The microalgae Isochrysis galbana and Chaetoceros muelleri were cultured with seaweed extract (SWE) from six different seaweeds and each SWE was used either as a supplement or as an alternative media to conventional f/2 medium. Microalgae cultured with only the f/2 media acted as the control treatment. I. galbana and C. muelleri were then fed to Penaeus indicus larvae, either singly or in combination, for 8 days to evaluate its nutritional quality. Shrimp larvae fed on microalgae cultured in f/2 media supplemented with SWE from Ulva lactuca or Enteromorpha intestinalis moulted faster (P < 0.05) to the second mysis stage compared with the control. Survival, total length and growth rates were better in shrimp larvae fed on microalgae previously cultured in media supplemented with SWE. In general, shrimp larvae fed microalgae cultured entirely in SWE performed equally well compared with the control. Shrimp larvae fed on mixed microalgae (I. galbana + C. muelleri) showed significantly better growth, survival and development compared with larvae on either single microalgal diet. It was concluded that microalgae cultured with SWE could be used as a low‐cost alternative media in producing live food for the hatchery production of shrimp larvae.  相似文献

The pathways by which pathogens invade Fenneropenaeus indicus, the potential colonization in various tissues and the disease transmission mechanisms are unclear. The aims of the present study were to visualize the colonization and pathogenesis of GFP‐tagged Vibrio parahaemolyticus, in various tissues of F. indicus to evaluate the pathogen interaction. Among the three strains isolated, a virulent strain VpDAHV2 was tagged with green fluorescent protein (GFP‐VpDAHV2) and validated for both its growth characteristics and its virulence as a genuine model for F. indicus infection. VpDAHV2 was positive for toxR and tlh genes and negative for tdh genes. CLSM images revealed that maximum colonization was observed in the haemolymph of the F. indicus challenged with GFP‐VpDAHV2. The haemolymph was the primary site for the colonization of GFP‐VpDAHV2 in F. indicus. The enteric localization occurred independently of the flagellum or motility of GFP‐VpDAHV2 through the intestinal route. The F. indicus infection model suggests that the haemolymph and the intestine represent the sites of infection by GFP‐VpDAHV2, and hence are the active sites of pathogen interactions. GFP tagging of V. parahaemolyticus is a new and systemic approach to determine the presence of bacteria in vivo for the confirmation of host pathogen interactions in aquaculture studies.  相似文献

Using the 70:30 replacement method and chromium as an inert marker, the digestibility of four fermented oilseed meals/cakes (soybean meal (FSBM), groundnut oil cake (FGNC), rapeseed meal (FRSM), and sunflower oil cake (FSFC)) were determined in Penaeus monodon and Penaeus indicus. Apparent dry matter digestibility (ADMD) of fermented ingredients was ranked as FSBM > FGNC > FRSM > FSFC. The critical variations in apparent protein digestibility (APD) in P. monodon (0.816%) and P. indicus (0.608%) were lower than the ADMD. Appaprent amino acid digestibility (AAD) was >90% in FSBM for both the species and was lower for other ingredients. Protein had a higher digestibility than total amino acids and was in the range of 0.69%–2.71% in P. monodon and 0.32%–2.75% in P. indicus. A correlation between the ADC of total amino acids and protein was found to be r = 0.8229 in P. indicus and r = 0.7447 in P. monodon. Data were further subjected to two way analysis of variance for assessing the digestibility variations between the species. It was observed that P. indicus had higher values of ADMD than P. monodon in FSBM (2.97%) and FRSM (1.22%) and the reverse was true in FGNC and FSFC. The APD was high in P. indicus for FSBM, FGNC and FRSM but not for FSFC. However, significant variations could be noticed in AAD between the species.  相似文献

Different levels of dietary chitosan on growth performance, survival and stress tolerance to air exposure was studied in tiger shrimp, Penaeus monodon. Shrimp (mean initial wet weight about 1.16 g) were fed with six different diets (C0, C0.05, C0.1, C0.2, C0.3 and C0.4) containing six level of chitosan (0%, 0.05%, 0.1%, 0.2%, 0.3% and 0.4% respectively) in triplicate for 60 days. Growth performance [final body wet weight (FBW); weight gain (WG); biomass gain (BG)] of shrimp fed chitosan‐containing diets were higher (< 0.05) than that of shrimp fed the basal diet, shrimp fed C0.1 diet showed the highest value of growth performance. Survival of shrimp in C0.1 and C0.2 diet groups were higher (< 0.05) than that of shrimp in C0, C0.05 and C0.4 diet groups but without statistical difference (> 0.05) in shrimp fed C0.3 diet group. Whole body and muscle lipid contents decreased with increasing dietary chitosan levels. Plasma total cholesterol and triglyceride contents of shrimp fed C0 diet was significantly higher (< 0.05) than that of shrimp fed chitosan‐containing diets. Plasma alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities of shrimp fed C0 diet were higher than those of shrimp fed chitosan‐containing diets. Digestive gland malondialdehyde (MDA) and carbonyl protein contents of shrimp fed chitosan‐containing diets were lower (< 0.05) than that of shrimp fed C0 diet. Total haemocyte count of shrimp fed C0 diet was lower (< 0.05) than that of shrimp fed chitosan‐containing diets. On the contrary, the haemolymph clotting time of shrimp fed C0 diet was higher (< 0.05) than that of shrimp fed chitosan‐containing diets. In conclusion, all results suggested that dietary intake containing 0.1% and 0.2% chitosan enhanced the growth of shrimp, whereas a higher level than 0.3% and 0.4% decreased growth of shrimp. Second‐degree polynomial regression analysis of WG and BG indicated that the optimum supplement of dietary chitosan level should be 0.19–0.21%.  相似文献

This study evaluated the genetic variance for survival at harvest of Indian white shrimp (Fenneropenaeus indicus) using a pedigreed synthetic population generated upon collecting broodstock from different fishery grounds in Egypt. The estimated heritabilities (h2 ± SE) of survival from generations G0 to G3 ranged from 0.144 ± 0.026 to 0.008 ± 0.030 using a model that included the common environmental effect (c2). Low heritability (0.014 ± 0.015) was estimated across generations with the inclusion of common environmental effect (c2). However, the h2 of survival from G0 to G1 ranged from 0.569 ± 0.071 to 0.410 ± 0.011 when c2 was not included. The genetic correlation for survival between different ponds from G0 to G3 was moderate to high (0.79 ± 0.042 to 0.83 ± 0.012) but significantly different (P < 0.05). There were low correlation coefficients (?0.237 ± 0.089 to 0.316 ± 0.073) between body weight estimated breeding value (EBV) and survival EBV of families in four generations.  相似文献

Pathology in penaeid shrimps relies on histology, which is subjective, time‐consuming and difficult to grade in a reproducible manner. Automated image analysis is faster, objective and suitable for routine screening; however, it requires standardized protocols. The first critical step is proper fixation of the target tissue. Bell & Lightner's (A Handbook of Normal Penaeid Shrimp Histology, 1988, The World Aquaculture Society, Baton Rouge) fixation protocol, widely used for routine histology of paraffin sections, is not optimized for image analysis, and no protocol for frozen sections is described in the available literature. Therefore, the aim of this study was to optimize fixation of the hepatopancreas (HP) from whiteleg shrimp (Penaeus vannamei) for both paraffin and frozen sections using a semiquantitative scoring system. For paraffin sections, four injection volumes and three injection methods were compared, for frozen sections, four freezing methods and four fixation methods. For paraffin sections, optimal fixation was achieved by increasing threefold the fixative volume recommended by Bell and Lightner, from 10% to 30% of the shrimp body weight, combined with single injection into the HP. Optimal fixation for frozen sections was achieved by freezing the cephalothorax with liquid nitrogen, followed by fixation of the section with 60% isopropanol. These optimized methods enable the future use of image analysis and improve classical histology.  相似文献

White spot disease (WSD) caused by white spot syndrome virus (WSSV) creates severe epizootics in shrimp aquaculture industry worldwide. Despite several efforts, no such permanent remedy was yet developed. Selective breeding using DNA markers would be a cost‐effective strategy for long‐term solution of this problem. In the present investigation, out of 30 random primers, only one primer produced a statistically significant (< 0.01) randomly amplified polymorphic DNA (RAPD) marker of 502 bp, which provided a good discrimination between disease resistant and disease susceptible populations of Penaeus monodon from three geographical locations along the East coast of India. Because RAPD markers are dominant, a sequence characterized amplified region (SCAR) marker was developed by cloning and sequencing of 502 bp RAPD fragment, which generates a single 457 bp DNA fragment after PCR amplification only in the disease resistant shrimps. Challenge experiment was also conducted to validate this 457 bp SCAR marker, and the results suggested that the WSSV loads were 2.25 × 103 fold higher in disease susceptible than that in disease resistant shrimps using real‐time PCR. Therefore, this 457 bp DNA SCAR marker will be very valuable towards the development of disease‐free shrimp aquaculture industry.  相似文献

Under normal farming conditions, shrimp can experience starvation periods attributable to disease outbreaks or adverse environmental conditions. Starvation leads to significant morphological changes in the hepatopancreas (HP), being the main organ for absorption and storage of nutrients. In the literature, limited research has described the effect on the HP of periods of starvation followed by refeeding and none in whiteleg shrimp (Penaeus vannamei) using computer‐assisted image analysis (CAIA). This study describes the effect of starvation and starvation followed by refeeding on the HP of whiteleg shrimp using CAIA. Visiopharm® software was used to quantify the following morphological parameters, measured as ratio to the total tissue area (TLA): total lumen area (TLA:TTA), haemocytic infiltration area in the intertubular spaces (HIA:TTA), B‐cell vacuole area (VBA:TTA), lipid droplet area within R cells (LDA:TTA) and F‐cell area (FCA:TTA). Significant changes were measured for HIA:TTA and LDA:TTA during starvation (increase in HIA:TTA associated with decrease in LDA:TTA) and starvation followed by refeeding (decrease in HIA:TTA associated with increase in LDA:TTA). In the future, HIA:TTA and LDA:TTA have the potential to be used in a pre‐emptive manner to monitor the health of the HP, facilitate early diagnosis of diseases and study the pathophysiology of the organ.  相似文献

Commercial de‐fatted groundnut oil cake (GNC) fermented with the fungus Aspergillus niger was evaluated as a fishmeal alternative in the diet of Penaeus vannamei. A 45‐day growth trail was performed using nine iso‐nitrogenous and iso‐lipidic diets. Untreated/fermented GNC was included at the rate of 0 (control), 25, 50, 75 and 100 g/kg by replacing fishmeal (w/w). Each diet was randomly assigned to triplicate group of 20 shrimps (initial weight of 3.09 ± 0.03 g). Results revealed that shrimp fed with diets having untreated GNC up to 50 g/kg has no significant difference in growth, whereas the inclusion level was enhanced to 100 g/kg with fermented GNC with no deleterious effect. The negative linear trend was found for SGR with increasing the inclusion of both untreated and fermented GNC. The feed and protein efficiency measures, viz. feed conversion ratio, protein efficiency ratio and apparent protein utilization, were better in shrimps fed with diets having fermented GNC than those fed the respective level of untreated GNC. The broken‐line analysis indicated that the inclusion of 72.5 g/kg fermented GNC showed the best FCR. No significant difference was observed in survival (86.67%–96.67%) between the dietary treatments. There was a significant difference in ether extract of shrimp between the treatments, whereas other parameters were not affected. Haemolymph indices showed a significant difference in total protein, glucose, cholesterol and triglycerides between control and test diets. The results conclude fermented GNC, which can be better than untreated one in the diet of shrimp.  相似文献

Dietary potassium requirement of juvenile grass shrimp Penaeus monodon   总被引:2,自引:0,他引:2  
ABSTRACT: A feeding trial was conducted to estimate the minimal dietary potassium (K) requirement for juvenile grass shrimp Penaeus monodon . Purified diets with seven levels (0, 0.3, 0.6, 0.9, 1.2, 1.5, 2.0 g/100 g) of supplemental potassium were fed to P. monodon (mean initial weight 0.75 ± 0.01 g) for 8 weeks. Each diet was fed to three replicate groups of shrimp. The rearing water contained 360 mg/L potassium. Weight gain and protein efficiency ratio (PER) of shrimp improved as dietary potassium supplementation level increased up to 1.5 g K/100 g diet and thereafter declined significantly ( P < 0.05). Feed efficiency (FE) of shrimp also showed a similar trend. However, whole body K concentration of the shrimp kept an increasing trend up to 2.0 g K/100 g diet. Analysis of the weight gain (percent) and PER of the shrimp by polynomial regression indicate that the minimal dietary potassium requirement in growing P. monodon is approximately 1.2 g/100 g.  相似文献

Marine invertebrates respond to fluctuations in their environment throughout development and growth. Homeostatic adaptation is mediated in part through the regulation of Na+‐K+ ATPase enzymes and members of this class of enzymes are localized to the antennal gland of many crustaceans. In this study, we investigated the morphological and biochemical changes of the antennal glands in eight families of cross‐breed post‐larval shrimp subjected to hypotonic (10 ppt) and isotonic (20 ppt) conditions. The greatest changes in the antennal glands were detected in the hypotonic condition with families two and five cross‐breed shrimp exhibiting the highest tolerance to this diluted salinity. The changes detected in hypotonic tolerant shrimp included enlargement of coelomosac and labyrinth spaces, the presence of many dense cytoplasmic globules in podocytes as well as highly wavy apical architecture of labyrinth cells. Several alterations were also observed at the subcellular level such as widening of the intercellular spaces between podocytes, organelle destruction, and decreased basal membrane in‐folding in labyrinth cells. In addition, shrimp exposed to hypotonic conditions displayed decreased expression of both α and β subunits of Na+‐K+ ATPase and decreased enzymatic activity. The changes observed in animals upon exposure to hypotonic stress are discussed.  相似文献

Evaluation of the dietary essentiality of vitamins for Penaeus monodon   总被引:1,自引:0,他引:1  
The effect of deletion of individual water-soluble (thiamin, riboflavin, pyridoxine, cyanocobalamin, pantothenic acid, folic acid, niacin, biotin, choline, inositol and ascorbic acid) and fat-soluble vitamins (vitamins A, D, E and K) in semipurified diets on growth and survival of juvenile shrimp, Penaeus monodon , was studied in the laboratory for 8 weeks. Diets lacking riboflavin and vitamin K did not affect growth and survival of shrimp. However, deletion of inositol and choline in the diet resulted in poor growth. Maximum growth was observed in the control diet (C1) supplemented with all vitamins. Diets deficient in ascorbic acid, biotin, folic acid, niacin, thiamin and α-tocopherol resulted in poor appetite and poorer feed conversion efficiency. All treatments except the control (C1) resulted in histopathological changes in the digestive gland cells. Detachment or destruction of the epithelial cells was observed in all treatments lacking individual vitamins but more severely in the treatment without a vitamin supplement, followed by inositol, choline and ascorbic acid deficient diets.  相似文献

The aim of this study was to estimate genetic parameters for spawning traits and growth traits in a breeding line of Pacific white shrimp, Penaeus (Litopenaeus) vannamei, selected for growth and survival. Traits studied were number of eggs (NE) and number of nauplii (NN) and female body weight at insemination (FWI) and body weight at 130 days of age (BW130). Genetic parameters were estimated using a multivariate animal model. Heritability for NE and NN were estimated as 0.13 ± 0.04 and 0.03 ± 0.04 respectively. The contribution to NN total variation due to ‘factors associated with male’ effect was estimated as 0.47 ± 0.07. In the cases of FWI and BW130, heritability was estimated as 0.44 ± 0.08 and 0.19 ± 0.03 respectively. Genetic correlation between FWI and NE was estimated as 0.49 ± 0.15, between FWI and NN as 0.54 ± 0.39 and between NE and NN as 0.27 ± 0.41, whereas the genetic correlations of FWI, NE and NN with BW130 were 0.30 ± 0.13, ?0.21 ± 0.19 and ?0.25 ± 0.38 respectively. Although it is important to perform more studies on this issue, our results found no evidence of a genetic antagonistic effect between female reproductive traits and body weight at harvesting (130 days of age) in P. vannamei.  相似文献

To evaluate effect of substrate integration in biofloc based system, a 52‐day growth experiment was conducted using black tiger shrimp, Penaeus monodon juveniles (3.32 ± 0.07 g). The factorial design consisted of floc, F (with or without) as first factor and substrate (bamboo mat, B; nylon mesh, N; and without substrate) as second factor. This resulted six treatments; F + B, F + N, F, B, N and a control without biofloc and substrate. Shrimps were stocked at 110 nos. m–3 in Fibre Reinforced Plastic (FRP) tanks and, rice flour was used as carbon source in biofloc based treatments. Incorporation of nylon mesh and bamboo mat in biofloc system trapped 31.3%–38.6% and 8.5%–13.5% total suspended solids respectively and reduced bottom solid deposition. Among the substrate based groups, significantly better development of biofilm with higher microbial population noticed in F + B compared with nylon mesh. Similarly, significantly higher final growth (p < 0.01) was recorded in F + B system followed by F + N while no significant difference in body weight recorded among floc, F or substrate based groups (B, N). Biofloc and substrate integration (F + B and F + N) resulted significantly (p < 0.01) lower feed conversion ratio compared to control and floc. Incoporation of bamboo substrate in biofloc, (F + B) improved shrimp immune responses through higher hemocyte counts and prophenoloxidase activity compared to other treatments. The study revealed that integration of substrate in the biofloc system improved growth performance, FCR and immune parameters in shrimp by trapping the suspended biofloc particles, better water quality parameters, enhanced biofilm growth and provision of quality natural food.  相似文献

The effect of ambient salinity on the haemolymph variables of Fenneropenaeus indicus and its susceptibility to Vibrio harveyi infection under salinity stress has been studied. Adult shrimps were acclimated to 5‰ (hypo osmotic), 25‰ (iso osmotic) and 35‰ (hyper osmotic) salinity levels and the animals were injected with a mid logarithmic culture of V. harveyi at sub lethal level and haemolymph parameters were analysed. Haemolymph proteins, intracellular superoxide anion production, phenoloxidase (PO), alkaline phosphatase (ALP) and acid phosphatase (ACP) activity were found to be at elevated level both at 5‰ and 35‰ post challenge. The haematological responses showed a progressive increase (P < 0.05) up to post challenge day 5 (PCD 5) followed by a considerable decline at all salinities with the lowest being at 35‰. The alterations in the variables were higher in shrimps held at 5‰. However, the V. harveyi infection was severe in animals held at 35‰. The reduction in the parameters could be correlated with the decrease in survival rate of shrimps at 35‰ with a concurrent increase in V. harveyi at this salinity. Multiple regression analysis revealed that ACP (P < 0.001), haemocyte protein HCP (P < 0.001) and PO (P < 0.05) could explain 91% variability in the shrimp survival. These parameters may be used as effective shrimp health indicators. It is evident from the study that ambient salinity alters the haemolymph variables, modulates the virulence in V. harveyi and makes the shrimps more vulnerable to infection at higher salinity. The virulence of V. harveyi is increased at 35‰ salinity as being evidenced from the high mortality at this salinity. The study emphasizes the importance of salinity as an important environmental factor both in terms of host susceptibility and virulence of the pathogen.  相似文献

The anaesthetic effects of eugenol on Penaeus monodon were investigated at the different eugenol concentrations (60, 110, 160 and 210 mg/L), water temperature (21, 26 and 31°C), air exposure time (3, 6, 9 and 12 min) and body weight (2.62 ± 0.27, 6.34 ± 0.36 and 11.43 ± 0.33 g). The anaesthesia and recovery time were recorded. The results showed that the anaesthesia time of the shrimp decreased with the increase in the eugenol concentration and water temperature, and the recovery time increased with the increase of the eugenol concentration and the decrease of water temperature. Under the same eugenol concentrations, the recovery time increased with the increase of air exposure time and body weight. Under the eugenol concentration range of 60–210 mg/L, the recovered rate was 100%. The results indicated that eugenol is a safe and efficient anaesthetic for P. monodon.  相似文献

The valine requirement of juvenile tiger shrimp, Penaeus monodon Fabricius, was determined. Shrimp postlarvae, PL20, with a mean weight of 14 mg, were randomly distributed in 36 oval 40-L capacity fibreglass tanks at 10 shrimp per tank in a flow-through seawater system and reared for 8 weeks. Postlarvae were fed amino acid test diets containing 400 g kg?1 protein with casein and gelatine as intact sources of protein. Crystalline L-amino acids were supplemented to simulate the amino acid profile of the shrimp muscle except valine. Valine was added in graded levels to obtain 7, 10, 13, 16, 19 and 22 g kg?1 of the diet or 18, 25, 33, 40, 48 and 55 g kg?1 of dietary protein. At termination of the feeding experiment, growth and survival were determined and nutritional deficiency signs noted. The relationship between weight gain and dietary valine level was analysed by the broken-line regression method to derive the valine requirement. The dietary valine requirement of Penaeus monodon postlarvae was found to be 13.5 g kg?1 of the diet or 34 g kg?1 of dietary protein. This value was lower than the level found in the shrimp tissue.  相似文献
