Vitamin A deficiency has rarely been reported in captive or free-ranging wildlife species. Necropsy findings in two captively housed southern sea otters (Enhydra lutris nereis) included irregular thickening of the calvaria characterized by diffuse hyperostoses on the internal surface. One animal also had moderate squamous metaplasia of the seromucinous glands of the nose. There was no measurable retinol in the liver of either sea otter. For comparison, hepatic retinol concentration was determined for 23 deceased free-ranging southern and northern (Enhydra lutris kenyoni) sea otters from California and Alaska. Free-ranging otters were found to have similar hepatic retinol concentrations (316 +/- 245 mg/kg wet weight) regardless of their location and subspecies. All of these values were significantly higher than the levels in the affected animals. Consumption of a diet with very low vitamin A concentrations and noncompliance in daily supplementation are hypothesized as the causes of vitamin A deficiency in these two sea otters.

Killer whales and sea otters maintained in captivity are the subjects of routine health monitoring programs, and interest in immunologic studies in sea otters has been rising recently in response to potential impacts from infectious disease and environmental pollution on the threatened southern sea otter population. Development of species-specific reagents for immunologic studies in these two marine mammals is currently in its infancy. In this study, killer whale and sea otter immunoglobulin-specific polyclonal antibodies were generated, and used to develop tests for serum Ig concentration in the killer whale (Orcinus orca) and the southern (Enhydra lutris nereis) and northern sea otter (Enhydra lutris lutris). Killer whale serum IgG was purified using caprylic acid/ammonium sulfate precipitation. Sea otter plasma IgG was purified using protein-A-agarose. Polyclonal anti-Ig antisera were produced in rabbits, and specificity confirmed by immunoelectrophoresis. Radial immunodiffusion was used to measure Ig concentration in serum or plasma samples derived from 21 captive killer whales, 18 wild and 4 captive southern sea otters and 15 wild and 4 captive northern sea otters grouped by age. Mean killer whale serum Ig concentration (+/-95% confidence interval) ranged from 15.04 +/- 3.97 g/l for animals aged 0-5 years to 26.65 +/- 9.8 g/l for animals aged >10 years. Mean sea otter serum Ig concentration (+/-95% confidence interval) ranged from 28.39 +/- 11.00 g/l for southern sub-adults to 32.76 +/- 11.58 g/l for southern adults. No significant difference in serum Ig concentration was found between southern and northern sea otters. Serum Ig concentrations in two northern sea otter pups were low compared to those of adult sea otters. The two serum Ig quantitation assays produced were highly specific and reproducible and will be useful additions to the limited number of tests available for immune function in these marine mammal species.

Feedlot producers often exceed NRC recommendations for vitamin A and D supplementation; however, increased concentrations of these vitamins have been shown to limit adipocyte differentiation in vitro. A feedlot trial was conducted using 168 Angus crossbred steers (BW = 284 ± 0.4 kg) allotted to 24 pens. The experiment had a 2 × 2 factorial arrangement of treatments: no supplemental vitamin A or D (NAND), 3,750 IU vitamin A/kg dietary DM with no supplemental vitamin D (SAND), no supplemental vitamin A and 1,860 IU vitamin D/kg dietary DM (NASD), and 3,750 IU and 1,860 IU vitamin A and D/ kg dietary DM (SASD), respectively. Serum, liver, and intramuscular and subcutaneous adipose tissue retinol concentrations were decreased in (P < 0.001) in cattle fed the no supplemental vitamin A diets (NAND and NASD combined) compared with those consuming supplemental vitamin A (SAND and SASD combined) diets. In addition, intramuscular retinol concentration was 38% less than in the subcutaneous depot. Serum 25(OH)D(3) concentrations were reduced (P < 0.001) during the first 70 d when cattle were fed no supplemental vitamin D diets (NAND and SAND combined); however, liver 25(OH)D(3) concentrations remained unchanged (P > 0.10) through d 184. Serum and liver 25(OH)D(3) concentrations increased (P < 0.001) with vitamin D supplementation (NASD and SASD combined). The DMI, ADG, G:F, and morbidity were not affected (P > 0.10) by dietary concentration of vitamin A or D. There were vitamin A and D interactions (P < 0.03) for backfat thickness and USDA Yield grade. Cattle fed the NAND diet had greater (P < 0.03) Yield grades than other treatments because of greater (P < 0.005) 12th rib backfat thickness in NAND steers than the NASD and SAND steers. Vitamin D concentrations were attenuated and minimal carcass adiposity responses to vitamin D supplementation were observed. Feeding a diet without supplemental vitamin A increased (P < 0.05) Quality grades and marbling scores and tended (P = 0.06) to increase ether extractable lipid of the LM. As retinol and 25(OH)D(3) concentrations in feedlot cattle declined as a result of a lack of dietary supplementation, adipose accretion increased, resulting in elevated Quality and Yield grades. Withdrawal of supplemental vitamin A, D, or both from the finishing diet of feedlot beef cattle had minimal impact carcass composition.

1. The effects of tomato powder supplementation on performance, egg quality, serum and egg yolk carotenoids, vitamins and malondialdehyde (MDA) concentrations in were investigated in laying hens in mid-lay.

2. A total of 90 laying hens, 49 weeks old, were divided into 3 groups consisting of 6 replicate cages, 5 birds per cage. Birds were randomly fed on one of three diets: basal diet and basal diet added with 5 or 10?g tomato powder per kg diet.

3. As tomato powder concentration increased, there were linear increases in feed intake, egg production, egg weight and yolk colour and a linear decrease in feed conversion. Shell weight, shell thickness and Haugh unit remained unchanged in response to dietary treatments.

4. Concentrations of serum and egg yolk lycopene, β-carotene, lutein and vitamin A increased for both diets including tomato powder, whereas MDA decreased linearly with increasing supplemental tomato powder concentration.

5. Tomato powder supplementation increased egg production persistency and increased carotenoids and vitamin A contents in egg yolk, accompanied by reduced yolk lipid peroxidation.

1. The effect of L-carnitine supplemented into experimental diets with varying dietary protein concentrations (50, 200 and 400 g/kg) on body weight gain and plasma insulin-like growth factor-I (IGF-I) concentration in chicks was examined. 2. Dietary L-carnitine supplementation provided 0, 200, 500 and 1000 mg/kg. Chicks were given the diet ad libitum for 10 d. 3. When L-carnitine was provided as 500 or 1000 mg/kg, body weight gain was significantly improved in birds receiving the 200 and 400 g protein/kg diets. 4. There was an interaction between dietary L-carnitine and protein content on plasma IGF-I concentration. L-carnitine supplementation had little influence on plasma IGF-I concentrations in birds receiving the low protein (50 g/kg) diet. When dietary L-carnitine concentrations were increased from 0 to 1000 mg/kg in the adequate protein (200 g/kg) diet, plasma IGF-I concentrations were also increased. However, when dietary L-carnitine content was more than 500 mg/kg in the 400 g/kg protein group, plasma IGF-I concentration decreased with increasing dietary L-carnitine content. 5. Body weight change correlated significantly with the alteration in plasma IGF-I concentrations in chicks given diets with adequate dietary protein. 6. In conclusion, the improvement in body weight gain caused by dietary L-carnitine supplementation was achieved when chicks were given their dietary protein requirement, which may be partially explained by an increase in plasma IGF-I concentration.

The influence of fish oil (highly unsaturated) and beef tallow (highly saturated) with vitamin E (100 IU/kg) supplementation on the antioxidant status of broiler chicken cockerels was investigated. Chicks were fed a control diet with no added fat, 40 g/kg each of fish oil and beef tallow diets, respectively, from 11 to 42 days of age. Tocopherol concentration and the rate of lipid peroxidation, thiobarbituric acid reactive substance (TBARS) in liver, fatty acid composition of the liver lipids, blood serum total antioxidant status (TAS), and reduced glutathione (GSH) content were determined. Vitamin E supplementation of the diet increased liver alpha-tocopherol content in chicks regardless of the type of dietary fat. Fish oil diet resulted in higher liver TBARS value while beef tallow diet showed lower values compared to the control diet. Vitamin E supplementation reduced liver TBARS as well as serum GSH, and raised serum TAS for all diets. Serum GSH was the same for vitamin E supplemented diets regardless of the fat supplement. Fish oil diets resulted in a significant increase in hepatic lipid n-3 PUFA content. A significant positive correlation was found between liver TBARS and n-3 PUFA content. No relationships were established, however, between liver TBARS and n-6 PUFA or saturated fatty acids. The results suggest that feeding oils rich in n-3 PUFA increases tissue concentration of these fatty acids, consequently increasing tissue lipid peroxidation and reducing the antioxidative status of broiler chickens. Supplementing high levels of vitamin E with such oils may increase tissue oxidative stability. Serum TAS or GSH may be used as a measure of antioxidative status in chickens.

An in vivo experiment was conducted to compare the effects of dietary supplementation with oleuropein and/or α‐tocopheryl acetate on growth performance, serum lipid concentrations and lipid oxidation of Japanese quail meat during refrigerated storage. Performance and slaughtering parameters were not affected by dietary treatments. The diets supplemented with oleuropein at the levels of 150 or 200 mg/kg were more effective in delaying lipid oxidation in breast and thigh meats compared with the control diet. The dietary inclusion of neither vitamin E nor oleuropein at different levels did not significantly affect the fatty acid compositions of the breast meat compared with the control diet. The diets supplemented with oleuropein at the levels of 150 or 200 mg/kg had significantly the highest polyunsaturated fatty acid and omega‐3 fatty acid contents in thigh meat compared with the vitamin E diet. The ratio of omega‐6 fatty acids to omega‐3 FAs in thigh meat of quails fed diet supplemented with vitamin E at the level of 200 mg/kg was equivalent to those of quails fed the diets supplemented with oleuropein at the levels of 100 and 150 mg/kg . The results showed that the dietary oleuropein supplementation at 150 mg/kg level may be used in quail diets enriched with the polyunsaturated fatty acids of vitamin E as a natural antioxidant.

Two experiments involving 496 cross-bred pigs evaluated the efficacy of various dietary levels of vitamin E, with or without supplemental fat, on postweaning pig performance and weekly serum and terminal tissue alpha-tocopherol concentrations. The first trial involved 248 pigs weaned at an average of 15 d of age and 4.8 kg BW. The experiment was a randomized complete block design conducted in seven replicates. Vitamin E was added as dl-alpha-tocopheryl acetate at 0, 20, 40, 60, 80, 100, 150, or 200 IU/kg diet. Pigs were bled initially and at 7-d intervals for a 42-d period. Liver and s.c. adipose tissue samples were collected from six pigs per treatment group at 42 d. In Exp. 2, a 2 x 4 factorial arrangement of treatments in a randomized complete block design was conducted in seven replicates. The experiment used a total of 248 pigs weaned at 19 d of age and averaged 6.4 kg BW. Four vitamin E levels (0, 20, 40, and 60 IU/kg diet) and two added fat levels of 0 or 5% were fed for 35 d. Four pigs per treatment pen were bled weekly, and at 35 d a total of four pigs per treatment group were killed and liver, heart, and s.c. adipose tissues were collected and analyzed for alpha-tocopherol. The basal diet in both experiments contained an average 7.9 IU for period 1, and later diets averaged 11.0 IU vitamin E/kg. In both experiments serum alpha-tocopherol concentrations declined from weaning to 7 d after weaning and continued to decline each week after weaning when the basal diets were fed. Serum alpha-tocopherol concentrations increased (P < 0.01) each week as the dietary vitamin E level increased in both experiments. In Exp. 2, when fat was added to the diet serum alpha-tocopherol concentrations were higher (P < 0.01) than in diets without added fat. Liver, heart muscle, and adipose tissue alpha-tocopherol concentrations increased (P < 0.01) as vitamin E level increased, but at the higher dietary vitamin E level the liver surpassed the adipose tissue in its alpha-tocopherol concentration. Liver and adipose alpha-tocopherol concentrations were higher (P < 0.01) when fat was added to the diet. These results indicate that supplementation of 40 to 60 IU/kg diet with added fat resulted in a relatively constant balance of serum and tissue concentration of alpha-tocopherol during the nursery period, but when fat was not supplemented a dietary vitamin E level of 80 to 100 IU/kg diet may be needed. The current NRC recommendations for vitamin E for the pig from 5 to 20 kg BW may need to be reevaluated.

1. The effect of a range of supplementations of vitamin A to the laying hen on the concentration of vitamins A, E, ascorbic acid and carotenoids in the maternal liver, the egg yolk and the embryonic liver were investigated. 2. Four groups of 25 Rhode Island Red hens were fed on standard layer-breeder diets with concentrations of supplemented vitamin A ranging from 0 to 120 mug/g retinol equivalents from 28 weeks of age. After 3 months, the concentration of vitamin A in the maternal liver was found to be greatly enhanced in proportion to the increasing rates of supplementation with the vitamin. However, the concentration of vitamin E in the maternal liver was markedly reduced by high dietary contents of vitamin A. 3. The concentration of vitamin A in the yolk of the hens' eggs was markedly increased by the dietary supplementation. However, the concentration of both vitamin E and carotenoids in the yolks were significantly reduced by high dietary contents of vitamin A. 4. The concentration of vitamin A in the liver of the embryo and the day old chick was greatly increased by the high concentrations of maternal vitamin A provision. However, the concentration of vitamin E, carotenoids and ascorbic acid in the embryonic/neonatal liver were significantly reduced by high contents of vitamin A in the maternal diet. 5. The susceptibility of the embryonic/neonatal liver to lipid peroxidation was significantly increased as a result of high provisions of maternal vitamin A. 6. It is concluded that excessive provision of vitamin A to the laying hen results in an adverse effect on vitamin E, carotenoids and ascorbic acid in the embryonic/neonatal liver and can compromise the antioxidant status of the progeny.

An experiment was conducted to evaluate the effects of a new phytase supplementation in Jinding laying ducks fed different concentrations of non-phytate P (NPP) on production performance, mineral retention, and bone and plasma minerals. A 14-wk experiment was conducted using 200-d-old female laying ducks. A total of 1,000 laying ducks were randomly allocated to 5 treatments and fed 5 diets: a control diet that contained an adequate concentration of NPP (0.45%) and 4 diets that were deficient in NPP (0.38, 0.32, 0.25, and 0.18%, respectively) but supplemented with phytase at 500 U/kg. Decreasing the NPP content from 0.45 to 0.18% in the diets with phytase supplementation had no detrimental effects on performance. However, Cu and Zn retention was significantly lower (P < 0.05) for laying ducks consuming the 0.18% NPP diet with phytase supplementation. Likewise, laying ducks fed the 0.18% NPP diet had a significant reduction in bone ash, Ca, P, and Cu contents, and in serum P and Cu (P < 0.05). Furthermore, the decrease in NPP content in the diet with phytase supplementation significantly increased (P < 0.05) P retention. Therefore, with the supplementation of this novel phytase at 500 U, it is possible to reduce dietary concentrations of NPP to 0.25% and maintain the normal performance of laying ducks.

In context with the frequent observations of excessive iron (Fe) storage in captive black rhinoceroses (Diceros bicornis), it has been suggested that both an excessive dietary Fe content and a lack of dietary Fe-chelating substances, such as tannins, is the underlying cause. Therefore, studies on the effects of tannin supplementation to captive diet are warranted. Six captive rhinoceroses were fed their normal zoo diet (N), and a similar diet supplemented with either tannic acid (T, hydrolysable tannin) or quebracho (Q, condensed tannins), and the total antioxidant capacity (TAC) was measured as mmol Trolox equivalents per kg fresh faeces. The TAC values on diets N (1.24 +/- 0.39 mmol/kg fresh faeces) and T (1.34 +/- 0.33 mmol/kg fresh faeces) were similar, but significantly higher on diet Q (2.32 +/- 0.61 mmol/kg fresh faeces). In contrast to expectations, faecal TAC increased with increasing faecal Fe, possibly as a result of the fact that the faecal Fe content was positively correlated to the proportion of concentrate feeds in the diet, which also contain antioxidants, such as vitamin E, in addition to Fe. Increased antioxidant status caused by the use of tannin substances could have a beneficial effect on animal health, but if tannins should be incorporated in designed diets, other tannin sources, such as grape pomace should be tested.

结果表明,45 000 IU/kg维生素A组的肉鸡血清BGP含量相对于1 500 IU/kg维生素A组有显著的降低趋势(P=0.08);随着日粮维生素A水平的增加(1 500~45 000 IU/kg),血清CaBP浓度呈趋于显著的一次线性降低趋势(P=0.092),血清维生素D3与CT含量也呈现降低趋势(P=0.12,P=0.18),其中,以15 000、45 000 IU/kg维生素A组较低,尤以45 000 IU/kg维生素A组最低;血清PTH含量呈显著的一次线性增加(P=0.029)。

An experiment was conducted to determine the effects of dietary concentrations of Co on vitamin B12 production and fermentation of mixed ruminal microbes grown in continuous culture fermentors. Four fermentors were fed 14 g of DM/d. The DM consisted of a corn and cottonseed hull-based diet with Co supplemented as CoCO3. Dietary treatments were 1) control (containing 0.05 mg of Co/kg of DM), 2) 0.05 mg of supplemental Co/kg of DM, 3) 0.10 mg of supplemental Co/kg of DM, and 4) 1.0 mg of supplemental Co/kg of DM. After a 3-d adjustment period, fermentors were sampled over a 3-d sampling period. This process was repeated 2 additional times for a total of 3 runs. Ruminal fluid vitamin B12 concentrations were affected by Co supplementation (P < 0.01), and there was a treatment x day interaction (P < 0.01). By sampling d 3, cultures fed the basal diet supplemented with 0.10 mg of Co/kg had greater (P < 0.05) vitamin B12 concentrations than those supplemented with 0.05 mg of Co/kg of DM, and increasing supplemental Co from 0.10 to 1.0 mg/kg of DM increased (P < 0.01) ruminal fluid vitamin B12 concentration. Ruminal fluid succinate also was affected (P < 0.10) by a treatment x day interaction. Cobalt supplementation to the control diet greatly decreased (P < 0.05) succinate in ruminal cultures on sampling d 3 but not on d 1 or 2. Molar proportions of acetate, propionate, and isobutyrate, and acetate:propionate were not affected by the addition of supplemental Co to the basal diet. However, molar proportions of butyrate, valerate, and isovalerate increased (P < 0.05) in response to supplemental Co. The majority of long-chain fatty acids observed in this study were not affected by Co supplementation. However, percentages of C18:0 fatty acids in ruminal cultures tended (P < 0.10) to be greater for Co-supplemented diets relative to the control. Methane, ammonia, and pH were not greatly affected by Co supplementation. The results indicate that a total (diet plus supplemental) Co concentration of 0.10 to 0.15 mg/kg of dietary DM resulted in adequate vitamin B12 production to meet the requirements of ruminal microorganisms fed a high-concentrate diet in continuous-flow fermentors.

1. Experiments were conducted independently at two stations to measure the requirement for methionine in chick diets with crude protein (CP) varying in 8 steps from 140 to 280 g/kg diet (experiment 1) or from 90 to 300 g/kg (experiment 2). 2. Protein composition was the same at all protein concentrations within a trial. The diet was designed to be first-limiting in methionine and DL-methionine was added to provide 5 ratios of methionine to CP at each protein concentration. 3. Methionine required for maximum growth rate or maximum efficiency of food utilisation was estimated at each protein concentration by fitting a quadratic regression equation to the relevant data. The requirement was also estimated by fitting the Reading model to data for growth rate and methionine intake. 4. In both trials and by all three methods of estimation, the methionine requirement (g/kg diet) for maximum performance increased as a linear function of dietary CP concentration and nearly in direct proportion to CP. 5. It is concluded that diets which contain surplus protein, beyond that needed to maximise growth rate or food efficiency, need supplementation with methionine beyond that required when dietary protein is just adequate. A suitable rule for practical formulation is that methionine concentration in chick diets should be not less than 0.025 times the dietary CP concentration.

The aim of the study was to investigate the ability of vitamin C to increase the antioxidative and immunomodulating potential in healthy dogs. Fifteen dogs were tested for the effects of orally administered vitamin E (60 mg dl‐α tocopheryl acetate) in combination with vitamin C (0, 30 or 60 mg ascorbic acid crystalline). Three treatments (0, 30, 60 mg vitamin C) were tested in a 3 × 3 cross‐over study in three periods of 36 days. Pre‐prandial blood samples were taken for analysis of vitamins C, E, A, retinyl palmitate and stearate, antioxidant status [Thiobarbituric acid reactive substances (TBARS) and uric acid], biochemical and haematological analysis. Subpopulations of lymphocytes, mitogen‐induced peripheral blood mononuclear cell proliferation (PBMC) and serum IgA and IgG concentrations were determined. There was a trend (p = 0.056) for an increased plasma vitamin C concentration by vitamin C supplementation. There was no evidence that dietary treatment altered neither the other plasma vitamin concentrations nor TBARS and uric acid concentrations nor the subpopulations of the lymphocytes except for the number of CD4+ lymphocytes that increased with vitamin C supplementation. There was no effect of vitamin C on serum IgA and IgG concentration. A significant time × treatment interaction was demonstrated on PBMC’s to pokeweed, with an increase observed by 30 mg vitamin C supplementation but a decrease by 60 mg vitamin C supplementation. There was no clear evidence for an effect of dietary vitamin C on antioxidative capacity in healthy dogs fed a diet with vitamin E concentrations well above the recommendations. Yet, a limited number of immunological parameters were slightly affected.

Free-ranging browsers such as the black rhinoceros (Diceros bicornis) consume a diet that contains tannins, whereas the diets offered to them in captivity consist mostly of items known to contain hardly any such secondary plant compounds. Tannins could have potentially beneficial effects, including the chelation of dietary iron (iron storage disease is a common problem in black rhinos). Here, we tested the acceptance, and the consequences on digestion variables, of a low-dose tannin supplementation in captive animals. Eight black rhinoceroses from three zoological institutions were used. Faecal output was quantified by total faecal collection. Diets fed were regular zoo diets supplemented with either tannic acid (T, hydrolysable tannin) or quebracho (Q, condensed tannins); overall tannin source intake increased at 5-15 g/kg dry matter (DM) in relation to regular zoo diets. Adaptation periods to the new diets were >2 months. Additional data were taken from one hitherto unpublished study. Data were compared to measurements in the same animals on their regular zoo diets. All animals accepted the new diets without hesitation. There was no influence of tannin supplementation on digestion coefficients of DM and its constituents, or faecal concentrations of short-chain fatty acids or lactate. Water intake did not increase during tannin supplementation. Should the inclusion of dietary tannin sources be an objective in the development of diets for captive rhinoceroses, moderate doses such as used in this study are unlikely to cause relevant depressions of digestive efficiency and will not interfere with bacterial fermentation in a relevant way.

A three-year-old Border collie was presented with a two-week history of lethargy, stiff gait, polydipsia and polyuria. Biochemical analysis revealed hypercalcaemia. Serum concentrations of 25-hydroxyvitamin D (25[OH]D) and 1,25-dihydroxyvitamin D (1,25[OH]2D) were markedly elevated and parathyroid hormone was undetectable. Subsequent analysis of the dog's diet revealed that the food contained excessive amounts of vitamin D. The hypercalcaemia resolved following treatment with bisphosphonates and dietary change. Hypervitaminosis D was diagnosed in a second unrelated dog, which had been fed the same brand of dog food as case 1. The dog was also hypercalcaemic and had markedly elevated serum concentrations of 25(OH)D and 1,25(OH)2D. Hypervitaminosis D in dogs has been reported to occur secondarily to ingestion of either rodenticides containing cholecalciferol or antipsoriatic ointments that contain vitamin D analogues. Hypervitaminosis D has also been reported following the treatment of hypoparathyroidism. To the authors' knowledge, this is the first report of hypervitaminosis D in dogs following the accidental over supplementation of a commercial diet with vitamin D.
