为了探寻稻瘟病菌无性世代DNA水平的变异,明确江西省稻区稻瘟病菌遗传宗谱与致病型之间的对应关系,利用rep-PCR(repetitive element-based PCR)分子指纹分析技术,对稻区稻瘟病菌的群体结构和遗传多样性进行分析,并用41株代表性菌株对35个水稻品种进行了致病性测定。结果表明,以相似度75%为界,可以将不同稻区采集的99个菌株划分为14个遗传宗谱,其中,宗谱4、1和10为优势宗谱,分别包含37、18和12个菌株,占总数的37.37%、18.18%和12.12%;稻瘟病菌遗传宗谱与致病型间存在复杂的关系,同一宗谱的菌株对应多个致病型,而同一致病型的菌株,分属于不同的遗传宗谱,两者之间不存在简单的对应关系。  相似文献

ABSTRACT The population genetic structure of Phytophthora infestans in Ecuador was assessed from 101 isolates collected from 1990 to 1992 and 111 isolates collected in 1993. All isolates were analyzed for mating type and allozyme genotype. Both samples were dominated (>95%) by a clonal lineage (EC-1) defined from neutral markers: 90/100 genotype for glucose-6-phosphate isomerase, 96/100 genotype for peptidase, A1 mating type, and a previously unreported nuclear DNA fingerprint. The remaining isolates belonged to the US-1 clonal lineage, which has a worldwide distribution. Isolates in the 1993 sample were analyzed for virulence and metalaxyl sensitivity. All representatives of EC-1 had complex patho-types, with three pathotypes representing >60% of the collection. There was variation for metalaxyl sensitivity. There was no evidence for geographical substructuring on the basis of neutral markers, but there was evidence for limited substructuring based on metalaxyl sensitivity and specific virulence. We hypothesize that EC-1 has been recently introduced to Ecuador.  相似文献

A previous study of the diversity and population structure of the rice blast fungus, Magnaporthe oryzae, over a 20-year period in Korea, found novel fingerprint haplotypes each year, and the authors hypothesized that populations might experience annual bottlenecks. Based on this model, we predicted that M. oryzae populations would have little or no genetic differentiation among geographic regions because rice blast is commonly found throughout Korea each year and M. oryzae would have to disperse from small populations surviving annually between rice crops. To test this hypothesis, we sampled M. oryzae from rice fields in eight provinces in Korea in a single year (1999). In four provinces, we sampled from a set of rice cultivars commonly grown in commercial fields (group I); because of low disease incidence in four other provinces, we could not sample from commercial fields and instead sampled from scouting plots of different cultivars set up for detecting new pathotypes of M. oryzae (group II). All isolates were genotyped with DNA fingerprint probes MGR586 and MAGGY, a telomere-linked gene family member TLH1, the PWL2 host specificity gene and mating type. Fingerprint haplotypes clustered into two distinct lineages corresponding to the two sets of cultivars (groups I and II), with haplotype similarities of 71% between lineages and >76% within lineages. Isolates from the same cultivar within group I were genetically differentiated among locations, and isolates within the same location were differentiated among cultivars. Differentiation for TLH1 and PWL2 was significant (P < 0.03), but not as strong as for fingerprint markers. Similar analyses were not possible among group II isolates because too few isolates were available from any one cultivar. All isolates were in the same mating type, Mat1-1, ruling out sexual reproduction as a source of novel haplotypes. When the 1999 samples were compared with the historical samples from the previous study, haplotypes of group I formed a separate cluster, while those of group II clustered with haplotypes from the historical sample. Altogether, geographic subdivision, monomorphism of mating type, and correlation of haplotypes to sets of cultivars are not consistent with the hypothesis of repeated turnover of haplotypes. Instead, the previous correlations of haplotypes to year might have been caused by inadequate sampling of haplotypes each year, highlighting the need for studies of population genetics to be conducted with systematic samples collected to address specific questions.  相似文献

根据93个单病斑中分离的227个单孢菌株样品的分析,研究了来自同一定殖系统(同一病斑)中稻瘟病菌不同单孢菌株(同源菌株)的遗传变异。在致病性分析的样品中,仅有5个病斑分离的10个单孢菌株在所有鉴别品种上表现同源菌株有完全相同的致病反应,并且有完全相同的DNA指纹图谱带型;另有13个病斑上分离的32个单孢菌株的同源菌株不仅属于不同的生理小种,而且在1~3个单基因品种上也有不同的致病反应。用rep—PCR指纹图谱和致病型分析67个病斑的157个单孢样品中的同源菌株具有完全相同的DNA指纹图谱;13个病斑中分离的27个单孢样品中同源菌株具有显著差异的指纹图谱;8个病斑分离的27个单孢菌株样品中同一来源样品中部分样品有相同的指纹图谱带型,部分样品却有不同的指纹图谱。同源菌株之间有完全相同致病性其指纹图谱亦相同或有很小的差异;通常,致病性差异很大的同源菌株其指纹图谱差异也很大。作者认为来自同一定殖系统中的不同单孢菌株存在的遗传变异与病菌的异核现象或自然状态下不同遗传背景菌株同时侵染造成同一病斑有关。  相似文献

Ninety-five isolates ofPhytophthora infestans collected throughout Poland during 1985–1991 and characterized for multilocus genotypes based on mating type, allozymes and DNA fingerprint, were analyzed for specific virulence to differential potato cultivars carrying ten major resistance genes. The multilocus analysis led to three groupings. The first group contained 22 isolates of a clonal lineage (PO-1) that is postulated to have been present in Europe during most of the twentieth century, but PO-1 isolates were recovered in Poland only during 1985–1988. This group contained, on average, virulence to 5.5 specific resistance genes per isolate. The second group consisted of 30 isolates in a clonal lineage (PO-4) that had not been detected before 1988. PO-4 isolates had virulence to a mean of 6.5 resistance genes per isolate. The third group was composed of 43 isolates representing 38 multilocus genotypes also not detected before 1988. These diverse genotypes had virulence to an average of 6.7 specific resistance genes per isolate. More than half (53%) of the PO-4 isolates shared a single pathotype. The group of 43 isolates was dominated by two pathotypes: the most common one (47% of the isolates) was the same pathotype that dominated PO-4 isolates; the next most common one (21%) differed from the most common one by the absence of virulence to resistance gene R5. The recent immigrant isolates (not detected before 1988) generally had virulence to a greater number of specific resistance genes than did isolates in the previous population [detected before 1988 (PO-1)]. Recent immigrant populations were dominated by one or two pathotypes, so their pathotypic diversity values were somewhat less than that of the previous population.  相似文献

应用rep-PCR分子指纹技术对2000~2002年采自四川6个籼稻自然生态区的137个稻瘟病菌菌株进行了DNA分子指纹扩增和聚类分析,共获得73个不同的DNA指纹图谱(单元型)和62条分子量不等的DNA带型.结果显示,无论以何种遗传相似水平划分,四川稻瘟病菌的群体结构都表现很突出的优势宗谱,又存在着具有较多遗传多样性的次要小宗谱和特异性宗谱,蕴含着极其丰富的遗传信息;在0.19遗传相似水平,所有供试菌株可以划分成37个遗传宗谱,层次较为丰富.四川稻瘟病菌群体结构具有明显的时空特点,不同年度间稻瘟病菌群体存在一定的亲缘关系,又各自拥有当年的特异性宗谱;在空间上,不同稻作区表现出从复杂到简单的病菌群体变化特点.稻瘟病菌的遗传宗谱与生理小种致病型不存在一一对应的关系,作者认为将二者横向比较没有可比性.  相似文献

The genetic variability among 41 isolates of the blast pathogen (Magnaporthe grisea) from five European rice growing countries was studied. The genealogy of the isolates was investigated by DNA fingerprinting and the results compared to the degree of similarity for (a)virulence factors. Fingerprinting grouped the isolates into five discrete lineages, that typically showed less than 65% band similarity. Within each lineage, two or more haplotypes were detected with a band similarity of 80% or higher. Each lineage showed a characteristic virulence pattern. All isolates of lineage E5 belonged to the same pathotype. The other lineages were composed of clusters of closely related pathotypes that showed variation for virulence to cultivars with certain known resistance genes, while remaining invariably (a)virulent to others. In most cases, lineage classification of an isolate could be easily inferred by its pathotype. Certain resistance genes and certain lineage-excluding resistance gene combinations appear to provide protection against all of the virulence factors sampled.  相似文献

ABSTRACT Amplified fragment length polymorphisms (AFLPs) were used to estimate phylogenetic relationships within Magnaporthe grisea and determine the genetic structure of M. grisea populations associated with tall fescue and St. Augustinegrass in Georgia. Sixteen clonal lineages were identified in a sample population of 948 isolates. Five lineages were isolated from tall fescue (E, G1, G2, G4, and H), with lineage G4 comprising 90% of the population. Isolates from tall fescue were closely related to those from perennial ryegrass, weeping lovegrass, and wheat. Two M. grisea lineages were isolated from St. Augustinegrass (C and K), with lineage C comprising 99.8% of the population. Populations from crabgrass were dominated (98%) by lineage K, but also contained a single lineage C isolate. Haplotype diversity indices ranged from 0.00 to 0.29 in tall fescue populations and from 0.00 to 0.04 in St. Augustinegrass populations. Selection due to host species was the primary factor determining population structure according to analysis of molecular variance; host cultivar and geographical region had no significant effect. The host range of M. grisea lineages from turfgrasses was determined in growth chamber experiments and supports the prominent role of host species in determining the genetic structure of M. grisea populations from turfgrasses in Georgia.  相似文献

西南地区稻瘟病菌对戊唑醇的敏感性基线建立及抗性监测   总被引:1,自引:0,他引:1  
于2013—2014年分别从重庆市及四川、贵州省的16个区县采集分离获得902株稻瘟病菌,采用菌丝生长速率法测定了其中150个菌株对戊唑醇的敏感性。结果表明:其EC_(50)值分布在0.047 5~0.599 6μg/m L之间,平均EC50值为(0.215 4±0.149 7)μg/m L。菌株的不同敏感性频率呈连续单峰曲线分布,因此其平均EC_(50)值0.215 4μg/m L可作为田间稻瘟病菌对戊唑醇敏感性检测的参考标准。戊唑醇对菌株的最低抑制浓度(MIC)值≤4μg/m L,以4μg/m L作为抗性菌株监测的区分剂量,测定了各地病原菌对戊唑醇的田间抗性,发现自各地采集分离的902株稻瘟病菌多数对戊唑醇仍表现为敏感,仅2株为低水平抗性。  相似文献

ABSTRACT Magnaporthe grisea (anamorph = Pyricularia grisea) causes blast on rice (Oryza sativa) and gray leaf spot on turfgrass. Gray leaf spot is a serious disease on St. Augustinegrass (Stenotaphrum secundatum), perennial ryegrass (Lolium perenne), and tall fescue (Festuca arundinacea). Virulence assays performed in this study revealed that M. grisea collected from rice could also cause disease on St. Augustinegrass and tall fescue. One rice isolate, Che86061, caused similar disease reactions on susceptible cultivars of rice and St. Augustinegrass and an incompatible interaction on resistant cultivars of both species. To explore whether similar defense-related genes are expressed in rice and St. Augustinegrass, a rice cDNA library was screened using pooled cDNAs derived from M. grisea-infected St. Augustinegrass. Thirty rice EST (expressed sequence tag) clones showing differential expression in St. Augustinegrass following M. grisea inoculation were identified and classified into six putative functional groups. Northern blot analyses of seven EST clones that collectively represented each putative functional group confirmed that the expression of five out of seven EST clones was similar in both rice and St. Augustinegrass. This study represents one of the first attempts to use a broad-scale genomic approach and resources of a model monocot system to study defense gene expression in St. Augustinegrass following M. grisea infection.  相似文献

ABSTRACT We applied DNA markers to determine whether parasexual recombination may contribute to the extreme genetic diversity and variability observed in Magnaporthe grisea, the causal agent of rice blast disease. Dispersed repetitive elements and mapped, low-copy restriction fragment length polymorphism (RFLP) probes were used to detect transfers of DNA between cultured isolates of M. grisea. Low-copy RFLP probes also were used to detect putative recombinants among isolates from well-characterized field populations of the pathogen. Microscopic examination of tufted mycelium between cocultured isolates revealed frequent hyphal fusions. Hyphal tips and conidia were recovered without selection from tufted zones in two separate vegetative pairings involving isolates with dissimilar haplotypes, based on the repetitive element MGR586. Haplotypic changes were observed at a higher frequency in tuft derivatives than in subcultures of each isolate alone. From 136 tuft derivatives analyzed, 5 putative recombinant haplotypes were identified. Introgression was demonstrated with two independent repetitive elements, fosbury and MGR586, as probes on DNA digested with several restriction enzymes. Introgressions were characterized by addition of 1 to 10 MGR586 bands, and 1 to 3 fosbury bands from one parent into the background of the other. Polymorphic single-copy probes were used to analyze putative recombinants. One probe detected an introgression event as predicted by analysis with MGR586. To assess the possible role of parasexual recombination in field populations of the pathogen, isolates in the Philippines previously grouped based on DNA fingerprinting were analyzed with low-copy RFLP markers. Polymorphism in single-copy loci typically was seen between, but not within, putative pathogen lineages. One lineage (designated lineage 4), however, was polymorphic for several probes. For some isolates, alleles at these loci comigrated with alleles characteristic of other lineages, suggesting the transfer of DNA fragments between lineages. One isolate was apparently a merodiploid, carrying an allele typical of lineage 4 plus another allele characteristic of a different lineage. In a survey of isolates from the Indian Himalayas, a merodiploid also was found with single- or low-copy probes. Examination of MGR586 profiles of the putative recombinant and its putative donor strains showed the expected introgression of MGR586 bands. The detection of parasexual DNA exchanges in wild-type strains under unselected conditions and the existence of merodiploids in nature suggest that parasexual recombination occurs in field populations of M. grisea. This raises questions concerning exclusive clonality in the blast fungus.  相似文献

ABSTRACT A rice isolate of Magnaporthe grisea collected from China was avirulent on rice cvs. Hattan 3 and 13 other Japanese rice cultivars. The rice cv. Hattan 3 is susceptible to almost all Japanese blast fungus isolates from rice. The genetic basis of avirulence in the Chinese isolate on Japanese rice cultivars was studied using a cross between the Chinese isolate and a laboratory isolate. The segregation of avirulence or virulence was studied in 185 progeny from the cross, and monogenic control was demonstrated for avirulence to the 14 rice cultivars. The resistance gene that corresponds to the avirulence gene (Avr-Hattan 3) is thought to be located at the Pik locus. Resistance and susceptibility in response to the Chinese isolate in F(3) lines of a cross of resistant and susceptible rice cultivars were very similar to the Pik tester isolate, Ken54-20. Random amplified polymorphic DNA markers and restriction fragment length polymorphism markers from genetic maps of the fungus were used to construct a partial genetic map of Avr-Hattan 3. We obtained several flanking markers and one co-segregated marker of Avr-Hattan 3 in the 144 mapping population.  相似文献

ABSTRACT During the 1995 wet season, harvested rice seed was collected from farmers' fields at different locations in Iloilo, Philippines. Bacterial isolations from crushed seed yielded 428 isolates. The isolates were characterized by BOX-polymerase chain reaction fingerprinting of total genomic DNA and represented 151 fingerprint types (FPT). Most FPTs were found on a single occasion, although matching fingerprints for isolates from different samples also were found. Identifications were made by cellular fatty acid methyl ester analysis and additional use of Biolog GN/GP MicroPlates and API 20E/50CHE systems. The predominant bacteria were Enterobacteriaceae (25%), Bacillus spp. (22%), and Pseu-domonas spp. (14%). Other bacteria regularly present were identified as Xanthomonas spp., Cellulomonas flavigena, and Clavibacter michiganense. Of the total number of isolated bacteria, 4% exhibited in vitro antifungal activity against Rhizoctonia solani or Pyricularia grisea. Two percent of isolates were pathogens identified as Burkholderia glumae and Burkholderia gladioli. Five percent of isolates induced sheath necrosis on only 50 to 90% of inoculated plants and were related to Bacillus pumilus, Paenibacillus spp., Pseudomonas spp., and Pantoea spp.  相似文献

 稻瘟病是福建省水稻生产中的重要病害之一,系统掌握稻瘟菌毒性类型组成和变化动态及其与主要抗病基因的互作特点,是制定抗病品种选育与合理利用的依据。本研究根据稻瘟病菌与6个CO39近等基因系品种互作亲和性的结果,将1995~2001年从福建采集分离的398个有效单孢菌株区分为26个毒性类型,其中毒性类型I34.1出现频率最高,为优势毒性类型,出现频率较高的还有I20.1、I04.1、I24.1、I0.1、I30.1等;结果还发现福建稻瘟菌群体对Pi1Pi2毒性频率较低,分别为7.53%和11.31%,特别是对Pi1Pi2基因联合毒性频率仅2.76%,说明在水稻抗瘟育种中可以考虑将Pi1Pi2基因累加利用。  相似文献

ABSTRACT Sixty Ecuadorian isolates of Phytophthora infestans from potato and 60 isolates from tomato were compared for dilocus allozyme genotype, mitochondrial DNA haplotype, mating type, and specific virulence on 11 potato R-gene differential plants and four tomato cultivars, two of which contained different Ph genes. Restriction fragment length polymorphism (RFLP) fingerprints of subsamples of isolates from each host were compared by using RG57 as the probe. All potato isolates had the allozyme genotype, haplotype, and mating type of the clonal lineage EC-1, which had been previously described in Ecuador. With the same markers, only one isolate from tomato was classified as EC-1; all others belonged to the globally distributed US-1 clonal lineage. RFLP fingerprints of isolate subsets corroborated this clonal lineage classification. Specific virulence on potato differentials was broadest among potato isolates, while specific virulence on tomato cultivars was broadest among tomato isolates. Some tomato isolates infected all tomato differentials but no potato differentials, indicating that specific virulence for the two hosts is probably controlled by different avirulence genes in P. infestans. In two separate experiments, the diameters of lesions caused by nine isolates from potato and 10 from tomato were compared on three tomato and three potato cultivars. All isolates produced larger lesions on the host from which they were isolated. No isolates were found that were highly aggressive on both tomato and potato. We conclude that there are two different populations of P. infestans in Ecuador and that they are separated by host.  相似文献

Eight Philippine races of Xanthomonas oryzae pv. oryzae have been identified based on virulence phenotypes observed on a set of five differential varieties. One of these, Race 3, was found to consist of two phylogenetically distinct lineages based on DNA fingerprinting analysis. To determine, if the two lineages could be differentiated based on host-specificity, 186 strains of Race 3 were analyzed with additional fingerprints and 76 selected isolates with additional differential rice varieties. The strains were separated into 36 haplotypes clustering in three groups (IS1113-B, -C, and -G) at the 75% similarity level. Isolates varied in their reaction to a rice line carrying the resistance gene Xa7, however, the variability was not consistent within lineage. Aggressiveness of isolates belonging to lineage IS1113-B and -G was significantly greater when tested during the dry season than when tested during the wet season. However, no such differences were evident for isolates from lineage IS1113-C, indicating that environmental effects presumably light and temperature are genotype-specific.  相似文献

从江苏省扬州、南通、常州和徐州等地水稻根、茎和种子分离获得内生细菌736个菌株,其中对稻瘟病菌、稻恶苗病菌、稻纹枯病菌和稻白叶枯病菌拮抗的菌株分别占20.7%、5.4%、3.1%和1.1%,且主要来自根和茎,并有24个和3个菌株分别对2种和3种病菌有拮抗活性。对稻瘟病菌和稻恶苗病菌拮抗的内生细菌转管培养20代后,多数菌株拮抗活性稳定,对其他两种病菌拮抗的菌株转管培养后则拮抗能力大都显著下降或丧失。经形态和生理生化鉴定,高拮抗菌株G87(对稻瘟病菌、稻恶苗病菌、稻白叶枯病菌拮抗)和J215(对稻瘟病菌、稻恶苗病菌拮抗)为枯草芽孢杆菌。针刺和剪叶接种试验表明,大多数水稻内生细菌不致病,少数(3.4%~4.8%)在人工接种条件下可有致病能力或潜在致病性。  相似文献

ABSTRACT In this survey, 799 isolates of Phytophthora infestans collected from potato crops in Israel during 1983 to 2000 were analyzed for mating type and sensitivity to metalaxyl, and 324 were analyzed for race structure. The A(2) mating type, first recorded in 1983, fully dominated the pathogen population from 1983 until 1991 (9 years). It was thereafter replaced by the A(1) mating type, which dominated the population during 1993 to 2000. Metalaxyl-resistant isolates were first recorded in 1982. During 1983 to 1991, the majority of the isolates were resistant. Isolates with intermediate sensitivity (I) to this fungicide were first observed in 1993, when both A(1) and A(2) mating types occurred in the population. The proportion of I isolates gradually increased, reaching 39 to 41% in 1997 to 1998, and then declined to approximately 15% in 1999 to 2000. Pathogenicity to nine potato differential cultivars was determined for 80 potato isolates collected in 1983 to 1991, to 11 potato differentials in 173 isolates collected in 1993 to 1998, and in 71 potato isolates collected in 1999 to 2000. The first population was composed of 5 races with race 1,3,4,7,8,10 predominating (76%), the second population was composed of 19 races with race 1,3,4,7,8,10,11 predominating (63%), and the third population exhibited 42 (34 new) races with no single predominating race. RG-57 DNA fingerprinting and allozymes loci assays of 23 isolates revealed that isolates collected during 1984 to 1986 belonged to the PO-57 lineage, whereas those collected during 1997 to 1999 belonged to the RFO-39 lineage. Among isolates collected during 1993 to 1995, two unreported DNA fingerprinting patterns were found. Severe late blight epidemics occurred in tomato crops during 1998 to 2000. Of 35 tomato isolates, 28 were A(1) and only 7 were A(2). Of these tomato isolates, 94% were sensitive to metalaxyl. Almost every isolate had a different race structure on the 11 potato differentials. When inoculated onto three tomato differential cultivars, tomato isolates showed a virulence much more enhanced than potato isolates. The data suggest the Israeli population of P. infestans has passed through three major genetic changes during the past 18 years: in 1983, 1993, and 1999. 从水稻作物根际分离筛选得到的 2 0 8个细菌菌株对稻瘟病病菌具有较强的抑制能力 ;抑菌圈直径在 2 0mm以上的有 65株。选用室内拮抗性较稳定的 8株细菌菌株经两年的田间防病试验 ,不同菌株间防效差异显著 ,其中两年防效较好的菌株是Ma 3 2 (蜡状芽孢杆菌 )和Xi 55(枯草芽孢杆菌 )。一些菌株处理种子对秧苗生长有促进作用。  相似文献

1993年我国小麦秆锈病发生较轻,从8个省18个区县中的47个品种上采集到的95个标样中分离到菌株336个,鉴定出21C3CKR、21C3CKH、21C3CTR、21C3CTH、21C3CFR、21C3CFH、34C1MKR、34C1MKH和34C2MKR等9个致病类型;21C3CKR的出现频率为65.2%,居于首位,21C3CDH为14.5%,21C3CTR为10.7%,21C3CFR为4.2%  相似文献
