The dimethylformamide extracts from sewage sludges from a chosen sewage — treatment plant in highly industrialized region of Poland were investigated. Using the column chromatography with Al2O3 and silicic acid as the stationary phases, the fraction of azaarenes has been separated. GC-MS analysis of azaarenes fraction made it possible to identify 10 azaarenes and several of their alkyl derivatives, which dominate in the organic matter extracted from sewage sludges. The silicic acid column chromatography in connection with the reduction of nitroarenes and derivatization with pentafluoropropionic acid anhydride yielded an enriched nitrofraction. Nitroarenes present in this fraction as amide derivatives were also analysed by GGMS method. There was found presence of 3 nitroarenes and their methyl derivatives.  相似文献

An international collaborative study of IUPAC methods II.D.19 and II.D.25 for preparation and GLC analysis of fatty acid methyl esters was begun in 1976. The IUPAC methodology, applicable to animal and vegetable oils and fats and fatty acids from all sources, contains special instructions for preparation and analysis of methyl esters of fatty acids containing 4 or more carbon atoms (analysis of milk fat). Twenty-three collaborators participated in the analysis of 5 known mixtures, 4 vegetable oils, 1 fish oil, and 2 butterfats. Several blind duplicate samples were included. The experimental data were subjected to statistical analysis to examine intra- and interlaboratory variation. Reproducibility and accuracy data for the higher fatty acid (14:0-22:1) mixtures and fish and vegetable oils were satisfactory and were in good agreement with results from an AOCS Smalley Committee check sample program involving analysis of the same samples. Typical coefficients of variation (%) at various concentrations were 15 (2% level), 8.5 (5% level), 7 (10% level), and 3 (50% level). Low recoveries and poor reproducibility were characteristics of results obtained for butyric acid in the butterfat and related known mixtures. A coefficient of variation of about 19% was found for analysis of butyric acid in butterfat, vs coefficients of variation in the range of 4-13% for similar levels of other components in butterfat and other samples. The IUPAC methodology for GLC analysis of fats and oils other than milk fats has been adopted by the AOAC as official first action to replace the current GLC method, 28.063-28.067.  相似文献

A rapid method is described for extraction and cleanup of raw and processed milk for determination of aflatoxins M1 and M2 by using a C18 Sep-Pak/silica gel cleanup column combination. Aflatoxins are separated by normal phase liquid chromatography and their concentrations are determined by fluorescence detection in a silica gel-packed flow cell. Recoveries ranged from 99 to 103% with coefficients of variation less than 2% for M1 levels of 0.117-1.17 ng/mL added to raw milk. Similar recoveries were obtained for M2. The coefficient of variation for analysis of 5 subsamples of naturally contaminated milk was less than 1%. Agreement with the official method is satisfactory. Each sample requires less than 25 mL solvent and 10 min actual handling time. Sample chromatograms show no interferences in the M1-M2 elution region and no late-eluting peaks, which permits spacing injections at 13-20 min intervals. Aflatoxin levels as low as 0.03 ppb may be determined by this procedure. Extracts have also been analyzed by thin layer chromatography.  相似文献

A simple procedure is described for the routine determination of pentachlorophenol (PCP) in various types of toy paints. PCP can be determined in water colors, body colors, showcard paints, gouache paints, and colored inks. PCP is extracted from the sample with acetone, followed by quantitative gas chromatographic analysis of the concentrated acetone extract on stainless steel columns containing 15% Carbo-wax 20M on 60-80 mesh Chromosorb P (AW). As little as 1 ppm PCP can be determined with recoveries ranging from 50 to 100%, depending on the type of paint. The presence of PCP in samples is confirmed by thin layer chromatography, using dansyl chloride as a fluorescent labeling compound. By this method as little as 4 ppm can be determined. The results of PCP determinations in 65 commerical samples are presented.  相似文献

A simple and rapid procedure for the separation and determination of inorganic, methyl, and ethyl mercury compounds was described using liquid chromatography (LC) followed by vapor generation inductively coupled plasma-mass spectrometry (VG-ICP-MS). Well resolved chromatograms were obtained within 5 min by reversed-phase liquid chromatography with a C8 column as the stationary phase and a pH 4.7 solution containing 0.5% v/v 2-mercaptoethanol and 5% v/v methanol as the mobile phase. The separated mercury compounds were converted to mercury vapors by an in situ nebulizer/vapor generation system for their introduction into ICP. The concentrations of NaBH4 and HNO3 required for vapor generation were also optimized. The method was applied for the speciation of mercury in reference materials NIST SRM 1568a Rice Flour and NIST SRM 1567a Wheat Flour and also rice flour and wheat flour samples purchased locally. The accuracy of the procedure was verified by analyzing the certified reference material NRCC DOLT-3 Dogfish Liver for methyl mercury. Precision between sample replicates was better than 13% for all the determinations. The detection limits of the mercury compounds studied were in the range 0.003-0.006 ng Hg mL(-1) in the injected solutions, which correspond to 0.02-0.06 ng g(-1) in original flour samples. A microwave-assisted extraction procedure was adopted for the extraction of mercury compounds from rice flour, wheat flour, and fish samples using a mobile phase solution.  相似文献

A postcolumn liquid chromatographic method to determine the extractable residues of glyphosate (GLYPH) and its principal metabolite, (aminomethyl)phosphonic acid (AMPA), in various cereals and beans is described. The finely ground sample is extracted with a mixture of chloroform and water, and the resulting aqueous layer is passed through a cation exchange column. The eluate is adjusted to pH 7-10 and passed through an anion exchange column. The second column is eluted with 0.3M HCl solution and the resulting acidic eluate is analyzed with liquid chromatography coupled with postcolumn fluorescence detection. The mean recoveries for GLYPH in barley, canola, dry pea, flax, soybean, wheat, and white bean ranged from 90.0 to 98.1%, with coefficients of variation (CV) from 2.9 to 10.0% and limits of detection (LOD) from 0.07 to 0.14 ppm. Similarly, mean recoveries for AMPA in the same crops ranged from 87.4 to 98.9%, with CV from 4.6 to 7.7 and LOD from 0.05 to 0.12 ppm. Using this method, an analyst can routinely analyze 6 samples per 1.5 days. The advantages of this procedure are discussed.  相似文献

A method has been developed for the quantitative determination of linoleic acid in infant formulas by gas chromatography (GC). A known amount of triheptadecanoin was spiked into the sample. Total lipid was extracted from the product by an ethyl ether-petroleum ether-ethanol system in a Mojonnier flask. The sample was saponified by methanolic KOH after the solvents were evaporated. Methyl esters of the fatty acids were prepared by boron trifluoride (BF3) in methanol and analyzed by gas chromatography. A glass column packed with 10% SP-2340 (75% cyanopropyl silicone) was used to separate and identify the methyl linoleate and the methyl heptadecanoate. The quantity of methyl linoleate was calculated by comparing the integrated peak areas of these 2 fatty acid methyl esters. This method was satisfactory for both milk protein-based and soy protein-based matrixes. The results obtained by this method are comparable to those obtained by the AOAC spectrophotometric method 28.082-28.085.  相似文献

A sensitive method is described for determining dicamba at low micrograms/L levels in ground waters by capillary column gas chromatography with electron-capture detection (GC-EC); compound identity is confirmed by gas chromatography-mass spectrometry (GC-MS) using selected ion monitoring. Dicamba residue is hydrolyzed in KOH to form the potassium salt. The sample is then extracted with ethyl ether which is discarded. The aqueous phase is acidified to pH less than 1 and extracted twice with ethyl ether. The combined ethyl ether extracts are concentrated, and the residue is methylated using diazomethane to form the corresponding dicamba ester. The derivatized sample is cleaned up on a deactivated silica gel column. The methylated dicamba is separated on an SE-30 capillary column and quantitated by electron-capture or mass spectrometric detection. Average recoveries (X +/- SD) for ground water samples fortified with 0.40 microgram/L of dicamba are 86 +/- 5% by GC-EC and 97 +/- 7% by GC-MS detections. The EDL (estimated detection limit) for this method is 0.1 microgram dicamba/L water (ppb).  相似文献

A high performance liquid chromatographic (HPLC) method for vitamin D in pet foods and feeds at low concentrations (2-8 IU/g = 50-200 ppb) was studied collaboratively. The procedure consists of the following purification steps: saponification, extraction of the unsaponifiable fraction, chromatography on alumina, cleanup on reverse phase HPLC, and quantitation with straight phase HPLC. The original method, developed by Knapstein, was simplified by deleting the quantitative TLC step. Six coded samples were distributed to 31 laboratories, along with a known sample containing 15 IU/g to allow practice of the rather complicated procedure. Eighteen collaborators returned their results. Results for the spiked samples show good recovery. The estimates of repeatability and reproducibility are 0.96 and 2.2 IU/g for spiked samples and 1.5 and 3.1 IU/g for commercial samples, respectively, which are considered acceptable for these low concentrations. The method has been adopted official first action.  相似文献

A method based on gas chromatography with electron capture detection was developed for the determination of ethylene dibromide (EDB) extracted from flour products. The procedure relies on the organic extraction of flour/water mixtures and uses an internal standard, 1-bromo-3-chloropropane. Recoveries of EDB at 10 and 100 ppb were 80.1 +/- 2.8% (SD) and 84.4 +/- 4.3%, respectively; recovery of the internal standard at the working concentration 500 ppb was 98.3 +/- 6.7%. Calibration curves were linear over the range 5-400 ppb, with a mean overall coefficient of variation of less than 5%. The reliability of the procedure was assessed by using gas chromatography combined with mass spectrometry. Results are shown for determination of EDB in locally milled flour products.  相似文献

Two strategies have been developed for Diuron determination by FTIR spectrometry, an off-line extraction and stopped-flow determination and a fully mechanized procedure, based on the on-line extraction of Diuron and FIA-FTIR measurement of the extracts. The aforementioned procedures have been compared with a reference chromatographic method. The off-line FTIR spectra were obtained at a nominal resolution of 4 cm(-1) from 4000 to 900 cm(-1) by accumulating 25 scans. Diuron was determined using peak height measurements at 1582 cm(-1) corrected using a baseline defined between 1562 and 1614 cm(-1). The waste generation of the off-line procedure was 3.4 mL chloroform for each sample, and the method provided a LOD of 40 microg g(-1), corresponding to 0.8% (w/w) Diuron in the original sample. The fully mechanized FIA method provided a LOD of 35 microg g(-1), which corresponds to 0.7% (w/w) in the solid sample and a maximum sampling frequency of the whole procedure of 30 h(-1), with a waste generation of 9.3 mL per sample, taking into account the volume of CHCl(3) required for sample dissolution and that need as a carrier. All those methods consume less organic solvent than a HPLC method, which involves the use of 39 mL of acetonitrile per sample and a sampling frequency of 12 h(-1).  相似文献

The Onley-Yip procedure for determining ethylenethiourea (ETU) in milk and crops was modified to reduce interferences by the ethylenebisdithiocarbamates (EBDCs). A 20 g crop-methanol extract is cleaned up by adsorbing the sample onto Gas-Chrom S. desorbing ETU, and eluting ETU from aluminum oxide with chloroform containing ethanol. ETU is converted to the S-butyl derivative for gas-liquid chromatography (GLC) and flame photometric detection (sulfur mode). For liquid chromatography (LC), ETU is cleaned up on another aluminum oxide column and injected directly. LC and GLC results are confirmed by thin layer chromatography. A cooking procedure based on conversion of EBDCs to ETU is included for surveying crops for possible EBDC content. Recoveries from 8 crops and milk fortified at 0.05 ppm ETU ranged from 73 to 100%.  相似文献

A gas chromatographic assay was developed to determine major residues of morantel in bovine milk over a range that is suitable for monitoring residues of the drug. The method is based on hydrolysis of the N-methyl-tetrahydropyrimidine portion of morantel and its metabolites to N-methyl-1,3-propanediamine, and converting the diamine to an N,N-bis-(2-nitro-4-trifluoromethylphenyl) derivative. The addition of an internal standard, the N-desmethyl-N-ethyl homolog of pyrantel, to the milk sample circumvents any potential problem that could arise from variable reaction yields, and eliminates the true recovery as a factor affecting the accuracy and precision of the procedure. The concentrations of the derivatives are determined by pulsed electron capture gas chromatography over a linear dynamic range that is equivalent to 12.5-50 ppb morantel. The method was evaluated at the 0, 12.5, 25, and 50 ppb levels in fortified bovine milk, and in a withdrawal sample containing physiologically incurred morantel residues. Mean values of 14 +/- 1.7, 24 +/- 3.7, and 47 +/- 6.9 were found for the fortified samples, approximately 3 ppb for control milk, and 16 +/- 1.7 ppb for the withdrawal sample.  相似文献

A method making use of turbulent flow chromatography automated online extraction with tandem mass spectrometry (MS/MS) was developed for the analysis of 4 quinolones and 12 fluoroquinolones in honey. The manual sample preparation was limited to a simple dilution of the honey test portion in water followed by a filtration. The extract was online purified on a large particle size extraction column where the sample matrix was washed away while the analytes were retained. Subsequently, the analytes were eluted from the extraction column onto an analytical column by means of an organic solvent prior to chromatographic separation and MS detection. Validation was performed at three fortification levels (i.e., 5, 20, and 50 microg/kg) in three different honeys (acacia, multiflower, and forest) using the single-point calibration procedure by means of either a 10 or 25 microg/kg calibrant. Good recovery (85-127%, median 101%) as well as within-day (2-18%, median 6%) and between-day (2-42%, median 9%) precision values was obtained whatever the level of fortification and the analyte surveyed. Due to the complexity of the honey matrix and the large variation of the MS/MS transition reaction signals, which were honey-dependent, the limit of quantification for all compounds was arbitrarily set at the lowest fortification level considered during the validation, e.g., 5 microg/kg. This method has been successfully applied in a minisurvey of 34 honeys, showing ciprofloxacin and norfloxacin as the main (fluoro)quinolone antibiotics administered to treat bacterial diseases of bees. Turbulent flow chromatography coupled to LC-MS/MS showed a strong potential as an alternative method compared to those making use of offline sample preparation, in terms of both increasing the analysis throughput and obtaining higher reproducibility linked to automation to ensure the absence of contaminants in honey samples.  相似文献

A method based on disposable pipet extraction (DPX) sample cleanup and gas chromatography with mass spectrometric detection by selected ion monitoring (GC/MS-SIM) was established for 58 targeted pesticide residues in soybean, mung bean, adzuki bean and black bean. Samples were extracted with acetonitrile and concentrated (nitrogen gas flow) prior to being aspirated into DPX tubes. Cleanup procedure was achieved in a simple DPX-Qg tube. Matrix-matched calibrations were analyzed, and the limits of quantification (LOQ) of this method ranged from 0.01 mg kg(-1) to 0.1 mg kg(-1) for all target compounds. Coefficients of determination of the linear ranges were between 0.9919 and 0.9998. Recoveries of fortified level 0.02 mg kg(-1) on soybean, mung bean, adzuki bean and black bean were 70.2-109.6%, 69.1-119.0%, 69.1-119.8%, and 69.0-120.8%, respectively, for all studied pesticides. Moreover, pesticide risk assessment for all the detected residues in 178 market samples at Beijing market area was conducted. A maximum 0.958% of ADI (acceptable daily intake) for NESDI (national estimated daily intake) and 55.1% of ARfD (acute reference dose) for NESTI (national estimated short-term intake) indicated low diet risk of these products.  相似文献

Rice (Oryza sativa, L.) degree of milling (DM) is a quantification of the amount of bran that has been removed from kernels during the milling process. Historically it has been determined using a time-consuming continuous solvent extraction (CSE) procedure to measure the surface lipid content. The objective of this research was to develop a gas chromatography (GC) method for rice surface lipids. The GC procedure was highly correlated with several other methods for DM including a commonly used CSE procedure and it demonstrated repeatability within acceptable analytical limits. Other aspects in support of the utility of this procedure are that it requires a relatively small amount of sample, organic solvent, and technician time. Consequently, this GC method for rice surface lipids measurement is suitable for use in determining rice DM in cultivar development programs and studies that require the analysis of a large number of samples.  相似文献

Accurate quantification of quizalofop-p-ethyl is essential for it may do harm to humans and animals through both water and food. Currently, detection of quizalofop-p-ethyl mainly relies on methods such as gas chromatography, high performance liquid chromatography, and gas chromatography-mass spectrometry. Although these techniques are reliable, they are relatively expensive and time-consuming because of multistep sample cleanup. To address this, we developed a competitive indirect enzyme-linked immunosorbent assay (ciELISA) with a polyclonal antibody against quizalofop-p-ethyl that was generated in our lab. The IC(50) of detection was 0.03495 microg/mL, and the lowest detection limit reached 0.00192 microg/mL. Furthermore, the method had high specificity for it did not cross-react with other structure-related compounds. When water and soil samples that were fortified with quizalofop-p-ethyl were analyzed by this ELISA, recoveries were in the range of 89-110% from water and 81-108% from soil. Good correlations between this immunoassay and gas chromatography data were obtained for residues of quizalofop-p-ethyl in water and soil. Our data indicate that this method is a convenient analytical technique for monitoring quizalofop-p-ethyl in waters without extraction and the extra cleanup step and in soil without the cleanup step.  相似文献

A micellar electrokinetic capillary chromatography (MECC) mode was applied to a capillary electrophoresis (CE) method, which was developed for detection and quantitation of patulin in apple ciders. This method used a small sample amount (2 mL) and consumed minimal organic solvent compared to the most commonly used HPLC methods. The sample preparation procedure of the CE method was also simpler than other chromatographic techniques developed for patulin analysis. Patulin was detected with a photodiode array detector at 273 nm. The standard curve was linear (r(2) = 0.9984) from 75 microgram/L to 121 microgram/mL with patulin working solutions corresponding to 3.8 microgram/L to 6.1 microgram/mL patulin in the sample. The linearity was better in a narrower range of concentrations (r(2) = 0.9999) from 75 microgram/L to 24.1 microgram/mL. The limit of detection of the method was 3.8 microgram/L. Patulin recoveries at 4 levels in spiked samples (10-121 microgram/L) ranged from 95.2 to 105.4%. The recoveries were 96. 9% and 99.2% for 2 levels (22.3 and 223 microgram/L, respectively) of patulin in infected apple samples. This method represents a unique alternative method for rapid and sensitive analysis of patulin in apple ciders.  相似文献

A sensitive method is described for determination of nivalenol (NIV) and deoxynivalenol (DON) in cereals by using reverse phase liquid chromatography and UV detection at 222 nm. The sample is extracted with acetonitrile-water (85 + 15) and an aliquot is purified by passage through a combined column of cation exchange resin and alumina-carbon (20 + 1). Analysis at this stage is possible with some samples but the method recommends passing an aliquot through a carbon minicolumn after evaporation and solubilization in methanol. Interference from coextracted compounds at this point is negligible. Recoveries of both NIV and DON from spiked extracts taken through the full method were in the range 83-94%. The relative standard deviation, based on 5 replicate determinations from each of 2 corn samples, was approximately 5% for both NIV and DON. With a 10 microL injection, the minimum contamination (3 X signal/noise ratio) able to be detected in cereal samples was about 0.015 micrograms NIV/g and 0.05 micrograms DON/g. The cleaned up extracts are also suitable for analysis by gas chromatography.  相似文献
