Summary The occorrence of potato mop-top virus (PMTV) was assessed by two different soil sampling methods. PMTV was prevalent in southwestern Sweden but was not detected in the northern part of the country. In storage trials more symptoms developed in tubers kept at 9°C than 4 °C. irrespective of the cultivar. Cutting the tubers enhanced post storage symptom develoment in some cultivars. Of 14 cultivars grown in Sweden, those used for crisp products were more inclined to develop spraing than the others. A comparison of the severity of PMTV symptoms in field trials in Sweden. Denmark and Finland indicated substantial geographic variation in the relative susceptibility of different cultivars suggesting variation among isolates of the virus. From glasshouse experiments it was found that inoculations done at flower bud development or at flowering resulted in most spraing. Cultivar trials in pots produced a low level of spraing symptoms in general.  相似文献

Summary The frequency of PMTV-spraing and TRV-spraing increased during storage in several cultivars. The only exception was cv. Saturna, where a decrease was observed in 1992. A spraing inducing treatment of one week at 18 °C followed by one week at 8 °C immediately after an early harvest increased the frequency and provided a rough estimate of the development of spraing in untreated tubers during storage. Under most circumstances wound healing at 25 °C compared with 15 °C resulted in a lower frequency of spraing during the storage period. The date of harvest had an important influence on development of spraing during storage and on the effect of post harvest temperature treatments. Therefore, physiological processes that induce or erase spraing are strongly related to tuber maturity. The mechanisms controlling development of symptoms seemingly are identical for the two spraing inducing vira.  相似文献

The unexpected detection ofPotato mop-top virus (PMTV) in potatoes growing at several locations in the U.S.A. and Canada in 2002 has led to the realization that this soil-borne virus may be widespread throughout the potato-producing regions of both countries. A lack of information concerning the response of U.S. cultivars to PMTV infection caused us to examine the effects of PMTV on field-grown plants at three locations in the Peruvian Andes where the virus and its fungal vector,Spongospora subterranea, are endemic. All 21 U.S. cultivars tested proved susceptible to PMTV infection, especially at La Victoria where the overall incidence of both PMTV and powdery scab was high. Symptom expression in the foliage proved to be an unreliable indicator of virus infection, and thus, more reliable serological (NCM-ELISA) and nucleic acid-based techniques (NASH and RT-PCR) were used to monitor tuber tissue for the presence of PMTV. As much as 25% of field-grown tubers of cvs Kennebec, Monona, and Norland were infected with PMTV, but no correlation between virus infection and either the incidence or severity of powdery scab for any variety was apparent. Although PMTV infection did not result in the appearance of spraing or other internal necrotic reactions resembling those reported from many North American and European locations, a disproportionate proportion of infected tubers did exhibit a reticulate surface cracking. In hydroponic culture, the proportion of cracked tubers increased dramatically when the nutrient solution was seeded with virus-infestedS. subterranea. Comparatively few tubers of cvs Monona and Russet Burbank exhibited this surface cracking, suggesting that these two varieties may be somewhat tolerant to PMTV infection.  相似文献

Summary In trials with potato tubers infected with tobacco rattle virus (TRV), symptoms of spraing in cvs Bellona, King Edward, Maris Bard, Matilda, Sv 82146 and Sv 82149 increased during storage when the tubers were cut. Storage of intact tubers at a constant temperature of 9°C or at fluctuating temperatures (2 weeks at 18°C, 2 weeks at 9°C and 2 weeks at 18°C) did not increase the frequency of symptoms.  相似文献

The necrotic symptoms (spraing) in tubers caused byPotato mop-top pomovirus (PMTV) have become a severe problem for potato production in Scandinavia. PMTV is transmitted by the powdery scab pathogen (Spongospora subterranea) and no methods for control are currently available. Two breeding lines (NY99 and NY103) from the Cornell University potato breeding program, twotuberosum ×acaule lines (TA3.8.3.3 and TA3.5.3.7) from the International Potato Center (CIP), and one line (IvP35) ofSolarium phureja were tested for resistance to PMTV in field trials in Sweden in 1998 and 1999. In both years, NY99 had a low incidence of PMTV infection in tubers, and the infected tubers showed a low accumulation of PMTV, indicating that NY99 is a promising new source of resistance to PMTV for breeding programs. Furthermore, it may be suitable for cultivation in Scandinavia, which, however, needs to be evaluated in more extensive field trials.  相似文献

Summary An indirect sandwich enzyme-linked immunosorbent assay (ELISA) was used to monitor potato stems of three cultivars for the presence of the ring rot pathogen,Clavibacter michiganensis subsp.sepedonicus, during the growing season and in progeny tubers after harvest. The highest ELISA values were obtained with the highest concentration of bacteria used to inoculate seed pieces in all cultivars tested. Low ELISA values were obtained for stems and progeny tubers selected from plants grown from seed inoculated at lower bacterial concentrations. Estimates of bacterial densities in stems and progeny tubers by immunofluorescence indicated that low ELISA values were most probably caused by low bacterial numbers. It is suggested that the sensitivity of ELISA for detecting the ring rot pathogen in potato stems and progeny tubers is a function of the concentration of bacteria in individual seed pieces.  相似文献

Summary Various haulm treatments and harvest times differentially influenced the incidence of storage rots in potato tubers (Solanum tuberosum L.) in field experiments in Sweden performed over 4 years. The incidence of gangrene caused byPhoma foveata Foister (Phoma exigua var.foveata (Foister) Boerema) increased in cv. Bellona after spraying haulms with diquat as did the amount of infection caused byFusarium avenaceum (Corda ex. Fr.) Sacc. in all three cultivars (Bintje, Bellona, and Magnum Bonum). The lowestP. foveata infection levels occurred after pulling haulms. Late harvesting increased the frequency ofP. foveata in cvs Bintje and Bellona both at 2 and 4 wks interval between haulm treatment and harvest. Tuber and visible stem infection caused byP. foveata were correlated for cvs Bintje and Bellona, but not for cv. Magnum Bonum. The possible relation between tuber and stem infection is discussed.  相似文献

Summary A reverse trancription-polymerase chain reaction (RT-PCR) assay was devised and shown to be sensitive and reliable for the detection of potato mop-top virus (PMTV) RNA sequences in the flesh of virus infected potato tubers, and in the roots and leaves of soil-bait plants. This assay was compared with an enzyme-linked immunosorbent assay incorporating PMTV specific monoclonal antibodies (TAS-ELISA). The tests were devised to improve the efficiency of detection of viruliferousSpongospora subterranea in agricultural soils, and PMTV in potato tubers. RT-PCR detected PMTV RNA sequences in the roots and leaves of bait plants after three weeks growth in viruliferous soil, three weeks before the bait plants themselves developed symptoms, and two weeks before the virus was detected by TAS-ELISA. Both RT-PCR and TAS-ELISA detected PMTV in the tubers of primary-infected potatoes. RT-PCR and TAS-ELISA were shown to be more sensitive and reliable than conventional baittests and sap inoculation methods for the detection and diagnosis of PMTV.  相似文献

Summary Reducing sugar and sucrose contents of potato tubers (Solanum tuberosum L.) from six cultivars were monitored both during the growing season and after a four months storage period at 10°C. Significant correlations were found between tuber sugar contents (sucrose, reducing and total) measured at harvest and the reducing sugar content after storage. Similarly, in five of the six cultivars, the sucrose loss and the corresponding reducing sugar gain during storage were significantly correlated.  相似文献

Summary Tubers of the potato cultivars Record, Wilja, Pentland Dell and Brodick (formerly clone 137371) were sampled before and after storage at either 4°C or 10°C. Reducing sugar content stayed constant during storage at 10°C in all four cultivars but rose greatly during the first 6–12 weeks of storage at 4°C in Record, Wilja and Pentland Dell but not in Brodick. Amylolytic activity was determined after 5 weeks storage using blockedp-nitrophenyl maltoheptaoside as substrate for α-amylase,p-nitrophenyl maltopentaoside as substrate for β-amylase, andp-nitrophenylglucopyranoside as substrate for α-glucosidase. The values obtained from tubers stored at 4°C were higher than those from tubers stored at 10°C, the differences being much less in Brodick than in the other three cultivars.  相似文献

Summary Optimum storage conditions to identify resistance to silver scurf among commercially grown potato cultivars were determined in a series of experiments. Inoculation of field-grown tubers with a conidial suspension ofH. solani (concentration 104 conidia per ml of water) and incubation at 15°C with 95% RH for 1 month followed by 2 months at 85% RH produced the greatest differences in disease severity between potato cultivars. Results with glasshouse-grown tubers were comparable with those from field-grown tubers, indicating that the test could be used to identify resistance in wild species of potato and to understand the inheritance of this resistance.  相似文献

Summary Infection with an M-type (particle producing) isolate of tobacco rattle virus (TRV) was detected in leaves and/or roots of some plants from 11 of 13 potato cultivars grown in soil containing viruliferous trichodorid nematodes. Virus was detected in tubers of 8 of 13 cultivars, although only two (Pentland Dell and Maris Bard) developed spraing symptoms. Six cultivars (Arran Consul, Home Guard, King Edward, Romano, Santé and Wilja) were infected with TRV without developing spraing symptoms. Plants grown from virus-containing, symptomless tubers became systemically infected with M-type TRV and produced symptomless infected daughter tubers. Virus was maintained through three generations of vegetative propagation, and the plants were sources for acquisition of the virus by trichodorid nematodes. Distribution of virus in both spraing-affected and symptomless tubers was erratic. Movement of symptomlessly infected seed tubers may be a means of dissemination of the virus and of its introduction to previously unaffected sites.  相似文献

Summary Pot and field experiments were carried out to test fertilizer treatments on the behaviour of potato tubers stores at 4°C and ≥90% relative humidity for 6 months. Weight losses (separated into water and dry matter) were enhanced after N and K fertilization and decreased by increasing P supply. Glucose and fructose contents were reduced at harvest by high N-fertilizer rates compared to no or low fertilization, but throughout storage reducing sugar accumulation was increased, sucrose reduction was decreased and ascorbic acid was increased. K fertilization affected the chemical composition of the tubers somewhat similar to that of nitrogen fertilization. High initial nitrate content of the tubers seemed to be increased and low nitrate values were decreased during storage. The tubers from pot experiments showed similar changes but of greater magnitude.  相似文献

Summary In 1983 and 1984 there were reports in Norway of more severe late blight infection than usual on tubers of cv. Pimpernel, a cultivar without R-genes known to be somewhat resistant to both haulm and tuber infection. Results from routine tests confirmed earlier experience that cv. Pimpernel shows stable resistance to tuber late blight, although the ranking of cultivars may vary between years. In experiments during three harvest seasons tubers of different cultivars were inoculated with isolates ofP. infestans obtained from stocks of cv. Pimpernel with severe tuber late blight and with other isolates. In these experiments the isolates from cv. Pimpernel caused more rot on tubers of Pimpernel than did other isolates.  相似文献

Summary The effects of soil-borneRhizoctonia solani on yield and quality of potato were studied by an extensive individual plant sampling procedure. From 1983 to 1986, stem canker and stolon pruning were examined in 10 768 plants growing on a sandy soil. Tuber yield and quality and haulm yield per plant were also recorded. The degree of attack byR. solani depended on the cropping frequency of potato. Severe and very severe stem and stolon attacks decreased fresh yield, dry matter yield and dry matter content of tubers and increased the number of deformed and small tubers, whereas the effect on haulm yield and stem number was small. Potato cultivars differed in susceptibility toR. solani, but yield response did not differ significantly between cultivars at the same levels of stem and stolon infection.  相似文献

Summary Total sugar, reducing sugar, glucose, fructose, and sucrose of scabbed (Streptomyces scabies) and scab-free harvested tubers of several potato cultivars were analysed in four portions (pell and tube flesh separtely, from basal and apical halves), and also in small whole growing tubers. The peel of scabby harvested tubes had higher reducing sugar (glucose and fructose) contents than that of corresponding scab-free tubers. The sugar contents in the flesh and in small growing tubers did not show significant difference beween the scabbed and scab-free state. Tubers of scab-susceptible cultivars had more scab and a higher reducing sugar content than those of resistant cultivars. The results indicate that the contents of reducing sugar (glucose and fructose) in tuber peel are positively correlated with scab severity.  相似文献

Summary In measuring fructose 1,6-bisphosphatase (EC 3.1.3.11; FBPase) in potato tubers, we used anti PFP antibodies to ensure that the assay was specific for FBPase and that PPi:fructose 6-phosphate 1-phosphotransferase (EC 2.7.1.90: PFP) did not contribute to the production of fructose 6-phosphate (Fru-6-P). The involvement of cytosolic FBPase in cold sweetening was determined by monitoring enzyme activity during storage ofSolanum tuberosum cv. Erntestolz tubers at 2 or 8°C. In contrast to the rapid increase in sucrose and reducing sugars, the specific activity of FBPase showed no change and PFP rose slightly during storage at 2°C. Sugar concentrations and enzyme activities remained virtually unchanged during storage at 8°C. These data allow the interconversion of fructose 1.6-bisphosphate (Fru-1,6-P2) to Fru-6-P and vice versa by FBPase and PFP, respectively, in potato tubers, but do not support a causal relationship between coarse metabolic control of FBPase and cold-induced sweetening.  相似文献

The effect of foliar symptomatic infection by Potato mop-top virus (PMTV) on yield of tubers, spraing and infection in daughter tubers, and foliar symptom development and tuber infection in the following generations of propagation was investigated in commercial seed potato crops in Scotland. Six crops covering cvs Atlantic, Hermes, Nicola and Cara were studied between 2000 and 2006 by labelling paired replicates of plants with foliar symptoms and plants with no symptoms. Tubers from plants with no symptoms rarely produced plants with foliar symptoms in the following generation. Plants with no symptoms produced more infected tubers if they had been derived from plants with foliar symptoms the previous year than from plants with no symptoms. The proportion of daughter plants with foliar symptoms produced by tubers from plants with foliar symptoms in year 1 ranged from 19–41% and seemed to be associated with the severity of foliar symptoms. The detection of PMTV by ELISA in samples of leaves from plants with foliar symptoms ranged from 13% for cv. Cara to 59% for a crop of cv. Atlantic in 2004. The amounts of spraing were generally low but tended to be greater for tubers from plants with foliar symptoms than those from plants with no symptoms. These results indicate that roguing plants with foliar symptoms in seed potato crops could achieve an improvement in crop health but might be impractical when diseased plants are too prevalent.  相似文献

Summary Available information has been reviewed on tuber resistance toP. infestans, its inheritance and breeding procedures used to obtain potato cultivars superior in this character. P. infestans is worldwide the most destructive potato pathogen. Tuber resistance is an essential component of potato resistance as this pathogen is often responsible for tuber rot in storage, and infected tubers of susceptible cultivars provide an important way for its overwintering. In Europe many cultivars and advanced breeders selections have been obtained with tuber resistance toP. infestans, but in the last decades no progress is noted in the mean level of this resistance in cultivars. The expression of tuber resistance depends to a large extent on testing conditions and therefore it can be difficult to evaluate. Consequently making progress in breeding is not easy, and also cultivar assessment data from various countries sometimes differ considerably. It is concluded from published data that it should be easier to make progress in breeding potato cultivars with resistant tubers if more attention is paid to combining the various types of resistance which are already known, and if the genetic determination of durable resistance toP. infestans is better understood.
