Full-thickness skin wounds were created on the dorsum of both metacarpi in 8 horses. Three topical treatment regimens were studied. All wounds were bandaged with a nonadherent dressing, which was held in place with a snug elastic wrap. Group-A wounds were treated with a proprietary topical wound medication that consisted of a spray and an ointment. Group-B wounds were treated with the same regimen, except the putative active ingredients in the ointment were omitted. Group-C wounds were treated with a dry nonadherent bandage only. Wound dressings were changed every day and the limbs were photographed every other day until the wounds were healed. Specimens of normal skin and biopsy specimens of healed wounds were examined histologically and were assayed for hydroxyproline content. Wound healing measurements quantitated for each wound were number of days to healing, maximal wound size attained, day wound contraction commenced, day epithelium first noticed, rate of wound contraction, final wound size, and fraction of the wound that healed by contraction. The cosmetic appearance of the healed wounds was also graded. Significant differences were not noticed in hydroxyproline content, histologic appearance, or any of the wound healing measurements between treatment groups. The cosmetic appearance of healed group-A and -B wounds was significantly better than the appearance of group-C wounds. The topical treatment regimens studied neither enhanced nor inhibited wound healing in this study.  相似文献

为研究不同浓度的芒硝对体外培养的驴皮成纤维细胞增殖、凋亡的影响,本试验采用组织块法体外培养驴皮成纤维细胞,并用HE、Masson染色鉴定;再添加高(2 000、1 500 μg/mL)、中(1 000、500、250 μg/mL)和低浓度(100、50、10 μg/mL)芒硝体外培养驴皮成纤维细胞,用MTT和Caspase-3法分别检测成纤维细胞增殖率和凋亡率;最后用实时荧光定量PCR、ELISA法检测不同浓度(1 500、250、10 μg/mL)芒硝培养液中影响驴皮成纤维细胞胶原蛋白合成的相关基因表达量及蛋白浓度。结果显示,培养5 d时,驴皮成纤维细胞开始从组织块边缘爬出,25 d后长满培养皿;HE染色呈典型的梭形,驴皮成纤维细胞中胶原纤维经Masson染色呈蓝色。中浓度芒硝培养液组成纤维细胞的增殖率显著高于高、低浓度组(P<0.05),相应地,中浓度组的凋亡因子Caspase-3活性显著低于其他两组(P<0.05)。相比于24 h,芒硝作用于成纤维细胞48 h能显著提高胶原蛋白合成相关基因的表达量(P<0.05),250 μg/mL浓度的效果最佳。综合上述结果,组织块培养法易于分离培养驴皮成纤维细胞,250 μg/mL浓度的芒硝在48 h能显著提高驴皮成纤维细胞的增殖率及其胶原蛋白合成相关基因的表达量,此结果可为芒硝促进驴皮成纤维细胞的增殖作用以及驴皮伤口愈合机制的研究提供理论依据。  相似文献

The effects of Zincojecol an ointment containing zinc oxide and cod liver oil on wound healing were compared with ointments that either contained no active ingredients or zinc oxide or cod liver oil alone. All formulations enhanced the epidermal proliferation after mechanical irritation of the tail skin. The combination of zinc oxide and cod liver oil was found to be superior to the vehicle control and formulations containing only one active ingredient. This combination was also found to be most efficient in accelerating wound healing being retarded by repeated dexamethasone treatment.  相似文献

OBJECTIVE: To determine the macroscopic effects of topical application of taurolidine on second intention healing of experimentally induced wounds in rats. ANIMALS: 32 adult Sprague-Dawley female rats. PROCEDURES: In each rat, 2 skin wounds were created in the lumbar area. Groups of 7 rats were assigned to have 1 wound treated topically with hydroxycellulose gel (HDCG), 2% taurolidine in HDCG (T-HDCG), 2% taurolidine-sodium citrate solution, or bacitracin-neomycin-polymyxin B ointment; the other wound was not treated. Four control rats (8 untreated wounds) were used. Wounds were monitored for contraction, epithelialization, and complete healing at 4, 8, and 14 days after wound creation. The number of days to complete healing was also recorded for each wound. RESULTS: Compared with other treatments or untreated wounds, wounds treated with T-HDCG had decreased total healing at day 8 and decreased epithelialization and decreased total healing at day 14. Wounds treated with T-HDCG required approximately 3 days longer to completely heal than all other treated and untreated wounds. Application of bacitracinneomycin-polymyxin B ointment did not enhance wound healing. Mean time to complete healing of untreated wounds in all treatment and control groups was 10.00 to 10.14 days. CONCLUSIONS AND CLINICAL RELEVANCE: In rats, topical application of T-HDCG to wounds had a negative effect on second intention healing by delaying the epithelialization process. In mammals, generally, wounds treated topically with taurolidine may need to be treated and monitored for a longer period than other wounds treated with other common woundhealing compounds or untreated wounds.  相似文献

试验旨在探究紫草素凝胶对驴伤口愈合的效果,为皮肤无瘢痕愈合提供参考。用不同浓度的紫草素对体外培养的驴巨噬细胞进行处理,分别检测细胞中TGF-β1、IL-10、TNF-α和IL-6的mRNA与细胞上清液中蛋白的表达水平,以确定紫草素的最佳添加浓度;建立驴皮伤口模型,在伤口表面涂抹紫草素凝胶,拍照、HE染色观察伤口愈合情况,并检测驴皮肤中伤口愈合相关基因和血清的表达水平。结果表明,与对照组相比,添加不同浓度的紫草素均能降低相关炎症因子的表达,其中10 μmol/L紫草素效果最佳。在驴皮伤口上涂抹紫草素凝胶第11天时,紫草素组的伤口收缩率极显著高于对照组(P<0.01);HE染色发现,第11天时紫草素中的组织逐渐排列整齐;第60天时,紫草素组中表皮层的厚度相对减少;实时荧光定量PCR结果表明,第11天时,紫草素组中TGF-β1、IL-10 mRNA表达极显著升高(P<0.01),TNF-α、IL-6 mRNA的表达极显著降低(P<0.01);第3天时,紫草素组血清中TGF-β1、IL-10的浓度升高,TNF-α、IL-6浓度极显著降低(P<0.01);第7天时,紫草素组TGF-β1浓度显著降低(P<0.05),TNF-α浓度极显著降低(P<0.01),且IL-10的浓度显著升高(P<0.05)。综上,与对照组相比,添加不同浓度的紫草素均能降低相关炎症因子的表达,其中10 μmol/L紫草素效果最佳,能促进驴皮伤口的愈合。  相似文献

The effects of topical administration of polysaccharides isolated from fungus, Phellinus gilvus (PG) on the healing of rat dermal wounds were assessed. In 10 Sprague-Dawley (SD) rats, six 6 mm diameter defects were made with a punch biopsy appliance. After 24 hr, test substances were applied to the defects twice a day: 0.025, 0.25, and 2.5% polysaccharides from PG (PG0.025, 0.25, and 2.5 groups), Madecassol ointment (MC group), aqueous gel (AG group) and no treatment (control group). Six days postoperatively, the contraction and reepithelialization of the wound surface were assessed. Wound diameter was significantly reduced in all PG groups (P<0.05). Complete epithelialization and macrophages were noted in the PG0.25 group, as compared to the control group. We conclude that polysaccharides isolated from PG have significant dermal wound healing effects, and this investigation suggests the potential clinical application of PG as a wound healing agent.  相似文献

The enantiomeric naphthoquinones alkannins and shikonins (A/S) have been established as potent wound healing agents. The purpose of this study was to evaluate the effectiveness of an A/S based ointment for humans on second intention wound healing in the dog, as compared to wound flushing with Lactated Ringer's solution (LRS). Ten mixed breed dogs, aged 2 to 5 y, were used. One 2.5 × 2.5 cm full-thickness skin defect was created on the lateral aspect of each arm for subjective evaluation, laser-Doppler flowmetry (LDF), and planimetry. Additionally, 3 matching 2 × 2 cm wounds were created on opposite sides of the dorsal midline for histologic evaluation. Wounds were treated once daily with the A/S based ointment on the right side and by flushing with LRS on the left until healed (about 20 d). During the healing process, tissue perfusion (mean LDF value) was found to be significantly higher on the side treated with the A/S based ointment compared with the LRS-treated side. Histologically, angiogenesis (on days 4 and 11), collagen production score (on days 4, 11, and 20), and epithelial thickness score (on day 11) were significantly higher in the wounds treated with the A/S based ointment. Wound size, as evaluated by planimetry, decreased significantly from day 0 to day 20 on both sides, but no significant differences were found between the A/S based ointment and LRS-treated wounds.  相似文献

Wound healing can be divided into immediate (zero to 1 hour), early (1 to 24 hours), intermediate (1 to 7 days), and late (greater than 7 days) stages. Many physical and physiologic events occur simultaneously and sequentially during these stages to produce the final wound scar. The processes of skin retraction, scab formation, would debridement, wound contraction, epithelial migration and proliferation, fibroplasia, and collagen maturation all must occur for healing to be successful. Many factors affect the size and shape of the resulting scar, including anatomic location and skin tension forces, systemic condition of the patient, blood supply to the wound, nutritional factors, environmental temperature, the presence of systemic drugs, wound infection, motion, wound oxygen gradient, wound moisture, and bandaging. Ideally, each of the factors would occur at a level compatible with optimal healing, but, in many clinical cases, one or more factors compromise normal, rapid healing. When we intervene with therapy, we probably adversely affect another factor in healing, while trying to correct the factor that is out of balance. In these decisions, the effects of our treatment on wound healing should be considered. The trade-off should be weighed and the treatment pursued only as long as necessary to allow healing to progress; then it should be discontinued or changed. With these considerations, it is hoped that we can attain healing at the most rapid physiologic rate.  相似文献

Background A two‐part study examined wound healing and contraction occurring after mulesing and two alternative methods of preventing breech flystrike in sheep. Objective To quantify wound healing using a scoring system and to assess the contractility of the wound bed of the breech after mulesing, cetrimide‐intradermal treatment and application of clips. Method The study group of 30 mulesed, 30 cetrimide‐intradermal treated, 30 control and 10 clip‐treated sheep were humanely killed at six time points from 3 to 47 days after each treatment. Wound healing post treatment was assessed using a scoring system, and contractility was assessed by the quantification of myofibroblast expression. Statistical analyses allowed comparisons of temporal wound healing and contraction between treatment groups. Results Mulesing wounds healed faster in the first 11 days, but by 19 days wound healing was similar between the mulesing and cetrimide‐intradermal groups. By 32 days, all three treatment groups had similar wound healing scores. There was greater myofibroblast expression in the mulesing group in the first 11 days after treatment, but by 19 days expression was similar in both the mulesing and cetrimide‐intradermal groups. The clip group had significantly less myofibroblast expression from 32 days after treatment. Conclusion Wound healing is initially most rapid after mulesing, but there are similar wound healing scores in the mulesing and cetrimide‐intradermal treatment groups by 19 days. Both mulesing and the cetrimide‐intradermal treatment induce a similar amount of wound bed contraction, with less contraction observed after application of clips.  相似文献

The effect of low reactive-level laser therapy (LLLT) with a He-Ne laser on operative wound healing was investigated in a rat model. 10-millimeter surgical wounds were created on the backs of Sprague Dawley rats, and animals were assigned to one of eleven groups (n=5). Ten groups received either 8.5 mW or 17.0 mW irradiation of 15 seconds LLLT a day with one of five different irradiation frequencies, i.e. daily (from the 1st to 6th day following surgery), every other day (the 1st, 3rd, and 5th day), on only the 1st day, on only the 3rd day, and on only the 5th day; the 1st day was the day following the surgery. The control group received no irradiation. A skin specimen was harvested from the dorsal thoracic region on the 7th day to measure the rupture strength. The control group had the lowest rupture strength (5.01 N), and the 17.0 mW every other day irradiation group had the highest rupture strength (13.01 N). Statistical differences were demonstrated in the 8.5 mW irradiation setting between the every other day irradiation group and the control group (p<0.05); and in 17.0 mW irradiation setting between the every day irradiation, the every other day, and the 1st day only groups vs. the control group (p<0.01). Histological examination demonstrated that wound healing in the 17.0 mW every other day irradiation group was promoted most significantly such as the prevention of excessive inflammation, increased formation of collagen fibers, and recovery in continuity of tissues. The control group showed poor wound healing and the other experimental groups showed intermediate healing. Thus LLLT with a He-Ne laser was found to promote the healing of operative wounds in the present rat model, in which the most favorable application of LLLT was the 17.0 mW setting of 15 seconds a day with a frequency of every other day.  相似文献

Three sets of paired circular and square full-thickness skin wounds were made on the dorsum of the metacarpus (n = 48) of 8 horses. Each wound was 6.25 cm2 in area. The wounds were treated topically with an ointment, nonadherent dressing, and bandaged with a snug elastic wrap. Wounds were photographed every other day until healing was complete. Wound areas were measured and exponential and linear wound healing models were applied to the wound healing data generated. Wound healing variables measured for each wound were: number of days to healing, maximal size attained, rate of wound contraction (calculated by use of first-order and linear models), final wound size, and percentage of wound that healed by contraction. The exponential model fit the data significantly better than the linear model. The maximal size attained by circular wounds was significantly smaller than the maximal size attained by square wounds. Wound shape did not influence the rate of wound healing. On the basis of our findings, conversion of circular defects to square defects would not speed wound healing.  相似文献

OBJECTIVE: To evaluate the effects of topical glycyl-L-histidyl-L-lysine tripeptide-copper complex (TCC; Iamin 2% Gel; Procyte Corporation, Redmond, WA) on healing in ischemic open wounds. STUDY DESIGN: Experimental study. SAMPLE POPULATION: Twenty-four adult male Sprague-Dawley rats. METHODS: Rats were divided into 3 groups: topical TCC, topical TCC vehicle (hydroxypropyl-methylcellulose), and no treatment (control). Six-mm-diameter, full-thickness wounds were created within an ischemic bipedicle skin flap on the dorsum of each rat. Each day, for 13 days, wound margins were traced, and the TCC and TCC vehicle groups were treated topically. Tracings were scanned, and wound perimeter and area were calculated. On days 6, 10, and 13, selected wounds were biopsied and analyzed for tumor necrosis factor alpha (TNF-alpha) and matrix metalloproteinases (MMP) 2 and 9. RESULTS: A significant decrease in wound area was seen in the TCC group, but not the vehicle group, when compared with the control group on days 3 to 5, 6 to 9, and 11 to 13 and when TCC was compared with TCC vehicle on days 3 and 9. On day 13, initial wound area had decreased by 64.5% in the TCC group, 45.6% in the vehicle group, and 28.2% in the control group. On days 6, 10, and 13, TCC-treated wounds contained significantly lower concentrations of TNF-alpha and MMP-2 and MMP-9 than control wounds. CONCLUSION: Topical TCC resulted in accelerated wound healing in ischemic open wounds. CLINICAL RELEVANCE: Topical TCC is an effective stimulant of healing of ischemic open wounds in rats and may have an application for the treatment of chronic wounds in other species. Clinical evaluation of topical TCC is warranted.  相似文献

Horses with severe and deep lacerations are represented by ten cases in which treatment emphasized the use of bovine collagen preparations to promote controlled second-degree repair. Traumatized areas were tarsal, metatarsal, neck, forearm, metacarpal and pastern. Wound size changes were recorded. Depending on the wound type, the site was treated with antibiotic-steroid ointment, organic acid cream, sterile collagen particles, suspension or dressings protected by hydrogel dressing, non-adherent pads, occlusive skin dressings, roll gauze and elastic tape. In three cases, a fiberglas cast was applied over a hind leg wound and lacerated tendon for stability. When controlled granulation of the deeper wounds reached skin level, the area often was stabilized by only cyanoacrylate spray. As these cases presented a wide range of trauma each with a unique history, healing rates were based on initial measurements. An overall progression of wound reduction occurred at a predictable rate. The exogenous collagen formulations were used to stimulate controlled granulation, ie. to “jump start” the healing process. Collagen particles, suspension or dressings were packed into depressions, placed under suture lines, secured over abraded tissue, and placed under protective bandage or cast. To further evaluate the use of cyanoacrylate tissue spray in wound treatments, an additional ten cases are presented. The variety of wounds were produced experimentally in Center-owned ponies or provided as clinical cases. Wound size changes and healing progress were recorded. Wounds occurred on the neck, abdomen, metacarpal, metatarsal, fetlock and pastern areas. Depending on wound type, the site was treated with cyanoacrylate only; or treated as above until controlled granulation attained skin level. In one case punch grafts of skin were transferred from one foreleg to the opposite with the horse standing. Cyanoacrylate spray provided a water proof barrier protecting the wound from dirt, debris and insects as well as stabilizing full-thickness skin lacerations by bridging normal to traumatized skin allowing uninterrupted granulation and epithelialization. The use of a neck cradle prevented wound disturbance and stall confinement aided stabilization.  相似文献

The effects of four topical medications on the rate and character of healing of cutaneous wounds were studied in six common garter snakes (Thamnophis sirtalis) held at an ambient temperature of 30 degrees C. Two sets of five 6 to 8 mm round excisional wounds, four test and one control site in each set, were created on the dorsolateral body wall of each snake. Wounds were examined daily and treated for ten days, then the snakes were killed and sections of all wounds were examined by light microscopy. Composite scores, derived by ranking each treatment group in relation to the control group (control score = 0) for each of 22 characteristics associated with wound healing, were used to compare the overall effects of each treatment. Statistical comparisons were made between groups for 20 characteristics. Wounds treated with a polyurethane film merited a score of +12 and had significantly more advanced healing than untreated controls for three characteristics. Wounds treated with an ointment containing scarlet red scored +6 but healing was not significantly greater than controls. Wounds treated with an antibacterial spray powder and an antibacterial ointment healed more slowly than controls and had scores of -6 and -12 respectively.  相似文献

The objective of this study was to evaluate the efficacy of topical application of a hyaluronan (HA) derivative in wound healing with respect to the rate of epithelialization, fibroplasia, angiogenesis and contraction, magnitude of the local inflammatory response, local expression of transforming growth factor-β 1 and 3 (TGF-β 1 and 3), tumor necrosis factor-α (TNF-α), and collagen type III deposition. In six healthy adult horses, six full-thickness skin wounds were created on the dorsal aspect of both metacarpi using a sterile template. Sites were sampled at 0, 1, 2, 5, 14, 21, and 35 days following wounding. Wounds on one limb were dressed with commercially available esterified HA fleece under a nonadherent dressing. The opposite limb was covered with the nonadherent dressing alone (control). Images of the most proximal wounds were used to determine the area of total healing and the relative contributions of epithelialization and contraction to healing. At each sample time, a control and treatment biopsy were taken for histological evaluation and special stains. All samples were evaluated for degree of inflammation, fibroplasia and angiogenesis; in situ hybridization for type III collagen, TGFβ1 and 3, and immunohistochemistry for TNF-α. Mean percentages of total wound healing, epithelialization, and wound contraction were not significantly different between control and treatment groups. In treated horses, initial wound expansion was significantly decreased during the first 2 weeks. Mononuclear cell numbers, counted in the granulation tissue, increased in both control and treated limbs over the entire course of the study. However at day 35 the macrophage numbers counted in the treated horses were significantly increased compared with the control limbs (P < .05). Although not statistically significant, relative staining for type III collagen in the treated wounds was less than that of control wounds. Results of the present study do not support a benefit of an exogenous HA-derivative in the healing of distal limb wounds in horses. The shortcomings of the study design are discussed.  相似文献

OBJECTIVE: To determine whether povidone iodine ointment or 2 forms of silver sulfadiazine applied topically to wounds of the distal aspect of the limbs in horses affect the rate of second intention healing and to evaluate the additional influence of bandaging with these antimicrobials on granulation tissue formation. ANIMALS: 6 healthy adult horses. PROCEDURE: Six standardized 2.5-cm2 skin wounds/horse were distributed between the dorsomedial surfaces of the metacarpi and metatarsi. One of the following 6 treatments was applied to each wound: 1% silver sulfadiazine cream with bandage, 1% silver sulfadiazine slow-release matrix with bandage, 1% silver sulfadiazine slow-release matrix without bandage, povidone-iodine ointment with bandage, untreated control with bandage, and untreated control without bandage. Wound area, granulation tissue area, and perimeter were measured by use of planimetry software applied to digital images. Exuberant granulation tissue was excised when present. Days until healing, rate of healing parameter, rate of contraction, and epithelialization were compared among wound treatment groups. RESULTS: Healing parameters and mean days to healing did not differ significantly among any of the wound treatment groups. Percentage wound contraction and rate of epithelialization were similar among wound treatments. All bandaged wounds produced exuberant granulation tissue, which was surgically excised; none of the unbandaged wounds produced exuberant granulation tissue. CONCLUSIONS AND CLINICAL RELEVANCE: When exuberant granulation tissue is removed, rates of epithelialization and wound contraction were not different among wound treatment groups, whether bandaged or unbandaged. Topical application of 1% silver sulfadiazine slow-release matrix on unbandaged wounds induced the same result as medications applied beneath bandages, but without exuberant granulation tissue formation.  相似文献

This study was carried out to evaluate the healing potential of water-soluble extract of Plantago lanceolata as a topical ointment on experimental tendinitis in burros. Tendinitis was induced by injection of 2,500 U of bacterial collagenase in the superficial digital flexor tendon of both forelimbs of six male burros. Tendinitis was confirmed ultrasonographically. 48 hours after collagenase injection, two grams of 10% P lanceolata ointment was applied on one limb (treatment group) and the same amount of eucerin (the vehicle of the ointment) on the other limb (control group), every other day for 6 weeks. Ultrasonographic examination of the tendon was performed weekly during the study and mean cross-sectional area of the superficial digital flexor tendon was measured, which was found to increase significantly after injection of collagenase in both groups. From the third week, it decreased significantly in the treatment group in comparison with the control group. At the end of the study, the animals were euthanized for histopathological study of the tendons. In control group, degenerative changes in tendon could be seen, whereas significant degree of healing was observed in the treatment group. It can thus be concluded that the P lanceolata ointment was effective in promoting the healing process of the tendon.  相似文献

In this study the clinical and histopathological effects of topically applied tripeptide-copper complex (TCC) and zinc oxide on open-wound healing in rabbits was evaluated. Eighteen New Zealand rabbits were divided into three groups: TCC, zinc oxide and no treatment. One full thickness wound was created on each side of the dorsal midline in each rabbit. Wound margins were traced on days 0, 7, 14, and 21 and topical TCC and zinc oxide were applied daily for 21 days to each rabbit in the respective treatment groups. The mean unhealed wound area was significantly smaller in the TCC than in the zinc oxide group on day 7, but it was significantly smaller in the TCC group than in to the control group on days 7, 14 and 21. The mean percentage of wound contraction on day 7 was significantly higher in the TCC than in the zinc oxide group; however, it was significantly higher in the TCC group than in the control group on days 7, 14 and 21. Median time for the coverage of the wound bed with granulation tissue was significantly shorter in the TCC group than in the other groups. Filling of the open wound with granulation tissue to skin level was significantly slower in the control group than in the other two groups. Neutrophil counts decreased regardless of the group in parallel with healing, while neovascularization was best observed in the TCC group. The results suggest that TCC is a better choice in the treatment protocols of open wounds in rabbits than zinc oxide.  相似文献
