Interpretation of laboratory results is based on comparison of the patient's own results against established decision thresholds or reference intervals in the context of the clinical presentation and history. Blood measurand analysis has pre‐analytical, analytical and physiological sources of variation, which may complicate interpretation of results. Biological variation describes the physiological random fluctuation of blood measurands around a homeostatic set point, which varies within and between individuals. This article reviews the practical applications of biological variation in the everyday clinical setting. Examples are offered to highlight how biological variation can be used to: (1) assess the usefulness of subject‐based reference intervals, (2) determine measurand homeostatic set points, (3) interpret single or serial results for diagnosis of disease and (4) evaluate changes in serial results during monitoring.  相似文献

Oxidative stress results from a disruption of the prooxidant/antioxidant cellular balance and monitoring free radical status becomes an interesting challenge in animal and human nutrition. In the present work, merits and limitations of different analytical techniques (HPLC, GC-MS, fluorometric and colourometric assays, ELISA, gel electrophoresis) for the measurement of radical mediated alterations in the cellular integrity of lipids (malondialdehyde, hydrocarbon gases, F2-isoprostanes) proteins (protein carbonyls, 3-nitrotyrosine) and DNA (8-hydroxy-2'-deoxyguanosine) are discussed. Besides these indirect methods, owing to the fact that free radicals are paramagnetic, electron paramagnetic resonance spectroscopy combined with spin trapping has become a valuable tool to directly assess and to better understand the mechanisms of free radical reactions. With this approach a radical that is too short-lived to be detected, adds to a spin-trapping agent to form a relatively long-lived radical adduct. Information obtained from the hyperfine splitting of the spin-trapped adduct can provide identification and quantification of the originally generated free radicals.  相似文献

Common goals of microarray experiments are the detection of genes that are differentially expressed between several biological types and the construction of classifiers that predict biological type of samples. Here we consider a situation where there is no training data. There is considerable interest in comparing expression profiles associated with successful pregnancies (SP) and unsuccessful pregnancies (UP) in model and farm animals. Successful pregnancy rate is known to be much higher in embryos generated by in vitro fertilization (IVF) than in nuclear transfer (NT) embryos, and higher under induced ovulation for large follicles (LF) than for small follicles (SF). The tasks of identifying genes differentially expressed between SP and UP, and predicting SP for future samples are not well accomplished by comparing IVF and NT, or LF and SF. A suitable method is finite mixture model analysis (FMMA), which models each observed class (IVF and NT, or LF and SF) as a mixture of two distributions, one for SP and one for UP, with different known or unknown proportions (here known to be 0.50 SP for IVF and 0.02 SP for NT). The means of the two distributions differ for the differentially expressed genes, which we identify via a likelihood ratio test. We confirm by simulation that FMMA strongly outperforms hierarchical clustering and linear discriminant analysis using the known class labels (NT, IVF). We apply FMMA to a real data set on IVF and NT embryos, and compute their posterior probabilities of SP, which confirm our prior knowledge of the SP proportions for IVF and NT.  相似文献

A pharmacokinetic study of doxycycline after intravenous administration at 5 mg/kg body weight in goats revealed that a concentration of 0.5 g/ml was maintained for 5 min-2 h, 4–12 h, 2–12 h and 5 min- >48 h in plasma, interstitial fluid, milk and urine respectively. The low t1/2 of 0.73±0.11 h and high t1/2 of 16.63±1.58 h show that the drug is rapidly distributed but slowly eliminated from the body. The tissue:plasma concentration of 4.86±1.06 during the elimination phase [K12/(K21-)] indicates a high expected tissue concentration, which is supported by similarly increased drug concentration in interstitial fluid and milk. The high Vdarea of 9.78±0.86 L/kg observed denotes that, apart from its wide distribution, the drug may be stored in fat depots as it is known to be highly lipophilic. As the drug maintained a therapeutic concentration for a shorter time in plasma, and the calculated dose rate for maintaining a minimal plasma concentration of 0.5–1.5 g/ml is relatively high, it may not be of much use in treating septicaemia in this species. Since the observed tissue:plasma concentration was higher and a therapeutic concentration was maintained in interstitial fluid and milk for longer, the drug can be used for other systemic infections at a lower dose rate than that required for treating septicaemia. As the drug maintained a very high concentration in urine, it may be of particular value in treating urinary tract infections caused by sensitive micro-organisms.  相似文献

Interrelationships among milk production, dam-calf feed efficiency to weaning, and other biological traits were evaluated on 411 first-calf heifers and their calves. Individual feed intakes were measured in a drylot environment for a 1-yr period until weaning of the first calf. Heifer breed types included crossbred Angus-Hereford, Simmental-Hereford, and Tarentaise-Hereford produced in two-breed rotational crossbreeding systems, F1 Salers-Hereford and straightbred Hereford. Production efficiency was defined as cumulative feed ME consumed by the dam-calf pair during the year divided by calf weaning weight. Milk production potential seemed to be unrelated to cow size, adjusted for breed-group effects. Lactational feed energy was increased for dams of increased milk production potential but not enough to offset the increased production of calf weaning weight in terms of production efficiency. Results indicated that increased levels of milk production of heifers were associated with improved production efficiency to weaning. However, the incremental improvement in efficiency per unit of increased milk was less for each additional unit of milk, within the range evaluated. These results are applicable to first-calf heifers that have not yet reached peak age for maximum milk production.  相似文献

Fifteen dogs were given doxorubicin, IV, at a dosage of 30 mg/m2 of body surface. A commercially available biological extract of Serratia marcescens (BESM) was administered SC to 9 of these dogs (0.04 mg/kg of body weight every third day, n = 2; 0.08 mg/kg every other day, n = 2; and 0.08 mg/kg daily, n = 5), beginning the day after administration of doxorubicin, in an attempt to find an optimal dosage and schedule of administration of BESM to reduce the duration and severity of chemotherapy-induced myelosuppression. Nine additional dogs were randomized into 3 groups of 3 dogs to receive 1 of the following dosages of BESM SC: 0.08, 0.16, and 0.32 mg/kg. Serum was harvested immediately prior to treatment and at 2, 4, 6, 8, 12, 24, 48, and 72 hours from this latter group of dogs for subsequent analysis of canine granulocyte colony-stimulating factor (G-CSF) by enzyme immunoassay. Increasing the dosage and schedule of administration of BESM reduced the duration and severity of doxorubicin-induced myelosuppression. Neutrophil counts of the group of dogs given BESM daily at a dosage of 0.08 mg/kg and the controls were evaluated statistically. The neutrophil count increased significantly (P < 0.05) above pretreatment values in BESM-treated dogs after day 7. Median neutrophil counts of the BESM-treated dogs were never significantly lower than pretreatment values, whereas the median counts of the dogs treated with doxorubicin alone were significantly below normal for 6 days (days 7-12).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

对化学方法和生物方法制备巨菌草(Pennisetum sp.)腐植酸和黄腐酸进行分析比较。结果表明,硝酸、盐酸、乙酸、草酸、氨水均能降解纤维素、半纤维素或木质素从而生产腐植酸和黄腐酸,其中10.0%氨水提取巨菌草得到的腐植酸含量最高,其次为37.5%硝酸和37.5%盐酸,草酸和乙酸提取得到的腐植酸含量最低。枯草芽孢杆菌(Bacillus subtilis)、毛霉(Mucor)、根霉(Rhizopus)、青霉(Penicillium)在巨菌草腐殖质的形成中均起到了积极作用,其中根霉对木质素的降解程度最高,生成总腐植酸的含量最高。根霉发酵制备得到的总腐植酸含量是10.0%氨水提取得到的总腐植酸含量的1.45倍,毛霉发酵制备得到的总腐植酸含量是10.0%氨水提取得到的总腐植酸含量的1.29倍,而枯草芽孢杆菌和青霉发酵制备得到的总腐植酸含量比10%氨水提取得到的总腐植酸含量低。37.5%硝酸提取得到的黄腐酸含量是毛霉发酵制备得到的黄腐酸含量的1.79倍,22.5%盐酸提取得到的黄腐酸含量是毛霉发酵制备得到的黄腐酸含量的1.98倍。综上可知,生物发酵法制备得到的总腐植酸含量比化学法提取得到的总腐植酸含量要高,而化学方法提取得到的黄腐酸含量比生物发酵法制备得到的黄腐酸含量要高。  相似文献

西北地区深居欧亚大陆腹地,是我国干旱最严重的地区之一,开展卫星遥感资料旱情监测工作,具有重要的现实意义。利用M()DIS合成产品数据MODllA2和M()D13A2获取的归一化植被指数(NDVI)和地表温度(Ts)构建Ts—NDVI特征空间,依据该特征空间计算的温度植被干旱指数(TVDI)作为土壤湿度监测指标,分析了西北地区2007年6—9月每16d的土壤湿度。研究表明,2007年6—9月,我国西北地区虽然没有发生大面积、较严重的旱情,但有较长时间和较大范围的轻旱到中旱。同时,对土壤湿度数据进行验证发现,TVDI指标基本上能够反映表层土壤湿度状况,利用温度植被旱情指数法对西北地区进行夏季干旱动态监测是可行的。  相似文献

We studied lambs' preferences for nonnutritive flavors that were paired with a glucose solution. On the 1st d of the experiment, lambs were offered a saccharin solution flavored with either orange or grape. The following day lambs received a glucose solution containing either orange or grape flavor. Lambs that had received grape and saccharin on d 1 received orange and glucose on d 2, and the reverse was true for the remaining lambs. Conditioning lasted 10 d; odd days were like the first and even days like the second. After conditioning, when lambs were offered a choice between orange- or grape-flavored water without sweeteners, lambs chose the flavor that had been paired with glucose. We also tested the initial hedonic response of naive lambs to the flavor of glucose and saccharin solutions. Lambs exhibited no initial preference. Our results indicate that lambs preferred the flavor that was paired with glucose (calories). Furthermore, results of this study suggest that ruminants may not innately recognize specific chemical constituents in foods or select diets based on initial hedonic value. Rather, learning plays a key role in the formation of dietary preference.  相似文献
