The effect of pregnant mare serum gonadotropin (PMSG) or estradiol cyclopentylpropionate (EC) on the induction of estrus, duration of estrus, and serum progesterone concentration after estrus was evaluated in 8 gilts with delayed puberty. Four gilts were given 500 IU of PMSG IM and 4 were given 2 mg of EC, IM. The inactive status of the ovaries at the time of treatment was verified by serum progesterone values of less than 0.5 ng/ml in serial samples collected before treatment. The 4 EC-treated gilts came into estrus at a mean of 3.5 days after treatment, but 1 of the gilts did not form corpora lutea. Three PMSG-treated gilts came into estrus at a mean of 4.0 days after treatment. The remaining PMSG-treated gilt remained anestrus and did not form corpora lutea. The mean duration of estrus in EC-treated gilts was 5.25 days compared with 2.0 days for PMSG-treated gilts (P less than 0.05). Serum progesterone concentrations were higher in PMSG-treated gilts than in EC-treated gilts at 8, 11, and 17 days after treatment (P less than 0.05).  相似文献

The possible roles of dietary protein level and feed restriction in regulating mammary development of prepubertal gilts were investigated. Cross-bred gilts were fed a commercial diet until 90 d of age and then divided into four nutritional regimens based on two pelleted diets (as-fed basis): a high-protein diet (HP = 13.8 MJ of ME, 1.0% total lysine, 18.7% CP) and a low-protein diet (LP = 13.8 MJ of ME, 0.7% total lysine, 14.4% CP). Nutritional regimens were as follows: 1) HP ad libitum until slaughter (n = 22, T1); 2) HP ad libitum until 150 d of age followed by LP until slaughter (n = 20, T2); 3) LP ad libitum until slaughter (n = 21, T3); and 4) HP with a 20% feed restriction until slaughter (n = 19, T4). Gilts were weighed, their backfat thickness was measured, and jugular blood samples were obtained on d 90, 150, and at slaughter to determine concentrations of prolactin, IGF-I, leptin, and glucose. Gilts were slaughtered 8+/-1 d after their first or second estrus (202.7+/-14.5 d of age). Mammary glands were excised, parenchymal and extraparenchymal tissues were dissected, and composition of parenchymal tissue (protein, fat, DM, DNA, protein/DNA) was determined. The T4 gilts weighed less (P < 0.01) and had less backfat (P < 0.01) than did gilts on other treatments on d 150 and at slaughter. Treatments had no significant effects on prolactin, IGF-I, or glucose concentrations, but there was a treatment x day interaction (P < 0.01) for leptin, with concentrations being lower at slaughter in restricted-fed (T4) vs. LP (T3) gilts (P < 0.05). There was less extraparenchymal mammary tissue (P < 0.01) in T4 gilts than in gilts from the other groups and a tendency (P = 0.13) for the amount of parenchymal tissue to be lower in T4 gilts. In conclusion, a lower lysine intake during prepuberty did not hinder mammary development of gilts, but a 20% feed restriction decreased mass of parenchymal and extraparenchymal tissues. The effect of feed restriction on extraparenchymal tissue is most likely associated with the lower fat deposition.  相似文献

Eleven Yorkshire x Landrace gravid gilts were used in two Latin square trials to determine the N requirement for pregnancy. Semipurified diets were formulated to be adequate in indispensable amino acids (IAA) and other nutrients. Diets were fed once daily (1.82 kg); L-glutamic acid, corn sugar, powdered cellulose, and soybean oil percentages differed to maintain isocaloric diets. In Trial 1, six gilts were fed diets containing 6.6 to 17.2% CP equivalent (19.2 to 50.1 g of N/d) during six 10-d periods beginning on d 40 postcoitum. In Trial 2, five gilts were fed diets containing 4.3 to 12.6% CP equivalent (12.5 to 36.8 g of N/d) during five 10-d periods beginning on d 50 postcoitum. Nitrogen balance trials were conducted during the last 5 d of each period. Blood samples were taken both before and 3 h after the last feeding of each period. Results from Trial 1 suggested that adequate N retention (10 g/d) could be attained by pregnant gilts fed less than 28 g of N/d intake when the diet contained indispensible amino acids at levels suggested by Nutrient Requirements of Swine (NRC, 1988). An intake of 20.6 g of N/d (7.1% CP equivalent) yielded near maximum N retention among pregnant gilts fed the semipurified diets in Trial 2. Other criteria measured (urine urea nitrogen and plasma urea nitrogen) had limited value in the evaluation of the N status of the pregnant gilts in these trials.  相似文献

Miniature and domestic sows at 108 to 110 days of the gestation were exposed to atmospheric carbon monoxide (CO) concentrations of 150 to 400 ppm for 48 to 96 hours. Overall stillbirth rates were 6.7%, 34.8%, 42.3%, and 80.0% in the sows exposed to CO in concentrations of 200, 250, 300, and 350 ppm, respectively. A significant linear relationship was determined between these concentrations of CO and the resultant maternal carboxyhemoglobin (COHb) concentration (P less than 0.01). The frequency of stillbirth increased significantly when maternal COHb concentration exceeded 23% saturation of hemoglobin. The COHb concentrations in new-delivered pigs (cesarean section) were greater than maternal COHb concentrations by 3 to 22%. Common gross lesions in stillborn pigs were cherry red discoloration of the subcutaneous tissues, muscle, and viscera and accumulation of a large volume of serosanguineous pleural effusion. Hypoxic ischemic leukoencephalopathy was found in new-delivered pigs from 3 of 14 litters. Lesions included focal leukoencephalomalacia, glial-vascular proliferation, multifocal hemorrhage, and vacuolation of the neuropile. The experimental material consisted of 42 sows divided into 3 feeding groups, each containing 14 sows fed complete compound feed varying in protein content. Protein and amino acid content of the feed fed to the control group was in compliance with Polish standards. Protein content in the experimental groups was reduced by 10% and 20%, respectively. At the same time, lysine, methionine, threonine and tryptophane content was supplemented to the level of the control group. All pregnant and lactating sows were fed individually. The condition of the sows was determined by measuring the thickness of backfat at the P2 position on days 30 and 105 of pregnancy and on day 25 of lactation. The data analyzed in the study included: the total number of piglets born, their body weight gain, milk composition, weaning-to-estrus interval and the farrowing rate. Protein content of the compound feed did not have a significant impact on the increment in backfat during pregnancy and losses during lactation. The sows fed compound feed with reduced protein content gave birth to 0.7 and 0.6 less piglets per litter than the control animals. However, due to lower losses, they bred 0.3 and 0.4 more piglets than the control sows. Average body weight of a piglet on day 21 was 0.5 kg lower in the experimental than in the control group. Dry matter, protein, fat and lactose content of the sow's milk did not depend on protein content of the feed. The weaning-to-estrus interval in the experimental groups was one day longer than in the control group. The reproductive rate accounted for 86% and was comparable in all the groups.  相似文献

Thirty-nine Landrace gilts, average age 240.6 days and mean body weight 117.1 kg, were mated and divided into five feeding groups. Gilts of groups 1 to 4 were fed daily, for 111 days of gestation, 1.5, 2.0, 2.5 or 3.5 kg of a diet containing 12% digestible protein (DP); gilts of group 5 were given daily 3.5 kg of a diet containing 14.7% DP. All gilts were slaughtered on day 112 of gestation and chemical composition of the carcass and internal organs, including the gestated utera, were analysed. Initial chemical body composition of gilts was estimated by equations obtained on 28 animals slaughtered at the beginning of the experiment.The level of feeding had no effect on the litter size, or on the chemical composition of the products of conception.Protein and fat deposition increased linearly with elevation of the amount of food per day. In gilts given daily 1.5 kg of the diet, retention of protein was accompanied by loss of fat from the body.Increase in protein content in the diet did not reduce the utilization of digestible protein for its retention in the body.The protein and energy costs of 1 g protein deposited in tissues was higher in the products of conception (6.57 g DP and 79.2 kJ ME) than in the maternal bodies of gilts (3.66 g DP and 48.4 kJ ME). The maintenance requirement of pregnant gilts was estimated at 490.6 kJ ME per kg0.75 per day.  相似文献

Xanthine oxidoreductase (XOR) is a key enzyme in the evolvement of reperfusion injury resulting from birth asphyxia, a common cause of decreased viability and perinatal mortality in newborn piglets under farm conditions. At present no standard pharmacological intervention strategy is available to reduce these adverse effects of birth asphyxia. In the present study we aimed to evaluate placental transfer of allopurinol, an inhibitor of XOR. For this purpose, fetal catheterization of the jugular vein was conducted in five late pregnant sows (one fetus per sow). At 24–48 h after surgery, sows received allopurinol (15 mg/kg body weight; i.v.) and pharmacokinetics of allopurinol and its active metabolite oxypurinol were measured in both late pregnant sows and fetuses. Maternal and fetal blood samples were collected during and after allopurinol administration. Maternal Cmax values averaged 41.90 μg/mL (allopurinol) and 3.68 μg/mL (oxypurinol). Allopurinol crossed the placental barrier as shown by the average fetal Cmax values of 5.05 μg/mL at 1.47 h after allopurinol administration to the sow. In only one fetus low plasma oxypurinol concentrations were found. Incubations of subcellular hepatic fractions of sows and 24‐h‐old piglets confirmed that allopurinol could be metabolized into oxypurinol. In conclusion, we demonstrated that allopurinol can cross the placental barrier, a prerequisite for further studies evaluating the use of allopurinol as a neuroprotective agent to reduce the adverse effects following birth asphyxia in neonatal piglets.  相似文献

The plasma kinetic profile of ivermectin during the last trimester of pregnancy was studied in ewes after a single subcutaneous administration of 0.2 mg/kg body weight (BW). Sheep were randomly distributed into two groups. Ewes in group 1 (control, n=6) were left unmated, whereas in group 2 (pregnant, n=6) ewes were estrus-synchronized and mated with rams. Both groups were housed under similar conditions of management and feeding. At 120 days of pregnancy, both groups were given a subcutaneous injection of 0.2 mg/kg BW of ivermectin. Blood samples were taken by jugular puncture according to a fixed protocol between 1 h and 40 days post-treatment. After plasma extraction and derivatization, samples were analyzed by high performance liquid chromatography with fluorescence detection. A computerized pharmacokinetic analysis was performed, and the data were compared by means of the Student t-test. The results showed that plasma concentrations of ivermectin remained longer in the pregnant than in the control group. The mean values of pharmacokinetic parameters C(max), t(max), and area under the concentration-time curve (AUC) were similar for both groups of sheep. The mean residence time (MRT) values for the pregnant group (8.8+/-1.4 days) were higher (P<0.05) than those observed in the control group (5.3+/-1.9 days). It can be concluded that pregnancy increases the residence time of ivermectin in the plasma of pregnant sheep when it is administered subcutaneously.  相似文献

The objectives of this study were to characterize the quantitative changes in various body tissues of high-lean type gilts during gestation and to determine the protein needs of pregnant gilts based on changes in tissue contents. Thirty-five gilts (158.2 +/- 8.3 kg) were housed in individual gestation crates with six unbred gilts randomly selected and slaughtered to provide data for d 0 of gestation. The remaining gilts were bred and assigned randomly to one of six slaughter groups: d 45, 60, 75, 90, 102, and 112. Gilts were fed 2 kg (as-fed basis) of gestation diet daily (3.1 Mcal/kg of ME and 0.56% lysine). Carcass soft tissue, bone, gastrointestinal tract, spleen, pancreas, kidney, liver, uterus, fetus, mammary gland, and the remaining viscera were separated and weighed. Carcass soft tissue, liver, remaining viscera, uterus, and gastrointestinal tract were ground, freeze-dried, and analyzed for composition. Body weights of the gilts increased quadratically (P < 0.001) during gestation. Weights of carcass soft tissue and uterus, including placenta, increased linearly (P < 0.001) during gestation. Weights of individual fetuses, fetal litters, individual mammary glands, and the entire mammary glands increased cubically (P < 0.001) during gestation. Crude protein in carcass soft tissue increased cubically (P < 0.01), whereas DM and ether extract (EE) in carcass soft tissue increased linearly (P < 0.01). The DM, CP, and EE in the entire mammary glands increased quadratically (P < 0.001) during gestation. The DM, CP, and EE in fetal litter increased cubically (P < 0.01) as gestation progressed. The accretion rates of the conceptus, fetal litter, individual fetus, individual mammary gland, and CP in fetal litter differed (P < 0.05) before and after d 70 of gestation. The CP daily gain from all maternal and fetal tissues was 40 and 103 g/d before and after d 70 of gestation, respectively, suggesting that pregnant gilts may require different quantities of dietary protein during gestation. Based on the maintenance requirement, maternal tissue gain, and conceptus gain, pregnant gilts require 6.8 and 15.3 g/d of true ileal-digestible lysine (or 147 and 330 g/d of true ileal-digestible protein) before and after d 70 of gestation, respectively, to support their true biological needs.  相似文献

The hypothesis of the present study was that a GnRH agonist application at early pregnancy would alter the pattern of the key reproductive hormones LH and FSH, and subsequently that of estradiol (E2) and especially progesterone (P4), and improve the conditions for embryo survival in early pregnant gilts. Therefore, the endocrine effects of a GnRH agonist (GnRHa) application to gilts (n=11 GnRHa treated, n=9 saline Controls) were studied in blood samples from the Vena cava caudalis. GnRHa injected on Day 12 after insemination induced elevated (P<0.01) LH and FSH levels for at least 180 min. However, subsequent LH concentrations were not altered up to Day 21 of pregnancy. LH pulse number, estimated in 6-h period samples on Days 13, 15 and 17, was not influenced by treatment and pregnancy. LH pulse amplitude was decreased (P<0.05) on Days 13 to 17 in pregnant gilts of both groups, but not in nonpregnant animals. In pregnant GnRHa-treated gilts, the basal LH level was elevated compared with the Controls (P<0.01). Additionally, differences (P<0.05) in basal LH were present between the pregnant and nonpregnant animals. The P4 and E2 secretion pattern was not affected by GnRHa. P4 concentrations increased (P<0.01) from Day 10 to Day 14 regardless of the treatment. P4 revealed a pulse-like pattern, but without a definite relation to the LH pulse characteristics. Also, pregnancy rate (73 vs. 67%) and the number of fetuses (12.8 ± 2.3 vs. 11.6 ± 2.3) were unaffected in the treated and Control gilts, respectively. The present study did not confirm the initial hypothesis that a GnRHa-mediated LH effect could alter ovarian steroid secretion and favorably support early embryo development and pregnancy outcome.  相似文献

Ovariectomized gilts were either placed on full feed (FF) or restricted to one-third of the full feed amount (RST) for 7 days. Blood samples were taken through jugular catheters every 15 min for 4 h at the end of the 7-day period. Then dietary treatments were reversed and 7 days later samples were taken as before. Serum concentrations of leptin, insulin and luteinizing hormone (LH) were determined by radioimmunoassay. LH pulse frequency and mean serum leptin and insulin concentrations were lower (P < 0.01) in RST than FF gilts. Reversal of treatment reversed the patterns of hormone secretion. These results confirm previous observations that feed restriction can inhibit pulsatile LH secretion and also decrease leptin and insulin secretion.  相似文献

Characterization of mammary gland development in pregnant gilts   总被引:2,自引:0,他引:2  
The purpose of this study was to quantify mammary gland (MG) growth during pregnancy in gilts and to determine the effect of anatomical location on gland growth. Size, composition, and histomorphology of MG were determined during gestation in 29 primigravid gilts. Gilts were allotted randomly to 6 slaughter groups: d 45 (n = 6), 60 (n = 4), 75 (n = 5), 90 (n = 4), 102 (n = 5), and 112 (n = 5) of gestation. Mammary glands were obtained at slaughter, and skin and extraneous fat pad were removed to obtain parenchymal MG tissue. Mammary glands were further separated into individual MG, and their locations were recorded. Individual MG were weighed and bisected in an approximate midsagittal section to measure cross-sectional area. Mammary glands were ground individually and pooled according to anatomical region: the first and second pairs of MG = anterior MG; the third, fourth, and fifth pairs of MG = middle MG; the sixth, seventh, and eighth pairs of MG = posterior MG. Contents of DM, CP, ether extract, and crude ash were measured. Wet weight, DM, CP, and ash content of total and individual MG increased (P < 0.01) between d 45 and 112 of gestation. Cross-sectional area of individual MG increased (P < 0.01) as gestation progressed. Percentage of CP and ash increased (P < 0.01), whereas percentage of ether extract decreased (P < 0.01) as gestation progressed. This inverse relationship between percentages of CP and ether extract (r = -0.999; P < 0.0001) was consistent with the histological shift from primarily an adipose tissue in early gestation to one containing extensive lobuloalveolar tissue in late gestation. Wet weight of middle MG was greater (P < 0.05) than that of posterior MG at d 102 and 112 of gestation, and amount of CP in middle MG was greater (P < 0.05) than that in anterior and posterior MG at d 102 and 112 of gestation, indicating that middle MG grow faster than other MG during late gestation. Rates of wet weight gain and protein accretion were accelerated (P < 0.01) after d 74 and 75 of gestation, respectively, indicating the importance of MG growth during the last trimester of gestation. The increase in rate of protein accretion after d 75 indicates a greater protein requirement for MG growth during later gestation.  相似文献

Two experiments were conducted to evaluate the relationship among the level of feed intake, chewing pattern, and diet digestibility in sheep fed a moderate‐concentrate diet. The first experiment was conducted using six male lambs at a replicated 3 × 3 Latin square design to evaluate diet digestibility and microbial N synthesis according to the level of intake: ad libitum, or restricted to either 70 or 55% of the ad libitum intake. In the second experiment, fifteen male lambs were housed in individual stalls, in a completely randomized design, and fed one of the three levels of dry matter intake (DMI). Chewing patterns were then evaluated for 24 h using a regular 5‐min interval observation technique, two times during the experimental period. Decreasing level of feed intake resulted in increased apparent digestibility of dry matter, organic matter, crude protein, and neutral detergent fibre, as well as of the true digestibility of organic matter. Total time spent eating and ruminating decreased with feed restriction. However, lambs fed at restricted levels of intake presented a higher rate of eating (g DMI/min) than those fed ad libitum, and spent more time ruminating each gram of DM (min/g DMI). In conclusion, our results suggest that a more effective chewing during rumination activity can have an important role on feed digestion in animals submitted to feed restriction.  相似文献

We studied luteinizing hormone (LH) pulsatility and episodic progesterone release of the corpus luteum (CL) on Day 11 and Day 21 in inseminated gilts and aimed to establish a relationship between these two hormones. Blood was collected at 15-min intervals for 12 hr on Days 11, 16 and 21 from a vena cava caudalis catheter. At euthanasia, eight gilts were pregnant and six gilts were not pregnant. Progesterone parameters (basal, mean, pulse frequency and pulse amplitude) did not differ between pregnant and non-pregnant gilts on Day 11, LH pulse frequency and amplitude tended to differ (p = .07 and p = .079). In pregnant gilts, basal and mean progesterone, progesterone pulse amplitude and frequency declined significantly from Day 11 to Day 21 (p < .05). A significant decline was also seen in the LH pulse amplitude from Day 11 to Day 21 (p < .05). None of the LH pulses was followed by a progesterone pulse within 1 hr on Day 21. On Day 11 and Day 21 appeared a synchronicity in the LH pulse pattern, as there were two or three LH pulses in 12 hr and these LH pulses appeared in the same time window. We conclude that on Day 11 and Day 21 of pregnancy in gilts, progesterone pulses do not follow an LH pulse within one hour. Further we demonstrated that the successful or not successful formation of a CL of pregnancy is independent of progesterone release on Day 11 after insemination. We confirmed the decline of progesterone from Day 11 to Day 21 in the vena cava caudalis and could demonstrate that this decline is partly due to lower progesterone pulse amplitude and frequency and that the decline occurs simultaneously with a decline in LH pulse amplitude.  相似文献
