A total of 378 adult Ixodes ricinus ticks were collected by flagging vegetation in four localities of two districts (Breclav, Znojmo) in south Moravia and examined microscopically. Borreliae were identified in Giemsa-stained midgut smears from 32 (i.e. 8.5%) ticks (9.4% females, 7.2% males); the infection rate varied between 0.0 and 11.4% in the four localities examined. Among female ticks, significantly more were found to be infected in autumn (19.7%) than in spring (5.8%). Dark-field (DF) and Giemsa-stained smears (GS) examinations were compared for their sensitivity in detecting borreliae in 128 field-collected ticks; GS method showed a little higher sensitivity (11.7% ticks were positive) than DF procedure (9.4% ticks positive). Two strains of Borrelia burgdorferi were isolated from a total of 150 adult I. ricinus ticks cultured in BSK medium. 相似文献

A statistically representative group of human population from the South Moravian region (CSSR) was examined on the presence of antibodies against the following arboviruses: Lednice, Sindbis, tick-borne encephalitis, West Nile, Tahyna and Calovo. The ascertained absence of antibodies to the Lednice virus is an evidence that at present this virus is of no epidemiological importance in the region studied. No antibodies to Sindbis, West Nile and Calovo viruses were demonstrated. 17.8 to 42.0% of antibodies to Tahyna virus were detected in all age groups examined. The total infection rate was 26.0%. Antibodies to the tick-borne encephalitis virus occurred primarily in persons over 40 years. The total infection rate was 5.5%. 相似文献

Migratory birds (swallow, Hirundo rustica; sand martin, Riparia riparia; house martin, Delichon urbica) caught in southern Moravia (Czechoslovakia) in 1984-87 were examined for arbovirus infections. Isolation experiments were carried out using blood samples of 183 birds (52 swallows, 107 sand martins, and 24 house martins). The results were negative. Serological examinations of 136 birds (36 swallows, 86 sand martins, and 14 house martins) were made by haemagglutination-inhibition test (HIT) using 6 arboviral antigens of the genera Alphavirus (Sindbis--SIN) and Flavivirus (tick-borne encephalitis--TBE, West Nile--WN) and of the family Bunyaviridae (Tahyna--TAH, Calovo, CVO, and Bhanja--BHA). Antibodies against all of the tested viruses were detected at different rates: SIN 2.9%, TBE 1.5%, WN 1.5%, TAH 4.4%, CVO 1.5%, and BHA 2.2%. The titres ranged from 1.20 to 1.80. 相似文献

Entamoeba polecki Prowazek, 1912 was recorded for the first time in Czechoslovakia in two students from Kampuchea. Uninucleated cysts of diameter 14.2-15.7 micron with nuclei diameter 3.2-4.2 micron were found-repeatedly in stool samples taken from them. The nucleus accounted in average for 24.3% of the cyst diameter. The isolation and two following subinoculations on Dobell-Leidlaw medium were achieved, more abundant growth was recorded on medium with pig serum. Sera of both students were positive in serological tests using the E. histolytica antigen. No serious clinical symptoms were observed, both patients were cured successfully by metronidazole and ornidazole. Electroimmunotransfer blots were used to characterize E. polecki as a separate species. The antigenic structure of polyxenically grown E. polecki was compared with the antigenic structure of E. histolytica (axenically grown HK-9 strain and 3 polyxenically grown strains). Different blot patterns of both species were obtained, but common fractions of 30-40 kD probably responsible for serological cross-reactions were found. 相似文献

为制备并鉴定番茄溃疡病菌(Clavibacter michiganensis subsp.michiganensis,Cmm)的单克隆抗体(McAbs),用全菌皮下免疫BALB/C小鼠,采用B细胞杂交瘤技术,经免疫、融合、间接ELISA筛选和克隆等,获得稳定分泌抗体的阳性杂交瘤细胞株,得到了抗番茄溃疡病菌的单克隆抗体。经免疫后获得3株单抗分别为1A4、1C3和1B7,经亚类鉴定分别是IgM、IgG1、IgG1;纯化腹水间接ELISA效价分别为1:3.2×106、1:8.1×105、1:3.2×106;与其他同属不同亚种无交叉反应。结果表明:3株单克隆抗体均具有较高特异性和敏感性,可作为番茄溃疡病菌的检测抗体,其中,1A4的效果最好。番茄溃疡病菌单克隆抗体的获得为进一步研发番茄溃疡病检测试剂盒奠定了基础。 相似文献

Two monoclonal antibodies (MAbs) were produced against soluble antigens from the 'Ascochyta complex' fungi. Specificity of MAbs was tested by ELISA using antigen-coated wells. MAbs secreted by the monoclonal hybridoma cell line JIM 44 recognized epitopes present in the antigen preparations from Mycosphaerella pinodes and Phoma medicaginis var. pinodella , but not those present in preparations from Ascochyta pisi . At high tissue culture supernatant concentration, MAbs produced by the monoclonal line JIM 45 recognized epitopes from all three fungi, however, on dilution of MAb the antigens from A. pisi were recognized preferentially to those from M. pinodes and P. medicaginis var. pinodella . On the basis of heat and periodate treatment of the antigens from the three fungi it can be concluded that the epitope recognized by JIM 44 is carbohydrate in nature, whereas that recognized by JIM 45 is proteinaceous in nature, carried on a glycoprotein antigen. Antigen preparations from other fungi, including other pea pathogens, non-pathogens associated with pea and other fungi closely related to the 'Ascochyta complex', were not detected with either of the two MAbs. Antigen preparations from peas could be used to differentiate healthy and infected seeds in a dot-blot assay, therefore indicating the potential of using the MAbs in the development of a diagnostic test for infection of Pisum seeds by the 'Ascochyta complex' fungi. 相似文献

测试了57种南药植物粗提物对螺旋粉虱成虫的杀虫活性.结果表明,在5 mg/mL浓度下,57种南药植物粗提物对螺旋粉虱成虫表现出较好的触杀活性.其中长春花、艾纳香、刺篱木、薄荷、广藿香、曼陀罗6种南药植物粗提物对成虫的触杀活性在60%以上,活性最好的是长春花粗提物,对成虫的触杀活性为67.3%.毒力测定结果表明:薄荷粗提物对成虫的EC50值最小,为3.308 9 mg/mL.因此薄荷对螺旋粉虱表现出较好的抑制活性,具有进一步研究开发的价值. 相似文献

The ambrosia beetle Gnathotrichus materiarius, which originally came from North America, was discovered in southern Finland in 1996. In 1997, using Norwegian drainpipe traps baited with pheromones of Gnathotrichus retusus and G. sulcatus, we collected beetles in the region where the first specimen had been caught in order to determine whether this potential pest species had become established in the area. Samples from a total of 16 traps included 79 species of beetles and 719 individuals, but no specimens of G. materiarius. The most abundant species in the samples were the ambrosia beetles Xyleborus dispar and Trypodendron lineatum. Several predators and other associates of bark beetles were also captured. Insecticides are the most commonly used tactic to control western flower thrips (WFT), Frankliniella occidentalis Pergande (Thysanoptera: Thripidae), on greenhouse cucumber. However, WFT has developed resistance to several of the insecticides presently in use. In addition, some of these insecticides adversely affect greenhouse biological control agents used to control WFT, resulting in subsequent pest resurgence. Therefore, there is a need to identify novel insecticides with unique modes of action for use in integrated pest management (IPM) programs to effectively control WFT with minimal impact on associated biological control agents. In laboratory bioassays conducted in 2001, immature and adult WFT and three associated greenhouse biological control agents: Amblyseius cucumeris Oudemans (Acarina: Phytoseiidae), Orius insidiosus Say (Hemiptera: Anthocoridae) and Encarsia formosa Gahan (Hymenoptera: Aphelinidae) were exposed to direct, direct/residual, and residual contact applications of the novel biopesticide, spinosad (Conserve 120 SC), and the industry standard for whitefly control, endosulfan (Thiodan 50 WP). In all three types of assay, spinosad was effective against immature and adult WFT life stages. It showed low toxicity to A. cucumeris, moderate toxicity to O. insidiosus and high toxicity to E formosa. Greenhouse studies involving exposure of immature and adult WFT and adult biological control agents to cucumber leaves sprayed previously with spinosad supported the laboratory data. Spinosad showed low toxicity to A. cucumeris exposed to leaves 1 day after treatment (DAT), moderate toxicity to O. insidiosus 1 and 8 DAT, and high toxicity to E. formosa up to 28 DAT. These data, along with spinosad's unique mode of action, suggest it would be a valuable reduced-risk control agent for greenhouse cucumber IPM programs. 相似文献

This paper constitutes an introduction to the forthcoming series of publications summing up the present store of knowledge on the incidence and ecology of avian haematozoa in Central Europe. The paper contains bibliography dealing with the incidence of these parasites in the region studied. The problem of the incidence of blood parasites in domestic birds is discussed in detail.
