Objective

The fixed-dose combination of butorphanol, azaperone and medetomidine (BAM; 30, 12 and 12 mg mL?1, respectively) with subsequent antagonism by naltrexone–atipamezole was evaluated for reversible immobilization of captive blesbok (Damaliscus pygargus phillipsi).

Study design

Prospective, clinical trial.

Animals

Sixteen blesbok (four males and twelve females), weighing 52.5?71.0 kg, were immobilized in South Africa.

Methods

The total dose of BAM ranged from 0.5 to 0.7 mL for females and 0.7 to 0.9 mL for males. In seven animals chosen randomly, 8000 units of hyaluronidase was added to the dart. Physiologic variables were recorded every 5 minutes beginning at 10?20 minutes after darting. Arterial blood samples were collected three times at 20, 30 and 40 minutes after darting for analysis of blood acid-base status.

Results

The mean administered doses of BAM were as follows: butorphanol (0.34 ± 0.08 mg kg?1), azaperone (0.14 ± 0.03 mg kg?1) and medetomidine (0.14 ± 0.03 mg kg?1). The inductions were calm and smooth. The mean induction time was 9.6 ± 3.2 minutes with just BAM and 5.1 ± 0.8 minutes with BAM and hyaluronidase combination. Heart rate (45 ± 6 beats minute?1) and respiratory frequency (38 ± 4 breaths minute?1) were stable throughout immobilization. The mean arterial blood pressure for all animals was stable but elevated (137 ± 7 mmHg). Rectal temperature slightly increased over time but remained within an acceptable range. The recovery time after administering naltrexone and atipamezole was 4.8 ± 0.7 minutes.

Conclusion and clinical relevance

The BAM combination proved to be reliable and effective in blesbok.  相似文献

Objective

The combination of butorphanol, azaperone and medetomidine (BAM) with subsequent antagonism by naltrexone–yohimbine or naltrexone–atipamezole was evaluated for reversible immobilization of captive African lions (Panthea leo).

Study design

Prospective, clinical trial.

Animals

Twenty lions, 11 males and nine females, weighing 38–284 kg were immobilized in South Africa.

Methods

The BAM volume dose rate administered was 0.005–0.008 mL kg?1 (0.6 mL 100 kg?1). Physiologic variables were recorded every 5 minutes. Four arterial blood samples were collected from all animals at 20, 30, 40 and 50 minutes after immobilization for analysis of blood-gases and acid-base status.

Results

The actual doses administered were as follows: butorphanol, 0.18 ± 0.03 mg kg?1; azaperone, 0.07 ± 0.01 mg kg?1; and medetomidine, 0.07 ± 0.01 mg kg?1. The inductions were calm and smooth, and induction time ranged from 4 to 10 minutes (7 ± 2 minutes). The amount of time needed to work with each lion was 70 minutes, and no additional drug doses were needed. Heart rate (40 ± 8 beats minute?1) and respiratory frequency (15 ± 4 breaths minute?1) were stable throughout immobilization. The mean arterial blood pressure of all animals was stable but elevated (142 ± 16 mmHg). The rectal temperature slightly increased over time but remained within acceptable range. The recovery time was significantly shorter when using naltrexone and atipamezole (9 ± 1 minutes) compared to using naltrexone and yohimbine (22 ± 7 minutes).

Conclusion and clinical relevance

The BAM combination proved to be reliable for general veterinary anaesthesia in lions. During anaesthesia, minor veterinary procedures such a blood collection, intubation, vaccination and collaring could safely be performed with no additional dosing required.  相似文献

ObjectiveTo characterize the effects of a combination protocol of dexmedetomidine–midazolam–ketamine (DMK) administered intramuscularly (IM) in ornate box turtles (Terrapene ornata ornata).Study designProspective experimental trial.AnimalsA total of 16 apparently clinically healthy adult ornate box turtles (eight male, eight female).MethodsEach turtle was treated with dexmedetomidine (0.1 mg kg−1), midazolam (1 mg kg−1) and ketamine (10 mg kg−1) administered IM. Time to first response, time to maximal effect, the plateau phase and time to recovery from reversal administration were recorded. Physiologic variables, muscle tone, reflexes and the ability to perform endotracheal intubation were recorded at 5 minute intervals. Movement in response to an IM injection of 0.1 mL sterile 0.9% NaCl administered in the left pelvic limb, using a 25 gauge needle to a depth of just past the bevel of the needle, was assessed every 15 minutes. Atipamezole (0.5 mg kg−1) IM and flumazenil (0.05 mg kg−1) SC were administered 60 minutes after the initial DMK injections.ResultsThe mean time to first response, time to maximal effect, the plateau phase and time to recovery were 2.1, 14.9, 38.7 and 7.8 minutes, respectively. A respiratory rate was not observed in most turtles. The body temperature significantly increased over time. The palpebral reflex was persistent in 43% of turtles and the tail pinch reflex remained intact in 13% of turtles. All turtles recovered with no observed adverse effects.Conclusions and clinical relevanceIn this study, this DMK protocol administered to ornate box turtles resulted in a rapid-onset, light anesthesia lasting approximately 40 minutes and a smooth recovery with no adverse effects noted.  相似文献

ObjectiveTo compare cardiovascular and ventilatory effects, immobilization quality and effects on tissue perfusion of a medetomidine–ketamine–midazolam combination with or without vatinoxan (MK-467), a peripherally acting α2-adrenoceptor antagonist.Study designRandomized, blinded, crossover study.AnimalsA group of nine healthy Patagonian maras (Dolichotis patagonum).MethodsMaras were immobilized twice with: 1) medetomidine hydrochloride (0.1 mg kg–1) + ketamine (5 mg kg–1) + midazolam (0.1 mg kg–1) (MKM) + saline or 2) MKM + vatinoxan hydrochloride (0.8 mg kg–1), administered intramuscularly. Drugs were mixed in the same syringe. At 20, 30 and 40 minutes after injection, invasive blood pressure, heart rate, respiration rate, end-tidal CO2, haemoglobin oxygen saturation, and muscle oxygenation were measured, arteriovenous oxygen content difference was calculated. Muscle tone, jaw tone, spontaneous blinking and palpebral reflex were evaluated. Times to initial effect, recumbency, initial arousal and control of the head were recorded. Paired t test, Wilcoxon matched-pairs signed rank test and analysis of variance were used to compare protocols; (p < 0.05).ResultsVatinoxan significantly reduced systolic (p = 0.0002), mean (MAP; p < 0.0001) and diastolic (p < 0.0001) arterial blood pressures between 20 and 40 minutes. MAPs at 30 minutes (mean ± standard deviation) with MKM and MKM + vatinoxan were 105 ± 12 and 71 ± 14 mmHg, respectively. Without vatinoxan, four animals were hypertensive (MAP > 120 mmHg), whereas with vatinoxan, four animals were hypotensive (MAP < 60 mmHg). Muscle and jaw tone were significantly more frequently present with MKM (both p = 0.039). Other measurements did not significantly differ between protocols.Conclusions and clinical relevanceIn Patagonian maras, vatinoxan attenuated the increase in blood pressure induced by medetomidine. Muscle and jaw tone were more frequently present with MKM, indicating that quality of immobilization with vatinoxan was more profound.  相似文献

Mycobacterium bovis recognizes as hosts a wide spectrum of animal species. In particular epidemiological situations, high prevalence of infection is found also in pigs. In the present study, we evaluated the capability of the interferon-gamma (IFN-γ) assay to identify pigs infected with M. bovis. The results of the immune-diagnosis were correlated to the findings of the post mortem inspection and the bacterial culture of lymph nodes. Blood samples of 146 pigs, belonging to a local breed of Sicily reared in free or semi-free roaming conditions, were collected to assess the specificity and the sensibility of the IFN-γ assay. Thirty-one pigs, from M. bovis free herds, did not react to the IFN-γ assay, yielding a specificity of 100%. The IFN-γ assay identified 15 out of 19 animals positive to the bacterial culture and 22 out of 26 animals with tuberculous lesions, with a sensibility of 78.9-84.6%, respectively. Out of 26 reactors to the test, 15 pigs (57.7%) confirmed to be infected after the bacterial culture and 22 (84.6%) had tuberculous lesions. The IFN-γ assay was able to reveal 4 animals with no visible lesions (NVL). Together, these findings support the feasible use of the IFN-γ assay as an intra vitam tool for the surveillance and management of M. bovis infection in swine populations.  相似文献

ObjectiveTo evaluate the effects and utility of tiletamine–zolazepam–medetomidine (TZM) and ketamine–medetomidine (KM) for anesthesia of Amur leopard cats (Prionailurus bengalensis euptailurus).Study designProspective, randomized experimental trial.AnimalsA total of six female (3.70 ± 0.49 kg) and six male (5.03 ± 0.44 kg; mean ± standard deviation) Amur leopard cats aged 2–6 years.MethodsEach animal was administered four protocols separated by ≥3 weeks. Each protocol included medetomidine (0.05 mg kg–1) combined with tiletamine–zolazepam (1 mg kg–1; protocol MTZLO); tiletamine–zolazepam (2 mg kg–1; protocol MTZHI); ketamine (2 mg kg–1; protocol MKLO); or ketamine (4 mg kg–1; MKHI) administered intramuscularly. At time 0 (onset of lateral recumbency) and 30 minutes, heart rate (HR), respiratory rate (fR), rectal temperature, noninvasive mean arterial pressure (MAP) and hemoglobin oxygen saturation (SpO2) were recorded. Times to onset of lateral recumbency, duration of anesthesia and time to standing were recorded.ResultsOverall, animals were anesthetized with all protocols within 10 minutes, anesthesia was maintained ≥57 minutes, and recovery (time from the first head lift to standing) was completed within 5 minutes. During anesthesia with all protocols, HR, fR, rectal temperature, SpO2 and MAP were 99–125 beats minute–1, 33–44 breaths minute–1, 37.6–39.4 °C, 90–95% and 152–177 mmHg, respectively. No adverse event was observed.Conclusions and clinical relevanceTZM and KM at various dosages resulted in rapid onset of anesthesia, duration of >57 minutes and rapid recovery without administration of an antagonist. Accordingly, all these combinations are useful for anesthetizing Amur leopard cats and for performing simple procedures. However, the low doses of the anesthetic agents are recommended because there was no difference in duration of anesthesia between the dose rates studied.  相似文献

Objective

To evaluate dexmedetomidine, midazolam and dexmedetomidine–midazolam for sedation and antinociception in tegus.

Study design

Prospective, crossover, randomized, blinded study.

Animals

Six healthy tegus (Salvator merianae) weighing 1.6 ± 0.3 kg.

Methods

Tegus were administered intramuscularly saline (0.5 mL; CON), dexmedetomidine (0.2 mg kg?1; DX), midazolam (1 mg kg?1; MZ) and dexmedetomidine–midazolam (same doses; DM). Heart rate (HR) and respiratory frequency (fR) were recorded before treatment (baseline) and 15, 30 minutes, 1, 2, 3, 4, 6, 8, 12 and 24 hours after the treatments. Sedation scores were recorded according to resistance to manual restraint, posture and response to noxious stimulus, at baseline and 5, 10, 15, 30 minutes, 1, 2, 3, 4, 6, 8, 12 and 24 hours after the treatments. Antinociception was evaluated by measurement of latency of limb withdrawal reflex (LWR) to thermal stimulus, recorded at baseline and 15 minutes, 1, 2, 4, 8, 12 and 24 hours after the treatments.

Results

Lower HR (DX and DM) and fR (MZ, DX and DM) than CON were measured 15 minutes after the treatment and for up to 6 hours. Sedation was mild to moderate in MZ, deep in DM and absent in DX, although animals showed behavioral changes in DX, with increase in aggressiveness. Median (interquartile range) duration of sedation were 170 (50; 235) minutes in MZ and 230 (115; 235) minutes in DM. Recovery period was prolonged in both treatments, surpassing the duration of the experiment. Higher LWR than CON was detected from 15 minutes until 12 hours in DX and DM.

Conclusions and clinical relevance

Midazolam provided sedation without antinociception, and dexmedetomidine provided antinociception without sedation. Drug combination increased the duration of sedation but not antinociception. Due to increased duration of sedation, reversal of effects with flumazenil and atipamezole should be considered after conclusion of clinical procedures.  相似文献

Objective

To evaluate three routinely used tidal volumes (VT; 10, 12 and 15 mL kg?1) for controlled mechanical ventilation (CMV) in lung-healthy anaesthetized dogs by assessing alveolar ventilation (VTalv) and dead space (DS).

Study design

Prospective, randomized clinical trial.

Animals

A total of 36 client-owned dogs.

Methods

Dogs were randomly allocated to a VT of 10 (G10), 12 (G12) or 15 (G15) mL kg?1. After induction CMV was started. End-tidal carbon dioxide tension was maintained at 4.7–5.3 kPa by changing the respiratory frequency (fR; 6 < fR < 30 breaths minute?1). After 29 minutes, cardiovascular and respiratory variables were recorded for 3 minutes using a multiparameter monitor, volumetric capnography (VCap) and a blood gas analyser. The ratios of VTalv to body weight (VTalv kg?1) and airway DS to VT (VDaw/VT), Bohr's DS (VDBohr), Enghoff's DS (VDBE) and the volume of expired carbon dioxide per breath (VTCO2,br) were calculated. Mean airway pressure (MawP), fR and peak inspiratory pressure (PIP) were recorded. Data were analysed using one-way anova and Student–Newman–Keuls tests with a statistical significance set at p < 0.05.

Results

No differences were observed for demographic data and cardiovascular variables between groups. A total of three dogs were excluded because of technical difficulties and one because of fR > 30. VTalv kg?1 (p = 0.001) increased and VDBohr (p = 0.002) decreased with greater VT. VTCO2,br (p = 0.017) increased and VDaw/VT (p = 0.006), VDBE (p = 0.008) and fR (p = 0.002) decreased between G10 and G15. PIP (p = 0.013) was significantly higher in G15 compared with that in G10 and G12. No changes were observed in MawP.

Conclusions and clinical relevance

A VT of 15 mL kg?1 is most appropriate for CMV in lung-healthy dogs (as evaluated by respiratory mechanics and VCap) and does not impair cardiovascular variables.  相似文献

The present study aimed to determine whether Cryptosporidium oocysts were present in stools from captive snakes at Funda??o Parque Zoológico (Zoological Park Foundation) in S?o Paulo, Brazil. Two collections were performed; the first in July 2008 and the second in February 2009. Fecal samples were collected from 74 enclosures that housed 101 individuals of 23 snake species. The stool specimens collected from 16 out of the 74 enclosures (21.6%) contained Cryptosporidium spp. oocysts; all of them were confirmed as Cryptosporidium serpentis, using molecular techniques. Only in three (18.7%) out of the 16 enclosures with positive samples were there animals with clinical signs compatible with infection by C. serpentis, such as regurgitation and significant progressive weight loss. From the results, it was concluded that diagnostic examinations need to be performed periodically, even on clinically healthy animals, as a preventive measure.  相似文献

1. An experiment was conducted with 360 Lohmann LSL-Classic White Leghorn layers (64 weeks old) to evaluate the effects of supplementation of microbial phytase on production, egg quality, bone, selected manure parameters and feed costs.

2. Experimental diets were formulated as follows: (1) maize–soybean (CS), (2) CS+300 units of phytase (FTU)/kg diet which was formulated to recoup only calcium and available phosphorus equivalency for phytase (CS+PHYCa+P), (3) CS+300 FTU/kg diet which was formulated to recoup total nutrient equivalency for phytase (CS+PHYtotal), (4) CS+100 g/kg distiller's dried grains with solubles (DDGS), (5) DDGS+300 FTU/kg diet which was formulated to recoup only calcium and available phosphorus equivalency for phytase (DDGS+PHYCa+P), or (6) DDGS+300 FTU/kg diet which was formulated to recoup total nutrient equivalency for phytase (DDGS+PHYtotal).

3. Each dietary treatment was assigned to 4 replicate groups with 3 cages and 5 hens per cage. The hens were provided with feed and water ad libitum. The experiment lasted for 8 weeks.

4. CS+PHYCa+P, CS+PHYtotal, DDGS+PHYCa+P and DDGS+PHYtotal diets supplemented with phytase provided similar percentage egg production, egg weight, egg mass, exterior egg quality, initial and final body weight compared with phytase-free diets.

5. However, supplementation of phytase to the experimental diets and calculation of the total nutrient equivalency for enzyme caused increased feed intake and decreased feed conversion ratio and Haugh unit.

6. No differences in manure dry matter, crude ash, total nitrogen, tibia crude ash, calcium and phosphorus contents were found among the experimental diets. On the other hand, manure total phosphorus content was significantly decreased in the DDGS diet and diets supplemented with phytase in comparison to the CS diet.

7. It was concluded that the addition of microbial phytase to the CS-based diets or diets with DDGS of hens in late lay and using Ca and available P equivalency of enzyme in feed; formulation may provide an economic benefit and decrease the amount of phytate P excretion in the manure without compromising production and egg quality parameters.  相似文献

In August 2005, 2 members of a group of 6 captive snowy owls (Bubo scandiacus) in central Saskatchewan died of West Nile virus infection. Objectives

To determine the endotracheal tube cuff pressure produced with two inflation techniques, in two brands of endotracheal tube in cats. To determine the inspiratory pressure which produces an audible leak when the intracuff pressure is 30 cmH2O.

Study design

Prospective, clinical, randomized study.

Animals

A total of 40 client-owned healthy adult cats.

Methods

Following induction of anaesthesia, endotracheal intubation was performed with a Parker Flex-Tip PFLP (Parker; n = 20) or Flexicare VentiSeal (Flexicare; n = 20) endotracheal tube. For each cat, the endotracheal tube cuff was inflated using two methods, minimum occlusive volume (MOV) and pilot balloon palpation (PBP). Intracuff pressure was recorded. Cuff pressure was then set at 30 cmH2O and the pressure within the breathing system when a manual breath first caused an audible leak was measured.

Results

PBP pressure was lower for Parker (36 ± 13 cmH2O) compared with Flexicare (45 ± 13 cmH2O, p = 0.048). MOV pressure was not different between tube types (56 ± 28 versus 66 ± 25 cmH2O for Parker and Flexicare, respectively, p = 0.247). MOV produced a higher pressure than PBP for Parker (56 ± 28 versus 36 ± 13 cmH2O, p = 0.001) and Flexicare (66 ± 25 versus 45 ± 13 cmH2O, p = 0.007). When intracuff pressure was set at 30 cmH2O, 95% of cats did not develop an audible leak until the inspiratory pressure was greater than 10 and 12 cmH2O for Parker and Flexicare tubes, respectively.

Conclusions

PBP produced lower cuff pressures than MOV, although both techniques produced a cuff pressure above that at which mucosal blood flow is believed to be restricted. A cuff pressure of 30 cmH2O may be sufficient to prevent audible leak in most cats if respiratory pressures are kept at 10–12 cmH2O or below.

Clinical relevance

To ensure a safe endotracheal tube cuff pressure, use of a specifically designed pressure gauge is recommended.  相似文献

Grain from transgenic corn line TC1507 (Herculex* I) from Pioneer Hi-Bred International Inc. and Dow AgroSciences LLC, which expresses the Cry1F protein from Bacillus thuringiensis to provide protection from lepidopteran pests of corn, was compared with its isoline equivalent and 2 conventional corn strains in a 16-wk laying hen feeding trial. Egg production and production efficiency of hens fed the diet formulated with transgenic grain TC1507 were similar to those of hens fed diets formulated with isoline or nontransgenic conventional corns. Hens fed TC1507 had similar egg qualities as those fed nontransgenic grain diets. Diet × phase interactions were noted for Haugh unit and Roche color fan score. Hens fed conventional corn 2 had a poorer Haugh unit score compared with hens fed the other 3 diets. Conventional corn 1 had greater levels of xanthophylls compared with the other corn treatments, resulting in increased Roche color fan score for eggs produced by hens fed this diet. Overall, hens fed the transgenic Herculex* I corn grain containing the Cry1F protein performed as well as hens fed the isoline equivalent of Herculex* I and hens fed the conventional corn grains.  相似文献

Parasitic diseases are a major constraint to optimum livestock production and are the major cause of economic loss in UK sheep flocks, with farmers remaining dependant on anthelmintics for control. In the UK, research and evidence based, “best practice” guidelines for sustainable control of parasites in sheep (SCOPS) were first produced in 2004 and have been regularly updated since. This study was designed to evaluate the effect of these best practice guidelines for worm control on lamb production and infection levels, compared with more traditional management. Sixteen farms were selected based on a 2 cube factorial design with 3 factors known to affect worm epidemiology: control regimen; farm type; and climatic region. A formalised plan for worm control using 7 potential resistance-delaying practices was prepared for each of the 8 best practice (SCOPS) farms, in conjunction with the farms veterinarians. The 8 farms in the traditional management group (CONTROL farms) were selected based on ongoing evidence of them using worm control strategies deemed to be “higher-risk”. A cohort of 40–50 study lambs at each farm was monitored from birth to finishing, allowing evaluation of lamb productivity, worm infection levels and for comparison of numbers of anthelmintic treatments. Birth and mid-season weights were used to calculate daily live-weight gain. Birth and finish dates were used to calculate time to finish and finish weights were also compared. Faecal egg counts, larval culture and species differentiation were undertaken throughout the year to assess the impact of the control strategies on worm burdens. There was no significant difference in results for any of the 3 production responses when comparing predicted means accounting for the differences in birth weight. In fact SCOPS farms had, on average, a higher daily weight gain and finish weight than CONTROL farms when comparing observed means. Statistical analysis of infection levels clearly showed no significant effect according to farm type (p = 0.71) or treatment (p = 0.81). In contrast the effect of region (p = 0.08), although not significant, had a much larger effect size (standardised mean difference) with lower parasite burdens based on faecal egg counts on Northern farms compared to Southern farms. For both ewes and lambs, significantly fewer treatments were carried out on the SCOPS farms. The data collected from this study suggests that farms implementing SCOPS principles use less anthelmintic than other farms, without loss of animal performance or increased worm burden.  相似文献

Group A animals were given alfaxalone alone at 3.5 mg kg–1. Group AM animals were given alfaxalone 2 mg kg–1 and medetomidine 40 μg kg–1, reversed with atipamezole at 0.16 mg kg–1. Blinded operators recorded heart rate (HR), respiratory rate (fR), cloacal temperature, depth of sedation and times to: first effect, sedation suitable for clinical interventions, first arousal and full recovery. Data were analysed using independent t test, Mann–Whitney U test, chi-square analysis and log-rank test at 5% level of significance.ResultsSuitable immobilization for clinical examination and sample collection was achieved in all animals. In groups A and AM, median time to working depth was 6.5 minutes (range: 3.4–15) and 8.1 minutes (range: 4.3–24) and time to complete recovery was 66 minutes (range: 12–138) and 34 minutes (range: 4–84), respectively, following reversal. Time to first effect was significantly shorter in group A (p = 0.013), whereas time to full arousal was significantly shorter in group AM (p = 0.007) probably due to the administration of atipamezole. Maximum HR was 117 ± 28 beats minute–1 in group A, which was a significant increase from baseline values (p < 0.0001), whereas group AM showed a significant tachypnoea of 67 ± 25 (normal fR 10–15; p < 0.0001).Conclusions and clinical relevanceBoth the protocols produced immobilization, enabling clinical examination and sample collection; however, protocol AM was more suitable for field work due to shorter recovery times.  相似文献
