Barrows with an average initial weight of 55 kg were fitted with simple T-cannulas at the distal ileum. The animals were fed a protein-free diet that consisted of 79.7% cornstarch, 10% sucrose, 3% Alphafloc (a source of cellulose), 3% canola oil and a vitamin-mineral premix. The pigs were fed 700 g of diet twice each day, at 0800 and 2000. A balanced amino acid mixture or a saline solution was administered intravenously while the protein-free diet was fed. Ileal digesta were collected for 24 h following a 7-d adaptation period. The administration of amino acids reduced (P less than .05) the recovery of endogenous protein from 18.5 to 12.7 g per kg dry matter intake. For the amino acids, the reduction was only significant (P less than .05) for proline, from 3.6 to .6 g per kg dry matter intake. If the total endogenous protein losses are assumed to be constant and the differences in the amino acid composition of non-reabsorbed endogenous protein, as observed in this study, are used to calculate true ileal digestibilities, differences in the digestibilities of the indispensable amino acids are large (up to 7.4 percentage units for threonine). The amino acid composition of endogenous protein determined in pigs fed a protein-free diet and parenterally administered with amino acids should provide a better estimate for the calculation of true amino acid digestibilities when based on the determination of true protein digestibility by the 15N-isotope dilution technique.  相似文献

Twelve gilts, with an average initial BW of 38 kg, were fitted with a simple T-cannula at the distal ileum and two catheters in the external jugular veins. The animals were fed 700 g of one of the experimental diets, twice daily at 0800 and 2000. Diets 1 and 2 were cornstarch-based diets formulated to contain 16% CP from soybean meal and canola meal, respectively. Diets 3 and 4 contained 93.9% wheat and barley, respectively. Chromic oxide was included to determine nutrient digestibilities. Following recovery from surgery, 15N-leucine was infused continuously via a jugular catheter for 9 d at a rate of 40 mg.kg BW-1.d-1. Blood samples were taken at every feeding. Ileal digesta were collected for 24 h on d 7 and 9 of the infusion period. Assuming that the 15N-enrichment excess in the trichloroacetic acid-soluble fraction of the blood was similar to that in endogenous N, the contribution of endogenous to total N in ileal digesta was calculated. The amount of endogenous protein (N x 6.25) recovered in digesta collected from the distal ileum were 25.5, 30.5, 27.4 and 27.7 g/kg DM intake for the pigs fed diets 1, 2, 3 and 4, respectively. In the same order for the diets, the real ileal protein digestibilities were 97.5, 84.1, 99.0 and 94.2%, respectively. In addition, the real ileal amino acid digestibilities were calculated. The recoveries of endogenous protein in ileal digesta and the real ileal protein and amino acid digestibilities were higher than those determined by feeding protein-free diets for determining the recovery of endogenous protein and amino acids in ileal digesta.  相似文献

The aim of the study was to determine and compare the endogenous ileal excretions of nitrogen and amino acids under protein-free and peptide alimentation by the dog and rat. Two diets were prepared, one that was devoid of protein and the other containing 23% enzyme hydrolysed casein. Chromic oxide was included in the diets as an indigestible marker. A total of 10 mixed breed dogs were fed hourly either a protein-free or enzymatically hydrolysed casein diet for a total of 10 days. A faecal sample was obtained from each dog on day 9 while digesta was obtained from the terminal 20 cm of the ileum directly after euthanasia on day 10. A total of 12 8-week-old Sprague-Dawley rats received the same diets as the dogs. A faecal sample from each rat was obtained on day 7 while ileal digesta samples were obtained on day 8. The endogenous ileal excretions of most amino acids were greater in the dogs and rats that received the enzymatically hydrolysed casein diet compared with those receiving the protein free diet. Whereas the pattern of endogenous amino acid excretion was similar in the rats and dogs, the dogs excreted a significantly greater amount of nitrogen (1.91 vs. 2.27 and 1.63 vs. 4.12 g/kg dry matter intake for the protein-free and peptide alimentation method, respectively) and all amino acids except for glycine, isoleucine and leucine. Endogenous ileal amino acid excretions are higher in dogs compared to omnivorous animals such as rats and pigs but similar to the carnivorous cat.  相似文献

An experiment was conducted to investigate the effects of BW, feed intake, and the physiological condition of the animal on the loss and amino acid composition of endogenous protein in swine. Ten growing barrows and five multiparous sows were equipped with a T-cannula in the distal ileum for digesta collection. A protein-free diet was fed to all animals. The barrows were given free access to the experimental diet. The sows also were allowed to consume the diet on an ad libitum basis, and digesta were collected during lactation and in the following gestation period. In addition, digesta from the gravid sows were collected after restricting the sows to 2 kg of feed per day. For each animal group, the endogenous losses of protein and amino acids were calculated in relation to DMI, and the amino acid composition of endogenous protein was calculated. The total endogenous gut protein loss at the distal ileum of growing pigs, lactating sows, and gestating sows, given free access to feed, was 12.4, 9.4, and 11.2 g/kg DMI, respectively. These values were not different (P > .10). However, when gestating sows were fed only 2 kg/d, 17.8 g of endogenous protein was lost per kilogram of DMI, which was higher (P < .05) than for any of the other groups. This difference was mainly caused by higher (P < .05) losses of glycine, proline, and serine. There were no differences (P > .05) in amino acid composition of endogenous protein between growing pigs, lactating sows, and gestating sows given free access to feed, but restricted-fed gestating sows had an amino acid composition of endogenous protein that was significantly different from that of the other groups. The results from the experiment showed that age, BW, and the physiological condition of the animal have little or no effect on the amount of endogenous protein and amino acids lost at the distal ileum of hogs if calculated in relation to DMI. Likewise, the amino acid composition was not affected by the BW or physiological condition of the animal. However, DMI had a significant effect on endogenous protein losses in sows as well as on amino acid composition of endogenous protein.  相似文献

Eight male Holstein calves (body weight 68 +/- 5 kg; age 75 +/- 6 d), each with a permanent re-entrant pancreatic cannula and T-type ileal and duodenal cannulas, were used in a crossover design with four animals per group to determine amino acid kinetics and digestibilities in the digestive tract of calves fed soybean meal (SBM) and canola meal (CM) protein. The SBM and CM diets were fed twice daily at a level of 900 g at each feeding time (air-dry basis). With the exception of methionine, crude protein and amino acid flows at the proximal duodenum, expressed as a percentage of intake, were not influenced by dietary protein source. Apparent ileal and total tract digestibilities of CP and amino acids were reduced (P less than .05) by feeding CM compared to SBM, but apparent ileal digestibility of methionine was not affected by dietary protein source. Except for methionine, net disappearance of all amino acids in the small intestine, relative to the amount fed, was higher for the SBM diet than for the CM diet. Net disappearance or synthesis of amino acids in the large intestine were not affected by dietary protein source. Similarly, dietary protein source did not affect (P greater than .05) the secretion of pancreatic juice or concentrations of protein, chymotrypsin and trypsin in pancreatic juice. Soybean meal protein has higher ileal and total gastrointestinal tract digestibility than CM protein for young, growing calves.  相似文献

Dietary guar gum and cellulose were studied as purified soluble and insoluble nonstarch polysaccharide (NSP) sources, respectively. A control diet containing 14% cornstarch was formulated. A 7% guar gum, a 7% cellulose, and a 7% guar gum + 7% cellulose diet were formulated by adding the NSP to the control diet at the expense of cornstarch (wt/wt), forming a 2 x 2 factorial arrangement. The objectives were to determine whether guar gum and cellulose altered 1) the passage rate of digesta through the small intestine and total tract; 2) the digestibility of energy and CP, characteristics of the digesta, and microbial populations in the ileum; 3) plasma glucose and ghrelin concentrations; and 4) short-term voluntary feed intake and growth performance of grower pigs. In Exp. 1, 12 pigs (27.0 +/- 1.5 kg of BW) were fitted with an ileal T-cannula and were used in a 2-period change-over design, providing 6 observations per diet. Each period included 18 d: a 12-d acclimation period followed by 2-d feces, 3-d digesta, and 1-d venous blood collection periods. In Exp. 1, guar gum and cellulose slowed the passage rate of digesta through the small intestine by 26 and 18%, respectively (P < 0.05). Guar gum increased total tract retention time of the digesta by 14% (P < 0.05). Guar gum and cellulose increased the viscosity of ileal digesta by 72 and 76%, respectively (P < 0.05). Cellulose reduced ileal energy and CP digestibility (P < 0.05), but guar gum only tended to decrease ileal energy digestibility (P < 0.10). Guar gum and cellulose reduced total tract energy and CP digestibility (P < 0.05). At 60 min after feeding, guar gum decreased plasma glucose by 10% (P < 0.10). Guar gum interacted with cellulose to reduce plasma ghrelin before and after feeding (P < 0.05). Guar gum and cellulose interacted to increase ileal bifidobacteria and enterobacteria (P < 0.05); however, guar gum, but not cellulose, increased ileal clostridia (P < 0.05). In Exp. 2, 20 individually housed grower pigs (5 pigs per diet) had free access to the 4 diets used in Exp. 1 for 14 d. Guar gum and cellulose decreased ADG and reduced ADFI on d 0 to 14 (P < 0.05). In summary, increasing purified NSP in the diet reduced the passage rate of digesta, energy and protein digestibility, and feed intake, but increased ileal bifidobacteria and enterobacteria populations. The effects of cellulose were similar to those of guar gum. In conclusion, monitoring of dietary NSP is a critical factor to achieve predictable digestible nutrient intake and intestinal bacterial populations.  相似文献

The apparent ileal digestibilities of amino acids and rate of passage were evaluated in pigs (BW = 78.3 +/- 7.4 kg) fed a semipurified diet. The pigs were fed 1.82, 2.73, or 3.65 kg DMI/d. The highest level of feed intake was considered to be ad libitum feeding. The pigs were fed according to a 3 x 3 Latin square design and were allowed to adapt to each experimental diet for 5 d. This was followed by 1 d of continuous collection of ileal digesta and a 2nd d of continuous collection separated into six 2-h postprandial time blocks. Ytterbium chloride hexahydrate was used to determine rate of passage. The ileal digestibilities of amino acids and rate of passage were unaffected (P > 0.05) by level of feed intake. The use of correction factors to more accurately express amino acid concentrations in the diet and digesta affected (P < 0.05) the apparent ileal digestibility coefficients of some amino acids.  相似文献

Six barrows of approximately 37 kg BW, fitted with two simple T-cannulas in the duodenum (25 cm posterior to the pylorus) and terminal ileum (12 to 15 cm anterior to the ileocecal junction), were fed two diets containing 2.1 g of P/kg in the form of phytic acid and a low intrinsic phytase activity (corn-soybean meal based diet [Diet A] or a typical Dutch diet [Diet B]) without or with supplementary microbial phytase from Aspergillus niger (var. ficuum) equal to 1,500 phytase units per kilogram of diet, in a crossover design. The apparent duodenal, ileal, and total tract (overall) digestibilities of DM, total P, and phytate P (phytic acid x .282) were calculated using both Cr-NDR (neutral detergent residue mordanted with Cr) and Co-EDTA as dual-phase markers. Concentration of total P in the ileal digesta (P less than .01) and feces (P less than .001) of pigs fed microbial phytase was lower than without this enzyme, irrespective of the diet. Ileal digestibility of total P was 18.5 and 29.8 percentage units higher (which was a 1.7- to 2.9-fold increase) due to added Aspergillus niger phytase (P less than .05). Also, total tract (overall) digestibility increased by 27.0 to 29.7 percentage units (P less than .01). Phytic acid concentration in the duodenal and ileal digesta of pigs receiving microbial phytase was lower (P less than .01 or .001), resulting in its higher ileal digestibility (dephosphorylation rate) by 50.1 percentage units for Diet A and by 75.4 percentage units for Diet B. Irrespective of the treatment, no phytase activity could be detected in the ileal digesta of pigs.  相似文献

Two trials were conducted with 15 wethers surgically equipped with duodenal and ileal cannulas to study the utilization of P, Ca, Mg, Cu, Fe and Zn from swine waste and broiler litter. For each trial, animals were fed a low-P basal diet until serum inorganic P averaged 5.5 mg/dl; then they were allotted at random to the following 50% DM ensiled diets: low-P basal, basal + swine waste, basal + broiler litter, basal + dicalcium phosphate and basal + soybean meal. Each trial consisted of a 7-d preliminary period, a 7-d collection of feces and urine and 6-d sampling of duodenal and ileal digesta and feces. Apparent P absorption was not different (P greater than .05) between sheep fed waste-supplemented diets (37%) and those fed the conventionally supplemented diets (28%). Phosphorus absorption, calculated by difference, tended (P less than .1) to be higher from the waste supplements (59%) than from dicalcium phosphate and soybean meal (37%). Less (P less than .05) Ca was absorbed from the waste diets (.62 g/d) than from the conventional diets (1.28 g/d). More (P less than .05) Cu (mg/d) was absorbed from the waste diets, but no difference was found when absorption was expressed as percentage of intake. Broiler litter and swine waste were good sources of available P and Mg for ruminants.  相似文献

Two experiments with growing pigs were conducted to determine the effects of dietary P and Ca level, phytase supplementation, and ileal pectin infusion on ileal and fecal P and Ca balance, chemical composition of fecal mixed bacterial mass (MBM), and bacterial metabolic activity. Pigs (initial BW = 30 kg) were fitted with simple T-cannulas at the distal ileum. They were fed a low-P corn-soybean meal control diet (3 g of P/kg) or the control diet supplemented with monocalcium phosphate (MCP; 7 g of P/kg; Exp. 1) or 1,000 FTU phytase/kg (Exp. 2). The daily infusion treatments consisted of 60 g of pectin dissolved in 1.8 L of demineralized water or 1.8 L of demineralized water as the control infusion, infused via the ileal cannula. In each experiment, 8 barrows were assigned to 4 dietary treatments according to a double, incomplete 4 x 2 Latin square. The dietary treatments in Exp. 1 were the control (Con-) diet with water infusion; the control (Con+) diet with pectin infusion; the MCP diet with water infusion; and the MCP diet with pectin infusion. In Exp. 2, the pigs received the same Con- and Con+ treatments as in Exp. 1 and, in addition, the phytase-supplemented diet in combination with water or pectin infusion. Studies were carried out with six barrows (initial BW 50 kg) to determine the effect of two fiber sources on the apparent ileal and fecal N and amino acid digestibilities and bacterial N excretion in feces. The pigs, fitted with a simple T-cannula at the distal ileum, were fed three cornstarch-based diets formulated to contain 16% CP from soybean meal according to a repeated 3 x 3 Latin square design. Diet 1 did not contain additional fiber. Diets 2 and 3 contained 10% powdered cellulose and 10% barley straw (ground through a 2-mm mesh screen), included at the expense of cornstarch, respectively. The pigs were fed 900 g at each meal twice daily, 0800 and 2000. Feces were collected for 48 h after a 10-d adaptation period. Thereafter, ileal digesta were collected for 24 h. Chromic oxide was used as a digestibility marker. With the exception of leucine, the inclusion of fiber did not affect (P greater than .05) the ileal digestibilities of the indispensable amino acids. The values for the average of the ileal digestibilities of the indispensable amino acids were 85.3, 82.9, and 83.2% for diets 1, 2, and 3, respectively. On the other hand, the inclusion of fiber decreased (P less than .05) the fecal digestibilities of all the indispensable amino acids. The values for the average of the fecal digestibilities of the indispensable amino acids were 91.7, 87.4, and 89.1% for diets 1, 2, and 3, respectively. These decreases resulted from an increase (P less than .05) in the excretion of bacterial N, which was measured using 2,6-diaminopimelic acid as a marker. Sixteen male growing pigs of about 24 kg BW were fitted with both a duodenal re-entrant and a post-valve T-shaped cannula inserted in the caecum. The animals were divided into four groups. Each group received one of the following diets: corn starch-soybean protein isolate-based diet without (diet C) and with carboxymethylcellulose (diet CMC) or a rye-wheat-based diet without (diet RW) and with xylanase addition (diet RWX). The diets provided similar levels of apparent precaecal digestible crude protein (CP), lysine, methionine + cystine, threonine and tryptophan. Additionally, [15N]-yeast was given with the diets during the first 10 days of the experiment. For estimation of digesta viscosity, N-flow of dietary and endogenous origin, apparent precaecal digestibilities of dry matter (DM), CP, amino acids and non starch polysaccharides (NSP) (only in pigs fed diets RW and RWX), ileal and duodenal digesta were quantitatively collected on day 16 and 17, respectively. The endogenous N-proportion was measured by the ratio of 15N enrichment in the digesta and urine. The duodenal and ileal digesta supernatant viscosity increased as carboxymethylcellulose was included into the diet. Xylanase addition to the rye-wheat based diet reduced the viscosity in the ileal digesta. There were no differences in precaecal digestibilities of DM, CP and amino acids between diet C and CMC. The precaecal digestibilities of DM and soluble and insoluble NSP increased from 69.5% to 73.9%, from 1.3% to 47.9% and from 17.0% to 35.4%, respectively, as xylanase was added to the rye-wheat-based diet. The apparent precaecal digestibility of most essential amino acids increased by 2 to 5 percent units. The amounts of endogenous N at the duodenal level were estimated to be 158, 233, 313 and 276 mg per 12 h per kg0.75 BW of pigs fed diets C, CMC, RW and RWX, respectively. The corresponding values at the ileal level were 95, 107, 164 and 150 mg per 12 h per kg0.75 BW. For endogenous N amounts, significant differences were observed between diets C and CMC (duodenum) and also between semi-purified and cereal-based diets (duodenum and ileum). Methodological aspects for the estimation of endogenous N using the isotope dilution technique are discussed. Obviously, the digesta viscosity per se does not affect the nutrient absorption and endogenous N flow within the small intestine of pigs. Other properties of complex dietary fibre, digesta passage rate or bacterial activity probably contribute to the observed changes.  相似文献

Ileal digesta samples were collected continuously for 24 h on d 7 and 9 of the infusion period. 15N-enrichment excess in leucine and isoleucine in the trichloroacetic acid (TCA)-soluble fraction of blood and in digesta were measured using single-ion monitoring gas chromatography-mass spectrometry. Assuming that the 15N-enrichment excess in leucine and isoleucine in the TCA-soluble fraction of the blood is similar to that in endogenous protein secreted into the digestive tract and that the amino acid composition of endogenous protein is constant, the contribution of endogenous to total protein in ileal digesta was calculated using the 15N-isotope dilution technique for leucine and isoleucine, respectively. These contributions were much smaller for the 15N-leucine than for the 15N-isoleucine isotope dilution technique: 7.1 to 11.0 vs 21.8 to 24.9 g of protein/kg of DM intake. The values obtained with the 15N-isoleucine isotope dilution technique were close to those obtained with the 15N-isotope dilution technique as described in a previous publication.  相似文献

Eight crossbred steers (avg wt, 350 kg) with duodenal and ileal cannulas were used in two experimental periods in a single-reversal design to determine the effects of monensin on site and extent of apparent absorption of Mg and Ca. Steers were fed a cottonseed hull:concentrate diet (30:70) with or without 25 mg monensin/kg. The diet contained .20% Mg and .38% Ca. Each diet contained .25% Cr2O3 as a digesta marker. During each period, steers were fed the control diet for 10 d before allotment to treatments. Each period consisted of 17 d for diet adjustment and 6 d for collection. During the collection phase, fecal samples were obtained beginning on d 1 and continuing through d 6 at 12-h intervals progressing 2 h each day. Apparent Mg availability (grams apparently absorbed expressed as a percentage of grams of intake) increased (P less than .05) from 18.4 to 32.5% when monensin was fed. Feeding monensin increased (P less than .05) the apparent Mg availability and amount of Mg absorbed in the preintestinal region (47.7 vs 29.8% and 8.5 vs 5.1 g/d, respectively). Larger quantities (P less than .05) of Mg were secreted into the small intestine when monensin was fed. Monensin did not alter (P less than .05) apparent Ca availability or absorption in any segment of the digestive tract. These data indicate that monensin increased Mg and did not change Ca availability in the preintestinal region.  相似文献

Experiments were conducted to determine the effect of the physiological condition of swine on standardized ileal digestibility coefficients (SID). The apparent ileal digestibility coefficients were determined for crude protein and amino acids in six feed ingredients (corn, barley, wheat, soybean meal, canola meal, and meat and bone meal) in growing pigs and in gestating and lactating sows. Growing pigs and lactating sows were given free access to their diets, whereas gestating sows were allowed to consume only 2 kg of feed daily. The nonspecific (basal) endogenous losses of protein and amino acids were determined under similar feeding regimens after feeding a protein-free diet. The SID for crude protein and amino acids were calculated by correcting the apparent ileal digestibility coefficients for the nonspecific endogenous losses of protein and amino acids. With a few exceptions, there were no differences (P > 0.05) in the SID for crude protein and amino acids between growing pigs and lactating sows. Overall, gestating sows had higher (P < 0.05) SID for crude protein and all amino acids, except for tryptophan and aspartate, compared with growing pigs. Likewise, the SID of most amino acids obtained by gestating sows were higher (P < 0.05) than those obtained by lactating sows. Interactions (P < 0.05) between animals and diets were observed for gestating sows compared with growing pigs as well as gestating sows compared with lactating sows. As a consequence, it is not possible to extrapolate data from one feed ingredient to another. On most occasions, the lowest SID among the indispensable amino acids was calculated for threonine, valine, and lysine. It is concluded that gestating sows fed 2 kg of feed per day have higher standardized digestibility coefficients than do growing pigs and lactating sows given free access to their diets. This difference may be due to differences in daily feed intake rather than to the physiological status of the animals.  相似文献

Two experiments were conducted to determine dietary factors affecting digesta viscosity. In experiment 1, six ileal-cannulated dogs were either full (607.1 kJ/BW0.67) or restricted (303.5 kJ/BW0.67) fed. In experiment 2, four dog foods were surveyed for effects on digesta viscosity based on protein source [animal protein (AP) or plant protein (PP)] or dietary fibre source [soluble dietary fibre (SDF) or insoluble dietary fibre (IDF)]. Dry matter (DM) intake was higher (p<0.05) when dogs were full (11.4 g/kg) compared with restricted fed (5.98 g/kg). Ileal digesta viscosity tended to be higher (p=0.07) for full fed [10,251 centipoise (cP)] compared with restricted-fed dogs (6677 cP). Ileal digesta viscosities were 6561, 11,074, 13,830 and 15,967 cP for dogs consuming IDF, SDF, AP and PP treatments respectively. Ileal DM concentrations were 11.4%, 13.6%, 14.8% and 14.8% for SDF, IDF, AP and PP treatments respectively. DM digestibility coefficients were lowest (71.2%) for the PP treatment and highest (88.7%) for the IDF treatment. Organic matter digestibilities followed a similar pattern. Ileal digesta viscosity of dogs appears to increase with increased diet intake, and increased ileal DM concentration. More digestible diets result in less ileal DM and lower viscosity constants.  相似文献

Studies were carried out with six growing barrows fitted with a simple T-cannula 5 to 10 cm anterior to the ileo-cecal sphincter. In Exp. 1, the digestibility of biotin was determined in three cornstarch-based diets formulated to contain 16% CP by supplementation with soybean meal (SBM), meat and bone meal (MBM) and canola meal (CM). In Exp. 2 the digestibility of biotin was determined in three diets that contained 96.8% barley, corn or wheat. Experiments 1 and 2 were conducted according to a replicated 3 X 3 latin square design. In Exp. 3 pigs were fed a cornstarch-based diet supplemented with 12% vitamin-free casein to determine the amount of endogenous biotin. In Exp. 4 the digestibility of supplemental biotin was determined. There was a small amount of endogenous biotin in ileal digesta, 11 micrograms/kg DMI. Digestibilities of biotin determined at the distal ileum (apparent digestibilities corrected for endogenous biotin) were 55.4, 2.7 and 3.9% in SBM, MBM and CM, respectively, and 4.8, 4.0 and 21.6% in barley, corn and wheat, respectively. The digestibility of supplemental biotin was 93.5%. There was a large increase in the level of biotin between digesta collected from the distal ileum and in feces, ranging from 138 to 324 micrograms/kg DMI. With the exception of the CM diet, this increase exceeded dietary biotin intake. Biotin in many feedstuffs was not available in the small intestine.  相似文献

Use of dietary AA in growing pigs reflects digestion and use of digested AA for various body functions. Before evaluating dietary effects on use of digestible AA intake for body protein deposition, a digestibility study was conducted to investigate true ileal AA digestibility and endogenous ileal AA losses in growing pigs fed graded levels of wheat shorts (WS) or casein (CS; control). A casein-based basal diet (basal) was formulated to contain 0.27 g of standardized ileal digestible (SID) Lys per MJ of DE, to which extra Lys was added from WS (WS2, +0.10 g of SID Lys per MJ of DE; WS3, +0.20 g of SID Lys per MJ of DE) or casein (CS3, +0.20 g of SID Lys per MJ of DE). A fifth diet was formulated to be similar in CP level and source as CS3 but in which 6% pectin, a source of soluble non-starch polysaccharides (NSP), was included at the expense of cornstarch (CS3 + pectin). Five Yorkshire barrows (17.5 +/- 1.5 kg of BW) were fitted with a T-cannula at the distal ileum and randomly assigned to 1 of the 5 experimental diets in a 5 x 5 Latin Square design. Apparent ileal digestibility (AID), true ileal digestibility (TID), and endogenous ileal protein losses (EPL) were determined using the homoarginine method. Diet CS level did not influence (P > or = 0.10) TID of most essential AA or EPL (10.4 g/kg of DM intake). Including pectin in the diet did not influence TID of AA (P > or = 0.10) but increased EPL (15.6 g/kg of DM intake; P > or = 0.01). Inclusion of WS in the diet reduced TID of most essential AA (P < 0.01). The TID values for most essential AA, however, were the same (P > or = 0.10) for both dietary WS levels, except for Lys and Met, which were further reduced at the greatest dietary WS level. Increased EPL (P < 0.01) was only observed for WS3 (16 g/kg of DMI). We concluded that (1) the effects of dietary protein source on AID of AA can be attributed both to reduced TID of AA and increased EPL, (2) the impact of dietary WS level on TID of AA and EPL does not seem to be linear, (3) soluble NSP from pectin or WS exerts a greater effect on EPL than insoluble NSP, and (4) because of the metabolic cost associated with EPL and the impacts of feed composition on microbial fermentation in the gut lumen, the effects of feed ingredients on the use of ileal digestible AA for protein deposition should be investigated further.  相似文献

The homoarginine technique has been suggested as a means to determine true ileal amino acid digestibilities in nonruminant animals fed protein-containing diets. Conditions for guanidinating lysine to homoarginine in barley and canola meal and the effect of this process on nutrient composition and ileal digestibilities in the resulting material were investigated. Conditions tested were methylisourea concentration (0.4, 0.5, or 0.6 M) and reaction time (4 or 6 d) at pH 10.5. Using 0.4 methylisourea M solution for 4 or 6 d gave guanidination rates of 72.5 and 78.5% for barley and 72.3 and 75.2% for canola meal, respectively. Using 0.5 M gave 88.0 and 84.6% guanidination rates in barley and canola meal, respectively, after a 6-d reaction time. Under these conditions, guanidination did not change the nutrient composition of barley (P > 0.10), whereas it increased CP (38.4 vs 49.0%), crude fiber (10.2 vs 16.0%), acid detergent fiber (30.0 vs 43.4%) and neutral detergent fiber (29.8 vs 49.4%) levels in canola meal (P < 0.05). Four 33.6-kg barrows fitted with a simple T-cannula at the terminal ileum were fed a 16% CP unguanidinated barley and canola meal-based diet for four consecutive 14-d periods. Ileal digesta were collected continuously for 24 h on d 12 and 14 to determine apparent nutrient digestibilities. On the morning of d 14, pigs were fed a diet in which half of the barley and canola meal was replaced with guanidinated material for determining true ileal amino acid digestibilities. Digesta samples were pooled by pig and by 24-h period to give 16 observations per diet. Apparent ileal digestibilities of DM, CP, and AA in the unguanidinated and guanidinated barley-canola meal diet were similar (P > 0.10) despite the changes observed in canola meal. Apparent ileal lysine digestibility was 73.9 and 74.5% in the unguanidinated and guanidinated diet, respectively. The true ileal lysine digestibility was 88.1%. The present results show that guanidination does not interfere with digestion and further support the use of the homoarginine method for determining true ileal amino acid digestibilities in pigs fed practical diets. A methylisourea solution of 0.5 M and a 6-d reaction time are recommended for converting lysine to homoarginine in barley and canola meal.  相似文献
