Trypanosomes     
Most immunological studies on trypanosomes have been made on species in the subgenus Trypanozoon which includes those causing sleeping sickness in man, but members of the subgenera Duttonella and Nannomonas are more important as the cause of disease in cattle. Wherever possible, studies on these subgenera in cattle are discussed in this paper. The antigenic structure of trypanosomes is complex and common antigens between species, strains and populations occur in homogenates. Antigens variation is responsible for trypanosome surface alter during infection and this antigenic variation is responsible for many of the immunological problems of trypanosmiasis. The characteristics humoral response to infection is a rapid and sustained rise in IgM followed by a later increase in IgG but little is known about cell-mediated immunity in trypanosomiasis. The altered immunoligical state of the infected animal may be of significance in the pathogenesis of the disease in which anemia is marked and kinin production occurs. Possible methods of stimulating protective immunity are discussed and the suppression of immune response to other antigens is analysed.  相似文献

Summary In two distinct areas of East Africa 28,238 and 16,303 non-teneral flies respectively were caught. Infected probosces containing mature trypanosome infections were macerated and inoculated into laboratory animals. Differences in the transmission of trypanosomes existed between different species of tsetse fly. The effect on transmission of trypanosome infection rate in the fly and species of host on which the flies fed is discussed. A method to estimate trypanosome challenge is proposed.
Sumario Se atraparon 28,238 y 16,303 moscas tsetse respectivamente en dos areas didtintas de Africa Oriental. Se maceraron proboscis infectadas con tripanosomas maduros y fueron inoculadas en animales de laboratorio. Existieron diferencias en la trasmisión de tripanosomas entre las distintas especies de moscas tsetse. Se discute el efecto obtenido sobre la transmisión por el grado de infección con tripanosomas en la mosca y en hospederos de los cuales la mosca se alimenta.

Résumé On a capturé dans deux régions différentes de l'Afrique de l'Est respectivement 28.238 et 16.303 mouches ayant pris au moins un repas. Les trompes infestées, contenant des trypanosomes métacycliques, ont été macérées et inoculées á des animaux de laboratoire. Il existait des différences dans la transmission de trypanosomes entre différentes espéces de glossines. L'effet sur la transmission du taux de l'infection á trypanosomes chez la mouche et l'espèce d'h?te sur lequel les mouches s'étaient nourries est discuté. On propose une méthode pour estimer l'intensité des inoculations trypanosomiennes.

The intracellular pathogen Anaplasma marginale (Rickettsiales: Anaplasmataceae), described by Sir Arnold Theiler in 1910, is endemic worldwide in tropical and subtropical areas. Infection of cattle with A. marginale causes bovine anaplasmosis, a mild to severe hemolytic disease that results in considerable economic loss to both dairy and beef industries. Transmission of A. marginale to cattle occurs biologically by ticks and mechanically by biting flies and by blood-contaminated fomites. Both male ticks and cattle hosts become persistently infected with A. marginale and serve as reservoirs of infection. While erythrocytes are the major site of infection in cattle, A. marginale undergoes a complex developmental cycle in ticks that begins by infection of gut cells, and transmission to susceptible hosts occurs from salivary glands during feeding. Major surface proteins (MSPs) play a crucial role in the interaction of A. marginale with host cells, and include adhesion proteins and MSPs from multigene families that undergo antigenic change and selection in cattle, thus contributing to maintenance of persistent infections. Many geographic strains of A. marginale have been identified worldwide, which vary in genotype, antigenic composition, morphology and infectivity for ticks. Isolates of A. marginale may be maintained by independent transmission events and a mechanism of infection/exclusion in cattle and ticks. The increasing numbers of A. marginale genotypes identified in some geographic regions most likely resulted from intensive cattle movement. However, concurrent A. marginale strain infections in cattle was reported, but these strains were more distantly related. Phylogenetic studies of selected geographic isolates of A. marginale, using msp4 and msp1α, provided information about the biogeography and evolution of A. marginale, and msp1α genotypes appear to have evolved under positive selection pressure. Live and killed vaccines have been used for control of anaplasmosis and both types of vaccines have advantages and disadvantages. Vaccines have effectively prevented clinical anaplasmosis in cattle but have failed to block A. marginale infection. Vaccines are needed that can prevent clinical disease and, simultaneously, prevent infection in cattle and ticks, thus eliminating these hosts as reservoirs of infection. Advances in genomics, proteomics, immunology and biochemical and molecular technologies during the last decade have been applied to research on A. marginale and related organisms, and the recent development of a cell culture system for A. marginale has provided a format for studying the pathogen/tick interface. Recent advancements and new research methodologies should provide additional opportunities for development of new strategies for control and prevention of bovine anaplasmosis.  相似文献

Despite the existence of an active vaccination program, recently emerged strains of nephropathogenic infectious bronchitis virus (IBV) in Korea have caused significant economic losses in the poultry industry. In this study, we assessed the pathogenic and antigenic characteristics of a K-IIb type field strain of IBV that emerged in Korea since 2003, such as Kr/Q43/06. Specific pathogen free 1-week-old chickens exhibited severe respiratory symptoms (dyspnea) and nephropathogenic lesions (swollen kidneys with nephritis and urate deposits) following challenge with the recent IBV field strain. The antigenic relatedness (R value), based on a calculated virus neutralization index, of the K-IIb type field strain and K-IIa type strain KM91 (isolated in 1991) was 30%, which indicated that the recent strain, Kr/Q43/06, is a new variant that is antigenically distinct from strain KM91. This report is the first to document the emergence of a new antigenic variant of nephropathogenic IBV in chicken from Korea.  相似文献

The effect of simultaneous infections with trypanosomes (Trypanosoma vivax and Trypanosoma congolense) and gastrointestinal nematodes on the productivity of sheep and goats was studied in 20 animals (6 male goats and 14 male sheep) at a farm near Maputo. The animals were divided into 4 groups which received either treatment against gastrointestinal nematodes, or against trypanosomes, or treatment against both, or no treatment at all.In two-weekly and later in weekly intervals body weight, packed cell volume, body temperature, worm egg burdens and occurence of trypanosomes in the peripheral blood were recorded. Body temperature and packed cell volume did not show the expected close relation to an infection with either trypanosomes or gastrointestinal nematodes, but the differences in the increase of body weight among the 4 groups were very considerable.Animals receiving both treatments gained an average of 13.5 kg in body weight in 40 weeks compared with 5.1 kg (surviving animals with treatment against worm parasites), 6.5 kg (surviving animals with treatment against trypanosomes) and 3.4 kg (surviving animals without any treatment). Of the last 3 groups 1 sheep (21 kg), 2 sheep (50 kg) and 1 sheep (21 kg), respectively, died.One goat, treated against gastrointestinal nematodes, but suffering from an infection with Trypanosoma vivax and Trypanosoma congolense was killed at the end of the experiment. A striking finding during the post-mortem examination was the complete hyperplasia of the red-marrow of the right and left femur.  相似文献

Mycobacterium avium subspecies paratuberculosis (Map) is an important pathogen that causes a chronic, progressive granulomatous enteritis known as Johne’s disease or paratuberculosis. The disease is endemic in many parts of the world and responsible for considerable losses to the livestock and associated industries. Diagnosis and control are problematic, due mostly to the long incubation period of the disease when infected animals show no clinical signs and are difficult to detect, and the ability of the organism to survive and persist in the environment. The existence of phenotypically distinct strains of Map has been known since the 1930s but the genetic differentiation of Map strain types has been challenging and only recent technologies have proven sufficiently discriminative for strain comparisons, tracing the sources of infection and epidemiological studies. It is important to understand the differences that exist between Map strains and how they influence both development and transmission of disease. This information is required to develop improved diagnostics and effective vaccines for controlling Johne’s disease. Here I review the current classification of Map strain types, the sources of the genetic variability within strains, growth characteristics and epidemiological traits associated with strain type and the influence of strain type on infection and pathogenicity.  相似文献

The aim of the present study was to reveal the characteristic features of genital Chlamydia suis infection and re-infection in female pigs by studying the immune response, pathological changes, replication of chlamydial bacteria in the genital tract and excretion of viable bacteria. Pigs were intravaginally infected and re-infected with C. suis strain S45, the type strain of this species. We demonstrated that S45 is pathogenic for the female urogenital tract. Chlamydia replication occurred throughout the urogenital tract, causing inflammation and pathology. Furthermore, genital infection elicited both cellular and humoral immune responses. Compared to the primo-infection of pigs with C. suis, re-infection was characterized by less severe macroscopic lesions and less chlamydial elementary bodies and inclusions in the urogenital tract. This indicates the development of a certain level of protection following the initial infection. Protective immunity against re-infection coincided with higher Chlamydia-specific IgG and IgA antibody titers in sera and vaginal secretions, higher proliferative responses of peripheral blood mononuclear cells (PBMC), higher percentages of blood B lymphocytes, monocytes and CD8+ T cells and upregulated production of IFN-γ and IL-10 by PBMC.

In the last decade, there has been a wide range of studies using a series of molecular markers to investigate the genotypic diversity of some of the important species of African trypanosomes. Here, we review this work and provide an update of our current understanding of the mechanisms that generate this diversity based on population genetic analysis. In parallel with field based studies, our knowledge of the key features of the system of genetic exchange in Trypanosoma brucei, based on laboratory analysis, has reached the point at which this system can be used as a tool to determine the genetic basis of a phenotype. In this context, we have outlined our current knowledge of the basis for phenotypic variation among strains of trypanosomes, and highlight that this is a relatively under researched area, except for work on drug resistance. There is clear evidence for ‘strain’-specific variation in tsetse transmission, a range of virulence/pathogenesis phenotypes and the ability to cross the blood brain barrier. The potential for using genetic analysis to dissect these phenotypes is illustrated by the recent work defining a locus determining organomegaly for T. brucei. When these results are considered in relation to the body of research on the variability of the host response to infection, it is clear that there is a need to integrate the study of host and parasite diversity in relation to understanding infection outcome.  相似文献

Summary The tsetse fly (Glossina spp.) inhabits 11 million km2 of Africa (Greekmore, 1989) where it is responsible for the transmission of trypanosomosis to man and animals. Because of its slow rate of reproduction, with females producing only four to five pupae per annum, control of the tsetse fly is the best means of controlling trypanosomosis. A number of different methods have been and are used but, whilst successes have been achieved, a long term solution has not been found.
Resumen La mosca tsetsé (Glossina spp.) infesta 11 millones de km2 (Greekmore, 1989) donde es responsable de la transmisión de tripanosomiasis al hombre y animales. Debido a la baja tasa de reproducción de la mosca (cuatro a cinco pupas por a?o), el control de la mosca per se es el método más efectivo, habiendose obtenido algún éxito, sin encontrer todavía una solución a largo plazo.

Foodborne salmonellosis is one of the most important bacterial zoonotic diseases worldwide. Salmonella Typhimurium is the serovar most frequently isolated from persistently infected slaughter pigs in Europe. Salmonella Typhimurium pathogenesis is host species specific. In addition, differences in in vitro behaviour of Salmonella Typhimurium strains have also been described, which may be reflected by a different course of infection within a host species. We compared the course of a Salmonella Typhimurium infection in pigs, using two Salmonella Typhimurium strains that were able to interfere with MHC II expression on porcine macrophages to a different extent in vitro. After experimental inoculation, blood and faecal samples from all pigs were collected at regular time points. At 40 days post inoculation (pi), animals were euthanized and tissue samples were bacteriologically analysed. The proportion of serologically positive piglets at 33 days pi was significantly higher in pigs that were inoculated with the strain that did not downregulate MHC II expression in vitro. Furthermore, this strain was less frequently shed and isolated in lower numbers from tonsils and ileocaecal lymph nodes than the strain that was able to markedly downregulate MHC II expression in vitro. We thus found that the delayed onset of seroconversion after oral inoculation of piglets with a particular Salmonella Typhimurium strain coincided with higher faecal shedding and increased persistence. Strain specific differences in Salmonella pathogenesis might thus have repercussions on the serological detection of Salmonella Typhimurium infections in pigs.  相似文献

为了探明伊氏锥虫在动物体内抗原变异,以伊氏锥虫新疆株克隆感染兔,每3天采兔血1次,共采血5次,并用环磷酰胺处理的小白鼠对各分离期锥虫予以克隆,获得5个锥虫克隆群体,经ABC-酶标试验和间接免疫荧光试验,对各克隆锥虫抗原进行鉴定,结果:获得5个表面抗原性互不相同的克隆群体,说明伊氏锥虫新疆株第1次抗原变异发生在4-6日内,以后每3天变异1次,本研究为进一步探明伊氏锥虫抗原变异规律打下了基础。  相似文献

Bordetella bronchiseptica (B. bronchiseptica) is associated with respiratory tract infections in laboratory animals. In our laboratory animal facility, B. bronchiseptica was isolated from 21 of 27 apparently healthy rabbits obtained from a breeding farm contaminated with B. bronchiseptica. Restriction fragment length polymorphism (RFLP) analysis showed that the flagellin genotype of isolates from the laboratory animal facility and breeding farm was type A, which is seen relatively frequently in rabbits in Europe. To examine its pathogenicity, guinea pigs, rats, and mice were inoculated intranasally with a representative strain isolated in the laboratory animal facility. Following inoculation of 107 colony forming unit (cfu), severe inflammation was observed in the lungs of guinea pig and mice, although the inflammation was less severe in rats. The strain was recovered from the trachea and lungs of these species after inoculation with lower dose such as 103 or 104 cfu. These results suggest that the isolated strain causes respiratory tract infection in guinea pigs, rats, and mice, and that its pathogenicity higher in mice than in rats. This study extends our knowledge of interpreting the microbiologic status of laboratory animals, which will contribute to the development of reliable and reproducible animal experiments.  相似文献

During two consecutive surveys (February and August/Sept 2002), a total of 970 cattle from the cattle population of Mafia Island (United Republic of Tanzania) were blood-sampled. All blood samples were microscopically screened for the presence of trypanosomes and a portion of these were checked for antibodies with an Ab-ELISA and for the presence of trypanosomal DNA with PCR. Microscopic evidence of trypanosomes of the congolense group (sub-genus Nannomonas) was found in 0.8% of the animals (8/970) and in two cases the species identified was confirmed by PCR as Trypanosoma congolense savannah type. Non-pathogenic Trypanosoma theileri were detected in 3.2% (31/970) of the samples using the Dark Ground-Buffy Coat (DG-BC) technique. For survey 1 (S1), detection of antibodies (Ab-ELISA) against pathogenic trypanosomes indicated a seroprevalence of 14.2% (68/480). Of the samples, either DG positive or with a PCV lower then 25, examined by PCR, a total of 8.4% (5/59) (selected from 970 samples), were found positive for T. congolense. The low prevalence of pathogenic trypanosomes on Mafia Island is intriguing, especially in view of the omnipresence of the tsetse fly Glossina brevipalpis. Although the presence of detected trypanosomal antibodies does not necessarily indicate a current infection, the combination of serological/parasitological examinations and the results of the PCR do support this low prevalence of trypanosomosis in cattle. Despite the low prevalence, pathogenic trypanosomes are present on Mafia Island and possible reasons for this low infection rate, taking account of the relation between Glossina species present, transmission risk and trypanosomes found in cattle, are discussed also in view of a future appropriate intervention strategy.  相似文献

Tropical canine pancytopenia (TCP), a disease of dogs caused by the rickettsial agent, Ehrlichia canis, is being recognized with increased frequency in many parts of the world. The brown dog tick, Rhipicephalus sanguineus, is an efficient vector of E. canis and transstadial transmission occurs. Infections persist and are characterized by rising antibody titers and hypergammaglobulinemia. Dogs cleared of infection with tetracycline are fully susceptible to reinfection with the homologous strain of E. canis and often develop severe disease. Experimental infections in beagle and German shepherd dogs have shown that severity of disease is related to the breed of dog. German shepherd dogs often develop a severe hemorrhagic syndrome 60 or more days following infection. Recent studies have shown that bone marrow hypoplasia is important in the pathogenesis of severe chronic TCP. Differences in response to infection may reflect underlying immunological defects, particularly in the cell-mediated system.  相似文献

Summary The host response of adult East African zebu cattle to trypanosome challenges of widely differing pathogenicity and antigenicity was studied. Two types of acquired immunity were distinguished; sterile and non-sterile. Sterile immunity was characterized by the absence of disease, the absence of circulating trypanosomes, a reciprocal titre of neutralizing antibodies of 102 or greater, and a high titre of immunofluorescent antibodies. Non-sterile immunity was characterized by the absence of disease, the presence of circulating trypanosomes; and again by a high titre of immunofluorescent antibody. Sterile immunity was achieved initially only when the immunizing and challenge infection consisted of one well defined antigenic type. Non-sterile immunity was induced when the antigenic types used to challenge were less well defined.
Sumario La respuesta de ganado Cebú adulto del Africa Oriental al desafío con tripanosomas de patogenicidad bastante variada y de antigenicidad fué examinada. La resistencia fué medida basándose en la presencia ó ausencia de tripanosomas y en desarrollo de anticuerpos neutralizantes y fluorescentes. Se demostró que desarrollaron dos tipos de inmunidad, una esteril y otra no esteril. Se discute la definición de los términos usados y la caracterización de la inmunidad.

Résumé On a étudié la réponse immunologique de bétail adulte zébu d'Afrique orientale à l'infection par des souches de trypanosomes de pouvour pathogène et d'antigénicité très différents. La résistance était évaluée selon les critères suivants: absence ou présence de trypanosomes, apparition d'anticorps neutralisants et d'anticorps recherchés en immunofluorescence. Deux types d'immunité puvaient être mis en évidence, le premier sans trypanosomes présents (stérile), le second associé à l'infection (non stérile). La définition des critères utilisés et la caractérisation de l'immunité sont discutées.

Dogs can be infected by a wide variety of Bartonella species. However, limited data is available on experimental infection of dogs with Bartonella strains isolated from domestic animals or wildlife. We report the inoculation of six dogs with Bartonella henselae (feline strain 94022, 16S rRNA type II) in three sets of two dogs, each receiving a different inoculum dose), four dogs inoculated with B. vinsonii subsp. berkhoffii type I (ATCC strain, one mongrel dog) or type II (coyote strain, two beagles and one mongrel) and B. rochalimae (coyote strain, two beagles). None of the dogs inoculated with B. henselae became bacteremic, as detected by classical blood culture. However, several dogs developed severe necrotic lesions at the inoculation site and all six dogs seroconverted within one to two weeks. All dogs inoculated with the B. v. berkhoffii and B. rochalimae strains became bacteremic at levels comparable to previous experimental infections with either a dog isolate or a human isolate. Our data support that dogs are likely accidental hosts for B. henselae, just like humans, and are efficient reservoirs for both B. v. berkhoffii and B. rochalimae.  相似文献

Monthly incidence rates for the detection of bovine leukemia virus (BLV) infection were calculated for a 15-month period for cattle over 12 months of age in a 200-cow dairy herd. Detection of BLV infection was based on the presence of persisting antibodies as measured by agar-gel immunodiffusion using the glycoprotein-51 antigen. A non-parametric procedure was used to examine 6-month trends in cumulative monthly incidence rates. Over the 15-month study, 25 heifers were detected as becoming infected with BLV after 12 months of age. No differences were found between the 12 monthly incidence rates (p = 0.77). Cumulative incidence rates between June and September (the fly season) were not higher than those for any other 6-month period (p = 0.29). Similarly, there was no evidence that BLV infection was transmitted during artificial insemination (p = 0.18) or during routine vaccination procedures (p = 1.0). Cumulative rates for a period corresponding to exposure of heifers to the dry herd were excessively high (p = 0.01). These findings suggest that transmission of BLV infection in the cattle studied was associated with close physical contact between susceptible and infected cattle, and not with biting flies, artificial insemination, or vaccination procedures.  相似文献

The use of transgenic plants as delivery system for antigenic proteins is attractive for its simplicity and increases likelihood for local immune response at sites of infection. The aim of this study was to evaluate the protective effect of oral administration of tobacco seeds, expressing the FedA, the major protein of the F18 adhesive fimbriae, and B subunit of verocytotoxin, against verocytotoxin-producing E. coli (VTEC) strain in piglets. Forty-three early weaned piglets, were randomly divided into 4 experimental groups: 3 test groups and a control. Treatment groups orally received a bolus, with different dose of tobacco seeds on 0, 1, 2, 14 days post primary administration. After challenge, with 1*1010 CFU of O138 Escherichia coli strain, piglets showed clinical scores significantly higher in the control group compared to orally immunized groups (P?<?0.05) and the latter showed a faster recovery than in CG. In conclusion, oral administration of recombinant tobacco seeds expressing antigenic proteins against VTEC strains can induce a protective effect against challenger strain in piglets.  相似文献

Habronemiasis is a parasitic disease of horses and other equids which is caused by the invasion of Draschia megastoma, Habronema majus, and H. muscae nematodes and is transmitted by house flies, face flies, and stable flies. The adult nematodes live in the horse's stomach, laying eggs, which pass out in the horse's feces. The carrier flies lay eggs in the horse's manure, which becomes infested with larvae while the flies feed in the manure. The nematode larvae molt while in the fly larvae and develop into infective L3 larvae by the time the adult flies emerge from their pupal stage. The infective larvae are then transmitted to the horse when flies feed on secretions or discharges from hosts' eyes, nostrils, mouths, wounds, and other openings. The L3 larvae are ingested by the horse and are then able to complete their lifecycle. The most common aberrant forms (conjunctival and cutaneous habronemiasis) are associated with the nematode larvae being deposited in these areas, not completing its life cycle, with resultant signs probably associated with local hypersensitivity. This article details the biology of nematode parasites, diagnosis and treatment of this condition, and methods of prevention and control of the transmitting flies.  相似文献
