Noncycling tumor cells: mitogenic response to antilymphocytic serum   总被引:2,自引:0,他引:2  
After continuous labeling with tritiated thymidine for a period several times the cell generation time, some ELD ascites cells remained unlabeled. Despite continued exposure to tritiated thymidine, unlabeled mitoses appeared promptly after administration of mouse antilymphocytic serum. Immunosuppression released some noncycling G(2) tumor cells into mitosis.  相似文献

The antigen receptor of lymphocytes destined to form antibody appears to have the characteristics of the immunoglobulin produced. Antibody directed against the combining region of this immunoglobulin should interact with the combining region of the cell receptor for the antigen. Purified Lewis rat alloantibody prepared against Brown Norway (BN) rat histocompatibility antigens was used to immunize L x BN F(1) hybrids. The resultant antiserum has anti-receptor activity because (i) it yields precipitin lines in gel diffusion when reacted against the immunizing alloantibody; (ii) it inhibits the hemagglutinin antibody response of Lewis rats to BN histocompatibility antigens; and (iii) it inhibits the local graft-versus-host response of Lewis lymphoid cells against BN antigens. This suggests that antireceptor antibody may inhibit cell-mediated responses as well as antibody responses to histocompatibility antigens and may play a role in the regulation of immune responses to such antigens.  相似文献

A local invasive-destructive reaction typical of that seen in allograft rejection occurs when Lewis rat spleen cells are inoculated under the capsule of Lewis kidney freshly grafted into F(1) hybrid hosts. Thus the donor lymphoid cells can be immunogenically stimulated by circulating host leukocytes and the interaction of these two cell populations results in nonspecific damage to kidney parenchyma. The results indicate that passenger leukocytes in organ allografts may be important immunogenic agents.  相似文献

Epithelial cells respond to physicochemical damage with up-regulation of major histocompatibility complex-like ligands that can activate the cytolytic potential of neighboring intraepithelial T cells by binding the activating receptor, NKG2D. The systemic implications of this lymphoid stress-surveillance response, however, are unknown. We found that antigens encountered at the same time as cutaneous epithelial stress induced strong primary and secondary systemic, T helper 2 (T(H)2)-associated atopic responses in mice. These responses required NKG2D-dependent communication between dysregulated epithelial cells and tissue-associated lymphoid cells. These data are germane to uncertainty over the afferent induction of T(H)2 responses and provide a molecular framework for considering atopy as an important component of the response to tissue damage and carcinogenesis.  相似文献

The cytomorphological reactions of lymphoid tissue and the immunobiological status of the cattle organism in the prenatal and postnatal periods were analyzed. The cytomorphological analysis was carried out by counting the lymphoid and plasma cells in 50 fields of vision according to the method in [10]. The level of metabolic processes in the experimental animals was determined by the content of the total protein in their blood serum estimated by refractometry [11]. Protein fractions were described with the electrophoresis in agar gel according to [12]. The immunoelectrophoresis was performed as described in [13]. The main shifts in the cellular composition of the lymph nodes and spleen, as well as other lymphoid tissues in early fetuses, were expressed in the accumulation of lymphoblasts, which were large basophilic cells. Electrophoretic separation and immune-based visualization of experimental animals' serum proteins were carried out simultaneously, and the phenomenon of complete antigenic identity was observed with both immune sera against cows’ and bulls’ blood proteins. From 18 to 22 precipitates were detected on their immunophoregrams.  相似文献

Mixed lymphocyte reactions and tissue transplantation tolerance   总被引:2,自引:0,他引:2  
The induction of tolerance of Lewis histocompatibility antigens in BN rats inoculated at birth with BNI Lewis F(1) hybrid bone marrow cells, as revealed by the prolonged survival of Lewis skin grafts, is accompanied by markedly decreased reactivity in the mixed lymphocyte interaction. Blood lymphocytes from animals inoculated with lymph node cell suspensions also display diminished proliferative reactivity to hybrid BN/Lewis cells in the interaction. However, these recipients are not tolerant of Lewis skin grafts. Blood lymphocytes from BN rats inoculated neonatally with Lewis thymocytes fail to display any level of unresponsiveness in vitro, and such animals are not tolerant of Lewis skin grafts. The results suggest that in rats skin and marrow cells have histocompatibility antigens that are absent or poorly expressed on lymph node cells and thymocytes.  相似文献

Ribonucleic acid extracts obtained from lymph nodes of immunized rabbits homozygous for the b(4)or b(5)allele of light chain immunoglobulin allotypes were injected intravenously into nonimmunized rabbits homozygous for the alternate allele. Approximately 30 percent of the plaque-forming cells in the spleen yielded plaques with immunoglobulin M antibody possessing the allotype of the RNA donor. The allotype of the RNA donor was also found in the IgG immunoglobulin of lymphoid cell lysates as well as in the IgG isolated from the serum. These results suggest that the injected RNA has an informational role in the in vivo synthesis of immunoglobulins by host lymphoid cells.  相似文献

The antibody response of young rabbits to the injection of killed Brucella abortus was abolished by prior extirpation of intestinal lymphoepithelial tissues combined with lethal xirradiation and reconstitution with rabbit fetal liver cells. Extirpative surgery alone, like lethal x-irradiation and reconstitution without surgery, did not abolish this response. Intestinal lymphoepithelial tissues of rabbits apparently play a central lymphoid function and are responsible for the differentiation of the lymphoid cell line which produces specific antibody and immunoglobulins.  相似文献

The blastogenic response of human lymphoid cells toward any individual's transplantation antigens can be deleted by the addition of tritiated thymidine of high specific activity during the incubation of the lymphoid cells in mixed leukocyte culture. After the immunocompetent clones which responded to histocompatibility antigens had been destroyed, the remaining population still retained its capacity to respond to unrelated antigens, including other transplantation antigens.  相似文献

The mechanism by which plasmacytomas (PC) depress the primary immune response to sheep red cells was investigated by determining the ability of normal spleen cells to produce antibody when enclosed in Millipore chambers and implanted in PC-bearing mice. Chamber-enclosed normal spleen cells implanted in PC-bearing mice responded poorly to the sheep red cells when compared to similar cells enclosed in chambers and implanted in normal mice or in mice with other lymphoid and nonlymphoid tumors. The data suggest that PC-induced immune suppression is mediated by a humoral factor.  相似文献

The immune response is initiated in organized lymphoid tissues where antigen-loaded dendritic cells (DCs) encounter antigen-specific T cells. DCs function as packets of information that must be decoded by the T cell before an appropriate immune response can be mounted. We discuss how the dynamics of DC-T cell encounter and the mechanism of T cell differentiation make the decoding of this information stochastic rather than determinate. This results in the generation of both terminally differentiated effector cells and intermediates that play distinctive roles in protection, immunoregulation, and immunological memory.  相似文献

Stable lymphoid cell lines expressing the human immunodeficiency virus type 1 (HIV-1) nef gene product, p27, were established. The presence of p27 in the lymphoid cells suppressed replication of some strains of both HIV-1 and HIV-2. This observation indicates that nef could be important in the establishment of HIV latency. In contrast, fast replicating and highly cytopathic HIV-1 isolates recovered from patients with advanced disease states were not affected by the negative effect of nef present in these lymphoid cell lines. This lack of response to nef appears to constitute another viral feature that correlates with disease progression. Thus, manipulating expression of the nef gene in vivo might influence pathogenesis in the host.  相似文献

Human bone marrow lymphoid cells, particularly when enriched with plasma cells, as in multiple myeloma, respond to pokeweed mitogen and to antiserum to immunoglobulin but not to phytohemagglutinin. Cells of patients with X-linked agammaglobulinemia of the bursal deficient type failed to respond to either pokeweed or to the antiserum to immunoglobulin. Leukocytes of the agammaglobulinemia patients however responded in a normal fashion to phytohemagglutinin. Just as the in vitro response to phytohemagglutinin is taken as an index of the thymus-dependent system, the in vitro response to both antiserum to immunoglobulin and pokeweed may be considered an index for the bursaldependent system. Human bone marrow, therefore, contains bursal cells and probably very few or no thymus cells.  相似文献

Many intracellular pathogens infect a broad range of host tissues, but the importance of T cells for immunity in these sites is unclear because most of our understanding of antimicrobial T cell responses comes from analyses of lymphoid tissue. Here, we show that in response to viral or bacterial infection, antigen-specific CD8 T cells migrated to nonlymphoid tissues and were present as long-lived memory cells. Strikingly, CD8 memory T cells isolated from nonlymphoid tissues exhibited effector levels of lytic activity directly ex vivo, in contrast to their splenic counterparts. These results point to the existence of a population of extralymphoid effector memory T cells poised for immediate response to infection.  相似文献

Mounting a protective immune response is critically dependent on the orchestrated movement of cells within lymphoid organs. We report here the visualization, using major histocompatability complex class I tetramers, of the CD8-positive (CD8) T cell response in the spleens of mice to Listeria monocytogenes infection. A multistage pathway was revealed that included initial activation at the borders of the B and T cell zones followed by cluster formation with antigenpresenting cells leading to CD8 T cell exit to the red pulp via bridging channels. Strikingly, many memory CD8 T cells localized to the B cell zones and, when challenged, underwent rapid migration to the T cell zones where proliferation occurred, followed by egress via bridging channels in parallel with the primary response. Thus, the ability to track endogenous immune responses has uncovered both distinct and overlapping mechanisms and anatomical locations driving primary and secondary immune responses.  相似文献

Kidneys were transplanted across a major genetic barrier (Ag-B locus), from LewisxBN F(1) hybrid rats into bilaterally nephrectomized Lewis rats. Survival of grafts is prolonged (indefinite?) in rats treated with a combination of (i) intravenous injection of donor spleen cells 1 day before the graft, and (ii) passive immunization with antiserum prepared in rats of the recipient strain against donor spleen and lymph-node cells. Dendritic cell-induced memory T cell activation in nonlymphoid tissues   总被引:1,自引:0,他引:1  
Secondary lymphoid organs are dominant sites of T cell activation, although many T cells are subsequently retained within peripheral tissues. Currently, these nonlymphoid compartments are viewed as sites only of effector T cell function, without the involvement of renewed induction of immunity via the interactions with professional antigen-presenting cells. We describe a method of reactivation of herpes simplex virus to examine the stimulation of tissue-resident T cells during secondary challenge. The results revealed that memory CD8+ T cell responses can be initiated within peripheral tissues through a tripartite interaction that includes CD4+ T cells and recruited dendritic cells. These findings lend evidence for the existence of a sophisticated T cell response mechanism in extra-lymphoid tissues that can act to control localized infection.  相似文献

Transfer of lymphoid cells from strain-2 guinea pigs immunized to bovine gamma globulin into syngeneic recipients immunized with dinitrophenyl ovalbumin markedly enhances the secondary antidinitrophenyl response of the recipient to challenge with dinitrophenyl-bovine gamma globulin. This function of the carrier bovine gamma globulin-specific cells is resistant to irradiation with up to 5000 rads, although the capacity of the irradiated cell population to transfer immunologic memory for bovine gamma globulins or to be stimulated by antigen to synthesize DNA in vitro is abolished by this treatment.  相似文献
