The present study investigated the basal levels and GnRH-induced responses of peripheral testosterone and estrogen in Holstein bulls with poor semen quality. On the basis of semen parameters, bulls (n=5) having poor semen quality were selected as experimental bulls, and good semen quality bulls (n=4) were used as control bulls. Both groups were treated intramuscularly once with GnRH (250 μg of fertirelin acetate). Blood samples were collected at -1 day (d), -30 min and 0 h (treatment) followed by every 30 min for 5 h and 1, 3 and 5 d post-GnRH treatment (PGT), and LH, testosterone and estradiol-17β (E(2)) concentrations were measured. The pretreatment concentrations were used as basal levels. The percentage increments based on the 0-h levels were calculated per bull for each sampling time until 5 h PGT, and differences were compared between the experimental and control groups. The PGT concentrations of testosterone and basal and PGT concentrations of E(2) were significantly lower in the experimental group. The testosterone increment in the experimental group was delayed and significantly lower from 1 to 5 h PGT than those in the control group. It can be suggested that bulls with poor semen quality have delayed and lower GnRH-induced testosterone response and may also have lower estrogen levels.  相似文献

In the present study, the correlations among age, body weight, scrotal circumference (SC), semen quality and peripheral testosterone and estradiol-17beta (E(2)) concentrations were investigated in pubertal (n=5) and postpubertal (n=7) groups of Holstein bulls over a 6 week period. There were significant positive correlations (P<0.01) among age, body weight and SC in both groups, and similar significant correlations between sperm motility and SC in pubertal bulls (P<0.01) and between sperm concentration and SC in postpubertal bulls (P<0.05). The sperm motility after collection (P<0.05) and after freezing and thawing (P<0.01) of the postpubertal bulls correlated positively with the E(2) concentration. Estrogen may be important for the function of postpubertal bull testes, in which it may regulate spermatozoa motility in vivo.  相似文献

As electroejaculation (EEJ) is prohibited for use on unanaesthetized animals in Sweden, there is a need for an alternative method of semen collection from bulls in the field. The aim of the present study was to evaluate the use of transrectal massage (TM) of the ampullae to collect semen from yearling beef bulls under field conditions in Sweden. Transrectal massage was performed on 52 yearling beef bulls. Volume of semen collected, duration of procedure, percentage progressively motile sperm, and sperm concentration were measured. Smears were prepared for sperm morphology examination. Semen samples were obtained from 47 of 52 bulls. Mean volume was 3.2 ml (SD +/- 3.7), mean duration of collection was 7.4 min (SD +/- 2.8), mean percentage progressively motile sperm was 43.5% (SD +/- 29.2) and mean concentration was 201.9 x 10(6) spermatozoa/ml (SD +/- 278 x 10(6)). Twenty-three of the 52 bulls were slaughtered 3-4 days after semen collection and aliquots of the cauda epididymal contents were collected for sperm morphology examination. The percentages of proximal droplets, abnormal tails and abnormal midpieces were significantly higher (p < 0.05) and the percentage of detached heads was significantly lower (p < 0.05) in the post-mortem samples compared with those in the TM samples. However, importantly there was no significant difference between the two sample types in the percentages of abnormal heads. This study demonstrates that semen can be collected from yearling beef bulls by TM. We think that TM constitutes a useful tool, when semen collection with EEJ or artificial vagina (AV) is not possible under field conditions, when included in the bull breeding soundness evaluation (BBSE) protocol. However, further studies are needed, and presently being carried out, to evaluate if semen samples collected by TM are comparable with semen samples collected by AV.  相似文献

This study was conducted to determine if growth hormone (GH) concentration in bovine seminal plasma would be proportional to but less variable than blood plasma GH. The relationship between GH in blood and seminal plasma was also examined critically. Blood samples were collected at 15-min intervals for 5.75 h, while semen was collected at 30-min intervals over the same time period. Average seminal plasma GH concentrations were 3.2 times higher (P less than .05) than blood plasma GH concentrations (40.4 +/- 15.8 ng/ml vs 12.6 +/- 1.2 ng/ml, respectively). The within animal correlation between blood and seminal plasma was consistently low and nonsignificant (P greater than .05). Overall blood plasma GH and seminal plasma GH concentrations were weakly correlated (r = .418; P greater than .05) among bulls. A predictable relationship between blood and seminal plasma GH concentration does not exist under the conditions of this study.  相似文献

The aim of the present study was to determine the effects of dietary supplements of vitamins on vitamin status, libido, and semen characteristics in young boars under normal and intensive semen collection. Sixty Landrace, Yorkshire, and Duroc boars were allocated randomly from 6 to 10 mo of age to one of the following diets: 1) basal diet (industry level) for minerals and vitamins (Control, n = 15); 2) basal diet supplemented with vitamin C (ASC, n = 15); 3) basal diet supplemented with fat-soluble vitamins (FSV, n = 15); and 4) basal diet supplemented with water-soluble vitamins (WSV, n = 15). After puberty (approximately 12 mo of age), semen was collected at a regular frequency (three times every 2 wk) for 5 wk. Thereafter, all boars were intensively collected (daily during 2 wk). A recovery period (semen collection three times every 2 wk) followed and lasted for 10 wk. Sperm quality (percentage of motile cells and percentage of morphologically normal cells) and quantity (sperm concentration, semen volume, and total sperm number) were recorded as well as direct and hormone related measurements of boar libido. Blood and seminal plasma samples were taken to monitor vitamin status. High concentrations of B6 (P < 0.05) and folic acid (P < 0.05) were observed in the blood plasma of WSV boars, whereas greater concentrations of vitamin E (P < 0.01) were obtained in FSV boars. In the seminal plasma, folic acid concentrations tended to be greater in WSV boars (P < 0.08). During the intensive collection period, there was a tendency (P < 0.06) for semen production to be greater in WSV boars, the effect being less pronounced (P < 0.10) in FSV boars. During the recovery period, the percentage of motile sperm cells was greater in WSV boars (P < 0.03) and, to a lesser extent, in FSV boars (P < 0.10) compared with Control boars. Sperm morphology and libido were not affected by treatments. These results indicate that the transfer of vitamins from blood to seminal plasma is limited and the dietary supplements of water-soluble and fat-soluble vitamins may increase semen production during intensive semen collection.  相似文献

Sixteen (mean age = 1.1 +/- 0.1 yr; mean BW = 478 +/- 34 kg) Brahman-influenced bulls were used to determine the influence of fescue type on sperm characteristics and serum concentrations of prolactin, cortisol, and testosterone. Bulls were blocked by BW, scrotal circumference (SC), and pregrazing sperm characteristics and randomly assigned to graze toxic endophyte-infected (EI; 4 bulls/pasture; 2 pastures) or novel endophyte-infected (NE; 4 bulls/pasture; 2 pastures) tall fescue for 121 d. Semen was collected by electroejaculation, and SC was measured and blood samples collected monthly. Sperm were evaluated for motility and morphology with an integrated visual optical system. Overall mean concentration of prolactin was decreased more (P < 0.01) in EI bulls than NE bulls from May to August. Scrotal circumference was not affected by fescue type (P = 0.58); overall SC averaged 36.7 +/- 2.3 cm. Percentage of live sperm was not different (P = 0.24) between NE bulls (80%) than EI bulls (67%) in July and August. Bulls grazing NE fescue had more (P < 0.06) motile sperm than EI bulls in July and August. Percentages of progressive (57 vs. 38%, NE and EI, respectively; P < 0.06) and rapid (67 vs. 46%, NE and EI, respectively; P = 0.04) sperm were greater from bulls grazing NE than EI bulls in July and August. Average velocity of the smoothed sperm path and progressive velocity in a straight line from the beginning to the end of the sperm track were slower (P < 0.09) in EI bulls than NE bulls and were slower (P = 0.04) in August compared with July. Mean width of head oscillation as the sperm swims was less (P < 0.06) in August than July. Concentrations of cortisol and testosterone were not (P > 0.10) influenced by fescue type. Semen from bulls grazing EI had reduced motility and morphology than bulls grazing NE. Detrimental effects of toxic fescue may not be mediated by cortisol, testosterone, or both. Semen quality of bulls grazing toxic EI tall fescue was decreased with increased maximum ambient temperatures.  相似文献

Zebu bulls are a shy breeder and they exhibit optimum libido in the presence of females with estrus phase. Continuous semen collection with the use of male dummy leads to lack of adequate sexual stimulation. Therefore, the present study was designed to test the effect of estrus-specific molecule(s) for effective sexual preparation of donor bulls. The bulls were divided into normal and poor libido group, five bulls in each group by taking 1-month control study data after collecting the information of individual bull’s sexual behaviour during semen collection by regular semen collector. The bulls were never being exposed to female animals and semen was collected by an artificial vagina. The ten animals were exposed to a glycerol-water solution (50/50 v:v) as control and then exposed to estrus-specific molecules one by one. The estrus-specific molecules like squalene, 1-iodoundecane, acetic acid, coumarin, propionic acid, oleic acid, and 2-butanone were purchased from Sigma-Aldrich Company, USA, and the molecules were solubilised individually in a non-pressurised aerosol dispenser as 1.0% concentration in glycerol-water solution (50/50, v:v). Identical bulls were used as the control and exposed to each molecule one by one by giving a refractory period of 14 days. A nasal spray of acetic acid or 2-butanone significantly (p < 0.05) reduced reaction time (RT) and total time taken to ejaculate (TTTE) in normal libido bull group. Semen volume, sperm concentration, and the total number of sperm per ejaculation obtained did not show significant improvement in the normal libido group of bulls after the application of estrus-specific molecules as compared to the control. In poor libido group, acetic acid, oleic acid, and 2-butanone application showed significant (p < 0.01) improvement in RT and TTTE as compared to the control group, whereas semen production variables like sperm concentration and total sperm output per ejaculation increased significantly (p < 0.05) except semen volume. There was significant (p < 0.01) reduction in RT (%) and TTTE (%) after the application of acetic acid followed by 2-butanone and oleic acid. The sperm concentration and total sperm output per ejaculation were more after the application of each molecule but significant increase (p < 0.05) in sperm concentration was observed with 2-butanone (11.42%), acetic acid (11.42%), and oleic acid (10.13%), whereas total sperm output per ejaculation increased significantly (p < 0.05) only after the application of acetic acid and 2-butanone (24.75% and 26.84%). Hence, it can be concluded that acetic acid, 2-butanone, and oleic acid are effective for better sexual preparation of Sahiwal bulls and total sperm output per ejaculation.

Artificial vagina (AV) and electroejaculation (EE) are the 2 methods used to obtain semen from bulls. The purpose in the present study was to evaluate these 2 methods of collection when 2 markers, cortisol and progesterone, were injected IV. During period 1 (control measurement), semen was obtained by EE at 0, 20, 60, 120, and 180 minutes. In period 2, bulls were injected (3 days later) with a mixture of cortisol (113 mg) and progesterone (100 mg), and then samples were obtained by EE. In period 3, cortisol and progesterone were injected (3 weeks later), and samples were obtained by AV. Seminal plasma concentrations of cortisol and progesterone were maximal at 20 minutes in EE and AV collections. Seminal plasma concentrations of progesterone and cortisol were roughly 50% less in EE than in AV collection. However, the total excretion of progesterone and cortisol per collection was similar in both techniques. Excretion of cortisol was 14 to 33 times greater than that of progesterone. It was concluded that concentrations of markers in the EE were significantly less than those in AV collection. For this reason, total excretion and concentration of marker in semen should be accounted for when conducting excretion studies.  相似文献

The objective of this study was to determine the effect of natural mating stimuli on serum concentrations of LH, testosterone (T) and estradiol-17 beta (E2) in beef bulls. Twenty sexually experienced, yearling beef bulls were bled every 15 min during a 9-h period, 4 h before and 5 h after exposure to estrual females. For exposure, each bull was placed individually in an isolated pen with two restrained estrual heifers for 10 min or until one service was achieved. Timing and number of all behavioral events, including flehmen responses, abortive mounts and services, were recorded for each bull by two independent observers. Of the 20 bulls, 9 bulls mounted and were removed immediately after achieving a service, 8 bulls mounted without achieving a service and 3 bulls exhibited no interest during exposure. Twelve bulls achieved fewer than three and eight bulls achieved three or more flehmen responses during exposure. Postexposure responses in LH, T and E2 were not consistently correlated with number of mounts or presence or absence of a service. However, postexposure LH and T, but not E2, responses were highly correlated with number of flehmen responses achieved (r = .40 to .66; P = .08 to .001). In bulls that achieved three or more flehmen responses, serum LH increased within 30 min after exposure (P = .02) and serum T was increased dramatically within 1 h after exposure (P less than .01), compared with preexposure hormone concentrations, regardless of the number of mounts and regardless of the presence or absence of a service. Natural mating stimuli had no effect on serum E2, and mounting activity alone and mounting that culminated in a service did not necessarily result in increased LH or T in beef bulls. However, number of flehmen responses achieved during exposure to females dramatically influenced postexposure serum LH and T concentrations in beef bulls.  相似文献

Effects of a short-term hyper- and hypoprolactinaemia on serum concentrations of LH, testosterone and semen quality in six male Beagles were investigated. Blood samples were collected at 3-day intervals for 12 weeks. The time span was divided into five 3-week periods: pre-treatment, metoclopramide (MCP) treatment (0.2 mg/kg orally three times daily), cabergoline (CAB) treatment (5 μg/kg orally once daily), post-treatment 1 and post-treatment 2. In the latter, only semen characteristics were evaluated. Semen parameters were analyzed once per week during the whole 15-week investigation time. At the end of each period, the effects of a single intravenous injection of thyrotropin-releasing hormone (TRH; 10 μg/kg) on the secretion of prolactin (PRL), LH, testosterone, thyroid-stimulating hormone and thyroxine (T4) were investigated. Pre-treatment serum PRL concentration increased under MCP (p < 0.05), followed by a decrease under CAB administration (p < 0.05). Luteinizing hormone and testosterone concentrations were not affected. Except for straight-line sperm velocity, semen quality did not differ between collection periods. A single iv TRH injection induced a significant PRL increase at 20 min in all experimental periods except during CAB treatment. Luteinizing hormone and testosterone did not show clear TRH-related changes. Basic T4 levels were significantly reduced after CAB treatment (p < 0.05). The results of the present study demonstrate that MCP-induced short-term hyperprolactinaemia in male beagles does not seriously affect the hypothalamo-pituitary axis and semen quality.  相似文献

In this study, the concentration of nickel in stallion, bull, ram, boar and fox semen, and its relation with spermatozoa quality was analyzed. The concentration of nickel in semen was 0.20 mg kg(-1) in stallion, 0.12 mg kg(-1) in bull, 0.31 mg kg(-1) in ram, 0.06 mg kg(-1) in boar and 0.36 mg kg(-1) in fox. Seminal nickel concentration was significantly higher (P < 0.05) in foxes than that in bulls and significantly higher (P < 0.01) in rams and foxes in comparison with boars. Evaluation of total pathological spermatozoa revealed the highest number in stallions followed by rams, bulls, boars and foxes. In bull, ram and boar semen, separated flagellum, flagellum torso and knob-twisted flagellum were predominant. Knob-twisted flagellum, separated flagellum and flagellum torso were found in increased number in stallion semen and broken flagellum in fox semen. Correlation analysis in bulls indicated a high positive correlation between seminal nickel and separated flagellum (r = 0.76) and medium positive correlation between nickel and flagellum torso (r = 0.62), and in rams a high positive correlation between nickel and separated flagellum (r = 0.77). Medium positive correlation was found between nickel and separated flagellum (r = 0.43) and between nickel and other pathological spermatozoa (r = 0.45) in boars.  相似文献

Postweaning growth, sexual development, libido, and carcass data were collected from two consecutive calf crops using 31 Brahman x Angus (B x A), 41 Senepol x Angus (S x A), and 38 Tuli x Angus (T x A) F1 bulls. Following weaning (by mid-September) and preconditioning, at the start of the study (late September) bulls were fed concentrate (three times each week at a rate equivalent to 4.5 kg/d) on bahiagrass pasture for approximately 250 d. At the start of the study and at 28-d intervals, BW, hip height, and scrotal circumference (SC) were measured. Concurrently at 28-d intervals, when the SC of a bull was > or = 23 cm, semen collection was attempted using electroejaculation. Ejaculates were evaluated for presence of first spermatozoa (FS), 50 x 10(6) sperm with at least 10% motility (PU), and 500 x 10(6) sperm with at least 50% motility (PP). After all bulls reached PP they were subjected to two libido tests. Carcass data were collected on all bulls (n = 110) and Warner-Bratzler shear (WBS) force values were assessed on a subset (n = 80). For both years, B x A bulls were heavier (P < 0.05) and taller (P < 0.05) than S x A and T x A bulls at the start and end of the study. However, breed type did not influence (P > 0.10) gain in BW or hip height during the study. Scrotal circumference of T x A bulls was larger (P < 0.05) than that of B x A or S x A bulls at the start of the study, but there was no effect (P > 0.10) of breed type by the end of the study. At PU and PP, B x A bulls were older (P < 0.05), heavier (P < 0.05), and taller (P < 0.05) and had larger (P < 0.05) SC than S x A and T x A bulls. Tuli x Angus bulls were younger (P < 0.05) than S x A bulls at PU and PP but had similar SC. Libido scores tended (P < 0.10) to be lower for B x A than for S x A and T x A bulls. Breed type affected (P < 0.05) carcass traits; B x A bulls had the heaviest (P < 0.05) hot carcass weight, greatest (P < 0.05) dressing percentage, larger (P < 0.05) longissimus muscle area than S x A bulls, and higher (P < 0.05) USDA yield grade than T x A bulls but greatest (P < 0.05) WBS force values. Breed type did not affect (P > 0.10) USDA quality grade. In conclusion, tropically adapted F1 bulls produced from Senepol (Bos taurus) and Tuli (Sanga) sires bred to Angus cows in Florida had lighter BW, shorter hip heights, and smaller carcasses than those from Brahman sires but reached puberty earlier and had higher libido scores and lower WBS force values.  相似文献

[目的]研究发情母牛粘液对采精种公牛交配能力以及精液品质的影响。[方法]从采精种公牛中随机抽出10头,进行2次采精试验,第一次按照正常方法采精,第二次在台牛尾根附近涂抹粘液,记录交配能力,将2次采精所采得的精液进行检测。[结果]第二次采精交配能力明显高于第一次。精液量、精液密度、活率、有效精子数、冻后活率、生产冻精数分别为:5.76 mL、19.23亿/mL、0.72、13.4、0.385、400.5,通过T检验P〈0.01,差异显著。[结论]在台牛后躯涂抹发情母牛粘液刺激公牛使交配能力和精液品质显著提高。  相似文献

The present study aimed to evaluate the effect of organic and inorganic selenium (Se) supplementation on semen quality and blood serum profiles of buffalo bulls. Nine mature buffalo bulls were divided into three groups: control (non‐supplemented); organic Se (10 mg Sel‐Plex®/head twice weekly) and inorganic Se (10 mg sodium selenite/head twice weekly). Semen was collected twice a week for 3 months during Se supplementation. Semen properties were evaluated from fresh ejaculate. Moreover, fructose concentration, aspartate and alanine transaminase (AST and ALT) activities, total protein and total cholesterol were assayed in seminal plasma. Additionally AST, ALT, testosterone and Se levels were determined in the blood serum. Results showed that Se supplementation significantly (P < 0.05) influences the semen parameters during 3 months of treatment. Organic Se significantly (P < 0.05) increased the percentage of viable sperms compared to inorganic Se and the control group. Fructose concentration was significantly higher (P < 0.05) in the seminal plasma of organic Se‐treated bulls. Serum testosterone and Se concentrations were significantly (P < 0.05) increased in the Se supplemented groups than the control group. In conclusion, Se supplementation improved the parameters of buffalo bull semen and more precisely, organic Se was more effective for the improvement of semen quality and some blood components than inorganic Se.  相似文献

Evaluation of two seminal collection regimens for mature Holstein bulls   总被引:2,自引:0,他引:2  
Twenty mature Holstein bulls (3 to 10 yr old) were used to test the effect of two semen collection regimens on spermatozoal output, post-thaw percentage spermatozoal motility, and time needed to make the collections/week. For both regimens, six ejaculates/wk were collected using either three ejaculates/d, 2 d/wk, or two ejaculates/d, 3 d/wk. A three-period switchback experimental design was used. Each collection period for which measurements were taken was 3 wk and was preceded by a 2 wk period of acclimation. The total number of spermatozoa harvested per week was not significantly different (P greater than .05): 33.2 X 10(9) when the bulls were collected two ejaculates 3 d/wk, compared with 33.9 X 10(9) three ejaculates 2 d/wk. Post-thaw progressive spermatozoal motility was 50.3 and 52.1% (P greater than .05), respectively. The average time per week to collect each bull was 73.6 and 83.7 min (P less than .05), respectively.  相似文献

Data from 34 yearling Hereford or Angus bulls were used to investigate relationships of testicular size, quantitative rates of sperm production, Sertoli cell numbers, numbers of germ cells supported per Sertoli cell, and the efficiency of spermatogenesis to daily sperm output and seminal quality. Two ejaculates were collected by electroejaculation from each bull on each of 2 days/week throughout the study. The percentage of progressively motile sperm and the percentage of morphologically normal sperm were determined from aliquots of fresh semen. Additional aliquots of semen were frozen in glass ampules or plastic straws and subsequently evaluated for postthaw motility and percentage of sperm with intact acrosomes. Sertoli cell numbers, the numbers of germ-cells per Sertoli cell, and the efficiency of spermatogenesis were unrelated to the quality of fresh or frozen semen (P greater than 0.05). In first ejaculates, the numbers of sperm and motile sperm were related (P less than 0.05) to testicular parenchymal weight (r = 0.38 and 0.50), daily sperm production (r = 0.45 and 0.53), and spermatids per gram of testicular parenchyma (r = 0.35 and 0.34). Testicular parenchymal weight and daily sperm production also were related to daily sperm output and to the average daily motile sperm output of these bulls (P less than 0.05), but could account for less than 25% of the variability in these end points among bulls.  相似文献

The testosterone and estrogen concentrations in venous serum of 4 mature Holstein-Friesian bulls were determined by radioimmunoassay before and after each of 3 consecutive semen collections taken at 1-hour intervals. The hormonal concentrations were highly variable for the 2 steroids, but uniform in their patterns after the bulls had ejaculated. After the 3rd semen collection, marked changes in hormonal concentrations did not occur.  相似文献

Repeatability (r) value of glutathione (GSH) content was estimated in semen of Tharparkar, Red Dane, their crosses, and Murrah buffalo bulls. Mean GSH values were higher in bovine bull semen as compared to mean GSH values in bubaline bull semen. The r of GSH concentration for the pooled data was 0.1278. This trait is 12.78% repeatable. GSH value in semen of bovine and bubaline bull differed insignificantly. R estimates are expressed for selection of bulls of higher fertility and semen quality.  相似文献

Selection of dairy sires is based on the production records of their female ancestors, half-sibs and daughters. No trait expressed by the sire is used. Concentrations of growth hormone (GH) and prolactin (PRL), hormones produced in both males and females that are fundamental in lactation, may be correlated with production. A study was conducted to determine whether measures of these hormones in the sire would be useful predictors of lactational ability of daughters. Blood samples were collected at 15-min intervals for 8 h from 26 Holstein bulls (5.5 yr of age) that had one progeny summary available. Plasma concentrations of GH and PRL were quantified and the mean and baseline concentrations and the frequency and mean amplitude of the secretory peaks were determined for each bull. Concentrations among these values and bulls' predicted differences (PD) were determined. Significant negative correlations were detected for frequency of GH peaks and PD for yield of milk, fat and protein; correlations were positive for PRL baseline concentrations and PD for fat and protein (P less than .10), and correlations were negative for frequency of PRL peaks and PD for milk, fat and protein (P less than .10). Addition of estimates of bull hormone secretory parameters to breeding values based on performance of relatives considerably improved the accuracy (R2) for predicting progeny performance from sire information. Certain characteristics of the patterns of GH and PRL secretion may be heritable and aid in identification of superior dairy animals.  相似文献
