This study investigated the effects of dietary exogenous protease on the growth performance, intestinal health, immune parameters and disease resistance of genetically improved farmed tilapia (GIFT, Oreochromis niloticus). Five test diets with commercial protease at the levels of 0, 1.38, 2.76, 5.52 and 11.04 U/g (named PE0, PE1, PE2, PE5 and PE11, respectively) were administered to triplicate tanks with 30 fish for 60 days, and then, the fish were challenged with Streptococcus agalactiae for 14 days. The results indicated that weight gain increased as exogenous protease increased from 0 to 5.52 U protease/g diet and then decreased significantly (p < .05) with a further increase in exogenous protease supplementation (p < .05). The height of the villi in the proximal intestine and distal intestine, the width of the villi in three segments of the intestine, and the thickness of the muscle layer in the proximal intestine and mid‐intestine (p < .05) were increased in the fish fed the PE5 diet. Immune and antioxidant indices (except malondialdehyde), and survival after challenged with S. agalactiae were higher in fish fed PE5 diets than in those fed other diets (p < .05). In conclusion, 5.52 U/g protease supplementation in a plant‐based diet could promote the growth performance, intestinal physical barrier function, innate immunity and S. agalactiae resistance of GIFT.  相似文献

An attempt was made to assess the effect of dietary pyridoxine supplementation on growth performance and cellular metabolic enzymes of Chanos chanos fingerlings under endosulfan induced stress. Two‐hundred and twenty‐five fish were randomly distributed into five treatments, each with three replicates and fed four isocaloric and isonitrogenous diets with graded levels of pyridoxine for 45 days. Endosulfan concentration in the water was maintained at sublethal level of 1/40th of 96 h LC50 (0.52 ppb). Dietary pyridoxine supplementation significantly enhanced growth performance viz. weight gain (%), specific growth rate, feed conversion ratio and protein efficiency ratio of fish compared to control diet fed group. The results of this experiment revealed that the activities of digestive and metabolic enzymes were significantly improved after feeding with dietary pyridoxine supplementation in C. chanos. Significantly increased muscle and brain ascorbic acid content was observed in fish fed diets containing pyridoxine. Histological studies on liver of fish fed pyridoxine‐supplemented diet showed the normal architecture of hepatocytes compared to non‐pyridoxine fed group. The results obtained in this study revealed that the inclusion of pyridoxine in diet has a protective effect against endosulfan induced stress in C. chanos fingerlings through metabolic enzyme activities and vitamin C content. This study has a potential in ameliorating the toxic effect of endosulfan induced stress using dietary supplementation of pyridoxine.  相似文献

A 20‐week feeding trial was conducted to measure growth, nutrient utilization and faecal/gut bacterial counts in triplicate groups of red hybrid tilapia, Oreochromis sp., when fed diets supplemented with 0.5% organic acids blend (OAB), 1.0% OAB, 0.5% oxytetracycline (OTC) or a control diet (no additives). At the end of the feeding trial, tilapia were challenged with Streptococcus agalactiae for 22 days. Fish fed the OTC diet had significantly higher (P < 0.05) growth than the control treatment, while growth between fish fed the OTC or OAB diets was not significantly different (P > 0.05). Phosphorus, dry matter and ash digestibility were significantly higher in the 1.0% OAB diet than the control diet. Fish fed the OAB diets had significantly lower colony‐forming units of adherent gut bacteria compared to the control or OTC treatments while those fed the 1.0% OAB diet had the lowest total faecal bacterial counts. Tilapia fed the 0.5% OTC or OAB diet had significantly higher resistance to S. agalactiae than those fed the control diet. This study indicates that dietary organic acids can potentially replace OTC as a growth promoter and antimicrobial in tilapia feeds.  相似文献

The present investigation aimed to evaluate the effect of dietary chitosan supplementation on growth performance, body composition, immune response and histopathology of Nile tilapia, and also the in vitro antibacterial activity of chitosan against Streptococcus agalactiae (S. agalactiae). About 180 fish (average body weight 39.3 ± 0.3 g) were randomly divided into three groups according to chitosan supplementation: control group (basal diet without chitosan), Ch3 group (3 g chitosan/kg diet) and Ch5 group (5 g chitosan/kg diet). Growth performance parameters and body proximate composition were measured before infection but biochemical parameters and lysozyme and antibacterial activities before and after experimental infection. Results of the present investigation showed dietary chitosan (5 g chitosan/kg diet) significantly (p < .05) improved growth performance parameters, body composition (dry matter, crude protein, ether extract, ash, and carbohydrate) and serum biochemistry (total protein, albumin, globulin, with no effect on AST, ALT, urea and creatinine) before infection in Ch5 group than the control. After infection, liver enzymes (serum AST and ALT) were maintained lower in fish fed Ch3 or Ch5 than the control. Serum lysozyme and bactericidal activities significantly increased (p < .05) in chitosan groups before and after the challenge. The mortality rate was markedly reduced in the Ch3 group and prohibited in the Ch5 group after the experimental infection. In conclusion, feeding 3 or 5 g chitosan/kg diet increased the growth rate and improved FCR of Nile tilapia. In addition, it reduced mortality by its antibacterial and immunostimulant effects.  相似文献

The effects of enzyme supplementation on growth, body proximate, and fatty acid composition of great sturgeon Huso huso fingerlings were investigated. Multienzyme (Kemin?) was added to diet in different concentrations (0, 250, 500, 750, and 1,000 mg/kg). Fingerlings were randomly stocked in tanks and fed with the experimental diets three times a day at 5–10% body weight for 46 days. Fingerlings fed diets containing 250 mg/kg exogenous enzyme exhibited higher weight gain and specific growth rate. This level of enzyme in the diet significantly improved feed conversion ratio. Body protein and moisture decreased slightly when fish fed with enzyme-supplemented diets while fat increased. Fingerlings fed with 500 mg/kg enzyme in diet had higher contents of n-3 essential fatty acids and lower n-6/n-3 fatty acid ratio. Optimum growth performance in great sturgeon fingerlings can be obtained by 250 mg/kg multienzyme in the diet.  相似文献

This study evaluated the effects of the probiotic Lactobacillus plantarum as dietary supplement on growth performance, haemato‐immunological responses, microbiology, histology and transmission electron microscopy of the intestinal epithelium of Nile tilapia challenged with Streptococcus agalactiae. Fish were distributed into two groups: control (unsupplemented) group and the group fed L. plantarum supplemented diet for a period of 58 days. We observed an increase in the concentration of lactic acid bacteria and a reduction in the number of Vibrionaceae in supplemented fish. A significant increase in the final weight, specific growth rate and feed efficiency was also observed in supplemented fish. After challenge, the number of thrombocytes and neutrophils also increased in supplemented animals. Transmission electron microscopy showed damage to the intestinal mucosa and the presence of bacteria similar to S. agalactiae in both infected groups. L. plantarum colonized the intestines of fish, enhanced the growth performance and modulated some haematological parameters.  相似文献

The effect of aqueous extract of Cratoxylum formosum on innate immune response and disease resistance in tilapia Oreochromis niloticus was investigated. The fish were fed diets containing 0% (control), 0.5% (diet 1), 1% (diet 2), and 1.5% (diet 3) of C. formosum aqueous extract for 30 days. At the end of the experimental period, parameters of innate immune response including phagocytosis of blood leukocytes, lysozyme activity in plasma, and respiratory bust activity were examined. Feeding the fish with diet 2 and diet 3 for 20 days enhanced phagocytic activity and for 30 days stimulated lysozyme and respiratory burst activities. Diet 1 increased the phagocytic activity at 30 days, but did not affect the other measured parameters. All parameters were not significantly changed (P > 0.05) in the control group throughout the experiment. Following 30 days of feeding, fish were infected with S. agalactiae. The cumulative mortalities of bacterial-infected tilapia that were fed diet 1, diet 2, and diet 3 were 56, 12, and 10%, respectively, compared with 85% in the control group. These results indicate that the aqueous extract of C. formosum may elevate the innate immune response and enhance disease resistance in tilapia.  相似文献

A feeding trial was conducted to study the effect of mannan oligosaccharide (MOS) supplementation on growth, body composition, haematology, biochemical parameters and histology of Asian seabass (Lates calcarifer) fingerlings (8.13 ± 0.06 g average weight). Mannan oligosaccharide was supplemented at five different concentrations viz., 0%, 0.5%, 1%, 1.5% and 2% levels in the diet (40% protein and 9% lipid) of L. calcarifer. The results of the 60 days feeding trial showed that there was a significant (P < 0.05) increase in the final body weight, weight gain (WG), WG%, average daily gain, survival, specific growth rate, daily growth coefficient, hepatosomatic index and viscerosomatic index in the fish fed diet containing 1% MOS. Whole body composition of post‐fed animals showed non‐significant differences (P > 0.05) among the various treatment groups. The analysis of haematological parameters showed that there was no significant (P > 0.05) differences among different treatments but for the haemoglobin content which was significantly (P < 0.05) higher in the fish fed 1% MOS. Biochemical parameters revealed that glucose, urea, cholesterol and triglyceride content showed significant (P < 0.05) difference between control and MOS‐supplemented group. Histological observations of post‐fed animals revealed that MOS supplementation resulted in increased absorptive surface area of the intestine and increased glycogen deposition in liver. The result of this experiment infers that MOS supplementation has got a beneficial effect in the diet of seabass and supplementation at 1% level is optimal for improving the growth.  相似文献

The objective of this study was to compare the supplemental effects of crystalline DL‐methionine (DL‐Met) and methionine hydroxy analogue calcium (MHA‐Ca) on growth performance of Pacific white shrimp. Eight isoproteinic (355.3 ± 2.0 g/kg diet) and isolipidic (70.0 ± 2.2 g/kg diet) diets were prepared as positive diet (20% fish meal), negative diet (15% fish meal) and DL‐Met, MHA‐Ca‐supplemented diets with the supplementation of 0.03%, 0.06%, 0.09% DL‐Met and 0.04%, 0.07%, 0.1% MHA‐Ca in negative diet respectively. Pacific white shrimp (0.92 ± 0.03 g) were fed one of the eight diets for 49 days. The results showed that dietary DL‐Met did not affect weight gain (WG) and feed conversion ratio (FCR) (p > 0.05), but the supplementation of 0.1% MHA‐Ca significantly increased WG, protein and lipid retention, and reduced FCR (p < 0.05) when compared to the negative group, and reached the similar levels as the positive control. The total free amino acids (TFAA) in haemolymph of MHA‐Ca groups and PC, NC group peaked at the 3rd hr after feeding, but the peaking time of DL‐Met groups was advanced to the 2nd hr. Activities of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) in haemolymph of all MHA‐Ca groups peaked at the 2nd hr, but DL‐Met groups peaked at the 1st hr after feeding. The above results indicated that the supplementation of 0.1% MHA‐Ca in a low fish meal diet could improve the growth performance and feed utilization of Pacific white shrimp, but dietary DL‐Met did not significantly affect the growth.  相似文献

Feeding experiment was conducted to determine whether fermentation and taurine supplementation of soybean meal (SBM) fed to yellowtail improve growth performance and lipid digestion. Six diets were formulated and designated as SBM, SBM supplemented with taurine (SBM + T), SBM fermented by Bacillus spp. (FSBM1), SBM fermented by Lactobacillus spp. (FSBM2), FSBM2 supplemented with taurine (FSBM2 + T), and fish meal (FM). Yellowtail fingerlings with an initial body weight of 42 g were stocked in 200‐L tanks. The fish were fed, each diet was fed twice daily in two separate tanks for 8 weeks. Results showed that lipid digestibility of the SBM diet was significantly lower than that of the FM diet, but SBM diet lipid digestibility was improved by taurine supplementation and fermentation. Lipase activity in anterior intestinal digesta, lipid contents in liver and muscle, and bile acid concentrations in gallbladder and anterior intestinal digesta were significantly lower in fish fed SBM diet than in fish fed FM diet; these parameters were not significantly different between fish fed SBM + T, FSBM2, FSBM2 + T diets and FM diet. Although taurine supplementation or fermentation improved lipid digestibility compared with FM diet, growth parameters were only improved in the fish fed FSBM2 + T diet, indicating that apart from a lack of taurine, SBM contains other factors responsible for inferior growth of yellowtail. These factors can be partially eliminated by Lactobacillus spp. fermentation.  相似文献

Streptococcus agalactiae is an important pathogen in fish, causing great losses of intensive tilapia farming. To develop a potential live attenuated vaccine, a re‐attenuated S. agalactiae (named TFJ‐ery) was developed from a natural low‐virulence S. agalactiae strain TFJ0901 through selection of resistance to erythromycin. The biological characteristics, virulence, stability and the immunization protective efficacy to tilapia of TFJ‐ery were determined. The results indicated that TFJ‐ery grew at a slower rate than TFJ0901. The capsule thickness of TFJ‐ery was significantly less (p < 0.05) than TFJ0901. When Nile tilapia were intraperitoneally (IP) injected with TFJ‐ery, the mortality of fish was decreased than that injected with TFJ0901. The RPS of fish immunized with TFJ‐ery at a dose of 5.0 × 107 CFU was 95.00%, 93.02% and 100.00% at 4, 8 and 16 weeks post‐vaccination, respectively. ELISA results showed that the vaccinated fish produced significantly higher (p < 0.05) antibody titres compared to those of control at 2 or 4 weeks post‐vaccination. Taken together, our results suggest that erythromycin could be used to attenuate S. agalactiae, and TFJ‐ery is a potent attenuated vaccine candidate to protect tilapia against S. agalactiae infections.  相似文献

The effect of organic acids (OA) and dicalcium phosphate (Pi) supplementation in the feed of Labeo rohita fingerlings was studied by formulating seven practical diets, designed as D1 with no feed additives {dicalcium phosphorus (Pi) and organic acid (OA) blend} while, D2, D3, D4 and D5 were supplemented with Pi at the graded levels of 5, 10, 15 and 20 g/kg, respectively, whereas D6 and D7 contained blend of OA at 15 and 30 g/kg, respectively. The diet supplemented with OA blend exhibited increase (p < .05) in growth of L. rohita fingerlings compared with the diet supplemented with Pi. The OA supplementation at both levels showed significant improvement in nutrients digestibility and minerals absorption in fingerlings. Moreover, in case of muscle proximate composition, crude fat (CF) and crude protein (CP) were increased (p < .05) with the inclusion of OA blend in the diet while crude ash (CA) was improved by Pi supplementation. Additionally, digestive enzyme activities were not affected (? .05) by OA blend supplementation while increased activities were observed in the fingerlings fed with Pi diet. Consequently, supplementation of OA blend in the diet improved the growth performance and nutrient status while Pi enhanced digestive enzyme activities of L. rohita fingerlings.  相似文献

A 9‐week feeding trial was conducted to estimate the dietary requirement of arginine in juvenile cobia in indoor flow‐through and aerated aquaria. Six isonitrogenous and isoenergetic practical diets were formulated to contain graded levels of arginine ranging from 1.76% to 3.75% (dry weight) at about 0.4% increments replaced by equal proportions of glycine. Survival was not significantly different among dietary treatments. Specific growth rate (SGR) and feed efficiency ratio (FER) increased with increasing dietary arginine up to the 2.96% diet (< 0.05), and thereafter declined. The whole body crude protein content was significantly affected by dietary arginine (< 0.05), while moisture, crude lipid and ash showed no significant differences among dietary treatments. The essential amino acid contents of muscle were not significantly affected by dietary arginine. The serum nitric oxide synthase activities in fish fed diets with arginine from 2.18% to 3.75% were significantly higher than activities in fish fed the diet with 1.76% arginine (< 0.05). On the basis of SGR and FER, the optimal dietary arginine requirements of juvenile cobia were estimated to be 2.85% of the diet (6.20% of dietary protein) and 2.82% of the diet (6.13% of dietary protein), respectively, using second‐order polynomial regression analysis.  相似文献

Juvenile channel catfish Ictalurus punctatus were fed casein–gelatin‐based semi‐purified test diets with graded amounts of arginine (ARG, 0.5%, 1%, 2% and 4% of dry diet) for a 6‐week period, to evaluate the effects of dietary ARG supplementation on performance indicators, levels of plasma ARG, ornithine, citrulline, glutamine and glutamate, as well as selected innate immune parameters. Incremental dietary ARG resulted in significant improvements in weight gain, feed efficiency and protein efficiency ratios, protein retention, protein deposition and arginine deposition. Fish fed the 4% ARG diet had significantly higher values for all performance indicators, although these values were not different from those of fish fed the 2% ARG diet with the exception of protein retention, protein deposition and arginine deposition. Conversely, fish fed the 0.5% ARG diet had significantly lower values for all analysed plasma amino acids. Similarly, both phagocyte superoxide anion production and neutrophil respiratory burst were significantly decreased in fish fed the 0.5% ARG diet. These results demonstrate that the indispensability of dietary ARG is not only related to optimized protein utilization for growth but also its supplementation to fish diets may significantly impact several aspects of the immune system, including phagocyte function and thus improve overall fish health.  相似文献

The current high demand and cost of fish meal (FM) necessities the evaluation of alternative plant protein ingredients in diets of farmed marine fish. A 56‐day feeding trial was performed to study the effects of replacement of FM with soy protein concentrate (SPC) in diets of Acanthopagrus schlegelii. Diets were prepared at levels of 0%, 30%, 47.5%, 65%, 82.5% and 100% SPC, respectively, replacing FM. The results indicated no significant differences (p > .05) in % weight gain (WG) and specific growth rate of fish fed S30% to S82.5% diets compared with the control diet while, further inclusion at 100% significantly depressed growth performance. SPC inclusion and phytase supplementation significantly affected the phosphorous discharge (P‐load) showing lowest value (3.83 ± 0.53 g/kg WG) in S100% compared to control (14.79 ± 0.37 g/kg WG) and in fish fed S30% diet (13.24 ± 0.89 g/kg WG) (p < .05). The results of this study showed that FM could be substituted up to 82.5% by SPC in the diet of Acanthopagrus schlegelii fingerlings (5.53 ± 0.12 g) without any adverse effects. Phytase supplementation SPC based diets could be effective in reducing the phosphorus load in the aquatic culture environment.  相似文献

Streptococcus agalactiae has become one of the most important emerging pathogens in the aquaculture industry and has resulted in large economic losses for tilapia farms in China. In this study, three pairs of specific primers were designed and tested for their specificities and sensitivities in quantitative real‐time polymerase chain reactions (qPCRs) after optimization of the annealing temperature. The primer pair IGS‐s/IGS‐a, which targets the 16S‐23S rRNA intergenic spacer region, was finally chosen, having a detection limit of 8.6 copies of S. agalactiae DNA in a 20 μL reaction mixture. Bacterial tissue tropism was demonstrated by qPCR in Oreochromis niloticus 5 days post‐injection with a virulent S. agalactiae strain. Bacterial loads were detected at the highest level in brain, followed by moderately high levels in kidney, heart, spleen, intestines, and eye. Significantly lower bacterial loads were observed in muscle, gill and liver. In addition, significantly lower bacterial loads were observed in the brain of convalescent O. niloticus 14 days post‐injection with several different S. agalactiae strains. The qPCR for the detection of S. agalactiae developed in this study provides a quantitative tool for investigating bacterial tissue tropism in infected fish, as well as for monitoring bacterial colonization in convalescent fish.  相似文献

An 8‐week feeding trial was conducted to evaluate the dietary arginine requirement of juvenile hybrid sturgeon. Seven isonitrogenous and isolipidic diets were formulated to contain graded levels of dietary arginine ranging from 1.74% to 3.54% (dry weight). The results indicated that the fish fed with 1.76% arginine diet had lower specific growth rate (SGR) and feed efficiency ratio (FER) than the fish fed the 2.64% to 3.24% arginine diets (< .05), and the fish fed the 2.64% arginine diet presented the highest SGR. The fish fed with 1.76% arginine diet had lower whole‐body crude protein content than the fish fed the 2.64% or 2.93% arginine diets (< .05). Compared with the 1.76% arginine diet, 2.36% to 2.93% arginine diets significantly increased the total amino acid (TAA) concentration and total nonessential amino acid (TNEAA) of carcasses. The fish fed with 1.76% arginine diet had lower activity of nitric oxide synthase (T‐NOS) and content of nitric oxide (NO) than the fish fed the 2.05% to 2.93% arginine diets in the liver. The fish fed with 1.76%, 2.05% or 2.36% arginine diets had lower gene expression of growth hormone (GH) and insulin‐like growth factor I (IGF‐I) in the liver than the fish fed the 2.64% to 3.53% arginine diets in the liver (< .05). A broken‐line analysis between SGR against the dietary arginine levels provided estimates for the optimal dietary arginine requirement of 2.47% corresponding to 6.18% of the dietary protein on a dry weight basis.  相似文献

This study aimed to examine the use of Streptococcus agalactiae polyvalent vaccine in tilapia broodstock and the effect of maternal immunity and resistance on their offspring against S. agalactiae strain. The broodstock was injected with polyvalent vaccine of S. agalactiae at a dose of 108 CFU per fish at 2nd gonadal maturity until spawning. Challenge test was carried out on the offspring at the 5, 10, 15 and 20 days after hatching using NK1, N17O, N14G, N3M, N4M strain respectively and combination of them. We observed immunological parameters in broodstock, eggs and larvae and relative per cent survival (RPS) of larvae after challenged with pathogenic S. agalactiae. The results showed that the leukocytes, phagocytic activity, respiratory burst, lysozyme activity and antibody levels of vaccinated broodstock had higher level compared with unvaccinated broodstock. The high level of the lysozyme activity, antibody levels and recombination activating gene 1 (RAG1) were also observed in eggs and larvae from vaccinated broodstock. Larvae produced from vaccinated broodstock when challenged with variety strain of pathogenic S. agalactiae had RPS value more than 50% until 20 days after hatching. In conclusion, polyvalent vaccine of S. agalactiae administrated in the broodstock could enhance immunity in the broodstock and protect their offspring from pathogenic S. agalactiae.  相似文献

The aim of this study was to evaluate the effect of dietary supplementation with the autochthonous probiotic bacteria Bacillus cereus on growth performance, haematological parameters and survival of tambaqui (Colossoma macropomum). Tambaqui fingerlings (0.94 ± 0.02 g) were fed for 120 days the following diets: basal (without probiotic) and three supplemented diets (4.2 × 104, 3.9 × 106 and 3.3 × 108 CFU/g). Growth performance and haematological parameters were evaluated every 30 days. Thereafter, fish were challenged against Aeromonas hydrophila and survival was evaluated. Probiotic B. cereus improved weight and length gains (p < 0.05), and increased neutrophils and thrombocyte counts (p < 0.05) in tambaqui supplemented with 3.9 × 106 CFU/g diet. Challenged fish fed unsupplemented diet presented the lowest survival rate (33.4%) while fish fed diets supplemented at 4.2 × 104, 3.9 × 106 and 3.3 × 108 CFU/g B. cereus had 88.8%, 80.5% and 80.5% relative per cent survival respectively. These results demonstrated that B. cereus supplemented as probiotics to C. macropomum for 120 days improved physiological and haematological responses, leading to enhanced survival in this fish species.  相似文献
