Tyrosinase-related protein 1 (tyrp1) and tyrosinase-related protein 2 (tyrp2) genes are important downstream regulatory factors for body colour formation in animals. The present study aimed to explore the relationship between tyrp1a and tyrp2 expression and body colour variation between wild-type (WT) and yellow mutant (YM) rainbow trout. The full-length cDNA sequences of trout tyrp1a and tyrp2 were obtained using rapid amplification of cDNA ends, and the bioinformatic analysis was performed. Quantitative real-time PCR (qRT-PCR) was used to investigate the different expression levels of tyrp1a and tyrp2 in different developmental stages and tissues. The full-length cDNAs of tyrp1a and tyrp2 were 2409 bp and 2219 bp encoding predicted proteins of 522 and 529 amino acid residues, respectively. Both proteins possess six conserved domains, including a signal peptide, an EGF-motif, a Pfam tyrosinase motif, a transmembrane structure, and two copper binding sites (CuA and CuB). Amino acid sequence comparisons showed higher conservation of Tyrp1a and Tyrp2 proteins amongst fishes than amongst other vertebrates, which was further confirmed by phylogenetic analysis. qRT-PCR analysis revealed that tyrp1a and tyrp2 expression levels in WT rainbow trout, and tyrpla in YM rainbow trout, were detected from the fertilised stage to 12 months post hatching (12 M), while tyrp1a did not expressed until the blastula in YM rainbow trout. In addition, the expression levels of both of the genes post hatching were significantly higher than those in the embryonic stages, which showed extremely low expression levels. Additionally, there were extremely significant differences (P < 0.01) in expression at the same stages between WT and YM rainbow trout, e.g., at the blastula, gastrula, 7 days post hatching (7 dph), 10 dph, 2 M, 3 M, 6 M, and 12 M stages for tyrp1a; and at the 4-cell, 16-cell, multicell, blastula, somites, heartbeating, 1 dph, 5 dph, 7 dph, 3 M, 6 M, and 12 M stages for tyrp2. Besides, variable expression levels of tyrp1a and tyrp2 were detected in all tissues; significantly high expression was detected in the dorsal skin and eye compared with that in the other tissues in WT and YM rainbow trout (P < 0.05). These results suggested that the expression level changes of tyrp1a and tyrp2 might be closely related to the variation of rainbow trout body colour, which will enrich our knowledge of the molecular mechanism of skin colour variation in rainbow trout and provide data for research into fish skin colour inheritance and improvement.

Cyp19a1b、Cyp11a1、Cyp11b2、Cyp17a1、Hsd3b1等细胞色素相关基因能够调节硬骨鱼类性类固醇的合成,对性腺发育和性别决定产生影响。本研究以全雌三倍体虹鳟(Oncorhynchus mykiss)为研究对象,正常雌性二倍体虹鳟为对照,选取31~68 dpf(days post fertilization)时间段的虹鳟仔鱼脑组织,采用q RT-PCR和酶联免疫的方法研究以上几种基因的表达状况和脑芳香化酶的活性变化,以期探明导致三倍体雌性虹鳟性腺发育异常的关键原因。q RTPCR结果显示,二倍体中Cyp19a1b在30~50 dpf时表达量上调并且维持在较稳定水平,但50~56 dpf时表达量逐渐下调,之后56~68 dpf表达量持续上调;三倍体中Cyp19a1b表达量在30~35 dpf开始上调,35~47 dpf逐渐下调,47~55 dpf开始第二次上调,之后维持在较稳定水平直至68 dpf,但三倍体Cyp19a1b的表达量显著(P0.05)低于同期二倍体的。二倍体Cyp11a1表达量在34 dpf出现峰值,三倍体Cyp11a1在38 dpf时出现峰值。二倍体Hsd3b1表达量在33~42 dpf时维持在较高水平,在38 dpf时出现高峰;三倍体Hsd3b1表达量在47~59 dpf时较高,在49 dpf出现高峰。二倍体中Cyp11b2在37 dpf出现峰值,之后开始下调;三倍体在40 dpf出现峰值,之后逐渐下调,但三倍体Cyp11b2表达量显著低于同期二倍体。二倍体Cyp17a1的表达量在35~46 dpf时逐渐上升,在45 dpf时达到高峰之后直至69 dpf逐渐下降,并且维持在较为平稳的水平上;但是在相同的实验条件下未检测到同一时期三倍体Cyp17a1的表达量。酶联免疫结果显示,在40 dpf时二者的脑芳香化酶活性到达高峰,但在40~60 dpf时期,二倍体虹鳟脑芳香化酶活性显著(P0.05)高于三倍体虹鳟,尤其在45~50 dpf时,该酶活性分别较三倍体的高1.15倍和1.12倍。以上结果表明三倍体虹鳟早期性腺发育迟缓的原因之一是Cyp19a1b、Cyp11a1、Cyp11b2、Cyp17a1、Hsd3b1等基因的表达晚于二倍体,且表达量低于二倍体,造成雌二醇不能正常合成,最终导致性腺发育迟缓。  相似文献

The nonnative rainbow trout Oncorhynchus mykiss has been an unsuccessful invader in North European streams, although it has been widely introduced. Here we studied whether early life history stages (egg incubation and hatching, first overwintering) act as filters for the establishment of hatchery rainbow trout. Survival of hatched alevins was approximately 80%, whereas only 47% of the embryos survived. However, the latter value was impacted by the high number of unfertilized eggs. Correlation coefficients with embryo survival rate and environmental variables (pH and temperature) were statistically insignificant. In the overwintering experiments, the survival of rainbow trout was 93%. The growth was generally slowed during the winter, but in the spring the growth of rainbow trout exceeded that of the native brown trout. Our data demonstrated that the survival and growth of rainbow trout during early life-history stages were relatively high and comparable to those of the native brown trout. Based on the variables considered in our study, our results suggest that environmental conditions during early life-history stages are not detrimental for rainbow trout in the study streams.  相似文献

Early life history traits of fish are very variable as a result of both genetic and environmental factors. In this study, we examine individual variation in early life history traits in progeny of seven females crossed with one male rainbow trout, Oncorhynchus mykiss (Walbaum). Individual eggs were followed from fertilization through hatching until death of the larvae by starvation. Larvae and yolksac size (size was shown to be a good indicator of energy content) increased with increasing egg size, but there were still differences between families after variation in egg size was accounted for. Incubation time was not correlated with egg size, and did not differ between families. The progeny from the different families utilized the available energy differently, as both longevity and growth-rate without food was independent of yolk-sac size, but strongly dependent on family. The observed between-family differences in early life history traits in rainbow trout were mainly caused by genetically based effects; egg size and thus probably egg quality, which differed strongly bet ween females, could not adequately explain these differences.  相似文献

Inhibitory concentrations of clove oil and ethanol against growth of Saprolegnia sp. hyphae were screened by a modification of the hemp (Cannabis sativa L.) seed MicroPlate (HeMP) method and their usability as antifungal agents during incubation of rainbow trout Oncorhynchus mykiss eggs was tested. In vitro experiment showed that in continuous static exposure, clove oil at 100 mg L?1 significantly inhibited the growth of Saprolegnia, whereas in bath exposures, clove oil at 500 mg L?1 had no significant effect at any exposure time tested (15, 60 and 240 min), but clove oil at 10 000 mg L?1 significantly inhibited growth at all exposure times. Clove oil and ethanol treatments had no visible effects on the onset or spread of the fungus during incubation of rainbow trout eggs. Clove oil at 1000 mg L?1 resulted in 95–100% mortality before the eyed stage was reached. Sublethal concentrations of clove oil and ethanol had no effects on the development rate of the embryo or growth and yolk utilization efficiency after hatching. This study suggests that clove oil and ethanol may not be options in controlling aquatic fungi infestations during incubation of rainbow trout eggs.  相似文献

The aim of this study was to evaluate probiotic properties and the aflatoxin B1 adsorption ability of yeasts isolated from rainbow trout intestine and fish feed to assess their use in the formulation of feed additives. Growth at pH 2, bacterial pathogens inhibition, bacterial pathogens co‐aggregation, autoaggregation, homologous and heterologous inhibition against lactic acid bacteria were evaluated. Moreover, aflatoxin B1 adsorption was tested. All strains were able to maintain viable (107 cells/ml) at pH 2. All strains isolated from intestine were identified as Kazaschtania exigua, while strains isolated from feed were all identified as Debaryomyces hansenii. Kazaschtania exigua RC035 and RC037 showed the strongest antimicrobial activity while K. exigua RC037 and RC038 were the most efficient co‐aggregating bacterial pathogens. All strains exhibited strong autoaggregation. None of the tested yeast strains showed homologous inhibition towards other yeasts and heterologous inhibition towards lactic acid bacteria strains. Debaryomyces hansenii RC031 demonstrated aflatoxin B1 adsorption capacity (21%). The results of the present study indicate that select strains of Kazaschtania exigua and D. hansenii showed potential to improve the health of rainbow trout by inhibiting pathogens and binding AFB1 and their use as probiotics may improve the production of rainbow trout in aquaculture systems.  相似文献

The parasitic ciliate Ichthyophthirius multifiliis has a low host specificity eliciting white spot disease (WSD) in a wide range of freshwater fishes worldwide. The parasite multiplies rapidly whereby the infection may reach problematic levels in a host population within a few days. The parasite targets both wild and cultured fish but the huge economic impact of the protozoan is associated with mortality, morbidity and treatment in aquacultural enterprises. We have investigated the potential for genetic selection of WSD-resistant strains of rainbow trout. Applying the DNA typing system Affymetrix® and characterizing the genome of the individual fish by use of 57,501 single nucleotide polymorphisms (SNP) and their location on the rainbow trout chromosomes, we have genetically characterized rainbow trout with different levels of natural resistance towards WSD. Quantitative trait loci (QTL) used for the selection of breeders with specific markers for resistance are reported. We found a significant association between resistance towards I. multifiliis infection and SNP markers located on the two specific rainbow trout chromosomes Omy 16 and Omy 17. Comparing the expression of immune-related genes in fish—with and without clinical signs—we recorded no significant difference. However, trout surviving the infection showed high expression levels of genes encoding IgT, T-cell receptor TCRβ, C3, cathelicidins 1 and 2 and SAA, suggesting these genes to be associated with protection.  相似文献

Triploid rainbow trout exhibit improved survival and extended growth during sexual maturation, compared to their diploid counterparts. However, there have been few benefits demonstrated prior to sexual maturation. This study was undertaken to investigate the possibility of improving growth and survival parameters in triploids through interstrain crosses. Triploids were induced by heat shocking fertilized eggs from intra- and interstrain crosses of two rainbow trout strains. The four triploid groups and their diploid counterpart groups were reared to 233 days post-hatching and analyzed for growth and survival characteristics. Compared with diploids, triploids had significantly (P < 0.05) higher mortalities during the first 100 days post-fertilization, primarily just prior to and after hatching. However, during the remainder of the study triploids exhibited significantly (P < 0.05) lower mortalities than diploids. During the first 50 days of rearing all four triploid groups were significantly shorter and lighter than their diploid counterparts. The growth of the triploid groups later in the study varied considerably. At the conclusion of the rearing phase, one interstrain triploid group was significantly (P < 0.05) longer than its diploid counterpart, although not significantly heavier. The other triploid groups were either significantly smaller than, or equal to the diploids. Analysis of variance indicated that the growth of triploid rainbow trout was significantly affected by maternal strain effects. These results suggest that the use of specific strains and crosses may improve the growth of triploid rainbow trout.  相似文献

The infectious salmon anaemia virus (ISAV) has not been observed to cause natural disease in farmed rainbow trout, Onchorhynchus mykiss (Walbaum), but may cause high mortality in farmed Atlantic salmon, Salmo salar L. In this study, ISAV was passaged 10 times in succession by intraperitoneal injections of serum from previous passage into naïve rainbow trout. The serum viraemia was monitored by real‐time qPCR. The rainbow trout in this study became infected but did not develop ISA. No clinical signs were observed in the rainbow trout in any passage, but replication of ISAV was detected from Day 4 post‐infection (p.i.). Neither increased relative virus loads nor histopathological and immunohistochemical findings consistent with ISA were observed. However, the expression of interferon type I and Mx genes were slightly up‐regulated in the hearts of some individual fish at day 17 p.i. Sequencing of all open reading frames in the ISAV genome of the 10th passage revealed two nucleotide mutations, one in segment 6 coding for the haemagglutinin–esterase (HE) and one in segment 1 coding for the basic polymerase 2 (PB2). The mutation in HE resulted in an amino acid substitution T/K312.  相似文献

The nutritional value of the kernel meals of three species of lupin (Lupinus albus, L. angustifolius and L. luteus) was compared against each other and solvent extracted soya bean meal and wheat gluten, when fed to either rainbow trout or red seabream. The digestible nutrient and energy values were determined for each ingredient, based on the diet substitution digestibility method. Each test ingredient was included in test diets at 300 g kg?1. As different faecal collection methods were used on each species, only ingredient comparisons within each fish species study are valid. Protein digestibility for all lupin kernel meals was significantly better than for soya bean meal when fed to red seabream, although only numerically so when fed to rainbow trout. The highest protein digestibility in both fish species was that from the wheat gluten (100.0% for both rainbow trout and red seabream). Energy digestibility for each of the lupin kernel meals (range 62.4%–64.9% in rainbow trout and 60.9%–69.5% in red seabream) was less than that obtained from soya bean meal (75.1% and 81.0% for rainbow trout and red seabream, respectively). However, the higher gross energy content of the lupin kernel meals still resulted in both L. albus and L. luteus providing equivalent levels of digestible energy as that of soya bean meal. Organic matter digestibility was generally poorer for each of the lupin kernel meals relative to that for the soya bean meal and particularly compared with the wheat gluten. Phosphorus digestibility was significantly better in all lupin kernel meals than that from both the soya bean meal and wheat gluten. Highest phosphorus digestibility was that obtained from L. albus kernel meal (100.0% for both rainbow trout and red seabream) although notably the phosphorus digestibility of all lupin kernel meals when fed to rainbow trout was determined at 100%. Similarly the digestibility of phosphorus from lupin kernel meals fed to red seabream was also very high. This study supported the good nutritional value to both rainbow trout and red seabream of kernel meals of all three species of lupin, although the digestible nutrient attributes of kernel meal from L. luteus were particularly favourable.  相似文献

Heat shock proteins (Hsps) can promote integrity of damaged intestinal epithelial tissues. The aim of this study was to determine the effects of a commercially available anti‐phospholipase A2 antibody on growth, feed efficiency, Hsp expression, and intestinal health of rainbow trout fed diets containing high levels of soybean meal (SBM). Six practical‐type diets (45% protein; 15% fat) were formulated to contain 0, 17.5, or 35% SBM with and without antibody. Each diet was fed to three replicate tanks of fish (20 fish/tank; initial weight 21 g) for 9 wk. RNA was isolated from liver and intestinal tissues (n = 3/tank) to detect alterations in Hsp 27, Hsp 70, and Hsp 90 expression. Fish fed a diet containing 35% SBM had decreased growth and intestinal damage. Hsp 27 and Hsp 90 mRNA was elevated in fish fed 35% SBM. Fish fed 35% SBM with antibody had lower Hsp 90 mRNA expression than fish without antibody. These data indicate that feeding rainbow trout high levels of SBM alters the expression of some Hsps and anti‐phospholipase A2 antibody supplementation does not normalize growth or prevent distal enteritis in fish fed 35% SBM.  相似文献

Although non‐native species can sometimes threaten the value of ecosystem services, their presence can contribute to the benefits derived from the environment. In the Great Lakes, non‐native brown trout (Salmo trutta) and rainbow trout (Oncorhynchus mykiss) support substantial recreational fisheries. With current efforts underway to restore once‐native Atlantic salmon (Salmo salar) to Lake Ontario, there is some concern that Atlantic salmon will impede non‐native contributions to the recreational fishery because Atlantic salmon exhibit niche overlap with brown trout and rainbow trout, particularly during the juvenile life stage. We therefore examined competition and growth of juvenile Atlantic salmon, brown trout and rainbow trout in semi‐natural streams. We found that brown trout were the most dominant and had the greatest growth rate regardless of what other species were present. Rainbow trout were more dominant than Atlantic salmon and consumed the most food of the three species. However, in the presence of brown trout, rainbow trout fed less frequently and exhibited negative growth as compared to when the rainbow trout were present with only Atlantic salmon. These data suggest that, outside of density‐dependent effects, Atlantic salmon will not impact stream production of brown trout and rainbow trout.  相似文献

通过生理性解偶联调节氧化磷酸化效率可能是进化早期发展起来的一种普遍策略,对生命的存在至关重要. 解偶联蛋白(uncoupling protein,UCP)家族是位于线粒体内膜的转运蛋白,哺乳动物基因组中已发现5个成员,其中UCP1~UCP3紧密相关,而UCP4和UCP5(即BMCP1,brain mitochondrial carrier protein-1)与它们差异较大. UCP1~UCP4不仅在哺乳类、鸟类等恒温脊椎动物中普遍存在,而且在鱼类等变温动物中也被证实存在,但UCP5目前仅在哺乳类中被发现. 哺乳动物褐色脂肪组织的UCP1,介导呼吸链产生的质子梯度泄漏,使氧化产生的能量以热的形式散发,但鱼类等变温动物UCP1的功能尚待进一步研究确立. UCP2和UCP3在产热、控制活性氧生成、脂肪酸氧化以及肥胖、糖尿病的发生中均发挥重要作用,二者的转录调节较复杂. UCP4和UCP5与其他UCP关系较远,UCP4仅在脑中表达,而UCP5在脑中高表达,它们的功能还不十分明确,但研究表明很可能与UCP2和UCP3的功能相似.  相似文献

Laboratory and in‐stream enclosure experiments were used to determine whether rainbow trout Oncorhynchus mykiss influence survival of longnose dace Rhinichthys cataractae. In the laboratory, adult rainbow trout preyed on longnose dace in 42% of trials and juvenile rainbow trout did not prey on longnose dace during the first 6 h after rainbow trout introduction. Survival of longnose dace did not differ in the presence of adult rainbow trout previously exposed to active prey and those not previously exposed to active prey ( = 0.28, = 0.60). In field enclosures, the number of longnose dace decreased at a faster rate in the presence of rainbow trout relative to controls within the first 72 h, but did not differ between moderate and high densities of rainbow trout (F2,258.9 = 3.73, = 0.03). Additionally, longnose dace were found in 7% of rainbow trout stomachs after 72 h in enclosures. Rainbow trout acclimated to the stream for longer periods had a greater initial influence on the number of longnose dace remaining in enclosures relative to those acclimated for shorter periods regardless of rainbow trout density treatment (F4,148.5 = 2.50, = 0.04). More research is needed to determine how predation rates will change in natural environments, under differing amounts of habitat and food resources and in the context of whole assemblages. However, if rainbow trout are introduced into the habitat of longnose dace, some predation on longnose dace is expected, even when rainbow trout have no previous experience with active prey.  相似文献

The effect of cortisol on the in vitro metabolism of [3H]17-hydroxyprogesterone ([3H]17OHP) was studied during embryonic development of Arctic charr (Salvelinus alpinus) and rainbow trout (Oncorhynchus mykiss). In the absence of cortisol, rainbow trout embryos metabolized [3H]17OHP largely to androstenedione (A4) and androstenetrione (11-KA) with a minor conversion to 17,20ß-dihydroxy-4-pregnen-3-one (17,20P). In the presence of cortisol, this biosynthesis was inhibited. On the other hand, cortisol had no apparent inhibitory effect on the nature of metabolism of [3H]17OHP by Arctic charr embryos. In these embryos [3H]17OHP was metabolized mainly to 17,20P with a minor conversion to A4 and without the formation of 11-KA that was seen in rainbow trout.When incubated in the presence of [3H]cortisol both Arctic charr and rainbow trout embryos produced 11ß-hydroxyandrostenedione (11ß-OHA) as the major metabolite, with a minor conversion to an unknown steroid. The catabolism of the cortisol by salmonid embryos may reflect the ability of the embryo to inactivate or detoxify cortisol to protect itself from the adverse effects of this biologically potent steroid hormone The study indicates the existence of species-specific differences in the nature of metabolism of [3H]17OHP and the inhibitory effect of cortisol on this metabolism.  相似文献

Abstract— Due to species introductions, brook charr (Salvelinus fontinalis) and rainbow trout (Oncorhynchus mykiss) occur together in many North American streams and typically exhibit a pattern of distribution in which brook charr numerically dominate headwaters and rainbow trout dominate downstream reaches. It has been suggested that 1) the two species compete or 2) the two species do not compete because they are differentially adapted to environmental conditions found in upstream and downstream zones. We assessed whether there were differences in growth and macrohabitat (pool, run and riffle) selection of brook charr and rainbow trout in upper, middle and lower stream zones of a small Pennsylvania stream. Brook charr and rainbow trout placed in replicate paired enclosures set in upstream and downstream reaches showed no significant differences in growth and survival rates upstream, but brook charr had significantly greater growth rates than rainbow trout downstream. Enclosed fish and free-ranging fish both had negative growth rates during the summer. Enclosed fish lost significantly less weight than free-ranging fish. Instantaneous growth rates of free-ranging adult brook charr and rainbow trout from May to August were negative for both species in all stream zones. Underwater observations of adult brook charr and rainbow trout showed both species occurred significantly more often in pool macrohabitats than expected on the basis of macrohabitat availability, except for rainbow trout in the upstream zone. The proportion of pool macrohabitat was not significantly different among stream zones. Brook charr do not appear to be better adapted to upstream environments in Powdermill Run based on growth, survival and macrohabitat selection during summer. Negative biotic interactions acting along with differential environmental adaptations may explain the pattern of distribution of brook charr and rainbow trout in streams, but long-term transplant experiments with additional life stages will be necessary to examine this hypothesis.  相似文献

Major histocompatibility complex (MHC) loci encode glycoproteins that bind to foreign peptides and initiate immune responses through their interaction with T cells. MHC class II molecules are heterodimers consisting of α and β chains encoded by extremely variable genes; variation in exon 2 is responsible for the majority of observed polymorphisms, mostly concentrated in the codons specifying the peptide‐binding region. Lactococcus garvieae is the causative agent of lactococcosis, a warm‐water bacterial infection pathogenic for cultured freshwater and marine fish. It causes considerable economic losses, limiting the profitability and development of fish industries in general and the intensive production of rainbow trout, Oncorhynchus mykiss (Walbaum), in particular. The disease is currently controlled with vaccines and antibiotics; however, vaccines have short‐term efficacy, and increasing concerns regarding antibiotic residues have called for alternative strategies. To explore the involvement of the MHC class II β‐1 domain as a candidate gene for resistance to lactococcosis, we exposed 400 rainbow trout to naturally contaminated water. One single nucleotide polymorphism (SNP) and one haplotype were associated with resistance (P < 0.01). These results are promising for using MHC class IIβ as a molecular marker in breeding rainbow trout resistant to lactococcosis.  相似文献

The early humoral responses of rainbow trout, Oncorhynchus mykiss (Walbaum), and brook trout, Salvelinus fontinalis (Mitchill), with sterile inflammation induced by intraperitoneal Lipogen Triple vaccination were compared to determine if genetic differences in susceptibility to furunculosis in salmonids correlated with different acute phase responses to vaccination. Similar severe acute sterile peritonitis occurred in response to Lipogen Triple in both species. Both species also had a rapid transient reduction in plasma iron concentration at 3 days. Moderate hypoferraemia persisted to day 14 in brook trout, but returned to normal by day 7 in rainbow trout. Plasma zinc decreased sharply 3 days after vaccination in rainbow trout and returned nearly to control levels by day 10; however, plasma zinc did not change in brook trout. Two-dimensional sodium dodecyl sulphate polyacrylamide electrophoresis of plasma proteins revealed that increased amounts of a 48-kDa protein group coincided with the hypoferraemic response in rainbow trout. In addition, a modest elevation in a 16-kDa protein group also occurred in rainbow trout. These studies demonstrated the rapid changes in plasma iron in both species and mild elevation of two putative acute phase plasma proteins associated with vaccine-induced inflammation in rainbow trout.  相似文献
