The effect of the perorally ingested pesticide bentazone (195 mg.kg-1) of Czechoslovak origin on the amino acid composition of proteins of bacteria adhering to the ventral and dorsal ruminal wall investigated in six sheep. Proteosynthesis of adherent bacteria was studied by a modified and quantified elution method (elution of bacteria by an isotonic buffered solution at 4 degrees C). By the latter, a concentrate of undamaged bacteria a adhering to the sheep ruminal epithelium could be obtained. The yield of the method was estimated by scanning electrone microscopy and it was 93.3% (Legáth et al., 1990). The high correlation coefficient (r = 0.94, p < 0.01) suggested that in comparison with the control group of sheep, bentazone in the diet did not have a marked effect on the amount of amino acids in hydrolyzates of bacteria adhering to the dorsal and ventral ruminal epithelium. From the analysis of the single amino acids, it however follows that bentazone in the feeding ration caused significant changes in the concentrations of some amino acids in the proteins of bacteria adhering to the epithelium in the ventral and dorsal part of the rumen (p < 0.05) (Tab. 试验旨在研究北方典型草原放牧蒙古羊瘤胃细菌菌群结构和多样性随季节变化的规律。随机选取北方典型草原上终年放牧不补饲的6只蒙古羊,在冬季(1月)、春季(4月)、夏季(6月牧草生长期和8月牧草旺盛期)和秋季(10月中旬),通过口腔采取瘤胃液,利用MiSeq测序技术分析了不同季节、不同牧草生长时期瘤胃细菌种群的结构和多样性。结果表明,放牧蒙古羊瘤胃细菌在夏季牧草生长旺盛期操作分类单元(OTU)和多样性最高,冬季最少,瘤胃细菌夏季和冬季OTU和多样性都差异显著(P<0.05)。在门水平上,厚壁菌门(Firmicutes,56.93%)和拟杆菌门(Bacteroidetes,21.52%)为蒙古羊瘤胃中的优势细菌种群;厚壁菌门OTU在8月达到最大值,拟杆菌门OTU在6月达到最大值,均与其他月份差异显著(P<0.05)。在科水平上,瘤胃菌科(Ruminococcaceae,19.09%)、毛螺菌科(Lachnospiraceae,16.98%)、克里斯蒂娜科(Christensenellaceae,12.11%)、普雷沃氏菌科(Prevotellaceae,8.51%)和理研菌科(Rikenellaceae,8.27%)是蒙古羊瘤胃中的优势种群,分别在夏季(6月或8月)数量达到峰值。在属水平上,普雷沃氏菌属(Prevotella,4.79%)、丁酸弧菌属(Butyrivibrio,1.77%)和瘤胃球菌属(Ruminococcus,1.27%)含量最高。因此,北方牧区放牧蒙古羊瘤胃细菌以厚壁菌门的瘤胃菌科数量最高,是典型的消化纤维素细菌,符合以牧草为饲的放牧羊的特征。  相似文献

Effects of dietary factors including kernel thickness of processed barley grain, ratio of forage to concentrate, and forage particle length on chemical composition of bacteria, bacterial colonization of feed particles and distribution in the rumen, and duodenal flow of bacteria in dairy cows were evaluated. The experiment was designed as a double 4 x 4 quasi-Latin square with a 2 x 2 x 2 factorial arrangement of treatments using eight lactating cows with ruminal and duodenal cannulas. Barley grain was steam-rolled to two thicknesses: coarse (1.60 mm) or flat (1.36 mm); ratio of forage:concentrate was low (35:65) or high (55:45) (DM basis); and forage particle length was long (7.59 mm) or short (6.08 mm). Cows were offered ad libitum access to a total mixed diet. Chemical composition was different (P < 0.01) between liquid-associated bacteria (LAB) and solid-associated bacteria (SAB). Reduced barley thickness increased (P < 0.05) N content and 15N enrichment, but a high ratio of forage:concentrate decreased (P < 0.01) 15N enrichment of both the LAB and SAB. Significant differences between AA composition of the LAB and SAB were observed for 15 out of 17 AA studied. Bacterial colonization was linearly increased (P < 0.01) from about 5 to 70% as particle length of rumen contents was reduced from 3.35 mm to 0.15 mm (sieve size). The degree of colonization on each fraction of the rumen particulate matter was only affected (P < 0.10) by the ratio of forage:concentrate, with consistently higher (P < 0.10) bacterial colonization noted for high than for low forage:concentrate diets. Of the total bacterial mass within the rumen, less than 20% was associated with the liquid and over 70% was associated with the small particles that passed through the 0.6-mm sieve. Although the bacterial pool in the rumen was lower (P < 0.04) when flatly rolled barley rather than coarsely rolled barley was fed, bacterial flow to the duodenum was greater (P < 0.10) with increasing ratio offorage:concentrate. The present results confirm the differences of chemical composition and biomass for LAB and SAB. Manipulation of dietary factors such as ratio of forage:concentrate have the potential to alter bacterial colonization of rumen particles and the relative proportion of LAB to SAB, which were positively correlated to bacterial flow to the duodenum.  相似文献

The method of the use of the HD 1200-E automatic amino acid analyzer for the separation of amino acids was modified for the determination of 2-6-diaminopimelic acid (DAPA) as a bacterial marker, besides the other amino acids in the acid hydrolyzates of samples of bacteria isolated from the rumen of sheep. The reproducibility of the determination of DAPA in a standard amino acid mixture found in the tests corresponded with the reproducibility of the determination of the other amino acids as indicated by the manufacturer of the apparatus. The lower limit of DAPA determination sensitivity is between 2 and 5 nmol. In samples of bacteria isolated from rumen wall, from feed particles of rumen contents and from rumen fluid, different contents of nitrogen and DAPA were obtained; this is ascribed to the different proportions of bacterial species in the bacterial populations having different functions.  相似文献

本试验旨在以玉米为样本筛选用绵羊小肠液冻干粉测定精饲料瘤胃非降解残渣淀粉小肠消化率的最佳培养条件。分别研究了小肠液冻干粉用量(0.2、0.3、0.4、0.5和0.6g)对16h玉米瘤胃非降解残渣干物质和淀粉小肠消化率的影响;离体培养时间(4、8、12、16、20和24h)对16h玉米瘤胃非降解残渣干物质和淀粉小肠消化率的影响。结果表明,在本试验条件下,绵羊小肠液冻干粉测定玉米瘤胃非降解残渣淀粉小肠消化率的最佳用量为0.45g小肠液冻干粉/0.56g玉米瘤胃非降解残渣,最佳培养时间为12h。利用绵羊小肠液冻干粉测定常用饲料过瘤胃残渣淀粉小肠消化率的方法是可行的。  相似文献

A quantitative method of analysis for 2-aminoethylphosphonic acid (AEP) was developed using reverse-phase HPLC. The detection limit for AEP was 15 nM, and the detector response (peak area) was linear from AEP levels up to 100 microM (R = .99). Mean recovery of AEP added to strained ruminal fluid from faunated sheep was 98.2%. When AEP was added to a fermentation mixture at a concentration of 22.6 micrograms/ml, 78% disappeared during a 24-h incubation. 2-Aminoethylphosphonic acid was readily detected in preparations of mixed ruminal ciliate protozoa as well as in mixed and pure strains of ruminal bacteria, feedstuffs, and ruminal fluid and duodenal digesta from defaunated sheep. The occurrence of AEP in feed and bacterial hydrolysates was confirmed by organic phosphorus analyses. The concentration of AEP in mixed ruminal protozoa was three times greater than its concentration in mixed ruminal bacteria (4,304 vs 1,383 micrograms/g DM, respectively). The AEP values for pure ruminal bacterial cultures ranged from 733 micrograms/g DM in Bacteroides succinogenes B21a to 1,166 micrograms/g DM in Butyrivibrio fibrisolvens H17c. Ruminal fluid and duodenal digesta from defaunated sheep contained AEP concentrations of 30 micrograms/ml and 90 micrograms/g DM, respectively. The concentration of AEP in feedstuffs ranged from 25 micrograms/g DM in wheat straw to 263 micrograms/g DM in oats. Because AEP occurrence is not limited to ruminal ciliate protozoa, it is of little value as a marker for protozoal presence in or passage out of the rumen.  相似文献

The present in vitro experiment was conducted to study the effect of two concentrations of short chain fatty acids [SCFA: 0 (control), 40 and 100 mmol/l in the buffer solution on mucosal side] on calcium ion (Ca2+) transport across the isolated rumen epithelium of two groups of sheep. One group was subjected to hay ad libitum, whereas the other to concentrate feed (800 g per day in equal portion at 7.00 and 15.00 hours and hay ad lib). The conventional Ussing chamber method was used for measuring the Ca2+ transport rates (45Ca), short-circuit current (Isc) and tissue conductance (GT) of isolated rumen epithelium. The SCFA significantly increased Isc of the epithelia of concentrate-fed sheep. In both hay- and concentrate-fed animals, 45Ca flux rates showed an almost linear increase in net flux rate () with rising concentrations of SCFA, as a result of a combined effect of a large increase in mucosal-to-serosal flux rates () and an almost linear, but small, decrease in serosal-to-mucosal flux rate (). In concentrate-fed sheep and were significantly higher in tissues incubated with SCFA compared with hay-fed animals. The well-known adaptable morphological and functional changes in the rumen epithelium attributable to concentrate feeding obviously include Ca2+ transport; such feeding therefore may be considered as a possible prophylactic measure in the prevention of milk fever.  相似文献

A series of in sacco and in vitro studies were carried out to evaluate bean husks for activation of fibrolytic rumen bacteria and rice straw digestion. First, lablab bean husk, chickpea husk and rice straw were suspended in the rumen of sheep to analyze the bacterial consortium developed on each fiber source. Known members of fiber‐associating bacteria were found on both lablab bean husk and rice straw, but some of these bacteria were lacking on chickpea husk. Second, a pure culture study was carried out using six strains of Fibrobacter succinogenes. Both husks stimulated the growth of all tested strains, including a strain that did not grow on rice straw. The strain OS128 that showed the highest growth on rice straw displayed even higher growth on lablab bean husk without a time lag. Finally, two‐step incubations were carried out to determine whether prior incubation of rumen fluid with husks stimulates subsequent rice straw digestion. Higher digestibility of rice straw was recorded in the second‐round incubation following the first incubation with bean husks. These results suggest that the tested bean husks improve the digestion of rice straw by activating fibrolytic F. succinogenes and other associated bacteria.  相似文献

本试验旨在研究饲粮中不同比例的桑叶粉对育肥湖羊瘤胃上皮组织结构的影响。选取3月龄育肥湖羊40只,随机分为5组,每组8只。各组用桑叶粉分别替代饲粮中精料的0(A组)、15%(B组)、30%(C组)、45%(D组)、60%(E组)。预试期2周,正试期8周。结果表明:1)B组和C组瘤胃重/复胃重显著高于A组(P0.05)。2)各组瘤胃乳头宽度的差异不显著(P0.05)。3)对于瘤胃上皮的角质层宽度,A组高于其他各组,其中显著高于C组和D组(P0.05);对于瘤胃上皮颗粒层宽度,除D组显著高于E组(P0.05)外,其余各组间差异不显著(P0.05);对于棘突层和基底层宽度,与A组相比,桑叶粉处理能提高棘突层和基底层的宽度,但是作用不显著(P0.05),D组和B组的基底层宽度显著高于C组(P0.05)。总之,用桑叶粉替代15%~45%的精料能在一定程度上提高湖羊复胃的重量,促进的棘突层和基底层细胞的分裂,从而促进瘤胃上皮组织发育,同时,也有效降低了瘤胃上皮角质层的宽度。  相似文献

Details of the morphology of the forestomach of the Japanese serow (Capricornis crispus) were examined macroscopically, and by light and scanning electron microscopy. The rumen was sac-like in shape, and the right accessory longitudinal groove was not visible externally. The reticulum was relatively small, while the omasum was oval and large. Inside the rumen, even in the dorsal sac, the ruminal papillae were densely distributed, although absent on the pillars. The caudodorsal coronary pillar was indistinct. In the ruminoreticular fold, ruminal papillae were observed even on the reticulum side. In the reticulum, the primary reticular fold was apparent and the ill-defined secondary folds were also detected in some reticular cells. In the omasum, the omasal laminae of four sizes were recognized. The fourth order of laminae was occasionally absent. Histological sections of ruminal papillae revealed that softly keratinized epithelium covered the entire ruminal mucosa. In the reticulum, soft keratinized epithelium was observed throughout, with the exception of the tips of reticular papillae on which the epithelium showed evidence of hard keratinization. The epithelium of the omasal laminae was moderately keratinized. On the floor of the reticular groove, interlaminar area and in the omasal groove, the accumulations of numerous eosinophilic cells were observed in the superficial region. Under the scanning electron microscope, shallow transverse grooves were seen on the surface of the ruminal papillae. In general, the surface structure seemed to be similar to that of sheep rather than cattle. Cellular formations of the collagen fibrils were observed at the subepithelial region. In conclusion, examination of the forestomach allows classification of the Japanese serows as requiring an intermediate type of feeding habit and being between concentrate selectors and grass-roughage eaters from a morphological view.  相似文献

本文旨在研究大豆寡糖对绵羊瘤胃细菌总数、纤维分解菌数、甲烷产生菌数和瘤胃原虫数的影响.选用9只带有永久性瘤胃瘘管的试验羊,随机分为3组,饲喂相同的基础日粮,并在此基础上分别灌注占干物质采食量(DMI)1.2%、0.6%和0.0%的大豆寡糖.细菌和纤维菌的计数采用Hungate的滚管法;甲烷菌的计数采用最大可能计数法进行;原虫计数采用血球计数室计数法.结果表明,灌注大豆寡糖提高了瘤胃纤维分解菌的数量,降低了甲烷产生菌的数量.这表明大豆寡糖对绵羊瘤胃微生物区系有着较为显著的影响,有利于中性洗涤纤维(NDF)在瘤胃内的降解和减少饲料能量的损失.  相似文献

The effects of low and high nitrogen diets on amino acid levels were studied in hydrolyzates of ruminal bacteria adhered to four topographically different anatomic parts of the ruminal wall (dorsal, ventral and caudal parts as well as reticulum) in 18 sheep + of the Slovak Merino breed divided into three experimental groups. The epimural bacteria of the dorsal and ventral parts of the ovine rumen revealed the most sensitive reaction to the varying amounts of nitrogen ingested with the diet. In hydrolyzates of ruminal bacteria adhered to the dorsal and ventral epithelium, 15 and 14 amino acids were changing (Figs. 1, 2). In hydrolyzates of epimural bacteria, a sensitive reaction was observed in the following amino acids: alanine, histidine, thyroxin, arginine and proline (Tabs. I-IV). In all topographical and anatomical parts of the rumen, both alanine and histidine levels in hydrolyzates of epimural ruminal bacteria significantly increased with the diet with high-nitrogen content fed, but was falling in sheep fed with low-nitrogen diet. Changes in alanine concentrations may be explained by the fact that alanine forms a part of the mechanism for short-time storage of ammonia in bacterial cells (Bartos, 1987). The fact that alanine is in its lack deaminated to pyruvate (Havassy, 1976) is explained by significant fall in alanine contained in hydrolyzates of bacterial proteins when fed low nitrogen diets. Significant fall in alanine in shortage of amino acid bound nitrogen can be explained by the fact that under these conditions, the alanine skeleton is being incorporated in to 80% of amino acids synthetized de novo by ruminal bacteria (Syv?oja and Kreula, 1980). When sheep flock was fed the high-nitrogen diet, thyroxin and proline levels were significantly reduced in hydrolyzates of epimural bacteria from all parts of the rumen, while low-nitrogen diet significantly increased the concentrations of both given amino acids in comparison with the control. Bartos (1987) gives in his study the table containing weight representation of different amino acids in proteins of bacteria of the ruminal content compiled on the basis of data of several authors. These data principally correspond to the results of our measurements in hydrolyzates of epimural bacteria. The highest weight representation of amino acids in hydrolyzates of epimural bacteria was found for glutamic acid, aspartic acid and arginine, while the lowest ones were detected for thyroxin, proline and phenylalanine.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

选择1.2岁左右藏绵羊30只,随机分3组,采用单因子分组设计,以青干草为基础日粮,按每只150,300,450 g/d 3种精料水平补饲,研究不同精料补饲水平对藏绵羊瘤胃代谢参数变化规律的影响。结果表明;随精料补饲水平提高,藏绵羊瘤胃液pH值下降,NH3-N浓度、NH3-N浓度平均值、总挥发性脂肪酸(VFA)浓度显著增加(P<0.05)。在本试验条件下,从藏绵羊瘤胃内环境的稳定方面考虑,藏绵羊每日补饲精料300 g效果最佳。  相似文献

Sulfur influences on rumen microorganisms in vitro and in sheep and calves   总被引:2,自引:0,他引:2  
When continuously cultured ruminal microbes were given orchardgrass hay and sufficient sulfuric acid or hydrochloric acid to maintain a pH of 5.5, fermentation and numbers of protozoa were reduced compared with cultures whose pH was controlled with phosphoric acid. Likewise, when sulfur-deficient, purified diets were supplied to cultures, less methane (mmol X liter-1 X d-1), 3.2 vs 32.6, was produced and fewer cellulolytic bacteria (log10/ml), 5.8 vs 7.2 were present than when cultures were given the same diet supplemented with .3% elemental sulfur. The rumen of sheep fed the .04% sulfur diet had reduced digesta weights (1.69 vs 3.2 kg) compared with sheep fed the diet with .34% sulfur at the same intake. There also was reduced methanogenesis 12.3 vs 25.8 mmol X liter-1 X d-1) and reduced numbers of cellulolytic bacteria (7.4 vs 8.4 log10/ml) in sulfur-deficient sheep in comparison to sulfur-supplemented sheep. In growing calves, the same types of bacteria predominated in the rumen, but more facultative anaerobic bacteria were isolated from calves fed .04% sulfur than from calves fed diets with .34 to 1.72% sulfur. None of the dietary levels of sulfur appeared toxic. Regardless of treatment, volatile fatty acids were more predominant than lactic acid as end-products of fermentation of ruminal microbes in fermenters, sheep and calves. The greater methanogenesis and the greater cellulolytic bacterial numbers of sulfur-supplemented sheep compared with sulfur-deficient in vitro cultures, is interpreted to be the result of recycling of sulfur to the rumen in sheep where it is efficiently scavengered by ruminal bacteria.  相似文献

This study was undertaken to investigate the relationship between chemical composition and microbial profile of rumen liquid‐associated bacteria (LAB) in vivo (Murciano‐Granadina goats) and in a rumen simulation system (single‐flow continuous‐culture fermenters). To achieve this aim, analyses of purine bases along with some molecular techniques (quantitative PCR to assess abundance and DGGE to identify biodiversity and bacterial profile) were carried out. A control diet (AHC) based on alfalfa hay (AH) and concentrate (C) in a 1:1 ratio and two experimental diets (AHCBI and AHCBII), in which concentrate was partially replaced with multinutrient blocks, were used. Diets AHCBI and AHCBII included multinutrient blocks differing in the relative amount of two‐stage olive cake and the source of protein (sunflower meal vs. fava beans). We aimed to investigate the effect of these blocks on rumen microbiota to evaluate their potential as safe substitutes of cereal‐based concentrates. Similar patterns of response to diet were found for chemical composition, microbial abundances and diversity in LAB isolated from goat's rumen and fermenters. Whereas bacterial density (log10 gene copies/g FM: 11.6 and 9.4 for bacteria and methanogens, respectively, in rumen) and diversity indexes (Shannon index: 3.6) were not affected by diet, DGGE analyses showed that bacterial community profile was affected. The cluster analysis suggested differences in bacterial profile between LAB pellets isolated from the rumen of goat and fermenters. A relationship between chemical composition and bacterial community composition in LAB pellets seems to exist. Changes in the former were reflected in the bacterial community profile. Further research is needed to clarify the relationship between chemical and microbial composition of ruminal bacterial pellets with diets of different quality.  相似文献

Na+/H+ exchanger (NHE), which catalyses the exchange of extracellular Na+ for intracellular H+, is of importance in the maintenance of Na+ and pH homoeostasis for rumen epithelial cells. Studies in ruminants showed that high concentrate diets could increase the expression of NHE in ruminal epithelium. Results of recent studies further indicated that insulin, as an important hormone closely related to dietary concentrate, could enhance the expression of NHE. In this study, we have investigated the mechanisms of insulin regulating the expression of NHE in rumen epithelial cells and its potential role in dietary modulation of NHE expression in ruminal epithelium of cows. In primary culture, insulin increased phosphorylation of ERK 1/2 and AKT in rumen epithelial cells. However, this promotion was diminished by insulin receptor inhibitor. Insulin also stimulated NHE1 and NHE3 expression. But this increase was suppressed by insulin receptor inhibitor, ERK inhibitor and AKT inhibitor. In the present animal experiment, NHE1 and NHE3 expression increased in rumen epithelium of cows ingesting a high concentrate diet (HC, 60% concentrate), accompanied by increased insulin concentration in plasma, compared to those feeding a low concentrate diet (LC, 20% concentrate). Furthermore, the phosphorylation of ERK1/2 and AKT was higher in the rumen epithelium of the HC group than those in the LC group. Collectively, these results indicate that diet‐dependent change of NHE1 and NHE3 abundance was mediated, at least in part, by plasma insulin through the ERK and AKT pathway.  相似文献

本试验旨在筛选用绵羊小肠液冻干粉评定瘤胃非降解精饲料残渣的有机物于小肠消化率的最佳培养条件。试验包括三部分 :第一个试验研究了小肠液冻干粉用量 ( 0 .2、0 .3、0 .4与 0 .5g)与缓冲液种类(McDougall缓冲液与磷酸盐缓冲液 )对 1 2h瘤胃非降解玉米与豆粕的有机物于小肠消化率的影响 ;第二个试验研究了缓冲液pH值 ( 7.0、7.5与 8.0 )与Ca2 + 浓度 ( 3 4 0、680与 2 0 0 0 μmol/L)对 1 2h瘤胃非降解玉米与豆粕的有机物于小肠消化率的影响 ;第三个试验研究了离体培养时间 ( 3、6、9、1 2、1 5、1 8、2 1与 2 4h)对1 2h瘤胃非降解玉米与豆粕的有机物于小肠消化率的影响。筛选的用绵羊小肠液冻干粉评定瘤胃非降解精饲料残渣的有机物于小肠消化率的适宜条件为 :0 .4 5g小肠液冻干粉 / 0 .56g饲料 ,McDougall缓冲液 ,pH =7.5,Ca2 + =680 μmol/L ,3 9℃ ,瘤胃非降解玉米与豆粕的最佳培养时间不同 ,分别为 1 2与 1 8h  相似文献
