ABSTRACT A meta-analysis of the effect of tebuconazole (e.g., Folicur 3.6F) on Fusarium head blight and deoxynivalenol (DON) content of wheat grain was performed using data collected from uniform fungicide trials (UFTs) conducted at multiple locations across U.S. wheat-growing regions. Response ratios (mean disease and DON levels from tebuconazole-treated plots, divided by mean disease and DON levels from untreated check plots) were calculated for each of 139 studies for tebuconazole effect on Fusarium head blight index (IND; field or plot-level disease severity, i.e., mean proportion of diseased spikelets per spike) and 101 studies for tebuconazole effect on DON contamination of harvested grain. A random-effects meta-analysis was performed on the log-transformed ratios, and the estimated mean log ratios were transformed to estimate the mean (expected) percent control for IND ( C(IND) ) and DON ( C(DON)). A mixed effects meta-analysis was then done to determine the effects of wheat type (spring versus winter wheat) and disease and DON levels in the controls on the log ratios. Tebuconazole was more effective at limiting IND than DON, with C(IND) and C(DON) values of 40.3 and 21.6%, respectively. The efficacy of tebuconazole as determined by the impact on both IND and DON was greater in spring wheat than in winter wheat (P < 0.01), with a 13.2% higher C(IND) and a 12.4% higher C(DON) in spring wheat than in winter wheat. ABSTRACT The association between Fusarium head blight (FHB) intensity and deoxynivalenol (DON) accumulation in harvested grain is not fully understood. A quantitative review of research findings was performed to determine if there was a consistent and significant relationship between measures of Fusarium head blight intensity and DON in harvested wheat grain. Results from published and unpublished studies reporting correlations between DON and Fusarium head blight "index" (IND; field or plot-level disease severity), incidence (INC), diseased-head severity (DHS), and Fusarium-damaged kernels (FDK) were analyzed using meta-analysis to determine the overall magnitude, significance, and precision of these associations. A total of 163 studies was analyzed, with estimated correlation coefficients (r) between -0.58 and 0.99. More than 65% of all r values were >0.50, whereas less that 7% were <0. The overall mean correlation coefficients for all relationships between DON and disease intensity were significantly different from zero (P < 0.001). Based on the analysis of Fisher-transformed r values ( z(r) values), FDK had the strongest relationship with DON, with a mean r of 0.73, followed by IND (r = 0.62), DHS (r = 0.53), and INC (r = 0.52). The mean difference between pairs of transformed z(r) values (z(d) ) was significantly different from zero for all pairwise comparisons, except the comparison between INC and DHS. Transformed correlations were significantly affected by wheat type (spring versus winter wheat), study type (fungicide versus genotype trials), and study location (U.S. spring- and winter-wheat-growing regions, and other wheat-growing regions). The strongest correlations were observed in studies with spring wheat cultivars, in fungicide trials, and in studies conducted in U.S. spring-wheat-growing regions. The effects of propiconazole, prothioconazole, tebuconazole, metconazole, and prothioconazole+tebuconazole (as a tank mix or a formulated premix) on the control of Fusarium head blight index (IND; field or plot-level disease severity) and deoxynivalenol (DON) in wheat were determined. A multivariate random-effects meta-analytical model was fitted to the log-transformed treatment means from over 100 uniform fungicide studies across 11 years and 14 states, and the mean log ratio (relative to the untreated check or tebuconazole mean) was determined as the overall effect size for quantifying fungicide efficacy. Mean log ratios were then transformed to estimate mean percent reduction in IND and DON relative to the untreated check (percent control: C(IND) and C(DON)) and relative to tebuconazole. All fungicides led to a significant reduction in IND and DON (P < 0.001), although there was substantial between-study variability. Prothioconazole+tebuconazole was the most effective fungicide for IND, with a C(IND) of 52%, followed by metconazole (50%), prothioconazole (48%), tebuconazole (40%), and propiconazole (32%). For DON, metconazole was the most effective treatment, with a [Formula: see text](DON) of 45%; prothioconazole+tebuconazole and prothioconazole showed similar efficacy, with C(DON) values of 42 and 43%, respectively; tebuconazole and propiconazole were the least effective, with C(DON) values of 23 and 12%, respectively. All fungicides, with the exception of propiconazole, were significantly more effective than tebuconazole for control of both IND and DON (P < 0.001). Relative to tebuconazole, prothioconazole, metconazole, and tebuconzole+prothioconzole reduced disease index a further 14 to 20% and DON a further 25 to 29%. In general, fungicide efficacy was significantly higher for spring wheat than for soft winter wheat studies; depending on the fungicide, the difference in percent control between spring and soft winter wheat was 5 to 20% for C(IND) and 7 to 16% for C(DON). Based on the mean log ratios and between-study variances, the probability that IND or DON in a treated plot from a randomly selected study was lower than that in the check by a fixed margin was determined, which confirmed the superior efficacy of prothioconazole, metconazole, and tebuconzole+prothioconzole for Fusarium head blight disease and toxin control.  相似文献

Stagonospora nodorum blotch, caused by Phaeosphaeria nodorum, is considered one of the most destructive foliar diseases of wheat in the United States. However, relatively little is known about the population biology of this fungus in the major wheat-growing regions of the central United States. To rectify this situation, 308 single-spore isolates of P. nodorum were analyzed from 12 populations, five from hard red spring wheat cultivars in Minnesota and North Dakota and seven from soft red winter wheat in Indiana and Ohio. The genetic structure of the sampled populations was determined by analyzing polymorphisms at five microsatellite or simple-sequence repeat (SSR) loci and the mating type locus. Although a few clones were identified, most P. nodorum populations had high levels of gene (H(S) = 0.175 to 0.519) and genotype (D = 0.600 to 0.972) diversity. Gene diversity was higher among isolates collected from spring wheat cultivars in North Dakota and Minnesota (mean H(S) = 0.503) than in those from winter wheat cultivars in Indiana and Ohio (H(S) = 0.269). Analyses of clone-corrected data sets showed equal frequencies of both mating types in both regional and local populations, indicating that sexual recombination may occur regularly. However, significant gametic disequilibrium occurred in three of the four populations from North Dakota, and there was genetic differentiation both within and among locations. Genetic differentiation between the hard red spring and soft red winter wheat production regions was moderate (F(ST) = 0.168), but whether this is due to differences in wheat production or to geographical variation cannot be determined. These results suggest that sexual reproduction occurs in P. nodorum populations in the major wheat-growing regions of the central United States, and that geographically separated populations can be genetically differentiated, reflecting either restrictions on gene flow or selection.  相似文献

Seven spring wheat, 13 barley, 14 oats and 20 winter wheat genotypes were inoculated at flowering in 1993 and 1994 with mixed conidial suspensions of 8 isolates of Fusarium culmorum. Four weeks after inoculation, head blight was recorded in the field. After harvest, seed infection was assessed by a Freezing Blotter Test in the laboratory. Seed samples were also analyzed for deoxynivalenol (DON) content. Differences were found in head blight rating, the levels of infected seeds and the DON content between wheat, barley, and oats and between cultivars. Highly significant correlations were found between the percentage of heavily infected seed and the DON content. The weighted mean value of infected seeds and DON content were also significantly correlated. No significant correlation was found between head blight rating in the field and DON content. The level of infected seeds, as determined by the Freezing Blotter Test, was a better indication of the DON content in the seeds than the head blight rating. This visual assessment of levels of infected seeds gives a reliable estimate of resistance to Fusarium.  相似文献

Fusarium head blight (FHB) resistance of 50 cultivars from the National List of winter wheat cultivars approved for sale (or were undergoing trails for approval in 2003) in the UK was compared with 21 reference cultivars from continental Europe which had previously been characterized for resistance. Only three UK National List cultivars (Soissons, Spark and Vector) had stable resistance over trial sites that was significantly greater than that of the FHB susceptible cultivar Wizard. In addition, under moderate disease pressure, 21 of the National List cultivars had levels of the trichothecene mycotoxin deoxynivalenol (DON) above the proposed European Union limit of 1·25 ppm in grain. Surveys show that levels of FHB and DON in the UK crop are currently very low, however, should disease pressure increase for any reason, then an improvement in the overall levels of FHB resistance of UK winter wheat germplasm will be required. In order to infer the origin of resistance and to identify potentially novel resistance, allele sizes of microsatellite (simple sequence repeat, SSR) markers linked to quantitative trait loci (QTL) for FHB resistance were compared between the test cultivars and known, characterized resistance sources. The major FHB resistance QTL from the Chinese cv. Sumai-3 (3BS, 5A and 6B), the Romanian cv. Fundulea F201R (1B and 5A) and the French cv. Renan (5AL) were screened with 17 SSRs. No National List cultivar had haplotypes similar to any of these QTL. However, the highly resistant German reference cultivar Petrus had an identical haplotype to cv. Fundulea F201R on 1B indicating that this cultivar has an allelic FHB resistance QTL at that location.  相似文献

Host plant preference of the Russian wheat aphid Diuraphis noxia (Kurdj.) was studied on 11 cultivars of 9 plant species: winter barley, spring barley, winter wheat, spring wheat, rye, oat, Triticale, canary grass, red millet yellow millet and maize. Seeds of the host plants were sown in a circle near the edge of pots. The host plant choice was evaluated 24 hours after releasing 55 Diuraphis noxia female adults in the middle of each pot. The suitability of different hosts for aphid development was evaluated 2, 7 and 14 days after infestation based on the mean number of Russian wheat aphid individuals per plant. Red millet, yellow millet and maize were chosen by significantly fewer aphids than grain crops. Winter and spring barley were chosen as hosts most frequently, and the progeny production was also the highest on these plants. The growth rate of D. noxia was significantly affected by the host plants and the date of assessment and their interaction.  相似文献

Pratylenchus thornei densities were monitored in field plots of two winter wheat cultivars in a dry farming area of southern Spain. Samples were taken fortnightly during the wheat-growing season and from the following dry fallow. Under bread wheat cv. ‘Yecora’, densities ofP. thornei increased for 5 to 6 months and then were maintained or slightly decreased thereafter, surviving the summer dry fallow in an anhydrobiotic state (78.2% and 85.3% survival in soil and roots, respectively). Under durum wheat cv. ‘Donpedro’, nematode densities decreased over the growing season, although densities within the roots increased during the first 2 months of the wheat-growing period, indicating that nematodes could penetrate the roots of this cultivar but were unable to reproduce. These observations suggest resistance of wheat cv. Donpedro toP. thornei.  相似文献

Twenty four isolates of Fusarium graminearum, half of which were 3-acetyldeoxynivalenol (3-ADON) and half 15-acetyldeoxynivalenol (15-ADON) chemotypes, were tested for their ability to produce deoxynivalenol and to cause Fusarium head blight (FHB) in spring wheat cultivars. The objectives of this study were to determine (1) whether 3-ADON isolates differ in aggressiveness, as measured by the FHB index, and DON production from 15-ADON isolates under field conditions, and (2) whether the performance of resistant host cultivars was stable across isolates. Field tests of all isolates were conducted with three replicates at each of two locations in Canada and Germany in 2008 with three host genotypes differing in FHB resistance level. The resistant host genotype showed resistance regardless of the chemotype or location. The differences between mean FHB indices of 3-ADON and 15-ADON isolates were not significant for any wheat genotype. In contrast, average DON production by the 3-ADON isolates (10.44 mg kg−1) was significantly (P < 0.05) higher than for the 15-ADON isolates (6.95 mg kg−1) at three of the four locations where moderately resistant lines were tested, and at both locations where susceptible lines were evaluated. These results indicate that 3-ADON isolates could pose a greater risk to food safety. However, as the mean aggressiveness and DON production of 3-ADON and 15-ADON chemotypes was similar on highly resistant lines, breeding and use of highly resistant lines is still the most effective measure of reducing the risks associated with DON in wheat.  相似文献

为明确自然发病条件下我国小麦主产区小麦籽粒镰刀菌毒素污染情况,用超高效液相色谱-串联质谱仪(UPLC-MS/MS)测定了长江中下游冬麦组(CZX)、黄淮冬麦区南片冬水组(HNS)、黄淮冬麦区北片水地组(HBS)、北部冬麦区水地组(BBS)180份小麦样品中DON、3ADON、15ADON、T-2、HT-2和ZEN共6种镰刀菌毒素含量并对其进行比较分析。结果显示:来自CZX小麦籽粒中的DON、3ADON与ZEN毒素含量显著高于其他麦区;T-2毒素在CZX小麦籽粒中含量显著低于其他麦区;15ADON毒素含量在HNS与CZX、HBS与BBS以及CZX与BBS差异不显著,其他麦区间该毒素含量差异显著,15ADON在HNS含量最高;检出率方面,HT-2毒素均未检出;ZEN、3ADON检出率较低,DON、15ADON和T-2毒素的检出率较高。  相似文献

Soilborne wheat mosaic virus (SBWMV) is one of the most important winter wheat pathogens worldwide. To identify genes for resistance to the virus in U.S. winter wheat, association study was conducted using a selected panel of 205 elite experimental lines and cultivars from U.S. hard and soft winter wheat breeding programs. Virus symptoms were evaluated twice in virus-infected fields for the panel at Manhattan, KS in spring 2010 and 2011 and for a subpanel of 137 hard winter wheat accessions at Stillwater, OK in spring 2008. At the two locations, 69.8 and 79.5% of cultivars were resistant or moderately resistant to the disease, respectively. After 282 simple-sequence repeat markers covering all wheat chromosome arms were scanned for association in the panel, marker Xgwm469 on the long arm of chromosome 5D (5DL) showed a significant association with the disease rating. Three alleles (Xgwm469-165bp, -167bp, and -169bp) were associated with resistance and the null allele was associated with susceptibility. Correlations between the marker and the disease rating were highly significant (0.80 in Manhattan at P < 0.0001 and 0.63 in Stillwater at P < 0.0001). The alleles Xgwm469-165bp and Xgwm469-169bp were present mainly in the hard winter wheat group, whereas allele Xgwm469-167bp was predominant in the soft winter wheat. The 169 bp allele can be traced back to 'Newton', and the 165 bp allele to Aegilops tauschii. In addition, a novel locus on the short arm of chromosome 4D (4DS) was also identified to associate with the disease rating. Marker Xgwm469-5DL is closely linked to SBWMV resistance and highly polymorphic across the winter wheat accessions sampled in the study and, thus, should be useful in marker-assisted selection in U.S. winter wheat.  相似文献

ABSTRACT The symptoms of tan spot of wheat, caused by Pyrenophora triticirepentis, include a tan necrosis component and an extensive chlorosis component. Since tan spot has become the major component of the leafspotting disease complex of wheat in western Canada, the need for resistant cultivars has increased. This study was conducted to determine whether the resistance to tan spot found in a diverse set of spring and winter wheat genotypes was due to resistance genes not previously reported. The genetic control of resistance to necrosis induced by P. triticirepentis race 1 and race 2 was determined, under controlled environmental conditions, for spring wheat genotypes Erik and 86ISMN 2137 and winter wheat genotypes Hadden, Red Chief, and 6B-365. Plants were inoculated at the two-leaf stage and disease reaction was assessed based on lesion type. Tests of the F(1) and F(2) generations, and of F(2:3) and F(2:8) families, indicated that one recessive gene controlled resistance to the necrosis component of tan spot caused by both race 1 and race 2 in each cross studied. Lack of segregation in crosses between the resistant cultivars indicated that the resistance gene was the same in all of the cultivars.  相似文献

通过23年(1970—1993)的研究,初步明确了东南方冬麦区黄矮病情、春麦区拔节初期麦二叉蚜量和东南方冬麦区麦蚜迁出迟早等是影响宁夏春麦区黄矮病流行的关键因素,黄矮病历年流行程度与春小麦生长期间的主要气象因子相关不密切。针对麦蚜远距离迁飞所致春麦黄矮病,首次提出了以2个预测式为基础的黄矮病流行趋势预测办法,并据此进行了多次成功的预报。  相似文献

A study was conducted to determine the susceptibility of popular United States (U.S.) and European winter wheat cultivars to the fungal pathogen Tilletia indica. Historically, the disease has been limited to autumn-sown spring-habit wheat areas and not associated with winter wheat. In 1997, Karnal bunt was observed on winter wheat in limited regions of Texas. This region marks the southern end of the contiguous U.S. central winter wheat belt, which extends north into Canada. The aim of this study was to assess the levels of disease resistance in winter wheat. Fifty U.S. and European winter wheat cultivars were tested using two different greenhouse inoculation procedures. For each cultivar, 12 spikes in boot were inoculated by boot-injection with a sporidial suspension (1.0 ml/boot, 10,000 spores ml−1), and 12 other emerged spikes were spray-inoculated with the same concentration. The experiment was repeated for three seasons. Among cultivars, mean seed infection ranged from 2.1 to 87.2% and 0 to 15.6% for boot-injected and spray-inoculated treatments, respectively. Results showed that the majority of winter wheat cultivars tested were susceptible to Karnal bunt.  相似文献

Pathogen development and host responses in wheat spikes of resistant and susceptible cultivars infected by Fusarium culmorum causing Fusarium head blight (FHB), were investigated by means of electron microscopy as well as immunogold labelling techniques. The studies revealed similarities in the infection process and the initial spreading of the pathogen in wheat spikes between resistant and susceptible cultivars. However, the pathogen’s development was obviously more slow in the resistant cultivars as in comparison to a susceptible one. The structural defence reactions such as the formation of thick layered appositions and large papillae were essentially more pronounced in the infected host tissues of the resistant cultivars, than in the susceptible one. β -1,3-glucan was detected in the appositions and papillae. Furthermore, immunogold labelling of lignin demonstrated that there were no differences in the lignin contents of the wheat spikes between susceptible and resistant cultivars regarding the uninoculated healthy tissue, but densities of lignin in host cell walls of the infected wheat spikes differed distinctly between resistant and susceptible cultivars. The lignin content in the cell walls of the infected tissues of the susceptible wheat cultivar increased slightly, while the lignin accumulated intensely in the host cell walls of the infected wheat spikes of the resistant cultivars. These findings indicate that lignin accumulation in the infected wheat spikes may play an important role in resistance to the spreading of the pathogen in the host tissues. Immunogold labelling of the Fusarium toxin DON in the infected lemma showed the same labelling patterns in the host tissues of resistant and susceptible cultivars. However, there were distinct differences in the toxin concentration between the tissues of the susceptible and resistant cultivars. At the early stage of infection, the labelling densities for DON in resistant cultivars were significantly lower than those in the susceptible one. The present study indicates that the FHB resistant cultivars are able to develop active defence reactions during infection and spreading of the pathogen in the host tissues. The lower accumulation of the toxin DON in the tissues of the infected spikes of resistant cultivars which results from the host’s defence mechanisms may allow more intensive defence responses to the pathogen by the host.  相似文献

Determination of the Fusarium protein equivalent (FPE) levels in kernels for better characterisation of genotypes showing Fusarium head blight (FHB) resistance, and better detection of susceptibility to kernel infection among genotypes with slight symptom expression was carried out. Twelve wheat cultivars and eight hexaploid winter wheat lines derived from a cross of Triticum aestivum with related species T. macha, T. polonicum, and T. dicoccoides were evaluated for levels of spike and kernel infection, the content of the mycotoxin deoxynivalenol (DON) and FPE in kernels after artificial inoculation with the fungus Fusarium culmorum in the field in 2006–2007. The ELISA immunochemical method was employed for the quantitative analyses of DON and FPE. Three wheat lines had a significantly low infection of spikes and kernels compared to cvs Sumai 3 and Nobeoka Bozu, indicating the presence of specific resistance mechanisms to FHB. The significantly low AUDPC (area under the disease progress curve) and the high level of FPE and DON content in kernels indicated a lack of resistance in one wheat line (crossed with T. polonicum). The results showed highly significant correlations (P < 0.01) between FPE and DON content and between FPE and AUDPC. In addition, correlations between FPE and reductions in yield components were also highly significant. Quantification of Fusarium spp. in wheat kernels can be helpful for evaluating wheat genotypes for their levels of resistance to FHB.  相似文献

ABSTRACT A susceptible synthetic winter rye population was inoculated with 42 isolates of Fusarium culmorum, originating from nine European countries and Australia, at two field locations in Germany. Significant (P = 0.01) genetic variation in aggressiveness of isolates of F. culmorum was observed across both field locations. Field samples were used to determine deoxynivalenol (DON), nivalenol (NIV), and ergosterol (ERG) contents. The 42 isolates also were incubated on rye grain in vitro, and DON and NIV contents were analyzed. Thirty-four isolates produced DON, and seven isolates produced NIV at both field locations and in vitro. Mean DON contents ranged from 0.5 to 64.6 mg/kg in grain from field trials and from 0.3 to 376.3 mg/kg in grain incubated in vitro; mean NIV contents ranged from 17.6 to 30.4 mg/kg in grain from field trials and from 0.8 to 381.0 mg/kg in grain incubated in vitro. No correlation was found between the DON content of field-grown grain and grain incubated in vitro. NIV-producing isolates originated from the Netherlands, Germany, Italy, and Australia. More aggressive isolates produced higher mean DON contents in grain in field trials (r = 0.69; P = 0.01). However, DON production rate per unit of fungal biomass, estimated as the DON/ERG ratio at harvest, was not correlated with aggressiveness. Toxin production seemed to be a common feature in F. culmorum. In vitro assays reliably distinguished DON- and NIV-producing types of F. culmorum; however, these assays could not predict production of DON by these isolates in the field.  相似文献
