This study determined the in vitro effects on the viability of internal parasites of grazing undrenched weaner deer on either chicory (Cichorium intybus) or perennial ryegrass (Lolium perenne)/white clover (Trifolium repens) pasture. One experiment investigated the hatching and development of gastrointestinal nematode eggs and larvae, and the development and motility of L1 lungworm (Dictyocaulus eckerti) larvae, and a second experiment used larval migration inhibition assays to test the viability of L1 lungworm larvae extracted from the faeces of weaner deer grazed on either chicory or pasture when they were incubated with rumen and abomasal fluids from fistulated deer also grazing on chicory or pasture. The incubations were undertaken with and without added condensed tannins purified from chicory and with or without polyethylene glycol (PEG) to bind the tannins. Chicory had no effect on the hatching and development of gastrointestinal nematode eggs and larvae. Grazing chicory reduced the number of lungworm larvae developing to the L3 stage, and L1 lungworm larvae from the faeces of chicory-grazed deer were less viable in rumen and abomasal fluid than larvae from pasture-grazed animals. Abomasal fluid was significantly (P < 0.001) less inhibitory to the migration of L1 lungworms than rumen fluid. When the larvae were incubated in rumen and abomasal fluids from chicory-grazed deer, their passage through sieves was significantly (P < 0.001) reduced in comparison with when they were incubated in the fluids from pasture-grazed deer Adding condensed tannins to rumen fluid increased the inhibition of the migration of L1 lungworm larvae but PEG removed this inhibition; this effect was not observed with abomasal fluid.  相似文献

In vitro and in vivo studies were conducted to determine possible direct anthelmintic effects of condensed tannins towards different ovine gastrointestinal nematodes. A larval development/viability assay was used to investigate the effect of a condensed tannin extract (Quebracho) towards larvae of Haemonchus contortus, Teladorsagia circumcincta and Trichostrongylus vitrinus. The development to infective larvae and their viability was assessed in all three species and LD 50 values were calculated. The presence of Quebracho extract in the cultures decreased the viability of L3 in all species; the LD 50 were not significantly different for the different species. Forty-eight sheep were allocated to one of eight groups and were infected with a single dose of either 4000 L3 H. contortus (groups 1 and 2) or 5000 L3 T. colubriformis and 5000 L3 Nematodirus battus simultaneously (groups 3-6) or 10,000 L3 of T. circumcincta (groups 7 and 8). From day 28 until day 31 of the experiment, sheep infected with the intestinal species were drenched with Quebracho extract at 4, 8 or 16% w/w of food intake, or remained as undrenched controls; sheep infected with the abomasal species were either drenched with Quebracho extract at 8% w/w of food intake or remained as undrenched controls. All sheep were slaughtered 4 days after the end of the drenching period. Sheep infected with the intestinal species and drenched with 16% w/w Quebracho had lower FEC compared to sheep drenched with 8% w/w (P<0.05), which in turn were lower than in sheep either drenched with 4% Quebracho or which remained undrenched (P<0.05). The lowest intestinal worm burden was recovered from sheep drenched with 8% w/w Quebracho extract (P<0.05). The administration of Quebracho extract at 8% of food intake for 3 days did not affect FEC or worm burdens in sheep infected with the abomasal species compared to controls.  相似文献

There was a significant negative linear relationship between dietary CT concentration and abomasal nematode burdens. No significant differences in faecal egg counts, lungworm burdens or voluntary feed intake were found. Deer fed sulla had higher liveweight gain, carcass weight and carcass dressing-out percentage, higher serum total protein and albumin concentration and lower serum gastrin concentration and faecal lungworm larval count, compared with lucerne-fed deer. Inclusion of sulla in diets for young red deer may reduce the impact of internal parasites and/or reduce the dependence on anthelmintic treatment.  相似文献

The susceptibility of Romanov and Lacaune sheep to nematode parasites was studied in a permanent flock grazing an extensive pasture. The output of strongyle eggs and protostrongyle larvae was higher in Romanov ewes. Faecal samples of the Romanov ewes more often harboured Nematodirus eggs while the larvae recovered from cultures of these samples contained a higher percentage of Teladorsagia. Romanov tracer lambs had a significantly heavier adult Teladorsagia burden. This higher natural susceptibility of the Romanov breed was confirmed by experimental infections with T. circumcincta and N. spathiger but not with Trichostrongylus colubriformis. In infections with Muellerius capillaris and Protostrongylus rufescens, the worm burdens were similar but parasite fertility (larvae per adult) seemed to be higher in Romanov tracer lambs.  相似文献

This research assessed the potential use of a low input forage containing a high amount of condensed tannins (CT) to reduce foodborne pathogens prior to slaughter of meat goats. In a completely randomized design, twenty Kiko × Spanish intact male kids (BW = 19.2 ± 0.74 kg) were fed ground sericea lespedeza [SL; Lespedeza cuneata (Dum-Cours) G. Don; 2 pens], a high-CT legume, or bermudagrass hay [BG; Cynodon dactyon (L.) Pers.; 2 pens], at 75% of daily intake with a corn-based supplement (25% of intake) for 14 weeks (n = 10 goats/treatment). At the end of the feeding trial, the animals were slaughtered using standard procedures. Immediately after evisceration, rumen and rectal samples were collected to assess bacterial loads and volatile fatty acids in the rumen. Concentrations of rumen volatile fatty acids were significantly different between dietary treatments. Goats fed SL hay had higher (P < 0.05) contents of butyric (8.66 vs 7.16 mM), isobutyric (1.94 vs 1.44 mM), isovaleric (3.03 vs 2.13 mM), and valeric (1.43 vs 1.07 mM) acids than those fed BG hay; however, the content of acetic acid (78.6 vs 64.4 mM) was higher (P < 0.05) in the BG-fed groups than in SL-fed groups. Escherichia coli (2.33 vs 1.13 log10 CFU/g) counts of rumen contents were higher (P < 0.05) in the SL-fed group compared with the BG-fed group. However, E. coli counts in feces were not different (P > 0.05) between dietary treatments. The high-CT influenced (P < 0.05) total plate counts in the feces; and the total plate counts in feces of SL- and BG-fed goats were 4.95 and 6.57 log10 CFU/g, respectively. The results indicated that high CT in the diet might influence rumen volatile fatty acid composition, but might not reduce the bacterial loads in gastrointestinal tracts of meat goats.  相似文献

One hundred and fifteen young red deer (Cervus elaphus), heavily infected with lungworm (Dictyocaulus viviparus) and lightly infected with gastro-intestinal nematodes, were divided into three groups. One group of 50 animals was treated with one adult sheep dose of a slow-release albendazole capsule, another group of 50 was dosed orally five times with liquid albendazole and 15 were left as untreated controls. The capsule eliminated faecal lungworm larvae during the 103-day trial period. There was a highly significant difference in faecal larval counts between the capsule-treated and control group. Over the trial period, the mean body weight gain of the untreated, liquid albendazole and capsule-treated animals was 0.1 kg, 4.5 kg and 7.8 kg respectively.  相似文献

OBJECTIVE: To evaluate efficacy of topically administered doramectin against eyeworms, lungworms, and gastrointestinal nematodes of cattle. ANIMALS: 400 cattle (20 cattle in each of 20 trials). PROCEDURE: Trials were conducted in North America; natural and experimentally induced infections were used. In each trial, cattle were allocated randomly to control (placebo [saline [0.9% NaCl] solution at 1 ml/10 kg of body weight] or untreated; n = 10) or doramectin-treated (500 microg/kg of body weight; 10) groups. Treatments were applied in a single passage along the midline of the back, from the withers to the tailhead. Cattle were euthanatized > or =14 days after treatment, and worm burdens were determined by use of standard techniques. RESULTS: Efficacy of doramectin was > or =95.3% against adults of Thelazia gulosa, T skrjabini, Dictyocaulus viviparus, Haemonchus contortus, H placei, Ostertagia lyrata, O ostertagi, Trichostrongylus axei, Bunostomum phlebotomum, Capillaria spp, Cooperia oncophora, C pectinata, C punctata, C spatulata, C surnabada, Nematodirus spathiger, Strongyloides papillosus, T colubriformis, Oesophagostomum radiatum, and Trichuris spp. Efficacy was 95.1% against fourth-stage larvae of D viviparus, H placei, O lyrata, O ostertagi, T axei, C oncophora, C punctata, C spatulata, C surnabada, N helvetianus, T colubriformis, O radiatum, and Trichuris spp. In addition, efficacy against inhibited fourth-stage larvae of O ostertagi and Ostertagia spp was > or =98.1%. CONCLUSIONS AND CLINICAL RELEVANCE: A single topical application of doramectin pour-on was efficacious against a broad range of nematode species in cattle.  相似文献

The use of tanniferous plants or tannins represents one alternative approach to the control of gastrointestinal parasites in ruminants but most data have been obtained in sheep. The current study was therefore performed in goats with two objectives: firstly, to investigate the effects of condensed tannins (CT) on adult populations of Trichostrongylus colubriformis and Teladorsagia circumcincta; secondly, to examine their effects on the establishment of infective larvae of these two species. In experiment 1, two groups of kids were infected with 6 000 L3 of T. colubriformis and 6 000 L3 of T. circumcincta. After 7 weeks, quebracho extracts were administered per os for 8 days to one group. A comparable group which did not receive tannins was included as the control. The kids were slaughtered on week 11. Parasitological and pathophysiological parameters were measured weekly. Worm counts were assessed and mast cells, globule leukocytes and eosinophils were counted in the abomasal and intestinal mucosae. Tannin administration was associated with a decrease in egg excretion, and a decrease in female fecundity, but with no changes in worm numbers. These changes were associated with an increased number of intestinal mast cells. In experiment 2, 24 goats were used according to a 2 x 2 factorial design, depending on infection and tannin administration. Two groups were either infected with 6 000 L3 of T. colubriformis or T. circumcincta. Within each group, the goats were either drenched or undrenched with tannin extracts. Pathophysiological parameters were measured weekly. Twelve days after the cessation of tannin administration, the goats were slaughtered. Worm counts and female worm fecundity were determined. Tannin consumption was associated with a significant reduction (P < 0.01) of Trichostrongylus populations and a close to significant reduction for Teladorsagia. No effect on fecundity was observed. Our results (1) confirm the consequences of condensed tannins on nematodes in goats as in sheep and (2) indicate divergent effects depending on the parasitic stage exposed to the condensed tannins.  相似文献

The effect of erythrosin B and visible light on 3rd stage larvae of gastrointestinal nematodes of naturally-infected cattle was studied. Cattle were treated orally with erythrosin B at dosages of 30 and 40 mg kg-1 day-1 for as many as 17 days. Feces from treated and untreated animals were collected and prepared for culture. Third stage larvae were then collected by Baermannization and exposed to light. Both sunlight and artificial fluorescent light were shown to be toxic to 3rd-stage-larvae after treatment with erythrosin B. This toxic reaction was significant after only 2 consecutive daily treatments.  相似文献

通过对红三叶(Trifolium pratense)、白三叶(Trifolium repens)、高丹草(Sorghum sudanense)、紫花苜蓿(Medicago sativa)接种AM真菌,测定植物根际AM真菌菌根侵染率、孢子密度,地上和地下部分生物量,了解宿主植物与AM真菌之间的协同作用,明确AM真菌对牧草的促生长效应,并筛选与4种牧草匹配的AM真菌。结果表明,AM真菌可侵染4种牧草且均能产生孢子,但不同牧草的侵染率和孢子密度存在差异。当对高丹草接种Glomus mosseae、Glomus intraradices、Glomus etunicatum、Glomus cladoideum、Glomus microagregatum、Glomus caledonium(简称G6),对红、白三叶接种Glomus intraradices(简称Gi),对苜蓿接种Glomus mosseae(简称Gm)时产量提高水平较对照组最为明显,接种AM真菌有效促进了植株的生长发育,提高了牧草的产量。  相似文献

Results of real-time PCR analysis of coproculture third stage larvae (L3) using genus specific TaqMan minor groove binder probes were compared with the results of morphological differentiation of L3 after coprocultured and direct morphological worm differentiation from gastrointestinal samples of eight sheep with naturally acquired nematodes infections. Faecal egg counts prior to postmortem confirmed infections with trichostrongyles with a geometric mean count of 4828 eggs per gram for all sheep. Individual egg counts correlated positively with total worm counts (correlation coefficient 0.794). Five different nematode species and one genus were found in the abomasi and small intestines: Cooperia curticei, Haemonchus contortus, Nematodirus spp., Teladorsagia (Ostertagia) circumcincta, Trichostrongylus axei and Trichostrongylus colubriformis. Coproculture of faecal eggs yielded five of these, Cooperia spp., Haemonchus spp., Ostertagia/Teladorsagia spp. and Trichostrongylus spp. Comparison between morphological L3 and worm differentiation data showed high congruence (94%). The agreement between PCR analysis of L3 after coproculture and direct morphological worm differentiation was 84%. Thus, real-time PCR was found to be suitable as a speedy and reliable diagnostic tool for the assessment of gastrointestinal nematode infections of ruminants in the field.  相似文献

The effects of condensed tannins extracted from seven forages on the viability of the eggs and first stage (L1) larvae of the sheep nematode Trichostrongylus colubriformis were evaluated in in vitro assays. The extracts of condensed tannins were obtained from Lotus pedunculatus (LP), Lotus corniculatus (LC), sulla (Hedysarum coronarium), sainfoin (Onobrychus viciifolia), Dorycnium pentaphylum (DP), Dorycnium rectum (DR) and dock (Rumex obtusifolius). Extracts containing 200 to 500 microg/ml reduced the proportion of eggs that hatched. The larval development assay was used to evaluate the effect of the extracts on the development of either eggs or L1 larvae to L3 infective larvae. Development was allowed to proceed for seven days by which time the larvae in control incubations had reached the infective L3 stage. Extracts containing 200 microg/ml from LP, DP, DR or dock prevented egg development, and only 11, 8 and 2 per cent of the eggs developed to L3 larvae with extracts from LC, sulla and sainfoin, respectively. When the concentration was 400 microg/ml no eggs developed to L3 larvae. The addition of the extracts after hatching also inhibited the development of L1 to L3 larvae; 200 microg/ml extracted from LP, LC, sulla, sainfoin, DP, DR and dock resulted in only 14, 18, 17, 15, 14, 16 and 4 per cent of L1 larvae developing to the L3 stage compared with 85 per cent for controls, and 400 microg/ml further reduced the development of L1 larvae. Statistical analyses showed that when the extracts were added before hatching they were significantly (P<0.001) more effective at inhibiting the larval development than when they were added after hatching. The condensed tannins from dock had the greatest inhibitory effect on egg development followed by the tannins from DR, sainfoin, DP, LP, sulla and LC.  相似文献

On a series of pasture plots, 2 kg pats of bovine faeces containing known numbers of strongylid (Haemonchus, Cooperia, Oesophagostomum and Trichostrongylus) eggs were deposited at intervals of 4 weeks from July 1995 to June 1996. The plots were sampled every 2 weeks after contamination and infective larvae were identified and counted. Larvae of all the genera developed throughout the year, but the pats exposed during the rainy season yielded more abundant larvae on the herbage. Irrespective of the season of deposition of the pats, larvae were found in larger numbers from 2 to 6 weeks after deposition and generally declined to below detectable levels within 12 to 16 weeks of contamination. The comparatively short survival times noted in this experiment may present opportunities for manipulation of the population dynamics of the gastrointestinal nematodes in the tropical environment of Kenya.  相似文献

The immune expulsion of gastrointestinal nematodes is impaired during lactation by the inhibition of the lymphocyte mediated component. As a result the population of worms may increase. At the same time, the fecundity of female worms in previously immune hosts may increase to approach that of worms in fully susceptible animals. There is good evidence that these events are primarily endocrinal in origin but although prolactin is probably involved, certain anomalies exist. These may reflect differences in the secretion pattern of the hormone in different phases of reproduction or they may indicate an interrelationship with a second hormone, for instance, adrenal glucocorticoids.  相似文献

The phototoxic effect of erythrosin B on the infective third-stage larvae (L3) of naturally acquired mixed populations of ovine gastrointestinal nematodes was investigated. This xanthene dye was phototoxic when administered orally to parasitized lambs or applied directly to feces containing nematode ova. Phototoxicity was assessed by the lack of motility (non-swimming) exhibited by the L3 following their collection by Baermannization from cultured feces and exposure to fluorescent light for 360 min. When lambs were administered erythrosin B orally at dosages of O (control), 40, 60 and 80 mg dye kg-1 body weight daily for 10 consecutive days, the percentages of non-swimming L3 were 16%, 46%, 55% and 62%, respectively. However, erythrosin B phototoxicity did not persist after administration of the dye was discontinued and the percentage of non-swimming L3 declined to a level similar to that of the untreated controls within 2 days. The highest percentage of non-swimming L3 was observed when erythrosin B was added directly to feces containing nematode ova. A dose-response curve was evident from the successively higher percentages of non-swimming L3 with increasing concentrations of erythrosin B. Xanthene dyes have the potential to control parasites acquired by livestock on pasture by inducing a phototoxic reaction in the infective L3.  相似文献

The influence of a priming infection with gastrointestinal nematodes on the subsequent establishment of lungworms was studied. Repeated inoculations of calves with Ostertagia ostertagi and Cooperia oncophora during 3, 5 or 7 weeks resulted in an establishment of lungworms that was 191% of the establishment found in na?ve controls. The higher take was associated with a higher faecal output of lungworm larvae and with lower weight gains of calves. The effect was not significantly influenced by the duration of the primary infections or by the actual presence or absence of worms in the gastrointestinal tract.  相似文献
