.01) reduced the accumulation of heterophils in the peritoneum after the injection of ILK/SE. Injections of preimmune rabbit IgG had no effect on peritoneal heterophil numbers. Within 2 h of injection of ILK/SE, a ten-fold increase in heterophil chemotactic activity was found in the peritoneal lavage fluid from these chicks compared to the saline control chicks. Pretreatment, with the anti-IL-8 antibody, of the peritoneal lavage fluids collected from the ILK/SE-treated chicks dramatically reduced this heterophil chemotactic activity. Treatment of the lavage fluids from all groups with preimmune IgG had no effect on heterophil chemotaxis. Additionally, pretreatment of ILK with the anti-human IL-8 antibody had no effect on heterophil chemotaxis. The results from these experiments suggest that IL-8 is produced locally by the host in response to both the SE infection and the ILK. With these studies, it was established that IL-8 is a major chemotactic factor produced by the host, which aids in mediating the ILK/SE-induced recruitment of heterophils to the site of SE invasion.  相似文献

During the first week post-hatch, chickens demonstrate an increased susceptibility to infection by bacteria such as Salmonella. The purpose of the present study was to evaluate the effects of immune lymphokines on phagocytosis and killing activities of heterophils in chicks during the first 1-7 days of life. Lymphokines isolated from chicken splenic T-cells harvested from Salmonella enteriditis (SE)-hyperimmunized hens (SE-ILK), have in past experiments, demonstrated augmentation of heterophil activity in day-of-hatch chicks resulting in protection from SE organ invasion. The present experiments reveal significant increases (p<0.05) in heterophil phagocytosis and killing when comparing chicks treated with SE-ILK to control groups in vitro. In SE-ILK-treated groups, a two-fold or greater increase is noted in heterophil phagocytosis within I h of incubation as compared to controls. Heterophils isolated from 1-day-old and 4-day-old chicks treated with SE-ILK killed significantly greater numbers (p<0.05) of SE than heterophils isolated from control groups. By Day 7 post-hatch, significance is not noted in the killing activity of heterophils from treated groups when compared to control groups. However, heterophils from SE-ILK groups continue to kill greater numbers of SE than control groups. These data support SE-ILK augmentation results in an enhanced heterophil function in chicks during the greatest period of susceptibility to Salmonella invasion.  相似文献

Long-term feed withdrawal has been shown to increase ileocecal intestinal colonization and fecal shedding of Salmonella enterica serovar Enteritidis in challenged hens. Less information is available regarding effects of fasting on crop colonization. Two trials were conducted to compare effects of 14-day feed withdrawal vs. full feed on crop colonization in hens challenged with Salmonella Enteritidis. The levels of Salmonella Enteritidis in the crops of fasted hens were significantly higher than in nonfasted hens on days 3 and 10 and days 3, 9, and 16 postinfection (PI) in trials 1 and 2, respectively. Fecal shedding of Salmonella Enteritidis was significantly increased in the fasted hens on day 10 PI in trial 1. Analysis of crop IgA anti-Salmonella Enteritidis lipopolysaccharide levels in crop lavage samples of hens in trial 1 revealed a humoral response PI in both treatment groups with no significant differences, although peak response for fasted hens occurred 1 wk later. Histologic evaluation of hematoxylin and eosin-stained crop sections from trial 1 birds revealed mild to moderate heterophilic infiltration within the crop lamina propria (LP) or LP and epithelium of nonfasted infected hens at 24 and 96 hr PI. In comparison, heterophils in crops of fasted hens infected at this time point were sparse, indicating a possible diminished heterophil response in the fasted birds. Multifocal areas of tissue inflammation, as indicated by marked heterophil infiltration, with necrosis and sloughing of epithelium, were observed in crops from fasted hens at day 11 PI (14th day of feed withdrawal) but not in the fed groups. This severe heterophilic inflammation was observed in both challenged and nonchallenged fasted hens, suggesting that some factor other than Salmonella Enteritidis was responsible. These results indicate that feed withdrawal can have a dramatic effect on the integrity of the crop and its ultimate response to infection.  相似文献

As laying hens age, egg production and quality decreases. Egg producers can impose an induced molt on older hens that results in increased egg productivity and decreased hen mortality compared with non-molted hens of the same age. This review discusses the effect of induced molting by feed removal on immune parameters, Salmonella enterica serovar Enteritidis (SE) invasion and subsequent production of SE-contaminated eggs. Experimental oral infections with SE show molted hens are more susceptible to SE infection and produce more SE-contaminated eggs in the first few weeks post-molt compared with pre-molt egg production. In addition, it appears that molted hens are more likely to disseminate SE into their environment. Molted hens are more susceptible to SE infection by contact exposure to experimentally infected hens; thus, transmission of SE among molted hens could be more rapid than non-molted birds. Histological examination of the gastrointestinal tracts of molted SE-infected hens revealed more frequent and severe intestinal mucosal lesions compared with non-molted SE-infected hens. These data suggest that induced molting by feed deprivation alters the normal asymptomatic host-pathogen relationship. Published data suggest the highest proportion of SE-positive eggs is produced within 1-5 weeks post-molt and decreases sharply by 6-10 weeks and dissipates to the background level for non-molted hens by 11-20 weeks. Appropriate treatment measures of eggs produced in the fist 5 weeks post-molting may decrease the risk of foodborne infections to humans.  相似文献

Oligodeoxynucleotides (ODN) containing CpG dinucleotides (CpG-ODN) mimic bacterial DNA and stimulate the innate immune system of vertebrates. Here, we investigated the effects of intraperitoneal (ip) administered CpG-ODN on the innate immune functions of chicken heterophils. Our results demonstrated CpG-ODN-dependent priming of chicken heterophil degranulation and oxidative burst. Heterophils from chickens treated with CpG-ODN exhibited significantly higher (p<0.05) degranulation activity compared to PBS and control ODN (ODN containing no CpG motif) treated groups when stimulated with opsonized Salmonella enterica serovar enteritidis. Similarly, oxidative burst activity, which generates bactericidal reactive oxygen species, was significantly higher (p<0.05) in heterophils from the CpG-ODN treated group than from PBS and control ODN groups when stimulated with formalin-killed S. enteritidis. The priming effects of CpG-ODN on heterophil immune functions continued at least 4 days post-treatment. In the infection study, newly hatched chickens were treated with CpG-ODN, control ODN or PBS for 24h then challenged with oral inoculation of S. enteritidis. A significant reduction (p<0.05) in colonization by S. enteritidis was observed in chickens treated with CpG-ODN. Our study provides evidence that immunostimulatory CpG-ODN potentiates the innate immune responses of heterophils and enhances resistance to infectious pathogens in neonatal chickens.  相似文献

Four trials were conducted to evaluate whether prior infection with Salmonella enterica serovar typhimurium (S. typhimurium) or Salmonella enterica serovar muenchen (S. muenchen) would modify the severity or the transmission of Salmonella enterica serovar enteritidis (S. enteritidis) challenge in hens undergoing molt via feed withdrawal. Hens were separated into two groups where one group received a prior S. typhimurium or S. muenchen infection, whereas the other group remained untreated until S. enteritidis challenge. In trials 1 and 2, one group of hens was infected with S. typhimurium 5 days prior to feed withdrawal. Both groups of hens were then challenged with S. enteritidis on day 4 post feed withdrawal. In trials 3 and 4, one group of hens received S. typhimurium or S. muenchen, respectively, 1 day after feed was withdrawn. Transmission of S. enteritidis was evaluated by challenging the center hen in rows of 11 hens per row with S. enteritidis at 4 days post feed withdrawal and following the progression of the S. enteritidis down the row of hens over time. In trials 1 and 2, where hens received S. typhimurium 5 days prior to feed withdrawal, shedding of the S. enteritidis challenge was significantly reduced in hens on day 10 postchallenge in trial 1 and on days 3 and 10 postchallenge in trial 2 compared with the hens subjected only to the molt procedure. Significantly fewer S. enteritidis were recovered in livers and spleens at day 9 postchallenge in trial 2 from hens receiving the prior S. typhimurium infection. In trial 3, where hens received S. typhimurium 1 day after feed withdrawal, S. enteritidis transmission was significantly reduced in these hens on days 3, 10, and 24 postchallenge. In trial 4, similar in methodology to trial 3 except that, rather than S. typhimurium, hens received S. muenchen, a Salmonella organism totally lacking any antigen cross-reactive with S. enteritidis, S. enteritidis transmission was significantly reduced on days 3, 10, 17, and 24 postchallenge, suggesting that factors other than specific immunity were involved in the observed resistance to S. enteritidis infection. These results indicate that prior infection of a flock with a non-S. enteritidis paratyphoid Salmonella can reduce S. enteritidis problems that may occur during a molt.  相似文献

Two trials were conducted to determine the effect of induced molt on the reisolation of Mycoplasma gallisepticum (MG) from commercial leghorn hens that had been eyedrop-inoculated with MG at 10 weeks of age. Chickens were maintained in a conventional floored chicken house on dry litter through 100 weeks of age. At age 64 weeks, 4 days (Trial 1), and at 100 weeks (Trial 2), hens were swabbed and cultured for MG and then molted in biological isolation units. Swabs were again taken at the end of each molt. No difference was observed in the number of MG isolations between molted hens and controls that did not undergo molting. However, a significant decrease in MG isolations was observed in both trials from swabs obtained when hens were housed on dry litter floors as compared with swabs taken from the same hens after 18 days (Trial 1) or 21 days (Trial 2) of confinement in isolation units.  相似文献

The purpose of this study was to determine the ability of three strains or isolates of Pasteurella multocida (serotype 3,4) to generate chemotactic factors for heterophils when exposed to pooled turkey serum. Results indicated that each bacterial strain or isolate (M-9, CU, and 86-1913) was associated with the production of chemotactic factors, but the more pathogenic bacterial isolate (86-1913) elicited greater heterophil migration in chemotaxis studies.  相似文献

A study involving 7 white-egg and 3 brown-egg commercial layer strains was conducted at the North Carolina Piedmont Research Station to assess the effects cage population 3-bird vs. 4-bird cages and alternative molt programs on performance through 113 wk of age (791 d). The flock was divided into 3 groups; not molted (NM), maintained in continuous production; molted using feed restriction (FR), an industry-type 14-d feed withdrawal molting program initiated at 463 d of age; and molted using a nonfasted/anorexic program (NF), in which an ad libitum supply of a low protein, low energy molt feed was provided. Productivity, egg size, and egg quality were monitored at 28-d periods throughout the test providing the performance data for the white-egg and brown-egg strains with regard to these management factors. The performance for the 3-bird or 4-bird cage groups for either type of layer was similar, except that birds in 4-bird cages had higher feed consumption in the first production phase than did the birds in 3-bird cages. In the same time period the white-egg birds in the 3-bird cages had a greater percentage of cracked eggs and a smaller percentage of Grade A large eggs than those in 4-bird cages. No differences were observed in egg income for the 2 population sizes for either type of layer. However, due to their higher feed consumption, the white egg birds in the 4-bird cages had higher feed cost per hen for the first cycle, which was offset by the egg income component due to their increased production rate. The molted layers outperformed the nonmolted layers for both types of layers in terms of overall income over feed costs. The FR molted hens, under a program that has traditionally been used by commercial producers, outperformed NF-molted hens, indicating that further refinements are needed to make the NF program economically competitive with the FR molting program.  相似文献

Previous studies demonstrated that Salmonella enteritidis infections in hens undergoing molt via feed withdrawal were more severe than in full fed hens. We conducted two trials to determine if immunizing specific-pathogen-free, Salmonella-culture-negative hens via aerosol exposure to MeganVacl, a commercially available attenuated Salmonella typhimurium vaccine, would reduce transmission of S. enteritidis from infected hens to uninfected but contact-exposed hens during a molt. In trial 1, one group of hens received two aerosol doses of vaccine 2 wk apart whereas a second group of hens remained nonvaccinated. In trial 2, the vaccinated group received only one dose of vaccine. Two weeks after the final immunization, feed was removed from all the hens, and on day 4, the center hen in rows of 11 hens received a dose of 3 x 10(5) (trial 1) or 1.3 x 10(6) (trial 2). Transmission to the unchallenged hens was followed 3, 10, 17, and 24 days later. Vaccination reduced the horizontal spread of S. enteritidis in vaccinated hens compared with their nonvaccinated counterparts, with vaccinated hens shedding significantly less S. enteritidis on day 10 postchallenge in trial 1 and on days 3, 10, 17, and 24 in trial 2. Recovery of S. enteritidis from ovaries was significantly reduced in the vaccinated hens in trial 1 and from livers/spleens, ovaries, and cecum in trial 2. These studies indicate that immunization of hens with a live S. typhimurium vaccine could help reduce S. enteritidis problems during a molt situation.  相似文献

An experiment was conducted to determine the effects of age at inoculation and induced molt on the reisolation of Mycoplasma gallisepticum (MG) from commercial leghorn hens that had been eyedrop-inoculated with F strain MG at either 10 or 66 wk of age. Chickens were maintained in biological isolation units from 10 wk of age through 78 wk of age. At 70 wk of age (premolt), hens were swabbed, cultured for MG, and molted. Swabs were taken both at the end of molt (postmolt [74 wk]) and again 4 wk later (postmolt+4 [78 wk]). A significant (P < or = 0.05) decrease in MG isolations was observed in the postmolt swabs as compared with the premolt swabs of hens inoculated at either 10 or 66 wk of age. A significant (P < or = 0.05) increase in isolations was observed in the postmolt+4 swabs as compared with the postmolt swabs of hens inoculated at either 10 or 66 wk of age. For the hens inoculated at 10 wk, no significant difference was found in premolt as compared with postmolt+4 MG isolations; however, for hens inoculated at 66 wk, a significant (P < or = 0.05) decrease was observed between premolt and postmolt+4 isolations. Significantly (P < or = 0.05) fewer MG isolations were obtained from the premolt swabs of hens inoculated at 10 wk as compared with hens inoculated at 66 wk. No significant difference in MG isolations was observed in either the postmolt or postmolt+4 swabs between hens inoculated at either 10 or 66 wk.  相似文献

Serial passage of Salmonella enteritidis (SE) yields heterophil-adapted SE (HASE) strains that have resulted in decreased shedding of SE in feces and reduced egg contamination. Additionally, increasing the number of heterophil passages further reduced the number and frequency of fecal shedding. To evaluate SE and heterophil interaction, nine SE strains were fluorescein isothiocyanate-labeled when viable. There were six wild-types: SE TK 474, SE TK 584, SE TK 599, SE TK 600, SE TK 655, and SE TK 657; and three HASE strains: TK 499 heterophil adapted five times, TK 598 heterophil adapted six times, and TK 605 heterophil adapted 11 times. Trials were repeated seven times in duplicate with heterophils isolated from seven healthy chickens. Heterophils were incubated with the bacterial strains at 41 C for 15 min, and 10,000 heterophils were analyzed by flow cytometry. Percentage of phagocytosis and mean channel number of fluorescence were compared. Both parameters were significantly increased for all HASE-type strains compared with wild-type, nonadapted SE strains. Increased phagocytosis of HASE bacterial strains may be significant in processing and elimination of the HASE strains and may be related to the protective effect of HASE by decreased shedding of wild-type SE challenge strains.  相似文献

In the commercial egg industry the management practice of using feed withdrawal to induce a flock to molt has been under extreme scrutiny. This is because animal rights groups have voiced their concern about using this practice. Thus, about 5 yr ago, the United Egg Producers (UEP) commissioned 5 universities to conduct experiments to develop alternative molting programs that used nonwithdrawal feeding programs to molt laying hens. The studies conducted to date used techniques ranging from feeding hens without added salt in the diet to using readily available, low-cost feed ingredients to develop molt diets that are low in energy level and protein content. The results of these studies indicated that molting laying hens without feed withdrawal could be done successfully. Research at the University of Illinois found that feeding laying hens diets consisting of wheat middlings, soybean hulls, and corn (low protein and low energy) were successful in providing for acceptable postmolt egg production performance and economic benefit compared with using a standard feed withdrawal method. Therefore, after 5 yr of experimenting with nonwithdrawal molting methods done in several university settings, the egg industry has successfully adopted these methods of molting laying hens. In addition, based on the finding of these experiments, the UEP has revised their recommended molting guidelines to state that only nonwithdrawal molting methods will be permitted after January 1, 2006. Thus, these guidelines will apply to approximately 82% of the US egg industry.  相似文献

Genetic line and diet affect chicken heterophil activity and gene expression, and the combination of these factors can enhance disease resistance. This study evaluated the effects of immune modulating diets on heterophil/lymphocyte (H/L) ratio and heterophil chemokine expression in distinct genetic lines. Fayoumi and Leghorn chickens were fed a basal diet or immune modulating diets enhanced with β-glucans, ascorbic acid, or corticosterone. H/L ratios and heterophil gene expression in response to in vitro stimulation with Salmonella enteritidis (SE) were evaluated on days 1, 3, 7, and 21 of diet treatment. The stress-mimicking corticosterone diet influenced H/L ratio in the Leghorn line, but not the Fayoumi line, suggesting resistance to stress-induced immunosuppression in the Fayoumi line. Leghorn line H/L ratios were increased on days 1 and 3 of corticosterone diet treatment, but not days 7 or 21. Expression of CXCLi2 by SE stimulated heterophils was higher in the Leghorn line, suggesting that Leghorns rely more heavily on inflammatory response than do Fayoumis. Corticosterone diet was associated with reduced CXCLi2 expression in heterophils from both lines. Dietary β-glucan or ascorbic acid did not affect H/L ratio or CXCLi2 expression, suggesting that benefits of these immunomodulators may not be evident in healthy birds.  相似文献

The aim of this study was to compare the ability of laying hen abdominal macrophages during the second production cycle by using two different methods of induced molting. Two groups of Single Comb White Leghorn hens were induced to molt at the end of their first production cycle using feed restriction and ZnO supplementation. Macrophages were isolated from the abdomen and in vitro cytotoxic ability, at which point macrophage bactericidal moiety nitric oxide (NO) was recorded. Serum IgM and IgG titers against sheep red blood cells (SRBC) were determined at various stages: before molting (BM), 5% production level (5P), peak production stage (PP) and at the end of production (EP) level after fast and Zn-induced molt. Macrophages adherence percentage remained unaffected (p ≤ 0.05) during all production cycles, whereas the macrophage engulfment percentage and engulfment/cell was significantly higher (p ≤ 0.05) at PP in both fast and Zn-induced molted groups, as compared to all other studied stages. Macrophage NO production was increased (p ≤ 0.05) at PP and after SRBC and lipopolysaccrides (LPS) stimulus, when molted with ZnO supplementation. Serum total antibody titer against SRBC increased serum IgG and IgM titers during the second production cycle by Zn-induced molt. However, molting stress greatly reduced IgG and IgM production at the 5P stage. Serum Zn concentration increased with the onset of production but decreased at the EP stage irrespective of their molting regimes. Our results validate the strengthened innate and acquired immune response during the second production cycle after Zn-induced molting instead of fasting.  相似文献

The objective of the present study was to evaluate the welfare level of hens in four molting methods by comparing their behavior, physical condition and performance. In total, 96 White Leghorn layers (72 weeks of age) were used. The four molting methods consisted of 10 days of feeding and the first 3 days of water withdrawal WFW); 10 day feed withdrawal (WF) followed by ad libitum access to a layer diet from day 11, 28 days of restricted feeding (RF) and 28 days without withdrawing their feed (NW). The RF and NW hens were fed a low energy diet. The WFW and WF hens showed two clear phases of behavior during the 10‐day fasting period. Explorative and stereotyped behavior increased for WFW and WF hens just after their fast. Standing‐resting then increased for WFW and WF hens while lying‐resting decreased. In contrast, RF and NW hens did not show clear behavioral changes during the 28‐day molt period. The weight loss of the NW hens was more gradual compared with the other three methods. No significant differences were seen in any productive trait among the four methods during the postmolt period. In conclusion, the welfare level of RF and NW, especially NW, was higher compared with WFW and WF.  相似文献

Heterophil function was evaluated in 16 healthy chickens and in 46 chickens with experimentally induced staphylococcal tenosynovitis. In paired blood samples, heterophils from chickens with tenosynovitis had a significant increase in adherence, chemotaxis, phagocytosis, and bacterial killing of Staphylococcus aureus compared to heterophils from healthy chickens. The percent adherence of heterophils to nylon fiber columns increased significantly from a 78.4% mean +/- 6.6% standard deviation to 87.6% +/- 3.2% after induction of staphylococcal tenosynovitis. Heterophil movement following in vitro exposure to saline or endotoxin was increased in chickens with tenosynovitis; 3 +/- 1 heterophils/0.25 mm2 to 10 +/- 6 heterophils/0.25 mm2 and 136 +/- 29 heterophils/0.25 mm2 to 340 +/- 74 heterophils/0.25 mm2, respectively. Endotoxin-activated serum was chemoattractive for heterophils from all chickens. Flow cytometry was used to define the heterophil population on light scatter histograms, evaluate individual cell phagocytosis of latex beads, and quantitate the number of beads phagocytosed per heterophil. When incubated with increased numbers of beads, only heterophils from chickens with tenosynovitis phagocytosed higher numbers of beads. At heterophil to bead ratios of 1:10, the percentage of heterophils that phagocytosed beads increased from baseline values of 37.8% +/- 9.0% to post-infection values of 67.3% +/- 7.5%. Using 1:20 heterophil to bead ratios, heterophil phagocytosis increased from 38.7% +/- 9.9% to post-infection values of 79.8% +/- 7.3%. Heterophils from all chickens were able to phagocytose and kill log phase staphylococcal bacteria. After phagocytosis, the heterophils from chickens with staphylococcal tenosynovitis rapidly decreased the number of viable bacterial colony forming-units per milliliter by approximately one log.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
