From black spots on winter mushroom (Flammulina velutipes), fluorescent bacteria were repeatedly isolated during surveys at places of production in the years 2009–2010 in Korea. From these lesions three bacterial strains (designated CHM13, CHM16, CHM17) were isolated which, following inoculation of mushroom stipes and caps, yielded characteristic black spots and sunken lesions, which developed into a severe black rot. Results of Gram stain and biochemical tests preliminarily identified these isolates as Pseudomonas tolaasii. This was confirmed by pathogenicity to winter mushroom, physiological and biochemical properties, analysis of the 16S rRNA and rpoB gene sequences, fatty acids profile, specific and sensitive PCR assays and, lipopeptide detection. This is the first report of the isolation of Pseudomonas tolaasii from cultivated winter mushroom in Korea.  相似文献

The Vat resistance gene (in Cucumis melo L.) inhibits the transmission of non-persistent viruses by Aphis gossypii Glover, but does not affect transmission by Myzus persicae (Sulz.). To see whether this difference was behaviourally determined, we investigated the stylet penetration behaviour of these two aphid species by recording EPGs (Electrical Penetration Graphs) of 8 and 20 min on two sets of susceptible and resistant isogenic melon lines. During the 20 min EPG study, inoculation with CMV (Cucumber Mosaic Virus) was also investigated. For both sets of isogenic lines, the two aphid species were able to detect the presence of Vat. The mean duration of individual intracellular punctures on the resistant genotypes was significantly reduced for both M. persicae and A. gossypii (-10% and -8% respectively for duration of pattern ‘pd’ in the 20 min experiment); this reaction appeared faster for M. persicae, a species for which melon was not a suitable host-plant. Therefore, in contrast to Vat's anti-transmission effect, this behavioural effect was not aphid species-specific. Also, the frequency of intracellular punctures on the resistant genotypes was significantly reduced in A. gossypii (but not in M. persicae): on average, this frequency dropped from ≈0.65 pd.min-1 on the susceptible genotypes to ≈0.5 pd.min-1 on the resistant ones. It was concluded that (intracellular?) chemical cues were detected very early by aphids probing on the resistant genotypes carrying the Vat gene. However, a comprehensive analysis of the behavioural traits of both aphids on the two genotypes could not alone account for the complete inhibition of transmission which was found only to occur for A. gossypii on resistant genotypes. None of the differences detected (between aphid species or between plant genotypes) could account for the Vat phenotype, although they may explain quantitative differences in transmission efficiencies between aphid species. It was thus concluded that Vat effect was primarily chemically mediated. Finally, some intracellular punctures bearing typical subphases have been identified in both aphid species and were designated as ‘long potential drops’ (pd-L). For A. gossypii, these were observed early after plant contact and their mean duration was twice that of standard intracellular punctures (≈8.5 s vs ≈4.2 s). Although not necessary for CMV inoculation, the duration of such phases was positively correlated with a high transmission efficiency by A. gossypii on the susceptible genotype. The nature of this pattern and a putative mechanism of action of the Vat gene are discussed.  相似文献

A detailed observation of the sensory apparatus of Opisthioglyphe ranae (Fr?lich, 1791) cercariae was carried out. The species was determined on the basis of an experimental study of the life cycle and morphology of sporocysts, cercariae, metacercariae, and adults. In contrast to other species of the family Plagiorchidae, no papillae were present in the positions CId1, CId2, CII0, CII3 and CII4. Papillae with markedly ventral situation on the postacetabular zone were found very rarely. Some of the cercariae possessed ventral papillae variously distributed in the preacetabular zone. A comparative analysis of the chaetotaxy revealed that the cercaria studied by us was very similar to Cercaria 4 of Richard, 1971, with the exception of the number of acetabular papillae and some groups of ventral papillae. Compared to the cercaria of O. ranae studied by Dobrovolskiy (1965). AID papillae and a part of ventral papillae were distributed differently. The chaetotaxy was identical with that of experimentally obtained cercariae of O. ranae from L. stagnalis and of cercariae of the same species obtained from Bulgaria.  相似文献

Two Hungarian virus isolates from sweet pepper (K8) and melon (S4) were identified as cucumber mosaic virus (CMV) on the basis of host plant reactions and serology. The isolates were purified and antisera prepared. Homologous antiserum titers in double-diffusion tests were 256 (K8) and 512 (S4). They were serologically closely related to each other and to other CMV isolates. On the basis of symptoms they belong to different symptomatological groups of CMV; this was supported by serological properties. Sedimentation coefficients were c. 93 S, at 2 mg ml–1. Purified preparations, stained with 2% uranyl acetate, showed spherical particles. In ELISA purified preparations reacted with each other's antisera.Samenvatting Twee hongaarse virusisolaten uit paprika (K8) en meloen (S4) werden geïdentificeerd als komkommermozaïekvirus (CMV) met behulp van toetsplanten en serologie. Beide isolaten werden gezuiverd en er werden antisera tegen bereid. De homologe titers van de antisera in de agar-geldiffusietoets bedroegen 256 (K8) en 512 (S4). K8 en S4 waren serologisch nauw verwant aan elkaar, evenals aan andere CMV-isolaten. Op grond van hun symptomen op toetsplanten behoren ze tot verschillende symptomatologische groepen van CMV. Dit laatste werd gesteund door de serologische eigenschappen. Beide isolaten hebben een sedimentatiecoëfficiënt van ca 93 S, bij een concentratie van 2 mg ml–1. Gezuiverde preparaten, gekleurd met 2% uranylacetaat, bleken bolvormige deeltjes te bevatten. In ELISA reageerden gezuiverde preparaten van K8 en S4 met elkaars antisera.  相似文献

A field colony of the Two-spotted spider mite, Tetranychus urticae (Koch), resistant to fenpyroximate was further selected with fenpyroximate 5SC for 20 generations at a selection pressure of 30-50% mortality (designated as FR-20 strain). Resistance and cross-resistance levels of the FR-20 strain to 18 acaricides were determined using a spray method. The FR-20 strain was extremely resistant to fenpyroximate [resistance ratio (RR) 252]. The strain exhibited extremely strong positive cross-resistance to acrinathrin (RR 196), and high levels of resistance to benzoximate (RR 55) and propargite (RR 64). Moderate levels of cross-resistance (RR 11-40) to abamectin, fenbutatin oxide, fenpropathrin, pyridaben, pyridaben + bifenthrin and tebufenpyrad were observed. The FR-20 strain showed low levels of resistance (RR < 10) to azocyclotin, bromopropylate, chlorfenapyr, chlorfenapyr + bifenthrin, chlorfenapyr + pyridaben, dicofol, fenazaquin and milbemectin. Synergist experiments with different metabolic inhibitors revealed that piperonyl butoxide had the greatest effect on the efficacy of fenpyroximate, followed by iprobenfos and triphenyl phosphate. In a comparative assay with detoxifying enzymes, the FR-20 strain showed 2.5-fold higher activity in p-nitroanisole-O-demethylation, and 2.5- and 2.2-fold higher activities in alpha- and beta-naphthyl acetate hydrolysis, respectively. These results suggested that enhanced activities of both mixed-function oxidases and esterases likely contribute to the fenpyroximate resistance of the FR-20 strain of T urticae.  相似文献
