ABSTRACT Experiments were conducted in three prune orchards in California. In each orchard, inoculations with Monilinia fructicola, the causal agent of brown rot of stone fruits, were performed on branches of trees at bloom and fruit developmental stages. Five inoculum concentrations were used in each inoculation. Six and four wetness durations were created for each inoculum concentration at bloom and fruit developmental stages, respectively. Fruit were harvested 3 weeks before commercial harvest. The overnight freezing incubation technique was used to promote sporulation and to determine incidence of latent infection (ILI) of fruit brown rot. No differences in ILI among locations were found. A seasonal pattern of bloom and fruit susceptibility to latent infection was determined. Susceptibility to latent infection at bloom stage was at a moderate level and increased to reach the highest level at pit hardening stage. Subsequently, fruit susceptibility to latent infection decreased, reaching the lowest level in early June at embryo growth stage. Thereafter, the susceptibility increased again with fruit development and maturity until harvest. Linear relationships between ILI and inoculum concentration were obtained for most combinations of growth stage and wetness duration. Incidence of latent infection increased linearly with increased wetness duration at bloom stage and increased exponentially with increased wetness duration at early and late fruit developmental stages. The optimum temperatures for latent infection at pit hardening stage ranged from 14 to 18 degrees C, but the effect of temperature on latent infection was reduced at resistant stages. The temperature range favorable to latent infection varied for different wetness durations.  相似文献

ABSTRACT The quantitative relationships between incidence of latent infection (ILI) of prune by Monilinia fructicola and wetness duration (WD) for different bloom and fruit developmental stages and different inoculum concentrations were obtained. Three levels of ILI were considered as criteria for low, moderate, and high risks of latent infection, respectively. Seasonal patterns of WD leading to different risk levels of latent infection were obtained for low (IP(L)) and high (IP(H)) inoculum potential conditions in orchards. Longer WD was needed at a resistant than at a susceptible fruit developmental stage to induce similar levels of latent infection. The curves of WD leading to different levels of ILI over the growing season (risky WD curves) were used in risk analysis for latent infection. Multi-year historical WD data from 10 prune-growing locations in California were compared with risky WD curves. The percentage of days (P) with WD leading to a certain risk level of latent infection was calculated for each month from historical weather data. Under the IP(L) condition, the P distributions for low risk of latent infection were higher in March and April than in May and were the lowest in June for most locations. Under the IP(H) condition, the number of days that WD leading to low risk of latent infection in May increased compared with those under the IP(L) condition. The risk analysis approach was evaluated by using separate experimental data as incidence of fruit brown rot obtained from different prune orchards over years. Consistency between predicted overall risk levels of latent infection and observed incidence of fruit brown rot was obtained. The results demonstrated the usefulness of the risk analysis in decision support system for disease management.  相似文献

Experiments under controlled environmental conditions were conducted during bloom of prune (Prunus domestica, L.) in 1999 and 2000 to assess the effects of inoculum concentration (IC), wetness duration (WD), temperature, and bloom stages on development of brown rot blossom blight of prunes. Branches from trees of a prune orchard were inoculated with Monilinia fructicola at different bloom stages and incubated at different temperatures with different periods of WD. The proportion of blighted blossoms (PBB) for each inoculated branch was determined. Bloom stage, IC, temperature, and WD significantly affected blossom blight of prunes. PBB at popcorn and full bloom stages was significantly greater than PBB at later bloom stages (P 30 degrees C and less than 4-h WD. However, longer than 4-h WD linearly increased incidence of blossom blight. A risk assessment table of blossom blight was produced for different environmental conditions to guide the control of prune brown rot.  相似文献

Stone fruit tree apoplexy in Greece is often caused by Phytophthora spp. Two types of Phytophthora apoplexy of stone fruit tree can be distinguished, one occurring during the hot summer period and usually caused by P. cactorum (Leb. et Cohn) Schröt. or P. citrophthora (R. et E. Smith) Leonian, and a second occurring in winter or early spring, and caused by P. syringae (Kleb.) Kleb. or sometimes P. megasperma Drechsler. Inoculation tests showed P. syringae to be highly pathogenic to apricot and almond, whereas certain clonal rootstock material showed resistance. ABSTRACT Blueberry fruit infected by Monilinia vaccinii-corymbosi, the causal agent of mummy berry disease, are unsuitable for use in processed food products. Fruit shipments that exceed a disease incidence threshold of 0.5% are redirected to alternative markets with substantial reductions in economic return to the producer. Because of this low tolerance, a sampling procedure with defined statistical properties is needed to determine disease incidence in the packinghouse. In this study, a sequential sampling plan was developed based on counts and dispersion patterns of infected fruit in 23 loads of mechanically harvested rabbiteye blueberries. Each load was sampled 20 to 100 times, with each sample containing 550 cm(3) of fruit. Various dispersion statistics (k of the negative binomial distribution, Lloyd' index of patchiness, and Iwao' b) were computed, all of which suggested aggregation of infected fruit. Because k was variable across loads, Iwao' regression procedure, which does not assume a single frequency distribution with fixed parameters describing the counts of infected fruit, was used to develop upper and lower stop lines for sequential sampling. For alpha = 0.05 and assuming a total of 250 fruit per 550-cm(3) sample, the resulting sampling plan would require only one sample to conclude that a load exceeds the threshold if the number of infected fruit in that sample is greater than four. A minimum of six samples would be needed to conclude that disease incidence in a load is below the threshold if the cumulative total of infected fruit in these samples is zero. Resampling analysis showed that most fruit loads could be classified reliably with <10 samples per load; for loads with a disease incidence very close to the 0.5% threshold, <50 samples were needed on average. To examine the duration of post-infectional activity of ergosterol biosynthesis-inhibiting fungicides (EBIs) against Botryosphaeria dothidea, the cause of apple white rot, a series of experiments was conducted for 3 years at different times during the apple-growing season using apples infected through artificial inoculation or natural infection. Disease incidence at harvest was gradually reduced as the time of chemical application was delayed until early or mid-August, regardless of the time of infection. Latent infections, detected after storing the fruit at 25°C for 4 weeks were also reduced by late season applications, but the reduction was less than that observed on the fruit at harvest. From these results, apple white rot can be effectively controlled by a single application of EBIs around mid-August, after which time, infections no longer occur in Korea. ABSTRACT The effect of environment on the infection of peach fruit by Taphrina deformans was investigated using orchard observations under natural conditions (in 2001 to 2004) or in trees managed in such a way to exclude rainfall. These conditions were then validated using pot-grown peach plants exposed to single infection events and independent orchard observations. Leaf curl incidence was related to rainfall, length of wet periods, and the temperature during wetness and during the incubation period, as well as to the developmental stage of flowers and fruit. Weather conditions before petal fall did not influence fruit infection. After petal fall, rainfall and the duration of the wet period triggered by rainfall played a key role in infection occurrence. The minimum rainfall required for infection was 12 mm, with at least 24 h of wetness interrupted by no more than 4 h. No infection occurred when temperature was >/=17 degrees C during the wet period or >19 degrees C during incubation. Disease symptoms appeared on fruit after approximately 3 weeks of incubation, which is equivalent to 240- to 290-degree-days (base 0 degrees C). The period for fruit infection was relatively short being from petal fall until air temperature remained greater than 16 degrees C. During this period, the incidence of fruit that developed symptoms was closely related to the number of favorable events and the total wetness duration during such events.  相似文献

Brown rot and twig canker of fruit trees are caused by Monilinia laxa, M. fructigena and M. fructicola. The Internal Transcribed Spacer (ITS) between the 18S and the 28S rRNA genes of four M. laxa and four M. fructigena isolates collected in France was amplified by Polymerase Chain Reaction (PCR) using universal primers and sequenced. Multiple alignment of the ITS sequences and comparison with published sequences revealed very little intraspecific variation and a low interspecific polymorphism clustered in two regions. Species-specific PCR primers were designed to amplify a 356bp fragment for each of the three species. The specificity of the three primer pairs was successfully tested with a collection of 17 M. laxa, 18 M. fructigena and 6 M. fructicola isolates collected from different hosts and different countries, unequivocally confirming the identification of each isolate based on morphological and cultural traits. Using stringent PCR conditions, no cross-reaction was observed with any of the isolates tested. The specificity of the PCR assays was also successfully confirmed with DNA extracted from different fungal species, either phylogenetically close to the genus Monilinia or commonly found on diseased fruits. Using this new reliable technique, doubtful isolates can be directly identified in a single PCR run. Moreover, detection and identification of the Monilinia species were successfully achieved directly on diseased fruits. This simple and rapid method can be particularly useful to detect M. fructicola which is a listed quarantine fungus in all European countries.  相似文献

ABSTRACT Infection of cranberry flowers by conidia of Monilinia oxycocci, the cottonball pathogen, was investigated using a squash-mount histological method. Conidia germinated on anthers, nectaries, petals, and stigmata, but not styles. The stigma was the only flower part penetrated by the fungus, but no specialized infection structures were noted. Both fungal and pollen germ tubes grew through the stylar canal and made contact with ovules and nucellar tissue by 72 h after inoculation and pollination. Cottonball incidence was greatest when stigmata were inoculated; the low level of cottonball that resulted from inoculation of other flower parts and in noninoculated flowers was attributed to contamination of stigmata. In greenhouse tests, cottonball incidence was 25, 28, 31, and 38% for cvs. Searles, Pilgrim, Ben Lear, and Stevens, respectively, and was greater for M. oxycocci isolate 593 than isolate 591. We conclude that the stigma is the sole floral infection court for conidia of M. oxycocci and that the most popular cranberry cultivars in Wisconsin do not differ in inherent resistance to cottonball. The relevance of these findings to the long-term management of cottonball is discussed.  相似文献

ABSTRACT Naturally established lowbush blueberry clones in four fields were evaluated for the incidence of leaf and flower blight, proportion of mummy berries, and yield reductions caused by Monilinia vacciniicorymbosi. The relationship between the phenology of flower and leaf bud development and susceptibility also was examined. Three fields were examined over one crop year and one field was studied in two subsequent crop years. The incidence of stems with blight was correlated to incidence of leaf blight in all fields and to incidence of flower blight in one field. Incidence of leaf and flower blight and the proportion of mummy berries produced were not correlated. Lowbush blueberry clones with higher incidence levels of leaf blight had reduced fruit set and lower berry weights. For healthy stems, leaf-to-fruit ratios had no effect on berry weight in most fields. In contrast, blighted stems with higher leaf-to-fruit ratios had higher berry weights in three fields. Stems with slowerdeveloping leaf and flower buds had less leaf and flower blight, respectively, than stems with faster bud development. Some blueberry clones may avoid infection by delaying production of susceptible tissue until after ascospore release by M. vaccinii-corymbosi.  相似文献

ABSTRACT Plant defense strategies against pathogen attack can be divided into either resistance or tolerance. Variation in tolerance is expressed as differences in the relationship between host fitness (or yield) and the degree of infection. Plant tolerance of pathogen attack remains poorly understood both in terms of its specific mechanisms and in terms of the evolutionary processes by which it has arisen. Theoretical models predict that it is the result of coevolution between host and pathogen, suggesting greater tolerance in interactions with native as opposed to introduced pathogens. Therefore, we quantified and compared the degree of tolerance expressed in the interaction of Senecio vulgaris with the rust fungus Coleosporium tussilginis, which is native to the UK, and the introduced rust fungus Puccinia lagenophorae. We used the reaction norm approach to quantify tolerance and its components. The S. vulgaris-C. tussilaginis interaction expressed a significantly greater degree of tolerance, as reductions in host growth and fitness per unit infection were significantly less than with P. lagenophorae. The key mechanism for this greater tolerance to C. tussilaginis was a significantly smaller reduction in photosynthesis per unit infection than with P. lagenophorae, at both leaf and whole plant scales. There was no significant difference in the relationship between whole plant photosynthesis and host reproduction. We discuss these responses in the context of coevolution for tolerance in host-pathogen interactions.  相似文献

As it is difficult to detect ring rot in its latent form as well as in badly decayed tubers and in potato haulm, a diagnostic technique has been developed. With the help of a Turmix blender and a little water, a suspension is made of the suspected material. The suspension is inoculated into eggplants. Gram-staining of bacteria in sap from petioles of leaves showing symptoms is used to confirm the diagnosis. The fungus Alternaria alternata, is considered to be the predominant fungus involved in moldy-core of Red Delicious strains of apple. In this paper, we report on the sensitivity of various phenologic stages to infection by A. alternata, and on the efficacy of various fungicides in controlling moldy-core disease in apple orchards. Artificial inoculations conducted in the orchard during 1999 and 2000 seasons revealed that the beginning of bloom (10–30%) and full bloom were the most susceptible developmental stages for infection. Natural infection with A. alternata in fruits was relatively high, reaching 44% and 46% of the fruits on control non-treated trees in 1999 and 2000. Four foliar applications of polyoxin B, difenoconazole and azoxystrobin, starting from the beginning of bloom until fruit set, reduced the number of infected fruits by 54–70%, 61–70% and 50–65%, respectively, compared with non-treated trees. Four or eight foliar applications of each fungicide provided similar levels of control. There were no significant differences between two, four or six foliar applications of difenoconazole, neither between two or four applications of polyoxin B. Adding CaCl2, as a tank mixture with difenoconazole at full rate, did not improve efficacy. Alternaria was recovered from the inner part of the core region of 71–88% of the fruits of the non-treated control, but was recovered less frequently from the outside part of the core region. Fruits of difenoconazole and polyoxin B treated trees were less colonized with A. alternata at both the inner and outside parts of the core region, as compared with controls. Results indicate that a control programme based on spray applications of difenoconazole or polyoxin B, during bloom period, can effectively reduce Alternaria on Red Delicious.  相似文献

Young leaves of 62 citrus cultivars were inoculated with conidia of three Spanish isolates of Alternaria alternata pv. citri, the causal agent of brown spot of citrus. Hybrids with Dancy mandarin, King mandarin or their derivates as a parent, grapefruit cultivars and the mandarin cultivars Guillermina, Emperor, Clemenpons and Esbal were highly susceptible to the pathogen. Satsuma cultivar Clausellina and orange cultivars, with the exception of Sanguinelli, were slightly susceptible. Lemon and lime cultivars were not susceptible, with the exception of Mexican lime (Citrus aurantifolia), which was slightly susceptible. Although this study shows a range of potential hosts for this pathogen, to date the only affected cultivars in Spain are Fortune and Nova mandarins, and Minneola tangelo. The susceptibility of Fortune fruits decreased as diameter increased, being susceptible through the whole season. This was confirmed with field observations in autumn where fruit infections have been detected when the diameter reaches 6–7 cm.  相似文献
