Argulus siamensis is a major pathogen in freshwater aquaculture. The immune responses of Indian major carp, Labeo rohita to experimental infection of A. siamensis was evaluated by quantitation of immune‐relevant gene expression in head kidney and skin, and serum innate immune parameters through the course of infection. In skin of infected fish, antioxidant genes like natural killer cell enhancing factor (NKEF‐B) and superoxide dismutase (MnSOD) were significantly up‐regulated in addition to lysozyme G and β2 microglobulin (β2M). Both tumour necrosis factor α (TNFα) and toll‐like receptor 22 (TLR22) genes were significantly down‐regulated in skin during early phases of the infection. Most of the genes exhibited significant down‐regulation in head kidney; immunoglobulin (IgM) and β2M genes being the exceptions which were significantly up‐regulated at 12 h and 3 days post infection. Most of the innate immune parameters like serum complement activity and ceruloplasmin levels showed significant reduction in infected fish. The observed results are indicative of A. siamensis modulating the immune response of rohu by down‐regulation of many immune factors which may explain the susceptibility of rohu to A. siamensis infection. The interaction of this parasite with the host need to be further explored to understand its pathogenesis.  相似文献

This study evaluated the growth and immune response of gibel carp (Carassius auratus gibelio) cultured under no feed addition biofloc technology (BFT) system at different total suspended solid (TSS) concentrations (10, 300, 600, 800 and 1,000 mg/L for group BF0‐NF, BF300‐NF, BF600‐NF, BF800‐NF and BF1000‐NF) for 30 days. The results demonstrated that bioflocs contained rich nutrients, and gibel carp eaten bioflocs showed higher weight gain, specific growth and survival. Digestive enzyme activities such as pepsin and amylase increased significantly in BF300/600/800/1000‐NF groups than those in BF0‐NF group. Antioxidant response including superoxide dismutase and total antioxidant capacity in serum and skin mucus was also enhanced significantly (< .05). In addition, six immune‐related genes were examined by RT‐qPCR. Compared with BF0‐NF group, expression levels of immune genes intelectin (ITLN), dual specificity phosphatase 1 (DUSP 1), keratin 8 (KRT 8), myeloid‐specific‐peroxidase (MPO), c‐type lysozyme (c‐lys) and interleukin‐11 (IL‐11) were up‐regulated by 78.1‐, 23.9‐, 13.8‐, 138.8‐, 401.8‐ and 91.1‐fold, respectively. The highest expression values were observed at TSS of 600–800 mg/L. This study suggested that bioflocs can be uptaken by gibel carp as a food source, and have a potential to be used as a supplemental food for aquaculture.  相似文献

To evaluate the effects of dietary nano‐selenium (Nano‐Se) on antioxidant capacity and hypoxia tolerance of grass carp fed with high‐fat diet, experimental fishes were fed Nano‐Se supplemented diets at doses of 0 (Control), 0.3, 0.6, 0.9 and 1.2 mg/kg for 10 weeks. After feeding trial, a part of the fishes were exposed to hypoxia stress. Results showed that the survival ratio of grass carp significantly increased in 0.6 and 0.9 mg/kg Nano‐Se group, and the content of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) significantly decreased in 0.6–1.2 mg/kg Nano‐Se groups compared with the control group. In addition, dietary Nano‐Se significantly enhanced glutathione peroxidase (GPX) activity and reduced the malondialdehyde (MDA) content in fishes fed diets with 0.3 and 0.6 mg/kg Nano‐Se. Dietary Nano‐Se significantly elevated mRNA expression of GPX1 and catalase (CAT) by promoting the mRNA expression of NF‐E2‐related nuclear factor 2 (Nrf2) in the hepatopancreas. After hypoxia stress, the GPX and superoxide dismutase (SOD) activities were significantly enhanced, and the MDA content and mortality rate consequently decreased in fishes fed diets with 0.3 and 0.6 mg/kg Nano‐Se. In summary, these results suggested that optimal Nano‐Se in diet enhanced the antioxidant capacity and hypoxia tolerance of grass carp.  相似文献

This study determined the effect of different lipid sources on growth, feed use, lipid metabolism and antioxidant status of grass carp (Ctenopharyngodon idellus). Juvenile fish (56.9 ± 4.7 g) were divided into four triplicate groups and fed diets containing 30 g/Kg of fish oil (FO), olive oil (OO), peanut oil (PO) and linseed oil (LO), respectively, for 60 days. Weight gain and feed conversion ratio were not significantly different between the dietary groups, but we observed changes in the fatty acid composition of muscle and intraperitoneal fat reflecting the fatty acid profile of the dietary lipid source. In the hepatopancreas, the highest mRNA level of fatty acid translocase CD36 (FAT/CD36) and carnitine palmitoyl transferase (CPT‐1A) was both observed in the FO group. In muscle, the expression of FAT/CD36 and CPT‐1A in the LO group was significantly higher than that in other groups, except for CPT‐1A in the PO group. In addition, the lowest and highest content of malondialdehyde in serum was observed in OO and FO groups, respectively. In summary, dietary lipid source altered the fatty acid composition, potential uptake (FAT/CD36) and oxidation (CPT‐1A) of fatty acids, and antioxidant status of grass carp, which should be considered when selecting a lipid source.  相似文献

This study was conducted to investigate graded levels of dietary zinc on the growth, flesh quality, and the relationship between flesh quality and muscle antioxidant status in young grass carp (Ctenopharyngodon idella Val.). Per cent weight gain (PWG), special growth rate (SGR), feed intake (FI), feed conversion ratio (FCR), anti‐hydroxy radical (AHR), superoxide dismutase (SOD), catalase (CAT), glutathione reducase (GR) activities and glutathione (GSH) content were significantly increased with increasing levels of Zn up to a point, and thereafter declined (P < 0.05). Serum zinc, alkaline phosphatase (AKP), muscle anti‐superoxide anion (ASA), glutathione peroxidase (GPx), glutathione‐S‐transferase (GST) activities and collagen content were significantly enhanced with dietary zinc levels up to a point (P < 0.05), beyond which it plateaued. Cooking loss, shear force and malondialdehyde (MDA) were significantly reduced with increasing level of zinc up to a point, and thereafter increased (P < 0.05). The pH value significantly increased with the increasing zinc levels, whereas the trend of protein carbonyl content was opposite. Flesh quality was positively related to the antioxidant enzymes activities in muscle of young grass carp. These results indicated that optimum zinc could improve growth, and improve flesh quality partly through improving muscle antioxidant status of young grass carp.  相似文献

This study was designed to investigate effects of dietary Chlorella meal (CM) additive on growth performance, immune responses and appetite regulation of juvenile crucian carp Carassius auratus (initial body weight: 1.27 ± 0.03 g). Four experimental diets were formulated to contain 0% (CM0), 1% (CM1), 2% (CM2) and 4% Chlorella meal (CM4), respectively. Each diet was randomly assigned to triplicate groups with 40 juvenile fish per fibreglass tank for 8 weeks. Weight gain rate, specific growth rate and feed intake increased with increasing dietary CM levels. In contrast, FCR (feed conversion rate) declined with dietary CM levels. No significant differences were observed in moisture, crude protein, crude lipid and ash contents of muscle and liver tissues. Dietary CM addition increased activities of acid and alkaline phosphatase in liver and kidney. Dietary CM up‐regulated the mRNA expression levels of NKEF‐B, MCHII and IgM in kidney, and increased the mRNA levels of NPY and agouti gene‐related protein in the brain, but down‐regulated mRNA levels of MC4R, LEP, LEPR, CART1, CART2 and CCK8 genes. Based on these observations above, this study indicated that dietary CM additive increased growth performance, immune responses and appetite of crucian carp. The results, for the first time, demonstrate a role for the central nervous system in the control of food intake in fish fed dietary Chlorella meal.  相似文献

The present study investigated the effects of dietary astaxanthin on the growth, blood biochemical, antioxidant, immune and inflammatory response in lipopolysaccharide‐challenged Channa argus. A total of 0, 50, 100 and 200 mg/kg of astaxanthin were added to the basal die for 56 days. After the feeding experiment, each group was subjected to a lipopolysaccharide challenge (except for the Control group). The results showed that adding astaxanthin to the diet can significantly increase the weight gain and specific growth rate and decrease the feed conversion ratio of C. argus; the highest weight gain, specific growth rate and minimum feed conversion ratio occurred in the 100 mg/kg group. Furthermore, dietary astaxanthin supplementation can alleviate the negative effects of lipopolysaccharides by increasing the levels of alkaline phosphatase, lysozyme, complement 3, complement 4, total serum protein, albumin, globulin, superoxide dismutase, catalase and glutathione peroxidase and decrease the serum cortisol, aspartate aminotransferase, alanine aminotransferase, glutathione peroxidase, and malondialdehyde. Dietary astaxanthin supplementation also can decrease the relative expression of inflammatory genes (nuclear factor κB, interleukin‐1, interleukin‐8 and tumour necrosis factor‐α) in the liver, spleen, kidney and intestine. To summarise, dietary astaxanthin addition can improve the growth performance and attenuate the negative effects of lipopolysaccharide challenge in C. argus. The optimal amount of astaxanthin is 100 mg/kg.  相似文献

The present research was designed to investigate the growth promoting and immunostimulating properties of Moringa oleferia leaf meal (MLM) in grass carp. Juvenile grass carp (22.03 g ± 1.164) were fed with diets supplemented with 0, 50, 100 and 150 g/kg MLM for 48 days. At the end of feeding trial, skin mucus was used for analysis of lysozyme, protease, antiprotease and peroxidase activity. Head kidney was used for expression analysis of tumour necrosis factor‐alpha, interleukin‐8 and interferon‐γ. The obtained results showed that fish fed with 100 and 150 g/kg MLM had significant increase in weight gain and specific growth rate (p < .05). However, condition factor was not altered. The MLM (50 and 100 g/kg) inclusion resulted in higher mucus lysozyme and protease activity (p < .05), while peroxidase activity increased only in fish fed with 100 g/kg MLM and antiprotease activity was not altered. Expression of tnf‐α increased in a dose‐dependent manner, and significant (p < .05) increase was recorded in fish fed with 150 g/kg MLM. The expression of il‐8 and ifn‐γ increased in fish fed with 50 and 150 g/kg MLM; however, the increase was not significant (p > .05). In conclusion, supplementing juvenile grass carp feed with MLM up to 150 g/kg has growth promoting and immunostimulating effects.  相似文献

The effects of taurine supplementation on the growth and intestinal immune function in young grass carp (Ctenopharyngodon idella) were investigated in this study. A total of 540 fish (initial average weights of 255.74 ± 0.65 g) were fed one fishmeal diet and five all‐plant protein source‐based diets with graded levels of taurine (0 to 1.98g/kg diet) for 60 days, and then challenged with Aeromonas hydrophila for 14 days. First, the results showed that the taurine supplementation improved growth (PWG, TGC, FI and FE), enteritis resistance, intestinal antimicrobial compounds (LZ, ACP, C3, C4, IgM, hepcidin, LEAP‐2A, LEAP‐2B, β‐defensin‐1 and MUC2) and attenuated intestinal inflammation in young grass carp under the all‐plant protein source‐based diet. Second, the taurine supplementation attenuated intestinal inflammation partially referring to nuclear factor kappa B (NF‐κB) and target of rapamycin (TOR) signallings. Finally, under the all‐plant protein source‐based diet, the comparable level of taurine supplementation based on growth and ability against enteritis relative to fishmeal diet was established as 0.50 g/kg diet, and the optimal levels of taurine supplementation based on thermal‐unit growth coefficient (TGC), ability against enteritis and acid phosphatase (ACP) activity were established as 0.97, 1.08 and 1.21 g/kg diet, respectively.  相似文献

A feeding trial was conducted to evaluate the results of replacing soybean meal (SBM) with other blend plant protein (BP) (rapeseed meal, cottonseed meal and peanut meal) sources on growth, fish body composition, biochemical parameters, non‐specific immune index and gene expression of growth hormone/insulin‐like growth factor‐1 in Yellow River carp Cyprinus carpio. The results showed that the 600 g/kg replacement with BP in diet did not affect the growth performance of Yellow River carp, but the 800 g/kg SBM replacement diet could depress the growth of fish (p < .05). The 800 g/kg SBM replacement diet significantly impacted the superoxide dismutase, malondialdehyde contents, lysozyme, alanine aminotransferase and aspartate aminotransferase activities (p < .05). Fish fed 800 g/kg SBM replacement diet showed lowest insulin‐like growth factor 1 and highest growth hormone level than that of other diets (p < .05). Both gene expression levels of GH and IGF‐I in hepatic showed significant difference among dietary treatments (p < .05), and the lowest GH and IGF‐I mRNA level in liver were found in fish fed 800 g/kg SBM replacement diet (p < .05). In conclusion, no more than 600 g/kg SBM could be replaced by BP in diet of Yellow River carp without adverse effects on the growth performance. However, 800 g/kg SBM replacement with BP in diet inhibited the growth performance, affected some blood parameters and immune response index, and down‐regulated GH and IGF‐1 gene expression of liver in Yellow River carp. Further, GH level in serum was negatively correlated with GH mRNA level in liver; meanwhile, serum concentrations of IGF‐I were positively correlated with hepatic IGF‐I mRNA expressions.  相似文献

To investigate the effect of dietary folic acid on fish flesh quality, muscle antioxidant status and the potential mechanism, young grass carp (Ctenopharyngodon idella) were fed diets containing 0.10–3.12 mg/kg diet of folic acid for 8 weeks. The results showed that optimum folic acid increased muscle contents of protein, lipid, pH, shear force, amino acids, unsaturated fatty acids, hydroxyproline and glutathione, whereas contents of moisture, cooking loss, lactate and saturated fatty acids and cathepsin L activity showed an opposite trend (p < .05). Moreover, optimum folic acid elevated antioxidant enzyme activities and mRNA levels, as well as NF‐E2‐related factor 2 and casein kinase 2 mRNA levels (p < .05). However, optimum folic acid decreased reactive oxygen species (ROS) content and Kelch‐like ECH‐associated protein 1a (Keap1a) and Keap1b mRNA levels (p < .05). Interestingly, excess folic acid induced negative effects on above‐mentioned parameters (p < .05). Summarily, this study indicated that optimum folic acid improved fish flesh quality and muscle antioxidant system associated with Nrf2‐Keap1 pathway. Based on muscle cathepsin L activity and ROS content, the folic acid requirements for young grass carp were 1.87 and 1.80 mg/kg diet, respectively, regarding to the flesh quality and antioxidant status.  相似文献

A 3‐hr experiment was conducted to investigate the effects of dietary myo‐inositol (MI) supplementation on survival, immune response and antioxidant abilities in Litopenaeus vannamei under acute hypoxia stress. Six practical diets were formulated with supplementation of graded levels (control group 0, 0.1, 0.2, 0.4, 0.8 and 1.6 g/kg dry diet) of MI and were randomly assigned to triplicate groups of L. vannamei (mean weight 0.40 ± 0.00 g) for 8 weeks. Ten healthy shrimp (final mean weight approximately 11–14 g) randomly selected from each tank were exposed to hypoxia stress after feeding trial. After 3‐hr acute hypoxia stress, survival of shrimp fed MI‐supplemented diets (except 0.1 and 0.4 g/kg diets) was significantly increased compared with the control group. Shrimp fed control diet had lower activities of alkaline phosphatase (AKP), acid phosphatase (ACP), total antioxidant capacity (T‐AOC) and glutathione peroxidase (GPX), and higher malondialdehyde (MDA) and protein carbonyl (PC) contents in hepatopancreas than those fed the MI‐supplemented diets. In addition, mRNA expression levels of heat shock protein 70 (Hsp70), catalase (CAT) and penaeidin were significantly differentially regulated in hepatopancreas. In summary, dietary MI supplementation may have a positive effect on improving resistance to acute hypoxia stress of L. vannamei.  相似文献

Grass carp reovirus (GCRV) is the primary cause of grass carp haemorrhagic disease. The major catechin in green tea, (–)‐epigallocatechin‐3‐gallate (EGCG), has been found to have anti‐GCRV activity in the C. idellus kidney cell line (CIK). The aim of this study was to test the potential application of EGCG as an anti‐GCRV agent in aquaculture. Here, we demonstrate that various concentrations (99%, 50% and 35%) of EGCG could inhibit GCRV infectivity. EGCG (50%) + GCRV treatment significantly reduced the number of dead fish at 1‐, 2‐, 3‐, 4 ‐and 5‐day post‐challenge compared with the negative control (GCRV challenge without EGCG treatment). The safety of EGCG compound products on cell survival was studied using four fish cell lines; we did not detect a significant change in cell viability within 24 hours of EGCG incubation. We also evaluated toxicity and concentrations of malondialdehyde (MDA), glutathione (GSH) and lysozyme (LZM) in the grass carp, and the results showed that even a high dose of EGCG did not induce toxicity. Following EGCG compound injection, the concentration of MDA decreased and the concentration of GSH and LZM increased compared with the control groups. We also detected EGCG concentration in grass carp plasma and kidney using HPLC with electrochemical detection after intraperitoneal injection at a dose of 150 mg/kg. The concentration of EGCG in the plasma and kidney reached the highest levels (20 μg/ml and 1.5 μg/ml) about 12 hr after injection and then decreased. Overall, EGCG is a safe, effective product that could inhibit GCRV infection and improve immunoactivity in aquaculture.  相似文献

The present study was conducted to evaluate the effects of dietary Eucommia ulmoides (EU) on growth, feed utilization, antioxidant activity and immune responses of turbot fed a basal diet (CON) or EU‐supplemented diets with 5.0 g/kg (EU1), 10.0 g/kg (EU2) and 20.0 g/kg (EU3) EU leaf powder. After 70‐day trial, EU supplementation did not affect nutrient utilization, but reduced feed intake (FI) and specific growth rate (SGR) of fish at doses above 5.0 g/kg. Superoxide dismutase (SOD), catalase (CAT) and total antioxidant capacity (T‐AOC) activities in the EU‐supplemented groups were significantly higher than those in the control group at a dose of 20.0 g/kg. Significantly reduced malondialdehyde (MDA) contents were observed in EU‐supplemented groups at doses over 5.0 g/kg. Furthermore, fish fed 20.0 g/kg EU showed the highest lysozyme (LZM) activity among groups. The EU‐supplemented diets with doses above 5.0 g/kg significantly enhanced the mRNA expressions of cytokines. The expression level of major histocompatibility complex II alpha (MHC IIα) was significantly upregulated compared to that of the control fish when the supplemental level was at 20.0 g/kg. Taken together, the present study indicated that the EU could remarkably enhance the antioxidant activity, non‐specific immunity and maintain an active immune response in turbot.  相似文献

Sodium butyrate is one of the most popular feed additives in animal husbandry. In recent years, sodium butyrate has been increasingly used as supplement in aquaculture. The present study is to investigate the intestinal mRNA and microRNA response to diet with sodium butyrate in grass carp (Ctenopharyngodon idella), an important aquaculture species in China. mRNA and microRNA profiles of intestine of grass carp fed with diet contained 0, 1.0, 2.5, 5.0, 7.5 and 10.0 g/kg sodium butyrate were obtained by RNA‐seq using Illumina Hiseq 2,500 platform. The feeding trial was performed using 18 individuals of 1‐year‐old grass carp (n = 3 for each group) and lasted for 40 days in tanks in laboratory. A total of 349,860,852 sequence reads were generated from six intestinal libraries. Functional analysis of differentially expressed genes showed that genes participated in immune pathways tend to be activated by sodium butyrate supplementation. A total of 700 microRNAs were obtained, including 275 conserved microRNAs and 425 novel microRNAs which are potentially involved in regulating 14,300 genes. Spearman's correlation analysis identified 18 pairs of microRNA‐mRNA associated with immune pathways (p < .01 and R<?0.5). The potential genes targeted by microRNAs include CXCL12, AKT1S1, Cab39 and MHCII which are important genes associated with intestinal immune pathways. To our knowledge, this is the first integrated profiling of both mRNA and microRNA in intestine with supplementation of sodium butyrate in grass carp. The present results suggest that sodium butyrate affects intestinal immune system by regulating microRNA‐mRNA interaction in fish.  相似文献

This study investigated the effect of xylanase supplementation of a plant protein‐enriched diet on the growth performance and intestinal immune function of grass carp grow‐out (Ctenopharyngodon idella). Five hundred forty healthy grass carp grow‐out (232.74 ± 0.12 g) were fed diets containing graded levels of xylanase for 60 days. The fish were then challenged with Aeromonas hydrophila for 14 days. The results indicated that xylanase supplementation (a) improved fish growth performance and optimized the intestinal microflora; (b) increased lysozyme and acid phosphatase activities, complement and immunoglobulin M levels and antibacterial peptide hepcidin mRNA levels, thereby enhancing fish intestinal immune function; and (c) attenuated intestinal inflammation associated with the inhibition of the NF‐κB signalling pathway and activation of the TOR signalling pathway, resulting in the down‐regulation of pro‐inflammatory cytokines [except IL‐1β and IL‐12p40 in the distal intestine (DI) and IL‐6 in the proximal intestine (PI)] and up‐regulation of anti‐inflammatory cytokines [except IL‐4/13B and TGF‐β1 in the mid intestine (MI) and DI]. Finally, on the premise of a basal diet containing 45.8 g/kg araboxylans (AXs), the appropriate supplementation levels based on the ability to improve per cent weight gain (PWG) and prevent enteritis were estimated to be 1,527 and 1,608 U/kg, respectively.  相似文献

The effects of astaxanthin supplementationon the growth, innate immunity and antioxidant defence system of loach (Paramisgurnus dabryanus) were investigated in this study. A total of 450 fish (initial average weights of 3.00 ± 0.10 g) were fed five diets with graded levels of astaxanthin (0.00, 50.00, 100.00, 150.00 and 200.00 mg/kg) for 56 days. The results showed that astaxanthin supplementation significantly stimulated the growth (FBW, WG, FER, HSI), innate immunity (AKP activity in the hepatopancreas, intestinal tract, muscle and skin and LZM activity in the hepatopancreas, intestinal tract and skin), antioxidant ability (T‐SOD, GSH‐PX and CAT activities and T‐AOC, GSH and MDA contents in the hepatopancreas, intestinal tract, muscle and skin) of loach. Moreover, dietary astaxanthin supplementation significantly up‐regulated the relative mRNA levels of Nrf2 and Maf, and down‐regulated the relative mRNA level of Keap1 in the hepatopancreas when the supplementation levels of astaxanthin were 50–200 mg/kg. In conclusion, the diets with 100–151.06 mg/kg astaxanthin supplementation were optimal for loach, which based on the growth, immunity and antioxidant‐related indicators, and the astaxanthin supplementation regulated the antioxidant ability partially referring to Keap1‐Nrf2 signallings.  相似文献

An 8‐week feeding trial was conducted to investigate the effect of dietary selenium (Se) on feed intake, weight gain and antioxidant activity in juvenile grass carp (11.2 ± 0.03 g). Six Se levels (0.13, 0.41, 0.56, 1.12, 2.18 and 4.31 mg/kg) of semi‐purified diets were assayed in triplicate. The maximum weight gain, specific growth rate and feed intake were obtained in fish fed with 1.12 mg Se/kg diet. Hepatic glutathione peroxidase activity was markedly increased when dietary Se ≤1.12 mg/kg diet and reached a plateau when dietary Se ≥1.12 mg/kg diet. Hepatic superoxide dismutase and serum catalase activities in juvenile grass carp fed with 0.56, 1.12 and 2.18 mg Se/kg diets were all significantly higher than those in the other groups. The malondialdehyde content in liver and serum was firstly decreased and then increased with increasing dietary Se content, and the lowest content was observed in fish fed with 1.12 mg Se/kg diet. With the increase in Se level, the activities of serum alanine aminotransferase and aspartate aminotransferase were reduced. In addition, serum alkaline phosphatase activity and albumin content were highest in fish fed with 1.12 mg Se/kg diet. This study indicated that both the Se deficiency and excess of Se caused negative effect on the oxidative stress in juvenile grass carp and suggested that the health‐giving concentration of dietary inorganic Se was 1.12 mg/kg diet. Moreover, based on the broken‐line regression analysis of weight gain, the optimal concentration of dietary inorganic Se was 0.83 mg/kg for juvenile grass carp.  相似文献
