In Europe, sugar beet is often produced in a 3‐year rotation with cereals, leaving stubble fields fallow from cereal harvest until primary tillage in autumn in the year prior to sugar beet production. The weed flora on such fields could include host plants of Heterodera schachtii that is one of the most important pests of sugar beet. Crop sequences with non‐hosts and cover cropping with resistant cruciferous hosts during this period have been crucial for its management. Availability of resistant and tolerant sugar beet cultivars could entice growers to forego cover cropping, exacerbating weed problems during the fallow period. The objective of this study was to determine the reproductive potential of H. schachtii on weeds that develop during this period. Under glasshouse conditions, reproduction on 39 plant species was compared with that on oilseed radish and sugar beet of differing nematode host status. In 2 years in field microplots, 18 previously tested species were grown in H. schachtii‐infested soil during the typical fallow period at 60 plants m?2, and nine of these species were also grown at 180 plants m?2. There were variable results between years after 8 weeks of growth, but most weeds allowed lower reproduction (<10%) than the susceptible sugar beet; only Stellaria media at 180 plants m?2 and Thlaspi arvense at both plant densities increased nematodes. Such weed densities may seldom occur under commercial conditions; thus, weed management for nematological considerations during the stubble period may have limited importance. 相似文献

The activity of arginase (ARGAH), which results in ornithine and urea production, is important for nitrogen metabolism in all organisms as well as for defence responses. The second‐stage juveniles of the cyst‐forming nematode Heterodera schachtii penetrate roots and induce the formation of a permanent feeding site. To determine whether infection with H. schachtii causes the induction of arginase, the expression was studied in Arabidopsis roots and shoots at the day of inoculation and at 3, 7 and 15 days post‐inoculation (dpi). Parasite infection caused a strong decrease of root arginase activity and ARGAH1 gene expression, which persisted over the entire examination period. In shoots, the mRNA expression of ARGAH2 increased at 3 and 7 dpi, but the enzymatic activity was significantly enhanced only at 3 dpi. Thus, while arginase down‐regulation occurs in roots, which is apparently due to the presence of nematode effectors, in shoots the activity is only transiently up‐regulated despite persistently high gene expression. As oxidative stress is possible during nematode infection, the activity and gene expression of glutathione reductase, a marker of the redox equilibrium, were estimated and found to be significantly enhanced at 7 dpi in shoots of infected plants. The level of proline, an amino acid known for its ability to scavenge free radicals, was increased 60‐fold. The results suggest that the disruption of redox homeostasis, as reflected by increased proline level and glutathione reductase expression and activity, accompanies changes in arginine metabolism in the shoots, indicating systemic changes induced by nematode infection. 相似文献

In Germany, sugar beet is often rotated with 2 years of cereal. Extensive fallow periods between cereal harvest and autumn primary tillage allow for a weed flora to develop. Broad‐leaved weeds could potentially be alternate hosts for the common nematode Heterodera schachtii, one of the most important pests of sugar beet. Between 2009 and 2012, annual weeds developing in cereal stubble fields during July to mid‐October in the season prior to sugar beet were surveyed, including known hosts of H. schachtii. Yearly weather patterns and agronomic practices possibly impacted weed species composition and weed population densities. During September, Chenopodium album, Cirsium arvense, Convolvulus arvensis, Mercurialis annua, Polygonum spp., Solanum nigrum and Sonchus spp. occurred at the highest frequencies. Weed hosts of H. schachtii were present, but densities, frequencies and uniformity were limited. In 2010 and 2011, staining for nematodes in roots revealed juvenile penetration of some weeds but few adult stages. No indication of nematode reproduction of H. schachtii was found on these weed hosts. A fairly stable weed flora was detected on stubble fields that could provide some carry over for weed species. An elevated risk for nematode population density build‐up on these weeds was not found and management of these weeds at the observed densities during the stubble period for nematological reasons appeared unnecessary. 相似文献

甜菜孢囊线虫(Heterodera schachtii Schmidt)是全世界重要检疫性有害生物,对甜菜具有毁灭性危害,该线虫已在全世界50多个国家或地区有分布,22个国家将其列为检疫对象。甜菜孢囊线虫寄主多达23科95属218种植物,可导致甜菜产量损失达25%~70%,甚至绝产,在欧洲每年造成的经济损失已超过9 000万欧元,严重威胁当地甜菜生产和制糖业。甜菜孢囊线虫是我国重要进境检疫性有害生物,因其对甜菜具有毁灭性危害,我国各级农业行政主管部门对甜菜孢囊线虫都高度重视,严防该线虫的暴发和危害,本文介绍国外甜菜孢囊线虫研究进展。 相似文献

小麦禾谷胞囊线虫(Heterodera avenae)的核糖体基因(rDNA)限制性片段长度多态性研究 总被引:10,自引:4,他引:10
采用PCR技术扩增出中国和摩洛哥禾谷胞囊线虫群体的核糖体基因(rDNA)的内转录间隔区(ITS)片段的长度约为1 060 bp。用11种限制性内切酶(RE)酶切禾谷胞囊线虫ITS扩增产物,共产生27个酶切片段。用AluI和RsaI酶切ITS扩增产物证明中国禾谷胞囊线虫ITS属于"B型",而摩洛哥禾谷胞囊线虫ITS属于"A型"。用HinfI酶切后,7个中国禾谷胞囊线虫群体产生2个RFLP片段(860和200 bp),而摩洛哥群体产生3个RFLP片段(520、340和200 bp),HinfI揭示出中国与摩洛哥禾谷胞囊线虫ITS之间存在差异。AvaI和HindⅢ不能酶切禾谷胞囊线虫ITS。用CfoI、Bsh1236I、MsrFI、ScrFI、HaeⅢ和MvaI 6种RE酶切中国和摩洛哥禾谷胞囊线虫群体的rDNA-ITS,均分别得到相同类型的RFLP分布型,因此这6种RE不能揭示中国与摩洛哥群体的rDNA-ITS的差异。 相似文献

为了解甘肃省陇东南地区大豆孢囊线虫病分布和发生程度,采用Z字形取样法,对甘肃省陇东南地区9个县(区)37个乡镇大豆孢囊线虫病的发生和分布进行调查,并根据形态特征和rDNA-ITS序列比对鉴定线虫种类。共采集土样411份,陇东地区大豆孢囊线虫检出率为87.2%,其中镇原、西峰、正宁、华池和环县的检出率最高,达100%,镇原孢囊发生量最大,平均孢囊数为11.4个/100 g土;陇南地区大豆孢囊线虫检出率为63.7%,成县和徽县分别为64.0%、63.0%,平均孢囊数分别为0.9和1.2个/100 g土。经形态学特征观测和rDNA-ITS序列比对,甘肃省陇东南采集到的孢囊线虫均鉴定为大豆孢囊线虫Heterodera glycines。 相似文献

为了明确河北省小麦主产区禾谷孢囊线虫的致病类型,采用27个国际鉴别寄主品种和2个感病小麦品种‘石新733’和‘温麦4’对河北省邯郸、保定、任丘和唐山的4个禾谷孢囊线虫群体致病型进行了鉴定。依据Andersen等鉴别标准,4个群体的致病型不同于国际上已命名的16个禾谷孢囊线虫致病型。唐山群体为一个致病型,它的致病型和山西太谷、安徽固镇群体的致病型相似;邯郸群体、保定群体和任丘群体的致病型属于同一致病型,这3个群体的致病型和定州群体的致病型很接近。对照小麦品种‘石新733’和‘温麦4’对4个禾谷孢囊线虫群体均表现感病。 相似文献

The induction and differentiation of feeding structures (syncytia) of the cyst nematode Heterodera schachtii in roots of Arabidopsis thaliana is accompanied by drastic cellular modifications. We investigated the formation of cell wall openings which occurred during syncytium differentiation. At the beginning of syncytium induction, a callose-like layer was deposited inside of the wall of the initial syncytial cell (ISC). First wall dissolutions developed by gradual widening of plasmodesmata between the ISC and neighbouring cells. As a general thickening of syncytial cell walls blocked existing plasmodesmata, other large openings were formed by enzymatic dissolution of intact walls by putative cellulase activity. 相似文献

Plant parasitic nematodes of the genus Heterodera show a high degree of sexual dimorphism, which is reflected by different nutritional demands and differences in the structure of the induced specific syncytial feeding site in the plant. The determination of the sex of the nematode Heterodera schachtii and other related species was repeatedly reported to be dependent on trophic factors, which are provided by the induced syncytia. The structural differences of syncytia induced by H. schachtii in roots of Arabidopsis thaliana were analysed at the anatomical and ultrastructural level. Syncytia of males were induced in the root pericycle. The developing syncytium then expanded into procambial or cambial cells of the vascular cylinder. Differentiated vascular elements were not included. The expansion of the syncytium triggered the proliferation of cambial and peridermal tissues, in a manner similar to secondary growth, and the formation of additional xylem and phloem elements. In comparison to syncytia associated with females, syncytia associated with males were less hypertrophied and were composed of more cells. Distinct cell wall openings were mostly found between the few strongly hypertrophied syncytial elements at the actual feeding site in the pericycle. The ultrastructure was very similar to female-associated syncytia but showed conspicuous differences in the structure and localization of cell wall ingrowths. These ingrowths were rare and weakly developed and occurred not only at the interface with xylem elements but also at the internal and external walls of the syncytia. After feeding had ceased at the end of the third developmental stage the syncytia degenerated. 相似文献

本文应用MAXENT与GARP两种生态位模型对甜菜孢囊线虫在中国的适生性进行了风险分析和预测。结果表明甜菜孢囊线虫可在中国17个省市生存,适生范围为26°N~48°N,77.6°E~136°E。甜菜孢囊线虫入侵的高风险区:内蒙古南部、新疆西部、河北中南部、山西东北部、宁夏、甘肃北部;中风险区:北京、天津、陕西、山西大部、内蒙古西部和东南部、吉林西部、新疆北部;低风险区:河南、陕西南部、山东、内蒙古中东部、新疆中部、辽宁、吉林大部、黑龙江南部、江苏、安徽、湖北、湖南、江西、浙江北部;而青海、新疆南部、西藏、四川、重庆、云南、贵州、广西、广东、福建、海南、内蒙古少数地区和黑龙江部分地区属于基本不发生区。我国内蒙古、新疆、辽宁等省市甜菜种植区均适合甜菜孢囊线虫发生,对上述地区的进出口岸应加强甜菜孢囊线虫的检疫工作。 相似文献

Genetic relationships and isozyme variability in the Heterodera avenae complex determined by isoelectrofocusing 总被引:1,自引:0,他引:1
Isozyme variability was assessed among the principal species of the cereal cyst nematode complex to complete and enhance the information provided by classical nematode systematics, in order to clarify inter- and intraspecific relationships within this complex. Twenty populations of cereal cyst nematodes ( Heterodera avenae , H. filipjevi , H. latipons and H. mani ) were compared by means of five different isoenzymatic systems (esterase, malate dehydrogenase, phosphoglucoisomerase, phosphoglucomutase and superoxide dismutase) using isoelectrofocusing (IEF) on the electrophoretic separation. The results are in agreement with previous morphological and biochemical characterizations, which established genetic diversity between the Gotland strain and H. avenae and identified the Gotland strain with H. filipjevi . Populations from Israel, all included in the H. avenae group, exhibited well-defined intraspecific dissimilarity. The highest degree of polymorphism was found in the H. avenae group for all five enzymatic systems studied. The H. mani population was also included in the H. avenae group by these isozyme analyses. Malate dehydrogenase, phosphoglucoisomerase and phosphoglucomutase isozymes, fractionated for the first time by IEF in the cereal cyst nematode complex, displayed a higher level of polymorphism than using conventional electrophoresis. Isoelectric focusing has proved to be a useful tool for detecting genetic diversity within and among species of the cereal cyst nematode complex and for taxonomic purposes. 相似文献

The development ofHeterodera schachtii Schm. (beet cyst nematode, BCN) juveniles in roots of resistant and susceptible genotypes belonging to cruciferous crop species and hybrids was studied from 4 to 28 days after inoculation. No difference in root penetration by larvae was observed between resistant and susceptible plants.The development of nematodes in roots from resistant plants ofRaphanus sativus L., resistant xBrassicoraphanus Sageret and a resistant hybrid xBrassicoraphanus x Brassica napus L. was similar. BCN resistance in these three sources of plant material appeared to be related to an increased male:female nematode ratio as compared to the ratio found in susceptibleR. sativus plants.Also in resistant plants ofSinapis alba L. and a resistant intergeneric hybridS. alba x B. napus the increase in male:female nematode ratio, as compared to the ratio found for susceptibleS. alba cultivars and a susceptible intergeneric hybridS. alba x B. napus, seemed to be related with the observed resistance. In roots of the resistantS. alba and of a resistant hybridS. alba x B. napus, however, BCN resistance might also be due to a slower development of larvae and increased necrosis of root cells at the site of larval penetration. 相似文献

为了解安徽皖北麦区及颍上县禾谷类孢囊线虫(cereal cyst nematode,CCN)发生和危害情况,用"zig-zag"法在皖北不同麦区采集小麦根际土样60份,利用Fenwick&Oostenbrink法分离CCN孢囊,统计卵密度发生频度。依据孢囊阴门锥等形态特征,鉴定颍上县CCN种类。利用扩散系数法判断CCN田间平面分布型,利用系统观察法,动态监测小麦生长季节0~300mm土层的2龄幼虫(J2)虫口变化。通过酸性品红染色方法监测早期侵染;选择CCN密度中等田块,以涕灭威作播前土壤处理,比较‘泛麦5’等品种的药剂处理间的株高和产量差异。结果表明,在皖北麦区,62%的田块CCN卵发生密度为1~10个/g土;经鉴定发生在颍上县的CCN为禾谷孢囊线虫(Heterodera avenae),颍上黄桥镇双楼村禾谷孢囊线虫在田间为块状分布;在颍上监测到J2对小麦侵染的最早时间在3月18日,当感染田块卵密度在2.5~20.5个/g土时,产量损失为9.8%~14.9%,其中‘泛麦5’产量损失显著,达14.9%。 相似文献

ABSTRACT Soil suppressiveness to Heterodera schachtii was demonstrated in a field at the research station of the University of California, Riverside. In two field trials planted to Swiss chard (Beta vulgaris), introduced H. schachtii multiplied 2.7 and 1.7 times more in preplant metam sodium-fumigated plots than in nontreated plots in 1994 and 1995, respectively. In greenhouse experiments, preplant treatments with metam sodium, methyl bromide, methyl iodide, formaldehyde, and aerated steam reduced suppressiveness of soil against H. schachtii to undetectable levels. H. schachtii multiplied significantly less in nontreated soil than in treated soil on Swiss chard. At harvest, the number of infective second-stage juveniles in suppressive soil was close to the lowest detection level, whereas high numbers were encountered in soils initially treated. In a crop rotation trial with host crops of H. schachtii, introduced H. schachtii populations were monitored for five cropping periods over 30 months in initially fumigated versus nontreated suppressive field plots. In fumigated plots, H. schachtii population levels increased in the first and second cropping periods and then declined in the third cropping period. In the fourth and fifth cropping periods, the nematode reproduction factor in the initially fumigated plots was not significantly different from that in suppressive plots. 相似文献

The gene CreAet for resistance to Heterodera avenae, transferred from Aegilops triuncialis to Triticum aestivum introgression line TR-353, was assessed for responses to Pratylenchus thornei under field conditions. After 2.5 months, P. thornei infestation on TR-353 was similar to those on its progenitors (T. aestivum H10-15, T. turgidum H1-1 and A. triuncialis A-1) and on the cultivar Capa. After 5 months, TR-353 hosted significantly more P. thornei per plant than A. triuncialis, T. aestivum H10-15 or cultivar Rinconada, being very close to the maximum value obtained from T. turgidum. The final infestation of line TR-353 by H. avenae was significantly lower than on the other plants, except for A. triuncialis. In addition, at 2.5 months, the abundance of P. thornei on TR-353 was intermediate between the introgression wheat line H93-8 (Cre2 gene) and the cultivar Loros (Cre1 gene), but the final number of P. thornei per plant on TR-353 was significantly greater than on Loros. Line TR-353 was not able to control P. thornei in the field, but it confirmed its resistance to H. avenae, which was significantly lower than those due to Cre1 and Cre2 genes. No competition was observed between P. thornei and H. avenae populations on line TR-353 or A. triuncialis. 相似文献

禾谷孢囊线虫(Heterodera avenae)是中国小麦上的重要病原线虫。纤维素结合蛋白基因是一种重要的植物线虫寄生和致病相关基因。用同源克隆的方法从禾谷孢囊线虫寄生前二龄幼虫中克隆出一种纤维素结合蛋白新基因Ha-cbp-1(GenBank 注册号GQ178086)cDNA序列。Ha-cbp-1基因cDNA包含1个开放阅读框,编码131个氨基酸残基的蛋白质,预测蛋白由1个长度为18氨基酸残基的信号肽和1个纤维素结合区域(CBD)组成。Ha-cbp-1的基因组DNA序列含有2个内含子。预测的禾谷孢囊线虫纤维素结合蛋白(HA-CBP-1)序列与大豆孢囊线虫纤维素结合蛋白(HG-CBP-1)序列有60%的同一性和76%的相似性,与甜菜孢囊线虫纤维素结合蛋白(HS-CBP-1)序列有60%的同一性和75%的相似性。本研究首次从禾谷孢囊线虫中成功克隆出CBP蛋白基因。 相似文献
