Effects of dexamethasone, levamisole, or combined dexamethasone-levamisole administration on polymorphonuclear neutrophil (PMN) function in healthy, adult female goats were studied. Goats were assigned to treated (n = 6) and control (n = 6) groups. In experiment 1, treated goats were given levamisole (6 mg/kg of body weight, IM). In experiment 2, treated goats were given 0.1 mg of dexamethasone/kg, IV, for 3 consecutive days, 1 mg of dexamethasone/kg, IV, for 6 consecutive days, and 6 mg of levamisole/kg, IM, with a 4th injection of 1 mg of dexamethasone/kg. All injections were administered 12 hours before blood collection. The PMN were evaluated for random migration and chemotaxis under agarose, ingestion of Staphylococcus aureus, cytochrome C reduction, iodination, and antibody-dependent cell-mediated cytotoxicity. Levamisole alone did not alter the function of caprine PMN. Both doses of dexamethasone caused increased random migration and decreased cytochrome C reduction and iodination. Dexamethasone resulted in no changes in chemotaxis, S aureus ingestion, and antibody-dependent cell-mediated cytotoxicity. Random migration and cytochrome C reduction returned toward base line in cells from dexamethasone and levamisole-treated goats. Although iodination activity in cells from dexamethasone-treated goats remained significantly (P less than 0.05) lower than those of controls after levamisole administration, a rebound toward base-line activity occurred. 相似文献

Background: Peripheral blood neutrophils of untreated human cancer patients have been shown to have normal, increased, and decreased phagocytic activity, killing capacity, and/or oxidative burst activities. Objectives: The objectives of this study were to evaluate oxidative burst and phagocytic activities of peripheral blood neutrophils from tumor‐bearing dogs before therapy and compare them with neutrophil function of healthy control dogs. Methods: Heparinized whole blood was obtained from dogs with high‐grade lymphoma (n=23), sarcoma (n=13), or carcinoma (n=11), and healthy control dogs (n=11) for flow cytometric evaluation of oxidative burst and phagocytic activities. Percentage of bursting cells and amount of oxidative burst activity were determined after stimulation with phorbol 12‐myristate 13‐acetate (PMA) or Escherichia coli. Percentage of phagocytic cells and amount of phagocytic activity were determined after incubation with fluorescent E. coli. Results: Compared with control dogs, dogs with sarcoma (P=.004) and carcinoma (P=.05) had a lower percentage of neutrophils exhibiting oxidative burst activity after stimulation with PMA. Phagocytic activity was significantly lower in dogs with sarcomas compared with control dogs (P<.0001) and dogs with lymphoma (P=.01). Conclusions: Untreated carcinomas and sarcomas in dogs may suppress the percentage of neutrophils capable of oxidative burst when stimulated by PMA. Furthermore, sarcomas also may suppress the amount of phagocytic activity per neutrophil. Until further studies can be performed, the clinical significance of these findings is unknown. 相似文献

erythrocytic lipid peroxidation were increased and reduced glutathione levels decreased significantly (p < 0.05) in group I goats as compared to group II healthy goats. The results of the present study suggest that scabies infection

SUMMARY Ventricular septal defects were diagnosed in 3 familially-related Saanen goats, a 4-year-old doe and 2 female kids aged 18 weeks, by clinical examination, electrocardiography, phonocardiography and thoracic radiography. Findings were contrasted with those from 3 clinically normal goats. Cardiac catheterisation was attempted on the 2 affected kids before euthanasia. Blood pressure tracings and selective contrast cardioangiography successfully demonstrated the defect in one case, but the other died shortly after anaesthetic induction. The adult goat, suspected paternal grand-dam of the affected twins, died without premonitory signs at 4.5 years of age. Necropsy confirmed the diagnosis in all 3 cases. 相似文献

Previously we showed that the primer pheromone responsible for the "male effect" was produced in specific skin regions of castrated male goats by androgen treatments. In the present study, we examined whether androgen can also induce production of the male effect pheromone in female goats. Capsules containing dihydrotestosterone (DHT) or testosterone (T) were subcutaneously implanted into six ovariectomized (OVX) goats for 28 days. Small skin samples were collected from the head and rump regions, and the pheromone activity of their ether extracts was examined using a bioassay that monitors the electrophysiological manifestation of the hypothalamic gonadotropin-releasing hormone pulse generator as multiple-unit activity. Behaviors of OVX goats towards ovary-intact estrous goats were also examined before and at the end of DHT or T treatment. Before androgen treatment, neither the head nor rump skin samples in OVX goats showed pheromone activity. DHT treatment induced pheromone activity in the head skin sample of six OVX goats and in the rump skin sample of two OVX goats. Similar results were obtained by T treatment. In addition, OVX goats treated with T showed masculine-type sexual behaviors such as courtship and mounting behaviors towards the estrous goats. These results demonstrate that androgen is capable of inducing primer pheromone activity in the female and suggest that the synthesis pathway of the male effect pheromone exists in both sexes in the goat. 相似文献

The left and right kidneys, ureters, urinary bladder and urethra of 29 female clinically healthy Saanen goats were examined via transcutaneous and transrectal ultrasonography. In order to establish reference values the examinations were performed using a 5.0 MHz linear transducer to scan the right caudal costal part of the abdominal wall, right and left dorsal flanks and right and left inguinal regions of standing goats. A 5.0 MHz intracavity probe was used for transrectal ultrasonographic examination of the urinary bladder and urethra. The kidneys were examined in longitudinal and cross section and assessed subjectively. They could usually be seen from the 12th intercostal space on the right side and dorsal right flank. The right kidney was 8.0 ± 0.67 cm long and the left was 8.4 ± 0.64 cm long. The ureters could not be visualized in any of the goats. The length of the urinary bladder was 5.1 ± 1.38 cm, and its largest cross-sectional diameter was 2.6 ± 1.01 cm. The urethra was seen in 23 goats and appeared as echogenic lines with no visible lumen. The transition from the neck of the bladder to the internal urethral orifice extended beyond the brim of the pelvis in only one goat. 相似文献

The effect of dietary selenium (Se) deficiency on the generation of neutrophil chemotactic factors from goat polymorphonuclear leukocytes (PMN) was studied. Polymorphonuclear leukocytes were isolated from 2 groups of crossbred goats. One group (n = 3) was fed a diet deficient in Se; the other (n = 3) was fed a diet adequate in Se. The PMN from each goat were stimulated with calcium ionophore A23187 and supernatants were collected. Each supernatant was immediately assayed for its ability to elicit neutrophil chemotaxis (NCT) and neutrophil chemokinesis by the Boyden chamber technique and neutrophil chemiluminescence (NCL) by the luminol-dependent chemiluminescence assay. Measurements from each group were compared, using the Student's t test. There was no significant difference (P greater than 0.01) in neutrophil chemokinesis elicited by supernatants derived from stimulated PMN from the 2 groups. However, supernatants derived from PMN of goats fed the Se-deficient diet had a significantly decreased capacity (P less than 0.01) to stimulate NCT and NCL. In vitro incubation of PMN with Se before ionophore treatment produced supernatants with increased NCL stimulation (P less than 0.05), but none with increased NCT stimulation (P greater than 0.05). However, both activities were restored after goats fed the Se-deficient diet (P less than 0.01) were given Se subcutaneously. Seemingly, dietary Se deficiency is associated with depressed PMN generation of factors that stimulate NCT and NCL. In contrast to NCL factors, synthesis of factors stimulating NCT probably was not associated directly with PMN glutathione peroxidase concentration. 相似文献

Growth-related changes in pulmonary function values were investigated in 20 healthy French Alpine goats, aged between 20 and 550 days, weighing 7–55 kg. Pulmonary ventilation, mechanics of breathing and arterial oxyten tension were measured using standardized techniques and methods adapted for goats of different body sizes. The Ppl values and the tI/tTOT ratio showed no significant changes with age and body size. The ventilation values (Vt, Ve, mVI and mVE) increased linearly with growth. There was a significant correlation of age and body weight with dynamic lung compliance (Cdyn), total pulmonary resistance (RL), viscous work of breathing (Wvis tot) and minute viscous work (Wvis min) throughout the age range studied. Cdyn, Wvis tot and Wvis min increased and RL decreased with age and body weight. Arterial blood gases (PaO2 and PaCO2) did not show significant changes over the age range studied. Regression equations for each pulmonary function parameter are given with body weight as the independent variable. Data for the mechanics of breathing were compared with those elsewhere for cattle, horses, man and dogs. 相似文献

为了检测雌性山羊颈动脉体中是否存在卵泡刺激素受体(FSHR),探讨卵泡刺激素(FSH)是否可以影响雌性山羊颈动脉体的活动,本试验采用免疫组织化学SP法观察卵泡刺激素受体(FSHR)在山羊颈动脉体内的分布特点,利用IPP 6.0图像半定量分析系统分析FSHR在颈动脉体中实质细胞和非实质细胞上的表达量差异。结果显示,FSHR主要分布在颈动脉体的实质细胞(Ⅰ型细胞和Ⅱ型细胞)。Ⅰ型细胞中FSHR强阳性产物分布于细胞核,中等阳性产物分布于细胞膜,弱阳性或阴性产物分布于细胞质;Ⅱ型细胞中有强阳性产物;血管内皮细胞中有中等阳性或强阳性产物。FSHR在颈动脉体的实质细胞与非实质细胞中相对表达量呈显著性差异(P<0.05)。结果表明,证实FSHR在雌性非妊娠山羊颈动脉体内广泛表达,提示颈动脉体可能接受FSH的调节,这为研究FSH作用于非生殖系统及颈动脉体神经内分泌调节机制提供形态学依据。 相似文献

Previous research on energy requirements of female Saanen goats, using the factorial approach, has not considered the specific requirements for maintenance and growth during the pubertal phase. Thus, the purpose of this study was to estimate energy requirements for maintenance (Trial 1) and growth (Trial 2) of non‐pregnant and non‐lactating female Saanen goats at the pubertal phase from 30 to 45 kg. In Trial 1, the net energy requirements for maintenance (NEm) were estimated using 18 female Saanen goats randomly assigned to three levels of intake: ad libitum, and 70% and 40% of ad libitum intake. These animals were pair‐fed in six slaughter groups, each consisting of one animal for each level of intake. In Trial 2, the net energy requirements for growth (NEg) were estimated using 18 female Saanen goats, which were fed ad libitum and slaughtered at targeted BW of 30, 38 and 45 kg. The NEm was 52 kcal/kg0.75 of BW. The NEg increased from 3.5 to 4.7 Mcal/kg of BW gain as BW increased from 30 to 45 kg. Our results suggest that the guidelines of the major feeding systems for the entire growth phase may not be adequate for females at pubertal phase. 相似文献

Neutrophil function was evaluated on 2 occasions in 5 calves from each of the following age groups: 4 to 5 weeks, 9 to 11 weeks, 16 to 19 weeks, and 12 to 14 months. Of the neutrophil functions examined, the iodination reaction, which evaluates the activity of the myeloperoxidase-hydrogen peroxide-halide antibacterial system of the neutrophil, reflected the most marked differences among age groups. The iodination values for the 2 youngest age groups were approximately 52% of the value for the oldest cattle. This difference could not be attributed to the amount of myeloperoxidase in the neutrophil granules. Antibody-dependent cell-mediated cytotoxicity also tended to be lower in neutrophils from the 3 younger age groups. Nitroblue tetrazolium reduction, a measure of superoxide anion generation by neutrophils, was lower in the youngest age group only. The capability of neutrophils to ingest Staphylococcus aureus was higher in the 3 youngest groups of calves than in the oldest group. The observed differences in neutrophil function in young vs older calves may be partially responsible for the increased susceptibility of young calves to infectious disease. 相似文献

Two experiments were conducted to determine the response of Creole male goats treated with long days and melatonin implants, and the response of the anovulatory does to male effect using males treated only with artificially long days. All animals were allocated to open sheds. In Exp. 1, one group of males was under natural photoperiod (CG; n = 7); the second group was submitted to 2.5 mo of long days followed by the insertion of two s.c. melatonin implants (LD+MEL; n = 7); the third group was subjected only to 2.5 mo of long days (LD; n = 7). Testicular weight was measured every 2 wk. Plasma testosterone concentrations were determined weekly. A treatment x time interaction was detected (P < 0.001) for testicular weight and plasma testosterone concentration. In the LD+MEL and LD groups, testicular size and plasma testosterone levels varied in a similar way, but differed from those observed in CG (P < 0.001). In this latter group, testicular weight displayed seasonal variations and peaked in June, whereas in treated groups this peak occurred in March. In CG, testosterone varied in a seasonal manner and plasma concentrations increased in June and remained elevated throughout the study. In experimental groups, testosterone increased in February and peaked in March. In Exp. 2, one group of males was left under natural photoperiod (CG, n = 5) and the other one was submitted to 2.5 mo of artificially long days (LD, n = 4). On March 16, two control and two treated males were put in contact with 20 and 19 females, respectively. Sexual behavior of the bucks was observed during the 5 d following male introduction. Progesterone assays and estrous behavior were used to determine ovarian and behavioral responses of the females to teasing. The anogenital sniffing, nudging, and mount instances registered in LD-treated males were greater than those observed in CG (P < 0.05). Of the does exposed to CG, none ovulated and only two of 20 females displayed estrous behavior. All does in contact with LD-treated males ovulated and showed at least one estrous behavior during the 15 d following joining (P < 0.001). These results indicate that the sexual activity of male goats from subtropical latitudes can be induced using only artificially long days. In addition, males treated in this way are capable of stimulating sexual activity in anovulatory females by the male effect. 相似文献

OBJECTIVE: To investigate the effects of short-chain fatty acids (SCFAs) and pH on neutrophil oxidative burst, phagocytosis, and morphology after exposure to acetate, propionate, butyrate, or succinate at pH 5.5 and 6.7. SAMPLE POPULATION: Neutrophils isolated from bovine blood samples and Porphyromonas levii, Prevotella spp, and Bacteroides fragilis isolated from lesions of cattle with acute interdigital phlegmon (foot rot). PROCEDURES: Bacteria were cultured in strictly anaerobic conditions. Bacterial SCFA production was measured with high-performance liquid chromatography. Neutrophils were isolated, stimulated with phorbol 12-myristate 13-acetate (PMA) or opsonized zymosan (OZ), and incubated with dihydroethidium or dichlorofluorescein diacetate to measure production of O(2)and H(2)O(2), respectively. Phagocytosis was assessed after exposure to serum-opsonized bacteria. Cellular morphology was assessed with differential staining. RESULTS: All bacteria produced at least 3 of the 4 SCFAs. Production of both O(2) and H(2)O(2) was markedly curtailed in PMA-stimulated neutrophils exposed to SCFA at pH 5.5, compared with production at pH 6.7. Succinate caused a significant dose-dependent decrease in O(2) production at pH 6.7 in OZ-stimulated neutrophils. Monoprotic SCFAs elicited a significant increase in H(2)O(2) production in OZ-stimulated neutrophils at pH 6.7 but a significant decrease at pH 5.5. Monoprotic SCFAs significantly increased phagocytosis at pH 6.7 but decreased phagocytic activity at pH 5.5. Cellular necrosis was observed in cells exposed to SCFAs at pH 5.5. CONCLUSIONS AND CLINICAL RELEVANCE: Establishment and persistence of anaerobic bacteria in cattle with foot rot infection may result in part from neutrophil dysfunction secondary to the effects of bacterially secreted SCFA in acidotic microenvironments. 相似文献

The correlations between heart-girths (measured as the smallest thoracic circumference) and liveweight were determined for 96 Saanen, 100 Angora and 101 feral female goats all of New Zealand origin. The correlation coefficients were 0.94, 0.94 and 0.90 for Saanens, Angoras and ferals respectively. To facilitate liveweight estimation for calculating doses of non-mineralised anthelmintics and to minimise under-dosing it is suggested that a straight-line equivalent of the upper 95% confidence curve be used. Care should be exercised in using girth measurements in small animals where the error could be high. They are not recommended for use when mineralised drenches or potentially dangerous drugs are being administered. 相似文献

The aim of the present study was to clarify the effect of photoperiod on the secretion of growth hormone (GH) in goats. Adult female goats were kept at 20°C with an 8‐h or 16‐h photoperiod, and secretory patterns of GH for 4 h (12.00 to 16.00 hours) were compared. In addition, the goats were kept under a 16‐h photoperiod and orally administered saline (controls) or melatonin, and the effects of melatonin on the secretion of GH were examined. GH was secreted in a pulsatile manner. There were no significant differences in pulse frequency between the 8‐ and 16‐h photoperiods; however, GH pulse amplitude tended to be greater in the group with the 16‐h photoperiod (P = 0.1), and mean GH concentrations were significantly greater in the 16‐h photoperiod (P < 0.05). The GH‐releasing response to GH‐releasing hormone (GHRH) was also significantly greater for the 16‐h photoperiod (P < 0.05). There were no significant differences in GH pulse frequency between the saline‐ and melatonin‐treated groups. However, GH pulse amplitude and mean GH concentrations were significantly greater in the saline‐treated group (P < 0.05). The present results show that a long photoperiod enhances the secretion of GH, and melatonin modifies GH secretion in female goats. 相似文献

OBJECTIVE: To evaluate the effects of a standardized exercise test to exhaustion in horses on leukocyte function ex vivo. ANIMALS: 6 Thoroughbred geldings. PROCEDURES: Blood samples were obtained from each horse before exercise; at exhaustion (termed failure); and at 2, 6, 24, 48, and 72 hours after exercise to evaluate hematologic changes, rate of leukocyte apoptosis, and leukocyte production of reactive oxygen species (ROS) ex vivo. To assess leukocyte function, leukocyte ROS production in response to stimulation with lipopolysaccharide, peptidoglycan, zymosan, and phorbol myristate acetate was evaluated. Apoptosis was evaluated via assessment of caspase activity in leukocyte lysates. RESULTS: In response to lipopolysaccharide, production of ROS by leukocytes was significantly increased at 2 hours and remained increased (albeit not significantly) at 6 hours after exercise, compared with the preexercise value. In the absence of any stimulus, leukocyte ROS production was significantly increased at 6 and 24 hours after exercise. In contrast, ROS production in response to phorbol myristate acetate was significantly decreased at 6, 24, and 72 hours after exercise. Leukocyte ROS production induced by zymosan or peptidoglycan was not altered by exercise. Leukocytosis was evident for 24 hours after exercise, and neutrophilia was detected during the first 6 hours. A significant increase in the rate of leukocyte apoptosis was detected at failure and 72 hours after exercise. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicated that strenuous exercise undertaken by horses causes alterations in innate immune system functions, some of which persist for as long as 72 hours after exercise. 相似文献

South African Angora goats (Capra aegagrus) are susceptible to stress conditions, possibly due to adrenal cortex malfunction. Selection for mohair production may reduce adrenal function and decrease cortisol production. Secretion of cortisol by the adrenal cortex is essential for the induction of several gluconeogenic enzymes that enable animals to survive stressful conditions, and adrenocortical insufficiency, therefore, precipitates a vulnerability to stress. In this study, Angora goats were compared with two breeds generally accepted as hardy, Boer goats (Capra hircus) and Merino sheep (Ovis aries). Adrenal steroidogenesis was studied using subcellular fractions prepared from the adrenal glands of freshly slaughtered animals. Adrenal microsomes and mitochondria were incubated with the relevant steroid substrates, and products were analyzed and quantified with TLC, HPLC, or RIA. Subsequently, the activity of individual enzymes involved in this pathway were further investigated. The cytochrome P450 content in the preparations was also compared. The results from these studies indicated that the activity of the cytochrome P450c17 enzyme in Angora goats differed (P < .01) from that of the other species investigated. This difference may contribute to the cause of the observed hypoadrenocorticism in Angora goats. 相似文献
