The objective of the present study was to examine the relationship between plasma leptin levels and fat distribution in crossbred steers. Twenty-four crossbred steers (Japanese Black × Holstein) were used in this study. Six steers belonged to the growing phase (11 months of age), and 18 steers to the finishing phase (from 21 to 28 months of age). Plasma leptin levels correlated positively with body fat weight ( r = 0.807, P < 0.01), body fat percentage ( r = 0.802, P < 0.01), visceral fat weight ( r = 0.807, P < 0.01), visceral fat percentage ( r = 0.780, P < 0.01), subcutaneous fat weight ( r = 0.745, P < 0.01), subcutaneous fat percentage ( r = 0.734, P < 0.01), intermuscular fat weight ( r = 0.793, P < 0.01), intermuscular fat percentage (0.775, P < 0.01), renal fat weight ( r = 0.694, P < 0.01), renal fat percentage ( r = 0.642, P < 0.01), subcutaneous fat thickness ( r = 0.755, P < 0.01), intermuscular fat thickness ( r = 0.721, P < 0.01), beef marbling score ( r = 0.706, P < 0.01) and the crude fat content of Musculus longissimus thoracis ( r = 0.592, P < 0.01). The location of fat deposits did not affect the correlation values. However, when steers were divided into growing ( n = 6) and finishing ( n = 18) groups, there was no significant correlation between plasma leptin levels and the carcass characteristics related to fat deposition (fat weight, fat percentage, carcass traits) in either group. These results suggest that plasma leptin levels in crossbred steers positively correlate with body fat content and carcass traits when these parameters vary widely. 相似文献

A study was undertaken in five draught horses of 648±33 kg body weight to find the effects of continuously pulling loads on their cardiovascular, respiratory and metabolic responses. A cart equipped with an odometer, for measuring distance, and a hydraulic dynamometer, for measuring draught force, was used. Heart and respiration rates and rectal temperatures were recorded. Blood samples for measuring arterial and venous pH and blood gases, haemoglobin, glucose and lactic acid concentrations and the serum activity of the enzymes creatine phosphokinase (CK), lactate dehydrogenase, aspartate aminotransferase and alkaline phosphatase were taken before exercise and immediately after each journey (morning and afternoon) of the daily work.Draught exercise, with loads which generated forces of between 0.57 and 0.59 kN, at speeds of 1.60 to 2.11 m/s, for 8 h daily for five consecutive days, with resting intervals of 10 min each hour, was well tolerated. Exercise tolerance was evaluated from the recovery from the changes observed in the biochemical and physiological parameters induced by the work. The analysis of these showed that, when the horses were subjected to prolonged periods of resting, their loss of fitness for work was shown by significant increases in the serum activity of muscle-derived enzymes and in blood lactate concentrations during the first day of work. However, over the following days the horses adapted to the work, so that the decreases in serum enzyme activities and blood lactate concentrations were reduced. Since similar observations have been described for racehorses, the determination of blood lactate concentrations and the serum activities of muscle-derived enzymes, specifically CK, seem to be good indicators of fitness in draught horses. 相似文献

In the United States, horses are used for a variety of purposes including recreation, exhibition, and racing. As farm, performance, and companion animals, horses are a unique species from a zoonotic disease risk perspective, and the risks of subclinical infections spreading among horses can pose challenges. Using a nanoscale real-time PCR platform, we investigated the prevalence of 14 enteric pathogens, 11 Escherichia coli genes, and 9 respiratory pathogens in fecal samples from 97 apparently healthy horses at a multi-day horse event. In addition, sugar flotation test was performed for fecal parasites. E. coli f17 was commonly detected, prevalent in 59% of horses, followed closely by Streptococcus equi subsp. zooepidemicus (55%). Additional pathogens recognized included betacoronavirus, Campylobacter jejuni, Cryptosporidium sp., E. coli O157, equine adenovirus 1, equine rhinitis B virus, and others. The use of PCR data may overestimate the true prevalence of these pathogens but provides a sensitive overview of common pathogens present in healthy horses. Our results prompt the continued need for practical biosecurity measures at horse shows, both to protect individuals interacting with these horses and to minimize transmission among horses. 相似文献

The purpose of this study was to evaluate the influence of exercise on plasma tryptophan (TRP) and free serotonin (f5-HT), whole blood-5-HT (WB-5-HT) and f5-HT/WB-5-HT ratio in Italian Saddle horses. Six clinically healthy Italian Saddle horses were subjected to a 450 meters obstacles course. Blood samples were collected from each horse by jugular venipuncture using vacutainer tubes with K3-EDTA at rest, immediately after exercise, and after 30 min. TRP, f5-HT and WB-5-HT were analyzed by HPLC. Immediately after exercise, statistically significant increases of f5-HT (p<0.001) and WB-5-HT (p<0.001) were observed. After 30 min, f5-HT and WB-5-HT decreased compared to immediately after exercise, but were still significantly higher than rest values (p<0.01 and p<0.05, respectively). A significant linear regression between f5-HT and WB-5-HT was observed during experimental conditions. f5-HT and WB-5-HT modifications after exercise suggest an important role of peripheral serotoninergic markers in response to physical activity. The possible source of extra serotonin detected after show jumping should be clarified by further investigation. 相似文献

Measurement of renal function in horses poses a particular challenge because plasma creatinine is influenced by muscle mass which is highly developed and variable between individuals, while conventional clearance methods involve potentially daunting problems, particularly urine collection and bladder washout. This paper provides data which enable technetium-diethyleneaminopentacetic acid (Tc-DTPA) clearance to be used to calculate glomerular filtration rate (GFR)/extracellular fluid volume (ECFV) as an expression of GFR in horses, as previously validated in humans, dogs and calves. Apart from being arguably a more physiological expression of GFR than using derivatives of body weight, the use of GFR/ECFV eliminates a source of delay and error, namely measurement of the injected dose, and offers the convenience of requiring only three blood samples. Videodermoscopy is a method that enables the examination of many parameters of the skin and its structures. The aim of this study was to assess specific dermoscopic parameters in purebred Arabian horses during the summer. The study involved 21 clinically healthy purebred Arabian horses (18 mares and three stallions) that had not been used for breeding and were 1 to 25 years old. The videoderoscopic evaluation was performed on seven selected areas of the body: forehead, mane, neck, chest, flank, rump, and tail. The tests were carried out with Vidix and Olympus cellSens specialised software. Videodermoscopy revealed that the skin was pigmented in all of the bay horses, in one of the seal brown horses, and in the 10 grey horses. Only one grey horse and one chestnut horse had unpigmented skin. Hair thickness ranged from 44.82 µm (neck) to 75 µm (mane). Regarding the amount of hair in the field of view, the highest and lowest numbers of hairs were found on the neck (3,004) and mane (990), respectively. A valuable insight obtained from our research is that it is possible to use digital image analysis for precise quantitative and qualitative evaluations of the skin and its structures. 相似文献

Data of 13 body measurements have been analysed from 166 Arabian horses, 123 mares and 43 stallions, ranging from 49 to 298 months of age, belonging to Alzahraa stud, Cairo, Egypt. General linear model was used to study age and gender effects on these measures. Gender was a significant source of variation for most studied traits, but not for neck girth, cannon bone circumference of fore and hind legs, and pastern girth of fore and hind legs. Age significantly affected pastern girths of fore and hind legs and cannon bone circumference of fore legs, while there was no significant effect on the other measurements. Pearson correlations, adjusted for age effect, between measurements were estimated and ranged from 0.02 to 0.84 for mares and from ?0.05 to 0.90 for stallions. Factor analysis with promax rotation for each gender was carried out to derive fewer independent common factors. Three factors were extracted which accounted for 66% and 67% of the total variance in mares and stallions respectively. The first, second and third factors in mares tended to describe body thickness, leg thickness and general size respectively; whereas in stallions they tended to differentiate among general size, leg thickness and body thickness respectively. The three extracted factors for each gender determine the main sources of shared variability that control body conformation in Arabian horses. These factors could be considered in selection programmes to acquire highly coordinated bodies in pure Arabian horses with fewer measurements. 相似文献

Leptin gene is expressed in the mammary tissue and the expression of both leptin and its receptor changes significantly during pregnancy and lactation, with high levels during the first half of pregnancy and a decrease at delivery. The aim of this work was to investigate into leptin concentration in plasma and in milk during pregnancy and the first week after parturition in dairy cow and to analyze the correlation between leptin and beta-lactoglobulin (beta-LG) concentrations in plasma and in milk. The trial was conducted on six Holstein dairy cows, reared in the same environmental conditions and evaluated with similar body condition score, during the complete reproductive cycle from insemination to the delivery. Blood from the jugular vein and milk samples were collected at weekly intervals. Plasma leptin concentration showed a lower level (p < 0.05) at the beginning of pregnancy. Milk leptin concentration showed a higher level (p < 0.01) than plasma level from week 23 to week 29 of pregnancy. Plasma beta-LG concentrations were higher (p < 0.01) compared to plasma concentrations during the first part of pregnancy, then milk levels rise and become higher than plasma levels during the last weeks before dry period. A positive correlation (p < 0.01) was observed between leptin and beta-LG both in plasma and in milk profiles. 相似文献

The time‐dependent changes in intramuscular amino acid (AA) levels caused by exercise and by feeding a protein/AA supplement were analysed in nine horses. Horses were submitted to a total of four standardized exercise tests (SETs). Amino acid concentrations were determined prior to, immediately after, 4 and 18 h after exercise. The experiment was subdivided into two consecutive periods of 3 weeks. In each period two SETs were performed. In the second period, horses were given a protein/AA supplement within 1 h after exercise. Significant changes in mean plasma AA levels similar to previous studies were noted to be time‐dependent and to be associated with feeding the supplement. The intramuscular concentrations of the free AA in relation to pre‐exercise levels showed significant time‐dependent changes for alanine, asparagine, aspartate, citrulline, glutamine, glycine, isoleucine, leucine, methionine, serine, taurine, threonine, tyrosine and valine. Feeding the supplement significantly increased the 4 h post‐exercise intramuscular concentration of alanine, isoleucine, methionine and tyrosine. At 18 h after exercise, apart from isoleucine and methionine, levels were still increased and also those of asparagine, histidine and valine in relation to none treatment. Hence, it was concluded that AA mixtures administered orally to horses within 1 h after exercise increased intramuscular AA pool. 相似文献

The ability to show standing oestrus and to ovulate within 10 days of weaning was studied in 240 purebred Swedish Yorkshire primiparous sows, fed according to a conventional feeding regime during lactation. The sows were weighted and backfat depth was recorded at farrowing and at weaning. Oestrus control was performed daily and blood samples for determination of plasma progesterone were drawn regularly in 205 sows. The distribution among the sows of the first standing oestrus after weaning had 2 peaks. The first peak occurred within 10 days of weaning and the second 24-30 days after weaning. Twelve per cent of the sows ovulated without showing standing oestrus within 10 days of weaning and 4% had an anovulatory first oestrus within the same time. Significant differences in age at farrowing and in loss of weight and backfat during lactation were found between sows which both showed standing oestrus and ovulated within 10 days of weaning and sows which neither showed standing oestrus, nor ovulated within the same time. The season during which weaning occurred significantly influenced the ability to show standing oestrus and ovulate within 10 days of weaning. Among the sows which both showed standing oestrus and ovulated within 10 days of weaning, significant positive correlations were found between weight loss, litter size, litter weight gain and the interval from weaning to first standing oestrus. 相似文献

Plasma concentrations of adiponectin and leptin were measured in 23 mature Standardbred mares (age: 10 ± 3 years) and 12 weanling fillies (10 Quarter Horse/Belgian crossbreds and two Quarter Horses; aged: 4 ± 3 months) to test the hypothesis that adipocytokines are proportional to adiposity in horses. Rump fat thickness was measured using B-mode ultrasound and percent body fat (% fat) calculated using a published formula for the estimation of fatness in horses. Plasma adiponectin and leptin were determined using radioimmunoassay. In the absence of purified equine adiponectin or leptin, results were expressed as human equivalents (HE) of immunoreactive (ir) adipocytokines.Plasma ir-leptin HE concentration was positively correlated (r = 0.543; P < 0.001) with percent body fat and (r = 0.556; P < 0.001) to fat mass in all horses. The plasma ir-leptin HE concentration was lower (P = 0.03) in weanlings (1.90 ± 0.34 ng/mL HE) than in mature mares (3.47 ± 0.50 ng/mL HE). The ratio of ir-adiponectin HE to ir-leptin HE was negatively correlated (r = −0.621; P < 0.001) to percent fat and (r = −631; P < 0.001) to fat mass in all horses. The ratio of ir-adiponectin HE to ir-leptin HE was higher (P < 0.001) in weanlings (3.04 ± 0.51) than mature mares (1.03 ± 0.13). These data suggest that leptin is proportional while adiponectin is inversely proportional to adiposity in horses. 相似文献

Objective To identify the incidence of adverse effects caused by morphine 100–170 µg kg?1 administration during surgery in horses. Design Retrospective case record analysis (1996–2000). Animals Eighty‐four healthy (ASA 1 or 2) horses, mean age 5.5 ± 3.1 (SD) years (2 months to 16 years), mean weight 524 ± 14 kg (100–950). Methods Physiological data and evidence of complications were collected from the anaesthetic records of all animals anaesthetized with romifidine, ketamine, diazepam and halothane and undergoing laryngeal surgery or orchiectomy at the Royal (Dick) School of Veterinary Studies. Cases were divided into those receiving (group M+; n = 18) and those not receiving morphine (M?; n = 29), and the data compared. Values for heart and respiratory rate and mean arterial pressure were compared at 15‐minute intervals between 30 and 120 minutes after induction using anova for repeated measures. The incidence of intraoperative problems was compared using Fisher's exact test. Recovery scores were compared using Student's unpaired t‐test. The records of a further 37 horses undergoing umbilical herniorrhaphy (n = 5), arthroscopy (n = 29) or tarsal arthrodesis (n = 3) were also studied but not analysed statistically due to disparate treatment distribution. Results There were no significant differences between the M+ and M? groups. The incidence of post‐operative complications such as box‐walking and colic were similar in each group. Conclusions Morphine doses of 100–170 µg kg?1 do not increase the risk of problems when used to provide perioperative analgesia in horses anaesthetized with romifidine, ketamine, diazepam and halothane. Clinical relevance Morphine provides an acceptable and relatively inexpensive way to provide perioperative analgesia in horses. 相似文献

This experiment investigates the changes in the plasma leptin levels of sheep fed a diet of three energy levels (low, moderate and high). Four mature wethers were used for this experiment. For the first 4 weeks, the sheep were fed diets to provide 1.2 times the maintenance metabolizable energy requirements, low energy levels (LE). During the second 4 weeks, the sheep were fed diets to provide 1.5 times the maintenance metabolizable energy requirements, moderate energy levels (ME). During third 4‐week period, the sheep were fed a diet to provide 1.8 times the maintenance metabolizable energy requirements, high energy levels (HE). Body weight was determined every week. Blood samples were collected prior to the morning meal at 3 days intervals throughout the experiment, and plasma leptin, insulin and nonesterified fatty acid (NEFA) concentrations were assayed. Body weight decreased in week 1 after the start of the experiment, it continued to decrease during the LE feeding, but it gradually increased until the end of HE period. Similarly, plasma leptin concentration decreased during LE feeding, but increased during the HE feeding. Additionally, positive correlation was obtained between leptin and insulin or glucose concentrations, whereas no clear relationship with circulating NEFA was observed. In conclusion, it was suggested that plasma leptin concentrations were affected by the metabolizable energy in feed. 相似文献

Serum enzyme activities, albumin, protein, urea, cholesterol, triglyceride, free fatty acid, glucose and lactate concentrations as well as hematocrit values were measured in standardbred and Finnish-bred horses at rest and after (i) a short controlled exercise and (ii) a trotting competition. There were no breed differences in the enzyme activities at rest and the 2 breeds responded in the same manner to the exercise. Only after the race proper significant increases in the enzyme activities were found. The activities rose more in the standardbred horses than in the Finnish-bred horses. Urea and cholesterol concentrations did not change after either exercise. Protein and albumin concentrations as well as hematocrit values increased significantly after the exercise. At rest hematocrit values were significantly higher in the standardbred horses and the difference persisted throughout the exercise. After the race proper also albumin and protein concentrations were higher in the standard-bred than in the Finnish-bred horses. Free fatty acid and triglyceride concentrations increased significantly during the exercise. Although glucose and lactate concentrations increased in both breeds, the behaviour of these parameters differed. Glucose concentrations remained increased for a longer period and the recovery from the increased lactate level was faster in the standardbred than in the Finnish-bred horses. The observed differences suggest that the standardbred horses have higher anaerobic capacity than the Finnish-bred horses. 相似文献

OBJECTIVE: To compare the effect of anesthesia alone with anesthesia and abdominal surgery on plasma thromboxane B(2) concentrations in horses. STUDY DESIGN: Non-randomized experimental study. ANIMALS: Six male mixed-bred horses (5-12 years, 350 +/- 18 kg). METHODS: All horses were anesthetized for 2.5 hours using halothane, and a month later abdominal surgery was performed using the same anesthetic technique with a similar duration. The schedule of anesthesia included pre-medication with diazepam (0.1 mg kg(-1) IM), followed by xylazine (2.2 mg kg(-1) IV), and 10 minutes later anesthesia was induced with ketamine hydrochloride (2.2 mg kg(-1) IV). After orotracheal intubation, anesthesia was maintained with halothane. Blood samples for the determination of thromboxane B(2) (TXB(2)) were obtained before, at induction, at 60 minutes after halothane was first inspired, and at recovery from anesthesia as well as at the corresponding stages of the experimental abdominal surgery (before induction, prior to laparotomy, enterectomy, enteroanastomosis, abdominal wall closure). RESULTS: Baseline value for the anesthesia group was 76 +/- 12 pg mL(-1) and increased (p < 0.001) after 1 hour of anesthesia to 265 +/- 40 pg mL(-1). With surgery, the corresponding value was 285 +/- 21 pg mL(-1) (hour 1, p < 0.001) and 210 +/- 28 pg mL(-1) (hour 2, p < 0.001), respectively. These were not different from anesthesia alone. CONCLUSION: The increased concentrations of thromboxane B(2) between 1 and 2.5 hours of halothane anesthesia and during the corresponding stages of the surgical intervention suggested that the anesthetic technique caused a significant increase in thromboxane B(2) and that surgery did not appear to contribute to this response. 相似文献

Physical activity and stress both cause an increase in cortisol release ratio. The aim of this study was to evaluate the use of saliva samples for the determination of cortisol concentrations indicating the work‐load level in horses during race training. Twelve Purebred Arabian horses aged 3–5 years were studied during the routine training session. After the warm‐up, the horses galloped on the 800 m sand track at a speed of 12.8 m/s. Three saliva samples, and three blood samples were collected from each horse. Both types of samples were taken at rest, immediately after return from the track and after 30 min restitution. The concentrations of blood lactic acid (LA), and cortisol in saliva and plasma samples were measured and analyzed. Blood LA, plasma and salivary cortisol levels increased significantly after exercise (P < 0.05). Salivary cortisol concentration determined 30 min after the exercise correlated significantly with plasma cortisol level obtained immediately after exercise (P < 0.05) as well as measured 30 min after the end of exercise (P < 0.05). The determination of cortisol concentration in saliva samples taken from racehorses 30 min after the end of exercise can be recommended to use in field conditions to estimate the work‐load in racehorses. 相似文献

Objective To establish the prevalence of intussusceptions involving the caecum in a population of horses admitted to a university hospital for colic. Design Retrospective clinical study Methods Medical records of all horses admitted to the Massey University Veterinary Teaching Hospital between 1991 and 2004 were examined for information of those horses diagnosed with an intussusception involving the caecum. Results A total of 135 horses were admitted for colic surgery during the study period and 61 horses had a diagnosis of ileocaecal (37), caecocaecal (5) or caecocolic intussusception (19) made either at surgery or necropsy. Of the horses with ileocaecal intussusception, 32 had an incomplete hand-sewn ileocaecostomy without reduction and 29 survived to discharge. All the horses with caecocaecal intussusceptions were diagnosed preoperatively via rectal examination and/or transabdominal ultrasound: 2 were euthanased at surgery and 3 survived to discharge. In the 19 horses with caecocolic intussusceptions, manually reduction at surgery was performed in 6 and 5 of them survived to discharge. A typhlectomy was performed via a colotomy in 6 horses, 3 of which survived to discharge. Conclusions The high prevalence of intussusceptions involving the caecum seen at this referral centre may indicate a higher prevalence in New Zealand than is reported elsewhere in the world. Clinical relevance Intussusceptions involving the caecum should be considered as a differential diagnosis in horses presenting with chronic low-grade colic. Transabdominal ultrasound is useful for identifying caecocaecal and caecocolic intussusceptions. Hand-sewn side-to-side incomplete ileocaecostomy is a quick, effective and safe method of surgical treatment of ileocaecal intussusceptions. 相似文献

Feed deprivation decreases plasma leptin concentrations depending on the amount of body fat reserves. While a greater response was observed in lean than in fat humans and rats, a few results for ruminants are inconsistent. The objective of this study was to determine the influence of feed deprivation on plasma leptin concentration in growing lambs with different body fat reserves and on the relationship between leptin and fatness. In addition, we included other hormones (growth hormone, GH; insulin-like growth factor-I, IGF-I and insulin) involved in tissue development. Thirty male lambs of 40 kg live weight were used. Blood was sampled before and after a fasting period of 24 h. The lambs were slaughtered and dissected into several fat and lean tissues. Feed deprivation reduced plasma leptin by an average of 34.6% (p < 0.001). Obese lambs exhibited a greater decline of leptin than lean lambs (2.50 vs. 1.36 ng/ml, p < 0.05). The correlations between leptin and several fat tissues were lower in those lambs than that were fasted. This indicates that leptin concentrations after short-term fasting scarcely reflect the extent of body fat reserves but reflect more the actual metabolic situation. Body fat did not significantly influence the response of GH, IGF-I and insulin to fasting in most cases. 相似文献
