This study was designed to determine what effects the birth process would have on development of the somatotrophic axis in neonatal pigs. Eight crossbred sows were selected (n = 4 natural birth and n = 4 Caesarian section) for the present study. Blood and tissue samples from 38 pigs were collected at birth. Twenty pigs were maintained with natural birth sows until sacrificed for blood and tissue collection at 2 wk of age. Gestational age at birth did not differ (P > 0.16) between natural birth and C-section pigs. Average daily gain (ADG) from birth until 2 wk of age was reduced (P < 0.0001) by 39.3% in the C-section pigs as compared to the natural birth pigs. Serum growth hormone (GH) did not differ (P > 0.86) at birth, but was greater (P < 0.024) at 2 wk in C-section pigs. Serum insulin-like growth factor 1 (IGF-1) was greater at birth (P < 0. 0025) and at 2 wk of age (P < 0.035) in the natural birth pigs. Serum concentration of IGF-2 did not differ at birth (P > 0.8) but was greater (P < 0.043) in natural birth pigs at 2 wk of age. Pituitary content of GH mRNA and GH-releasing hormone receptor mRNA did not differ (P > 0.90) between groups regardless of age; however, expression of both mRNAs declined (P < 0.0003) from birth until 2 wk of age. There tended to be a birth type X age interaction (P < 0. 082) for liver IGF-1 mRNA such that C-section pigs had a greater expression at 2 wk of age. Liver IGF-1 mRNA expression increased (P < 0.0001) in both groups from birth to 2 wk of age. Liver expression of GH receptor mRNA was greater in C-section pigs at birth (P < 0. 04) and 2 wk of age (P < 0.03). These data provide evidence that the natural birth process affects postnatal development and/or function of the somatotrophic axis in the neonatal pig.  相似文献

This study was designed to examine the physiology and behavior of pigs whose dams were snared and then injected with ACTH during gestation. Administration of ACTH to dams during pregnancy has been shown to replicate the effects of prenatal stress in other species. Control sows (n = 8) were given no treatment, whereas the treatment sows (ACTH, n = 8) were immobilized by snaring the snout and then administered an i.v. injection of ACTH (1 IU/kg BW) weekly from 6 to 12 wk of gestation. A pig was killed from each sow at 1, 30, and 60 d of age. The hypothalamus, pituitary gland, adrenal glands, and liver were immediately obtained to determine the amounts of corticotropin-releasing hormone (CRH), beta-endorphin, and mRNA for pro-opiomelanocorticotropin (POMC), the ACTH receptor (ACTH-R), and insulin-like growth factor I (IGF-I). Pituitary corticotrope and somatotrope cell numbers and adrenal cortex-to-medulla area ratios (CORT:MED) were also determined. Pigs' behaviors were recorded at 6 and 8 wk of age. At 75 d of age, a blood sample was taken and a biopsy puncture was created on one pig from each litter, then pigs were stressed by mixing. Blood was sampled every other day for 10 d to determine plasma cortisol concentrations and differential leukocyte counts. Biopsy damage was evaluated for healing. At 1 d of age, control pigs tended to weigh more (P = .09), have a lower expression of ACTH-R mRNA (P = .01) and IGF-I mRNA (P = .01), and a lower CORT:MED (P = .04) than ACTH pigs. At 30 d of age, control pigs had a greater concentration of beta-endorphin (P = .01) and tended to have a lower concentration of CRH (P = .09) and IGF-I mRNA (P = .10) than ACTH pigs. At 60 d of age, control pigs tended to have lighter pituitary glands (P = .08), a lower expression of POMC mRNA (P = .02), and a CORT:MED (P = .003) than ACTH pigs. At 8 wk of age, control pigs performed a higher frequency of belly nosing (P = .07) and oral vice behaviors (P = .01) than ACTH pigs. In response to mixing stress, control pigs had lesser concentrations of plasma cortisol (P = .03) and healed faster (P = .006) than ACTH pigs. Thus, exogenous ACTH and restraint during gestation alters the HPA axis of the sow's offspring, and during stressful situations later in life health, and therefore welfare, may be compromised.  相似文献

Three experiments were conducted to test the hypothesis that supplementing nursery pig diets with a mixture of carbohydrases (CS) will improve pig performance and nutrient digestibility. The CS used in these experiments contained 7 units/g of alpha-1,6-galactosidase, 22 units/g of beta-1,4-mannanase, beta-1,4 mannosidase, and trace amounts of other enzymes. In Exp. 1, 108 pigs weaned at d 21 of age were fed one of three diets containing 0 (control), 0.1, or 0.2% CS for 5 wk, based on a three-phase feeding program (1, 2, and 2 wk). Over the entire 35-d period, ADG was not affected (P > 0.05) by treatment, but supplementing 0.1% CS increased (P < 0.05) gain:feed by 9%. Experiment 2 used 10 gilts fitted with simple T-cannula in the terminal ileum at 3 wk of age. After cannulation, pigs were fed the same control Phase I and II diets, but the Phase III diet contained either 0 or 0.1% CS. Ileal samples were collected for the 3 d following the 5-d adjustment period during Phase III. Apparent ileal digestibility of GE, lysine, threonine, and tryptophan was greater (P < 0.05) in the CS diet. In Exp. 3, 90 pigs weaned at 21 d of age were fed the same control Phase I and II diets, but the Phase III diet contained either 0 or 0.1% CS. Phase III diets were fed for 3 wk. Average daily gain of the CS group was greater (P < 0.05) than the control group during wk 3. Gain:feed ratio was greater (P < 0.05) for the carbohydrase group during the entire Phase III period. Four pigs per treatment were killed at the end of Exp. 3 to measure villus height and to determine the concentration of raffinose and stachyose in different parts of the gastrointestinal tract. Average villus height was greater (P < 0.05) in pigs fed the CS diet. Carbohydrase supplementation decreased (P < 0.05) the concentration of stachyose in freeze-dried digesta from the proximal and distal small intestine. Raffinose concentration, on the other hand, was decreased (P < 0.05) by CS supplementation only in the distal small intestine. These lower concentrations suggest that CS improved the digestibility of carbohydrate in soybean meal. In conclusion, the addition of CS to Phase I and Phase II nursery diets containing low levels of soybean meal did not improve pig performance, but its addition to corn-soybean meal-based Phase III nursery diets improved gain:feed ratio and energy and AA digestibility.  相似文献

A study was conducted to compare growth performance and behaviors of group-farrowed pigs with those of confinement-farrowed pigs. A total of 216 pigs (8 wk of age) were used, with an equal number of group-farrowed or confinement-farrowed pigs from 1 farrowing batch. Group-farrowed pigs were farrowed in bedded, individual pens and mingled into a group of 8 litters at 12 (±1.3) d of age. Pigs were weaned at 33 (±1.3) d and remained in the farrowing room until 8 wk of age. Confinement-farrowed pigs were farrowed in farrowing crates. At weaning (32 ± 2.0 d of age), confinement-farrowed pigs were mixed and moved to pens of 9 pigs in a confinement nursery and remained there until 8 wk of age. At 8 wk of age, pigs from the 2 housing systems were allocated to 24 pens of 9 pigs in a confinement growing-finishing barn, with 12 pens from each of the housing systems. Within farrowing system, pigs from different groups or pens were mixed upon entering the growing-finishing barn. Individual BW was recorded at allotment (wk 0) to the growing-finishing barn and every 2 wk thereafter for 14 wk. Feed intake and G:F were monitored on a pen basis every 2 wk between wk 0 and 14 of the study period. Behaviors of pigs were video recorded in 6 pens of each housing treatment for 24 h on the day of mixing (d 0), d 7, and d 14 after mixing in the finisher barn. The video recordings were scanned at 5-min intervals to calculate behavioral time budgets for lying, standing, eating, drinking, and belly nosing. Data were analyzed using the Proc Mixed model of SAS with repeated measures. Compared with confinement-farrowed pigs, group-farrowed pigs spent more time lying (85.7 vs. 82.7%; SE = 0.75; P < 0.001) and belly nosing (0.05 vs. 0.02%; P < 0.05) and less time standing (5.8 vs. 7.5%; SE = 0.49; P < 0.01) and eating (7.3 vs. 9.3%; SE = 0.40; P < 0.001). The difference in behavioral time budgets was associated with differences in performance of pigs from the 2 housing systems. Group-farrowed pigs exhibited greater ADG (866 vs. 814 g; SE = 10.3; P < 0.01) for the initial 2 wk after mixing, less ADFI (2,004 vs. 2,188 g; SE = 42.5; P < 0.05), and improved G:F (0.431 vs. 0.393; SE = 0.0078; P < 0.01) for the entire 14-wk study period compared with confinement-farrowed pigs. These results suggest that group-farrowed pigs were more efficient than confinement-farrowed pigs in utilizing dietary energy for BW gain by lying more and standing and eating less during the growing and finishing period.  相似文献

Full term crossbred sows were selected to study the interaction of the immune system, hypothalamus-pituitary-adrenal axis, and growth in pigs born by Cesarean section (c-section; n=4 sows) or vaginal birth (n=4 sows). Gestation length and birth weight did not differ between vaginal birth and c-section pigs (P=0.34 and 0.62, respectively). Blood and tissue samples were collected from 44 pigs at birth. Forty-five pigs were weaned at 13 d. On d 14, pigs received an i.p. injection of lipopolysaccaride (LPS; 150 microg/kg) or saline at min 0, and blood samples were collected at -20, -10, 0, 5, 10, 20, 40, 60, 90, and 120 min. Vaginal birth pigs had 21% greater average daily gain than c-section pigs on d 14 (P<0.01). Basal serum concentrations of adrenocorticotrophin (ACTH) and cortisol were greater in c-section than vaginal birth pigs at birth (P<0.01) but were not different at 14 d (P=0.99 and 0.80, respectively). LPS increased serum concentrations of ACTH, cortisol, interferon-gamma (IFN-gamma), and tumor necrosis factor-alpha (TNF-alpha; P<0.01) but the response was not different between c-section and vaginal birth (P>0.22). Basal serum concentrations of TNF-alpha tended to be greater in c-section vs vaginal birth pigs at 14 d (P=0.0967); however, basal serum concentrations of IFN-gamma tended to be lower in c-section pigs vs vaginal birth pigs at 14 d (P=0.0787). Expression of interleukin (IL)-6, IL-6 receptor, IL-1beta, and TNF-alpha mRNA did not differ between vaginal birth and c-section pigs but changed in an age and tissue dependent manner. Thus, reduced growth rate of c-section pigs is associated with altered immune system function.  相似文献

The objective of this study was to determine the association between body weight of pigs at 7 wk of age, specific management factors, and previous body weight. Among 3736 pigs from 8 commercial farms, greater 7-wk weight was associated with greater birth weight (P = 0.001), greater weaning weight (P = 0.001), and earlier age at weaning (P = 0.001). Farms with the earliest average weaning age had the highest health status. Farms with later weaning ages tended to use nurse sows for light-weight pigs. Pigs from farms using all-in/all-out management in the nursery had greater 7-wk weights (P = 0.01). The total mortality and culling rate during the nursery phase (weaning to approximately 7 wk of age) was 6.8%. Higher losses were observed among pigs with low weaning weights (< 4.1 kg) compared with those with higher weaning weights. More of the variation due to 7-wk weight was due to individual sow rather than to farm of origin.  相似文献

Two studies were carried out in different wean-to-finish barns to determine the effects of double stocking on pig growth performance. In Study 1, pigs (n = 1,560) were used in a randomized complete block design with a 2 x 2 factorial arrangement of treatments: initial stocking treatment (Single [52 pigs/pen] vs Double [104 pigs/pen] stocked for 10 wk after weaning) and weighing frequency (High [12 times during the study] vs Low [3 times]) on pig performance from weaning (5.9+/-0.01 kg BW; 17 d of age) to harvest (114+/-0.67 kg BW). Floor and feeder space per pig were 0.650 m2 and 4 cm and 0.325 m2 and 2 cm for the single- and double-stocked treatments, respectively. In Study 2, pigs (n = 1,458) were used in a randomized complete block design to evaluate two initial stocking treatments (Single [27 pigs] vs Double [54 pigs] stocked for 10 wk after weaning) on pig performance from weaning (4.8+/-0.01 kg BW; 15 d of age) to harvest (24 wk after weaning). Floor and feeder space per pig were 0.640 m2 and 3.4 cm and 0.320 m2 and 1.7 cm for single- and double-stocked pens, respectively. In both studies, double-stocked pigs were split at the end of wk 10 into two equal-sized groups of similar mean BW and CV of BW, and one group was moved to a different pen in the same building. In Study 1, performance was not affected (P > 0.10) by frequency of weighing. For the first 10 wk after weaning, the Double compared to the Single treatment had lower ADG (7.7 and 7.9%, for Studies 1 and 2, respectively; P < 0.001) and lighter pigs at wk 10 (6.8 and 7.3%, respectively; P < 0.001). During the first 10 wk in Study 1, Double compared to the Single pigs had lower ADFI (7%; P < 0.001) but similar gain:feed (P > 0.10). From wk 11 to harvest, pigs on Double and Single treatments had similar (P > 0.10) ADG in both studies and, in Study 1, ADFI was unaffected by initial stocking treatment, but double-stocked pigs had greater gain:feed (4%, P < 0.01). Double-stocked pigs required an additional 2 d to reach a fixed harvest BW (P < 0.05) in Study 1 and were lighter (4%; P < 0.05) at 24 wk after weaning in Study 2. Carcass measures were similar (P > 0.10) for double- and single-stocked pigs. Double-stocked pigs that were moved at the end of 10 wk had growth performance similar (P > 0.10) to those that remained in the original pen. In summary, double stocking reduced growth rate to 10 wk after weaning but subsequently had no effect on growth rate and improved feed efficiency.  相似文献

The present study was designed to evaluate the effects of synthetic ACTH (1-24, tetracosactid) and porcine CRH on the plasma levels of cortisol and PGF2alpha metabolite in cycling gilts (n = 3) and castrated boars (n = 3). The experiments were designed as crossover studies for each gender separately. Each animal received, during three consecutive days; 1) ACTH (Synacthen Depot) at a dose of 10 microg/kg body weight in 5 ml physiological saline, 2) porcine CRH at a dose 0.6 microg/kg body weight in 5 ml physiological saline or 3) physiological saline (5 ml). The test substances were administered via an indwelling jugular cannula in randomized order according to a Latin square. The administration of ACTH to cycling gilts resulted in concomitant elevations of cortisol and PGF2alpha metabolite with peak levels reached at 70.0 +/- 10.0 and 33.3 +/- 6.7 min, respectively. Similarly, the administration of ACTH to castrated boars resulted in concomitant elevation of cortisol and PGF2alpha metabolite with peak levels reached at 60.0 +/- 0.0 and 20.0 +/- 0.0 min, respectively. Cortisol peaked at 20 min after administration of CRH in both cycling gilts and castrated boars with maximum levels of 149.3 +/- 16.5 nmol/l and 138.3 +/- 10.1 nmol/l, respectively. It can be concluded that administration of synthetic ACTH (tetracosactid) to pigs caused a concomitant elevation of cortisol and PGF2alpha metabolite levels in both cycling gilts as well as castrated boars. The administration of CRH to pigs resulted in an elevation of cortisol levels in both cycling gilts and castrated boars. Conversely, PGF2alpha metabolite levels were not influenced by the administration of CRH either in cycling gilts or in castrated boars.  相似文献

It was hypothesized in these studies that differences would exist in baseline immune and performance measures among different breeds of pigs, and that these differences would be affected by age of the pig. Baseline immune, plasma cortisol (CORT) concentrations, and performance measures were determined among Berkshire (n = 36), Duroc (n = 18), Meishan (n = 54), Landrace x Yorkshire (White X; n = 36), and Yorkshire (n = 36) pigs at 4, 8, and 12 wk of age. All piglets were weaned at 17 to 21 d of age and moved to a common nursery environment. Total white blood cell (WBC), leukocyte differential, plasma CORT, immunoglobulin G (IgG) concentrations, natural killer cytotoxicity, neutrophil phagocytosis (PHAGO), and chemotaxis (CHTX) were evaluated. At all ages, plasma CORT was greatest in Meishan pigs, and least in Yorkshires (P < 0.05). Plasma IgG increased with age for all breeds (age: P < 0.01; breed x age: P < 0.005), except that in Meishans, IgG decreased. Natural killer cytotoxicity was greatest (P < 0.05) among Meishan pigs. There were breed x age interactions for neutrophil PHAGO (P < 0.001) and CHTX (P < 0.001). Overall, Yorkshire pigs showed the greatest (P < 0.05) percentage of PHAGO but the least (P < 0.05) CHTX. White X pigs had the greatest (P < 0.05) CHTX response. Berkshire pigs had the greatest (P < 0.001) numbers of neutrophils. At 12 wk of age, Meishan pigs had the least BW gain (P < 0.001), and Durocs had the greatest G:F (P < 0.001). There were no significant sex differences for immune (P > or = 0.15), performance (P > or = 0.20), or CORT (P = 0.70) measures. Pig breed and age influenced both baseline immune measures and plasma CORT in growing pigs, suggesting that pig breed and age are important factors influencing the response to various stressors or infectious challenges.  相似文献

A study was conducted with 20 weaned barrows (14 d, 4.98 +/- 0.21 kg) to determine the effect of feeding spray-dried plasma (SDP) after weaning on the pig's stress response to a lipopolysaccharide (LPS) challenge. After weaning, pigs were fed a diet containing 0 or 7% SDP for 7 d. On d 6 after weaning, all pigs were nonsurgically fitted with a jugular catheter. On d 7 after weaning, the pigs were given i.p. injections of either saline or LPS (150 microg/kg BW) followed by serial blood collection every 15 min for a 3-h period. Following the 3-h blood collection, all pigs were killed and tissue was collected for mRNA analysis. Pig weight on d 7 after weaning was not affected by dietary treatment (P > 0.21). Pigs fed the diet with SDP had lower (P < 0.05) levels of hypothalamic corticotropin-releasing hormone (CRH) mRNA, pituitary gland CRH receptor mRNA, and adrenal gland adrenocorticotropin-releasing hormone (ACTH) receptor mRNA. Dietary treatment did not affect pituitary gland proopiomelanocortin (POMC) mRNA. No effect of LPS treatment was observed in any of the mRNA levels examined. For both serum ACTH and cortisol, there was a significant diet x LPS treatment interaction (P < 0.01) such that both the ACTH and cortisol responses to the LPS challenge were greater in the pigs fed the diet with SDP than in the pigs fed the diet without SDP. For pigs given the saline injection, diet did not affect basal serum cortisol concentration; however, basal serum ACTH concentration was lower in those pigs fed the diet with SDP (P < 0.0001). A diet x LPS treatment interaction (P < 0.024) was observed for adrenal gland mRNA expression for steroidogenic acute regulatory (StAR) protein such that the LPS-induced increase in StAR mRNA was greater in the pigs fed SDP than in pigs fed the diet without SDP. These results demonstrate that pigs fed a diet with SDP have an increased activation of the pituitary-adrenal axis following an LPS challenge compared to pigs fed a diet without SDP.  相似文献

Two hundred and twenty-four pigs (112 boars, 112 gilts) housed in pens of seven pigs per pen were used in a 2 x 2 x 2 factorial design, with the factors of vaccination with a gonadotropin-releasing factor (GnRF) vaccine (Improvac; 0 or 2 mL at 13 and 17 wk of age), porcine somatotropin (pST; 0 or 5 mg/d from 17 wk of age), and gender. Pigs were weighed and feed intake was measured from 17 wk of age until slaughter at 21 wk of age. Body composition was estimated by dual-energy X-ray absorptiometry in two focus pigs per pen at 17 and 21 wk of age. Testes and ovary weights at slaughter were decreased by Improvac treatment (P < 0.001), but were not altered by pST treatment (P > 0.44). Daily gain was lower for gilts than boars (1,128 vs. 1,299 g/d, P < 0.001) and was increased by pST (1,172 vs. 1,255 g/d, P = 0.003) and Improvac (1,150 vs. 1,276 g/d, P < 0.001) treatments. Feed intake (as-fed basis) was lower in gilts than in boars (2,774 vs. 3,033 g/d, P = 0.002), was decreased by pST (3,037 vs. 2,770 g/ d, P = 0.002), and was increased by Improvac treatment (2,702 vs. 3,105 g/d, P < 0.001). As a result of the differences in feed intake and daily gain, feed conversion efficiency (gain:feed) was lower for gilts than for boars (0.403 vs. 0.427 P = 0.025), was improved by pST (0.385 vs. 0.452, P < 0.001), but was unchanged by Improvac treatment (0.423 vs. 0.410, P = 0.22). Carcass weight was lower in gilts than in boars (75.3 vs. 77.0 kg, P = 0.012), was unchanged by pST treatment (75.9 vs. 76.4 kg, P = 0.40), and was increased by Improvac treatment (75.1 vs. 77.2 kg, P = 0.003). Lean tissue deposition rate was lower in gilts than in boars (579 vs. 725 g/d, P < 0.001), was increased by pST (609 vs. 696 g/d, P < 0.001) and by Improvac treatment (623 vs. 682 g/d, P = 0.014). Fat deposition rate tended to be lower in gilts than in boars (214 vs. 247 g/d, P = 0.063), decreased by pST treatment (263 vs. 198 g/d, P < 0.001), and increased by Improvac treatment (197 vs. 264 g/d, P < 0.001). For pigs treated with both pST and Improvac, daily gain and lean tissue deposition rate was greater than for pigs that received either treatment alone, whereas fat deposition rate and feed intake did not differ from untreated control pigs. In conclusion, Improvac increased growth rate through increased lean and fat deposition, but concomitant use of Improvac and pST increased lean gain above either alone, while negating the increase in fat deposition in pigs treated with Improvac.  相似文献

It is difficult to predict the size of pituitary corticotroph tumors in dogs with Cushing's disease (pituitary-dependent hyperadrenocorticism [PDH]) without pituitary imaging techniques. The purpose of this study was to examine the relationship between plasma adrenocorticotropin hormone (ACTH) precursor concentration and pituitary size in dogs with Cushing's disease. Plasma concentrations of ACTH precursors (pro-opiomelanocortin [POMC]/pro-ACTH) and pituitary tumor height/brain area were measured in 36 dogs with pituitary corticotroph adenomas of various sizes. There was a correlation between tumor size (measured as the pituitary tumor height/brain area ratio [P/B]) and POMC/pro-ACTH concentration (r = .70; P < .0001). Dogs with P/B > or = 0.40 x 10(-2) mm(-1) had higher concentrations of ACTH precursors than dogs with P/B < 0.40 x 10(-2) mm(-1) (median concentration 85 pmol/L, range 15-1,350 pmol/L, n = 14 versus 15 pmol/L, range 15-108 pmol/L, n = 22; P < .0001). With a threshold of 35 pmol/L of POMC/pro-ACTH concentration, the estimated sensitivity and specificity of the kit were 93% (95% confidence interval [CI], 79-100%) and 86% (95% CI, 73-100%), respectively. We interpret these data as indicating that measurement of POMC and pro-ACTH might be of value in the characterization of tumor size in dogs with Cushing's disease. Low POMC/pro-ACTH concentrations make it unlikely that a large pituitary tumor exists in dogs with PDH.  相似文献

Sows subjected to prenatal stress have been found to produce offspring that have altered responses to stress. Our objective was to determine if exposing a sow to stress would alter the response of the offspring to lipopolysaccharide (LPS) at 2 mo of age or their response to mixing stress at 4 mo of age. Sow treatments consisted of intravenous injections of ACTH (1 IU/kg of BW), exposure to rough handling for a 10-min duration (rough), or no treatment (control) once per week from d 42 to 77 of gestation. At 2 mo of age, pigs from each treatment, 1 per litter (n = 21, 17, and 15 for the ACTH, rough, and control treatments, respectively), were challenged with 2 μg of LPS/kg of BW or saline, or served as a noninjected control. Their behavioral response to a human approach test and salivary cortisol were measured. At 4 mo of age, 1 pig from each treatment (n = 14, 14, and 15 for the ACTH, rough, and control treatments, respectively) was taken from its home pen and placed in a pen of unfamiliar pigs. At this time, a punch biopsy wound (6 × 6 mm) was created to measure the ability of the pig to heal the wound. At this same time, each pig received a 1-mL intramuscular injection of 20% ovine red blood cells (oRBC), and then a second injection of oRBC at 21 d postmixing. Blood samples were collected 3 times per week for 2 wk and then once a week for 4 more weeks. Blood samples were analyzed for cortisol, porcine corticosteroid-binding globulin, antibody response to oRBC, and nitric oxide production by macrophages. Behavior was recorded during the first 5 d after mixing. All pigs in the LPS challenge responded with characteristic sickness behavior; however, pigs in the rough treatment showed less sickness behavior than those in the other 2 treatments (P < 0.05). Maternal stress treatment did not affect (P < 0.43) salivary cortisol. Pigs from all treatments responded similarly to mixing stress with regard to cortisol, porcine corticosteroid-binding globulin, antibody titers, nitric oxide production, and hematology measures, and all pigs experienced the same amount of aggression in response to mixing. Without altering peripheral measures of stress responsivity, prenatal stress enhanced the ability of pigs to cope with a simulated immune challenge, which could prove to be an adaptation to challenging environments.  相似文献

We evaluated the effect of maternal obesity before and throughout gestation on offspring hypothalamic-pituitary-adrenal axis function. Multiparous Rambouillet by Columbia crossbred ewes were fed either 100% of National Research Council (NRC) recommendations (control, C) or 150% of NRC recommendations (obese, OB) from 60 d before mating until lambing. Ten lambs born to OB ewes (five males and five females), and eight lambs born to C ewes (three male and five female) were studied. From delivery to weaning lambs were maintained with their mothers, who were all fed 100% NRC recommendations. After weaning, all lambs were group housed and fed the same diet to meet NRC requirements. At 19 mo of age lambs were placed in individual pens and fed a pelletized diet to meet maintenance requirements. Jugular vein catheters were placed and 2 d later lambs received an intravenous (i.v.) adrenocorticotropic hormone (ACTH) challenge followed by an i.v. corticotropin-releasing hormone (CRH)/arginine vasopressin (AVP) challenge 1 d later. Thirty d later offspring were again catheterized and placed into metabolism crates for 2 d before receiving an isolation stress test. ACTH and cortisol responses to the isolation stress test and CRH/AVP challenge and cortisol responses to ACTH challenge were determined. Cortisol was quantified via radioimmunoassay and ACTH was quantified using an Immulite 1000; both were analyzed using repeated measures using the MIXED procedure of SAS. Offspring from OB ewes had elevated basal plasma ACTH and cortisol compared with C offspring before all three challenges (P < 0.05). Offspring from OB mothers tended (P = 0.06) to have a greater ACTH response after an i.v. CRH/AVP injection than offspring from C mothers (12,340 ± 1,430 vs 8,170 ± 1,570 area under the curve, respectively). Cortisol response to the CRH/AVP and ACTH challenges was not influenced by maternal nutrition (P = 0.46) and averaged 4.77 ± 0.2 μg/dL and 1.94 ± 0.01 μg/dL, respectively. The ACTH response following the isolation stress test was also similar (P = 0.82) for OB and C offspring (147 ± 20 pg/mL), and cortisol response during the isolation stress test was similar between C and OB offspring (P = 0.64, 5.25 ± 0.3 μg/dL). These findings suggest that maternal obesity before and during gestation does not affect stress responses by the offspring, but has an impact on hypothalamic-pituitary-adrenal sensitivity. The lack of differences in cortisol release under the influence of difference concentrations of ACTH during the CRH/AVP challenge could indicate adrenal dysfunction in OB offspring.  相似文献

Four experiments were conducted to determine the interactive effects of pharmacological amounts of Zn from ZnO and Cu from organic (Cu-AA complex; Cu-AA) or inorganic (CuSO(4)) sources on growth performance of weanling pigs. The Cu was fed for 4 (Exp. 1) or 6 (Exp. 2, 3, and 4) wk after weaning, and Zn was fed for 4 (Exp. 1) or 2 (Exp. 2, 3, and 4) wk after weaning. Treatments were replicated with 7 pens of 5 or 6 pigs per pen (19.0 ± 1.4 d of age and 5.8 ± 0.4 kg of BW, Exp. 1), 12 pens of 21 pigs per pen (about 21 d of age and 5.3 kg of BW, Exp. 2), 5 pens of 4 pigs per pen (20.3 ± 0.5 d of age and 7.0 ± 0.5 kg of BW, Exp. 3), and 16 pens of 21 pigs per pen (about 21 d of age and 5.7 kg of BW, Exp. 4). In Exp. 1 and 2, Cu-AA (0 vs. 100 mg/kg of Cu) and ZnO (0 vs. 3,000 mg/kg of Zn) were used in a 2 × 2 factorial arrangement. Only Exp. 1 used in-feed antibiotic (165 mg of oxytetracycline and 116 mg of neomycin per kilogram feed), and Exp. 2 was conducted at a commercial farm. In Exp. 3, sources of Cu (none; CuSO(4) at 250 mg/kg of Cu; and Cu-AA at 100 mg/kg of Cu) and ZnO (0 vs. 3,000 mg/kg of Zn) were used in a 3 × 2 factorial arrangement. In Exp. 4, treatments were no additional Cu, CuSO(4) at 315 mg/kg of Cu, or Cu-AA at 100 mg/kg of Cu to a diet supplemented with 3,000 mg/kg of Zn from ZnO and in-feed antibiotic (55 mg of carbadox per kilogram of feed). In Exp. 1 and 2, both Zn and Cu-AA improved (P < 0.001 to P = 0.03) ADG and ADFI. No interactions were observed, except in wk 1 of Exp. 2, where Zn increased the G:F only in the absence of Cu-AA (Cu-AA × Zn, P = 0.04). A naturally occurring colibacillosis diarrhea outbreak occurred during this experiment. The ZnO addition reduced (P < 0.001) the number of pigs removed and pig-days on antibiotic therapy. In Exp 3, ADFI in wk 2 was improved by Zn and Cu (P < 0.001 and P = 0.09, respectively) with no interactions. In wk 1, G:F was reduced by ZnO only in the absence of Cu (Cu × Zn, P = 0.03). Feeding Zn decreased fecal microbiota diversity in the presence of CuSO(4) but increased it in the presence of Cu-AA (Cu source × Zn, P = 0.06). In Exp. 4, Cu supplementation improved the overall ADG (P = 0.002) and G:F (P < 0.001). The CuSO(4) effect on G:F was greater (P < 0.001) than the Cu-AA effect. Our results indicate that pharmacological amounts of ZnO and Cu (Cu-AA or CuSO(4)) are additive in promoting growth of pigs after weaning.  相似文献

The importance of birth-to-weaning average daily gain as a determinant of weight at a final age and yield of marketable pork was investigated. Treatments were imposed to create variation in birth-to-weaning ADG independent of birth weight. Newborn pigs were cross-fostered to create litters of four through 14 pigs/litter. Creep feed was offered to pigs from 5 d of age or during last 2 d before weaning at 13 to 20 d (average = 17 d). Growth rate and carcass dissection data were obtained from 195 barrows that were slaughtered at an average age of 170 d (SD = 7.5), weight of 109 kg (SD = 10.5). All traits measured were influenced by birth dam and sire (P < 0.01). Quadratic and cubic effects (P < 0.09) of litter size on birth-to-weaning ADG and weaning weight were different between the creep feeding treatments. Data revealed a positive influence (P < 0.04) of creep feeding from 5 d of age on birth-to-weaning ADG and weaning weight in larger size (> 8) litters. Importance of the independent variables birth weight, birth-to-weaning ADG, weaning weight, and birth weight plus birth-to-weaning ADG in determination of measures of postweaning growth and yield of marketable pork were examined by step-down regression analysis. Initial models included the linear and quadratic effects of the independent variables. In general, R2 for models ranked birth weight < birth-to-weaning ADG < d-17 weaning weight < birth weight + birth-to-weaning ADG. The R2 of models for BW at 170 d of age were 0.11 (P < 0.01) using birth weight as the independent variable, 0.16 (P < 0.01) using birth-to-weaning ADG, 0.19 (P < 0.01) using d-17 weaning weight, and 0.21 (P < 0.01) using birth weight + birth-to-weaning ADG. The model for effect of birth-to-weaning ADG on BW at 170 d of age indicated that a 10-g advantage in birth-to-weaning ADG produced a 0.94-kg advantage in BW at 170 d of age. Positive relationships (P < 0.05) between birth-to-weaning ADG and measures of postweaning growth and carcass yield suggest management practices that increase birth-to-weaning ADG may be advantageous in pork production.  相似文献

The objective of the present study was to evaluate the temporal aspects associated with corticotropin-releasing hormone (CRH) and vasopressin (VP) stimulated bovine adrenocorticotropic hormone (ACTH) secretion in vitro and in vivo. For the in vitro studies, bovine anterior pituitary glands were enzymatically dispersed to establish primary cultures. On day 5 of culture, cells were challenged for 3 h with medium alone (Control) or various combinations and concentrations of bovine CRH (bCRH) and VP. Both CRH and VP each increased (P < 0.05) ACTH secretion. Maximal increases in ACTH secretion occurred in response to 0.1 microM CRH (5.5-fold) and 1 microM VP (3.7-fold), relative to Control cells. The in vivo portion of the study examined possible temporal differences in the activation of the pituitary-adrenal axis by CRH and VP. Jersey cows were randomly assigned to one of four groups (n = 8 cows/group): (i) Control (saline); (ii) bCRH (0.3 microg/kg BW); (iii) VP (1 microg/kg BW) and (iv) bCRH (0.3 microg/kg BW) + VP (1 microg/kg BW). Jugular blood samples were collected at 15-min intervals for 4 h pre- and for 6 h post-treatment; samples were also taken at 1, 5 and 10 min post-treatment. Plasma concentration of ACTH did not differ among treatment groups for the 4-h pre-treatment period. At 1 min post-treatment, bCRH + VP, VP and bCRH increased ACTH secretion by 22.4-, 9.6- and 2.2-fold, respectively, relative to Control (32.7 +/- 7.2 pg/ml). Maximal plasma concentration of ACTH occurred at 5, 10 and 15 min post-treatment for the VP (1017.7 +/- 219.9 pg/ml), bCRH + VP (1399.8 +/- 260.1 pg/ml) and bCRH (324.8 +/- 126.2 pg/ml) treatment groups respectively. Both the in vitro and in vivo data demonstrated that while VP acutely activates the bovine pituitary-adrenal axis, CRH-induced ACTH secretion is slower in onset but of longer duration. The present study also provides insight into the dynamics of ACTH and cortisol (CS) responsiveness to CRH and VP in cattle.  相似文献

An experiment involving 118 pigs was conducted to evaluate the influence of weaning pigs at four different ages on in vivo and in vitro cell-mediated immunity. One-half of each litter was weaned at 2, 3, 4 or 5 wk of age; the other one-half remained with the sow as nonweaned controls. Phytohemagglutinin skin-test responses were determined on all pigs. Blastogenic responses of mitogen-stimulated lymphocyte cultures were determined before and after weaning. The intradermal response to phytohemagglutinin was reduced (P less than .001) in pigs weaned when 2 or 3 wk old and was suppressed (P less than .05) in those weaned when 4 wk old. In vivo cellular immunity was not altered by weaning in 5-wk-old pigs. The capability of lymphocytes to undergo blastogenesis in response to phytohemagglutinin was decreased in pigs weaned at 2 and 3 wk of age (P less than .001 and P less than .01, respectively). Pokeweed mitogen-stimulated blastogenesis was lower (P less than .01) in pigs weaned at 2 wk of age. Mitogen-stimulated lymphocyte blastogenesis was similar (P greater than .10) in control pigs and those weaned when 5 wk old. These data suggested that weaning pigs when younger than 5 wk old causes physiological changes detrimental to cellular immune reactivity. Those changes could alter disease susceptibility in young pigs.  相似文献

An experiment was conducted to determine the efficacy of dietary conjugated linoleic acid (CLA) as a growth promotant in weanling swine. Weanling pigs (n = 192; 7.6 kg and 29 d of age) were randomly assigned to four treatments that were arranged as a 2 x 2 factorial. Concentrations of dietary CLA (0 or 0.6%) and antibiotics (+/-) constituted the main effect variables. Dietary CLA treatments consisted of a 1% addition of an oil containing 60% CLA isomers or 1% soybean oil, and dietary antibiotic treatments were antibiotics or no antibiotics. The experimental diets were fed for 9 wk in four phases (1, wk 1; 2, wk 2 and 3; 3, wk 4 through 6; and 4, wk 7 through 9), after which all pigs were fed identical medicated diets for the duration of the finishing phase. Live weights were recorded at wk 17 postweaning and at marketing to determine any residual effects of dietary treatments on finisher ADG and days to market. Medicated diets fed during phases 1 and 2 contained 55 mg carbadox/kg; during phase 3 contained 299 mg tilmicosin/kg; and during phase 4 contained 110 mg tylosin and 110 mg sulfamethazine/kg. Pigs fed medicated diets had higher overall ADG than pigs fed unmedicated diets for wk 0 through 9 (P < 0.03). Gain:feed (G:F) was greater for pigs fed medicated diets than for pigs fed unmedicated diets during phase 1 (P < 0.03) and for the duration of the nursery phase (P < 0.03). There were no effects of CLA on ADG, ADFI, or G:F. There were no residual effects of nursery CLA or antibiotics on finisher ADG and days to market. Blood samples collected from a subset of pigs (n = 72) at the completion of phases 2, 3, and 4 were assayed for serum IGF-I and antibody concentrations to porcine reproductive and respiratory syndrome virus (PRRSV) and Mycoplasma hyopneumoniae. There was a tendency for pigs fed medicated diets to have greater IGF-I concentrations than pigs fed unmedicated diets at the completion of phase 4 (P < 0.06). Pigs fed CLA had greater antibody titers (P < 0.02) to Mycoplasma hyopneumoniae at d 63 than pigs fed diets without CLA. These results indicate that feeding 0.6% dietary CLA did not enhance growth performance in weanling swine and that the use of dietary antibiotics can increase production efficiency in nursery pigs. Furthermore, there were no interactions between CLA and dietary antibiotics on the variables addressed in this study.  相似文献

The objective of this study was to investigate the development of small intestinal size and digestive capacity of the jejunum in growing pigs. The weight, length, surface area, and mucosa weight of the small intestine were measured when pigs were 1, 3, 5, and 9 wk of age. Sucrase and alkaline phosphatase (ALP) activities of the jejunal brush-border membrane, prepared by differential centrifugation and Mg2+ precipitation, were determined at the respective postnatal stages. Body weights increased 7-fold from 2.7 kg at 1 wk to 23.32 kg at 9 wk postnatal. Body weight gains were greater (P < 0.05) from wk 3 to 5 than from wk 1 to 3. Weights of the small intestine and of the intestinal mucosa increased faster (P < 0.05) from 3 to 5 wk than from 1 to 3 wk; the slowest increase occurred from 5 to 9 wk. Weights of the duodenum, jejunum, and ileum, and mucosa from the respective sections increased (P < 0.05) as pigs grew from 3 to 9 wk. Mucosa weight relative to the weight of the section was greater (P < 0.05) for the duodenum and jejunum than for the ileum at 9 wk of age. Between the ages of 3 and 9 wk, the increase in mucosa weight was highest for the jejunum followed by the duodenum and the ileum. The increase was greatest for the duodenum followed by the jejunum and the ileum when mucosal weight was expressed per unit of appropriate intestinal section weight. There was a 55-fold increase in jejunal sucrase activity from 1 to 9 wk; the greatest rate of increase occurred between 5 and 9 wk. Total jejunal ALP activities in pigs at 9 wk was greater (P < 0.05) than at 5 wk, which in turn was greater than at 1 wk of age. In summary, increases in BW during the first 9 wk of postnatal growth in pigs are accompanied by significant developmental changes in digestive capacity including intestinal weights, length, and area as well as jejunal brush-border sucrase and ALP activities.  相似文献
