Summary The pre-and post-fertilization barriers in the interspecific crosses between Vigna umbellata and V. minima were investigated. In the reciprocal crosses (V. minima as the parent) the entry of pollen tubes into the ovary was delayed by about 4 h, and no seed set was observed. However, no pre-fertilization barriers were encountered in crosses involving V. minima as the parent and V. umbellata as the parent (normal cross). The delay/absence of divisions in the endosperm and the failure of embryo to divide were the post-fertilization barriers responsible for somatoplastic sterility in normal crosses which yielded a few hybrid seeds. The hybrid seeds showed poor germinability. The F1 hybrids were intermediate between the parents in most morphological characters, and are completely sterile for pollen. Backcrossing of F1 hybrid with both the parents did not restore fertility in the progenies. V. minima is considered as the tertiary gene pool of the rice bean.  相似文献

Bruchid beetles or seed weevils are the most devastating stored pests of grain legumes causing considerable loss to mungbean (Vigna radiata (L.) Wilczek). Breeding for bruchid resistance is a major goal in mungbean improvement. Few sources of resistance in cultivated genepool were identified and characterized, however, there has been no study on the genetic control of the resistance. In this study, we investigated the inheritance of seed resistance to Callosobruchus chinensis (L.) and C. maculatus (F.) in two landrace mungbean accessions, V2709BG and V2802BG. The F1, F2 and BC generations were developed from crosses between the resistant and susceptible accessions and evaluated for resistance to the insects. It was found that resistance to bruchids in seeds is controlled by maternal plant genotype. All F1 plants derived from both direct and reciprocal crosses exhibited resistance to the bruchids. Segregation pattern of reaction to the beetles in the F2 and backcross populations showed that the resistance is controlled by a major gene, with resistance is dominant at varying degrees of expressivity. Although the presence of modifiers was also observed. The gene is likely the same locus in both V2709BG and V2802BG. The resistant gene is considered very useful in breeding for seed resistance to bruchids in mungbean.  相似文献

Summary Restriction fragment length polymorphism (RFLP) analysis has several advantages over traditional methods of genetic linkage mapping, one of these being the starting point for map-based cloning. The recent development of an RFLP map of cowpea (Vigna unguiculata L. Walp) has allowed the investigation of associations between genes of interest and RFLP markers. A cross between an aphid (Aphis craccivora Koch) resistant cultivated cowpea, TT84S-2246-4, and an aphid susceptible wild cowpea, NI 963, was screened for both aphid phenotype and RFLP marker segregation. One RFLP marker, bg4D9b, was found to be tightly linked to the aphid resistance gene (Rac 1) and several flanking markers in the same linkage group (linkage group 1) were also identified. The close association of Rac 1 and RFLP bg4D9b presents a real potential for cloning this insect resistance gene.  相似文献

Summary Extent of heterosis for seed yield is an indicator for transgressive segregation in cross of self-pollinated crops. However, the extent of heterosis is thought to be related to the genetic diversity among the parents. Isoenzymic variation was used for estimating the genetic diversity. Eight varieties of green gram (Vigna radiata) were characterized for esterase, amylases, phosphatases, catalase and peroxidase isoenzymes. Differences in these systems were used to calculate a diversity index (D.I.). Hybrids produced from an inbred pair having higher D.I.'s outyielded those with low D.I.'s. However, the correlation (r) between D.I. and heterosis for seed yield was not consistent in different seasons but some of the desirable crosses could be singled out on the basis of D.I.  相似文献

Inheritance of resistance to Yellow Mosaic Virus (YMV) was studied in crosses of mungbean, black-gram and their interspecific crosses with Vigna sub-lobata. Resistance to YMV was recessive in the three Vigna species. The segregation ratios in F2 and back crosses indicated that the resistance was digenic recessive in the crosses of mungbean and in interspecific crosses of mungbean with blackgram and Vigna subiobata but YMV resistance was monogenic recessive in blackgram crosses.  相似文献

Inheritance of yield and yield contributing characters were investigated using generation mean analysis, utilising the means of six basic populations viz., P1, P2, F1, F2, BC1P1 and BC1P2 in four crosses of Vigna sesquipedalis. The analysis reiterated that the importance of dominance (h) gene effects for pod yield/plant and pods/plant as compared to additive (d) gene effects. However, significant and positive additive effects were noticed for pod yield/plant, pods/plant, pod weight and seed weight in different crosses. The three types of gene interactions (additive, dominance and epistasis) were significantly involved for pods/plant in cross KU 7 ×KU 8. Among the digenic epistatic interactions, both additive ×additive (i) and dominance × dominance (l) contributed more for pod yield/plant and pods/plant, however, it varied among the crosses. Populations having earliness can be developed as indicated by reducing dominance effects. Pedigree selection and heterosis breeding is suggested to exploit the fixable and non fixable components of variation respectively in Vigna sesquipedalis. Summary Physiological and morphological characteristics of the two wild and three domesticated subspecies of cowpeas are compared. The wild accessions are alike in having small, hard seeds borne in dehiscent pods, but differ in other characteristics. We suggest that the wild subsp. dekindtiana, from the seasonally-arid tropics, is more likely to have been the progenitor of modern cowpeas than the other wild subspecies (subsp. mensensis), but that subsp. dekindtiana was first cultivated in the humid tropics where its pods are slow to dehisce. Domestication has been associated with changes in the structure of pod valves and seed coats which reduce pod dehiscence and seed hardness. Pods and seeds have increased in size, mainly by increases in the rate of dry weight accumulation, and their increase has been only partly paralleled by increase in the area of subtending leaves. There has been no increase in the maximum photosynthetic rate of leaves, but the duration of their photosynthetic activity has increased. Domesticates are less sensitive than are wild plants to some environmental controls, such as in the response of germination to temperature, but in their flowering responses to daylength both wild and cultivated forms retain sensitivity under conditions where this is of adaptive value.  相似文献

A recombinant inbred line (RIL) mapping population (F8) was generated by crossing Vigna mungo (cv. TU 94‐2) with Vigna mungo var. silvestris and screened for mungbean yellow mosaic virus (MYMV) resistance. The inter simple sequence repeat (ISSR) marker technique was employed to identify markers linked to the MYMV resistance gene. Of the 100 primers screened, 54 showed amplification of which 36 exhibited polymorphism between the parents TU 94‐2 (resistant) and V. mungo var. silvestris (susceptible). Individual plants from 53 RIL populations were analysed and one marker (ISSR8111357) was identified as tightly linked to the MYMV resistant gene at 6.8 cM. Both the phenotype as well as the ISSR8111357 marker segregated in a 1 : 1 ratio. The ISSR8111357 marker was sequenced and sequence characterized amplified region (SCAR) primers were designed (YMV1‐F and YMV1‐R) to amplify the marker. Screening for the SCAR marker in the RIL population distinguished the MYMV resistant and susceptible plants, agreeing well with the phenotypic data. The ISSR8111357 marker was validated using diverse blackgram genotypes differing in their MYMV reaction. The marker will be useful for the development of MYMV‐resistant genotypes in blackgram.  相似文献

Summary Twenty-one genotypes of cowpea (Vigna unguiculata), comprising landraces and varieties, were grown in 22 photothermal environments in Nigeria and Niger, West Africa, and a stability analysis of days from sowing to flowering (f) was carried out. Cowpeas are rarely insensitive to photoperiod; they are typically quantitative shortday plants wherein f is delayed when photoperiod (P) is longer than the critical photoperiod (P c ). Therefore, in order to quantify genotypic variation in temperature sensitivity, genotype f was regressed against the mean trial f in circumstances where P c (i.e. approximately 13 hd-1) and mean temperature (T) was between 19° and 28° C. Correspondingly, in order to assess genotypic variation in photoperiod sensitivity, trials where T was near optimal (25°–28° C) but where P ranged from 10–14.5 hd-1 were used. These stability analyses detected no significant differences (P>0.05) between genotypes 9n temperature sensitivity but revealed significant differences (P<0.001) in photoperiod sensitivity. Regression coefficients from the stability analysis were strongly correlated (r=0.94, 19df) with a photoperiod sensitivity constant, c, determined from a photothermal flowering model. Controlled crosses in bambara groundnut were attempted between a range of thirty-six bambara groundnut landraces (thirty domesticated (V. subterranea var. subterranea) and six wild (V. subterranea var. spontanea)). Ten F1 seed were produced. Of these, eight germinated producing F2 populations. On seed set, four populations could be unambiguously confirmed as true crosses by F3 seed coat colour. A single F2 population, derived from a domesticated landrace from Botswana (DipC; female parent) crossed with a wild accession collected in Cameroon (VSSP11; male parent) was used to study a range of agronomic and domestication traits. These included; days to emergence, days to flowering, internode (fourth) length at harvest, number of stems per plant, leaf area, Specific Leaf Area (SLA), Carbon Isotope Discrimination (CID), 100 seed weight, testa colour and eye pattern around the hilum. On the basis of variation for internode length and stems per plant, 14 small F3 families were selected and grown under field conditions to further investigate the genetic basis of the ‘spreading’ versus ‘bunched’ plant character, a major difference between wild and cultivated bambara groundnut. Results presented suggest that traits including leaf area, SLA, CID and 100 seed weight are controlled by several genes. In contrast, the variation for traits such as internode length, stems per plant, days to emergence and seed eye pattern around the hilum are likely to be under largely monogenic control. The results of this work are discussed in relation to the domestication of bambara groundnut.  相似文献

Summary The nature and extent of the gene pools in cultigens of the genera Phaseolus and Vigna, as indicated by studies of experimental interspecific hybridization, are reviewed. In all cases a primary gene pool exists comprising conspecific wild and cultivated forms.Secondary gene pools have been established for Phaseolus vulgaris-Ph. coccineus, Vigna radiata-V. mungo and V. angularis-V. umbellata but not for V. unguiculata and for Voandzeia subterranea. Tertiary gene-pools apparently exist for all Phaseolus cultigens and the Asiatic grams but not for V. unguiculata and V. subterranea as far as is known.  相似文献

Summary The mean aim of this study was to identify an effective method of predicting cross-potential in respect of yield improvement of the cowpea (Vigna unguiculata (L.) Walp.) through hybridization and subsequent selection.Three prediction tests based on data collected from experiments with ten cowpea lines and the F1 and F2 generations of all possible crosses between these lines were evaluated. The three prediction tests were based on: i Yields of prospective parents. ii. General combining abilities estimated from F1 and parental data iii. Frequencies of superior plants in F2 populationsEach of the three procedures was used to identify the crosses with the lowest or highest yield potential, i.e. those crosses most likely to produce the greatest number of lines with yields higher or lower than those of either parent. Crosses selected on the basis of these criteria were evaluated in the following growing season on the basis of biometrical analysis of F2 and backcross populations and the performance of the respective F3 and F4 generations.Six different crosses were selected from the 45 tested on the basis of the three prediction tests. Analyses of F2 and backeross populations from each cross provided estimates of [d] the difference in parental means and 1/2D the additive genetic component which, in turn, were used to predict the frequency of transgressive inbred lines obtained by single-seed descent from the F2. Both crosses selected on the basis of F2 data were expected to produce a much higher frequency of transgressive lines than those selected on the basis of parental yields. The high potential cross selected on the basis of general combining ability was expected to produce a higher frequency of transgressive lines than the corresponding low yield potential cross. Replicated trails of F3 bulks of the selected crosses showed that high yield potential crosses were significantly higher yielding than low yield potential crosses for all prediction criteria. In a trial with 22F4 lines of each of the two crosses selected on the basis of parental yields and frequency of high yielding F2 plants, the high yield potential cross in each case had a greater mean and variance for seed yield than the corresponding low yield potential cross.It was concluded that parental yields provide a sound basis for an initial screening of prospective parents. This screening should identify a high yielding line which can be crossed with a number of contrasting lines to produce F2 populations for the final screening phase.  相似文献

Summary The resistance sources among various test cultivars of urdbean to Colletotrichum truncatum, a leaf spotting pathogen, were identified and genetics of resistance was worked out by studying F1, F2 and F3 generations of crosses between resistant cultivars and the susceptible cv. Summary Protoplasts were isolated from leaves of a single plant of Vigna aconitifolia, a drought resistant grain legume. The protoplasts regenerated and formed colony and calli from which 50 entire plants were regenerated and transferred to field conditions. Only 7 plants survived upto maturity and they flowered and produced pods with seed. The protoplast derived plants showed variation in important characters. Two groups of characters (one with 7 sets and another with 6 sets) were studied in the protoclones. In first group protoclones showed variations in seed germination, maturity age, pod length, pod and seed colour, number of abortive seed per pod and response to field rots, however, not much difference was recorded in pollen stainability and meiotic behaviour in these protoclones. In second group analysis of variants showed significance difference for plant height, rachis-length, length and breadth of mature odd leaflets, seed per pod and weight of seed. The results indicate that protoplast can be source of variation in this crop. However, detailed biochemical and genetical analysis of protoclones are required.  相似文献

Summary A dominant or partially dominant gene for sensitivity to photoperiod in mungbean strain PI 180311 was identified and labeled Ps. The gene was expressed when strain P1180311, or crosses involving PI 180311 were grown in 16- or 14-hour photoperiods but was not expressed when the populations were grown in a 12-hour photoperiod or in the field at Columbia, Missouri. Dominance × dominance epistatic effects were indicated as governing days to flower in absence of the obvious effect of the Ps gene. Summary The inheritance of seed weight in cowpea was examined in a field planting of the parents, reciprocal F1s, F2s and backcrosses to both parents of a cross between TVu 1977-OD (small seeded) and ACC 70002 (large).Seed weight was inherited quantitatively and small seed was partially domiminant to large seed size. Gene action was predominantly additive but dominance and additive × additive epistatic effects were also significant.Broad and narrow sense heritabilities were 85.1±5.3% and 75.4±18.6% respectively. The minimum number of loci involved in the inheritance of seed size was eight, and each gene pair contributed up to 1.02 g increase to seed weight. The wild Ceratotropis species which occur in Japan consist of Vigna angularis var. nipponensis, V. nakashimae, V. riukiuensis and V. reflexo-pilosa. The main aim of the present study is to detect cross-compatibility of adzuki bean and rice bean with these wild relatives. The cross of V. nakasbimae with adzuki bean was successful in both directions and viable seeds were produced, while the cross with rice bean was not successful. V. riukiuensis is cross-compatible with adzuki bean and rice bean only when V. riukiuensis is used as male parent and the hybrid seeds showed normal germination. The cross between V. reflexopilosa and the two cultigens adzuki bean and rice bean did not succeed in either direction. The present results suggest that V. nakashimae and V. riukiuensis could be used as gene sources for the adzuki bean breeding programme through interspecific hybridization. Successful hybrids between adzuki bean and rice bean cannot easily be produced. However, since V. riukiuensis is cross-compatible with both adzuki bean and rice bean, V. riukiuensis could be useful as a bridge species between adzuki bean and rice bean.  相似文献

Degenerate oligonucleotides designed to recognize conserved coding regions within the nucleotide binding site (NBS) and hydrophobic region of known resistance (R)genes from various plant species were used to target PCR to amplify resistance gene analogs (RGAs) from a cowpea (Vigna unguiculata L. Walp.) cultivar resistant to Striga gesnerioides. PCR products consisted of a group of fragments approximately 500 bp in length that migrated as a single band during agarose gel electrophoresis. The nucleotide sequence of fifty different cloned fragments was determined and their predicted amino acid sequences compared to each other and to the amino acid sequence encoded by known resistance genes, and RGAs from other plant species. Cluster analysis identified five different classes of RGAs in cowpea. Gel blot analysis revealed that each class recognized a different subset of loci in the cowpea genome. Several of the RGAs were associated with restriction fragment length polymorphisms, which allowed them to be placed on the cowpea genomic map. The potential for using these sequences to isolate R genes, and subsequent direct manipulation of disease and pest resistance using genetic engineering is discussed. Summary Seedcoat colour in greengram (Vigna radiata (L.) Wilczek.) is a useful marker for genetic studies and varietal identification. Its mode of inheritance was examined in five crosses among nine parents which differed for seedcoat colour. Four of the parents had sap green seedcoat colour while the others had raw sienna, brownish green, densely black spotted, black and greenish yellow seedcoat colour, respectively. At the F2 generation, no more than 20 different colour classes were observed. The segregation in F3 and backcross generations indicated that at least five major genes were involved in seedcoat colour inheritance. Sap green seemed to be dominant over raw sienna. The segregation ratios further indicated the role of non-allelic gene interactions (epistasis) in inheritance of seedcoat colour. Gene symbols were assigned to each colour and genotypes to each parent.  相似文献
