The metabolism of [(14)C] phenoxyacetic acid (POA) and the formation of bound residues were studied in soybean leaves and stems. POA was metabolized to 4-HO-POA and to 4-HO-POA glucoside, and a significant fraction of the radioactivity was incorporated in the cell walls (CW). An extraction procedure of CW polymers was developed to specifically isolate the radioactivity associated with each of them. In leaves, the radioactivity showed a preferential distribution into the hemicelluloses and lignins, while pectins and lignins were the most radioactive CW polymers in stems. The identified bound metabolites were 4-HO-POA, POA, and phenolic residues. The latter and POA were essentially incorporated into the lignin fractions and were linked to the benzylic carbons of lignin monomers. 4-HO-POA, the major bound residue, was more evenly distributed in CW polymers. It was esterified to noncellulosic polysaccharides and lignins, but in the latter, contrary to other POA residues, it was mainly linked at the gamma-carbon of propanoid side-chains of lignin monomers. That type of linkage suggested an enzymatic incorporation of 4-HO-POA in CW, contrary to others residues which have an opportunistic lignin incorporation. That incorporation of 4-HO-POA in CW polymers looks like that of endogenous hydroxycinnamic acids.  相似文献

Chufas (Cyperus esculentus) are edible tubers that, like Chinese waterchestnut (CWC), are very crisp when raw and do not soften when cooked. The present study compares the mechanical properties of chufas with those of potato and CWC in relation to the carbohydrate and phenolic compositions of the cell walls. The cutting toughness of raw chufa was higher than that of raw CWC and potato; its value decreased on boiling, as also observed with CWC, but remained over twice that of raw potato. Chufa cell walls were rich in xylose, arabinose, glucose, uronic acid, and galactose, with minor quantities of mannose. The cell walls of the parenchyma exhibited a uniform pH-dependent autofluorescence indicating the presence of cinnamic acid derivatives. Analysis of these revealed that peeled tuber cell walls are rich in ferulic acid, whereas p-coumaric acid dominates the monomeric phenol fraction of the skin. Cell wall material from both skin and peeled tubers contains a significant amount of different diferulic acids ( approximately 20% of the wall ferulic acid), consisting mainly of the 8-O-4'-, 8-5'-, and 5-5'-dimers. These are potentially available to form thermally stable cross-links between polysaccharides within the wall and between cells. This may confer thermal stability of texture.  相似文献

The postharvest changes of the white asparagus cell wall have been studied in relation to the toughening process along the length of a spear that was divided into three sections: apical, middle, and basal. Polysaccharides underwent significant turnover during storage. Uronic acid concentration decreased in all sections and in almost all polysaccharide fractions, while neutral sugars increased very slightly in the apical section, decreased in the middle, and increased in the basal one. Xylose, glucose, and galactose are the main neutral sugars implicated in the turnover; xylose accumulated in the hemicellulose fractions of the middle and basal sections, glucose decreased in the hemicellulose and cellulose fractions of the middle section and increased in the cellulose fraction of the basal one, and galactose disappeared mostly from the cellulose fractions of the three sections. Lignin increased most in the middle section and least in the basal one. No increase was detected in the apical one. No important increases of wall phenolics were detected in any part of the spear. The hardening process was limited only to the basal section.  相似文献

研究耐缺磷程度不同的长豇豆品种在缺磷胁迫下脱落酸(ABA)含量的变化。结果表明缺磷胁迫下长豇豆幼苗根系ABA含量都增高,缺磷前期下降后升至1个高峰后再降低,高峰出现的迟早与品种耐缺磷性有密切关系,不耐缺磷的二芦白比耐缺磷的芦花白出现早,平均升幅香港青二芦白芦花白。缺磷胁迫下长豇豆幼苗老茎叶和最嫩完全展开叶ABA含量升高,平均升幅香港青二芦白芦花白,但缺磷后期ABA含量二芦白香港青芦花白;缺磷胁迫下幼苗嫩茎叶ABA含量各品种都升高,升幅芦花白二芦白香港青。缺磷胁迫下长豇豆幼苗茎叶ABA含量的增加可能促进磷和水分的运输、减轻膜质过氧化,是对缺磷胁迫的适应机制之一。  相似文献

Pore size distributions in the 10/104 nm e.c.d. range in aggregates from three New Zealand soils with largely monomineralic clay fractions, were determined by mercury porosimetry after oven drying and also after critical point drying following methanol and then CO2 exchange from a range of water contents. A soil containing halloysite showed considerable porosity in the fine pores (10–30 nm) regardless of the method of drying. A smectitic soil showed virtually no porosity in the 102–104 nm range when oven dry. A soil containing allophane was dominated by large pores (> 103 nm). The change from a fine (clayey) to a coarser (clay loam) texture within the profile of one soil was reflected in an increase in large pores.  相似文献

Thirty-one peatlands from two areas of central Ontario were sampled to assess the influence of acid deposition on peatland water chemistry. Factor analysis differentiated peatland water chemistry along three major axes of chemical variation, interpreted as axes of organic concentration, mineral concentration, and deposition influence. Water from the surface mats had a higher organic concentration than water from open pools. Mineral influence in peatland waters was reflected by higher concentrations of Ca, Mg, Na, and silica in fen pools compared to bog pools. The influence of high acid deposition in the Wanapitei study area was indicated by high concentrations of sulphate, Ni, Mn, and Cu, and lower pH compared to an area that has received less acidic deposition (Ranger). Regression analyses indicated that H+ variation in bogs could be largely explained by organic C concentration, but that sulphate concentration was also positively associated with acidity, while Ca was negatively associated with acidity.  相似文献

Carrot cell walls have been shown to contain significant quantities of esterified p-hydroxybenzoic acid, which is presumed to be esterified to cell wall polymers. The purpose of this study was to investigate the distribution of p-hydroxybenzoic acid and related phenolics among carrot cell wall polysaccharides. Cell wall material was prepared from fresh carrot root tissues and extracted sequentially with water, imidazole, cyclohexane- trans-1,2-diamine- N, N, N', N'-tetraacetate, Na 2CO 3, and KOH (0.5, 1, and 4 M) to leave a cellulose-rich residue. The fractions were analyzed for their carbohydrate and phenolic acid components. Selected soluble fractions were subfractionated further by graded precipitation in ethanol. The majority of the polymer fractions comprised pectic polysaccharides, with varying quantities of neutral sugars (arabinose and galactose). Hemicellulosic polymers were generally found only in the strong alkali extracts (4 M KOH). p-OH-benzoic acid was the predominant phenolic ester and was associated with most fractions analyzed; p-OH-benzaldehyde was also detected in the fractions at much lower levels. Principal components analysis of the chemical data indicated that the p-OH-benzoic acid was associated predominantly with the branched pectic polysaccharides, in contrast to the p-OH-benzaldehyde. The possible roles and functional properties of these phenolics are discussed.  相似文献

Ecosystems in Finnish Lapland are threatened by heavy metal pollution and acid deposition derived from emissions of Cu-Ni smelters in Kola Peninsula and to varying extent to pollution from southern Fennoscandian and Central European sources. Extensive chemical analyses of small lake waters collected in Finnish Lapland have demonstrated that a significant number of lakes are acidified (ANC < 0μcq/l) or their buffering capacity is critical (ANC = 0–50μe/l). The relative abundance of mafic, ultramafic and carbonate rock components in the catchment were the chief factors controlling ANC and the main base cation (Ca2+, Mg2+) concentrations of lake waters. Both humic and clearwater lakes with low buffering capacity (ANC < 50μeq/l) were mainly located in the catchment areas identified as sensitive to acidification on the basis of low content of base cations (Ca2+ +Mg2+ +K+ < 500 meq/kg) in till. The ratio of the catchment area to the lake area was distinctly smaller for acidic lakes than for the well-buffered lakes, indicating the importance of catchment processes in determining the ANC and main base cations. The high sulphur concentrations (median 60μeq/1) of acidic lakes in northeastern Lapland, near the Finnish-Norwegian border, were strongly correlated with the highest deposition of sulphur derived from smelters of Kola Peninsula. The anomalously high concentrations of sulphur of well-buffered lakes in the western part of Lapland were due to sulphide minerals of soil and bedrock. The acidity of humic lakes in southern Lapland was in large part due to the organic acidity derived from peatlands.  相似文献

The concentration of ferulic acid (FA), the major phenolic acid in the wheat kernel, was found to differ significantly in the mature grain of six wheat cultivars known to have a range of tolerance to the orange wheat blossom midge (Sitodiplosis mosellana). Differences in FA content were correlated with floret infestation level of the cultivars. The wheat cultivars ranked similarly in FA content at the four locations where they were tested, despite a significant effect of environment. Ferulic acid was synthesized mainly during the early stages of grain filling but at different rates among cultivars. Ferulic acid was concentrated primarily in the shorts and bran fractions in an insoluble-bound form. A high correlation was obtained between FA contents as determined by GLC, fluorometry, UV, and colorimetry. The colorimetric procedure was modified as a qualitative, simple, and rapid test for identifying midge-resistant wheat and evaluated in several field trials. The method should provide a rapid tool in the preliminary screening of experimental lines in the development of midge-resistant wheat cultivars.  相似文献

Because pectins are released from potatoes and other plants under conditions that cleave ester linkages, it has been suggested that there are other galaturonoyl ester cross-links between pectin chains in addition to the known non-cross-linking methyl esters. A microscale titration method and a copper binding method were developed for the measurement of total polymer carboxyl (essentially pectic) ester content in potato cell walls. Relative to the uronic acid content of the cell walls, the degree of total esterification was 57-58%. Comparison with levels of methanol released on ester hydrolysis allowed nonmethyl uronoyl esters to be estimated to be 14-15% relative to total uronic acid. The possibility of nonmethyl-esterified linkages being formed in potato cell walls by a side-reaction catalyzed by pectin methyl esterase (PME) was investigated, but no increase in nonmethyl-esterified pectin was observed under conditions where pectin was being effectively de-esterified by endogenous PME activity.  相似文献

Determination of water-insoluble cell walls in feeds: interlaboratory study   总被引:2,自引:0,他引:2  
A collaborative study was conducted to test a new rapid procedure for determination of water-insoluble cell wall (WICW) content in feeds. In the method, starch is solubilized near boiling temperature with Termamyl, a heat-stable alpha-amylase, and proteins are solubilized at 40 degrees C with sodium dodecylsulfate and Pronase. Then, the organic matter of the residue is determined by incineration. Three hours were required to treat 12 different samples, including solubilization treatments, filtrations, and rinses. Eleven unknown products including 9 common feedstuffs of various origin and 2 mixed diets for poultry were analyzed by 7 analysts in France. Coefficients of variation ranged from 2.3 to 6.1%. The results were compared to those for water-insoluble dietary fiber (WIDF), total dietary fiber, and neutral detergent fiber. Agreement was best with the water-insoluble dietary fiber procedure. For most samples, the ratios of WIDF/WICW ranged from 0.981 to 0.842. The differences between WICW and WIDF values correspond to cell wall protein which is accounted for in WICW, but not in WIDF.  相似文献

Determination of glucosamine and N-acetyl glucosamine in fungal cell walls   总被引:1,自引:0,他引:1  
A new method was developed to determine glucosamine (GlcN) and N-acetyl glucosamine (GlcNAc) in materials containing chitin and chitosan, such as fungal cell walls. It is based on two steps of hydrolysis with (i) concentrated sulfuric acid at low temperature and (ii) dilute sulfuric acid at high temperature, followed by one-step degradation with nitrous acid. In this process, chitin and chitosan are converted into anhydromannose and acetic acid. Anhydromannose represents the sum of GlcN and GlcNAc, whereas acetic acid is a marker for GlcNAc only. The method showed recovery of 90.1% of chitin and 85.7-92.4% of chitosan from commercial preparations. Furthermore, alkali insoluble material (AIM) from biomass of three strains of zygomycetes, Rhizopus oryzae, Mucor indicus, and Rhizomucor pusillus, was analyzed by this method. The glucosamine contents of AIM from R. oryzae and M. indicus were almost constant (41.7 +/- 2.2% and 42.0 +/- 1.7%, respectively), while in R. pusillus, it decreased from 40.0 to 30.0% during cultivation from 1 to 6 days. The GlcNAc content of AIM from R. oryzae and R. pusillus increased from 24.9 to 31.0% and from 36.3 to 50.8%, respectively, in 6 days, while it remained almost constant during the cultivation of M. indicus (23.5 +/- 0.8%).  相似文献

The cell wall sap is a very dilute solution with a low and usually negative hydrostatic pressure that decreases further as the moisture stress increases. From a readily acceptable cell model it follows that, under these conditions of water in cell walls, plant cells will develop negative turgor pressures under severe moisture stress. Cell walls are not built to withstand the buckling loads resulting from negative turgor pressures. Consequently, the cell will collapse and shrivel. This is an important pattern of plant injury during severe moisture stress. Plasmolysis is rather an artifact that is observed under laboratory conditions. It will occur whenever solutes accumulate in cell walls and the osmolality in the walls exceeds that of the vacuole. Under natural conditions, this may e. g. occur in plants growing on salt affected soils.  相似文献

Present acid forming emissions to the atmosphere have the potential to alter significantly the chemistry of rain, snow, and surface water of weakly buffered lakes in the Upper Midwest. Average precipitation pH from field measurements during 1979–1983 declined from west to east from 4.8, 4.6, and 4.3 along a cross-section of sites in Minnesota, Wisconsin, and Michigan respectively where 990 lake and stream sampling sites were studied. Measurements of weakly buffered lakes show a parallel decline in lake water pH with the lowest values measured, 5.1, 4.6 and 4.4, respectively in the same regions. Correspondingly, the percentage of lakes sampled with little or no acid neutralizing capacity (ANC) was found to increase from 0 to 4 and 13%, respectively. The geographic patterns in ionic composition of airborne acids and bases, and the resultant surface water concentrations are compared. The acid forming capacity (AFC) from airborne inputs is calculated using mass balance and in-lake processes. Stoichiometric acid-base reactions are used to balance the observed chemical differences between airborne inputs and surface water composition considering nitrification, denitrification, other oxidation-reduction reactions, and the evaporation concentration process. Microbial activity in surface water can result in a net decrease in ionic strength from the conversion of most of the ammonium and nitrate to neutral compounds and biomass, but only a partial reduction of about 20% of the sulfate inputs to weakly buffered lakes. The resulting AFC of airborne inputs are calculated to range from 30 to 60, 50 to 90, and 80 to 130 μeq H+L-l, respectively, in northeastern Minnesota-Ontario, northcentral Wisconsin and northern Michigan-Ontario. The differences in AFC of airborne inputs from west to east, and differences in in-lake processes explain the observed acidity of weakly buffered lakes across the region.  相似文献

Under conditions of salt stress, plants show qualitative and quantitative alterations in various organic compounds, such as nitrogen (N) compounds and organic acids. In this work, the effect of different saline levels as well as various N levels, supplied as nitrate (NO3) or as ammonium (NH4)+NO3 on the concentration of amino acids and organic acids in the leaves of tomato and cucumber plants has been studied. The effect of the source of N on individual amino acid contents varied with plant species. Most of the amino acids increased when the concentration of N in the nutrient solution was increased, except when N was added as NH4+NO3 for tomato. The effect of salt stress depended on which amino acid was considered. The data also indicate that the effect of salinity on each particular amino acid was greatly dependent on the plant species and N source. Organic acids were differently affected by salinity and by the N source, depending on the plant species. In tomato, the concentrations of short‐chain organic acids were 2–3 times higher in NO3‐supplied plants than in those grown with NH4+NO3. Finally, in cucumber, malic acid concentration increased as a function of the saline level in the medium.  相似文献

Ferulic acid bioavailability is dependent on its form present in food. This necessitates a methodology to quantify different groups of ferulic acid derivatives in food products, especially cereal‐based products. The aim of the proposed methodology is to separate and quantify ferulic acid ester‐linked to mono‐ and/or oligosaccharides (OF), to soluble polysaccharides (SPF), and to insoluble polysaccharides (IPF) as well as in its free form. Development and partial validation of this method, which was widely based on liquid/liquid extraction and precipitation steps, was performed using characterized standard materials isolated from corn bran. As the determination of OF was one of the major goals of this methodology, three different feruloylated mono‐ and oligosaccharides were used for method development and validation. To determine the accuracy of the method, ferulic acid–containing standard materials added to a starch matrix were extracted and separated according to the developed protocol. The separated ferulic acid esters were saponified before ferulic acid was analyzed by reversed phase HPLC. Recovery rates were generally between 70 and 103%, with the lowest recovery rates for SPF and highest recovery rates for IPF and OF. Finally, the applicability of the method to unprocessed and processed wheat bran samples was demonstrated.  相似文献

In a lysimeter experiment the overall amino acid concentration in dry matter of ryegrass (Lolium perenne) increased in response to nitrogen application. Lack of water reduced the overall concentration of most amino acids in dry matter except proline the concentration of which was increased. Nitrogen application as well as lack of water significantly affected the overall amino acid composition, and in particular the relative concentration of glutamic acid and proline, respectively, which might be attributed to variations in the free amino acid composition, and resulting in a decreasing overall ratio essential/not essential amino acids.  相似文献

Proanthocyanidins were isolated from the skins of Cabernet Sauvignon grapes at different stages of grape development in order to study the effect of proanthocyanidin modification on the interaction with grape cell wall material. After veraison, the degree of proanthocyanidin polymerization increased, and thereafter was variable between 24 and 33 subunits as ripening progressed. Affinity of skin cell wall material for proanthocyanidin decreased with proanthocyanidin ripeness following veraison. A significant negative relationship (R2=0.93) was found for average proanthocyanidin molecular mass and the proportion of high molecular mass proanthocyanidin adsorbed by skin cell wall material. This indicated that as proanthocyanidin polymerization increased, the affinity of a component of high molecular mass proanthocyanidins for skin cell wall material declined. This phenomenon was only associated with skin proanthocyanidins from colored grapes, as high molecular mass proanthocyanidins of equivalent subunit composition from colorless mutant Cabernet Sauvignon grapes had a higher affinity for skin cell wall material.  相似文献
